The effects of alarm substance on feeding behaviour of zebra danio fish (Brachydanio rerio) were tested by offering them high and low densities of enclosed waterfleas (Daphnia magna). Normally the fish attacked high densities of prey, but when exposed to alarm substance they preferred lower and presumably less confusing prey densities — also lowering their feeding rate.  相似文献

Inbreeding depression was observed in Half and Full sib matings (Inbreeding Coefficient 0.125 and 0.25) of Zebra Danios ( Brachydanio rerio ). Inbreeding at these levels reduced fertility, survival to 30 days, and length at 30 days. There was an increase in the incidence of crippled fry. No effect of inbreeding on either number of eggs per spawn or hatchability were found.  相似文献

The zebrafish has become a popular model for studies of vertebrate development and toxicology. However,in vitro approaches utilizing this organism have not been fully exploited due to the absence of suitable cell culture systems. Previously, we developed methods for the culture of cells derived from zebrafish blastula-stage embryos. One of these cultures, ZEM-2, was derived in a complex medium containing trout embryo extract, trout serum and medium conditioned by buffalo rat liver cells. In this study we describe a zebrafish embryo cell line, ZEM-2A, derived from ZEM-2 following selection for growth in a simplified medium. Optimal growth of ZEM-2A cells is attained in nutrient medium supplemented with 5% fetal bovine serum.  相似文献

采用斑马鱼胚胎发育技术,对环境激素类物质五氯酚的毒性进行测定,结果表明,五氯酚(PCP)对胚胎的特定作用时间段是卵产出至发育6 h之内;PCP对胚胎发育有明显的抑制作用,会造成胚胎发育的畸形或死亡,不同时间染毒产生的可观察毒理学终点各异;随着PCP对发育48 h斑马鱼胚胎作用时间的减短,其致死效应敏感性降低,其中0 hpf组的LC0值最小,为70.8μg·L-1,24hpf组LC0值最大,为831.8μg·L-1;斑马鱼胚胎对孵化后0时染毒的PCP最为敏感,PCP对胚胎产生急性毒性效应的敏感指标:心胞囊肿、血液循环障碍、无心律>孵化率降低>停滞发育作用;斑马鱼胚胎最敏感的指标为48 h血液循环障碍和48 h半致死效应.  相似文献

Zebra Danio ( Brachydanio rerio , Hamilton-Buchanan) tropical fish are easily maintained and bred under laboratory conditions; procedures for maintenance and breeding are described. Intensive inbreeding of this fish gave rise to a high incidence of spontaneous skeletal deformities which appear to reflect a genetic defect rather than an environmental stimulus. This finding casts doubt on the suitability of the Zebra Danio for fish toxicity testing.  相似文献

Summary Blastoderm explants fromBrachydanio rerio (Teleostei: Cyprinidae) high blastulas exhibited limited differentiation of optic structures in culture. A number of explants showed migration of pigmented retinal epithelial cells and formation of monolayers. The findings permit comparative studies in vitro on phenomena pertaining to pigmented retinal epithelial cell morphology, function, and differentiation. Timidity, vigilance and response to alarm substance were examined in shoals consisting of one to 12 zebra danios, Brachydanio rerio . Measures of timidity were not clearly related to shoal size. Fish in larger shoals spent more time in central areas of the tank away from cover than fish in smaller shoals. However, fish in small shoals appeared to be as aggressive as those in larger shoals. Vigilance and foraging rates did not appear to be related to shoal size. Per capita foraging rates and shoal size were not correlated. After being frightened by alarm substance, danios in larger shoals did not return to foraging sooner than those in smaller groups. Zebra danios in all shoal sizes responded behaviourally to alarm substance. It appears that the presence of conspecifics is unnecessary for alarm behaviours to occur and that the nature of the behaviours are independent of shoal size.  相似文献

Pigment cell precursors of the embryonic zebrafish give rise to melanophores, xanthophores and/or iridophores. Cell signaling mechanisms related to the development of pigmentation remain obscure. In order to examine the mechanisms involved in pigment cell signaling, we treated zebrafish embryos with various activators and inhibitors of signaling pathways. Among those chemicals tested, LiCl and LiCl/forskolin had a stimulatory effect on pigmentation, most notable in the melanophore population. We propose that the inositol phosphate (IP) pathway, is involved in pigment pattern formation in zebrafish through its involvement in the: (1) differentiation/proliferation of melanophores; (2) dispersion of melanosomes; and/or (3) synthesis/deposition of melanin. To discern at what level pigmentation was being effected we: (1) counted the number of melanophores in control and experimental animals 5 days after treatment; (2) measured tyrosinase activity and melanin content; and (3) employed immunoblotting techniques with anti-tyrosine-related protein-2 and anti-melanocyte-specific gene-1 as melanophore-specific markers. Although gross pigmentation increased dramatically in LiCl- and LiCl/forskolin treated embryos, the effect on pigmentation was not due to an increase in the proliferation of melanophores, but was possibly through an increase in melanin synthesis and/or deposition. Collectively, results from these studies suggest the involvement of an IP-signaling pathway in the stimulation of pigmentation in embryonic zebrafish through the synthesis/deposition of melanin within the neural crest-derived melanophores.  相似文献

