Changes in biliverdin-binding cyanoprotein content in whole body and tissue extracts during development of nymphal and adult (non-diapause) bean bugs, Riptortus clavatus were analyzed by rocket immunoelectrophoresis (RIE). RIE using anti-CPegg serum can be used to determine the content of CP-A (Cp-1, 2 and 3) and CP-B (CP-4) separately. During the nymphal stage CP content of whole body changes cyclically in each instar. In the first nymphal instar, CPegg is the main CP which disappears during the first-second instar ecdysis. In nymphal bugs from the 2nd to 4th instars only CP-B (CP-4) is detected, and at the beginning of each instar the CP content is very low but increases toward the next ecdysis, after which CP decreases and disappears very rapidly. In the 5th nymphal instar, CP-B is the major CP but CP-A (CP-1, 2 and 3) is also detected. These changes in whole body CP content of 5th instar nymphs are observed in both females and males. The content of total CPs in the 5th instar nymph reaches about 1000 μg in the whole insect. During nymphal-adult ecdysis, nymphal CPs decrease and disappear at day 2 after emergence. In female adults CP-A (CP-1 only) increases rapidly after day 4 of adult emergence, while no CP is detected in male adults. In females CPs were detected only in the fat body, hemolymph and ovary. In the mid-5th-instar nymphs, CPs (CP-A and B) are mainly distributed in the hemolymph. CPs in the Hemolymph decrease during nymphal-adult ecdysis, whereas they increase in the fat body. CPs disappear from both the hemolymph and fat body by 2 days after ecdysis. Subsequently in the adult stage only CP-A increases again in the fat body and ovary. By tracer experiments using [35S]-methionine, the fat body was shown to be the site of CP synthesis. CP-A and B synthetic activity was detected in nymphal females whereas, only CP-A synthesis was observed in adult females, while no CP synthesis was seen in adult males.  相似文献

Injection of azadirachtin into females of Locusta migratoria at the beginning of the last nymphal instar prevented molting to the adult stage, and many of these locusts survived for long periods as overage fifth-instar nymphs. Overage female nymphs synthesized vitellogenin; maximum vitellogenin content in their hemolymph was 6–7 times higher than that found in normal adult females. The overage female nymphs developed vitellogenic oocytes, but development was retarded to some extent: although vitellogenin did accumulate in the proximal oocytes, their maximum average length was only about 2.8 mm (compared to 6.2 mm in normal adult females) and extensive oocyte resorption was observed. Thus, attainment of adult competence of the organs and processes involved in female reproduction is independent to a considerable extent from the process of overt adult morphogenesis.  相似文献

Aspects of the nymphal/adult developmental change were investigated in biometric studies of several species of Plecoptera: Nemouridae near Schlitz, Hesse, Germany. Preliminary information on the mayfly, Baetis vernus Curtis, is also provided. Nemourid nymphs pass through 3 wing bearing stages before reaching adulthood. Instars can be identified by their characteristic shapes, as expressed by the wing length/head width (WL/HW) ratio. Size does not allow instar discrimination, mainly due to sexual size differences. HW is ca 10% larger in last instar female than in male nemourid nymphs; exuviae shed at the moult to adult represent about 14% of nymphal ash free dry weight (AFDW). Biomass lost with exuviae during the many larval moults should be accounted for in estimates of production. Freshly emerged nemourid females are about 6% larger and 30% heavier than males. The HW/AFDW relationship is the same in both sexes. Through terrestrial feeding during adult life, males double their weight on average. Mature females are up to three times heavier than freshly emerged ones. They invest about 30% of their final AFDW in reproduction.Shape of last instar nymphal Baetis was expressed as the ratio wing length/mesonotum length. It is size-dependent, a characteristic, instar-specific shape may not occur in this mayfly. Nymphal and subimaginal exuviae together represent about 14% of last instar nymphal dry weight. Females of Baetis are about 55% heavier than males. Unlike in Plecoptera, the size/weight (ML/AFDW) relationship differs between sexes.  相似文献

