Abstract. The effect of short-term SO2 fumigation on photosynthesis and transpiration of Vicia faba L. was measured at different irradiances and SO2 concentrations. At high irradiances photosynthetic rates were reduced when leaves were exposed to SO2 and the magnitude of the reduction was linearly related to the rate of SO2 uptake through the stomata. Photosynthetic rates stabilized within 2 h after the start of fumigation.
The effect of SO2 on photosynthesis was measured at different CO2 concentrations to analyse the contribution of stomatal and non-stomatal factors to photosynthetic inhibition. Mesophyll resistance to CO2 diffusion increased as a result of SO2 exposure and caused a rapid reduction in photosynthesis after the start of fumigation. Stomatal resistance was not affected directly by SO2 fumigation, but indirectly as a result of a feedback loop between net photosynthesis and internal CO2 concentration.
Analysis of gas-exchange measurements in biochemical terms indicated that photosynthetic inhibition during SO2 exposure can be explained by a stronger reduction in the affinity of RBP carboxylase/oxygenase for CO2 than for O2.  相似文献

The dose- and time-response effects of 3 days of 6 h day-time sequential exposures to NO2, SO2 and SO2+NO2 of 0.45–1.81 μl l−1 (ppm) SO2 and 1.50–7.65 μl l−1 NO2 on photosynthesis, transpiration and dark respiration were examined for nine Carpatho-Ukrainian half-sib families and a population from the GFR ('Westerhof') of Norway spruce [ Piecea abies (L.) Karst.], all in their 5th growing season.
SO2+NO2 inhibited photosynthesis and transpiration and stimulated dark respiration more than SO2 alone. SO2 and SO2+NO2 at the lowest concentrations inhibited night transpiration, but increased it at the highest concentration, the strongest effects being obtained with combined exposures. Photosynthesis of the different half-sib families was affected significantly differently by SO2+NO2 exposures. NO2 alone had no effects.
Sensitivity to transpiration decline correlated negatively with branch density. Height of trees correlated postitively with decline sensitivity in the seed orchard. The distribution of photosynthesis and transpiration sensitivities over all tested half-sib families correlated negatively with the distribution of decline sensitivity of their parents in a rural Danish seed orchard. The relative photosynthesis and transpiration sensitivities may thus serve as diagnostic parameters for selecting against novel spruce decline.  相似文献

Picea sitchensis (Bong.) Carr. seedlings were exposed to SO2, NO2 and SO2+ NO2 during dormancy in controlled environments, and were taken to night temperatures of 4, 0, −5, −10 and −15 °C in a freezer. Conditions in the freezer were carefully monitored during the low–temperature treatments. In two experiments, different photoenvironments and temperature regimes were imposed prior to the cold treatments, and different effects were observed. In the first, only limited frost hardiness was achieved and night temperatures of −15 °C were lethal. Temperatures of −5 and − 10 °C led to poor survival of lateral buds, particularly in plants exposed to 45 ppb SO2. The poor bud break in plants exposed to SO2 and to − 5 °C resulted in a loss of the effectiveness of this temperature as a chill requirement. Pressure-volume analysis showed that the shoots of plants exposed to NO2 had greater elasticity (lower elastic moduli, e), so that loss of turgor occurred at lower relative water contents. In contrast, a hardening period (2 weeks in night/day temperatures of 3/10 °C and 8 h days at 50 μmol m−2 s−1 PAR) gave decreased elasticity and lower solute potentials of spruce shoots. In the second experiment, exposure to 30 ppb SO2 and SO2+ NO2 led to slight, but consistent, increases in frost injury to the needles of plants frozen to − 5 and − 10 °C. The results suggest that the main interaction of low temperatures and winter pollutants may be on bud survival rather than on needle damage, but that effects are subtle, only occurring with certain combinations of pollutant dose and cold treatment.  相似文献

The effect of SO2 on adenosine 5'-phosphosulfate sulfotransferase activity and various other parameters of needles from spruce ( Picea abies L.) was studied using potted grafts in outdoor fumigation chambers and trees growing near a factory. In summer and autumn fumigation of grafted spruce, SO2, decreased the extractable activity of adenosine 5'-phosphosulfate sulfotransferase to 12–50% of the controls, and reduced the amount of 35S from sulphate incorporated into protein by excised branches to a comparable degree. SO2 treatment in January and February inhibited the increase in adenosine 5'phosphosulfate sulfotransferase activity measured in the controls during this time. ATP-sulfurylase activity was less affected by SO2. fumigation. In trees growing near a factory with high SO2. emission, the activity of adenosine 5'-phosphosulfate sulfotransferase was about 35% of that of trees from a control area. The low enzyme activity was correlated with a high content of sulfate and compounds containing thiol groups.  相似文献

