Soybean root rot, caused primarily by Fusarium solani f. sp. phaseoli in a complex with F. oxysporum and Rhizoctonia solani, has become an increasing problem for soybeans, dry beans, and other rotation crops in central Minnesota due to soil conditions associated with reduced tillage. This study was conducted, in two field sites in central Minnesota located near Staples and Verndale, to develop methods for nondestructive assessment of root rot severity using plant radiometric properties. Soybean canopy reflectance was measured with a hand-held multi-spectral radiometer. Prior to the radiometer measurements, attempts were made to create differing root rot situations with moldboard or chisel tillage, and with or without a biological seed treatment. Root rot severity was estimated using a visual disease severity scale. Colony-forming units (CFU) were determined to estimate soil populations of pathogenic F. solani and F. oxysporum. Results from the Verndale site consistently showed significant treatment effects in the measured canopy radiometric parameters, and in the visual disease rating and yield (significant for seed treatment). Values of a simple ratio vegetation index from this site exhibited negative relationships with disease rating and F. oxysporum CFU, and a positive linear relationship with yield. Treatment effects were generally not significant at the Staples site because of low initial F. oxysporum populations. The results indicate that remote sensing is potentially a rapid, nondestructive means for assessment of root rot diseases in soybean.  相似文献

Field evaluation of six cassava genotypes for resistance to root rot disease was compared with three rapid laboratory methods (whole root inoculation, root slice inoculation, and stem inoculation) for resistance screening. Both the field evaluation and the three laboratory methods separated the varieties into resistant and susceptible groups. Genotypes 30572 and 91/02324 were resistant while 92/0247, 92/0057 and TME-1 were susceptible. One genotype (30001) was not consistent in its reaction between field evaluation and laboratory assays. In the laboratory assays with three fungal pathogens, different pathogens varied in their levels of virulence on host genotypes. With the most virulent pathogen (Botryodiplodia theobromae), the majority of the genotypes reacted in the same way across trials with the root slice and whole root assays. Due to the good correlation between the whole root assay and the field results, we recommend this for the routine assessment of cassava resistance to root rot disease and for the analysis of virulence of pathogen isolates. However, because of the advantages in terms of economy of labour, space, time, quantity of root and inoculum required, the root slice assay could be used for the preliminary screening of large cassava accessions. The selected genotypes can then be further screened with the whole root inoculation method.  相似文献

The effect of seed‐borne pathogens of wheat and barley on crown and root rot diseases of seven barley cultivars (Jimah‐6, Jimah‐51, Jimah‐54, Jimah‐58, Omani, Beecher and Duraqi) and three wheat cultivars (Cooley, Maissani and Shawarir) was investigated. Bipolaris sorokiniana and Alternaria alternata were detected in seeds of at least eight cultivars, but Fusarium species in seeds of only two barley cultivars (Jimah‐54 and Jimah‐58). Crown rot and root rot symptoms developed on barley and wheat cultivars following germination of infected seeds in sterilized growing media. Bipolaris sorokiniana was the only pathogen consistently isolated from crowns and roots of the emerging seedlings. In addition, crown rot and root rot diseases of non‐inoculated barley cultivars correlated significantly with B. sorokiniana inoculum in seeds (P = 0.0019), but not with Fusarium or Alternaria (P > 0.05). These results indicate the role of seed‐borne inoculum of B. sorokiniana in development of crown rot and root rot diseases. Pathogenicity tests of B. sorokiniana isolates confirmed its role in inducing crown rot and root rot, with two wheat cultivars being more resistant to crown and root rots than most barley cultivars (P < 0.05). Barley cultivars also exhibited significant differences in resistance to crown rot (P < 0.05). In addition, black point disease symptoms were observed on seeds of three barley cultivars and were found to significantly affect seed germination and growth of some of these cultivars. This study confirms the role of seed‐borne inoculum of B. sorokiniana in crown and root rots of wheat and barley and is the first report in Oman of the association of B. sorokiniana with black point disease of barley.  相似文献

