Free-moving rats chronically implanted in the striatum with multifiber carbon electrodes selective to dopamine were compressed in a helium-oxygen mixture to 80 bars. Extracellular dopamine level and behavioral symptoms of high-pressure neurological syndrome were simultaneously recorded. Under these conditions, the extracellular level of dopamine monitored by differential pulse voltammetry was found to be pressure dependent, and hyperlocomotor activity, a behavioral symptom of high-pressure neurological syndrome, was found to be linked to these pressure-induced changes in dopamine release.  相似文献

Background

Prolonged weaning from mechanical ventilation has a major impact on ICU bed occupancy and patient outcome, and has significant cost implications. There is evidence in patients around the period of extubation that helium-oxygen leads to a reduction in the work of breathing. Therefore breathing helium-oxygen during weaning may be a useful adjunct to facilitate weaning. We hypothesised that breathing helium-oxygen would reduce carbon dioxide production during the weaning phase of mechanical ventilation.

Materials/patients and methods

We performed a prospective randomised controlled single blinded cross-over trial on 19 adult intensive care patients without significant airways disease who fulfilled criteria for weaning with CPAP. Patients were randomised to helium-oxygen and air-oxygen delivered during a 2 hour period of CPAP ventilation. Carbon dioxide production (VCO2) was measured using a near patient main stream infrared carbon dioxide sensor and fixed orifice pneumotachograph.

Results

Compared to air-oxygen, helium-oxygen significantly decreased VCO2 production at the end of the 2 hour period of CPAP ventilation; there was a mean difference in CO2 production of 48.9 ml/min (95% CI 18.7-79.2 p = 0.003) between the groups. There were no significant differences in other respiratory and haemodynamic parameters.

Conclusion

This study shows that breathing a helium-oxygen mixture during weaning reduces carbon dioxide production. This physiological study supports the need for a clinical trial of helium-oxygen mixture during the weaning phase of mechanical ventilation with duration of weaning as the primary outcome.

The study was carried out on 17 healthy males aged 20-27 years subjected for 15 minutes to submaximal effort on a cycle ergometer (Elema-Schonander) under conditions of breathing ambient atmospheric air or a helium-oxygen mixture (20% O2 + 80% He) and under hypobaric pressure simulating an altitude of 3500 m above sea level. During the experiment the heart rate was recorded with ECG, and determinations were performed of the minute volume, respiratory rate, tidal volume and systolic arterial blood pressure. In the serum of venous blood obtained before and 3 minutes after the exercise the concentrations were measured of lactate (LA), pyruvate (PA) and glucose. High-altitude hypoxia caused unifavourable changes in the adaptation to effort manifesting themselves as an increase of the values of the determined physiological and biochemical indices. On the other hand, favourable changes were observed of the reaction to exercise while the subjects were breathing the helium-oxygen mixture during high-altitude hypoxia. The minute volume increased owing to increased tidal volume, and the exercise-induced rise of lactate (LA), pyruvate (PA) and the LA/PA ratio was lower. This may suggest reduced energy cost of respiration and reduced anaerobic metabolism under these conditions.  相似文献

Carotid blood flow was measured in rats by implanted transit-time ultrasonic flowprobes during hyperbaric experiments at up to 70 bar (7 MPa) using an helium-oxygen hyperoxic (partial pressure of O2 = 400 mbar) mixture. Before the hyperbaric experiment, an intracerebroventricular injection of phosphate saline buffered solution (PBS) or aminophylline, an adenosine receptor blocker, in PBS was given. Throughout the hyperbaric experiment carotid blood flow increased with ambient pressure in both PBS, i.e. control, and aminophylline treated rats. The increase in carotid blood flow was significantly attenuated in aminophylline treated rats. Additional experiments showed that the increased carotid blood flow was independent of hyperoxia as well as of temperature. The hypothesis that the hyperbaric dependent increase in carotid blood flow was mediated by brain adenosine receptors and its implication regarding a cerebral vasodilatation are discussed.  相似文献

The reported investigations were carried out on 17 healthy men aged 20-27 years subjected to a 15 minute submaximal exercise on an Elema-Schonander cycle ergometer while breathing ambient air or a helium-oxygen mixture (O2 20% and He 80%). During the exercise test the heart rate was recorded from the ECG tracings, with the respiratory rate and respiratory volume, minute ventilation and arterial blood pressure. The concentrations of lactate (LA), pyruvate (PA) and glucose were determined in the serum of venous blood obtained before and 3 minutes after the exercise. Favourable changes of the reaction of the organism to exercise were observed while the subjects breathed the helium-oxygen mixture. The minute ventilation increased owing to increased respiratory volume, and the exercise caused lower rises in LA, PA and the LA/PA ratio. This may suggest a reduction of respiration cost and a decrease of anaerobic metabolism under these conditions.  相似文献

