The herbicide Simazine is widely used in forestry nurseries.Its effect on the incidence of ectomycorrhizae on pine seedlingroot systems was investigated. Under field (nursery) conditions,at routine rates of application, Simazine does not inhibit mycorrhizaldevelopment in Pinus sylvestris and P. nigra, and it may, undersome conditions, actually enhance mycorrhizal formation. Sucha stimulatory effect was also obtained with pot-grown seedlingstreated with the herbicide. It is suggested that soil type andclimatic factors might be involved in determining the responseto Simazine. The mode of stimulation of mycorrhizal developmentby this herbicide is not understood, but it is not via suppressionof weed growth. Field and laboratory investigations were carried out to determine the influence of two commercial herbicides Lindex and Simazine on symbiotic N2 fixation, the photosynthetic apparatus, percentage of proteins, and grain yield of lupins (Lupinus albus L. cv. Multolupa). The herbicides were added (3 kg commercial product in 9001 per ha) two weeks after sowing. The nodulated roots were tested for nitrogenase activity by the acetylene reduction assay (ARA) at the first and second flowering. The ARA values decreased with herbicide application. The decrease was smaller in the Simazine treatments with inoculated seeds. The ARA for plants treated with Lindex did not increase with inoculation. Nitrogenase activity was greater in the non-inoculated plants growing together with weeds. The effect of Simazine on the photosynthetic apparatus proved to be more toxic than that of Lindex, not only considerably reducing the size of the chloroplasts but also affecting the grana structure. The latter appeared as an electron-dense compact mass. There were no visible alterations in the photosynthetic apparatus of plants treated with Lindex. However starch, which was not present in the control leaves, accumulated. No ultrastructural changes were observed in the nodule cells treated with Lindex. Simazine altered nodule cells by causing vesicle formation, degeneration of bacteria, and by decreasing the number of N2-fixing bacteroids. The grain yield decreased with both herbicide treatments, the decrease being lower in the inoculated plants, especially in the Simazine treatment. Our results suggest that each herbicide had a different effect on N2 fixation, the photosynthetic apparatus and grain yield. Laboratory studies indicated that cyanazine and simazine did not adversely affect the growth of Bradyrhizobium.  相似文献

Summary Production of lysine and methionine byAzotobacter chroococcum strain H23 andA. vinelandii strain ATCC 12837 was studied in chemically-defined medium and dialysed-soil medium, amended with different concentrations of Simazine. Responses on production due to Simazine were different for each strain and were fairly conditioned by culture media composition. Quantitative production of amino acids was significantly affected by the xenobiotic only at higher doses (50–100,g/ml). The effect of Simazine on methionine production by strain H23 was very pronounced when bacteria were grown in dialysed-soil medium, which was specially formulated to reproduce the natural habitat of the organisms.  相似文献

Extruded pellets containing activated carbon (AC) can be used to sow native seeds while simultaneously applying herbicide to control invasive species. Incorporating AC in pellets has been demonstrated to protect native seeds; however, there may be unintended detrimental impacts to seedling emergence. We aimed to optimize seed position within pellets to maximize emergence and survival of the perennial shrub Jacksonia furcellata. Seeds were positioned at 2 mm (top), 6 mm (middle), and 12 mm (bottom) within pellets (with or without AC), sown on or below the soil surface, and compared to non-pelleted seeds sown under the soil surface in the equivalent positions (2, 6, and 12 mm depth). Trays were treated with a pre-emergent herbicide (Simazine) or left unsprayed. Emergence (without herbicide) was significantly higher from seeds positioned at the bottom of pellets without AC sown on the soil surface (70%), compared to non-pelleted seeds sown at the bottom (12 mm below the soil surface; 57%). However, emergence was inhibited when seeds were positioned in the middle (6 mm) of pellets with AC (32%). When treated with Simazine, survival was highest from seeds positioned at the bottom of pellets with AC (60%), compared to pellets without AC (15%) and non-pelleted seeds sown at the bottom (12 mm below the soil surface; 15%). Jacksonia furcellata seeds positioned at the bottom of pellets, sown on the soil surface, shows promise to minimize negative impacts to emergence, and maximize herbicide protection. Further testing with additional species is required to refine pellet production (e.g. recipe, extrusion, and shape) for optimal emergence.  相似文献