Immunological properties and content changes of cyanoprotein (CP) were investigated in the bean bug, Riptortus clavatus. Anti-CPegg serum was prepared by immunizing a rabbit with CP purified from eggs (CPegg). In the Ouchterlony double diffusion test, the precipitin line between CPegg and anti-CPegg serum fused with that of non-diapause and diapause female hemolymph and anti-CPegg serum. Rocket immunoelectrophoresis (RIE) using anti-CP serum showed two types of rockets (A and B) depending on the samples. Namely, CPegg and non-diapause female adult hemolymph formed A rockets (heavy-stained with Coomassie Brilliant Blue) and early diapause female adult hemolymph formed B rockets (light-stained), but hemolymph from fifth instar nymphs formed both A and B rockets. Both rockets A and B were demonstrated by SDS-PAGE analysis of the precipitin lines to be formed from the same CP subunit (MW, 76,000). CP-1, 2 and 3 bands from native PAGE of nymphal hemolymph formed A rockets and CP-4 formed B rockets. The contents of CP-A (CP-1 to 3) and CP-B (CP-4) were separately determined by measuring the sizes of rocket A and B. CP-A and CP-B content were demonstrated to increase during the development of the last instar nymph and decrease at adult emergence by RIE analysis of non-diapause female whole body extracts. CP-B is predominant in the nymphal stage. In the early adult stage (day 2 and 3 after emergence), neither CP-A nor CP-B were detected. Only CP-A appeared again at day 4 after emergence and increased during development and vitellogenesis of non-diapause females.  相似文献

The concentration of the juvenile hormone-binding protein (JHB) in hemolymph was determined throughout the last nymphal instar. It was found to be 3.9 μM at the molt to the instar, rising to 13 μM by mid-instar, and dropping to 6.7μM the day before emergence. Endocrine control of its production during the last nymphal instar could not be established. The apparent juvenile hormone esterase (JHF) activity was low at the molt to the last instar, but rose about fivefold by mid-instar, and then modestly declined. On the day of emergence, JHF activity rose to the highest level observed. A four- to fivefold increase in absolute JHF activity was determined during the first half of the last nymphal instar. This increase is not regulated by JH. Removal of the JHB from hemolymph samples by precipitation with a polyclonal specific antibody increased the JHF activity up to 1,000-fold. Thus, changes in the concentrations of JHB can affect the apparent activity of JHE, which is unrelated to the production or degradation of the JHF.  相似文献

This study examined the effects of strain Beauveria bassiana (LPSC 1067) on nymphal development time, fecundity and adult survival in Dichroplus maculipennis and Ronderosia bergi under laboratory conditions. It was observed that infection with 1×103 conidia/ml altered nymphal development time, fecundity and adult survival in both species. Mortality of D. maculipennis during third through the last instar (sixth) was significantly higher among treated nymphs (66±3.8%) than in controls (15±1.7%). Similarly, mortality in R. bergi during third through the last instar (fifth) was higher in treated nymphs (71±2.8%) than in controls (19±1.5%). Nymphal development times of both infected D. maculipennis and R. bergi were longer than controls. On the other hand, among survivors of both the species, control adults lived longer than infected adults. Finally, control grasshoppers of both species were much more successful reproductively than infected grasshoppers.  相似文献

Summary The singing muscles of the katydid Neoconocephalus robustus develop adult ultrastructure late in the last nymphal instar and during the first few days of adult life. The ultrastructural changes during early adulthood were not affected by unilateral axotomy shortly after the adult molt. Both denervated and innervated muscles developed adult proportions of mitochondria, myofibril, and sarcoplasmic reticulum and transverse tubules.  相似文献

