Value-added abalone Haliotis discus hannai containing bioactive phlorotannins is produced by simply changing the feed to phlorotannin-rich brown seaweed Ecklonia cava 2 weeks prior to harvesting. We assessed the accumulation of phlorotannins by feeding with the seaweed after 4 days of starvation. Reverse-phase high-performance liquid chromatography afforded isolation of the major phlorotannins, which were identified by mass spectrometry and 1H-nuclear magnetic resonance to be 7-phloroeckol and eckol. Throughout the E. cava feeding period of 20 days, 7-phloroeckolol accumulated in the flesh (foot muscle tissue), up to 0.85?±?0.21 mg g?1 dry weight of tissue after 12 days. Eckol reached 0.31?±?0.08 mg g?1 dry tissue after 14 days. Feeding Laminaria japonica as a control, we detected no phlorotannins in the abalone muscle tissue. Abalone seaweed consumption and growth rate were similar when fed with E. cava or L. japonica for 20 days. Reduction in phlorotannins to half-maximal accumulation took 1.0 and 2.7 days for 7-phloroeckol and eckol, respectively, after replacement of the feed with L. japonica.  相似文献

The mass cultivation of Ecklonia stolonifera Okamura was studied as a possible summer feed for the abalone industry in Korea for the period between August and November when Undaria and Laminaria are not available. Experiments were conducted to investigate the optimal conditions for artificial seed production and mass cultivation of this species. Seedlings of E. stolonifera were reared in an indoor tank for 60 days until they were around 500 μm in length. Following indoor tank culture, the seedlings were transferred in situ to a nursery culture area for 2 months, before begin transferred to the main grow-out area. The maximum growth and development of young thalli in nursery culture area occurred at 2 m depth, whilst maximum growth of thalli in the main culture area occured at 1.5 m depth. Production of E. stolonifera was between 3 and 9 kg wet wt. m−1 in the first year of culture after seeding and 12 to 13 kg wet wt. m−1 in the second year of culture, after management (depth control and fouling organism removal, etc.) of the holdfast. The relationship between optimal water depth for culture and underwater irradiance during the E. stolonifera cultivation was defined as: y = −0.331x + 8.198 (r 2 = 0.9903). The growth rates achieved in this trial indicate that E. stolonifera cultures could produce sufficient biomass to supply summer feed for the Korean abalone industry.  相似文献

The culture of abalone is a growth industry in Australia that primarily utilises terrestrial crops to produce formulated pellet feeds. The use of cultivated macroalgae in place of such feeds could provide for better environmental, nutritional and/or economic outcomes for this industry. However, direct comparison trials using macroalgae and formulated crop feeds are rare, and it is therefore difficult to ascertain the benefits and costs of each feed type. This study compares the benefits to growth and performance of the cultivated hybrid abalone cross (Haliotis rubra 1814 Leach and Haliotis laevigata 1808 Donovan) which was fed one of eight dietary treatments, including two commercially formulated pellet feeds and six mixed macroalgae dietary treatments. Macroalgae dietary treatments comprised the three macroalgae species Grateloupia turuturu Yamada, Ulva australis Areschoug and/or Ulva laetevirens Areschoug. Four replicate tubs, each containing 40 juvenile abalone (10–15 mm), were used to test each dietary treatment over a 12-week period. Macroalgae dietary treatments provided for significantly higher specific growth rates of abalone compared to formulated feeds, by orders of magnitude, for both length (>0.2 % compared to <0.1 %?day?1, F 7, 31?=?22.3, p?<?0.0001) and weight (from <0.4 to >0.8 %?day?1, F 7, 31?=?24.4, p?<?0.0001). In addition, abalone health and condition increased, and the proximate composition of abalone tissue had a higher carbohydrate/protein ratio, higher ash content and lower lipid content. These findings suggest that the juvenile abalone may benefit from macroalgae diets in comparison to two formulated feeds as a result of optimal proximate composition of the algae biomass and improved condition of the abalone.  相似文献