The Cox proportional hazards regression model is the most popular approach to model covariate information for survival times. In this context, the development of high‐dimensional models where the number of covariates is much larger than the number of observations ( $p \,{\gg }\, n$ ) is an ongoing challenge. A practicable approach is to use ridge penalized Cox regression in such situations. Beside focussing on finding the best prediction rule, one is often interested in determining a subset of covariates that are the most important ones for prognosis. This could be a gene set in the biostatistical analysis of microarray data. Covariate selection can then, for example, be done by L1‐penalized Cox regression using the lasso (Tibshirani ( 1997 ). Statistics in Medicine 16 , 385–395). Several approaches beyond the lasso, that incorporate covariate selection, have been developed in recent years. This includes modifications of the lasso as well as nonconvex variants such as smoothly clipped absolute deviation (SCAD) (Fan and Li ( 2001 ). Journal of the American Statistical Association 96 , 1348–1360; Fan and Li ( 2002 ). The Annals of Statistics 30 , 74–99). The purpose of this article is to implement them practically into the model building process when analyzing high‐dimensional data with the Cox proportional hazards model. To evaluate penalized regression models beyond the lasso, we included SCAD variants and the adaptive lasso (Zou ( 2006 ). Journal of the American Statistical Association 101 , 1418–1429). We compare them with “standard” applications such as ridge regression, the lasso, and the elastic net. Predictive accuracy, features of variable selection, and estimation bias will be studied to assess the practical use of these methods. We observed that the performance of SCAD and adaptive lasso is highly dependent on nontrivial preselection procedures. A practical solution to this problem does not yet exist. Since there is high risk of missing relevant covariates when using SCAD or adaptive lasso applied after an inappropriate initial selection step, we recommend to stay with lasso or the elastic net in actual data applications. But with respect to the promising results for truly sparse models, we see some advantage of SCAD and adaptive lasso, if better preselection procedures would be available. BACKGROUND: Nitrofen is a diphenyl ether that induces congenital diaphragmatic hernia (CDH) in rodents. Its mechanism of action has been hypothesized as inhibition of the retinaldehyde dehydrogenase (RALDH) enzymes with consequent reduced retinoic acid signaling. METHODS: To determine if nitrofen inhibits RALDH enzymes, a reporter gene construct containing a retinoic acid response‐element (RARE) was transfected into HEK‐293 cells and treated with varying concentrations of nitrofen in the presence of retinaldehyde (retinal). Cell death was characterized by caspace‐cleavage microplate assays and terminal deoxynucleotidyl transferase dUTP nick end‐labeling (TUNEL) assays. Ex vivo analyses of cell viability were characterized in fetal rat lung explants using Live/Dead staining. Cell proliferation and apoptosis were assessed using fluorescent immunohistochemistry with phosphorylated histone and activated caspase antibodies on explant tissues. Nile red staining was used to identify intracellular lipid droplets. RESULTS: Nitrofen‐induced dose‐dependent declines in RARE‐reporter gene expression. However, similar reductions were observed in control‐reporter constructs suggesting that nitrofen compromised cell viability. These observed declines in cell viability resulted from increased cell death and were confirmed using two independent assays. Ex vivo analyses showed that mesenchymal cells were particularly susceptible to nitrofen‐induced apoptosis while epithelial cell proliferation was dramatically reduced in fetal rat lung explants. Nitrofen treatment of these explants also showed profound lipid redistribution, primarily to phagocytes. CONCLUSIONS: The observed declines in nitrofen‐associated retinoic acid signaling appear to be independent of RALDH inhibition and likely result from nitrofen induced cell death/apoptosis. 【目的】套索肽作为一类核糖体翻译后修饰肽(RiPPs)广泛分布于放线菌中,以其独特的修饰结构和多样的生理活性受到了广泛的关注。为了更好地研究未知的套索肽,期望开发基于链霉菌的无细胞转录翻译平台(下称“无细胞平台”)实现无细胞合成套索肽或其前体肽。【方法】首先尝试以不同的链霉菌构建无细胞合成平台,并以绿色荧光蛋白为报告蛋白对平台产率进行优化;在构建合适稳定的表达体系后,将包含有套索肽生物合成基因的质粒引入体系中以探索套索肽的无细胞合成。【结果】在对基于模式菌株Streptomyces lividans TK24的无细胞体系进行制备工艺、体系组分、反应条件等多个参数进行优化后,该体系最高能达到90μg/mL的荧光蛋白表达量;基于该体系成功表达了目标套索肽的前体肽,并通过融合SUMO标签增加前体肽在该体系中的稳定性。【结论】本研究成功构建了一类链霉菌无细胞平台,为丰富来源的基因表达提供了可能性。尽管该体系在对表达套索肽未知蛋白的适用性上仍有待进一步提升,但无细胞平台在天然产物的探索中将起到越来越重要的作用。  相似文献