Objective: Previous research has suggested a genetic contribution to the development of insulin resistance and obesity. We hypothesized that the same genes influencing insulin resistance might also contribute to the variation in adiposity. Research Methods and Procedures: A total of 601 (200 male, 401 female) adult baboons (Papio hamadryas) from nine families with pedigrees ranging in size from 43 to 121 were used in this study. Plasma insulin, glucose, C‐peptide, and adiponectin were analyzed, and homeostasis model assessment of insulin resistance (HOMA IR) was calculated. Fat biopsies were collected from omental fat tissue, and triglyceride concentration per gram of fat tissue was determined. Body weight and length were measured, and BMI was derived. Univariate and bivariate quantitative genetic analyses were performed using SOLAR. Results: Insulin, glucose, C‐peptide, and adiponectin levels, HOMA IR, triglyceride concentration of fat tissue, body weight, and BMI were all found to be significantly heritable, with heritabilities ranging from 0.15 to 0.80. Positive genetic correlations (rGs) were observed for HOMA IR with C‐peptide (rG = 0.88 ± 0.10, p = 0.01), triglyceride concentration in fat tissue (rG = 0.86 ± 0.33, p = 0.02), weight (rG = 0.50 ± 0.20, p = 0.03), and BMI (rG = 0.64 ± 0.22, p = 0.02). Discussion: These results suggest that a set of genes contributing to insulin resistance also influence general and central adiposity phenotypes. Further genetic research in a larger sample size is needed to identify the common genes that constitute the genetic basis for the development of insulin resistance and obesity.  相似文献

Aphanomyces root rot, caused by Aphanomyces cochlioides Drechs., is one of the most serious diseases of sugar beet (Beta vulgaris L.). Identification and characterization of resistance genes is a major task in sugar beet breeding. To ensure the effectiveness of marker-assisted screening for Aphanomyces root rot resistance, genetic analysis of mature plants’ phenotypic and molecular markers’ segregation was carried out. At a highly infested field site, some 187 F2 and 66 F3 individuals, derived from a cross between lines ‘NK-310mm-O’ (highly resistant) and ‘NK-184mm-O’ (susceptible), were tested, over two seasons, for their level of resistance to Aphanomyces root rot. This resistance was classified into six categories according to the extent and intensity of whole plant symptoms. Simultaneously, two selected RAPD and 159 ‘NK-310mm-O’-coupled AFLP were used in the construction of a linkage map of 695.7 cM. Each of nine resultant linkage groups was successfully anchored to one of nine sugar beet chromosomes by incorporating 16 STS markers. Combining data for phenotype and molecular marker segregation, a single QTL was identified on chromosome III. This QTL explained 20% of the variance in F2 population (in the year 2002) and 65% in F3 lines (2003), indicating that this QTL plays a major role in the Aphanomyces root rot resistance. This is the first report of the genetic mapping of resistance to Aphanomyces-caused diseases in sugar beet.  相似文献

Fusarium spp. in maize can contaminate grain with mycotoxins harmful to humans and animals. Breeding and growing resistant varieties is one alternative to reduce contamination by mycotoxins. Little is known about the population parameters relevant to resistance breeding. The objectives of this study were to draw conclusions on breeding of reduced mycotoxin concentrations of deoxynivalenol, zearalenone and fumonisins, and resistance to ear rot after silk channel inoculation with F. graminearum or F. verticillioides, respectively. For that, variation and covariation of line and testcross performance and correlations between both species and between mycotoxin concentrations and ear rot resistance were calculated. Means of ear rot after infection with F. graminearum were higher than with F. verticillioides. Moderate phenotypic correlations (r = 0.46–0.65) between resistances to both Fusarium spp. implicate the need of separate testing. Analyses of variance revealed significant (P < 0.01) differences among lines in line and testcross performance for 30–60 entries per maturity group. Multi-environmental trials for accurate selection are necessary due to significant (P < 0.1) genotype × environment interactions. High genotypic correlations between ear rots and mycotoxins (r ≥ 0.90), and similar heritabilities of both traits, revealed the effectiveness of indirect selection for mycotoxin concentrations based on ear rot rating after inoculation. Moderate genotypic correlations between line and testcross performance were found (r = 0.64–0.83). The use of one moderately to highly susceptible tester is sufficient since genotypic correlations between testcrosses of different testers were high (r = 0.80–0.94). Indirect selection for testcross performance based on line performance is less effective than selection based on mycotoxin concentrations. Consequently, selection for resistance to ear rot and mycotoxin accumulation should be started among testcrosses tested first for general combining ability based on ear rot data in parallel with a negative selection for line per se performance.  相似文献