Five-year-old Picea abies L. plants were grown in growth cabinets in the presence (3.1 μmol m−3) or absence of SO2. After 5 weeks, the photosynthetic capacity of mature needles produced in the year was the same in both conditions. Trees were then submitted in situ to drought stress by withholding water. The decline of leaf photosynthetic capacity was greatest in the presence of SO2. Chlorophyll decreased only when trees were submitted to dehydration in the presence of SO2; however, this al-one could not account for the large decline in photosynthetic capacity observed under that condition. Needle water content was the lowest during dehydration in the presence of SO2. It is concluded that the critical factor in the interaction between pollution by SO2 and drought stress is the greater dehydration of the tissue found in stressed plants grown in the presence of SO2. The large decline in photosynthetic capacity under such conditions might be due to this greater dehydration.  相似文献

The effect of temperature and humidity on SO2–induced photosynthetic depression was determined in gas exchange experiments with leaves of Vicia faba , L. Stomatal behaviour was sensitive to humidity resulting in higher uptake rates of SO2 and stronger reductions of photosynthesis at low VPD (vapour pressure deficit). After a fumigation period of 2 h, when the photosynthetic rate had stabilized, photosynthesis of leaves exposed to SO2 at 8°C was reduced much more than at 18°C at the same rate of SO2 uptake. Data analysis with a mechanistic model revealed that this effect was due to the slower rate of S(IV) oxidation at lower temperatures, resulting in higher accumulation of S(IV) and thus stronger reduction of photosynthesis. These results were confirmed by experimental analyses of the S(IV) concentration in leaves following fumigation, which showed that more S(IV) accumulated in leaves exposed at a lower temperature. This may explain the high sensitivity of plants exposed to SO2 under winter conditions, when both VPD and temperature are low.  相似文献

The effect of SO2 on the extractable activity of ATP sulfurylase (EC 2.7.7.4.). adenosine 5'-phosphosulfate sulfotransferase, ribulosebisphosphate carboxylase, chlorophyll, protein, sulfate, and amino acids was examined in leaves of potted grafts of beech ( Fagus sylvatica L.) treated in outdoor fumigation chambers. Addition of 0.025 and 0.075 μl SO2 1−1 to unfiltered ambient air caused a decrease in the extractable activity of adenosine 5'-phosphosulfate sulfotransferase to about 20 to 30% of the controls. Neither the extractable activity of ATP sulfurylase and ribulosebisphosphate carboxylase nor the content in chlorophyll, total amino acids and protein were significantly affected by SO2, but there was an increase in the sulfate content. Leaves treated with 0.075 μl SO2 1−1 contained more alanine and cysteine and less serine than the controls. After transfer of the SO2-treated beech trees to control chambers there was an increase in adenosine 5'-phosphosulfate sulfotransferase activity, but no significant decrease in SO2−4-sulfur.  相似文献

Abstract. Red spruce ( Picea rubens Sarg.) seedlings were overwintered in two controlled environment chambers designed to simulate sub-zero winter conditions. One of the chambers was fumigated throughout the 5-month period with low concentrations of SO2+ NO2. Extracts of extracellular fluid from trees in this treatment revealed accumulations of sulphite and nitrite, but not of sulphate or nitrate. Analysis of the chloroplast membrane lipid monogalactosyl diglyceride (MGDG) indicated a large increase in fatty acid saturation in both treatments during mid-winter, with a subsequent recovery to earlier levels. Although at the end of the experiment the MGDG of polluted trees contained significantly less linolenic acid, there was no overall treatment effect on fatty, acid content. In the following spring, there was some indication that flushing of leader buds began earlier and proceeded at a slower rate in polluted trees compared to controls, but no other growth parameters were affected by the winter treatment. The absorption of SO2 and NO2 and the accumulation of their products during dormancy is discussed as a potential mechanism for metabolic disruption, resulting in changes to seasonal responses which could be critical to the survival of the plant.  相似文献

