The phase-response curves (PRC) for light pulses in continuous darkness (DD) have been described in many mammals, especially in nocturnal rodents. The PRC for dark pulses in continuous light (LL), however, has been described in a few mammals only, in nocturnal for bat and for hamster and in diurnal for Octodon degus, suggesting that this PRC is mirror imaging the PRC for light pulses. Therefore, the effect of 1-h and 3-h lasting dark pulses on the circadian wheel-running activity rhythm of mice in continuous light was investigated and then the PRC for dark pulses in LL was drawn up. For comparison, the effect of 1-h lasting light pulses on the circadian wheel-running activity rhythm of mice in DD was examined and the PRC for light pulses in DD was constructed. It appeared that the PRC for dark pulses, to a certain degree, represents a mirror image of the PRC for light pulses in mice. However, the advance region of this PRC is longer than that of delay. The mechanism of dark pulses action is discussed.  相似文献

EARLIER studies of the location of the single cysteine residue and the two disulphide bridges in bovine β-lactoglobulins A and B1, for each of which the monomer is a single chain of 162 residues and 18,000 molecular weight2,3, led to the conclusion that the sulphydryl group is at position 69 and that the disulphides bridge positions 123 to 160 and 57 to 70. These results were based on diagonal peptide studies4 and on the composition of peptides in which the sulphydryl group had been labelled with 14C-iodoacetamide, the disulphide bridges being left intact. Use was made of the partial amino-acid sequence given by Frank and Braunitzer5 and the reasonable assumption was made that the sulphydryl occurred in only one position. Subsequently, Shaw6 has shown that the sequence of Frank and Braunitzer5 showing Cys residues adjacent at positions 69 and 70 is incorrect and that they are separated by a glutamine, the sequence for positions 67 to 71 for the Bvariant being Ala.Cys.Gln.Cys.Leu. Autoradiography of the dansyl amino-acid derivatives formed during the sequence determination of this pentapeptide indicated that both residues 68 and 70 seemed to have been labelled and so we have given further consideration to the sulphydryl location. It has been found that although it does occur at 68, with 57 and 70 disulphide bridged, there is also an equal amount of protein present with the sulphydryl at 70, with 57 and 68 disulphide bridged. We discuss this additional finding here and the significance for the determination of the location of sulphydryl groups in other proteins.  相似文献

Mushrooms for Life among the Jotï in the Venezuelan Guayana. This paper explores the relevance of mushrooms in mythology and daily life among the Jotï, an indigenous group inhabiting the rain forests of the Venezuelan Guayana. It is reported here how a personal experience of the author in the field stimulated deeper research into the religious symbolism surrounding mushrooms and their role in Jotï cosmology, ritual, and life dynamics. Different categories of mushroom usage are described, including edible, medicinal, and ritual uses. Mushrooms seem to be more important to the Jotï than to most Amazonian groups. Most significantly, certain species of mushrooms are key ingredients in medicinal and ritual preparations used throughout the Jotï life cycle in ways that reflect their worldview and mythology.  相似文献

Many glycine peptides support growth of a glycine auxotroph of Escherichia coli. If the alpha-amino group of these peptides is methylated, the products are still utilized for growth, and also retain comparable ability with the unsubstituted peptides to compete with natural peptides for transport into the cell. In contrast, glycine peptides devoid of an alpha-amino group, or that have the alpha-amino group substituted by one of a number of acyl groups are not utilized, although E. coli possesses intracellular enzymic activity able to release glycine from such compounds; further, these derivatives do not compete with natural peptides for transport into the cell.  相似文献

Measurement of the content of polyamines in pancreatic islets indicated that no significant change in their concentration took place during glucose-stimulated insulin release. The finding, together with the absence of any effect of -difluoromethylornithine on glucosestimulated insulin release suggested that rapid synthesis of polyamines is not involved in stimulus-secretion coupling in the -cell. The concentration of polyamines found in islets were high enough for them to act as substrates for the Ca2+-dependent islet transglutaminase during insulin release. This was further demonstrated by the ability of islet transglutaminase to incorporate [14C]putrescine into proteins from islet homogenates and by the demonstration of an increase in the covalent incorporation of [14C]putrescine into the proteins of intact islets following their challenge with glucose. Unlike monoamine substrates of transglutaminase, putrescine failed to effectively inhibit insulin release when its intracellular concentration was increased. A role for polyamines in the secretory process through their incorporation into islet proteins is suggested.  相似文献

