A small isometric virus has been isolated from larvae of the Guatemala potato tuber moth, Tecia solanivora (Povolny), collected in Ecuador. It was designated the Anchilibi virus (AnchV). The non-enveloped viral particles have an estimated diameter of 32 ± 2 nm. Three major proteins were found in virions, with estimated sizes of 102.0 ± 2.1, 95.8 ± 2.0 and 92.4 ± 1.5 kDa for AnchV as determined by polyacrylamide gel electrophoresis. After denaturing agarose gel electrophoresis, the genome of AnchV appeared to be a tri-segmented single-stranded RNA with fragment sizes of 4.1 ± 0.2, 2.8 ± 0.2 and 1.65 ± 0.2 kb. In addition to a high virulence towards its original host, AnchV also caused high mortality in larvae of two other potato tuber moth species, Phthorimaea operculella (Zeller) and Symmetrischema (tangolias) plaesiosema (Turner). Electron microscopy confirmed that AnchV replication occurs in the cell cytoplasm, mainly in vesicles. Several important characteristics exhibited by this virus differ from those reported for known families of insect viruses. Thus, AnchV might be member of a new taxonomic group.  相似文献

The moth Tecia (Scrobipalpopsis) solanivora Povolny (Lepidoptera: Gelechiidae) is the most important pest of potato, Solanum spp., in Central America and adjacent South American countries. Insecticide treatments are not sufficiently effective; therefore, we investigated the feasibility of pheromone-mediated mating disruption for control of T. solanivora. Pheromone dispensers were formulated with 70 mg of the three sex pheromone compounds (E)-3-dodecenyl acetate, (Z)-3-dodecenyl acetate, and dodecyl acetate, in a ratio of 100:56:100, respectively. Male attraction to these compounds is optimal at a ratio of 100:1:20, thus the mating disruption dispensers contained an off-blend, which attracted only a few males. Nonetheless, one mating disruption dispenser suppressed male attraction to calling females in a flight tunnel and reduced male activation in response to female pheromone. Communication disruption is accordingly due to camouflage of the female signal and possibly due to a reduction of male responsiveness by sensory imbalance. Only a few males were observed in a 3-ha potato field treated with 84 g pheromone/ha, compared with an untreated control field. During 2 mo, male attraction to traps baited with calling females or synthetic pheromone was strongly reduced. This reduction confirms the potential of mating disruption for management of T. solanivora. The efficacy of the pheromone treatment can be further improved by earlier dispenser application, by increased dispenser load, and by treatment of larger fields to reduce immigration of mated females.  相似文献

Nine polymorphic microsatellite markers were isolated from Tecia solanivora, one of the most serious pests of potato tubers in Central and South America. As found in other studies of Lepidoptera, development of microsatellites is a difficult task: in our case, despite the large number of clones sequenced (796), of which 70 were unique, only nine loci were found to be both variable, and in Hardy-Weinberg equilibrium, No null alleles were detected. The loci were tested in three other co-occurring Gelechiidae species, one of which was variable. These loci will be used to provide a greater understanding of the genetic changes occurring during the invasive process in this species.  相似文献

The biochemical properties of the digestive alpha-amylase from Tecia solanivora larvae, an important and invasive insect pest of potato (Solanum tuberosum), were studied. This insect has three major digestive alpha-amylases with isoelectric points 5.30, 5.70 and 5.98, respectively, which were separated using native and isoelectric focusing gels. The alpha-amylase activity has an optimum pH between 7.0 and 10.0 with a peak at pH 9.0. The enzymes are stable when heated to 50 degrees C and were inhibited by proteinaceous inhibitors from Phaseolus coccineus (70% inhibition) and P. vulgaris cv. Radical (87% inhibition) at pH 6.0. The inhibitors present in an amaranth hybrid inhibited 80% of the activity at pH 9.0. The results show that the alpha-amylase inhibitor from amaranth seeds may be a better candidate to make genetically-modified potatoes resistant to this insect than inhibitors from common bean seeds.  相似文献