Long term fumigation of 4-year-old spruce trees with ozone concentrations up to 200 nl l−1 has only minor effects on the photosynthetic activities measured as chlorophyll a fluorescence. Nevertheless, it drastically changes the turnover of the D-1 reaction center polypeptide of photosystem II. During summer, fumigation with ozone for 2 weeks resulted in an almost 4-fold stimulation of the light dependent incorporation of [14C] leucine into the D-1 protein in the exposed trees. The amount of immunodetectable D-1 protein remained constant when based on chlorophyll. This indicates that exposure to ozone stimulates both the synthesis and the degradation of the D-1 protein. When spruce trees were exposed during winter for 4 weeks to 100 and 200 nl l−1 ozone, respectively, an almost 3-fold increase of the amount of immunodetectable D-1 protein per chlorophyll in the exposed trees was observed. This can be explained by a varying stimulation of D-1 protein synthesis and degradation depending on the different physiological conditions. Since so far the D-1 protein has been found only as a component of photosystem II reaction centers, one has to assume that the relative content of photosystem II reaction centers also increases under certain stress conditions. The increased turnover of the D-1 protein in trees exposed to ozone explains the synergistic effects of stress conditions and high light intensities often observed in the field.  相似文献

The plant fraction of alfalfa ( Medicago sativa L. cv. Aragon) nodules contained both nitrate reductase (NR) and nitrite reductase (NiR). Specific activity of NADH-NR from the cytosol of nodules not treated with NO3- was about 30 nmol (mg protein)-1-h-1 and was not basically affected by NO3 addition. In contrast, typical specific activity for cytosolic NiR was 1.5 umol (mg protein)-1h-1 using methyl viologen as electron donor. This activity strongly increased with NO3 concentration, probably due to substrate induction. Maximal activity was 3.5 μmol (mg protein)-1h-1 at 50 to 200 mM NO3.
Estimates indicate that the contribution of cytosol to the overall NR and NiR activities of alfalfa nodules is distinctly different: less than 10% and about 70%, respectively. The increasing amounts of NO2 accumulating in the cytosol upon NO3, supply, and the different response to NO3 of bacteroid and cytosolic NRs support the concept that most of this NO2 comes from the bacteroids.  相似文献

Fumigation of spinach (Spinacia oleracea L. cvs Estivato and Monosa) with H2S or SO, for 1 to 6 days resulted in accumulation of sulfhydryl (SH) compounds in the shoots of both H2S- and SO2-exposed plants. The sulfate concentration in shoots of SO2-exposed plants increased linearly with time. SH accumulation showed saturation kinetics as a function of time as well as H2S concentration, ascribed to the internal H2S concentration in the plant and the availability of substrates for glutathione synthesis, respectively. SH compounds accumulated more at lower exposure temperatures, whereas sulfate accumulation was more pronounced at higher temperatures. These results are discussed in relation to the possible foliar uptake of H2S and SO2, the temperature dependence of uptake and the water solubility of these gases. The possibility of SO2-induced H2S emission rather than sulfate accumulation as a source for SH accumulation is also discussed. Cessation of fumigation resulted in a decrease in SH compounds and sulfate content that could be accounted for by sulfur metabolism and growth, respectively.  相似文献

The induction of activity of the enzyme nitrate reductase (NR, EC 1.6.6.1, 1.6.6.2) in needles of Norway spruce ( Picea abies [L.] Karst.) by nitrogen dioxide (NO2) was studied under laboratory and field conditions. In fumigation chambers an increase in nitrate reductase activity (NRA) was detected 4 h after the start of the NO2 treatment. During the first 2 days with 100 µg NO2 m−3, NRA reached a constant level and did not change during the following 4 days. At the same level of NO2, NRA was lower in needles from trees grown on NPK‐fertilized soil than on non‐fertilized soil. After the transfer of spruce trees from fertilized soil to NPK‐rich nutrient solution, NRA was transiently increased. This effect was assigned to root injuries causing nitrate transport to the shoot and subsequent induction of NRA. Neither trees on fertilized soil nor trees transferred to NPK‐poor nutrient solution had increased NRA unless NO2 was provided. The NO2 gradient in the vicinity of a highway was used to test the long‐term effect of elevated levels of NO2 on needle NRA of potted and field‐grown spruce trees. Compared with less polluted sites, permanently increased NRAs were detected when NO2 concentrations were above 20 µg m−3. Controls of field measurements some 10 years after the introduction of catalytic converters in cars showed no significant change neither in NO2 levels nor in the decreasing NRA of spruce needles with the distance from the highway.  相似文献

