An ultrastructural study of the boundary tissue of the seminiferous tubule of the camel reveals that it consists of three lamellae; inner fibrous, inner cellular and outer cellular. The inner lamella is subdivided into two homogeneous layers enclosing a third one that contains collagenous fibres and fine filaments. The inner cellular lamella consists of several layers of myoid cells; each layer is separated from the adjacent layer by homogeneous material and varying amounts of collagen. The outer cellular lamella consists predominantly of fibrocytes together with some fibroblasts and scattered collagen.  相似文献

Typical pinocytic vesicles were visible in electron micrographs in both outer and inner cellular layers of rat boundary tissue. FF technique revealed that they formed characteristic ribbons of foveoli. Foveoles less numerous were present in outer lamina for the most part of the investigated material. So we believe that this phenomenon speaks in favor of myoblastic origin of that layer.  相似文献

Structurally the boundary tissue of the vampire bat seminiferous tubuli showed 2 to 5 layers of connective tissue in which elongated contractile cells and lamellar and/or fibrillar collagen were noticed. This boundary tissue forms the seminiferous tubular lamina propria. Its structure was more complex around the seminiferous tubuli near the Capsula testicularis than between the adjacent and contiguous tubuli into the testicular lobuli. The arrangement and fine structure of peritubular myoid cells was studied in the testes of three species of reptiles (Lacerta dugesi, Testudo graeca and Natrix natrix) during two short periods of the seasonal cycle (European spring and autumn) and correlated with some ultrastructural properties of Leydig cells. The lamina propria consists of myoid cells, fibroblasts and non-cellular components comprising collageneous and non-striated microfibrils. Both components are arranged in alternating layers surrounding seminiferous tubules. In spring the lamina propria of lacertilian testis shows 1-5 layers of myoid cells which are rich in 50-70 A filaments and exhibit plasmalemmal and intracellular dense patches, smooth vesicles along the cell membrane and a concentration of organelles in a juxtanuclear position. Leydig cells are rich in smooth ER profiles and have few lipid droplets. In atumumn most myoid cells are replaced by fibroblast-like elements. Leydig cells display large numbers of lipid droplets and dense bodies, but only small amounts of agranular ER. Similar changes are noted in Leydig cells of Testudo and Natrix. However, in these species the boundary tissue of seminiferous tubules fails to show significant alterations comparing spring and autumn animals. In both species the lamina propria exhibits a few fibroblast-like cells interspersed among myoid cells.  相似文献

Cellular architecture of the lamina propria of human seminiferous tubules   总被引:3,自引:0,他引:3  
Summary The lamina propria of human seminiferous tubules is composed of 5 to 7 cellular layers separated by laminae of extracellular connective-tissue components. By means of immunocytochemical methods the different nature of the cellular layers could be defined for the first time. Based on the light-microscopic demonstration of both desmin-like and vimentin-like immunoreactivity in the inner 3 to 4 layers of the lamina propria, these cells can be identified as myofibroblasts. The outermost one or two cellular layers, on the contrary, only show a vimentin-like immunoreactivity indicating the pure fibroblastic nature of these cells. Therefore, the outermost cellular layers are suggested to be derivatives of the interstitium. In cases of disturbed spermatogenesis, the lamina propria is frequently considerably thickened by an increase in the extracellular matrix components between the cellular layers. Whereas the ultrastructural localization of laminin-, collagen type-IV- and fibronectin-like immunoreactivity remains unaffected in the thickened lamina propria, the desmin-like immunoreactive cells of the inner layers strongly decrease in number and staining intensity. Most probably, the myofibro-blasts lose their myoid characteristics to participate in the secretion of increased amounts of extracellular matrix components, which in turn presumably block the mediation of the lamina propria between the interstitium and the germinal epithelium. It is still unclear whether the thickened lamina propria provokes the disturbance of spermatogenesis or vice versa.  相似文献

