Summary The formation of adventitious shoots from potato tuber discs explanted onto a modified Murashige and Skoog (MS) medium containingN 6-benzylaminopurine (BAP) (3.0 mg/l), and α-naphthaleneacetic, acid (NAA) (0.01 mg/l), was affected by gibberellic acid (GA). The presence of GA in the explant medium was required for shoot formation and 3×10−10 M GA appeared optimum. However, microscopic examination of the tissue protuberances on the surface of the tuber discs from which shoots arose revealed that GA inhibited the formation of shoot meristems. Tuber discs cultured for 6 wk on MS medium containing BAP and NAA without GA did not initiate adventitious shoots that could be determined visually, but microscopic examination of the tissue protuberances revealed the presence of numerous shoot meristems. Subsequent transfer of these tuber discs to medium with GA but without BAP or NAA resulted in the formation of shoots from 100% of the recultrued dises. Thus it appears that although GA inhibits shoot meristem initiation from potato tuber discs, it is required for shoot development once meristems are initiated. Mutations in the alpha-synuclein gene have recently been identified in families with inherited Parkinson's disease and the protein product of this gene is a component of Lewy bodies, indicating that alpha-synuclein is involved in Parkinson's disease pathogenesis. A role for normal alpha-synuclein in synaptic function, apoptosis or plasticity responses has been suggested. We show here that in rat pheochromocytoma PC12 cells synuclein-1, the rat homolog of human alpha-synuclein, is highly and selectively up-regulated at the mRNA and protein levels after 7 days of nerve growth factor treatment. Synuclein-1 expression appears neither sufficient nor necessary for the neuritic sprouting that occurs within 1-2 days of nerve growth factor treatment. Rather, it likely represents a component of a late neuronal maturational response. Synuclein-1 redistributes diffusely within the cell soma and the neuritic processes in nerve growth factor-treated PC12 cells. Cultured neonatal rat sympathetic neurones express high levels of synuclein-1, with a diffuse intracellular distribution, similar to neuronal PC12 cells. These results suggest that levels of synuclein-1 may be regulated by neurotrophic factors in the nervous system and reinforce a role for alpha-synuclein in plasticity-maturational responses. In contrast, there is no correlation between synuclein expression and apoptotic death following trophic deprivation.  相似文献

Prolonged or excess stimulation of excitatory amino acid receptors leads to seizures and the induction of excitotoxic nerve cell injury. Kainic acid acting on glutamate receptors produces degeneration of vulnerable neurons in parts of the hippocampus and amygdala, but the exact mechanisms are not fully understood. We have here investigated whether the anti-apoptotic protein Bruce is involved in kainic acid-induced neurodegeneration. In the rat hippocampus and cortex, Bruce was exclusively expressed by neurons. The levels of Bruce were rapidly downregulated by kainic acid in hippocampal neurons as shown both in vivo and in cell culture. Caspase-3 was activated in neurons exhibiting low levels of Bruce causing cell death. Likewise, downregulation of Bruce using antisense oligonucleotides decreased viability and enhanced the effect of kainic acid in the hippocampal neurons. The results show that Bruce is involved in neurodegeneration caused by kainic acid and the downregulation of the protein promotes neuronal death.  相似文献

The internodes of Cephaelis ipecacuanha elongated when cultured on Gamborg B5 solid medium supplemented with 0.5 or 1 mg/L gibberellic acid (GA3). The size of the elongated internode doubled in length compared to the untreated shoots, and the adventitious shoots formed on the elongated internodes. The shoots grew easily into plantlets without the use of auxin for rooting. The ex vitro regenerates cultivated in the greenhouse showed normal characteristics. Emetic alkaloids were detected in the leaves of in vitro shoots and the roots of regenerates cultivated in the greenhouse. This method using GA3 propagated numerous plants at a rate of more than 100 times compared to the method without GA3.  相似文献

