Lateral gene transfer is any process in which an organism incorporates genetic material from a phylogenetically distant and reproductively isolated organism. A previous survey demonstrated that two Japanese populations of Monochamus alternatus Hope carried many Wolbachia genes that had been acquired by lateral gene transfer in their own genomes, but were not infected with Wolbachia. To understand the prevalence of Wolbachia infection and laterally transferred Wolbachia genes in this beetle, we performed a broader survey of natural populations covering two subspecies of M. alternatus from Japan (M. alternatus endai) and Taiwan (M. alternatus alternatus). We detected laterally transferred Wolbachia genes in all Japanese and Taiwanese populations of M. alternatus, but no Wolbachia infection in any population. In addition, we confirmed the absence of Wolbachia infection and of transferred Wolbachia genes in two Japanese populations of Monochamus saltuarius Gebler, which is a congeneric relative of M. alternatus. Our findings suggest that the Wolbachia endosymbiont as gene donor has disappeared from M. alternatus, and that the transfer of part of its genome to M. alternatus occurred in the ancestor of M. alternatus before the subspeciation event.  相似文献

The Japanese pine sawyer, Monochamus alternatus , is an important pine forest pest and vector transmitting the pine wilt nematode that causes pine wilt disease. Low temperatures in autumn, winter and spring often differentially affect mortality of M. alternatus larvae. In this paper, we mainly compared the differences of mortality and cold hardening of larvae from different seasons, based on supercooling point (SCP) and cumulative probability of individuals freezing (CPIF). The cold hardening of the larvae from autumn, winter and spring seasons were largely different. Correlations between mortality and CPIF of autumn and spring larvae were highest on day 1/4, and gradually decreased with prolonged exposure duration. This beetle's death mainly resulted from freezing in short exposure duration. However, the correlation between mortality and CPIF of winter larvae increased gradually with the prolonged exposure duration. Death did not mainly result from freezing in long exposure duration. Autumn larvae are more susceptible and adaptable than winter and spring larvae. Winter larvae have a slight freeze-tolerance trend. Our research showed that M. alternatus came into complex cold-hardening strategies under natural selection. Freeze avoidance is the primary strategy; with prolonged exposure duration to above SCP or < 0 °C, chill tolerance is more important; this is followed by freeze tolerance during harsh winters.  相似文献

We evaluated the responses of male and female Monochamus alternatus Hope (Coleoptera: Cerambycidae) to various terpenes commonly associated with host trees. Electroantennogram (EAG) tests were conducted with 12 plant volatile compounds and ethanol. Antennae of both sexes were highly sensitive to (R)-(+)-alpha-pinene, (+)-3-carene, (-)-beta-pinene, and terpinolene. Both sexes of M. alternatus were attracted by traps baited with (+)-alpha-pinene, (-)-beta-pinene, (+)-3-carene, or terpinolene. Our results support the first of the three-stage hypothesis posed by Ginzel and Hanks that suggests that location of stressed trees by cerambycids involves three stages: (1) both sexes locate larval hosts by using plant volatiles as kairomones; (2) males produces sex pheromones to attract females after both sexes land on the larval hosts; (3) males and female recognize each other by contract pheromones in their epicuticular wax layer. Males and females showed differences in their EAG responses to several compounds, including (R)-(+)-alpha-pinene, (-)-beta-pinene, myrcene, (+)-3-carene, (R)-(+)-limonene, terpinolene, and trans-caryophyllene. In all cases, males exhibited greater sensitivity than females. In laboratory assays, male M. alternatus showed strong preference for 1% (+)-alpha-pinene and 1% (-)-beta-pinene over other compounds. In field assays, traps baited with (+)-alpha-pinene, (-)-beta-pinene, (+)-3-carene, or terpinolene caught more beetles than control traps. We found strong male bias in beetle catches in baited traps and those captured on the stem of stressed trees despite a strong female bias in emerging beetles in 2004. We hypothesize that male M. alternatus are more responsive than females to plant volatiles and that males have more capacity than females in finding mating locations.  相似文献