At the protein level, the P1 protamine is the predominant form of mammalian protamine, present in all mammalian spermatozoa analyzed to date. An additional variant, the P2 protamine, has been detected only in spermatozoa of the mouse, hamster and human. Southern blot analysis of a group of restriction enzyme-digested mammalian DNAs has revealed the presence of sequences homologous to the P1 and the P2 mouse protamine genes in diverse species. In agreement with protein studies, nucleotide sequences homologous to the mouse P1 protamine cDNA are widespread, being present in the genomic DNAs of human, rat, dog, ram, horse, bull, hamster, baboon, flying fox (megabat), microbat, boar, North American opossum, and wallaby. Although we detect genomic sequences with strong homology to the mouse protamine 2 cDNA in rat and hamster, we also find weaker but reproducible hybridization to the genomic DNA of human, boar, dog, bull, microbat, wallaby, and platypus. With the exception of the human, the P2 protamine has not been detected in the spermatozoa of these latter species.  相似文献

The notochord and notochordal sheath of 10 adult amphioxus were investigated ultrastructurally and histochemically. The notochord in amphioxus consists of parallel notochordal cells (plates) and each plate consists of parallel thicker and thinner fibrils and numerous profiles of smooth endoplasmic reticulum situated just beneath the cell membrane. Histochemical staining shows that the notochordal plates resemble neither the connective tissue notochordal sheath nor the typical muscular structure myotomes. The notochordal sheath has a complex three-layered organization with the outer, middle and inner layer The outer and middle layer are composed of collagen fibers of different thickness and course, that correspond to collagen type I and collagen type III in vertebrates, respectively, and the inner layer is amorphous, resembles basal lamina, and is closely attached to the notochord by hemidesmosome junctions. These results confirm the presence of collagen fibers and absence of elastic fibers in amphioxus.  相似文献

SYNOPSIS. Oocysts of Eimeria nieschulzi from the laboratory rat, Rattus, norvegicus , were studied by scanning and transmission electron microscopy. Oocysts had a rough outer wall with apparent random depressions. The oocyst wall is composed of 2 layers: an osmiophilic outer layer consisting of a rough external and smooth internal surface, and a relatively thick, electron-lucent inner layer. The outer layer is composed of a dense, coarsely granular matrix. The inner layer consists of homogeneous fine granular material interspersed with coarse osmiophilic granules and contains one closely applied membrane on the outermost surface. Several raised lenticular areas are seen on the coarse outer surface of the inner layer. These layers are 102 (75–128) and 176 (135–204) nm thick, respectively.
The sporocyst wall is thin, consisting of 3 to 4 unit membranes, and measures 27 (18–34) nm thick.  相似文献

The perinuclear theca is a novel cytoskeletal consisting of a densely layered lamina that surrounds the nucleus of mammalian sperm. Using antibodies specific for the multiple band polypeptides present in the perinuclear theca of bull sperm, we show that a heterogeneous group of immunological related proteins are present in the sperm heads of other mammals with greatly different morphologies, including guinea pig, hamster, rat, and mouse. In none of the species were identical groups of immunoreactive polypeptides found, although immunoreactive proteins of molecular weights 65,000 to 80,000 were present in the sperm heads of all species examined. Immunoreactive proteins less than Mr 55,000 were prominent in rat sperm heads and mouse sperm: guinea pig, hamster, and rat sperm heads and mouse sperm had one band in common at approximately Mr 50,000. Different immunoreactive proteins were present in isolated sperm tails. The perinuclear theca first appeared in the subacrosomal space of round to elongating spermatids. Later, with the caudal movement of the manchette, the postacrosomal segment of the perinuclear theca was deposited in a cephalad to caudal direction along the sperm nucleus. Concomitantly, the cytoplasmic space between the nuclear envelope and the plasma membrane narrowed such that only the theca occupied this portion of the sperm head. Immunoreactivity accompanied the ultrastructural appearance of the subacrosomal layer and the postacrosomal segment. The periods of spermiogenesis, in which sub- and post-acrosomal components of the perinuclear theca are formed and the morphogenesis of sperm organelles with which these elements are associated, suggest that components of this cytoskeletal structure function to join the acrosome and the postacrosomal plasma membrane to the nucleus.  相似文献