1. This is the first study on the life cycle, growth and production of Sigara selecta, a Palearctic corixid species typical of brackish and saline waters, at the warmest limit of its European distributional range. The study combines field and laboratory approaches. 2. The S. selecta population studied was multivoltine, producing four asynchronous cohorts from early spring to December and overwintering in the adult state. Development time from egg to first adult ranged from 2 to 3 months. A minimum temperature threshold of 10 °C and diel amplitude of ≥10 °C were observed for reproduction and oviposition. 3. Maximum density and biomass were reached in mid spring and early autumn. The sex ratio was unbalanced, females dominating during most of the year, except in spring, when the sex ratio was balanced or dominated by males during the first adult emergence. 4. Laboratory rearing experiments at constant temperatures (18, 22 and 26 °C) pointed to a significant effect of temperature on egg development and nymphal growth. In the range of temperatures tested, both egg and nymphal instar duration decreased with increasing temperature. Mean nymphal development time varied from 43 days at 26 °C to 71 days at 18 °C, with a mean of 57 days. Survivorship was independent of temperature. 5. A reduction in nymphal and adult length was observed with increasing temperature. 6. Growth rates decreased with increasing body mass and increased as temperature increased. The first nymphal instar had the highest length increments and growth rates in all temperature treatments. 7. Satisfactory agreement was found between the field and laboratory degree‐days required for complete development from egg to first adult. At constant and variable thermal regimes, degree‐days decreased with increasing temperature. 8. Rate of growth in the field could be predicted with reasonable accuracy from a simple model obtained as a function of body mass. The model explained 67% of the variability in growth rates. 9. Annual production and production/biomass ratio (P/B) of S. selecta estimated by the Instantaneous Growth method were 1.28 g m?2 year?1 and 13.71, respectively. Spring and autumn cohorts contributed 32% and 54%, respectively, of total annual production. Maximum production corresponded to intermediate temperature periods, although summer production may have been underestimated because of the longer sampling interval relative to cohort interval production. The Size Frequency method underestimated production by at least 18% with respect to the Instantaneous Growth method.  相似文献

Membracids (family: Membracidae), commonly known as treehoppers, are recognizable by their enlarged and often elaborated pronota. Much of the research investigating the development and evolution of this structure has focused on the fifth instar to adult transition, in which the pronotum undergoes the largest transformation as it takes on adult identity. However, little is known about the earlier nymphal stages, the degree to which the pronotum develops at these timepoints, and how development has changed relative to the ancestral state. Here, we studied the nymphal stages and adults of five morphologically distinct membracid species and of Aetalion reticulatum (family: Aetalionidae), the outgroup which was used as an ancestral state proxy. We found that shape differentiation in the pronotum of membracids can start as early as the second instar stage. Most shape differentiation occurs within the nymphal stages and not in the embryo since the shape of the first-instar pronotum did not differ from the outgroup species in all but one species we investigated. We found the anterior–posterior axis of the pronotum elongated at a faster relative rate in membracid species than in A. reticulatum, which contributed to the development of exaggerated pronotal size. Finally, we found differences in the morphogenesis of shape across species. We suggest this is due to the developmental and evolutionary divergence of differential growth patterning of the dorsal surface of the pronotum, not only across species, but also between stages within the same species. This lability may contribute to the evolvability and diversification of the membracid pronotum.  相似文献

The wood‐feeding cockroach Panesthia angustipennis spadica Shiraki (Blaberidae) is a gregarious species, and its groups containing both nymphs and adults have often been observed in the field. To clarify the effects of aggregation with a parent and siblings on nymphal development in P. angustipennis spadica, we raised nymphs of this species in the laboratory under the following three experimental categories: (a) aggregate with a female adult; (b) aggregate without a female adult; and (c) solitary. Survival rates did not significantly differ among the three categories. Unexpectedly, our results clearly showed that nymphs raised in aggregates with a female adult were smaller, lighter, and reached a lower instar than those raised without a female adult. These results indicate that the presence of a female adult does not positively affect nymphal development; i.e. there may be no direct parental care in this species as indicated by previous studies. Moreover, solitary nymphs grew faster and larger than nymphs in aggregates, showing that the aggregation with siblings also has no positive effect on nymphal development. Thus, it seems likely that external factors, e.g. predation and environmental conditions, may drive aggregating behavior in P. angustipennis spadica.  相似文献