BACKGROUND : Pulmonary hypoplasia (PH) is the main cause of mortality in newborns with congenital diaphragmatic hernia (CDH). Prenatal administration of retinoic acid (RA) stimulates alveologenesis in the nitrofen‐induced pulmonary hypoplasia. Insulin‐like growth factor receptors (IGFRs) play a crucial role in alveologenesis during lung development. We recently demonstrated that IGFRs were downregulated in later stages of lung development in the nitrofen CDH model. Several studies suggest the ability of RA to regulate insulin‐like growth factor signaling. We hypothesized that IGFRs pulmonary gene expression is upregulated after the administration of RA in the nitrofen‐induced CDH model. METHODS : Pregnant rats were exposed to either olive oil or nitrofen on day 9 (D9) of gestation. RA was given intraperitoneally on days D18, D19, and D20. Fetal lungs were dissected on D21 and divided into control, control + RA, CDH, and CDH + RA group. IGFRs gene and protein expression were determined using RT‐PCR and immunohistochemistry. RESULTS : mRNA expression levels of IGFRs were significantly increased in control + RA and CDH + RA compared with CDH group. Immunoreactivity of IGFRs was markedly increased in control + RA and CDH + RA compared with CDH lungs. CONCLUSIONS : Upregulation of pulmonary gene and protein expression of IGFRs after prenatal RA treatment in the nitrofen model suggests that RA may promote lung growth by stimulating IGFRs mediated alveologenesis. To investigate nematode establishment and persistence, dauer juveniles (DJs) of Heterorhabditis bacteriophora were applied at 50 cm-2 in different crops in June and July with conventional spraying equipment and 420 l water ha-1. Application hardly had any effects on survival and infectivity. The number of DJs reaching the soil was assessed and the establishment and persistence recorded by baiting soil samples with larvae of the wax moth Galleria mellonella. The better the plant canopy was developed the fewer DJs reached the soil during application. Whereas in pasture 77% and in potatoes 78% of the applied nematodes reached the soil, in wheat and peas little less than 50%, in oil-seed rape only 5% and in lupine 6% were recorded. Between 50 and 60% of the soil samples contained H. bacteriophora a month after application with the exception of wheat (>90%) and potatoes (<5%) indicating that the number of nematodes reaching the soil during application had no influence on their establishment in the soil. Probably DJs can survive in the plant canopy and reach the soil later after application. The percentage of nematode-positive soil samples dropped considerably after tillage. In potatoes no nematodes were recovered after two months, which probably was also due to the intensive movement of the soil. Although nematodes are susceptible to freezing, temperatures below 0°C during the winter did not extinguish the H. bacteriophora population. In field crops EPN usually persisted not much longer than one year. The longest persistence of H. bacteriophora was detected 23 months after release in beans followed in rotation by wheat with red clover as cover crop. In this field larvae of the pea weevil Sitona lineatus (Coleoptera: Curculionidae) were detected in soil samples and found infected with the released nematode population. In the laboratory the field soils were tested for persistence of H. bacteriophora at 8°C and a half-life of 24.8 days was recorded in the absence of host insects and plants. Thus long-term persistence in the field was a result of recycling in host insects, which could not be detected in other crops than beans and clover. As H. bacteriophora seems to be restricted in its host potential, this species disappears after release once the host population is not available anymore.  相似文献