Dactylis glomerata was grown hydroponically in a controlled environment at ambient (360 μl l−1) or elevated (680 μl l−1) CO2 and four concentrations of nitrogen (0.15, 0.6, 1.5 and 6.0 m M NO3), to test the hypothesis that reduction of photosynthetic capacity at elevated [CO2] is dependent on N availability and mediated by a build-up of non-structural carbohydrates. Photosynthetic capacity of the youngest fully expanded leaf (leaf 5, 2 days after full expansion) was reduced in CO2-enriched plants at low, but not high N supply and so the stimulation of net photosynthesis by CO2 enhancement was less at low than at high N supply. CO2 enrichment resulted in a decrease in ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) content on a leaf area basis at 0.6 and 1.5 m M NO3, but not at 0.15 and 6.0 m M NO3, and had no effect on the total N content of the leaf on an area basis. However, decreases in Rubisco content could be primarily accounted for by a decrease in total N content of leaves, independent of [CO2]. A doubling of the Rubisco content by increasing the N supply beyond 0.6 m M had only a marginal effect on the maximum carboxylation velocity in vivo, suggesting that the fraction of inactive Rubisco increased with increasing N supply. Although CO2-enriched plants accumulated more non-structural carbohydrates in the leaf, the reduction of photosynthetic capacity at low N supply was not mediated simply by a build-up of carbohydrates. In D . glomerata , the photosynthetic capacity was mainly determined by the total N content of the leaf.  相似文献

The water fern Azolla pinnata R. Br. was fumigated for 1 week with either 25, 50 or 100 nl 1−1 SO2. The symbiosis of Azolla with Anabaena azollae (spp.) was severely damaged by atmospheric SO2 even at concentrations as low as 25 nl 1−1, with significant reductions in growth, reduction of C2H2, NH3 assimilation, protein synthesis, and heterocyst development. These disturbances appear to be mainly responsible for the extreme sensitivity of this fern to atmospheric SO2. Changes in violaxanthin/antheraxanthin and epoxy-lutein/lutein ratios also indicate that free radical products are induced by atmospheric SO2. These results suggest that the Azolla-Anabaena symbiotic system is a very responsive and reliable lower plant model to study the detailed effects of total sulphur deposition upon the balances between various important plant metabolic processes.  相似文献

Bark beetles, especially Ips typographus L. represent a severe biotic threat for spruce ( Picea abies [L.] Karst.) at low altitudes in Europe. We compared sulphur (total S, SO42−, glutathione, cysteine, methionine), nitrogen (total N, NO3, total protein, free amino acids), carbon, total phosphorus and PO43−, tree vigour index (TVI) and water content of the phloem after felling, and their dependent changes (tdc) with the breeding success of I. typographus . Twenty trees were classified according to age (34/90 years) and crown density (high/intermediate/low). Water content was higher in young trees than in old trees, higher in the crown than at breast height, and decreased significantly within the 8-week study period. In old trees, breeding success, length of mother galleries and SO42− were significantly higher, while total protein, NO3 and water content were significantly lower than in young trees. Trees with intermediate crown density provided the best breeding success for I. typographus and had significantly higher arginine content and C/N ratio as well as low amounts of phosphate and glutamine. During the period of bark beetle breeding, total sulphur, glutathione, protein, NO3, aspartate, glutamine, glutamate, arginine and γ-aminobutyrate decreased significantly. The results support previous investigations that I. typographus develops best in physiologically weakened trees.  相似文献

In order to investigate effects of limited NO3 availability in corn ( Zea mays L. cv. Brulouis) 17-day-old plants were grown for a further 25 days on sand in a growth chamber. The plants received frequent irrigation with a complete nutrient solution containing 0.2, 0.6, 1.5 or 3.0 mM NO3. With 0.2 mM NO; nitrate levels in both roots and leaves diminished rapidly and were almost zero after 10 days treatment. Concurrently, as signs of nitrogen deficiency appeared, shoot growth was restricted, whereas root growth was enhanced. In addition, the concentration of reduced nitrogen and malate in the leaves declined, and in vitro nitrate reductase activity (NRA. EC 1.6.6.1), soluble protein and chlorophyll levels of leaf tissue were depressed and starch concentration was enhanced. With 0.6 mM NO3 in the nutrient solution, the decrease in NO3 levels in the tissues and the increase in root development were similar to those observed with 0.2 mM NO3. However, shoot growth, reduced nitrogen concentration in leaves, and the above-mentioned biochemical characteristics were almost identical to those obtained at 1.5 and 3.0 mM NO3. This indicates that when supplied with 0.6 mM NO3, corn plants were able to absorb sufficient NO3 to support maximal biomass production without appreciable NO3 accumulation in roots or shoot. It is, thus, suggested that the plants responded to low NO3, availability in medium by enhancing root growth and by maximizing NO3 reduction relative to NO3 accumulation.  相似文献