To study the dispersal process of the pine sawyer Monochamus alternatus (Hope) in frontier populations, a microsatellite marker-based genetic analysis was performed on expanding populations at the northern limit of its range in Japan. In Asian countries, M. alternatus is the main vector of pine wilt disease, the most serious forest disease in Japan. Sawyers were collected from nine sites near the frontier of the pine wilt disease damage area. A mountain range divides the population into western and eastern sides. Five microsatellite loci were examined and a total of 188 individuals was genotyped from each locus with the number of alleles ranged from two to nine. The mean observed heterozygosity for all loci varied from 0.282 to 0.480 in the nine sites, with an overall mean of 0.364. None of the populations have experienced a significant bottleneck. Significant differentiation was found across the mountain range, but the genetic composition was similar amongst populations of each side. It is believed that the mountain range acts as a geographical barrier to dispersal and that gene flow without a geographical barrier is high. On the west side of the mountain range, a pattern of isolation by distance was detected. This was likely to be caused by secondary contact of different colonizing routes on a small spatial scale. Based on these data, a process linking genetic structure at local (kilometres) and regional spatial scales (hundreds of kilometres) was proposed.  相似文献

我们评价了聚集性信息素和植物源信息素对松墨天牛Monochamus alternatus Hope(Coleoptera:Cerambycidae)在林间的联合诱捕作用,以期研制出更优的引诱剂控制松墨天牛成虫的危害。结果显示,樟子松墨天牛M.galloprovincialis雄虫挥发出来的聚集性信息素2-undecyloxy-1-ethanol对松墨天牛雌雄成虫均有引诱效果,而且樟子松墨天牛性信息素与植物源信息素之间相互增效作用十分显著,比性信息素或植物源信息素单独使用效果提高45倍。松墨天牛雌虫卵巢发育情况显示,成虫期在浙江地区大约持续4个月,松墨天牛成虫期可以分为两个阶段,第一个阶段为取食期(卵巢未成熟),从5月9日到7月9日;第2个阶段为产卵期(卵巢成熟),从6月3日到9月18日,两个阶段在一段时期有重叠。聚集性信息素和植物源信息素联合应用将在防控松墨天牛危害,遏制松材线虫病传播方面发挥重要作用。  相似文献

A mitochondrial DNA (mtDNA) study, based on 43 European populations (33 of them sampled in France) of Monochamus galloprovincialis , vector of the pinewood nematode, and 14 populations of its sister species Monochamus sutor was realized. Sequencing of 792 bp of the cytochrome oxidase I (COI) and 521 bp of the COII genes revealed numerous ambiguities on multiple nucleotide sites for half of M. galloprovincialis specimens studied (44.8%). Hypotheses of heteroplasmy and pseudogenes ( Numts ) were examined. The mtDNA isolation by alkaline lysis and cloning (for three successfully used individuals) both support the hypothesis that the ambiguous sequences amplified were not of mtDNA nature and validate the presence of Numts in the nuclear genome of M. galloprovincialis . Multiple copies of mtDNA-like sequences were found paralogous to COI, tRNA leucin and COII regions. Phenetic analysis placed different recently diverged mtDNA-like sequences as a close relative of mtDNA sequences, and grouped 10 closely related mtDNA-like sequences as a more basal clade, closer to ancestral states and to M. sutor . This result supports that this nuclear family of pseudogenes arose independently of the other events and may represent mitochondrial haplotypes sampled from M. galloprovincialis ancestral populations. This is the first time that Numts are proved for a longhorned beetle, whereas no Numts were found within its sister species M. sutor. The incorporation mechanism of Numts in unknown for M. galloprovincialis , however, excess of ambiguous sites corresponding to synonymous mutations placed on third codon position as well as the absence of Numts in M. sutor , conducted to the hypothesis of a recent transfer of these Numts in the nuclear genome of M. galloprovincialis .  相似文献

Transmission of pinewood nematode, Bursaphelenchus xylophilus, to mature, field grown Scots pines through feeding wounds of Monochamus carolinensis was investigated by caging nematode-infested beetles on pine branches for 24 hours. Nematodes were transmitted to 31 of 64 branches. Frequency of successful transmission was independent of the sex of the beetle but dependent upon beetle age. Transmission frequencies were highest for beetles 2, 3, and 4 weeks after emergence as adults. The number of nematodes transmitted per branch was low and did not differ between beetle sexes or among beetle age categories. The number of nematodes extracted per branch was correlated with the number of nematodes carried per beetle but was not correlated with the feeding area on the branch.  相似文献