Pathogen density and genetic diversity fluctuate in the outside-host environment during and between epidemics, affecting disease emergence and the severity and probability of infections. Although the importance of these factors for pathogen virulence and infection probability has been acknowledged, their interactive effects are not well understood. We studied how an infective dose in an environmentally transmitted opportunistic fish pathogen, Flavobacterium columnare, affects its virulence both in rainbow trout, which are frequently infected at fish farms, and in zebra fish, a host that is not naturally infected by F. columnare. We used previously isolated strains of confirmed high and low virulence in a single infection and in a co-infection. Infection success (measured as host morbidity) correlated positively with dose when the hosts were exposed to the high-virulence strain, but no response for the dose increase was found when the hosts were exposed to the low-virulence strain. Interestingly, the co-infection resulted in poorer infection success than the single infection with the high-virulence strain. The rainbow trout were more susceptible to the infection than the zebra fish but, in both species, the effects of the doses and the strains were qualitatively similar. We suggest that as an increase in dose can lead to increased host morbidity, both the interstrain interactions and differences in infectivity in different hosts may influence the severity and consequently the evolution of disease. Our results also confirm that the zebra fish is a good laboratory model to study F. columnare infection.  相似文献

Complex aquatic systems of karst harbour a rich but little‐investigated biodiversity. In Croatia and Bosnia–Herzegovina karst, temporal springs are inhabited by a group of minnow‐like fishes that retreat to the associated ground water during dry seasons and spend several months underground. The most abundant species in this group is Delminichthys adspersus (Heckel 1843), which also has the most fragmented distribution range. To determine the population composition and dispersal patterns, and to detect potential underground migration, a large genetic data set comprising 544 specimens of D. adspersus covering most of its distribution area was analysed. Analysis of mitochondrial cytochrome b sequences (~1000 bp) and eight microsatellite loci showed that D. adspersus comprises at least three subpopulations with gene flow occurring among them. Coalescent‐based analysis revealed a complex migration pattern, with several unidirectional dispersal paths, including between temporal springs that share no surface connection. The results of this study suggest the existence of recurrent underground migration of fish in a karst environment and demonstrate the complexity of its hydrological network. The findings are relevant to conservation strategies for endemic karst organisms and karst ecosystems as a whole.  相似文献

A fish expression vector, FRM, was constructed by fusing the carp β-actin promoter and first intron to the ocean pout antifreeze protein terminator and putative boundary element. Mutant forms of the green fluorescent protein (GFP) were engineered into this vector, and the resultant series of vectors, FRMwg, FRM3wg (green GFP), and FRM2bl (blue GFP), were used to make transgenic zebra fish. After microinjection of either supercoiled or linearized DNA into one-celled eggs, GFP-expressing cells could be monitored by fluorescence microscopy commencing with the midblastula transition and continuing through embryogenesis. From adult fish, which retained scorable GFP either as patches or as a uniform fluorescence, 11 green and 1 blue GFP-expressing lines of zebra fish have been established. Expression of GFP was nearly ubiquitous and similar among all of these lines. Embryonic expression could be scored at 15 to 30 hours postfertilization and was seen throughout the embryo with the exceptions of the yolk, red blood cells, and in some lines, portions of the head. Adult expression was in a majority of tissues (e.g., muscle, brain, intestine, and heart, but not red blood cells). The notable difference between lines was that fluorescent eggs were scorable in seven of the lines. Adult homozygotes from a different subset of eight lines could be selected by the relative intensity of the GFP marking when compared with that in sibling heterozygotes. All 12 lines contain apparent single locus, multicopy, tandem integrations (1.5–100 copies per cell) of the transgenic DNA. The fish expression vector FRM could be used to drive nearly ubiquitous and strong expression of gene products other than GFP. The GFP expression vectors, FRMwg, FRM2wg, FRM3wg, and FRM2bl, may be useful for optimization of transgenesis and as a comarker. Morphogenetic movements accompanying formation of the neural keel and neural tube in the zebrafishDanino (Brachydanio) rerio were studied by labelling single neural plate cells with fluoresceinated dextran (FDA) during late gastrula stages (95–100% epiboly) and localizing their progeny with an anti-fluorescein antibody on histological sections throughout neurulation. The mediolateral extent of the neural plate correlates directly with the dorso-ventral extent of the neural tube. That is to say, the progeny of cells located medially in the neural plate come to lie ventrally in the neural tube; cells located laterally in the neural plate give rise to progeny that populate dorsal levels in the neural tube. Fixation of labelled cells at various stages reveals that neural keel and nerve rod are organized as monostratified epithelia and that they maintain this organization during neurulation. These observations strongly suggest that the neural keel in the zebrafish forms by way of infolding of the neural plate and, therefore, utilizes a mechanism similar to primary neurulation in other vertebrates. The folding process juxtaposes the apical surfaces of both flanks of the neural plate at the midline. Mitoses occur preferentially in this zone, leading very frequently to formation of bilaterally symmetrical clones of progeny cells. The size of the clones that develop from injected cells suggests that neural plate cells divide an 1.5 times on average between late gastrula and the end of neurulation. Correspondence to: J.A. Laboratory bred Zebra Danio ( Brachydanio rerio , Hamilton-Buchanan) tropical fish are prone to skeletal deformities resembling scoliosis and lordosis. This condition appears related to diet and has been studied by breeding from different broods of fish for three generations. Two broods bred using two commercially available tropical flaked foods developed severe spinal curvature three to six weeks post-hatching. A third brood of fish fed exclusively on live food did not develop any deformity. The results demonstrate that a dietary factor is responsible for the deformities and that the Zebra Danio is especially susceptible to this factor as other species breed normally when fed the same commercial diet. Analysis of the diets showed no deficiency in either ascorbic acid (Vitamin C) or tryptophan, deficiency of either being associated with scoliosis and lordosis in fish. The analysis did show, however, a relationship between the lead content of the diet and the incidence of deformities. Lead has previously been implicated in skeletal deformities in second and third generation Brook trout, Salvelinus fontinalis.  相似文献

