Local infections of either TMV or TNV in tobacco plants cv. Havana 425 (hypersensitive to TMV) proved effective in inducing systemic resistance to subsequent inoculation with the powdery mildew fungus Erysiphe cichoracearum DC. The proportion of leaf surface invaded by this pathogen and the amount of conidia it produced were both significantly lower in virus inoculated plants than in non-inoculated controls. However, the decrease in sporulation rate was less regularly observed than the reduction in leaf area infected. TMV was more effective than TNV in protecting tobacco plants from powdery mildew. E. cichoracearum is thus added to the list of challenge pathogens to which TMV or TNV are known to induce resistance in the host plants. Necrotic lesions caused to the leaves by local treatment with Ethephon (an ethylene-releasing compound) also conferred to tobacco some degree of systemic resistance to the same fungal pathogen, more frequently visible as a reduction of leaf area invaded. The protection due to the Ethephon lesions was in present experiments less marked than that of TMV. No effects against subsequent powdery mildew infection were obtained when point freeze necrotic lesions were provoked on the plants.  相似文献

Pectic polysaccharides elicit chitinase accumulation in tobacco   总被引:3,自引:0,他引:3  
Upon infection of leaves of tobacco ( Nicotiana tabacum L. ev. Havana) with Erwinia carotovora (Jones) Holl, strain 3912, a phytopathogenic bacterium that secretes pectinolytic enzymes, chitinase (EC 3.2.1.14) levels increased 12-fold within 48 h. Heat-killed E. carotovara cells did not induce this response. In young excised tobacco plants supplied with pectic polysaccharides, chitinase activity increased to about the same level as in leaves infected with E. carotovora . The amount of pectic polysaccharides required for half-maximal induction was about 160 μg (g fresh weight)−1. Using in vivo labeling of plants with [35S]-cysteine, it could be demonstrated that elicitormediated chitinase induction is due to enhanced de novo synthesis of the enzyme.  相似文献

The possibility of inducing systemic disease resistance in carrot leaves was investigated with the pathogen Cercospora carotae as the agent for both induction and challenge. The inoculation of plants with C. carotae induced resistance in newly emerged leaves challenged 23 days later. There was an optimum inoculum concentration of 3 × 104 conidia ml-1 for inducing resistance, the response decreasing with higher concentrations. Infection of carrot leaves by C. carotae could thus reduce susceptibility to subsequent infections by foliar pathogens later in the growing season. However, the inoculation of leaves with C. carotae did not increase the resistance of roots after harvest when they were wounded and challenged with Botrytis cinerea .  相似文献

A previously undescribed cDNA family was isolated from tobacco challenged with tobacco mosaic virus (TMV). A cDNA library was constructed with mRNA from upper leaves of Xanthi nc tobacco plants that had been inoculated with TMV on the lower leaves 11 days previously. The library was screened differentially with radiolabeled cDNA synthesized with mRNA from upper, uninoculated leaves of either TMV-inoculated or mock-inoculated tobacco plants. The new cDNA family, designated SAR8.2, had at least five expressed members, one or more of which were inducible by TMV inoculation and by salicylic acid treatment. The cDNAs encoded small, highly basic proteins containing N-terminal hydrophobic signal peptides and highly conserved cysteine-rich C-terminal domains. One of the SAR8.2 family members contained a direct repeat of the C-terminal domain in tandem. Hybridization of SAR8.2 cDNA to tobacco genomic DNAs indicated a gene family of 10-12 members.  相似文献

Conditions of electroporation were optimized for introduction of tobacco mosaic virus (TMV) particles into tobacco mesophyll protoplasts (Nicotiana tabacum L. cv. Petit Havana SR1). Compared with conditions for TMV-RNA uptake, a longer electric pulse was necessary at the same voltage to induce TMV particle entry. Up to 80–90% of the protoplasts were infected with TMV particles after exposure to a 10 msec pulse at 200 V (0.67 KV/cm) in a 0.5 M mannitol solution. Protoplast viability was slightly lower than for controls which did not undergo electroporation. The presence of buffer in the mannitol solution reduced the net voltage in the solution which resulted in a significant decrease of the level of infection. These results suggest that the membrane pores resulting from an electrical pulse were wide enough for TMV particles (300 × 18 nm) to enter protoplasts.  相似文献

