为了揭示臭氧(O3)浓度升高对植物活性氧代谢系统的影响机理,从代谢生理角度,总结了近年来国内外关于臭氧浓度升高对植物活性氧自由基代谢速率、细胞膜脂过氧化程度、抗氧化系统及生物量和产量影响的研究进展,同时,就臭氧浓度升高与二氧化碳浓度升高的复合作用对植物活性氧代谢系统的影响,及阐明二者相互作用对植物抗氧化系统影响机理的研究进行了综述。在此基础上指出在未来研究中,要在分子水平上进一步深入研究植物活性氧代谢系统对高浓度臭氧、二氧化碳复合作用的响应机理,并应加强高浓度二氧化碳对臭氧胁迫下植物抗氧化系统影响的研究,为解决如何减轻臭氧浓度升高对植物造成的氧化伤害提供基础理论依据。  相似文献

Epidemiological and experimental evidence indicated that hyperhomocysteinemia is associated with neurodegeneration. However, homocysteine neurotoxic effects have been so far investigated mostly by employing homocysteine concentrations (≥100 µM) much higher than homocysteine mean plasma levels (20 µM) observed in patients with neurodegenerative disorders. While evaluating the effects of a prolonged exposure to ~20 µM homocysteine in neuronal-like differentiated SH-SY5Y cells, we observed a 35 % loss of cell viability and a four-fold increase in reactive oxygen species levels in cells incubated with homocysteine for five days compared with controls. Moreover, homocysteine increased by 30 % and around two-fold, respectively, the Comet-positive cell number and DNA damage indexes (tail length, T-DNA, olive tail moment) compared with controls. Cell response to homocysteine-induced DNA damage involved the up-regulation of Bax and, at a greater extent, Bcl-2, but not caspase-3, in association with a p53-independent increase of p21 levels; concomitantly, also p16 levels were increased. When looking at time-dependent changes in cyclin expression, we found that a significant up-regulation of cyclins D1, A1, E1, but not B1, concomitant with p21 down-regulation, occurred in cells incubated with homocysteine for three days. However, in line with the observed increase of p21 and p16 levels, a five days incubation with homocysteine induced cyclin down-regulation accompanied by a strong reduction of phosphorylated pRB amounts. These results suggest that, when prolonged, the exposure of neuronal-like cells to mildly elevated homocysteine concentrations triggers oxidative and genotoxic stress involving an early induction of cyclins, that is late repressed by G1-S check-point regulators.  相似文献

SUMMARY. At any given temperature level, the rate of oxygen poisoning increases proportionally with an increase in oxygen tension. But the toxicity of oxygen does not bear a proportional relationship to temperature. Using the examples of cadmium and lead, the review considers the various toxic effects exerted by these heavy metals. Putative specific and nonspecific mechanisms of the toxic effects of the heavy metals and plant responses are discussed together with the issue of Cd and Pb accumulation in various plant organelles, cells, tissues, and organs. The basic mechanisms providing for plant resistance to excess Cd and Pb are elucidated. These data are used to schematically outline the changes in plant metabolism produced by these heavy metals.  相似文献

Pseudomonas sp. strain ST-200 isolated from a humus soil effectively oxidizes cholesterol dissolved in organic solvents but not that suspended in the growth medium. The organism does not assimilate cholesterol. This organism oxidized a variety of 5α- or 5-ene-sterols dissolved in organic solvent. First, the 3β-OH group was oxidized to a ketone group. The 3α-OH group was scarcely oxidized. Successively, C-6 position of 5-ene-steroids was hydroxylated, and a double bond of 5-ene-steroids was transferred from Δ5 to Δ 4. Then, the 6-OH group was oxidized to a ketone group. Persolvent fermentation with ST-200 would provide an effective, convenient, and stereospecific method to oxidize the C-3 and C-6 positions of steroids.  相似文献

