Shoots of three F1 hybrids of Sorghum bicolor (L.) Moench withtheir inbred parents were analyzed for chlorophyll a and chlorophyllb contents as well as for ascorbic acid turnover during theearly stages of seedling growth. In all the hybrids both chlorophylland ascorbic acid turnover showed better-parental and/or mid-parentalheterosis. Since ascorbate participates in photosynthesis andnitrate reduction, it is suggested that a well co-ordinatedsystem incorporating photosynthetic efficiency and nitrate assimilationmay be associated with the manifestation of hybrid vigour insorghum. Aniline blue fluorescence was used to study the growth of maizepollen tubes in the stigmas of 13 diverse sorghum accessions.In 12, only short maize pollen tubes were formed, but in thesingle exception (Sorghum nervosum Nr481) maize pollen tubesgrew at least as far as the base of the style. The S. bicolorgenotypes S9B and CMS (a cytoplasmic male sterile line) werehybridized with Nr481, and analysis of maize pollen tube growthin F1 plants, and BC1 plants using Nr481 as the recurrent parent,suggested that differences in inhibition of pollen tube growthwere due to variation at a single locus, which we propose todesignate lap (Inhibition of alien pollen tubes). AccessionNr481 appears to be homozygous for a recessive allele permittingmaize pollen tube growth. Attempts were made to produce sorghumx maize hybrids using Nr481 and CMS derivatives which were knownto allow maize pollen tube growth to the base of the style.A putative hybrid endosperm was obtained in one Nr481 x Seneca60 maize cross, but this was not repeatable and no hybrid plantswere produced. A fundamental problem may be the large size ofthe maize pollen tube, which could have difficulty growing throughthe sorghum ovary and in entering the micropyle. Sorghum bicolor spp. bicolor (L.) Negative linear relations were detected (P < 0·005)between the rate of progress from sowing to panicle initiationand CO2 concentration (210-720 µmol CO2 mol-1 air) fortwo genotypes of sorghum [Sorghum bicolor (L.) Moench]. Relationsbetween CO2 concentration and the rate of progress from sowingto first flowering were also negative in soyabean [Glycine max(L.) Merrill] (P < 0·025), but positive in cowpea[Vigna unguiculata (L.) Walp.] (P < 0·025), albeitthat in both grain legumes sensitivity was much less than insorghum. Thus CO2 elevation does not delay flowering in allshort-day species. The considerable effect of CO2 concentrationon times to panicle initiation resulted in large differencesamong the sorghum plants at this developmental stage; with increasein CO2 concentration, plants were taller with slightly moreleaves and more pronounced apical extension. At the same timeafter sowing however, sorghum plants were heavier (P < 0·05)at 210 than at 360 µmol CO2 mol-1 air. In contrast, relationsbetween the dry masses of the soyabean and cowpea plants andCO2 concentration were positive and curvilinear (P < 0·05).It is suggested that the impact of global environmental changecould be severe for sorghum production in the semi-arid tropics.Copyright1995, 1999 Academic Press Sorghum bicolor (L.) The Physiology of Heterosis in Sorghum with Respect to Environmental Stress   总被引:1,自引:0,他引:1  
The hypothesis that heterosis in biomass production of sorghum(Sorghum bicolor L. Moench) may be ascribed to stability incarbon exchange rate (CER) over a wide range of environmentalconditions was evaluated. This hypothesis was based on previousresults from detached leaves that hybrids sustained greaterCER over a wider temperture range than their parents. Two grain sorghum hybrids (ATx378/RTx430 and ATx378/RTx434)and their parental lines were grown in the greenhouse in a gradientof ambient temperatures under two water regimes (well-irrigatedand drought up to heading). Plant water-use (estimated by weighingpots), leaf area, leaf gas exchange, grain yield, and above-groundbiomass were determined. Significant heterosis was found for biomass, grain yield perplant, and grain number per panicle. No heterosis occurred forharvest index, indicating that heterosis in grain yield wasdue to heterosis in biomass. Neither growth duration nor leafarea could explain heterosis in biomass. CER and stomatal conductancefor hybrid ATx378/RTx430 in the controls were greater than forboth its parents at leaf temperatures above 38 °C. This,however, was