A novel flow injection-chemiluminescence (FI–CL) approach is proposed for the assay of pioglitazone hydrochloride (PG-HCl) based on its enhancing influence on the tris(2,2′-bipyridyl)ruthenium(II)–silver(III) complex (Ru(bipy)32+-DPA) CL system in sulfuric acid medium. The possible CL reaction mechanism is discussed with CL and ultraviolet (UV) spectra. The optimum experimental conditions were found as: Ru(bipy)32+, 5.0 × 10−5 M; sulfuric acid, 1.0 × 10−3 M; diperiodatoargentate(III) (DPA), 1.0 × 10−4 M; potassium hydroxide, 1.0 × 10−3 M; flow rate 4.0 ml min−1 for each flow stream and sample loop volume, 180 μl. The CL intensity of PG-HCl was linear in the range of 1.0 × 10−3 to 5.0 mg L−1 (R2 = 0.9998, n = 10) with limit of detection [LOD, signal-to-noise ratio (S/N= 3] of 2.2 × 10−4 mg L−1, limit of quantification (LOQ, S/N = 10) of 6.7 × 10−4 mg L−1, relative standard deviation (RSD) of 1.0 to 3.3% and sampling rate of 106 h−1. The methodology was satisfactorily used to quantify PG-HCl in pharmaceutical tablets with recoveries ranging from 93.17 to 102.77 and RSD from 1.9 to 2.8%.  相似文献

Effect of dietary supplementation of n‐3 and n‐6 polyunsaturated fatty acids (PUFA) on reproductive performances in both sexes of an Indian major carp, Catla catla (Ham.), were investigated in terms of sperm quality, spawning response, fecundity and spawn recovery over a consecutive 2‐year period. Sperm count and spermatocrit value were in the range of 3.2–3.8 × 107 ml−1 and 80–82%, respectively, in fish fed the PUFA‐enriched test diet, and significantly higher (P < 0.01) than in fish fed control diet (2.3–2.5 × 107 ml−1 and 64–70%). Spawning response in the test diet group was quite high (96%) compared to 76% in the control. Egg and larval quality were superior in the test diet group as evidenced by the significantly higher fertilization rate and larval survival in the range of 91–92% and 93.3%, respectively. Fatty acid profile of fertilized eggs and larvae showed that the quantities of 18:3 (n‐3), 20:5 (n‐3) and 22:6 (n‐3) were significantly (P < 0.01) increased in the test group while 18:1, 18:2 (n‐6) and 20:4 (n‐6) remained at higher levels in control. The results indicated that enrichment of n‐3 PUFA in the broodfish diet not only improved the quality of eggs and larvae in terms of recovery, but also enhanced the physiological response of broodfish to induced spawning. Oil enrichment in the broodfish diet should therefore be mandatory to ensure the optimum n‐3 PUFA content for warmwater carp for quality seed production.  相似文献

Proteomic approaches have unique advantages in the identification of biological pathways that influence physical frailty, a multifactorial geriatric syndrome predictive of adverse health outcomes in older adults. To date, proteomic studies of frailty are scarce, and few evaluated prefrailty as a separate state or examined predictors of incident frailty. Using plasma proteins measured by 4955 SOMAmers in the Atherosclerosis Risk in Community study, we identified 134 and 179 proteins cross-sectionally associated with prefrailty and frailty, respectively, after Bonferroni correction (p < 1 × 10−5) among 3838 older adults aged ≥65 years, adjusting for demographic and physiologic factors and chronic diseases. Among them, 23 (17%) and 82 (46%) were replicated in the Cardiovascular Health Study using the same models (FDR p < 0.05). Notably, higher odds of prefrailty and frailty were observed with higher levels of growth differentiation factor 15 (GDF15; pprefrailty = 1 × 10−15, pfrailty = 2 × 10−19), transgelin (TAGLN; pprefrailty = 2 × 10−12, pfrailty = 6 × 10−22), and insulin-like growth factor-binding protein 2 (IGFBP2; pprefrailty = 5 × 10−15, pfrailty = 1 × 10−15) and with a lower level of growth hormone receptor (GHR, pprefrailty = 3 × 10−16, pfrailty = 2 × 10−18). Longitudinally, we identified 4 proteins associated with incident frailty (p < 1 × 10−5). Higher levels of triggering receptor expressed on myeloid cells 1 (TREM1), TAGLN, and heart and adipocyte fatty-acid binding proteins predicted incident frailty. Differentially regulated proteins were enriched in pathways and upstream regulators related to lipid metabolism, angiogenesis, inflammation, and cell senescence. Our findings provide a set of plasma proteins and biological mechanisms that were dysregulated in both the prodromal and the clinical stage of frailty, offering new insights into frailty etiology and targets for intervention.  相似文献