Adverse climate change attributed to elevated atmospheric carbon dioxide concentration (CO2) and increased temperature components of global warming has been a central issue affecting economic and social development. Climate change, particularly global warming, imposes a severe impact on the terrestrial ecosystem. Elevated CO2, drought, and high temperature have been extensively documented individually; however, relatively little is known about how plants respond to the interaction of these factors. To summarize current knowledge on the response of plants to global change factors, we focus on the interactive effects of CO2 enrichment, warming, and drought on plant growth, carbon allocation, and photosynthesis. Stimulation due to elevated CO2 might be suppressed under other negative climatic/environmental stresses such as drought, high temperature, and their combination. However, elevated CO2 could alleviate deleterious effects of moderate drought via reducing stomatal conductance, altering leaf surface, and regulating gene expression. High CO2 levels and rising temperatures may result in opposite responses in plant water use efficiency. Stimulation of plant growth due to elevated CO2 for C3 species occurs regardless of water conditions, but only under a water deficit for C4 species. The positive effect of elevated CO2 on C4 species is derived mainly from the improved water status. Plant adaptive or maladaptive responses to multivariate environments are interactive; thus, researchers need to explore the ecological underpinnings involved in such responses to the multiple factors involved in climate change.  相似文献

通过固定不同溶氧浓度(DOT)对短小芽孢杆菌(Bacillus pumlus)进行分批发酵的过程参数变化的比较,发现发酵前期与后期对氧的需求不尽相同,探讨了氧代谢途径及溶氧浓度对核糖发酵的影响机理,并提出分阶段供氧模式。结果表明,发酵时间44h后,整个发酵过程保持了较高的核糖产率和葡萄糖消耗率,最终核糖产量和细胞生成量分别提高了5.0%和18.8%。  相似文献

The aim of this work was to study the proliferation pathological perturbations of cultured chondrocytes in response to menadione, an oxygen free radicals producing drug. Rabbit articular chondrocytes in monolayer culture were treated with 10-5, 1.5.M-5 and 2.10-5M of menadione during three days. A dose dependent decrease of the proliferative capacity was observed. Flow cytometry analysis revealed a perturbation of the cell cycle progression consisting in an accumulation of cells in the S and G2 + M phases. This growth perturbation was due to oxygen radicals production since a treatment with catalase suppressed these toxic effects. Furthermore, to identify oxygen derived radicals in the cellular suspension of cultures treated with menadione, we used a technique of spin-trapping coupled with electron spin resonance (ESR). The ESR signal corresponding to the DMPO hydroxyl radical adduct (DMPO-OH) has been detected. The spectra observation indicated the actual production of hydroxyl radical. However, superoxide anions have not been identified; this fact can be explained by the low reactivity of these anions with DMPO and by the decomposition of signal DMPO-OOH to DMPO-OH.  相似文献

城市绿化植物是城市生态系统的重要组成部分,在美化城市、保持其生态平衡及维持其可持续发展等方面作用不可低估。本研究以贵州省安顺市城区为研究实例,在对绿化植物种类和数量进行调查的基础上,对香樟等16种典型植物的释氧和降温的生态效应进行分析。结果表明:贵州安顺城区内绿化植物种类较少,结构单一,物种均匀度低;不同植物在不同月份的释氧能力虽有差异,但都表现出6月和8月上升,7月和9月下降的规律;8月份各种植物的降温效果与释氧规律相一致,乔木以法国梧桐降温0.32℃最明显,灌木以小叶女贞降温0.26℃最明显,攀援植物以爬山虎降温最强,达0.46℃。本研究可以为同类型城市的生态建设、改善人居环境以及生态可持续发展提供基础性的研究参考。  相似文献

Continuous cultures of Saccharomyces cerevisiae grown over a range of oxygen and glucose concentrations were used to determine the effects of these two physiological regulators on mitochondrial protein synthesis in vivo. Quantitative estimates were obtained of the contribution of the mitochondrial protein-synthesizing system to the formation of mitochondrial membranes in cells grown over a wide range of dissolved oxygen concentrations, under conditions of glucose limitation or glucose excess in the cultures. The nature of the products of the mitochondrial system formed under these conditions was examined by selectively labeling membrane proteins in vivo in the presence of cycloheximide, and fractionating the products by gel electrophoresis under dissociating conditions. These results have been correlated with changes in the lipid composition of the cells and with the synthesis and assembly of components of the mitochondrial adenosine 5'-triphosphatase complex.  相似文献