Inhibitory effects of Bursaphelenchus mucronatus on the number of B. xylophilus carried by an adult Monochamus alternatus were investigated using artificial pupal chambers. When pupal chambers were infested with either B. xylophilus or B. mucronatus, the load of B. xylophilus onto the beetle was greater (P < 0.001) than that of B. mucronatus. However, within the pupal chamber there was no difference in the abundance of the third-stage dispersal juveniles, which would molt to the fourth-stage dispersal juveniles to board beetles. The nematode load on beetles that emerged from pupal chambers infested with both Bursaphelenchus species was smaller (P = 0.015) than that of beetles with B. xylophilus alone but greater (P < 0.001) than that of beetles with B. mucronatus alone, suggesting an inhibitory effect of B. mucronatus. As a result of this study, the rate of inhibition of B. mucronatus on molting of third-stage dispersal juveniles of B. xylophilus to fourth-stage dispersal juveniles was 0.65, which resulted in great inhibition on boarding beetles at a rate of 0.7.  相似文献

Abstract:  Transmission of the pinewood nematode (PWN) Bursaphelenchus xylophilus (Steiner & Bührer) Nickle to Pinus pinaster Aiton branches through feeding wounds of its vector in Portugal, Monochamus galloprovincialis Olivier, was studied under laboratory conditions. All the B. xylophilus -infected beetles transmitted nematodes to branches they fed. The transmission was more frequent during the first 6 weeks after emergence, with transmission peaks during the second and the sixth week. The adult M. galloprovincialis transmitted nematodes for a mean of 5 weeks, independently of the beetle's sex or longevity. No relation was found between beetle feeding intensity and effective transmission of B. xylophilus to the branches. The nematode transmission ceased after the ninth week, even in insects which still had B. xylophilus on their bodies. The longevity of the nematode-free insects (control group) was slightly higher than the B. xylophilus -infected beetles, although with no significant difference. The results emphasize the necessity to control the immature stages of M. galloprovincialis prior to emergence and develop efficient strategies to capture and eliminate the recently emerged beetles, as majority of the nematode infection of healthy pine trees occurs during a short period of few weeks after beetle emergence.  相似文献

Monochamus alternatus Hope is an important vector of the pinewood nematode Bursaphelenchus xylophilus (Steiner et Buhrer) Nickle, which causes pine wilt disease. To determine canopy-related differences in the activity density of M. alternatus adults in pine stands, sticky screen traps with no lure were suspended under and in the canopy at 3.8 and 10 m above the ground of a Pinus densiflora Siebold et Zuccarini stand during a flight season. The five upper and five lower traps caught 338 and 54 adults, respectively, during a flight season from 9 June to 12 October 2011, indicating that activity density was six times as high in the canopy as under it. Male-biased sex ratio was observed for trap catches during a season: The sex ratio was male-biased in the early half of the flight season and 1:1 in the late half. There was no difference in sex ratio between the inside and outside of the canopy. To determine the sex-related difference in flight activity, the adults were captured by two different measures in two other mixed stands of P. densiflora and P. thunbergii Parlatore. Kicking tree trunks to cause beetles to drop revealed a 1:1 sex ratio of catches, whereas sticky screen traps tended to show a male-biased sex ratio in a 4-year study, suggesting that males were more active fliers than females.  相似文献

Summary The seasonal occurrence ofMonochamus alternatus and newly weakened trees were investigated in aPinus thunbergii stand for 4 years. Adult beetles were present between June and September with a peak in their population occurring in early July followed by a decline then a period of about one month being in a steady number. The average number ofBursaphelenchus xylophilus (Nematoda), which is the causal agent of pine wilt disease, within beetles decreased as the season advanced. Pine trees newly weakened byB. xylophilus appeared between June and October, especially from August to October. Abstract:  The pine sawyer Monochamus galloprovincialis is the vector of the introduced pine wood nematode Bursaphelenchus xylophilus in Portugal and has a fixed univoltine life cycle, overwintering as larvae inside the wood. We investigated the possibility of the existence of a phase of winter dormancy affecting the larvae, its induction and termination under controlled conditions and its incidence and termination in the field. In the laboratory, the majority of the newly hatched larvae which were maintained at a constant temperature of 23°C and different photoperiods became dormant in <70 days. Part of the population completed dormancy and reassumed development until adult emergence at the constant temperature, without the need for a chilling period. Although no critical photoperiod for dormancy maintenance or termination could be determined, dormancy termination was more frequent for larvae experiencing changes on daylength duration. In the field, dormancy incidence grew progressively from a minimum of 26% in October to a maximum of 100% in April, and dormancy appeared to be completed only during late winter. The results suggest that M. galloprovincialis larvae underwent an obligatory dormancy associated with the last larval instar, which conforms to the general definition of diapause, although presenting some affinities with oligopause. The role of this dormancy on the regulation of the pine sawyer's seasonal development and the number of generations is discussed.  相似文献