Background: We hypothesized that due to the absence of a dietary source of omega-3 fatty acids, the essential fatty acid (EFA) deficiency model leads to an overestimate of linoleic acid (LA) requirements. Methods: over 7 wk, young rats consumed an EFA diet containing either 0 en% linoleate (0LA) and 0 en% α-linolenate (0LNA) or a diet containing 0.5 en% LNA plus one of seven levels of added LA (0.12–4.0 en%; n=6/group).Results: Rats consuming the 0LA–0LNA diet had the lowest final body weight, 34–68% lower LA and arachidonate in plasma and liver, 87% lower LA in epididymal fat, and an 8–20 fold higher eicosatrienoate in plasma, liver and muscle lipids. 0.5LNA completely prevented the lower growth and partly prevented the rise in eicosatrienoate seen in the 0LA–0LNA group.Conclusion: Providing dietary LNA at 0.5 en% reduces the rat's physiological requirement for LA by an estimated factor of at least four (0.5 en% instead of 2 en%). Since LA requirements in humans are also based on the same flawed model of EFA deficiency, it is plausible that they too have been overestimated and should therefore be reinvestigated.  相似文献

In this paper we examine the bias towards weekend recording (the weekend effect) in volunteer phenology, using over 14,000 bird migration phenological observations from eight locations in the UK as a data source. Data from 45 bird species were used. Overall, 44% of all records were taken at weekends in contrast to the 28.6% (i.e. two out of seven days) that would be expected if records were evenly spread throughout the week. Whilst there is documented evidence of environmental differences at weekends, particularly in large urban areas, we believe the weekend effect is mostly a consequence of greater recorder effort at weekends. Some birds, likely to be obvious by their behaviour or abundance, had fewer weekend records than the remaining species. The weekend effect, to some extent, differed between locations and between seasons. There was some evidence that, particularly in autumn, the weekend bias may be lessening. If so, this will increase the accuracy of phenological records, making the detection of changes and responses to temperature easier.  相似文献

1. The ability of a range of phenothiazines to inhibit activation of brain phosphodiesterase by purified calmodulin was studied. Trifluoperazine, prochlorperazine and 8-hydroxyprochlorperazine produced equipotent dose-dependent inhibition with half-maximum inhibition at 12mum. When tested at 10 or 50mum, 7-hydroxyprochlorperazine was a similarly potent inhibitor. However, trifluoperazine-5-oxide and N-methyl-2-(trifluoromethyl)phenothiazine were ineffective at concentrations up to 50mum, and produced only a modest inhibition at 100mum. 2. The same phenothiazines were tested for their ability to inhibit activation of brain phosphodiesterase by boiled extracts of rat islets of Langerhans. At a concentration of 20mum, 70-80% inhibition was observed with trifluoperazine, prochlorperazine, 7-hydroxyprochlorperazine or 8-hydroxyprochlorperazine, whereas trifluoperazine-5-oxide and N-methyl-2-(trifluoromethyl)phenothiazine were less effective. 3. The effect of these phenothiazines on insulin release from pancreatic islets was studied in batch-type incubations. Insulin release stimulated by glucose (20mm) was markedly inhibited by 10mum-trifluoperazine or -prochlorperazine and further inhibited at a concentration of 20mum. 8-Hydroxyprochlorperazine (20mum) was also a potent inhibitor but 7-hydroxyprochlorperazine (20mum) elicited only a modest inhibition of glucose-stimulated insulin release; no inhibition was observed with trifluoperazine-5-oxide or N-methyl-2-(trifluoromethyl)phenothiazine. 4. Trifluoperazine (20mum) markedly inhibited insulin release stimulated by leucine or 4-methyl-2-oxopentanoate in the absence of glucose, and both trifluoperazine and prochlorperazine (20mum) decreased insulin release stimulated by glibenclamide in the presence of 3.3mm-glucose. 5. None of the phenothiazines affected basal insulin release in the presence of 2mm-glucose. 6. Trifluoperazine (20mum) did not inhibit islet glucose utilization nor the incorporation of [(3)H]leucine into (pro)insulin or total islet protein. 7. Islet extracts catalysed the incorporation of (32)P from [gamma-(32)P]ATP into endogenous protein substrates. Sodium dodecyl sulphate/polyacrylamide-gel electrophoresis resolved several phosphorylated bands, but incorporation was slight. However, calmodulin in the presence of Ca(2+) greatly enhanced incorporation: the predominant phosphorylated band had an estimated mol.wt. of 55000. This enhanced incorporation was abolished by trifluoperazine, but not by cyclic AMP-dependent protein kinase inhibitor protein. 8. These results suggest that islet phosphodiesterase-stimulating activity is similar to, although not necessarily identical with, calmodulin from skeletal muscle; that islet calmodulin may play an important role in Ca(2+)-dependent stimulus-secretion coupling in the beta-cell; and that calmodulin may exert part at least of its effect on secretion via phosphorylation of endogenous islet proteins.  相似文献