The expression of PR-protein b1' in plants and cell suspension cultures of Nicotiana glutinosa L., Nicotiana debneyi Domin, and an amphidiploid cross of these two species, a hybrid, has been investigated. An enzyme linked immunosorbent assay has been employed to determine the concentration of PR-protein b1' in extracts. The PR-Protein b1' was constitutively produced in intact plants of the hybrid (around 25 μg g−1 leaf tissue), while only trace amounts of the protein (< 50 ng g−1 leaf tissue) were found in plants of the two parents. In suspension culture, the concentrations of PR-protein b1' were 8, 0.4 and less than 0.1 mg l−1 medium for the hybrid. N. debneyi and N. glutinosa , respectively. Only trace amounts of the protein were found in extracts from cells. Seven days after infection by tobacco mosaic virus (TMV) the concentration of PR-protein b1' in leaves of N. glutinosa was 22.5 μg g−1 leaf tissue. In N. debneyi and the hybrid a relatively limited induction of PR-protein b1' by TMV was observed. The influence of various phenoxyacetic acids on the expression of PR-protein b1' in the 3 cell cultures has been investigated. Cultures of N. glutinosa responded to treatments with 2,4-D and 2,4,5-T while cultures of N. debneyi and the hybrid were essentially unaffected. In the former case a concentration of 5–10 mg l−1 2,4,5-T was optimal and cells were most responsive to the treatment 4 days after subcultivation. The concentration of PR-protein b1' in elicited cell cultures of N. glutinosa was 2 to 4 mg l−1 medium.  相似文献

The response of tobacco (Nicotiana tabacum L. cv. Xanthinc) plants, epigenetically suppressed for phenylalanine ammonia-lyase (PAL) activity, was studied following infection by tobacco mosaic virus (TMV). These plants contain a bean PAL2 transgene in the sense orientation, and have reduced endogenous tobacco PAL mRNA and suppressed production of phenylpropanoid products. Lesions induced by TMV infection of PAL-suppressed plants are markedly different in appearance from those induced on control plants that have lost the bean transgene through segregation, with a reduced deposition of phenofics. However, they develop at the same rate as on control tobacco, and pathogenesis-related (PR) proteins are induced normally upon primary infection. The levels of free salicylic acid (SA) produced in primary inoculated leaves of PAL-suppressed plants are approximately fourfold lower than in control plants after 84 h, and a similar reduction is observed in systemic leaves. PR proteins are not induced in systemic leaves of PAL-suppressed plants, and secondary infection with TMV does not result in the restriction of lesion size and number seen in control plants undergoing systemic acquired resistance (SAR). In grafting experiments between wild-type and PAL-suppressed tobacco, the SAR response can be transmitted from a PAL-suppressed root-stock, but SAR is not observed if the scion is PAL-suppressed. This indicates that, even if SA is the systemic signal for establishment of SAR, the amount of pre-existing phenylpropanoid compounds in systemic leaves, or the ability to synthesize further phenylpropanoids in response to the systemic signal, may be important for the establishment of SAR. Treatment of PAL-suppressed plants with dichloro-isonicotinic acid (INA) induces PR protein expression and SAR against subsequent TMV infection. However, treatment with SA, while inducing PR proteins, only partially restores SAR, further suggesting that de novo synthesis of SA, and/or the presence or synthesis of other phenylpropanoids, is required for expression of resistance in systemic leaves.  相似文献