Bacillus subtilis RB14‐C, which produces the antibiotic iturin A, was investigated for its effectiveness as a biocontrol agent against Rhizoctonia solani infecting tomato using seed coating and/or direct introduction of the bacteria to the soil. The ability of RB14‐C to colonize plant roots and produce iturin A in soil, depending on the method of bacterial application, was also determined. Seed coating and the combined treatment (soil and seed bacterization) did not protect seedlings against damping‐off caused by R. solani. By contrast, RB14 introduced only to the soil controlled the disease. The total number of RB14‐C bacteria on the roots of plants grown from coated seeds was significantly lower than on the roots of plants grown in soil mixed with the bacteria. In the combined treatment, application of B. subtilis with seeds to soil preinoculated with this bacterium, at first suppressed the population of RB14‐C in the soil. Then the colonization was generally uniform. The concentration of iturin A in non‐planted soil was highest at the beginning of the experiment (i.e. after application of the bacterial suspension) but then decreased, and was undetectable 3 days after incubation. However, after seed planting the antibiotic was produced again around young roots. Bacteria introduced to the soil as a seed coating also released the antibiotic around the seeds.  相似文献

The persistence of the nematophagous fungus Paecilomyces lilacinus Samson strain 251 (PL251) and the effect of application rate, substrate type, as well as the presence of the nematode host on its dynamics after application to the soil were investigated under controlled conditions. In all experiments, increase of P. lilacinus colony forming units after application was not found. In contrast, a gradual decline in fungal densities over time was observed. Application rate had no significant effect on the dynamics of the fungal population. Likewise, P. lilacinus density decline in soil was not significantly affected by the presence of the nematode host. Substrate type had a significant effect on P. lilacinus persistence in soil. The fungal agent persisted longer in silty loam and clay soil, with reduced persistence when sand was added to field soil. Conversely, when organic substrate was added to pure sand, persistence was significantly increased. Although persistence of fungal biocontrol agents in soil depends on various biotic and abiotic conditions, baseline data on persistence such as those reported in this study are helpful for biocontrol and environmental risk assessment and merit further study.  相似文献

Soil solarization in combination with introduction of biocontrol agents (BCA) was evaluated as a potential disease management strategy for tomato damping-off caused by Pythium spp. A rifampicin resistant Pseudomonas fluorescens strain (PfT-8) and a carbendazim resistant Trichoderma harzianum strain (ThM-1) were introduced into soil following solarization. Tomato seeds were planted into treated field plots. The influence of soil solarization and application of biocontrol agents on damping-off incidence, plant biomass, rhizosphere population of introduced antagonists, and native Pythium spp. was assessed by two consecutive field trials. Damping-off incidence was significantly reduced in solarized plots compared to control. Soil inoculation of biocontrol agents into solarized plots resulted in the highest suppression of damping-off incidence (PfT-8 up to 92%; ThM-1 up to 83%), and increase in plant biomass (PfT-8 up to 66%; ThM-1 up to 48%) when compared to un-solarized control plots. Rhizosphere population of introduced biocontrol agents gradually increased (PfT-8 up to 102% and ThM-1 up to 84%) in solarized soils when compared to unsolarized control. The population of Pythium spp in rhizosphere soil was reduced up to 55% in solarized plots; whereas, application of BCA to solarized soils reduced the rhizosphere population of Pythium spp. by 86 and 82% in P. fluorescens and T. harzianum applied plots respectively.  相似文献

Summary The rhizosphere microflora of arecanut palm under continuous application of organic manures and inorganic fertilizers was studied. The nutrients applied are 100 g N, 40 g P2O5 and 140 g K2O/palm/year in the form of organics and inorganics. The application of organic manure increased the microbial population. The increase in microbial population was observed between the rhizosphere samples collected at 0–30cm and 30–60 cm depths. The effects of triazines on growth, 14CO2-fixation and translocation of 14C-assimilates by young Pinus seedlings were investigated. Post-emergenceroot application of moderate and high concentrations of simazine and atrazine resulted in severe toxicity to the seedlings of Pinus nigra and P. coniorta as depicted by reduction in dry weight, chlorosis, wilting and mortality. Atrazine was more toxic than simazine and P. nigra was more susceptible than P. contorta to both the triazines. Simazine treatment to the roots of P. nigra seedlings, not only caused inhibition of 14CO2-fixation but also reduced the short-term downward transloca-tion of l4 C-assimilates. Various possible mechanisms whereby simazine reduced the downward movement of I4C-assimilates in Pinus seedlings are discussed.  相似文献