The Guatemalan potato moth Tecia solanivora (Povolny) recently invaded part of South America, colonizing zones where Phthorimaea operculella (Zeller), another potato moth species belonging to the same group, was previously established. T. solanivora is now the major insect pest of potato in this area encompassing Venezuela, Colombia and Ecuador. P. operculella granulovirus (PhopGV) (Betabaculovirus) is a biocontrol agent to be considered for the simultaneous management of these two potato pests, instead of classical chemical insecticides. In a previous work, five PhopGV isolates were isolated in Colombia from T. solanivora and were tested against larvae of the same species showing variable efficacies. Infections with mixtures of different genotypes of Baculoviruses had been carried out in a wide range of species and several showed interesting results. In the present study, the effect of sequential passages of PhopGV in P. operculella and T. solanivora larvae was analyzed through biological assays. Three different mixtures containing a Peruvian PhopGV isolate (Peru) adapted to P. operculella and a Colombian PhopGV isolate (VG003) adapted to T. solanivora were tested. A preliminary analysis of the correlation between the genotypic marker egt gene and the level of pathogenicity after a variable number of replication cycles was made. Mixtures of virus isolates showed a higher efficacy in both hosts compared to individual PhopGV isolates. This higher pathogenicity was maintained through passages. In P. operculella the mixtures were between 2.8 and 23.6-fold (from 7.15 OB/mm(2) to 0.10 OB/mm(2)) more pathogenic than isolate Peru applied alone. In T. solanivora they were between 2.3 and 4.9-fold (from 12.29 OB/mm(2) to 1.25 OB/mm(2)) more pathogenic than isolate VG003 alone. Viral biopesticide containing a mixture of selected genotypes active against each hosts seemed suitable for the development of a biopesticide aimed to simultaneously control P. operculella and T. solanivora.  相似文献

Transgenic maize, Zea mays L., expressing the Bacillus thuringiensis (Bt) CrylAb toxin has been planted to extensive areas across the United States and several other countries, but no resistance has been documented in field populations oflepidopteran target pests. This article describes the first report of resistance alleles to commercially available Cry1Ab Bt maize in a Louisiana population of sugarcane borer, Diatraea saccharalis (F.) (Lepidoptera: Crambidae). Two hundred thirteen two-parent isolines of D. saccharalis were screened for Cry1Ab resistance on Bt maize leaf tissue using an F2 screening technique. Larvae representing three isolines survived >15 d on Bt tissue in the F2 generation. The second generation backcross progeny (B1F2) derived from isoline 52 completed larval development on Bt maize in the greenhouse. Segregation and resistance frequency analysis associated with isoline 52 suggested that Bt resistance is probably determined by a nearly completely recessive allele at a single locus. With this assumption, the estimated resistance allele frequency in this population is 0.0023, within a 95% confidence interval of 0.0003-0.0064.  相似文献

Sensitivities of Helicoverpa armigera (Hübner) field populations to Bacillus thuringiensis (Bt) insecticidal protein CrylAc were monitored during 1998-2000 in China. A total of 41 strains was sampled, and most of them were collected from Bt cotton planting regions. The range of IC50 values (concentration producing 50% inhibition of larval development to third instar) among different populations in 1998,1999, and 2000 were 0.020-0.105 microg/ml, 0.016-0.099 microg/ml, and 0.016-0.080 microg/ml, respectively. Diagnostic concentration studies (IC99) showed that the percentage of individuals reaching third instar ranged from 0 to 4.35%, with only eight of the 41 tested populations showing values above 0%. Also interesting was a trend over successive years in which fewer populations contained individuals that survived the diagnostic concentration by reaching third instar. Considering these data, it was determined that the field populations sampled during the 3-yr study were susceptible to CrylAc protein, and that movement toward resistance among H. armigera populations was not apparent.  相似文献