Indole-3-acetic acid (IAA), fusicoccin and weak acids all lower the cytoplasmic pH (pHi) and induce elongation growth of maize (Zea mays L.) coleoptiles. Gibberellic acid (GA3) also induces elongation growth and we have used confocal laser scanning microscopy to study the effects of GA3 on pHi employing the pH-indicator dyes, 2,7-bis(2-carboxyethyl)-5-(and-6) carboxyfluorescein and carboxy-semi-naphthorhodafluor-1. We confirm that GA3 induces growth significantly in light-grown but only slightly or not at all in dark-grown coleoptiles. The growth induced by IAA treatment was similar in light- and dark-grown coleoptiles. The pHi decreased by up to 0.6 units during the first 7 min of GA3 or IAA treatment of both light- and dark-grown coleoptiles. Gibberellic acid inhibited IAA-induced growth of dark-grown coleoptiles. Hence, in dark-grown coleoptiles GA3 may activate either directly or indirectly reactions that interfere with the signalling pathway leading to elongation growth. 为探索宁夏枸杞对枸杞瘿螨致瘿过程诱导的激素响应及外源水杨酸对枸杞瘿螨的影响,利用LC-MS/MS技术对枸杞瘿螨为害后宁夏枸杞叶片中水杨酸(SA)、茉莉酸(JA)、生长素(Auxin)和脱落酸(ABA)4种内源激素进行测定,并通过浸叶法研究外源SA对枸杞瘿螨侵染过程、虫瘿生长和种群增长的影响。结果表明:枸杞瘿螨的致瘿行为可引发枸杞叶片中多种内源激素产生变化,其中SA和JA的变化最显著,分别是对照组含量的4.0和13.0倍,而Auxin和ABA的变化不显著;外源SA对枸杞瘿螨虫瘿生长有显著抑制作用,且随时间的延长,抑制作用增强,同时虫瘿内瘿螨种群数量受到显著抑制。枸杞瘿螨能够同时诱导枸杞产生SA和JA抗性,外源SA对枸杞瘿螨虫瘿生长和瘿螨种群数量均具有显著抑制作用,利用外源SA防治枸杞瘿螨具有一定的应用前景。  相似文献

The production of hydrogen peroxide by isolated hepatocytes in response to lauric, palmitic and oleic acids, a measurement of peroxisomal fatty acid oxidation, is inhibited by phenothiazines under conditions in which ketone body production, a measurement of mitochondrial fatty acid oxidation, does not reveal inhibition of mitochondrial activity. This novel finding provides a pharmacological tool for the study of peroxisomal function in whole cells. The mechanism of this effect of phenothiazines, detected in hepatocytes from rats treated with a peroxisome proliferation inducing drug, is not yet known.  相似文献

Summary Gibberellic acid inhibition of betacyanin biosynthesis has been studied in Amaranthus caudatus L. using the pigment precursors L-tyrosine and L-dihydroxyphenylalanine. Precursors fed to gibberellic acid (GA) treated seedlings completely recovered betacyanin synthesis while the GA induced growth enhancement remained unaltered. Cancer cachexia is characterized by skeletal muscle wasting that is mainly supported by hypercatabolism. Muscle atrophy has been suggested to depend on impaired IGF-1 signal transduction pathway. The present study has been aimed at investigating the IGF-1 system in rats bearing the AH-130 hepatoma, a well-characterized model of cachexia. IGF-1 mRNA expression in the gastrocnemius of tumor hosts progressively decreases to approximately 50% of controls. By contrast, both IGF-1 receptor and insulin receptor mRNA levels increase in day 7 AH-130 hosts. IGF-1 and insulin circulating levels, as well as IGF-1 expression in the liver, are reduced. Muscle wasting in the AH-130 bearers is associated with hyperactivation of the ubiquitin-proteasome system. Consistently, the mRNA levels of ubiquitin and of the ubiquitin ligases atrogin-1 and MuRF1 are significantly increased in the gastrocnemius of day 7 AH-130 hosts. Exogenous IGF-1 administered to tumor bearers does not prevent cachexia. IGF-1 mRNA levels also have been evaluated in the gastrocnemius of AH-130 hosts treated with pentoxifylline, an inhibitor of TNF-alpha synthesis, alone or combined with formoterol, a beta(2)-adrenergic agonist. Both treatments partially correct muscle atrophy without modifying IGF-1 and atrogin-1 mRNA levels, whereas MuRF1 hyperexpression is reduced by the combination of pentoxifylline with formoterol. These results demonstrate for the first time that the IGF-1 system is downregulated in cancer cachexia, although the underlying mechanism remains unknown. Moreover, no simple relation linking IGF-1 and/or atrogin-1 mRNA levels and muscle atrophy could be observed in these experimental conditions. Further studies are thus needed to clarify both issues.  相似文献

In a mouse melanoma metastasis model it has been recently shown that ADAM15 overexpression in melanoma cells significantly reduced the number of metastatic nodules on the lung. Unfortunately, the expression of ADAM15 in human melanoma tissue has not been determined so far. In our study, we characterized the expression of ADAM15 in tissue micro-arrays of patients with primary melanoma with melanoma metastasis. ADAM15 was expressed in melanocytes and endothelial cells of benign nevi and melanoma tissue. Importantly, ADAM15 was significantly downregulated in melanoma metastasis compared to primary melanoma. We further demonstrate that IFN-γ and TGF-β downregulate ADAM15 protein levels in melanoma cells. To investigate the role of ADAM15 in melanoma progression, we overexpressed ADAM15 in melanoma cells. Importantly, overexpression of ADAM15 in melanoma cells reduced the migration, invasion and the anchorage dependent and independent cell growth of melanoma cells. In summary, the downregulation of ADAM15 plays an important role in melanoma progression and ADAM15 act as a tumorsuppressor in melanoma.  相似文献