The tiny parasitoid wasp, Encarsia formosa, has been used successfully to control greenhouse whiteflies (GHWFs) in greenhouses in many countries throughout the world. Therefore, there has been considerable interest in developing methods for artificially rearing this wasp. However, little information is available concerning the regulation of its development including the host-parasitoid interactions that are required for the parasitoid to complete its life cycle. Here we confirm that parasitoid developmental rates differ significantly based upon the host instar parasitized. Development was faster when 3rd and 4th instar GHWFs were offered for parasitization than when 1st or 2nd instars were used. Our results show that it is primarily the embryo and the first two parasitoid instars that exhibit prolonged developmental times when 1st and 2nd instar whiteflies are parasitized. Although percent emergence was not affected by host age at the time of parasitization, adult longevity as well as adult emergence pattern varied greatly depending upon the instar parasitized. When 3rd and 4th instar GHWFs were selected for oviposition, adult wasps lived significantly longer than when 1st or 2nd instars were used; also, there was a sharp emergence peak on the 2nd day after emergence was first observed (reduced or absent when 1st or 2nd instar GHWFs were parasitized) and the emergence period was reduced from between 8 and 11 days to 5 days. In general, the younger the host instar parasitized, the less synchronous was parasitoid development. Previous reports that E. formosa will not molt to the 2nd instar until the host has reached its 4th instar were not confirmed. When 1st instar host nymphs were parasitized, 2nd instar parasitoids were detected in 3rd instar hosts. Importantly, however, no matter which instar was parasitized, the parasitoid never molted to its last instar until the host had reached Stage 5 of its last instar, a stage in which host pharate adult formation has been initiated. It appears, then, that a condition(s) associated with host pharate adult formation is required for the parasitoid's final larval molt. Results reported here should facilitate the development of in vitro rearing systems for E. formosa.  相似文献

Photoperiodic regulation of nymphal development was examined in four species of cricket collected in the Xinjiang‐Uygur Autonomy Region, China (approximately 43°N, 81–89°E). Fifty percent of individuals of Modicogryllus frontalis reared at 28°C reached adulthood in approximately 80 days in conditions of 11 h light : 13 h dark (LD 11:13) to 14:10, and in approximately 95 days under LD 15:9 to 16:8. Melanogryllus desertus started adult emergence earliest under LD 16:8 at 28°C, but some individuals required much longer to mature, and thus two peaks of adult emergence occurred at approximately 60 and 160 days after hatching. More individuals hatched during the late peak in LD 18:6 than in LD 16:8. The mean nymphal period was approximately 100 days in LD 11:13 to 14:10. Both species showed short‐day type photoperiodic responses, but Mo. frontalis developed faster than did Me. desertus. The latter occupied a wider range of habitat conditions and was more variable in life cycle than the former. Modicogryllus burdigalensis started adult emergence earliest in LD 16:8 at 28°C in the six photoperiods tested, most adults emerging within 60 days. The mean nymphal period was 80 days in LD 15:9, 135 days in LD 14:10 and 80–100 days in LD 11:13 to 13:11, showing an intermediate type of photoperiodic response. Acheta domesticus was a long‐day type species, and the proportion of delayed individuals increased with decreasing photoperiod. In the dry climate of Xinjiang‐Uygur, nymphal overwintering seems to be successful in all of the four different patterns of nymphal development.  相似文献

With the aim of developing better procedures for rearing the microtype tachinid fly Pales pavida (Meigen), we performed ecological studies in the laboratory using the natural host Mythimna separata (Walker), investigating larval development, mating behaviour, individual oviposition patterns and relationships between parasitisation and number of eggs ingested (NEI) per host. The host instar at the time of parasitoid egg ingestion significantly affected the development time of the immature parasitoid: development took longer when the hosts ingested eggs when at the fifth instar than at the sixth (last) instar. There was no difficulty obtaining mated females in the laboratory when day 0–1 female flies were kept with day 2–4 males. Mean lifetime fecundity was 5805?±?568 eggs per female. Daily rates of oviposition by individual females varied widely; the greatest number of eggs laid in a day was exactly 1700. When the NEI by day 1 fifth instars or day 0 or 3 last instars was 1, 3, 6 or 10, the parasitisation percentage tended to increase with increasing NEI, although it did not differ significantly between NEI 6 and 10. Therefore, the percentage adult emergence per egg ingested decreased from NEI 6 to 10, particularly in the case of last instars. Using day 0 last instars, with six eggs ingested per host, should increase parasitisation rates and shorten the development time of the parasitoid for rearing.  相似文献