Bacterial wilt caused by Curtobacterium flaccumfaciens pv. flaccumfaciens is an important new disease of common bean (Phaseolus vulgaris) in western Canada. Both yellow and orange variants of the pathogen were found in the region. A controlled environment study was conducted to assess 124 common bean cultivars and lines from eight market classes for resistance to the yellow and orange variants of the pathogen, using the hilum injury/seed inoculation method. Results of the screening tests showed significant (P < 0.05) differences in resistance to bacterial wilt among the cultivars or lines. The great northern line L02E317, the great northern cultivar Resolute and pinto lines L02B662 and 999S‐2A, were highly resistant to both variants of the pathogen, with disease severity indices of 0 on a rating scale of 0 (no wilt symptoms) to 5 (dead seedling). Resistant cultivars or lines were found among black, great northern, pink, pinto, small red and Flor de Mayo bean market classes. The study concludes that new bacterial wilt‐resistant germplasm exists among Canadian bean cultivars and lines, and constitutes a valuable resource for breeding common beans for resistance to both yellow and orange variants of C. flaccumfaciens pv. flaccumfaciens.  相似文献

The inheritance of resistance to parathion in the Pacific spider mite was investigated by means of a backcross. A 1:1 ratio of a susceptible and a resistant class indicated the existence of a single dominant gene for resistance.Since the susceptible parental strain was marked with a recessive for albinism, information was obtained of the genetic relationship between the gene for resistance (R p ) and the gene for albinism (p). The segregation of resistant (R p r p ) and susceptible (r p rp) classes in pigmented (p+p) and albino (pp) animals revealed that there was no linkage between these genes.  相似文献

 We report the molecular mapping of the py-1 gene for resistance to corky root rot [Pyrenochaeta lycopersici (Schneider and Gerlach)] in tomato using RAPD and RFLP marker analysis. DNA from near-isogenic lines (NILs) of tomato differing in corky root rot resistance was screened with 575 random oligonucleotide primers to detect polymorphic DNAs linked to py-1. Three primers (OPW-04, OPC-02, OPG-19) revealed polymorphisms between the NILs. Twelve resistant and eight susceptible DNA pools derived from segregating F3 families were used to confirm that the RAPD markers were linked to the py-1 gene. Two of the linked amplified fragments, corresponding to OPW-04 and OPC-02, were subsequently cloned and mapped on the tomato molecular linkage map as RFLPs. These clones were located between TG40 and CT31 on the short arm of chromosome 3. Further analysis with selected RFLP markers showed that 7% (8.8 cM) of chromosome 3 of the resistant line ‘Moboglan’ was introgressed from the L. peruvianum donor parent. Three RFLP markers (TG40, TG324, and TG479) from the introgressed part of chromosome 3 were converted to cleaved amplified polymorphism (CAP) markers for use in a polymerase chain reaction (PCR) assay. The adaptation capacity of olive trees to different environments is well recognized. However, the presence of microorganisms in the soil is also a key factor in the response of these trees to drought. The objective of the present study was to elucidate the effects of different arbuscular mycorrhizal (AM) fungi coming from diverse soils on olive plant growth and water relations. Olive plants were inoculated with native AM fungal populations from two contrasting environments, that is, semi‐arid – Freila (FL) and humid – Grazalema (GZ) regions, and subjected to drought stress. Results showed that plants grew better on GZ soil inoculated with GZ fungi, indicating a preference of AM fungi for their corresponding soil. Furthermore, under these conditions, the highest AM fungal diversity was found. However, the highest root hydraulic conductivity (Lpr) value was achieved by plants inoculated with GZ fungi and growing in FL soil under drought conditions. So, this AM inoculum also functioned in soils from different origins. Nine novel aquaporin genes were also cloned from olive roots. Diverse correlation and association values were found among different aquaporin expressions and abundances and Lpr, indicating how the interaction of different aquaporins may render diverse Lpr values.  相似文献