not observed in the other hybrid which was lessheterotic for biomass and grain yield in the controls. WhenCER data were subjected to a stability analysis by joint linearregression, the two hybrids had greater CER than their respectiveparents especially under conditions favouring high CER. An understanding of volatilization of nitrogen (N) from leavesof crop and weed species may be important to the improvementof crop production. Foliar N loss (both reduced and oxidizedforms), net CO2 uptake, and transpiration rates were measuredconcomitantly at 30°C on soyabean (Glycine max (L.) Merr.)and sorghum (Sorghum bicolor (L.) Moench) leaves at low (1 percent), ambient (20 per cent), and high (40 per cent) levelsof oxygen. In soyabeans, maximum reduced and total N losseswere found at the highest O2 concentration, and the lowest Nlosses were measured at the lowest O2 level. Net CO2 assimilationwas significantly reduced with increasing O2 during two of threesamplings. Quantities of oxidized N lost were not altered. Sorghumshowed no significant effects from O2 on N loss or net CO2 assimilation.The increased ammonia released from soyabean foliage in thepresence of higher concentration of O2 probably affects metabolicpathways that contribute to the total reduced N volatilization. We report here an in vitro culture system that provides reliable, highly efficient regeneration from immature embryos of pearl millet [Pennisetum glaucum (L.) R. Br.] and sorghum [Sorghum bicolor (L.) Moench]. Immature embryos were isolated 10-20 days after pollination and cultured on various L3 media. The influence of different parameters during the callus induction phase was examined with respect to the regeneration rate: (1) the concentrations of 2,4-dichlorophenoxyacetic acid (2,4-D) and various cytokinins; (2) the addition of AgNO3; (3) the use of maltose or sucrose as a carbon source. Modifications in the phytohormones alone resulted in the regeneration of fertile sorghum plants at high efficiency. Significant increases in the regeneration rates of pearl millet genotypes were achieved by the combination of sucrose as a carbon source and silver nitrate as a potential ethylene inhibitor.  相似文献

Floret opening of excised spikelets in Sorghum bicolor (L.) Moench and Sorghum Sudanesis (Piper) Stapf was significantly stimulated by immersing into 2 mM methyl jasmonate (MeJA) solution. In male sterile (MS) lines of Sorghum bicolor (L.) Moench, floret opening was more sensitive to MeJA than that in maintainer (MT) lines. Salicylic acid (SA) could abolish the effect of MeJA on the opening of spikelets. These data indicate that MeJA plays a role in floret opening in sorghum, and that SA interacts with MeJA in regulating of this process.  相似文献

The addition of nickel (4 × 10−3m) to the extracting buffer enhances the nitrate reductase activity in preparations of young grain sorghum (Sorghum bicolor L. [Moench] leaf tissue by as much as 6-fold. Activities comparable to other plant species are obtained over an extraction pH range of 7 to 8 with tris buffer and reduced nicotinamide adenine dinucleotide as a cofactor for the reaction when the ratio of plant material to extraction medium is 1:20. The aim of this research was to describe the temperature responseof gas exchange in grain sorghum (Sorghum bicolor L. Moench)leaves as temperature was increased in a fashion similar toits daily increase in a hot environment. The hypothesis thatsorghum hybrids manifested heterosis for carbon dioxide exchangerate on a wider temperature range as compared with their parentallines was also evaluated. Gas exchange of detached turgid leaveswas measured in four sorghum hybrids and their parents as leaftemperature rose steadily from 32 °C to 43 °C in 4 h.CER was maximal at about 37–40 °C depending on thegenotype. In one genotype (Tx378), CER was maximal at 42 °C.In three out of four hybrids significant heterosis was foundfor CER at the lower, the higher or at the full range of temperaturestested depending on the hybrid. Consequently, all three heterotichybrids displayed a greater temperature range for high CER ascompared with their parents. Heterosis in CER was largely explainedby heterosis in stomatal conductance, though the effects ofnon-stomatal components could not be elucidated here. Heterosisin transpiration was revealed in the same three hybrids, especiallyat moderate