The potato tuber moth, Phthorimaea operculella (Zeller), in tropical and subtropical countries, is the most destructive pest of potato, Solanum tuberosum L. The larvae attack foliage and tubers in the field and in storage. The purpose of this study was to evaluate the efficacy of a Bt-cry5 transgene to control the potato tuber moth in tuber tissues. Tuber bioassays using stored (11-12 mo old) and newly harvested tubers of Bt-cry5-Lemhi Russet and Bt-cry5-Atlantic potato lines showed up to 100% mortality of 1st instars. Mortality was lowest in the newly harvested tubers of Bt-cry5-Atlantic lines (47.1-67.6%). Potato tuber moth mortality was 100% in the Bt-cry5-Spunta lines that were transformed with Bt-cry5 gene controlled by the CaMV 35S promoter (pBIML5 vector) and in 2 of 3 lines transformed with Bt-cry5 gene controlled by the Gelvin super promoter (pBIML1 vector). The transgenic Spunta lines expressing Bt-cry5 controlled by the patatin promoter (pBMIL2 vector) showed the lowest tuber moth mortality (25.6 and 31.1%). The Bt-cry5 transgenic lines with high tuber expression of B. thuringiensis have value in an integrated pest management system to control potato tuber moth.  相似文献

Fitness costs associated with resistance to transgenic crops producing toxins from Bacillus thuringiensis (Bt) could reduce male response to pheromone traps. Such costs would cause underestimation of resistance frequency if monitoring was based on analysis of males caught in pheromone traps. To develop a DNA-based resistance monitoring program for pink bollworm, Pectinophora gossypiella (Saunders) (Lepidoptera: Gelechiidae), we compared the response to pheromone traps of males with and without cadherin alleles associated with resistance to Bt cotton (Gossypium hirsutum L.). When irradiated males from two hybrid laboratory strains with an intermediate frequency of resistance alleles were released in large field cages, the probability of capture in pheromone traps was not lower for males with resistance alleles than for males without resistance alleles. These results suggest that analysis of trapped males would not underestimate the frequency of resistance. As the time males spent in traps in the field increased from 3 to 15 d, the success of DNA amplification declined from 100 to 30%. Thus, the efficiency of a DNA-based resistance monitoring program would be improved by analyzing males remaining in traps for 3 d or less.  相似文献

The larva of Scrobipalpuloides absoluta , a South American moth, is the most devastating insect pest of tomato production in Chile. The potential for using bacterial insecticides was studied analysing the relative toxicity of native Bacillus thuringiensis (BT) isolates belonging to the Chilean collection. The polymerase chain reaction (PCR) technique was used in order to facilitate the prescreening. Mixtures of homologous specific primers to regions within genes encoding CryI , CryIII and CryIV crystal proteins were employed to generate a PCR product profile of each BT isolate. Four isolates were selected and further characterized by means of SDS-PAGE, Western blot and bioassays on fourth-instar S. absoluta larvae. Relative toxicities were evaluated by LD50 determinations. The entomocidal activity of isolate 121e, an autoagglutinating strain, was threefold higher than toxin synthesized by B. thuringiensis var. kurstaki . This native strain was also active against Culex pipiens larvae, although much less than towards S. absoluta .  相似文献

Abstract  Trichome-based host plant resistance of Lycopersicon species to potato moth, Phthorimaea operculella (Zeller) (Lepidoptera: Gelechiidae), was examined in a laboratory bioassay. Neonate P. operculella were placed on the foliage of accessions of L. hirsutum , L. hirsutum f. glabratum , L. pennellii , L. cheesmanii f. minor and L. esculentum for 48 h . At the end of this period, larval mortality, the numbers of larvae emigrating from, and mining into, the leaf surface were recorded as were leaf area and the densities of trichomes. Analysis by categorical logistic regression found accessions differed in levels of resistance to P. operculella ( P  < 0.001). Further analysis using generalised linear models showed mortality was associated with high densities of type VI and low densities of type V trichomes; emigration from the leaf was associated with high densities of type I and type VII trichomes; and the numbers of mines associated with low densities of type I and type VI trichomes and increased leaf area. Although results indicate that certain accessions of L. hirsutum and L. hirsutum f. glabratum may be most appropriate for inclusion in a breeding program aimed at introducing trichome-based host plant resistance of wild Lycopersicon species into the tomato, further research is necessary to fully understand the mechanisms.  相似文献