Summary The boundary tissue of bovine testicular seminiferous tubules exhibits remarkable regional differences at the level of the seminiferous tubule proper, as compared with its terminal segment. The basal lamina of the seminiferous tubule proper is multilayered and possesses knob-like protrusions. At the level of the terminal segment the basal lamina is highly specialized; in the region of the terminal plug candelabrum-like projections of the tubular basal lamina invade the bases of the modified supporting cells up to a depth of 3.5 m. The adjoining surface of these supporting cells is densely studded with hemidesmosomes. The elongated peritubular cells are arranged in 3–5 concentric layers around the tubulus seminiferus proper but form a loose association at the level of the terminal segment. Where the terminal segment joins the testicular straight tubule, peritubular cells may assemble to constitute a contractile spiral. Elastic tissue is situated mainly subjacent to the tubular basal lamina and to a lesser degree between the peritubular cell layers. A peritubular space lined by endothelium-like cells may surround the seminiferous tubule proper and also the transitional zone of the terminal segment.Supported by a grant from the Deutsche Forschungsgemeinschaft  相似文献

Summary Four different types of spermatogonia were identified in the seminiferous tubules of the Japanese quail: a dark type A (Ad), 2 pale A type (Ap1 and Ap2), and a type B. A model is proposed describing the process of spermatogonial development in the quail. The Ad spermatogonia are considered to be the stem cells. Each divides to produce a new Ad spermatogonium and a Ap1 spermatogonium during Stage IX of the cycle of the seminiferous epithelium. An Ap1 spermatogonium produces two Ap2 spermatogonia during Stage II of the cycle, Ap2 spermatogonia produce four type B spermatogonia during Stage VI of the cycle, and type B spermatogonia produce eight primary spermatocytes during Stage III of the cycle. Consequently, 32 spermatids can result from each division of an Ad spermatogonium. Spermatogonial development in the quail differs from the process described in mammals in that there are fewer mitotic divisions and they are all synchronized with the cycle of the seminiferous epithelium. It is suggested that the fewer mitotic divisions explain why a smaller area of the seminiferous tubule is occupied by a cellular association in the quail than in mammals like the rat, ram and bull. The duration of spermatogenesis from the division of the Ad spermatogonia to sperm release from the seminiferous epithelium was estimated to be 12.77 days.  相似文献

Epididymal fluid, which is derived from testicular fluid, contains several unusual compounds. Little information is available on the composition of the testicular fluid of primates, but the fluid of the ram, bull, boar, and rat contains high concentrations of inositol and certain amino acids. Analyses have been made of epididymal fluid collected from the cauda epididymis of the Rhesus monkey and several nonprimate species (e.g., ram, bull, dog, stallion, rabbit, guinea pig, rat, and hamster), but similar information on the human is lacking. Cauda epididymal fluid appears to be similar in composition from one mammalian species to another. However, the epididymal plasma differs considerably from blood, lymph, and other extracellular fluids. The environment of spermatozoa in the epididymis is, therefore, highly specialized, and presumably in some way contributes to the prolonged survival of spermatozoa in this organ, and provides substrates for the metabolism of the spermatozoa. The chief characteristics of the cauda epididymal plasma are the low concentration of inorganic ions and the high levels of several unusual organic constituents namely, glycerylphosphorylcholine, carnitine, sialic acid, amino acids, glycosidases, and phosphatases. At least one antifertility compound, namely, orally administered α-chlorohydrin, appears to be concentrated in the epididymis. Studies on laboratory animals, domestic species, and man, suggest that it inhibits enzymes of the glycyolytic pathway in spermatozoa, and this may be the basis for its antifertility activity.  相似文献

Prenatal development of the thoracic aorta of the rat during the period ranging from gestational days 12 to 21 was examined by transmission electron microscopic and morphometric studies. The process of wall formation occurred in four major phases. At phase I (gestational day 12), the dorsal aorta consists of an endothelium and loosely surrounding mesenchymal cells. Collagen fibrils and fine filamentous materials are sparsely present in the intercellular space. At phase II (days 13 to 16), the mesenchymal cells begin to differentiate to myoblasts, which have small clusters of myofilaments with dense bodies, rough endoplasmic reticulum, and a discontinuous basal lamina. The differentiating cells form a few compact cell layers around the endothelium. Elastic fibers first occur sparsely in juxtacellular spaces at days 13-14. The thickness of the aorta increases rapidly from 1-3 layers of cells at day 13 to 5-8 layers at day 17, leading to a maximum of 5-9 cell layers at day 20. The differentiation of myoblasts and elastogenesis are initiated in the inner layers, and later progress toward the outer layer of the aortic wall. At phase III (days 17 to 19), the myoblasts continue to develop into typical smooth muscle cells, and elastic fibers rapidly increase in both size and number. At phase IV (day 20 and later), smooth muscle cells have well-developed myofilaments in the cell periphery, and rough endoplasmic reticulum and other organelles tend to accumulate in the apical portion of the cytoplasm. Elastic laminae appear in a few inner layers of the aortic wall.  相似文献