In order to utilize the by-product of Ecklonia cava (the remaining biomass of E. cava), microbial fermentation which may result in the production of bioactive compounds using the by-product was applied in this study. The fermentation broth of E. cava by-product fermented with Candida utilis showed enhanced antibacterial activity against methicillin-resistant Staphylococcus aureus (MRSA) and food-borne pathogenic bacteria compared to that of control. To perform a more detailed investigation on the antibacterial activity, the broth was extracted with methanol and further fractionated with organic solvents. After 1 day of fermentation, the ethyl acetate (EtOAc) fraction exhibited the highest anti-MRSA activity with minimum inhibitory concentration ranging from 64 to 256 μg mL?1, suggesting that the fermentation of E. cava by-product with C. utilis could enhance antibacterial activity against MRSA. In addition, high-performance liquid chromatography analysis revealed that dieckol, eckol, eckstolonol, and triphlorethol-A contents in the EtOAc-soluble extract increased significantly. The anti-MRSA activity of E. cava by-product most probably originated from phlorotannins, and the fermentation of C. utilis may have stimulated the breakdown of phlorotannins or have increased the efficiency in extracting phlorotannins.  相似文献

The kelp Undariopsis peterseniana is warm-water-tolerant, and consequently, there is currently considerable interest in developing commercial cultivation techniques for this species in Korea. U. peterseniana plants have been successfully transferred to the northern coast of Korea beyond their original habitat in Jeju Island (33°30′08.65″N, 126°55′39.02″E). In this study, we cultured a hybrid kelp consisting of a cross between free-living gametophytes of U. peterseniana and U. pinnatifida in an attempt to extend the culture period of Undaria which is an important species for both the abalone industry and for commercial seaweed mariculture for human food applications. Morphological characters and cultivation period were compared between the parent thalli and the hybrid. The cultivation experiment was conducted in Wando, on the southern coast of Korea (34°26′18.68″N, 127°05′43.88″E). The morphological characteristics of the hybrid thalli were intermediate between the two species having shallow pinnated blades and a reduced reproductive organ. Hybrid thalli showed faster growth rates, 1.5 times greater biomass, and a longer cultivation period than the parent thalli. The hybrid strain possessed characteristics that indicate it could be used as an alternative kelp source to supply the abalone feed industry.  相似文献

Fragments of Ecklonia cava Kjellman were cultured under controlled laboratory conditions of light irradiance, water temperature, and photoperiod. To clarify the relationship between the maturation of E. cava and the photosynthetic products, laminaran, the content in the fragments was measured with the progress of maturation. The culture conditions ranged from 12.5 to 100 µmol m?2 s?1, 10–25°C, and 14 : 10 h LD (light : dark) to 10 : 14 h LD. In the case of low light conditions, despite an optimum temperature for maturation, the fragments did not form sori and laminaran was not accumulated during the culture period. In the case of sufficient light and non‐optimum temperature conditions, the fragments did not form sori, but laminaran was accumulated. When the fragments were cultured under optimum light and temperature conditions for maturation, laminaran was accumulated in the early stage of maturation, just before or after cortex of the bladelets thickened, and decreased with the progress of maturation, and all fragments matured regardless of the length of the photoperiod. So, these results support the idea that laminaran is used as the main respiratory substrate in the maturation of E. cava.  相似文献

Experiments in the subtidal zone of the southeastern coast of Korea examined the role of competition in determining the upper limit of perennial kelp, Ecklonia stolonifera, which is a dominant species in the subtidal community, but rarely found shallower than 5 m depth. Replicate clearings of dense algal turfs simulating natural disturbance at 1 and 3 m depth zones showed that E. stolonifera is able to settle and develop adult sporophytes in the upper zone. However, these adult sporophytes were eventually excluded from the clearings within one year after clearing. Two-year succession patterns at the clearings fell into three sequential categories: E. stolonifera and polychaete worms (Serpula vermicularis Linnaeus), mussels (Mytilus edulis Linnaeus), and turf-forming algae. A competitor removal treatment revealed that competition with M. edulis for primary space is the direct cause resulting in exclusion of E. stolonifera from the clearings. Recolonization of E. stolonifera after exclusion did not occur at clearings due to preemptive competition with other sessile organisms, which occupied 75-99% of bottom cover during the reproduction period of E. stolonifera (Autumn). Algal turfs, the final colonizer of the clearings, recovered more than 80% of bottom cover within 10 months after exclusion of M. edulis, and their species composition and abundance were not significantly different to those of adjacent controls. Experiments using artificial substrata showed that recruitment of E. stolonifera from spores is essentially impossible on the plates occupied by other sessile organisms (>86% cover). These results indicate that preemptive competition with dense algal turfs for the settlement and competition with M. edulis for survival in the course of succession are two most important factors determining the vertical distribution limits of E. stolonifera.  相似文献

