Among the antimicrobial phytoalexins produced by Phaseolus vulgaris (French bean) are the prenylated isoflavonoids kievitone and phaseollidin. Two enzyme activities, kievitone hydratase and phaseollidin hydratase, occur in culture filtrates of the bean pathogen, Fusarium solani f. sp. phaseoli, and catalyse similar hydration reactions on the dimethylallyl moieties of the phytoalexins. The enzymes nearly co-purified during hydroxyapatite chromatography followed by preparative native gel electrophoresis. Eluates from successive slices taken from the native gel were assayed for both activities. Although they were not completely separated in the native gel, the activity profiles indicated that the two activities were distinct. The Km of phaseollidin hydratase for phaseollidin was approximately 7 microM. 相似文献

During the September‐December season of 1990, severe symptoms of Fusarium wilt were for the first time observed on a popular climbing bean (Phaseolus vulgaris L.) cultivar. G 2333. introduced within the previous 5 years. Seventy‐three bean genotypes were screened for resistance lo the disease, using artificial inoculation. The effect of inoculation density on the reaction of four selected genotypes was also investigated. Of the 29 climbing bean genotypes evaluated, 19 were resistant, including 11 of the 15 pre‐release or released cultivars. Of the 44 bush bean cultivars evaluated, 28 were resistant, five were intermediate and 11 were susceptible. All susceptible cultivars showed vascular discoloration. In both susceptible and resistant genotypes, the fungus spread almost equally from the entry points in inoculated roots to the base of the plants, but colonization and vertical spread within the vascular system were markedly less in resistant than in susceptible cultivars. At 20 and 30 cm above soil level, the fungus was only recovered from susceptible cultivars. Increasing inoculum density from 102 to 107 conidia/ml did not affect the resistance of cultivars RWR 950 and G 685 but. in the susceptible cultivars G 2333 and MLB‐48‐89 A. it resulted in early appearance, high incidence and severity of the disease. 相似文献

Leaf yellowing and brown discoloration was observed in tobacco plants cv. Burley TN97 in tobacco fields of central Greece in 2002. Fusarium oxysporum f. sp. nicotianae was isolated from symptomatic plants and Koch's postulates were fulfilled. The pathogenicity of the isolated fungus was examined on five tobacco cultivars (Burley TN97, BurleyB21, VirginiaBE9, Virginia Niki and Anatolika KE26/2). The pathogen was present in tobacco seed batches imported in 2000 and 2001, which indicates that the infected seed is most probably the primary source of the disease in Greece. As Fusarium oxysporum f. sp. vasinfectum can also cause vascular wilt in tobacco, the hypothesis that the isolated F. oxysporum strain belongs to f. sp. vasinfectum was excluded by a pathogenicity test to cotton cv. Acala SJ‐2. This is the first report of F. oxysporum f. sp. nicotianae in Greece and the second in the European Union, although the seedborne nature of the pathogen has not been previously reported in Europe. 相似文献

Abstract A gene fusion between the Saccharomyces cerevisiae actin gene promoter and the cDNA of the Fusarium solani f. sp. pisi pelA gene has been constructed. This expression cassette has been introduced into the industrial wine yeast strain T73 . The resulting recombinant strain is able to secrete active PELA enzyme into the culture medium. In preliminary microvinification experiments the wine produced by this pectinolytic strain is indistinguishable from wine produced using the non-transformed strain on the basis of the chemical analyses. Large scale fermentations need to be carried out in order to assess the effects on filtrability. 相似文献

An improved in vitro test is described for determining the pathogenicity of Fusarium solani f.sp. pisi isolates on pea. This technique involves the use of polypropylene fibre Milcap plugs to suspend peas in boiling tubes containing spore suspensions in 0.1% water agar. Results were available after 14 days of incubation at 25°C. Four levels of pathogenicity were detected on pea cultivars Little Marvel and Dark Skinned Perfection using a total of eight isolates and strains of F. solani f.sp. pisi. 相似文献