Liquid suspensions and dry formulations of a granulovirus (family Baculoviridae, genus Granulovirus, PoGV) derived from infected larvae and the bacterium Bacillus thuringiensis subsp. kurstaki (Berliner) (Btk) were evaluated for control of the potato tuberworm, Phthorimaea operculella (Zeller) (Lepidoptera: Gelechiidae), in stored tubers. Laboratory bioassays at 25 degrees C showed that both PoGV and a wettable powder (WP) formulation of Btk incorporated with carriers (water, talc, sand, diatomaceous earth, and kaolin clay), were effective against neonate larvae. Depending on the technique, 100% larval mortality was achieved at concentrations as low as 0.025 larval equivalents (LE) PoGV per kg tuber and 150 mg Btk WP per kg tuber. However, 100% mortality was never achieved with tests on preinfested tubers, ostensibly due to the higher dosage required to kill older instars inside tubers. The most effective PoGV formulations were dipping (water) and talc, with dipping most effective for postinfestation treatments, causing up to 91.6% mortality at 0.4 LE per kg. There was no significant effect of formulation in the Btk treatments. The protective effects of residues were also evaluated under longer-term storage conditions. Batches of tubers treated with PoGV or Btk via dipping (up to 0.1 LE and 150 mg WP per kg tuber) were stored in cages containing an initial potato tuberworm infestation (10% of tubers). Although potato tuberworm populations were reduced by up to 98.4% after 2 mo at 25 degrees C, no treatments prevented the development and reproduction of the F1 generation. The sprouting of stored tubers seemed to be a limiting factor for sustained control. No significant treatment effects were detected in similar cages held at 12 degrees C for 4.5 mo. Improved strategies for the application of PoGV and Btk for long-term potato tuberworm control in tuber stores, including the use of chemical sprout suppressants, are discussed.  相似文献

Gamma irradiation doses applied to inhibit potato sprouting were tested against potato tuber moth larval and pupal stages. Young larvae and pupae were more susceptible to irradiation injuries than older ones. When larvae of different ages were exposed to doses > or = 100 Gy, only 13-35% pupated, but no adult emergence was obtained. The exposure of 1-1.5- or 3-3.5-d-old pupae to 150 Gy induced high level of sterility and remarkable reduction in female mating ability and fecundity, whereas the reduction was less noticeable for 5-5.5-d-old pupae. Postharvest gamma irradiation of potatoes could be considered as an efficient control approach against potato tuber moth larval and pupal infestations.  相似文献

Our laboratory has demonstrated previously that Bacillus thuringiensis (Bt) crystal (Cry) proteins present in the Cry5 and Cry6 subclades intoxicate free-living nematodes. In this study, we tested whether the expression of nematicidal Cry6A in transgenic plants provided protection against plant-parasitic nematodes. As bacterial codon usage is incompatible with expression in plants, two different codon-modified cry6A genes were synthesized for expression in plants. One was designed by maintaining codon diversity whilst removing codons not common in plants, and the other was designed by selecting the optimal codon for each amino acid based on the Arabidopsis genome. Both versions of the cry6A gene, driven by the constitutive cauliflower mosaic virus 35S promoter, were introduced into tomato roots via Agrobacterium rhizogenes . Although both were found to express Cry6A protein, the codon diversity gene generated superior expression. These Cry6A-expressing roots were then challenged with root-knot nematode, Meloidogyne incognita . Three different infection parameters were compared between Cry6A-expressing roots and control roots transformed with empty vector or green fluorescent protein (GFP). These data demonstrated that M. incognita was able to ingest the 54-kDa Cry6A, and that Cry6A intoxicated the parasitic nematode, as indicated by a decrease in progeny production of up to fourfold. These results indicate, for the first time, that a Bt Cry protein can confer plant resistance to an endoparasitic nematode, and that Cry proteins have the potential to control plant-parasitic nematodes in transgenic plants.  相似文献