s-Triazine herbicides are widely used for weed control, and are persistent in soils. Nitrification is an essential process in the global nitrogen cycle in soil, and involves ammonia-oxidizing Bacteria (AOB) and ammonia-oxidizing Archaea (AOA). In this study, we evaluated the effect of the s-triazine herbicide simazine on the nitrification and on the structure of ammonia-oxidizing microbial communities in a fertilized agricultural soil. The effect of simazine on AOB and AOA were studied by PCR-amplification of amoA genes of nitrifying Bacteria and Archaea in soil microcosms and denaturing gradient gel electrophoresis (DGGE) analyses. Simazine [50?μg g(-1) dry weight soil (d.w.s)] completely inhibited the nitrification processes in the fertilized agricultural soil. The inhibition by simazine of ammonia oxidation observed was similar to the reduction of ammonia oxidation by the nitrification inhibitor acetylene. The application of simazine-affected AOB community DGGE patterns in the agricultural soil amended with ammonium, whereas no significant changes in the AOA community were observed. The DGGE analyses strongly suggest that simazine inhibited Nitrosobacteria and specifically Nitrosospira species. In conclusion, our results suggest that the s-triazine herbicide not only inhibits the target susceptible plants but also inhibits the ammonia oxidation and the AOB in fertilized soils.  相似文献

Cross-validation is the standard method for hyperparameter tuning, or calibration, of machine learning algorithms. The adaptive lasso is a popular class of penalized approaches based on weighted L1-norm penalties, with weights derived from an initial estimate of the model parameter. Although it violates the paramount principle of cross-validation, according to which no information from the hold-out test set should be used when constructing the model on the training set, a “naive” cross-validation scheme is often implemented for the calibration of the adaptive lasso. The unsuitability of this naive cross-validation scheme in this context has not been well documented in the literature. In this work, we recall why the naive scheme is theoretically unsuitable and how proper cross-validation should be implemented in this particular context. Using both synthetic and real-world examples and considering several versions of the adaptive lasso, we illustrate the flaws of the naive scheme in practice. In particular, we show that it can lead to the selection of adaptive lasso estimates that perform substantially worse than those selected via a proper scheme in terms of both support recovery and prediction error. In other words, our results show that the theoretical unsuitability of the naive scheme translates into suboptimality in practice, and call for abandoning it.  相似文献

The effects of three selected agrochemicals on bacterial diversity in cultivated soil have been studied. The selected agrochemicals are Cerox (an insecticide), Ceresate and Paraquat (both herbicides). The effect on bacterial population was studied by looking at the total heterotrophic bacteria presence and the effect of the agrochemicals on some selected soil microbes. The soil type used was loamy with pH of 6.0–7.0. The soil was placed in opaque pots and bambara bean (Vigna subterranean) seeds cultivated in them. The agrochemicals were applied two weeks after germination of seeds at concentrations based on manufacturer’s recommendation. Plant growth was assessed by weekly measurement of plant height, foliage appearance and number of nodules formed after one month. The results indicated that the diversity index (Di) among the bacteria populations in untreated soil and that of Cerox-treated soils were high with mean diversity index above 0.95. Mean Di for Ceresate-treated soil was 0.88, and that for Paraquattreated soil was 0.85 indicating low bacterial populations in these treatment-type soils. The study also showed that application of the agrochemicals caused reduction in the number of total heterotrophic bacteria population sizes in the soil. Ceresate caused 82.50% reduction in bacteria number from a mean of 40 × 105 cfu g−1 of soil sample to 70 × 104 cfu g−1. Paraquat-treated soil showed 92.86% reduction, from a mean of 56 × 105 cfu g−1 to 40 × 104 cfu g−1. Application of Cerox to the soil did not have any remarkable reduction in bacterial population number. Total viable cell count studies using Congo red yeast-extract mannitol agar indicated reduction in the number of Rhizobium spp. after application of the agrochemicals. Mean number of Rhizobium population numbers per gram of soil was 180 × 104 for the untreated soil. Cerox-treated soil recorded mean number of 138 × 104 rhizobial cfu g−1 of soil, a 23.33% reduction. Ceresate- and Paraquat-treated soils recorded 20 × 104 and 12 × 104 cfu g−1 of soil, respectively, representing 88.89% and 93.33% reduction in Rhizobium population numbers. Correspondingly, the mean number of nodules per plant was 44 for the growth in untreated soil, 30 for the plant in the Cerox-treated soil, 8 for the plant in Paraquat-treated soil and 3 for the plant in Ceresate-treated soil. The study has confirmed detrimental effect of insecticide on bacterial populations in the soil. Total heterotrophic counts, rhizobial counts as well as the number of nodules of all samples taken from the chemically treated soils were all low as compared to values obtained for the untreated soil. However, the effect of the insecticide was minimal in all cases as compared to the effects of the herbicides on the soil fauna. Indiscriminate use of agrochemicals on farms can therefore affect soil flora and subsequently food production.  相似文献