Photoacclimation was studied in tobacco leaves (Nicotiana tabacum cv Xanthi) infected with two strains of tobacco mosaic virus (TMV) and grown under different light and nitrogen nutrition regimes. Photosynthetic acclimation measured by the quantum yield and the maximum rate in saturating light of CO2-saturated photosynthesis was impaired to a greater extent in tobacco leaves infected with TMV strain PV230 than in those infected with TMV strain PV42. Infection with TMV strain PV230 severely impaired photosynthetic acclimation at high light/low nitrogen and during transfer from low to high light. Expanding leaves showing chlorotic-mosaic symptoms had greatly reduced capacity to acclimate to high light compared with controls and with developed leaves without visible symptoms. We conclude that the failure of expanding leaves to acclimate was largely due to the destruction of chloroplasts in yellow areas of the tissue, accompanied by severe reduction in ribulose-1,5-bisphosphate carboxylase/oxygenase levels, and corresponding reduction in photosynthesis on a leaf-area basis. When corrected for areas of healthy green tissue, photoacclimation of infected leaves was the same as that of controls. Visible symptom development was greatest in high light/low nitrogen treatments. In developed leaves without visible symptoms, virus accumulation, which was as extensive as in expanding leaves, accelerated senescence and impaired photoacclimation during transfer from low light to high light. Generally, infection with TMV strain PV42 did not impair photosynthetic acclimation and even enhanced it in some treatments, even though virus accumulated to the same concentration as in PV230-infected leaves. These data show that TMV does not simply impair photoacclimation in tobacco by competing with chloroplasts for leaf nitrogen reserves. Rather, specific properties of severe strains, such as PV230, which lead to visible symptom development and patchy loss of photosynthetic activity in expanding leaves as well as general acceleration of chloroplast senescence in developed leaves, contribute to impaired photoacclimation, which is generally exacerbated by low nitrogen nutrition.  相似文献

Carbendazim applied at the rate of 2 g per plant to the roots of tobacco (Nicotiana tabacum cv. White Burley) plants before infection with tobacco mosaic virus (TMV) caused very considerable reduction in the severity of disease symptoms in systemically infected leaves but did not affect their virus content. Leaves of untreated, infected plants had a greatly reduced chlorophyll content 100 days after infection whereas the chlorophyll content of leaves of infected plants treated with carbendazim was similar to that of normal uninfected leaves. Carbendazim had no effect on the infectivity of TMV in vitro or on the local lesion reaction of N. glutinosa plants when inoculated with TMV. Carbendazim was applied to lettuce cv. Cobham Green at a total rate of o-i g per plant before and after they were infected with beet western yellows virus and the plants were then grown on in the field. At harvest time (50 days after infection) almost all the treated virus-infected plants were of a normal green appearance, whereas the untreated controls were almost all very severely yellowed and unmarketable.  相似文献

Systemic acquired resistance (SAR) is an inducible defence mechanism which plays a central role in protecting plants from microbial pathogen attack. Guided by bioassays, a new chemical inducer of SAR was isolated from the extracts of Strobilanthes cusia and identified to be 3-acetonyl-3-hydroxyoxindole (AHO), a derivative of isatin. Tobacco plants treated with AHO exhibited enhanced resistance to tobacco mosaic virus (TMV) and to the fungal pathogen Erysiphe cichoracearum (powdery mildew), accompanied by increased levels of pathogenesis-related gene 1 ( PR-1 ) expression, salicylic acid (SA) accumulation and phenylalanine ammonia-lyase activity. To study the mode of action of AHO, its ability to induce PR-1 expression and TMV resistance in nahG transgenic plants expressing salicylate hydroxylase, which prevents the accumulation of SA, was analysed. AHO treatment did not induce TMV resistance or PR-1 expression in nahG transgenic plants, suggesting that AHO acts upstream of SA in the SAR signalling pathway. In addition, using two-dimensional gel electrophoresis combined with mass spectrometry, five AHO-induced plant proteins were identified which were homologous to the effector proteins with which SA interacts. Our data suggest that AHO may represent a novel class of inducer that stimulates SA-mediated defence responses.  相似文献