Ecklonia stolonifera is distributed along the coast facing the Sea of Japan. The size of various parts of the shoot (blade length and width and stipe length and diameter) and the age were determined at Ooma, Aomori Prefecture. The smaller the holdfast, the higher the percentage of one-year-old shoots. Holdfasts 10 cm in diameter seemed to be three years old, whereas holdfasts 40 cm in diameter seemed to be five or more years old. Zoosporangial sori were observed on blades three or more years old. Ecklonia stolonifera holdfast diameter expands only vegetatively by stoloniferous rhizoids. Zoospores, formed on shoots three or more years old, serve for the formation of new populations.  相似文献

Net production of theEcklonia cava community was monitored on a monthly basis for a year, and annual net production was estimated. Growth rate of blades reached a maximum of about 13 g dry wt·m?2·day?1 in spring and a minimum of about 2 g dry wt·m?2·day?1 in late summer. Annual production of blades was calculated to be 2.84 kg dry wt·m?2·year?1. If the growth of stipes is taken into account, annual net production is estimated to be about 2.9 kg dry wt·m?2·year?1. Standing crop was monitored monthly for two and a half years, and a close negative correlation was found between seasonal change in standing crop and net production. Standing crop reached a maximum of about 3 kg dry wt·m?2 in summer and a minimum of about 1 kg dry wt·m?2 in winter. Low productivity in summer at a period of maximum biomass may be explained by the dense canopy and the large area of reproductive portion occupying a blade, which diminish net assimilation.  相似文献

The effects of different diets on growth in the cultured South African abalone, Haliotis midae (Linnaeus), was investigated. Growth of juvenile Haliotis midae was monitored on a commercial abalone farm over a period of 9 months in an experiment consisting of 9 treatments with 4 replicates (n = 250 individuals per replicate). The treatments were: fresh kelp (Ecklonia maxima) blades (seaweed control); Abfeed® (formulated feed control); kelp + Abfeed® dried kelp pellets; dried kelp blades; dried kelp stipes; fresh kelp with the epiphyte Carpoblepharis flaccida; a mixed diet (Gracilaria gracilis, Ulva lactuca, and kelp) and a rotational diet (abalone were fed 1 of the 9 treatments for the first week and them kelp for the next 3 weeks). Results show that abalone grow well on all fresh seaweed combinations, but grow best on a mixed diet. The likely reason for the success of the mixed diet is that the red and green seaweed was farm grown, with an increased protein content. Dried kelp in any form produced poor growth. Abalone fed on the mixed diet grew at 0.066 mm day?1 shell length and 0.074 g day?1 body weight; this corresponds to 24.09 mm shell length and 27.01 g body weight increase per annum. Abalone fed on dried kelp grew at only 0.029 mm day?1 shell length and of 0.021 g day?1 body weight. Abalone grown on Abfeed® grew at 0.049 mm day?1 shell length and 0.046 g day?1 body weight which corresponds to 17.88 mm and 16.79 g increase per annum; this is better than the dried seaweed feeds, but poorer than the fresh seaweed combinations. This study shows that seaweed diets, particularly if the diets include seaweeds grown in animal aquaculture effluent, are good substitutes for the formulated feed generally used today.  相似文献