The suitability of cysts of the brine shrimp Artemia sp. as a factitious food for the predator Macrolophus pygmaeus Rambur was investigated. The influence of decapsulation time and hydration of the cysts on the performance of the predator were studied in the absence of plant material. A longer time of decapsulation had a positive influence on the development of the predator. Hydration of cysts had a significant impact on nymphal survival when cysts where non‐decapsulated or poorly decapsulated. An experiment in which nymphs were switched from a diet of hydrated cysts to non‐hydrated cysts showed that in the absence of plant material the relative importance of hydrating the cysts decreased with nymphal age. Especially, the first instar and to a lesser extent the second instar appear to be susceptible to water shortage. Effects of prolonged rearing on development and reproduction on brine shrimp cysts from different origins were tested in the presence of plant material. Rearing M. pygmaeus on Artemia sp. (Jingyu Lake) cysts yielded similar survival, development, adult weight and fecundity in the fourth as in the second generation. In contrast, for Artemia franciscana cysts, an increase in nymphal development was notable. Biochemical analyses showed that total amino acid content and the concentration of the different amino acids did not differ among diets and generations. There were, however, differences in total fatty acid content between the different diets and generations and in the concentration of certain fatty acids, indicating that insects fed brine shrimp cysts may show nutritional deficiencies compared to those reared on a diet of Ephestia kuehniella eggs. Our results indicate that decapsulated brine shrimp cysts are an economically viable alternative food source in at least part of the rearing process for M. pygmaeus.  相似文献

The effect of the dark-colour-inducing neurohormone (DCIN=[His(7)]-corazonin) of locusts was investigated in nymphs of the grasshopper, Oedipoda miniata, which exhibit strong homochromy, but neither green-brown, nor phase colour polymorphism. Graded doses of synthetic DCIN were injected in 1 μl of olive oil into 0-24-h-old penultimate nymphs. DCIN induced dose-dependent darkening in the recipients, seen 4 days after injection (still in the penultimate nymphal instar), as well as in the subsequent last-instar nymphs. The dose-range obtained between discernible and maximum (almost black) darkening extended over three orders of magnitude, from 1 pmol to 1 nmol of DCIN. In spite of the darkening observed already in the penultimate nymphal instar, the exuviae in the moult from this to the last nymphal instar did not show marked dark patches. However, the exuviae of the next moult (from last-instar nymph to adult) showed such dark patches, which increased with the increase of the dose, revealing the presence of exocuticular melanin. We conclude that DCIN, or a very close peptide, is the endogenous hormone which plays the major or sole role in the control of homochromy of O. miniata and possibly also of other acridids which exhibit homochromy.  相似文献

Nutritional status regulates different processes, such as growth and development, through TOR (target of rapamycin) and insulin receptor signalling pathways. The ribosomal S6 protein kinase (S6K) is a downstream element of both pathways. Using cDNA from the German cockroach Blattella germanica (L.), two S6K isoform sequences (BgS6K) are cloned. The long isoform differs from the short one by the insertion of a 22‐amino acid duplication, involving the key phosphorylation position Thr390. As a result of this, the long isoform presents a new, potentially regulatory phosphorylation site. RNA interference knockdown of both BgS6K isoforms induces an increase in the length of the last nymphal instar, together with a reduction in the mRNA levels of a number of enzymes of the Juvenile Hormone biosynthetic pathway in the corpora allata, vitellogenin mRNA levels in the fat body and basal oocyte length. Thus, BgS6K is important for nymphal development and is necessary for the full induction of Juvenile Hormone synthesis and vitellogenin production in adult females.  相似文献