1 The oviposition behaviour and response of Monochamus alternatus females to oviposition scars were investigated in the laboratory. 2 Prior to oviposition, females gnawed at the bark surface of Pinus densiflora bolts to make a wound. Then females turned their bodies 180° to position their ovipositors over the wounds and inserted them under the bark through the wounds. After an oviposition, a jelly was deposited while the ovipositor was still inserted. The females then withdrew their ovipositors and rubbed the oviposition scars with the tips of their abdomens. 3 When searching females encountered oviposition scars, they stopped walking and drummed the surface and inside of the oviposition scars with their maxillary and labial palpi. 4 Eighty-six percent of females left oviposition scars containing single eggs after the palpation. By contrast, when females encountered oviposition scars containing no eggs, 76% of them began to gnaw at the scars and 64% deposited single eggs. The response to artificial oviposition scars was similar to that to vacant oviposition scars made by the females. 5 The results of various observations and experiments showed that the females could recognize oviposition scars and discriminate the scars occupied by single eggs from vacant ones, and suggested that the palpation of oviposition scars was the critical discrimination behaviour, indicating mediation by chemical cues.  相似文献

1 Sawyer beetles (Coleoptera: Cerambycidae) and metallic wood‐boring beetles (Coleoptera: Buprestidae) attack recently killed or felled trees. They are attracted to potential hosts by volatiles and possibly by visual cues. We examined traps of six different designs to determine the role of silhouette and shape in the capture of pine sawyer beetles, Monochamus L. 2 Traps with a round black cylinder, a single vane (clear or black), or cross vanes (clear or black) all mounted above a pan were compared to an open pan. Each pan contained soapy water to retain insects. 3 Traps with a black silhouette were significantly more effective in capturing male and female Monochamus scutellatus (Say) than traps with or without clear vanes. For both sexes of M. mutator LeConte, pan traps were as effective as traps with a black silhouette. Silhouette was not important for the capture of male and female M. notatus (Drury). Trap catches for the six trap types were not significantly different for male Buprestis maculativentris Say. 4 A pan trap with a black silhouette is recommended for general use in capturing wood‐boring cerambycids and buprestids.  相似文献

松材线虫病是一种重要的检疫性病害,松褐天牛是传播松材线虫的主要媒介昆虫.本实验研究了温度对松褐天牛传递松材线虫数量的影响.设置22℃、28℃和34℃ 3个温度梯度,结果表明松褐天牛成虫的寿命随温度升高而缩短,其中雌虫寿命短于雄虫,且羽化进度不太一致,多数松褐天牛携带的松材线虫量在1000~9999条之间.通过对松褐天牛取食传入松枝的松材线虫量的追踪记录发现,随着温度升高,传播线虫的高峰时段随之提前.34℃高温下松褐天牛的传递率最低,表明高温对松材线虫病的发生具有抑制作用.  相似文献

To investigate the seasonal occurrence of the pine sawyer, Monochamus saltuarius, its emergence rate was observed in pine log and Korean white pine log conditions after the logs were cut to a regular size and placed in a netted cage. Numerical measurement was also conducted for sex ratios, fresh weight, body length and width, head width, and length of adult antennae. In the Korean white pine logs, the first day of emergence occurred on May 2, the day of peak emergence observed on May 29, and the last day of emergence was on June 10. In the pine logs, the first day of emergence occurred on May 7, the day of peak emergence was observed on May 22, and the last day of emergence was on May 30. The period of seasonal occurrence observed for the pine logs was shorter than that of Korean white pine logs. Sex ratios were 0.58; the average fresh weights of males (331) and females (379) were 0.21 g and 0.23 g; body lengths were 17.6 mm and 18.4 mm; body widths were 5.47 and 5.81 mm; head widths were 3.30 mm and 3.42 mm; and antennal lengths were 40.28 mm and 26.40 mm, respectively. The ratio of the length of the antenna to the body length was measured to be 2.0 in males and 1.4 in females.  相似文献