Abstract. Short-term fumigation of Spinacia oleracea with 380 μg m−3 H2S (250 ppb) resulted in a rapid accumulation of water-soluble SH-compounds in the shoots. After 1 h exposure a substantial increase in the SH-content was already detectable and maximal accumulation, three- to four-fold that in control plants, was observed after 24 h of exposure. Irradiation during H2S exposure only slightly affected the rate and level of SH-accumulation. H2S fumigation did not affect the water-soluble SH-content of the roots. Glutathione was the sole water-soluble SH-compound accumulating upon exposure to H2S. It was calculated that during the first hour of exposure to 380 μg m−3 H2S 39% of the possible absorbed H2S was converted into glutathione. The SH-content of the water-soluble proteins of the shoots was not affected by H2S exposure. When fumigation was stopped, a rapid decrease in glutathione content was observed and after 48 h the content was comparable to that of the control plants. Contrary to H2S, SO2 fumigation did not result in a rapid accumulation of glutathione in spinach shoots. The possible role of glutathione accumulation during H2S fumigation is discussed.  相似文献

Three 9-day-old cultivars of Hordeum vulgare L. (Barberousse, Gerbel and Panda) were exposed to low levels of SO2 fumigation (40 ± 5 and 117 ± 20 ppb). After 48 days of treatment the seedlings were harvested for lipid analysis. In comparison to the control (plants exposed to charcoal-filtered air), the total lipid content of fumigated seedlings declined at 40 ppb SO2 and even more so at 117 ppb in all three cultivars. A large reduction in diacylglycerols. polar lipids and free sterols was also observed after fumigations at both SO2 concentrations, whereas the treatments resulted in an increase in triacylglycerols and free fatty acids. The percentage composition of total fatty acids and that of each lipid class were changed by the fumigations. resulting in an increase in the degree of unsaturation. No changes in the percentage composition of sterols occurred in the fumigated leaves. These results suggest that even if SO2 may not directly oxidize unsaturated fatty acids at the low concentrations used here (which do not cause visible injury). it may alter lipid metabolism. This alteration. which was particularly evident in the polar lipids and sterols, could affect the functions associated with membrane stabilization, in which lipids plus sterols play a key part.  相似文献

Abstract. Presence of 2.4-diaminobutyric acid (A2bu), a neurotoxin, in tissues of flatpea ( Lathyrus sylvestris L.) necessitates a thorough understanding of the regulation of this nonprotein amino acid before the species can be recommended to livestock producers for forage applications. To determine how different concentrations and ratios of NO3 and NH+4 in growth media influence the levels of A2bu and other free amino acids in the 'Lathco'flatpea cultivar, plants were grown hydroponically in controlled environments. The concentration of A2bu was highest in tissues when the NO3 to NH+4 ratio in the nutrient solution was low. Responses of amides and other nonprotein amino acids, especially in the roots, followed a similar trend. Free protein amino acids in leaves and stems were generally unaffected by changes in NO3 to NH+4 ratios. In roots, protein amino acids increased as the NO3 to NH+4 ratio in the growth medium increased. Ammonium inhibited shoot and root growth; NO3 alleviated the toxic effects of NH+4. Soluble protein concentrations were higher in the shoots of NO3-fed plants and in the roots of plants supplied with NH+4. These results suggest that accumulation of A2bu and other nonprotein amino acids, as well as asparagine and glutamine, plays a role in detoxification of NH+4 and storage of N.  相似文献

In the 1990's, evidence has accumulated that various unfavourable environmental conditions substantially affect the turnover of the D1 protein of reaction centre II, the psb A gene product. Biochemical, molecular and physiological studies in higher plants indicate that alterations of D1 protein turnover occur under drought, nutrition deficiency, heat, chemical stress, ozone fumigation as well as UV-B and visible photo-stresses. The behaviour of photosystem II under these various stress conditions indicates that the response of D1 protein turnover can be interpreted as a general adaptive response to environmental extremes.  相似文献