Melone Vine Decline is a severe rot root disease of increasing world‐wide importance. In Eastern Spain it is related to the presence of Acremonium cucurbitacearum and Monosporascus cannonballus. The strong influence of environmental conditions on the progress of this disease has made its study difficult. A field screening of Cucumis melo accessions has been conducted over four years. Simultaneously, the pathogenicity of isolates of the two fungi recovered from the screening field was studied. These were more aggressive than other Spanish and American isolates. Percentage of vine decay was scored, togeter with root damage, the latter being evaluated by using four scoring systems based on root characteristics and disease severity. Root inspection alowed the selection of resistance sources, even when aboveground symptoms did not appear, due to the lack of environmental stresses at time of fruit maturity. The root damage scoring if environmental stresses occur during fruit maturity. The accession C. melo var. agrestis Pat 81 consistently exhibited high field resistance level, expressed as a higher percentage of symptomeless plants, together with a significant delay in symptoms appearance. The F1 hybrids derived from the cross Pat 81 ×C. melo susceptible varienties showed an intermediate level of resistance between the parents, suggesting a partial dominance gene action. The high resistance level found in Pat 81, and also in its derived hybrids, against the aggressive isolates found in this area, makes it promissing for breeding melon verieties resistant to melon vine decline.  相似文献

Two field trials with two groups of cassava (Manihot esculenta Crantz) cultivars were conducted under rainfed conditions in seasonally dry and semiarid environments at the northern coast of Colombia, South America, to evaluate the genetic diversity in photosynthesis and productivity, and to determine their interrelationship. There were significant differences among cultivars in both environments, in average net photosynthetic rates (P N ) of upper canopy leaves and in final dry root yields. Both P N and dry root yields were much higher in the seasonally dry environment than in the semiarid one. Highly significant correlation (r 2 = 0.90, p < 0.001) between leaf P N and dry root yield was observed across environments, suggesting that selection in parental plants for high photosynthesis might lead to high yields if combined with other yield determinants, such as leaf area duration, high harvest index and strong root sink. The strength of coupling between canopy gas exchange and root respiration was examined in ~15-yr-old ponderosa pine (Pinus ponderosa Doug. Ex Laws.) growing under seasonally drought stressed conditions. By regularly watering part of the root system to reduce tree water stress and measuring soil CO2 efflux on the dry, distant side of the tree, we were able to determine the strength of the relationship between soil autotrophic (root and rhizosphere) respiration and changes in canopy carbon uptake and water loss by comparison with control trees (no watering). After ~40 days the soil CO2 efflux rate, relative to pre-treatment conditions, was twice that of the controls. This difference, attributable to root and rhizosphere respiration, was strongly correlated with differences in transpiration rates between treatments (r2 = 0.73, p<0.01). By the end of the period, transpiration of the irrigated treatment was twice that of controls. Periodic measurements of photosynthesis under non-light limited conditions paralleled the patterns of transpiration and were systematically higher in the irrigated treatment. We observed no evidence for a greater sensitivity of soil autotrophic respiration to temperature compared to the response of heterotrophic respiration to temperature; the Q10 for total soil respiration was 1.6 (p>0.99) for both treatments. At the ecosystem scale, daily soil CO2 efflux rate was linearly related to gross primary productivity (GPP) as measured by eddy-covariance technique (r2 = 0.55, p<0.01), suggesting patterns of soil CO2 release appear strongly correlated to recent carbon assimilation in this young pine stand. Collectively the observed relationships suggest some consideration should be given to the inclusion of canopy processes in future models of soil respiration.  相似文献