temperatures. Since heterosis in CER was relativelygreater than heterosis in transpiration, significant heterosisin transpiration ratio at moderate temperatures was seen intwo of the four hybrids. An effect of heat hardening on CERwas observed in parental line Tx430 and its two hybrids as seenin the reduction of CER at 36–37 °C and its subsequentrecovery as temperatures rose slowly to 43 °C. The effectof heat hardening on photosynthesis was also observed in someof the genotypes as a hysteresis in the association betweenstomatal conductance and CER as temperatures increased. Thishysteresis indicated that, for the same stomatal conductance,CER was greater after than before leaves were exposed to hightemperature. The length of the seminal root (SR) axis and the number andlength of lateral roots (LRs) of sorghum (Sorghum bicolor Moench)were markedly inhibited by taro [Colocasia esculenta (L.) Schott]residues incorporated into a sand growing medium. The sand profilewas divided equally into zones with and without residues. Productionand elongation of the first-order LRs of the SR axis facingthe zone containing taro residues were severely suppressed.On the side facing the zone that was free of residues, productionand elongation of LRs was not inhibited. SR and LR growth wasdrastically impaired and many plants were killed when taro residueswere incorporated in large amounts into the uppermost 2 cm ofthe growing medium. Cobaltous ions (Co2+) inhibited light-mediated anthocyanin synthesisin excised first internodes of Sorghum bicolor (L.) Moench.Studies with precursors/intermediates of the anthocyanin biosyntheticpathway, such as acetate, phenylalanine, t-cinnamic acid, tyrosine,and p-coumaric acid were undertaken to identify the metabolicsite at which Co2+ ions inhibit anthocyanin synthesis. p-Coumaricacid partially restored anthocyanin synthesis. No other intermediatewas effective in bringing about recovery. The F1 progeny of Chowghat Dwarf Orange (CDO) and West CoastTall (WCT) cross tend to be highly heterozygous with respectto the colour of petiole and seedling vigour. The study indicatedthat the greenish brown coloured vigorous plants had higherchlorophyll (a + b) content and nitrate reductase (NR) activitycompared to the orange coloured vigorous plants and the dwarfs.The greenish brown seedlings were also characterized by a largerleaf area and shoot d. wt. Abstract

Grain sorghum [Sorghum bicolor (L.) Moench] is a gluten-free grain that is gaining attention as a food crop that can be used in the management of celiac disease. At present, sorghum is widely grown in many semiarid regions of the world. New food-grade sorghum cultivars are of particular interest in Mediterranean countries due to improved quality characteristics and gluten-free status of the grains. Until now very few studies have examined the grain yield (GYLD) and agronomic performance characteristics of food-grade sorghum hybrids in Italy. A 2 year study was conducted to evaluate the agronomic performance of eight food-grade sorghum hybrids representing different maturity classes in trials conducted in Southern Italy. The results showed wide variation in adaptation of these hybrids as measured by differences in GYLD (2.35–8.50 t ha?1) and other pheno-morphological characteristics. Of particular interest was the fact that many of the early-flowering hybrids (e.g. SP-X303) performed better than the later-flowering hybrids (e.g. ArchX-02). These results demonstrated that flowering time of hybrid and crop cycle length are important factors to consider in selecting cultivars for production in the Mediterranean region.  相似文献

Summary Restriction endonuclease patterns of chloroplast DNA (cpDNA) were consistently distinguishable between fertile and male-sterile cytoplasms of sorghum [Sorghum bicolor (L.) Moench], whereas no differences in restriction patterns of cpDNA among male-sterile (A1) lines, including six isocytoplasmic strains, were revealed in this study. Previous studies have demonstrated that pollen of sorghum [Sorghumbicolor (L.) Moench] loses capacity to both germinate in vitroand to set seed in vivo soon after being shed. The current studyevaluates the capacity for dehydrated pollen to effect in vitrogermination, reduce tetrazolium chloride, and set seed on cytoplasmicmale sterile plants. Morphological changes during pollen germinationwere examined