Specimens of dispersed Middle Devonian megaspores have been isolated from core samples from the Miastko 1 borehole in Western Pomerania. Comprehensive investigations using light, scanning and transmission electron microscopy supplement previous information on morphology and gross structure and provide data on spore wall ultrastructure of four megaspore species. Corystisporites acutispinosus is azonate; the inner layer is laminate, and the lumen is lined by a thick, laterally continuous lamina. The outer layer consists of small, tangentially aligned tabular elements that become wider, more extensive and irregularly arranged toward the outside. Coronispora variabilis is a coronate megaspore; the inner layer appears homogenous and is probably lamellate. The outer layer consists of elongate, cylindrical, branching elements that are overlaid within the proximal part of the body by a lamellate, compact, almost homogenous layer. Grandispora ciliata is pseudosaccate. The inner body is laminate with laminae thickening and becoming less continuous and less tightly packed toward the outside. The outermost region of the inner body and the innermost region of the outer envelope consist of tabular and cylindrical elongate units. The bulk of the outer wall is almost homogenous, and near the surface it is granular. Pomeranisporites subtriangularis is pseudozonate. The inner layer appears homogenous except for the presence of a single innermost lamina. The inner part of the outer layer may represent small tabular and cylindrical elements, and the outer part comprises folded laminae. The megaspores studied share numerous features of morphology and wall ultrastructure with the lycopsids, putative lycopsids, and some enigmatic Devonian plants.  相似文献

During early development of the mammalian eye, invagination and differential growth result in the formation of a cleft, the optic fissure, through which the hyaloid artery reaches the interior of the optic cup. Closure of this fissure was studied by electron microscopy in hamster embryos from days 10 through 12 of gestation. Closure occurred only when and where the basal lamina, which invests the entire wall of the optic cup, had disappeared. No morphological evidence was found that indicated a mechanism for the breakdown of the basal lamina lining the fissure, the fusion of surface cells of opposing sides and restoration of the basal lamina along inner and outer margins of the wall after closure. While in previous light microscopic investigations of the developing human eye eversion of the inner layer into the fissure has been reported, an inversion of the outer layer was found in this study. During inversion cells of the outer layer temporarily changed their orientation. While most of these cells remained within the layer when it returned to its normal position, some cells became separated and degenerated. Inversion of the outer layer, breakdown of the basal lamina and degeneration of superfluous cells appear to be necessary events for a normal closure to occur.  相似文献

The fine structure of the heart and connective tissue sheath surrounding the stomach of the brachiopod Rhynchonella psittacea has been studied. The stomach wall is lined externally with peritoneal epithelium. Between the bases of the peritoneal epithelial cells and those of the stomach epithelial cells is an extracellular amorphous matrix. The exterior part of the matrix is occupied by smooth muscle cells and the interior part by fibroblasts. The heart wall shows continuity with the peritoneal epithelium covering the stomach wall and consists of three layers: an outer layer of smooth myoepithelial and epithelial cells, an intermediate thick layer of extracellular matrix, and an inner discontinuous layer of fibroblasts. In myoepithelial cells, nucleated heads protruding freely into the coelom and contractile parts embedded in the extracellular matrix can easily be distinguished. These cells contain no sarcoplasmic reticulum or any elements of a T system. The epithelial cells are non-muscular mononucleated cells scattered among the myoepithelial cells and closely associated with these basally. They possess a well-developed rough endoplasmic reticulum. In rare cases, a small amount of myofibrils occurs basally in the epithelial cells. Morphologically the epithelial cells in the myocardium are very similar to the peritoneal epithelial cells covering the stomach wall. Both epithelial and myoepithelial cells are ciliated. No nerve elements have been found in the brachiopod heart. The structure of the brachiopod heart is compared with that of other invertebrates; similarity of cellular composition of the brachiopod heart and stomach cover is considered evidence of origin of the heart cells from the cells of the connective tissue sheath of the stomach. In this study, we examined the age-related changes on morphometric parameters and ultrastructure of seminiferous tubules, and on the expression of extracellular matrix proteins in lamina propria of Syrian hamsters. A significant decrease in the percentage of normal tubules and an increase in the percentage of hypospermatogenic and arrested maturation tubules was observed with aging. Aged animals showed a decrease in tubular diameter, tubular lumen, seminiferous epithelium volume and total tubular volume. However, the total length of seminiferous tubules was significantly increased with aging. The most important ultrastructural changes with aging were the thickening of the lamina propria, the presence of diverse abnormalities in the spermiogenesis process, degeneration of germ cells, and vacuolization and flattening of Sertoli cells showing abundant lipofucsin droplets and residual bodies. Laminin immunoreactivity was found along the lamina propria of seminiferous tubules both in young and aged animals. Fibronectin immunoreactivity was found along the lamina propria and blood vessels. Both laminin and fibronectin total volume of immunostaining per testis was increased in aged hamsters. In conclusion, the age-related changes in seminiferous tubules of hamster include: a decrease in tubular width and an increase in tubular length; widening of the lamina propria caused by a more extensive connective matrix between the peritubular cells and the basal membrane; and a strong disarrangement of the seminiferous epithelium, including germ cell degeneration and important alterations in both spermiogenesis and Sertoli cell structure.  相似文献