Wilt of Psidium guajava L., incited by Fusarium oxysporum f. sp. psidii and Fusarium solani is a serious soil-borne disease of guava in India. Forty-two isolates each of F. oxysporum f. sp. psidii (Fop) and F. solani (Fs) collected from different agro climatic zones of India showing pathogenicity were subjected to estimate the genetic and molecular characterisation in terms of analysis of microsatellite marker studies. Out of eight microsatellite markers, only four microsatellite markers, viz. MB 13, MB 17, RE 102 and AY212027 were amplified with single band pattern showing the character of identical marker for molecular characterisation and genetic identification. Microsatellite marker MB 13 was amplified in F. oxysporum f. sp. psidii and F. solani isolates. Product size of 296 bps and 1018 bps were exactly amplified with a single banding pattern in all the isolates of F. oxysporum f. sp. psidii and F. solani, respectively. Microsatellite markers, viz. MB 17, RE 102 and AY212027 were also exactly amplified with a single banding pattern. MB 17 was amplified in F. oxysporum f. sp. psidii isolates with a product size of 300 bp. RE 102 and AY212027 were amplified in F. solani isolates with the product size of 153 bp and 300 bp, respectively. Therefore, amplified microsatellite marker may be used as identifying DNA marker. 相似文献

Localized infection in cucumber cotyledons with Colletotrichum lagenarium induced resistance against infection after challenge inoculation with Rhizoctonia solani AG2–2 and Fusarium oxysporum f. sp. cucumerinum in the roots. The plants were unprotected in soil that was infested heavily with R. solani or in contact with the mycelium, and induced resistance was not observed. Wounding of the root also negated the effect of induced resistance to F. oxysporum . 相似文献

Pot trials were carried out under controlled conditions to evaluate the effectiveness against Fusarium wilt of rocket (Fusarium oxysporum f.sp. conglutinans) and basil (F. oxysporum f.sp. basilici) of soil amendments based on a patented formulation of Brassica carinata defatted seed meal and compost, combined or not with a simulation of soil solarization. The soil solarization treatment was carried out in a growth chamber by heating the soil for 7 and 14 days at optimal (55–52°C for 6 h, 50–48°C for 8 h and 47–45°C for 10 h/day) and sub‐optimal (50–48°C for 6 h, 45–43°C for 8 h and 40–38°C for 10 h/day) temperatures similar to those observed in summer in solarized soil in greenhouses in Northern Italy. Two subsequent cycles of plant cultivation were carried out in the same soil. Even at sub‐optimal temperature regimes, 7 days of thermal treatment provided very valuable results in terms of disease control on both rocket and basil. In general, the thermal treatment was more effective against F. oxysporum f.sp. basilici than against F. oxysporum f.sp. conglutinans. Control of Fusarium wilt of rocket is improved with 14 days of thermal treatment. The combination of organic amendments with a short period of soil solarization (7 or 14 days), although not providing any improvement to the level of disease management, did significantly increase biomass and positively affected yield. 相似文献

A total of 250 endophytic fungal isolates, representing 30 morphotaxa, were isolated and characterised, they were collected from the different living symptomless parts of date palm trees of orchards of six Egyptian governorates. Colonisation was greater in samples from the midrib than in those from laminar tissue and slightly greater at the tip of the lamina compared with the base of the leaf. Acremonium spp. were frequently isolated as date palm root endophytes. Acremonium isolates were screened in Petri dishes to select the highest antagonistic one against an Algerian isolate of Fusarium oxysporum f.sp. albedinis. Two-week-old axenically reared date palm seedlings grown in Petri dishes were directly injected with spore suspension (1.5?×?107 spores/ml) of a pure culture of the virulent antagonistic isolate of Acremonium sp. One week after endophytic colonisation, date palm seedlings were then challenged with the pathogen, Fusarium albedinis. The challenged seedlings exhibited a significant reduction in wilt symptom percentage (by 87.0%), while the seedlings exposed to Fusarial toxin without pathogen exhibited the wilt disease symptoms. This indicates that the endophyte ably depresses any toxic action of F. albedinis. The endophytic fungus was recovered from sites distant from the point of inoculation after six?months from the application, indicating that the Acremonium sp. has the potential to move throughout the tissue plant, even the end time of trial. The Acremonium mode of action, as a biocontrol agent, was discussed. 相似文献