Four strains of marine microalgae commonly used as live feeds in hatcheries (Isochrysis sp. T.ISO, Tetraselmis suecica, Phaeodactylum tricornutum, Nannochloropsis sp.) were grown in a novel solid-state photobioreactor, the twin-layer system. Microalgae were immobilized by self adhesion to vertically oriented twin-layer modules which consisted of two different types of ultrathin layers, a macroporous source layer (glass fiber nonwoven) through which the culture medium was transported by gravity flow, and a microporous substrate layer (plain printing paper) which carried the algae on both surfaces of the source layer. This simple open cultivation system effectively separated the immobilized microalgae from the bulk of the growth medium and permitted prolonged cultivation of microalgae with average biomass yields of 10–15 g dry weight m?2 growth area after 14–25 days of cultivation. Algal biomass was harvested as fresh weight (with 72–84 % water content) without the need to pre-concentrate algae. No aeration or external CO2 supply was necessary, and due to the microporous substrate layer, no eukaryotic contaminations were observed during the experiment. All experiments were conducted in Germany under greenhouse conditions with natural sunlight. Small-scale growth experiments performed under the same conditions revealed that growth over most of the experimental period (24 days) was linear in all tested algae with growth rates (dry weight per square meter growth area) determined to be 0.6 g ?m?2?day?1 (Isochrysis), 0.8 g? m?2?day?1 (Nannochloropsis), 1.5 g ?m?2?day?1 (Tetraselmis), and 1.8 g? m?2?day?1 (Phaeodactylum). Due to its cost-effective construction and with further optimisation of design and productivity at technical scales, the twin-layer system may provide an attractive alternative to methods traditionally used to cultivate live microalgae.  相似文献

Coastal structures are constructed principally to protect the coast line. However, these structures also can act as artificial substrata for seaweeds. In particular, armor blocks, such as tetrapods, prove to be good algal substrata. Our field observations on the vegetation and standing crop of seaweed communities on armor blocks led us to the following conclusions: 1) Ecklonia cavacommunities grew on tetrapods that had been placed to coincide with the maturation period of E. cava. The communities have been maintained for more than four years. 2) The standing crop of seaweeds on an offshore breakwater composed of tetrapods placed seven years before was almost the same as that found under natural conditions. 3) Variations in roughened surfaces (pebbles or scores and grooves 3 or 46 mm wide) did not affect the growth of Sargassum spp. However, a greater number of Ecklonia stolonifera plants were observed attached to roughened surfaces compared to smooth surfaces.  相似文献

In this study, fermentation was employed as a tool to further increase the bioactive potential of processing by-product from a brown seaweed, Ecklonia cava, which can be obtained from food and cosmetic industries after its polyphenol extraction. The fermentation process was done for 24 h using an industrially important microorganism Candida utilis prior to being extracted with 80 % ethanol. The anti-inflammatory potential of the fermented E. cava processing by-product extract (FEPBE) was evaluated in vitro. The phlorotannin-rich FEPBE dose-dependently inhibited the nitric oxide production, prostaglandin-E2 production and suppressed the inducible nitric oxide synthase and cyclooxygenase-2 expressions in lipopolysaccharide-stimulated RAW 264.7 cells. The release of pro-inflammatory cytokines, interleukin-1β and interleukin-6, was significantly suppressed by the extract in a dose-dependent manner. Due to the profound anti-inflammatory activity, FEPBE appears as a value-added biomass fraction that can be exploited in numerous industrial applications as a source of functional ingredients.  相似文献

Brown algae of the order Laminariales, commonly referred to as kelps, are the largest and most productive primary producers in the coastal inshore environment. The genus Ecklonia (Lessoniaceae, Phaeophyceae) consists of seven species with four species in the Northern Hemisphere and three in the Southern Hemisphere. It was recently transferred to the family Lessoniaceae based on phylogenetic analyses of nuclear and chloroplastic markers, though the type of the genus was not included and its relationship with allied genera Eckloniopsis and Eisenia remained unresolved. The present study is the first to produce a phylogeny focussed on the genus Ecklonia. It included sequences from nuclear, mitochondrial, and chloroplastic DNA, for most of the distribution range of the three current Southern Hemisphere species (Ecklonia radiata, Ecklonia maxima, and a sample of a putative Ecklonia brevipes specimen), sequences for East Asiatic species (Ecklonia cava, Ecklonia kurome, and Ecklonia stolonifera), as well as the closely related genera Eckloniopsis and Eisenia. Results confirmed E. radiata and E. maxima as two distinct species in South Africa, E. radiata as a single species throughout the Southern Hemisphere (in South Africa, Australia, and New Zealand) and East Asiatic species as a distinct lineage from the Southern Hemisphere clade. Results further pointed out a close sister relationship between Eckloniopsis radicosa and two Eisenia species (including the type species: Eisenia arborea) to the genus Ecklonia suggesting that the genera Eckloniopsis and Eisenia are superfluous.  相似文献