Intercrossing red clover plants, selected for reaction to Fusarium root rot (F. avenaceum), produced progenies with lower root rot scores than the parental cultivar Hermes II. Eleven percent of the progenies had less than 2 mm root rot after two cycles of selection, compared to less than 1% in the parental cultivar. Root rot severity in two old local strains of red clover, as well as in two populations of wild red clover, was lower than in cv. Hermes II. This could indicate that a natural selection for root rot resistance can occur in red clover.  相似文献

Water and solute relations of young roots of Phaseolus coccineus have been measured using the root pressure probe. Biphasic root pressure relaxations were obtained when roots were treated with solutions containing different osmotic test solutes. From the relaxations, the hydraulic conductivity (Lpr), the permeability coefficient (Psr), and the reflection coefficient (σsr) of the roots could be evaluated. Lpr was 1.8 to 8.4 . 10?8 m . s?1 . MPa?1 and Psr (in 10?10 m . s?1): methanol, 27–62; ethanol, 44–73; urea, 5–11; mannitol, 1.5; KCl, 7.1–9.2; NaCl, 2.1; NaNO3, 3.7. The hydraulic conductivity was similar when using osmotic and hydrostatic pressure gradients as driving forces. The hydraulic conductivity of individual root cortex cells (Lp) was by two orders of magnitude larger than Lpr (Lp = 0.3 to 4.7 . 10?6 m . s?1 . MPa?1) which indicated a predominant cell-to-cell rather than an apoplasmic transport of water in the Phaseolus root. Except for distances shorter than 20 mm from the root apex, the hydraulic resistance of the roots was limited by the radial movement of water across the root cylinder and not by the hydraulic resistance within the xylem. Reflection coefficients were low: methanol: 0.16–0.34; ethanol: 0.15–0.47; urea: 0.41–0.51; mannitol: 0.68; KCl: 0.43–0.54; NaCl: 0.59; NaNO3: 0.54. The transport coefficients (Lpr, Psr, σsr) have been critically examined for influences of unstirred layers and active transport. The low σsr suggests that the common treatment of the root as a rather perfect osmometer (σsr = 1) analogous to plant cells should be treated cautiously. The reasons for the low σsr and the possible implications of the absolute values of the transport parameters for the absorption of water and nutrients are discussed.  相似文献

In common bean (Phaseolus vulgaris L.), Fusarium root rot (caused by Fusarium solani f. sp. phaseoli) disease severity is increased by environmental factors that stress the plant. The current study used reciprocal grafting techniques with the resistant cultivar FR266 and the susceptible cultivar Montcalm to determine if the genetic control of resistance is conferred by the rootstock (root genotype) or the scion (shoot genotype) and if root vigor played a role in resistance. The influence of a compacted layer on root and shoot genotype response and root rot resistance was studied. Root rot resistance was found to be controlled by the root genotype, such that on a scale of 1 to 7 (severe disease) the FR266 root had an average score of 2.3 and the Montcalm root had an average score of 4.4. However, when grafted plants were grown in the presence of a compacted layer, the FR266 root and/or shoot genotype in any graft combination with the susceptible Montcalm had reduced root rot (score = 2.4 average) than the Montcalm self graft (score = 4.5). Root mass was shown to be controlled by the root genotype in the absence of compaction such that the FR266 root was 26% larger that the Montcalm root when grafted onto a FR266 shoot or a Montcalm shoot. When a compacted layer was present the root and shoot genotype both contributed to root mass. Average root diameter was controlled by the shoot genotype, as the FR266 shoot grafted to Montcalm or FR266 roots had thicker roots (average diameter 0.455 mm) than the Montcalm shoot (average diameter 0.418 mm). This study shows evidence that root vigor in the presence of Fusarium disease pressure should be evaluated to effectively develop common bean lines resistant to Fusarium root rot across a range of environments.  相似文献