The activity of superoxide dismutase (SOD: E.C. 1.15.1.1) was evaluated on Nicotiana tabacum and Nicotiana glutinosa leaf tissue after Tobacco Mosaic Virus (TMV) infection. Significant increase in extracted SOD appeared to be directly related to the appearance of necrotic and systemic symptoms in hypersensitive (N. glutinosa and N. tabacum cv. Havana 425) and susceptible (N. tabacum cv. Bright BC 60) leaves, respectively. SOD activity did not change significantly during the replication of TMV in the inoculated susceptible leaves up to 4 days after inoculation. Both cyanide-insensitive (2 days after inoculation) and sensitive (3–4 days after inoculation) enzymes increased during the expression of the hypersensitivity. Only cyanide-sensitive enzyme increased in systematically infected leaves. SOD and peroxidase increased simultaneously and the enhancement of peroxidase was higher than that of SOD. The values of peroxidase greatly exceeded that of SOD only in the hypersensitive leaves during local lesion differentiation. In N. tabacum leaves 4 clear SOD bands were separated by polyacrylamide gel electrophoresis: 3 cyanide-sensitive (Cu,Zn enzyme) and 1 cyanide-insensitive, while N. glutinosa had 3 bands: 2 cyanide-sensitive and 1 cyanide-insensitive. The cyanide-insensitive band, both in N. tabacum and N. glutinosa, was sensitive to H2O2 and insensitive to chloroform-ethanol treatment and thus supposed to be Fe enzyme. The infection did not induce change in the electrophoretic pattern of SOD enzymes.In summary, our results indicate that the pathogenic alteration caused by TMV infection both in the compatible and in the incompatible combinations are characterized by an induction of SOD activity, particularly cyanide-sensitive Cu,Zn-SOD. The connection between the induction of SOD and a possible activation of O2 production in the hypersensitive tissue following TMV infection is discussed.  相似文献

The aim of this work was to study the effect of ultraviolet-B (UV-B) exposure on oxidative status in chloroplasts isolated from soybean ( Glycine max cv . Hood). Chloroplasts were isolated from soybean leaves excised from either control seedlings or those exposed to 30 and 60 kJ m−2 day−1 of UV-B radiation for 4 days. Chloroplastic oxidative conditions were assessed as carbon-centered radical, carbonyl groups and ascorbyl radical content. Treatment with UV-B increased the carbon-centered radical-dependent EPR signal significantly by 55 and 100% in chloroplasts from leaves exposed to 30 and 60 kJ m−2 day−1 UV-B, respectively, compared to radical content in chloroplasts from control leaves. The content of carbonyl groups increased by 37 and 62% in chloroplasts isolated from soybean leaves irradiated for 4 days with 30 and 60 kJ m−2 day−1 UV-B, respectively. The content of soluble metabolites in isolated chloroplasts should not be taken as absolute in vivo values; however, these data are valuable for comparative studies. UV-B exposure did not significantly affect ascorbyl radical content compared to controls. The content of ascorbic acid and thiols in chloroplasts isolated from leaves exposed to 60 kJ m−2 day−1 UV-B was increased by 117 and 20.8%, respectively, compared to controls. Neither the content of total carotene nor that of β -carotene or α -tocopherol was affected by the irradiation. The results presented here suggest that the increased content of lipid radicals and oxidized proteins in the chloroplasts isolated from leaves exposed to UV-B could be ascribed to both the lack of antioxidant response in the lipid soluble fraction and the modest increase in the soluble antioxidant content.  相似文献

In regions with a long cultural history, past land-use may strongly determine floristic composition in seemingly natural vegetation. Here we examine if past land-use affects understory plant distribution in near-natural forest in a historically and environmentally heterogeneous setting, namely the 17 ha freshwater island of Borgnr (Denmark). In 107 randomly located 100 m2 plots we recorded species composition and various environmental variables. The plots were classified into three groups according to past land-use: (1) Disturbed sites close to 12th and 20th century ruins and associated open areas; (2) formerly grazed meadow areas; and (3) forested sites without indications of strong anthropogenic influence. Past land-use, environmental variables and species traits were used to interpret floristic gradients. Unconstrained and constrained ordinations clearly separated plots with different past land-use, and showed that the main floristic gradients on Borgø correlated with hydrological-edaphic conditions and past land-use. Partial constrained ordination using the environmental parameters as covariables showed that past land-use had a unique influence on floristic composition.  相似文献