Chitosan has been considered an environmental-friendly polymer. However, its use in agriculture has not been extended yet due to its relatively low solubility in water. N-Methylene phosphonic chitosan (NMPC) is a water-soluble derivative prepared by adding a phosphonic group to chitosan. This study demonstrates that NMPC has a fungicidal effect on the phytopathogenic fungus Fusarium solani f. sp. eumartii (F. eumartii) judged by the inhibition of F. eumartti mycelial growth and spore germination. NMPC affected fungal membrane permeability, reactive oxygen species production, and cell death. Also, this chitosan-derivative exerted antifungal effects against two other phytopathogens, Botrytis cinerea, and Phytophthora infestans. NMPC did not affect tomato cell viability at the same doses applied to these phytopathogens to exert fungicide action. In addition to water solubility, the selective biological cytotoxicity of NMPC adds value in its application as an antimicrobial agent in agriculture. 相似文献

We present the first procedure for extracting intercellular fluids of potato ( Solanum tuberosum L. cv. Spunta) tubers. Intercellular washing fluids were isolated from healthy and Fusarium ‐infected potato tissue. The electrophoretic pattern using SDS‐PAGE indicated differences between the fluids from the two tissues. A significant extracellular proteolytic activity was accumulated during the infection with Fusarium solani f. sp. eumartii . A major proteolytic band with an apparent molecular mass of 70 kDa and another of approximately 30 kDa were detected after separation of intercellular fluids by casein gel electrophoresis. Proteolytic activity was principally inhibited by diisopropylfluorophosphate, which is indicative of the involvement of serine protease(s). In vitro degradation assay indicated that specific potato proteins from healthy tubers were hydrolyzed by fluid proteases from infected tubers. The biological role of such activity in potato‐ Fusarium interaction is still unknown. Our results suggest that the intercellular serine protease has a fungal origin. 相似文献

From 2002 to 2004, wilted plants of different species of rocket (Eruca vesicaria and Diplotaxis spp.) were found for the first time in Europe, in greenhouse cultivations in Piedmont and Lombardy, northern Italy. The causal agent of the disease was found to be Fusarium oxysporum. Vegetative compatibility analysis was carried out on 46 isolates of the fungus, 41 of them obtained from wilted rocket (E. vesicaria and D. tenuifolia) and five reference strains, in order to increase the knowledge on the causal agent of recent epidemics of Fusarium wilt on rocket in Italy. The analysis showed the presence of two vegetative compatibility groups (VCGs) (VCG 0101 and VCG 0220) pathogenic on both kinds of rocket. The two VCG populations, which were classified as formae specialesconglutinans and raphani, respectively, are spread in the area of epidemics but are not related to the host species from which they were isolated (D. tenuifolia or E. vesicaria). This finding shows the heterogeneity of the causal agent of Fusarium wilt on rocket in Italy. 相似文献