Relative resistance of paddy varieties to the Angoumois grain moth, Sitotroga cerealella was investigated in the laboratory at a temperature of 28 ± 1.0°C and relative humidity of 65 ± 1.5%. The study was carried out on Indian Agricultural Research Institute (IARI), New Delhi, India varieties and a Taiwan variety. The larvae of S. cerealella were allowed to feed and developed inside the grain while adults did not feed. Results showed that the performance of S. cerealella was least in T (N)-1 (Taichung Native-1), with the longest developmental time (26.3 days) and egg to adult survival of 38% making the variety the most resistant. The performance was best in Pusa 44 with the shortest developmental time (23.0 days) and egg to adult survival of 68.7% making the variety the least resistant. The heaviest females and males with 4.04 g and 2.38 g, respectively, were reared on Pusa sugandh 3, which are large seeds. Moths reared on T (N)-1 had the lowest weights for males and females. Unmated females lived longer than their mated counterparts while females generally lived longer than males. Fecundity was highest (164.2 eggs/female) in Pusa 44 and lowest (108.1 eggs/female) in T (N)-1. Hatchability ranged from 81.2% in T (N)-1 to 96.2% in Pusa 834. Generally, grain characters tested in this study influenced moth herbivore performance but the influence was not significant except for grain width which appeared to be the most important trait. With advances in biotechnology, it is possible to transfer desirable characters to other varieties to improve their resistance to S. cerealella and other stored products insects. However, care must be taken since this could change the grains susceptibility to some pests affecting pest-host relationships.  相似文献

Bacillus thuringiensis var. kurstaki (Berliner) (Bt) has been suggested as a biological control agent for Tuta absoluta (Meyrick). The objective of this study was to determine the interaction between abamectin, azadirachtin, indoxacarb, chlorantraniliprole, dichlorvos and metaflumizone with Bt. Effect of recommended doses of the chemical insecticides on colonisation of Bt was also investigated in culture medium. Except for metaflumizone, none of the chemicals tested reduced the colonisation of Bt compared with control. Interaction between Bt and the chemical insecticides on 2nd-instar larvae was also assessed. In interaction tests, Bt was applied at LC50 level, 0, 12, 24 or 36 h after treating the larvae with LC10 or LC25 of the chemical insecticides. An antagonistic effect was observed in all treatments where Bt was applied immediately after the chemical insecticide. Also, antagonism was observed when treatment with Bt was done 12 h after azadirachtin and metaflumizone applications. Applying Bt 12 and 24 h after treatment with LC25 of chlorantraniliprole, dichlorvos and abamectin resulted in synergism. But, synergism with LC10 of dichlorvos and abamectin was observed only after 12 h. Additive effect was observed in the rest of the time and concentration combinations. Based on the results obtained, simultaneous use of the chemical insecticides tested and Bt is not recommended for T. absoluta control; and an appropriate time interval should be taken into consideration accordingly.  相似文献

Selection of resistance in Spodoptera exigua (Hubner) to an HD-1 spore-crystal mixture, CryIC (HD-133) inclusion bodies, and trypsinized toxin from Bacillus thuringiensis subsp. aizawai and B. thuringiensis subsp. entomocidus was attempted by using laboratory bioassays. No resistance to the HD-1 spore-crystal mixture could be achieved after 20 generations of selection. Significant levels of resistance (11-fold) to CryIC inclusion bodies expressed in Escherichia coli were observed after seven generations. Subsequent selection of the CryIC-resistant population with trypsinized CryIC toxin resulted, after 21 generations of CryIC selection, in a population of S. exigua that exhibited only 8% mortality at the highest toxin concentration tested (320 (mu)g/g), whereas the 50% lethal concentration was 4.30 (mu)g/g for the susceptible colony. Insects resistant to CryIC toxin from HD-133 also were resistant to trypsinized CryIA(b), CryIC from B. thuringiensis subsp. entomocidus, CryIE-CryIC fusion protein (G27), CryIH, and CryIIA. In vitro binding experiments with brush border membrane vesicles showed a twofold decrease in maximum CryIC binding, a fivefold difference in K(infd), and no difference in the concentration of binding sites for the CryIC-resistant insects compared with those for the susceptible insects. Resistance to CryIC was significantly reduced by the addition of HD-1 spores. Resistance to the CryIC toxin was still observed 12 generations after CryIC selection was removed. These results suggest that, in S. exigua, resistance to a single protein is more likely to occur than resistance to spore-crystal mixtures and that once resistance occurs, insects will be resistant to many other Cry proteins. These results have important implications for devising S. exigua resistance management strategies in the field.  相似文献

The processes of female searching by male potato tuber moths,Phthorimaea operculella, were analyzed. The behavioral components to copulation were antennal cleaning, quiescence, walking, wing fanning, contact with female, hair brush display, copulation attempt, and copulation. Males did not always succeed in mating on their first attempt. Searching behavior of males changes to “area-restricted searching” after contact with a female.