Aims:  To investigate the infection biology of Colletotrichum sublineolum (isolate CP2126) and defence responses in leaves of resistant (SC146), intermediately resistant (SC326) and susceptible (BTx623) sorghum genotypes.
Methods and Results:  Infection biology and defence responses were studied quantitatively by light microscopy, H2O2 accumulation by DAB staining and HRGP accumulation by immunological methods. Inhibition of conidial germination and appressorium formation may represent prepenetration defence responses on the leaf surface. Inducible defence responses in the resistant genotypes included decreases in formation of appressoria as well as accumulation of H2O2, HRGPs and phytoalexins. Concomitant with these inducible responses, fungal growth was stopped during or just after penetration in genotypes SC146 and SC326. High levels of H2O2 accumulating at late infection stages (5 days after inoculation) in the susceptible genotype BTx623 correlated with necrosis and tissue degeneration.
Conclusions:  The early accumulation of H2O2 and HRGPs indicates roles in defence whereas the late accumulation in genotype BTx623 correlated with successful pathogenesis.
Significance and Impact of the Study:  The fact that there is a significant correlation between induced accumulation of H2O2, papilla formation and cell wall cross-linking, as evidenced by HRGP accumulation, and cessation of pathogen growth in resistant genotypes may help exploit host resistance in sorghum.  相似文献

The effects of CO2 enrichment on growth of Xanthomonas campestris pv. pelargonii and the impact of infection on the photosynthesis and export of attached, intact, 'source' leaves of geranium ( Pelargonium x domesticum, 'Scarlet Orbit Improved' ) are reported. Two experiments were performed, one with plants without flower buds, and another with plants which were flowering. Measurements were made on healthy and diseased leaves at the CO2 levels (35 Pa or 90 Pa) at which the plants were grown. There were no losses of chlorophyll, or any signs of visible chlorosis or necrosis due to infection. Lower numbers of bacteria were found in leaves at high CO2, suggesting growth at elevated CO2 created a less favourable condition in the leaf for bacterial growth. Although high CO2 lowered the bacterial number in infected leaves, reductions in photosynthesis and export were greater than at ambient CO2. The capacity of infected source leaves to export photoassimilates at rates observed in the controls was reduced in both light and darkness. In summary, the severity of infection on source leaf function by the bacteria was increased, rather than reduced by CO2 enrichment, underscoring the need for further assessment of plant diseases and bacterial virulence in plants growing under varying CO2 levels.  相似文献

Tomato plants at the four-leaf stage were inoculated on thefirst leaf with TMV in a growth room and the effects studiedin systemically-mfected leaves with reference to growth, virusmultiplication and changes in water, nitrogen, nitrate and chlorophyllcontents. Parallel changes in the free amino compounds werealso studied in the growth room (incident radiation 152 mwhcm-2 day-1) and in two experiments in a glasshouse (352 and226 mwh cm-2 day-1) Dry matter accumulation and leaf expansion in leaves 3, 4 and5 were checked by TMV 5–7 days after inoculation but notin leaf 2. In the period 7–25 days after inoculation therelative growth rates of whole plant and leaves 3 and 6 andnet assimilation rate were not affected by TMV. Stem heightand dry weight were not affected by TMV but ‘root’dry weight was reduced from days 5–15. Virus was presentin the stem and in leaves 2 and 6 by days 3, 5 and 15 respectivelyso that infection per se did not always check growth. Chlorophyllcontent of systemically-infected leaves was reduced 10 daysafter inoculation. Total N and ammonia contents were not affectedby TMV but infected leaves contained less nitrate. At the two lower levels of incident radiation the initial effectof TMV was to reduce the content of total free protein aminoacids and amides, which were minimal 5–7 days after inoculation.In the glasshouse experiment a reduction could be measured only1 day after inoculation or before virus was present in the youngerleaves. With high incident radiation there was no initial reductionbut an increase at day 13 when mottling symptoms were visible.Total non-protein amino acids, of which amino butyric acidwas the major constituent, were increased by TMV in all threeexperiments for up to 13 days after inoculation. It is suggested that inoculation of a leaf with TMV temporarilyinterferes with export of photosynthates and import of root-synthesisedamino acids and that the results reported above can be interpretedin this context. Evidence in support of this is adduced froman experiment in which 13 foliar sprays of gibberellic acid(2·5 ppm) were combined with TMV inoculation and thechanges in free amino compounds followed. It is concluded that analyses of the changes in individual freeamino compounds are unlikely to provide useful information concerningthe sources of virus coat protein.  相似文献