Cultivations of the brown seaweed, Saccharina japonica, was developed, promoted, and industrialized in the 1970s and is highly valued in Korea both for human consumption and as a feed for the rapidly developing abalone farming industry. Production has continually increased, and approximately 442,637 tonnes wet weight was harvested in Korea in 2015. Abalone is a highly sought-after delicacy in Korea, and the abalone farming industry has been developed based on a stable production of seaweed. Korean abalone farmers prefer to feed their stock on locally cultured S. japonica; however, between August and November, the supply of farmed S. japonica declines because higher seawater temperatures reduce S. japonica productivity. In an attempt to overcome this temperature-induced period of low production, cultivation trials with a strain of S. japonica selected to withstand higher cultivation temperatures were undertaken. Strain selection involved using individual parent thalli that were found to remain productive under high seawater temperature (26 °C) at Wando. Male and female gametophytes were isolated through 3 cycles of free-living gametophyte culture to produce the F3 strain used in the production trials. Production trials using the selected strain were initiated every month between December 2014 and March 2015. This delayed the initiation of culture beyond the latest initiation time currently used by farmers (December). Delaying initiation of cultivation resulted in delayed maximum growth compared to the control. Growth of the F3 strain continued for up to 3 months longer than normally achieved on farms for non-selected thalli. The mean length, growth rate, and biomass were also greater than those achieved by the control strain. The use of the F3 strain of S. japonica coupled with delayed initiation of culture can therefore be used to help to ensure a stable year round algal feed supply for abalone industry in Korea.  相似文献

Prolific algal growth in sewage ponds with high organic loads in the tropical regions can provide cost-effective and efficient wastewater treatment and biofuel production. This work examines the ability of Euglena sp. growing in wastewater ponds for biofuel production and treatment of wastewater. The algae were isolated from the sewage treatment plants and were tested for their nutrient removal capability. Compared to other algae, Euglena sp. showed faster growth rates with high biomass density at elevated concentrations of ammonium nitrogen (NH4-N) and organic carbon (C). Profuse growth of these species was observed in untreated wastewaters with a mean specific growth rate (μ) of 0.28 day?1 and biomass productivities of 132 mg ?L?1? day?1. The algae cultured within a short period of 8 days resulted in the 98 % removal of NH4-N, 93 % of total nitrogen 85 % of ortho-phosphate, 66 % of total phosphate and 92 % total organic carbon. Euglenoids achieved a maximum lipid content of 24.6 % (w/w) with a biomass density of 1.24 g ?L?1 (dry wt.). Fourier transform infrared spectra showed clear transitions in biochemical compositions with increased lipid/protein ratio at the end of the culture. Gas chromatography and mass spectrometry indicated the presence of high contents of palmitic, linolenic and linoleic acids (46, 23 and 22 %, respectively), adding to the biodiesel quality. Good lipid content (comprised quality fatty acids), efficient nutrient uptake and profuse biomass productivity make the Euglena sp. as a viable source for biofuel production in wastewaters.  相似文献

Two abalone species: green Haliotis fulgens and yellow Halioti corrugata represent nearly 97% of the total production in the Mexican abalone fishery. It has been assumed that abalone feed on the kelp algae Macrocystis pyrifera. Regional hatcheries use this species as a main source of natural food. M. pyrifera does not occur at the southern limit of the distribution of abalone species along the Baja California Peninsula. In this study, growth rates of juveniles H. fulgens, 17.3 ± 2.2 mm shell length and 0.4 ± 0.2 g body weight, were evaluated. Juveniles were fed with common species in the benthic environments inhabited by abalone along the western coast of Baja California during 191 days. Three diets were based on algae: palm kelp, Eisenia arborea, giant kelp, M. pyrifera and Gelidium robustum, and one on seagrass, Phyllospadix torreyi. Shell length and body growth rates varied between 21.5 μm day?1 and 2.2 mg day?1 for E. arborea and between 45.9 μm day?1 and 6.7 mg day?1 for M. pyrifera. Higher specific growth rates (SGR) in length and weight were determined for M. pyrifera: 0.2% and 0.7% day?1. Significant differences between values of juveniles fed M. pyrifera with the rest of the diets were found. The highest mortality (21%) was in juveniles fed the red algae G. robustum.  相似文献