The effect of inert helium and argon gases on the tissue respiration has been studied on lymphocyte suspensions of white rats. It is shown that normoxic helium-oxygen mixture induces almost a two-fold increase of the O2 uptake by lymphocytes as compared with the control (air). No deviations in the value of the studied parameter are revealed in case of replacement of nitrogen from air by argon. Significance of the membrane structure in realization of effects of inert gases is under discussion.  相似文献

Both the physical behavior of aerosols and survival of airborne Serratia marcescens in hyperbaric chambers with a helium-air mixture at 20 atm of pressure was approximately the same as in the system at ambient pressures. Exposure of mice to aerosols of Klebsiella pneumoniae at 1-, 2-, and 17-atm (ca. 101-, 203-, and 1,722-kPa) pressures of helium-oxygen mixture showed that the number of viable organisms constituting a 50% lethal dose was not significantly affected by the hyperbaric conditions.  相似文献

Abstract: In vivo electrochemical detection with a Nafion-coated carbon fiber working electrode, which provides information on the spatial and temporal dynamics of dopamine overflow, was used to investigate the involvement of nitric oxide (NO) in the dopaminergic transmission in the striatum of urethane-anesthetized Sprague-Dawley rats. A mixture of N -methyl- d -aspartate (NMDA) and nomifensine, a dopamine uptake blocker, was locally pressure-ejected to elicit a transient dopamine overflow from the dopamine-containing nerve terminals in the striatum. Local application of N ω-nitro- l -arginine methyl ester ( l -NAME), which blocks endogenous NO formation, increased the magnitude of dopamine release evoked by a subsequent NMDA and nomifensine application but resulted in no significant alteration in the time course. Furthermore, microejection of l -arginine, an NO precursor, or sodium nitroprusside (SNP), an NO generator, did not cause detectable changes in dopamine level in the striatal extracellular space. However, NMDA-induced dopamine release was profoundly inhibited with l -arginine or SNP pretreatment. In addition, NO affects dopamine uptake in rat striatum. Exogenous dopamine applied through a micropipette, reversibly and reproducibly, elicited an electrochemical signal. The time course of these signals was significantly prolonged by l -NAME treatment. These data suggest that NO is diversely involved in regulating dopaminergic transmission in rat striatum.  相似文献

N-Methyl-D-aspartate (NMDA) stimulated the release of endogenous dopamine from striatal slices prepared from adult Sprague-Dawley rats. A mixture of sodium fluoride and aluminum chloride (AlF4-) added to the slices significantly potentiated the NMDA-stimulated release of dopamine in a concentration- and time-dependent manner. The AlF4- mixture had no effect on the nonstimulated basal efflux of dopamine, and no increases in NMDA-stimulated release were observed when NaF was replaced with NaCl. Similarly, AlCl3 or a mixture of NaCl and AlCl3 had no effect on NMDA-stimulated release. The AlF(4-)-induced increase in NMDA-stimulated dopamine release was totally blocked by magnesium or the selective NMDA glycine antagonist 7-chlorokynurenic acid. Striatal slices depolarized with KCl (15 mM) also released dopamine and this release was similarly potentiated by AlF4-. However, KCl-stimulated dopamine release from striatal synaptosomes was not potentiated by concentrations of AlF4- that greatly increased release from striatal slices. NMDA did not stimulate the release of dopamine from striatal synaptosomes in the absence or presence of aluminum fluoride. Modulators of adenylate cyclase (forskolin) and protein kinase C (phorbol esters) did not enhance NMDA-stimulated dopamine release. The protein kinase C inhibitor H-7 also did not reduce the potentiating effects of AlF4-. The mixed cholinergic agonist carbachol and the calcium ionophore A23187 mimicked the AlF4- effect although the increase in NMDA-stimulated dopamine release produced by these agents was less than that seen with AlF4-.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