This study was undertaken to ascertain if the soft rot inciting Pectobacterium carotovorum/Erwinia carotovora would pass through the micropropagated bananas as a latent pathogen and cause disease during or post acclimatization. In vitro cultures of ‘Grand Naine’ were exposed to the pathogen by providing 100 μl of inoculum (0.001–1.0 at OD600 nm) at the lower leaf axil. These cultures showed a gradual development of soft rot symptoms coupled with obvious bacterial colony growth on banana proliferation medium and consequent plant mortality within a month irrespective of the inoculum level employed. Plants carried forward to acclimatization following inoculation in vitro failed to establish ex vitro. Monitoring the normal field-grown suckers at culture initiation through PCR screening employing soft rot Erwinia primers did not show the amplification of the 119-bp fragment as seen with the pure cultures of pathogen. Further testing of micropropagated banana plants through soil inoculation, in vitro culturing and PCR screening ruled out the possibility of the pathogen surviving in micropropagated stocks in latent form as the organism outgrew and killed the cultures. It emerged that the infection possibly takes place in the nursery. This information will be of particular value for the plant tissue culture industry, plant pathologists and quarantine agencies.  相似文献

Cassava (Manihot esculenta Crantz), is an important tropical tuber crop with global importance and plays a significant role in the food, nutritional and livelihood security of around 500 million people. In India, the low productivity of cassava attributes to the soil borne disease, particularly tuber rot caused by Phytophthora palmivora (Butl.) which is destructive and the attack is spreading in alarming rate in all the cassava growing regions causing heavy yield loss of more than 50%. Introduction of disease resistant varieties may alleviate the problem to a certain extent. This paper describes the screening procedures and findings on the disease resistant variety of cassava accession against tuber rot. Variety Sree Padmanabha imparted high resistance against tuber rot, while Sree Sahya was moderately resistant and all other accessions studied were found to be susceptible in in vitro and in field trials. In screening studies, a reproducible positive correlation was obtained between attached tubers in live plant with detached tubers which showed that detached tuber part can be used for the prediction of resistance in attached live plants of cassava for cultivar resistance. The procedure described here could be used as a simple, rapid and efficient method for screening of cassava accessions against tuber rot of cassava.  相似文献

Cigarette smoke is known to generate free radicals by various mechanisms. In this study involving 30 non-smokers and 30 smokers, we show that urinary excretion of 5-(hydroxymethyl) uracil (HMUra) was not different in the two groups (6.54±2.07 vs. 6.70±1.68 nmol/mmol creatinine). In contrast, 8-oxo-7,8-dihydro-2′-deoxyguanosine (8-oxo-dGuo) excretion increased by 16% (1.16±0.35 vs. 1.35±0.50 nmol/mmol creatinine, p=0.039). Results concerning 8-oxo-dGuo are in agreement with those of previous studies. We observed significant multiple correlations between HMUra and creatinine (rp=0.44), BMI (rp=-0.27) and nicotine derivatives (rp=0.26). Multiple correlation analysis showed relations between 8-oxo-dGuo on the one hand, and: creatinine (rp=0.36), nicotine derivatives (rp=0.29), BMI (rp=-0.24) on the other.  相似文献

Twelve widely grown cultivars of subterranean clover (Trifolium subterraneum) were screened both under controlled environment conditions for their resistance to five fungi commonly associated with root rot and under field conditions for their resistance to natural root infections. All cultivars showed decreased seedling survival (particularly from Pythium irregulare and Rhizoctonia solani), tap and lateral root rot (particularly from Fusarium avenaceum, P. irregulare, and R. solani) and reduced plant size (particularly from R. solani and P. irregulare). Individual cultivars generally differed in their response to the five pathogens and for any one pathogen there was generally a range of cultivar susceptibilities. Cultivars with the best resistance to individual root pathogens were identified. The results for the five individual pathogens under controlled conditions only showed correlation with field data for some of the parameters compared.  相似文献