Pollen and anthers of the F1 hybrid with the restored fertility and the parental forms, a maternal male-sterile line harboring the mitochondrial orfH522 gene and a paternal male-fertile line harboring dominant alleles of the nuclear Rf gene, were studied. In order to find protein markers for the genetic CMS–Rf system in sunflower, we analyzed cytoplasmic proteins by disc electrophoresis under denaturing conditions. As a result, a protein fraction with a mol wt of 43 kD specific for cytoplasmic proteins of the paternal anthers, and a protein fraction with a mol wt of 16 kD specific for cytoplasmic proteins of the maternal anthers, were detected. The electrophoretic patterns of water-soluble proteins from the hybrid and paternal lines did not differ.  相似文献

1. The relationship between coarse particulate organic matter (CPOM) standing stock and benthic macroinvertebrate assemblages in Kenyan highland streams was determined by sampling seven sites on three rivers (2000–2700 m a.s.l.). Taxa recorded were allocated to functional feeding groups using published literature, mouthpart analysis and examination of gut contents. Patterns were compared with five structurally similar streams in three areas of Europe (south-west France, south-east England, north-east England).
2. Number of individuals and proportion of detritivores in Kenyan streams were equivalent to, or greater than, those in European sites. Shredders were, however, almost completely absent from Kenyan sites, despite high standing stocks of CPOM. Shredders were abundant in all European sites.
3. The phenomenon of low shredder abundance has been observed in other tropical streams in south-east Asia and Central and South America but, in contrast to these regions, the African rivers studied were devoid of shrimps or fish which may occupy the shredding niche elsewhere.
4. These preliminary data suggest that shredder-mediated detritus processing, which is a key functional component of streams in the North Temperate Zone, does not operate in East African streams. There are three possible reasons for this. The first is that tropical African rivers are functionally different to those in temperate regions. This could be because of enhanced microbial activity replacing shredder activity at high temperatures. Alternatively, it could be a result of low palatability of detrital inputs from dominant riparian trees in the region. The second and third are methodological: that our allocation to functional feeding groups is incorrect, and that our sampling methods missed a potentially key shredding taxon – the freshwater crab Potamonautes sp.  相似文献

This article focuses on the Vision Documents of the Arab civil society organizations in Israel as an act of citizenship and an expression of the politics of contention used by the Palestinians in Israel. We argue that these documents challenge both the political inclusiveness of the identity of Israel as a ‘Jewish and democratic’ state, and the political continuity of collective identity of the Palestinian people. With these documents, the Arab civil society organizations reclaim responsibility over their political future by clinging to the Israeli citizenship framework, but at the same time attempt to change its nature from within, by re-associating the Palestinians in Israel with the core issues of the stumbling peace process, especially in regard to the ‘Right of Return’. The paper contends that for the Palestinians in Israel, the national and the civic frameworks do not circulate in separate orbits, but constitute and reframe each other.  相似文献