QUESTION: To determine flow pattern and critical Reynolds numbers in endotracheal tubes submitted to different helium-oxygen mixtures under laboratory conditions. MATERIALS AND METHODS: Flow-pressure relationships were performed for seven endotracheal tubes (rectilinear position, entry length applied) with distal end open to atmosphere (predicted internal diameters: 6-9 mm). Nine helium-oxygen mixtures were tested, with FIHe varying from zero to 0.78 (increment: 10%). Nine flows were tested, with rates varying from 0.25 to 1.60 l s(-1) (increment: 0.15 l s(-1)). Gas flow resistance was calculated, and for each endotracheal tube, a Moody diagram was realised. Flow regime and critical Reynolds numbers were then determined (fully established laminar, nonestablished laminar, smooth turbulent, or rough). RESULTS: Even low concentration of helium in inspiratory mixture reduces endotracheal tubes resistance. Effect is maximal for high flows, small tube and high FIHe. Critical Reynolds numbers are inversely correlated to tube diameter. ANSWER: Under laboratory conditions, flow pattern in endotracheal tubes varies from fully established laminar to rough. Knowledge of the critical Reynolds numbers allows correct application of fluid mechanic formula when studying tube or gaseous mixture effects on respiratory mechanisms.  相似文献

To determine whether marsupial mammals increase their metabolic capabilities during cold acclimation, the metabolism of both warm and cold acclimated Dasyuroides byrnei was examined by exposure to cold in a helium-oxygen atmosphere. Mean values of heat production and conductance were significantly higher in a helium-oxygen atmosphere than in air. Body temperature did not change until metabolic capacity was exhausted. Both cold and warm acclimated groups could maintain a metabolic scope of 10-11 times the basal or standard level for this species. Such a metabolic scope is much higher than levels recorded for placental mammals. At very low ambient temperatures cold acclimated D. byrnei could sustain a high level of heat production longer than could warm acclimated animals. While there are some similarities between marsupial mammals and placental mammals in their responses to cold acclimation, an increase in maximum metabolism, as reported for placentals, does not seem to occur in marsupials.  相似文献

The regional distribution of inhaled gas within the lung is affected in part by normal variations in airway geometry or by obstructions resulting from disease. In the present work, the effects of heterogeneous airway obstructions on the distribution of air and helium-oxygen were examined using an in vitro model, the two compartments of a dual adult test lung. Breathing helium-oxygen resulted in a consistently more uniform distribution, with the gas volume delivered to a severely obstructed compartment increased by almost 80%. An engineering approach to pipe flow was used to analyze the test lung and was extrapolated to a human lung model to show that the in vitro experimental parameters are relevant to the observed in vivo conditions. The engineering analysis also showed that helium-oxygen can decrease the relative weight of the flow resistance due to obstructions if they are inertial in nature (i.e., density dependent) due to either turbulence or laminar convective losses.  相似文献

1-methyl-4-phenylpyridinium ion (MPP+) was tested for its effects upon dopamine level after incubating striatal synaptosomes in medium with and without reserpine. In the absence of reserpine, MPP+ enhanced the total incubation mixture dopamine level when tyrosine was present in the medium but that enhancing effect was considerably weaker when tyrosine was replaced by alpha methyl p-tyrosine. 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) also had effects upon dopamine but likely due to MPP+, which was formed from MPTP by free mitochondrial MAO present in the tissue preparation. The incubation mixture dopamine level was drastically reduced by the addition of only reserpine and its presence in the medium markedly raised the ability of MPP+ to increase dopamine; the effects of MPTP in this medium were weaker than those of MPP+. Pargyline also raised dopamine levels under these conditions but only at concentrations much higher than those of MPP+. The particulate uptake of MPP+, at several medium concentrations, and the corresponding value of dopamine increase above the basal level were determined; the dopamine increase in p-moles was much greater than the p-moles of MPP+ uptake. These results indicate that, in the presence of reserpine, MPP+ has a potent action and that may lead to a release of intraneuronal free dopamine; this action is also likely to be independent of the countertransport from MPP+ uptake. The possibility of MPP+ being a potent inhibitor of intraneuronal MAO may have to be considered.  相似文献

J. Appl.Physiol. 83(2): 575-582, 1997.To study the effects of nitrogen addition to the breathing mixture on sleep disturbances at pressure, two dives were performed in whichhelium-nitrogen-oxygen mixture was used up to 450 m sea water (msw). Intotal, sleep of 12 professional divers was analyzed (i.e., 184 nightrecords). Sleep was disrupted by compression and by stay at 450 msw: we observed an increase in awake periods and in sleep stages I and II anda decrease in stages III and IV and in rapid-eye-movement sleepperiods. These changes, which were more intense at thebeginning of the stay, began to decrease from the seventh day of thestay, but the return to control values was recorded only during the decompression at depths below 200 msw. These changes were equivalent tothose recorded in other experiments with helium-oxygen mixture in thesame range of depths and were independent of the intensity of changesrecorded in electroencephalographic activities in awake subjects.