Fusarium wilt, caused by Fusarium oxysporum f. sp. cubense (Foc), is one of the most devastating diseases of banana (Musa spp.). Apart from resistant cultivars, there are no effective control measures for the disease. We investigated whether the transgenic expression of apoptosis-inhibition-related genes in banana could be used to confer disease resistance. Embryogenic cell suspensions of the banana cultivar, 'Lady Finger', were stably transformed with animal genes that negatively regulate apoptosis, namely Bcl-xL, Ced-9 and Bcl-2 3' UTR, and independently transformed plant lines were regenerated for testing. Following a 12-week exposure to Foc race 1 in small-plant glasshouse bioassays, seven transgenic lines (2 × Bcl-xL, 3 × Ced-9 and 2 × Bcl-2 3' UTR) showed significantly less internal and external disease symptoms than the wild-type susceptible 'Lady Finger' banana plants used as positive controls. Of these, one Bcl-2 3' UTR line showed resistance that was equivalent to that of wild-type Cavendish bananas that were included as resistant negative controls. Further, the resistance of this line continued for 23-week postinoculation at which time the experiment was terminated. Using TUNEL assays, Foc race 1 was shown to induce apoptosis-like features in the roots of wild-type 'Lady Finger' plants consistent with a necrotrophic phase in the life cycle of this pathogen. This was further supported by the observed reduction in these effects in the roots of the resistant Bcl-2 3' UTR-transgenic line. Abstract An exopolygalacturonase produced by Fusarium oxysporum f. sp. radicis lycopersici , a fungus that produces root rot, was purified by gel filtration and ion exchange chromatography. It had a M r 68 K, a pH optimum of 5.6 and an optimum temperature of 60°C. This polygalacturonase was inhibited by calcium ions and had a K m of 0.64 mM using sodium polypectate as substrate. Damaging threshold levels of root-knot nematode Meloidogyne incognita and root-rot fungus Fusarium solani on plant growth parameters, viz., plant length, fresh and dry weight of chilli were determined by conducting their pathogenicity trials in pot experiments. The results revealed a significant reduction in the plant growth parameters at and above the inoculum level of about 1000?J2 per plant of M. incognita and the highest reduction was recorded at 8000?J2 per plant. Significant reduction in plant growth parameters was recorded at 1.00?g mycelial mat of F. solani per plant, while the highest reduction was observed at 8.00?g mycelial mat per plant. Black root rot, caused by Fusarium solani f.sp. pisi, is a devastating soil‐borne disease in chickpea in Iran with no effective control measures. With the aim of finding applicable biocontrol agents to alleviate the malady, isolates of Actinomycetes isolated from soil and their antagonistic effect against F. solani f.sp. pisi were evaluated both in vitro and in vivo. More than 100 Actinomycetes isolates were screened for their antifungal activities against the pathogen. The most active isolates were evaluated in greenhouse for their biocontrol performance. Based on the results of dual cultures in screening evaluations, the size of inhibition zone of fungal growth, and the most effective antagonist isolates (S3, S12 and S40) were selected for further studies. Identity of active isolates was determined, in this regard, 16S rDNA of isolates were amplified using universal bacterial primers FD1 and RP2. The PCR products were purified and sequenced. Sequence analysis of 16S rDNA was then performed using NCBI BLAST method. Comparison of the near full length 16S rRNA sequence of isolates to GenBank sequences demonstrated that isolates S3 and S12 were most similar to Streptomyces antibioticus, while isolate S40 was most similar to Streptomyces peruviensis. Biocontrol studies of these isolates in control of the disease in greenhouse significantly decreased the disease severity. Actinomycetes isolate S12 demonstrated the greatest effect in reducing disease than the other two. Results of this research are at preliminary stage for developing biocontrol agents. These data can be utilized as a platform for future studies with the aim of commercializing these biocontrol products and hoping to step towards sustainable agriculture. 相似文献

Fusarium oxysoporum f. sp. radicis-cucumerinum (Forc) is able to cause disease in cucumber, melon, and watermelon, while F. oxysporum f. sp. melonis (Fom) can only infect melon plants. Earlier research showed that mobile chromosomes in Forc and Fom determine the difference in host range between Forc and Fom. By closely comparing these pathogenicity chromosomes combined with RNA-sequencing data, we selected 11 candidate genes that we tested for involvement in the difference in host range between Forc and Fom. One of these candidates is a putative effector gene on the Fom pathogenicity chromosome that has nonidentical homologs on the Forc pathogenicity chromosome. Four independent Forc transformants with this gene from Fom showed strongly reduced or no pathogenicity towards cucumber, while retaining pathogenicity towards melon and watermelon. This suggests that the protein encoded by this gene is recognized by an immune receptor in cucumber plants. This is the first time that a single gene has been demonstrated to determine a difference in host specificity between formae speciales of F. oxysporum. 相似文献