The interaction between chick embryo fibroblasts and various lectins has been studied at different stages of embryo development. There is evidence that Robinia lectin, Dolichos lectin, and Conca navalin A decrease cell number and proportion of cells incorporating [3H] thymidine in case of 8and 10day-old chick embryo fibroblasts, whereas they stimulated the proliferation of 16-dayold embryo cells. No effect was noticed in 12-day cells.
These results suggest that some cell surface changes occur during embryo development. The site number of Dolichos lectin remains the same during embryo development, and the affinity constant decreases. The site number of Robinia lectin and Concanavalin A decreases from the 8th to the 12th day of development, and slowly increases on the 16–day cells, the affinity constant remaining rather constant.
The results indicate that the age–dependent effect of lectin on embryo cells could not be directly related to the number of lectin–binding sites. Competitive binding experiments revealed that Dolichos receptor sites were distincts from binding sites of Robinia lectin and Concanavalin A, and Robina receptor sites distinct from those of Concanavalin A.
Lectin effects on embryo fibroblasts were very specific as determined by inhibitory assays.  相似文献

Effects of ozone on spring wheat ( Triticum aestivum L. cv. Satu) were studied in an open-top chamber experiment during two growing seasons (1992–1993) at Jokioinen in south-west Finland. The wheat was exposed to filtered air (CF), non-filtered air (NF), non-filtered air+35 nl l−1 ozone for 8 h d−1 (NF+) and ambient air (AA). Each treatment was replicated five times. Two wk after anthesis, after 4 wk of ozone treatment (NF+, 45 nl l−1 1000–1800 hours, seasonal mean) the net CO2 uptake of wheat flag leaves was decreased by c . 40% relative to CF and NF treatments, both initial and total activity of Rubisco and the quantity of protein-bound SH groups were decreased significantly. Added ozone also significantly accelerated flag leaf senescence recorded as a decrease in chloroplast size. The effect was significant 2 wk after anthesis, and senescence was complete after 4 wk. In the CF and NF treatments senescence was complete 5 wk after anthesis. The significant effect of ozone on the chloroplasts and net CO2 uptake 2 wk after anthesis did not affect the grain filling rate. However, since the grain filling period was shorter for ozone fumigated plants, kernels were smaller. The decrease in 1000-grain weight explained most of the yield reduction in the plants under NF+ treatment. The results indicate that wheat plants are well buffered against substantial decrease in source activity, and that shortened flag leaf duration is the major factor causing ozone-induced yield loss.  相似文献

Abstract: Disruption of Ca2+ homeostasis often leads to neuron death. Recently, the function of calcium-binding proteins as neuronal Ca2+ buffers has been debated. We tested whether calbindin D28k functions as an intracellular Ca2+ buffer by constructing bicistronic herpes simplex virus vectors to deliver rat calbindin cDNA to hippocampal neurons in vitro. Neurons were infected with vectors delivering calbindin or a negative control or were mock-infected. After 12 or 24 h of hypoglycemia, infected cells were made aglycemic during fura-2 calcium ratiometric imaging. In response to this challenge, neuronal overexpressing calbindin had less Ca2+ mobilized as compared with negative controls or mock-infected cells. Cells were assayed for survival after 12- or 24-h hypoglycemia or aglycemia. The calbindin vector decreased neuronal death due to hypoglycemia but not aglycemia. Here we demonstrate, in response to hypoglycemic challenge, both decreased Ca2+ mobilization and increased survival of cells infected with the calbindin vector.  相似文献

Although Solanum brevidens could be infected with potato virus X (PVX), potato virus Y0 (PVY0) and PVYN, no symptoms of infection were apparent and tests by double antibody sandwich ELISA, electron microscopy and sap transmission to local lesion test plants indicated that the titres of PVX were less than a tenth of those of PVY0 and PVYN were less than a hundredth of those in infected plants of PDH40, a susceptible dihaploid clone of S. tuberosum cv. Pentland Crown. Furthermore, PVY0- and PVYN- infected leaves of S. brevidens were a poor source of inoculum in aphid transmission tests. The possibility of a common mechanism and genetic basis of resistance to PVY, PVX and potato leaf roll virus in S. brevidens is discussed.  相似文献