Pigment contents and proximate compositions are important traits to evaluate the nutritional value of microalgae. The environmental condition and nutrient availability in the culture medium are hypothesised to regulate algal colour, nutritional composition and production. This study aimed to compare the pigment and proximate compositions in Tisochrysis lutea under phototrophic and mixotrophic conditions in an attempt to improve algal product quality. Algae were grown in the phototrophic and mixotrophic conditions at 50 μmol photons m?2 s?1 for 12 h daily, but 50 mM glycerol was added as a carbon source in the mixotrophic culture. The pigment contents in algae significantly increased in the mixotrophic condition, and the contents of chlorophylls a and c and carotenoids increased nearly 60 % compared with those in the phototrophic condition. The contents of saturated and polyunsaturated fatty acids in algae were significantly higher while the content of monounsaturated fatty acids was significantly lower in the mixotrophic condition than those in the phototrophic condition. Although the contents of protein, lipid and carbohydrate in T. lutea were not different, their overall production was significantly enhanced in the mixotrophic condition compared to the phototrophic condition. This study indicates that mixotrophic culture promotes pigment and proximate production and the change of fatty acid profile in algae depends on the addition of organic carbon to the culture medium.  相似文献

Bio-fixation of carbon dioxide (CO2) by microalgae has been recognised as an attractive approach to offset anthropogenic emissions. Biological carbon mitigation is the process whereby autotrophic organisms, such as microalgae, convert CO2 into organic carbon and O2 through photosynthesis; this process through respiration produces biomass. In this study Dunaliella tertiolecta was cultivated in a semicontinuous culture to investigate the carbon mitigation rate of the system. The algae were produced in 1.2-L Roux bottles with a working volume of 1 L while semicontinuous production commenced on day 4 of cultivation when the carbon mitigation rate was found to be at a maximum for D. tertiolecta. The reduction in CO2 between input and output gases was monitored to predict carbon fixation rates while biomass production and microalgal carbon content are used to calculate the actual carbon mitigation potential of D. tertiolecta. A renewal rate of 45 % of flask volume was utilised to maintain the culture in exponential growth with an average daily productivity of 0.07 g L?1 day?1. The results showed that 0.74 g L?1 of biomass could be achieved after 7 days of semicontinuous production while a total carbon mitigation of 0.37 g L?1 was achieved. This represented an increase of 0.18 g L?1 in carbon mitigation rate compared to batch production of D. tertiolecta over the same cultivation period.  相似文献

The aim of the present study was to survey the growth and astaxanthin production of E17, an astaxanthin-rich mutant of Chlorella zofingiensis, through feeding the low-cost carbon source cane molasses. In heterotrophic batch cultivation, E17 fed with pretreated molasses achieved biomass (1.79 g L?1 day?1) and astaxanthin (1.99 mg L?1 day?1) productivities comparable to those with glucose, which were about 2- and 2.8-fold of those fed with untreated molasses, respectively. Molasses-induced astaxanthin accumulation may be attributed to the elicited expression of carotenogenic genes, in particular the genes specifically responsible for the ketolation and hydroxylation of β-carotene to form astaxanthin. A two-stage fed-batch strategy was employed to grow E17 and induce astaxathin accumulation, resulting in 45.6 g L?1 biomass and 56.1 mg L?1 astaxanthin, the highest volumetric astaxanthin yield ever reported for this alga. In addition, the astaxanthin production by E17 was tested with a semi-continuous culture method, where the directly diluted raw molasses (giving 5 g L?1 sugar) was used as the carbon source. Little growth inhibition of E17 was observed in the semi-continuous culture with a biomass productivity of 1.33 g L?1 day?1 and an astaxanthin productivity of 0.83 mg L?1 day?1. The mixotrophic semi-continuous cultures enhanced the biomass and astaxanthin productivities by 29.3 % and 42.2 %, respectively. This study highlights the potential of using the industrially cheap cane molasses towards large-scale cost-saving production of the high-value ketocarotenoid astaxanthin.  相似文献