The pine sawyer Monochamus galloprovincialis Olivier (Coleoptera: Cerambycidae) is a vector of the pine wood nematode Bursaphelenchus xylophilus (Steiner and Buhrer) Nickle. Male and females of this species have a kairomonal attraction to host volatiles and Ips semiochemicals. Once on the host tree, males and females copulate and oviposition occurs. Bioassays using Y-tube olfactometry revealed that females were attracted to volatile compounds produced by males, but not to volatiles produced by females. However, immature males did not seem to release attractants for mature females and immature females did not show any attraction for mature males. A experiment designed to know about mating behaviour in this specie revealed that most of the males encountered females while they were walking. In all pairs both sexes contacted with antennae before copulation, however, in some occasions copulation did not proceed just after antennal contact. On the contrary, in all pairs observed, the male licked the elytra of the female with his mouth palpi just before copulation (licking); immediately afterwards, the male grabbed the female with his forelegs and mounted her suggesting a chemically mediated mate recognition by males.  相似文献

Summary Oviposition schedules under laboratory conditions, survivorship curves, and mortality factors within trees of two cerambycid beetles, Japanese pine sawyer (JPS),Monochamus alternatus Hope, and sugi bark borer (SBB),Semanotus japonicus Lacordaire, were investigated. Average longevities of reared adults of JPS were 38.1 days for males and 42.3 days for females; those of SBB were 15.7 days for males and 23.8 days for females. It was confirmed that the JPS must feed on the pine branches for full maturation after emergence, but SBB need not; they are able to lay eggs soon after emergence. The average fecundities of JPS and SBB were 32.9 and 90.5, respectively. Thus, the JPS lay fewer eggs for a long time with continuous maturation feeding, whereas the SBB lay more eggs for short time without maturation feeding. Average survivorship curves of JPS within dead pine trees in 8 pine forests were theDeevey's B type, showing a constant mortality through the pre-imaginal stages in the trees. On the other hand those of SBB in 4 cedar stands approached theDeevey's type, suggesting that the high mortality occurred at an early stage in the trees. Average mortalities of JPS between the appearance of oviposition scars and adult emergence in 8 forests ranged from 62.3% to 95.2%. Intraspecific ompetition of JPS resulting from overcrowding in dead pine trees appeared to be main mortality factor. For SBB, as most larvae were killed by resin flow in living trees, this appeared to be the main mortality factor for this species.  相似文献

The Japanese pine sawyer, Monochamus alternatus Hope (Coleoptera: Cerambycidae), is an important forest pest as well as the principal vector of the pinewood nematode (PWN), Bursaphelenchus xylophilus (Steiner et Buhrer), in mainland China. Despite the economic importance of this insect-disease complex, only a few studies are available on the population genetic structure of M. alternatus and the relationship between its historic dispersal pattern and various human activities. The aim of the present study was to further explore aspects of human activity on the population genetic structure of M. alternatus in mainland China. The molecular data based on the combined mitochondrial cox1 and cox2 gene fragments from 140 individuals representing 14 Chinese populations yielded 54 haplotypes. Overall, a historical (natural) expansion that originated from China’s eastern coast to the western interior was revealed by the haplotype network, as well as several recent, long-distant population exchanges. Correlation analysis suggested that regional economic status and proximity to marine ports significantly influenced the population genetic structure of M. alternatus as indicated by both the ratio of shared haplotypes and the haplotype diversity, however, the PWN distribution in China was significantly correlated with only the ratio of shared haplotypes. Our results suggested that the modern logistical network (i.e., the transportation system) in China is a key medium by which humans have brought about population exchange of M. alternatus in mainland China, likely through inadvertent movement of infested wood packaging material associated with trade, and that this genetic exchange was primarily from the economically well-developed east coast of China, westward, to the less-developed interior. In addition, this study demonstrated the existence of non-local M. alternatus in new PWN-infested localities in China, but not all sites with non-local M. alternatus were infested with PWN.  相似文献

To understand the mechanism of spread of pine wilt disease caused by the pinewood nematode, Bursaphelenchus xylophilus, which is vectored by a cerambycid, Monochamus alternatus, the spatial distribution of trees weakened by the nematode was examined within a Pinus thunbergii stand from June to October for 4 years. The weakened trees were distributed in a clumped pattern in 1980 and 1981, at an early stage of infestation. In many cases, they showed a double-clumped pattern. The degree of aggregation was higher in June or July than after August. They were uniformly distributed in June or July 1982 and in June 1983 whereas they showed a double-clumped pattern after August. The trees were frequently weakened in June or July when they were near the trees weakened during the previous year. At quadrat sizes of more than 25 m2, spatial overlapping was pronounced between trees weakened during June–July of the current year and those weakened in the previous year. The seasonal changes in spatial distribution of weakened trees were explained by the interaction among M. alternatus, B. xylophilus and Pinus trees.  相似文献