Superior antagonistic activity against axenic Microcystis aeruginosa PCC7806 was observed with Paucibacter sp. B51 isolated from cyanobacterial bloom samples among 43 tested freshwater bacterial species. Complete genome sequencing, analyzing average nucleotide identity and digital DNA–DNA hybridization, designated the B51 strain as Paucibacter aquatile. Electron and fluorescence microscopic image analyses revealed the presence of the B51 strain in the vicinity of M. aeruginosa cells, which might provoke direct inhibition of the photosynthetic activity of the PCC7806 cells, leading to perturbation of cellular metabolisms and consequent cell death. Our speculation was supported by the findings that growth failure of the PCC7806 cells led to low pH conditions with fewer chlorophylls and down-regulation of photosystem genes (e.g., psbD and psaB) during their 48-h co-culture condition. Interestingly, the concentrated ethyl acetate extracts obtained from B51-grown supernatant exhibited a growth-inhibitory effect on PCC7806. The physical separation of both strains by a filter system led to no inhibitory activity of the B51 cells, suggesting that contact-mediated anti-cyanobacterial compounds might also be responsible for hampering the growth of the PCC7806 cells. Bioinformatic tools identified 12 gene clusters that possibly produce secondary metabolites, including a class II lasso peptide in the B51 genome. Further chemical analysis demonstrated anti-cyanobacterial activity from fractionated samples having a rubrivinodin-like lasso peptide, named paucinodin. Taken together, both contact-mediated inhibition of photosynthesis and the lasso peptide secretion of the B51 strain are responsible for the anti-cyanobacterial activity of P. aquatile B51.  相似文献

Hirsutella rhossiliensis, a nematophagous fungus, has shown potential in biocontrol of plant-parasitic nematodes. Monitoring the population dynamics of a biocontrol agent in soil requires comprehensive techniques and is essential to understand how it works. Bioassay based on the fungal parasitism on the juveniles of soybean cyst nematode, Heterodera glycines, can be used to evaluate the activity of the fungus but fails to quantify fungal biomass in soil. A real-time polymerase chain reaction (PCR) assay was developed to quantify the fungal population density in soil. The assay detected as little as 100 fg of fungal genomic DNA and 40 conidia g−1 soil, respectively. The parasitism bioassay and the real-time PCR assay were carried out to investigate the presence, abundance and activity of H. rhossiliensis in soil after application of different inoculum levels. Both of the percentage of assay nematodes parasitized by H. rhossiliensis based on the parasitism bioassay and the DNA yield of the fungus quantified by real-time PCR increased significantly with the increase of the inoculum levels. The DNA yield of the fungus was positively correlated with the percentage of assay nematodes parasitized by H. rhossiliensis. The quantitative determination of Macrophomina phaseolina population in an arid soil was carried out utilizing a selective medium. The population of the fungus increased under continuous monocropping with a susceptible host, but considerable reduction was recorded with crop rotation or a change in crop sequence. The effects of fungi, bacteria, actinomycetes, total microbial population, minimum and maximum soil temperature on the survival of M. phaseolina population were studied in the absence of a crop at different soil depths. Highest population and survival rates were recorded at 0–5 cm soil depth. Correlation and path coefficient analyses were carried out to determine the role of individual factors influencing M. phaseolina population. In winter months, low temperature and high actinomycete population were found to be the major factors governing the reduction of the population of M. phaseolina.  相似文献