The endochondral layer of the osseous labyrinth in the rat, golden hamster, mouse, guinea pig, pig, rabbit, cat, dog and monkey was studied and compared with that of man. (1) With the exception of the mouse and golden hamster, interglobular spaces were found. (2) In all species but the rat, the interglobular spaces contain acid mucopolysaccharides. An analogy between these structures and the 'basophilic islands' (basophile Inseln) is discussed. (3) Extension, arrangement, direction, occurrence and frequency of interglobular spaces vary within each species so that no constant relations could be found, which are also lacking in man. Possible reasons for the persistence throughout life of interglobular spaces in the osseous labyrinth of man and some mammals are discussed.  相似文献

The changes occurring in the testicular lamina propria (LP) of the seasonal breeder Octodon degus were analyzed. Four groups of animals were studied using electron microscopic procedures. The animals in group I were reproductively active, whereas those in groups II, III, and IV were in periods of recrudescence, regression, and resting, respectively. These changes, observed in animals maintained under laboratory lighting conditions, resembled those known to be elicited in feral populations by natural photoperiods. Testicular changes in each group were monitored by calculating the gonadosomatic and spermatogenetic indexes, and by obtaining averages of the diameter of the seminiferous tubules and the height of the seminiferous epithelium. The LP of group I animals consisted of a basal membrane formed by two to four lamellae, inner and outer acellular layers containing moderate numbers of collagenous fibrils, and single or double layers of smoothly contoured myoid cells encircled by a discontinuous lymphatic epithelium. The LP of group IV animals differed considerably from the LP of group I animals: The inner acellular layer was enlarged, and the basal membrane appeared composed of a variable number of lamellae with numerous folds and indentations. Myoid cells were very irregular in contour and enveloped by a well-developed surface coating. The LPs of animals in groups II and III possessed relatively similar characteristics, which appeared to be intermediate between those of groups I and IV. The significance of these morphological changes in the LP are discussed in relation to testicular changes induced by photoperiod and other normal or pathological processes. © 1995 Wiley-Liss, Inc.  相似文献

SYNOPSIS. Electron microscope observations of the complex cyst wall of Didinium nasutum are reported. The cyst wall is composed of 3 major coats. The outermost coat, the ectocyst, consists of short strands of filamentous material which forms a diffuse, amorphous layer approximately 8–9 μ thick. Culture debris, bacteria and unidentified inclusions have been observed adhering to the outer coat. The mesocyst, approximately 2.5 μ thick, has 2 distinct regions. The outer region is differentiated into several slightly thicker layers which in face view have a honeycomb appearance. The deeper region of the mesocyst consists of compact lamellae lacking the obvious honeycomb appearance of the layers of the outer region. Finally, the endocyst (0.3 μ thick), which arises in the mature cyst in the space that develops between the pellicle and the mesocyst, consists of delicate fibrils in a compact matrix. Both mesocyst and endocyst may be undulant and folded. The structure, origin and possible relationships of the various coats composing the cyst wall are discussed. The present study also contributes information on the role and fate of mucocysts and other cytoplasmic structures during the formation of the cyst wall.  相似文献