目的探讨心钠肽(ANP)在小鼠卵泡发育过程中的表达定位以及其对体外受精的影响。方法应用免疫组织化学对小鼠卵巢切片进行染色,检测ANP在卵泡不同发育阶段的表达情况。利用ANP受体A的拮抗剂anantin检测ANP在小鼠体外受精中的作用。结果在卵泡生长和分化期,ANP广泛地存在于间质细胞、内膜细胞、颗粒细胞、卵丘细胞、卵母细胞以及黄体细胞的胞质中,特别是在排卵前卵泡中的卵丘细胞上其表达最强;经ANP受体A的拮抗剂anantin孵育后的小鼠精子,其体外受精的卵裂率与对照组相比显著下降(58.7±4.3 vs.92.3±2.1,P〈0.01),但是对胚胎后来的发育没有影响。结论卵丘细胞中表达的ANP可能通过受体A参与了小鼠的受精过程。  相似文献

In fishes, arsenic (As) is absorbed via the gills and is capable of causing disturbance to the antioxidant system. The objective of present study was to evaluate antioxidant responses after As exposure in gills of zebrafish (Danio rerio, Cyprinidae). Fish were exposed for 48 h to three concentration of As, including the highest As concentration allowed by current Brazilian legislation (10 μg As/L). A control group was exposed to tap water (pH 8.0; 26 °C; 7.20 mg O2/L). As exposure resulted in (1) an increase (p < 0.05) of glutathione (GSH) levels after exposure to 10 and 100 μg As/L, (2) an increase of the glutamate cysteine ligase (GCL) activity in the same concentrations (p < 0.05), (3) no significant differences in terms of glutathione reductase, glutathione-S-transferase and catalase activities; (4) a significantly lower (p < 0.05) oxygen consumption after exposure to 100 μg As/L; (4) no differences in terms of oxygen reactive species generation and lipid peroxidation content (p > 0,05). In the gills, only inorganic As was detected. Overall, it can be concluded that As affected the antioxidant responses increasing GCL activity and GSH levels, even at concentration considered safe by Brazilian legislation.  相似文献

Isoenzyme patterns were studied in local populations of the carp ( Cyprinus carpio ) and the bream ( Sarotherodon mossambicus ) of the Cyprinid family. and in the trout ( Sabno giardneri ) of the family Salmonidae. Homogenates of heart muscle extracts were used in the identification of PGI. NADP-IDH. PGM and 6-PGD.
Polymorphisms found and gene frequencies obtained are discussed separately for each enzyme and species or population.
In the bream, variation was found only at the PGI locus. In the carp only the PGM locus was polymorphic whereas both PGM and IDH showed variation in the trout. The variation at the PGM locus in the trout cannot support the three locus model suggested elsewhere and clearly indicates a single locus for the mo-nomeric enzyme.
The large variety of NADP-IDH types in trout not only illucidates the complexity of this locus but confirms a disomic mode of inheritance. Genetic differences in the trout populations could be related to possible advantages for management purposes.  相似文献