The yellow protein of the takeout family (YPT) controls the development of yellow body color in the desert locust. This study focused on two aspects related to YPT in the locust. We first examined the expression pattern of YPT during nymphal stages because yellowing was not obvious during the early instars. YPT expression levels were extremely low in the second and third instars compared with the last two nymphal instars. Warm rearing temperature and juvenile hormone (JH) injection, which stimulated YPT expression in the late instars, had little effect in the second instar, suggesting that YPT expression during the early instars was suppressed and could not be stimulated by either of these factors. We also investigated delayed yellowing in fasting male adults, under the hypothesis that fasting decreased the JH titers and delayed the onset of YPT expression. Yellowing was delayed in fasting adults compared with well‐fed adults and YPT expression was stimulated by JH injections at Day 15. However, we failed to obtain evidence that fasting significantly influenced the expression levels of YPT and the JH early‐inducible gene Krüppel homolog 1 at Days 15 and 20 post‐adult emergence. Results suggest that a YPT‐independent mechanism possibly induces delayed yellowing in fasting males.  相似文献

The planthopper Delphacodes kuscheli is the main vector of Mal de Río Cuarto virus in Argentina, disease that severely affects maize production. In this study, we investigated the effects of heat stress on fitness traits and on the number of its obligate yeast‐like symbionts (YLS). The exposition of newly‐hatched nymphs to 35°C for 3 days, a well‐known procedure used to reduce the number of YLS in planthoppers, was applied. To compare different fitness components between control and heat‐treated insects, we estimated nymphal instars development time, nymphal survival, adult body length, longevity, fecundity and fertility. Also, correlates of fitness, as proportion of sexes and wing forms of the emerging adults, were evaluated. In heat‐treated group, the nymphal developmental time increased due to an increase in the fifth instar duration, and the nymphal survival, body length of adults and fecundity were reduced when compared to control. There was a significant association between treatments (control and heat‐treated insects) and wing morphs. The heat treatment successfully reduced the number of YLS in third instar nymphs of D. kuscheli. Our results revealed the negative effect of heat stress on development, survival and reproduction of D. kuscheli and on the load of its YLS endosymbionts suggesting that YLS could play a crucial role in the development and reproduction of these planthoppers.  相似文献

The rate of oxygen consumption was investigated in fed larval, nymphal and adult Ornithodoros turicata ticks and in starved nymphal and adult ticks. Oxygen consumption rate of fed adult ticks increased with increasing temperature. The metabolic rate of adult ticks was affected by starvation whereby starved adult ticks showed a significantly lower oxygen consumption than their fed counterparts. The oxygen consumption rate of fed female ticks was significantly higher than that of fed males but, there was no significant difference between the oxygen consumption rates of starved female versus starved male ticks. Oxygen consumption of fed larvae was significantly greater than those of fed first through third instar nymphs. Fed and starved nymphal ticks as well as fed adult ticks ventilated continuously. In contrast, starved adults ventilated discontinuously. The ability to reduce metabolic rate, plus the capability to ventilate discontinuously allow O. turicata adults to cope with prolonged starvation.  相似文献

The firing behaviour of an identified neurone in the cricket was studied using extracellular recording from the axon. In the last nympal instar (preadult developmental stage), the contralateral dorsal longitudinal motor neurone (CDLM) showed spontaneous activity and was excited by air puffs to the head and cerci and by single shocks to the anterior nerve cord. In the normal adult the CDLM did not exhibit these properties. However, responses which were characteristic of the last instar appeared in the adults which had been subjected to any one of the following surgical procedures: (1) central nervous system injuries which separated the CDLM arborization and axon from the soma; (2) operations which injured the central nervous system without cutting the CDLM; and (3) operations which damaged the cuticle only. Since cuticle damage alone was as effective as the more extensive operations, it is suggested that a sufficient cause for the appearance of nymphal firing behaviour in the adult CDLM is cuticle damage. The factor associated with cuticle damage which mediates the changes in activity of the CDLM neurone is not known, but its action does not require the mediation of the CDLM soma.  相似文献