Allochronic isolation between the two host races of the bisexual generation ofA. mukaigawae (Mukaigawa) (Hymenoptera: Cynipidae) was demonstrated in the field. Assortative mating seems to occur in the bisexual generation of each race. Genetic changes of ovipositional preference probably played an important role in the host race formation; it is likely that little or no genetic change in survival was required in this process. A difference in host phenology seems to have caused the allochronic isolation of the bisexual generation between the parent population on the original host tree species and its daughter population on the new host tree species. Gene flow between them may have been restricted ever since the initial host shift. The host race formation of this wasp probably represents an example of sequential evolution.
Résumé L'isolement allochronique de 2 races spécifiques aux hôtes différents a été prouvé dans la nature pour la génération bisexuée deA. mulaigawae (Hymen., Cynip.). Des accouplements assortis semblent exister à l'intérieur de la génération bisexuée de chaque race. Des différences génétiques dans le choix du lieu de ponte doivent jouer un rôle important dans la formation de ces races; il n'est probablement pas nécessaire qu'il y ait des changements génétiques dans la survie, pour obtenir cette genèse. Une différence dans la phénologie des hôtes semble avoir entraîné l'isolement allochronique dans la génération bisexuée entre les populations parentales suivant l'espèce originelle d'arbre hôte, et dans ses populations filles suivant les nouvelles espèces d'arbres hôtes. Le flux génique entre ces populations peut avoir été limité depuis le changement initial d'hôtes. Chez ce cynipide la formation de races, en fonction de l'hôte, représente probablement un exemple d'évolution séquentielle.

Crown gall tumors were initiated in a variety of plant species by infection with Agrobacterium tumefaciens strain B6 and the concomitant changes in the tissue levels of phytohormones, mainly indole-3-acetic acid (IAA) and cytokinins, were analyzed. A comparison was made of these hormones with those produced by virulent and avirulent strains of the bacterium in liquid culture and with those of bacteria-free crown gall callus cultures. Specific radioimmunoassays were employed for hormone determinations. An assay for the quantitation of femto-mol amounts of isopentenyladenosine and related cytokinins was newly developed and is described in detail. The results can be summarized as follows: Virulence in strain B 6 is associated with the ability to release trans-zeatin and increased amounts of IAA into the surrounding environment. In many, but not all plants analyzed, the development of crown gall tumors is also associated with a sharp rise in the levels of trans-zeatin-type zytokinins and IAA (e.g., Euphorbia lathyris, Catharanthus roseus). Crown gall calli growing on hormone-free media varied greatly in their cytokinin levels. In a culture of Nicotiana tabacum, both trans-zeatin and isopentenyladenine or related cytokinins were not detected. Gibberellic acid (GA), a plant hormone stimulating plant growth and development, is a tetracyclic di-terpenoid compound. GAs stimulate seed germination, trigger transitions from meristem to shoot growth, juvenile to adult leaf stage, vegetative to flowering, determines sex expression and grain development along with an interaction of different environmental factors viz., light, temperature and water. The major site of bioactive GA is stamens that influence male flower production and pedicel growth. However, this opens up the question of how female flowers regulate growth and development, since regulatory mechanisms/organs other than those in male flowers are mandatory. Although GAs are thought to act occasionally like paracrine signals do, it is still a mystery to understand the GA biosynthesis and its movement. It has not yet confirmed the appropriate site of bioactive GA in plants or which tissues targeted by bioactive GAs to initiate their action. Presently, it is a great challenge for scientific community to understand the appropriate mechanism of GA movement in plant’s growth, floral development, sex expression, grain development and seed germination. The appropriate elucidation of GA transport mechanism is essential for the survival of plant species and successful crop production.  相似文献

Adiponectin protects the liver from steatosis caused by obesity or alcohol and therefore the influence of adiponectin on human hepatocytes was analyzed. GeneChip experiments indicated that recombinant adiponectin downregulates aldehyde oxidase 1 (AOX1) expression and this was confirmed by real-time RT-PCR and immunoblot. AOX1 is a xenobiotic metabolizing protein and produces reactive oxygen species (ROS), that promote cell damage and fibrogenesis. Adiponectin and fenofibric acid activate peroxisome proliferator-activated receptor-alpha (PPAR-alpha) and both suppress AOX1 protein and this is blocked by the PPAR-alpha antagonist RU486. Obesity is associated with low adiponectin, reduced hepatic PPAR-alpha activity and fatty liver, and AOX1 was found induced in the liver of rats on a high-fat diet when compared to controls. Free fatty acids and leptin, that are elevated in obesity, failed to upregulate AOX1 in vitro. The current data indicate that adiponectin reduces AOX1 by activating PPAR-alpha whereas fatty liver disease is associated with elevated hepatic AOX1. High AOX1 may be associated with higher ROS well described to induce fibrogenesis in liver tissue but may also influence drug metabolism and activity.  相似文献