by scanning electron microscopy (SEM). Close to70% of the pollen germinated in 5 min, or less, when collectedat 80% relative humidity (RH) and stored in sealed glass vials.Pollen tubes elongated autotropically with atmospheric humidityapparently being a controlling factor in the process. Pollendehydrated at 50% RH and 25°C for 15-30 min neither germinatedin vitro, reduced tetrazolium chloride, nor set seed on malesterile plants. Rehydrating the pollen did not restore the capacityfor germination. SEM micrographs demonstrated that elongatingpollen tubes encircled the pollen grain and were contiguousto the surface. The Mexican rice borer, Eoreuma loftini (Dyar) (Lepidoptera: Crambidae), is an economic pest of sugarcane (Saccharum spp. hybrids) and other graminaceous host crops, and it attacks grassy weeds. Oviposition preference has been known to be for plants with leaves that form folds. This study is the first to associate the nutritional quality of crop and forage plant hosts with Mexican rice borer injury. Three experiments were conducted to determine the levels of selected biochemical nutrients, including free amino acids and sugars, in four grass weeds [barnyardgrass, Echinochloa crus‐galli (L.); broadleaf signalgrass, Urochloa platyphylla (Munro ex C. Wright); johnsongrass, Sorghum halepense (L.) Pers.; and Vasey's grass, Paspalum urvillei Steud.], a forage grass [sudangrass, Sorghum bicolor (L.) Moensch spp. drummondi (Nees ex Steud.) de Wet & Harlan], three crop species [maize, Zea mays L.; sorghum, Sorghum bicolor (L.) Moench; and sugarcane, Saccharum spp.], and a sorghum‐sudangrass forage hybrid (all Poaceae). Of 16 free amino acids detected among plants in the first two experiments, only high accumulations of free histidine in sudangrass and maize were associated with increased infestations by Mexican rice borer larvae. In all three experiments, high levels of fructose were associated with heightened Mexican rice borer infestation. Ramifications of these findings on the potential dispersal of this invasive pest in the USA and possible applications of fructose in baits are discussed.  相似文献

A molecular genetic analysis of soriz genotypes (Sorghum oryzoidum), its parental form Sorghum bicolor (L.) Moench (grain sorghum), possible parents (Sorghum sudanense (Piper.) Stapf. (Sudan grass) and Oryza sativa L. (Rice planting), as well as its closest relatives, has been carried out with the use of microsatellite loci of sorghum and rice. Based on the obtained data, the genetic distances were calculated and the examined species were clustered. It was shown that soriz did not carry rice DNA fragments, but its genome contained DNA fragments, which belonged to Sudan grass. This confirms that the origin of soriz is associated with representatives of Sorghum sudanense.  相似文献

Leaf surfaces of seven genotypes of Sorghum bicolor, two ofmaize, Zea mays, and two pearl millet, Pennisetum americanum,were examined by scanning electron microscopy for possible morphologicaldifferences. Leaves 1, 3, 5 and 7 were photographed and printswere used to estimate waxiness, hairiness or pubescence andstomatal density. Glossiness was determined by spraying water,which adhered to the glossy leaves. Cuticular transpirationof detached third and fifth leaves was estimated from the rateof water loss after abscisic acid induced stomatal closure.Sorghum lines SC283, CSM63, CSM90, and pearl millets Souna andTiotioni (all from Mali), were non-glossy, well covered withwax, and exhibited variable hairiness. Older leaves of sorghumvarieties Martin and Redlan were glossy and, like older leavesof the other glossy lines SC1096 and SC90, had little or nowax deposits on their cuticles. The two maize cultivars, NB611and N7A, were non-glossy with dense wax covering; no trichomeswere observed until the 5 to 7 leaf stage. Thus, the glossycharacter was correlated with the reduction or absence of waxdeposits on the leaf surfaces, while hairiness might occur ineither glossy or non-glossy genotypes. Unlike sorghum and maize,in which all leaves after the fifth or seventh were glossy,pearl millet showed no glossiness through the ninth leaf. Measurementsshowed that cuticular transpiration of glossy leaves was oftenmore than double that of non-glossy leaves. Comparisons amongsorghums showed that non-glossy lines had higher stomatal densitiesthan glossy lines.