Effect of the medium with high partial pressure of helium and nitrogen on the content of di- and triphosphinositides in the rat brain great hemispheres. The stay of the rats in helium-oxygen mixture under the pressure of 40 kg/cm2 for 1 h is accompanied by the true decrease in the content of the both components of polyphosphoinositides whose value does not depend on the decompression conditions. Nitrogen-oxygen mixture under the pressure of 15 and 25 kgf/cm2 also evokes a decrease of the content of di- and triphosphoinositides. The character of reduction of the content of the both components for 4 h after the stay of the rats under the pressure of 25 kgf/cm2 depends on the decompression conditions.  相似文献

In older children and adults, density dependence (DD) of forced expiratory flow is present over the majority of the full flow-volume curve. In healthy subjects, DD occurs because the pressure drop from peripheral to central airways is primarily dependent on turbulence and convective acceleration rather than laminar resistance; however, an increase in peripheral resistance reduces DD. We measured DD of forced expiratory flow in 22 healthy infants to evaluate whether infants have low DD. Full forced expiratory maneuvers were obtained while the subjects breathed room air and then a mixture of 80% helium-20% oxygen. Flows at 50 and 75% of expired forced vital capacity (FVC) were measured, and the ratio of helium-oxygen to air flow was calculated (DD at 50 and 75% FVC). The mean (range) of DD at 50 and 75% FVC was 1.37 (1.22-1.54) and 1.23 (1.02-1.65), respectively, values similar to those reported in older children and adults. There were no significant relationships between DD and age. Our results suggest that infants, compared with older children and adults, have similar DD, a finding that suggests that infants do not have a greater ratio of peripheral-to-central airway resistance.  相似文献

Hyperbaric oxygen induced in rats a decrease in striatal dopamine levels. Such decrease could be a result of changes in glutamatergic and GABAergic controls of the dopaminergic neurons into the Substantia Nigra Pars Compacta. The aim of this study was to determine the role of gluatamatergic and Gama-Amino-Butyric-Acid neurotransmissions in this alteration. Dopamine-sensitive electrodes were implanted into the striatum under general anesthesia. After one week rest, awaked rats were exposed to oxygen–nitrogen mixture at a partial pressure of oxygen of 3 absolute atmospheres. Dopamine level was monitored continuously (every 3 min) by in vivo voltammetry with multifiber carbon electrodes before and during hyperbaric oxygen exposure. Hyperbaric oxygen induced a decrease in dopamine level in relationship with the increase in partial pressure of oxygen (?40% at 3 ATA). The used of N-Methyl-d-Aspartate, agonist of glutamatergic N-Methyl-d-Aspartate receptors did not improve considerably this change and gabazine antagonist of Gama-Amino-Butyric-Acid-a receptors induced some little alteration of this change. Brain ion homeostasis is severely perturbed during spreading depression of Leao and during anoxia. The ionic composition of the extracellular space changes abruptly and approaches the intracellular concentrations owing to an increase in cell permeability. In spreading depression, synchronous transmitter efflux caused by a depolarization of the presynaptic terminals has been implicated as a possible mechanism that would explain the concomitant movement of ions. Anoxia, having many features in common with spreading depression, may follow the same mechanism. We have measured the concentrations of extracellular potassium with ion-selective microelectrodes and dopamine by in vivo voltammetry with carbon fiber microelectrodes during spreading depression and anoxia to compare the temporal relationship between the release of dopamine and ion movements in the striatum. There is a pronounced release of dopamine during both spreading depression and anoxia. In spreading depression, the sharp increase of potassium concentration that follows an initial smaller and slower increase of potassium is accompanied by the release of dopamine. In anoxia, the dopamine release clearly precedes the fast rise of extracellular potassium concentration. We conclude that in striatum, there is a pronounced dopamine release during spreading depression and anoxia, but that the relationships between ionic changes and transmitter release for these two phenomena are different and probably reflect different mechanisms.  相似文献

We have studied the physical properties of monolayers formed by calix[4]resorcinarene and in mixtures with dipalmitoyl phosphatidylcholine (DPPC) in various molar ratios formed at the air-water interface and at presence of dopamine in water subphase by means of measurements of surface pressure and dipole potential. We showed that both calix[4]resorcinarene as well as its mixture with DPPC form stable monolayers at the water subphase. The presence of dopamine resulted in an increase of the mean molecular area and in a decrease of the compressibility modulus of the monolayers. For mixed monolayers at higher content of calix[4]resorcinarene (> 0.2 molar fraction) a deviation from ideal miscibility took place especially for monolayers in a solid state. This can be connected with formation of aggregates of calix[4] resorcinarene. Lowest miscibility and weakest interaction of dopamine with a monolayer was observed for calix[4]resorcinarene molar fraction of 0.33 in the monolayer.  相似文献