Brachymystax tsinlingensis Li is a threatened fish species endemic to China. With the problems of environmental factors and seeding breeding diseases, it is important to further improve the efficiency of seeding breeding and the basis of resource protection. This study investigated the acute toxicity of copper, zinc and methylene blue (MB) on hatching, survival, morphology, heart rate (HR) and stress behaviour of B. tsinlingensis. Eggs (diameter: 3.86 ± 0.07 mm, weight: 0.032 ± 0.004 g) of B. tsinlingensis were selected randomly from artificial propagation and developed from eye-pigmentation-stage embryos to yolk-sac stage larvae (length: 12.40 ± 0.02 mm, weight: 0.03 ± 0.001 g) and exposed to different concentrations of Cu, Zn and MB for 144 h in a series of semi-static toxicity tests. The acute toxicity tests indicated that the 96-h median lethal concentration (LC50) values of the embryos and larvae were 1.71 and 0.22 mg l−1 for copper and 2.57 and 2.72 mg l−1 for zinc, respectively, whereas the MB LC50 after 144-h exposure for embryos and larvae were 67.88 and 17.81 mg l−1, respectively. The safe concentrations of copper, zinc and MB were 0.17, 0.77 and 6.79 mg l−1 for embryos and 0.03, 0.03 and 1.78 mg l−1 for larvae, respectively. Copper, zinc and MB treatments with concentrations greater than 1.60, 2.00 and 60.00 mg l−1, respectively, led to a significantly low hatching rate and significantly high embryo mortality (P < 0.05), and copper and MB treatments with concentrations greater than 0.2 and 20 mg l−1 led to significantly high larvae mortality (P < 0.05). Exposure to copper, zinc and MB resulted in developmental defects, including spinal curvature, tail deformity, vascular system anomalies and discolouration. Moreover, copper exposure significantly reduced the HR of larvae (P < 0.05). The embryos exhibited an obvious change in behaviour, converting from the normal behaviour of emerging from the membrane head first to emerging tail first, with probabilities of 34.82%, 14.81% and 49.07% under copper, zinc and MB treatments, respectively. The results demonstrated that the sensitivity of yolk-sac larvae to copper and MB was significantly higher than that of embryos (P < 0.05) and that B. tsinlingensis embryos or larvae might be more resistant to copper, zinc and MB than other members of the Salmonidae family, which benefits their resource protection and restoration.  相似文献

SYNOPSIS. Paramecium multimicronucleatum was exposed to various external concentrations of adenosine triphosphate (Na2ATP) to determine the effects thereof on the cycling rate of the nephridial apparatus. Normal rate was found to vary from 3.46 to 4.28 cycles/min with a mean rate of 3.85 cycles/min at 20 C. Concentrations of ATP of less than 5 × 10−4 M caused only slight, very temporary acceleration of the cycling rate. At 5 × 10−4 M the cycling rate was accelerated less than 15%. At 3 × 10−3 M cycling rate was accelerated, varying from 5.35 to 7.24 cycles/min, with a mean accelerated rate of 6.25 cycles/min, a mean aoceleration of 88.3%. Changes in rate after addition of 5 × 10−3 M ATP ranged from a decrease of 6.2% to an increase of 1.8%, with the nephridial apparatus ultimately stopping. At higher concentrations, stoppage was almost immediate. Paramecium is rapidly dehydrated by the ATP-accelerated cycling of its nephridial apparatuses, with a net loss of 27% of its volume in 6 minutes in the 3 × 10−3 M ATP.  相似文献

Abstract

Bacterial symbionts are one of the most toxic bacteria to insect pests. They have been isolated from the intestine of the entomopathogenic nematodes. Ten strains of the entomopathogenic nematodes, that may be different, have been isolated out of different soil fauna having different crops from different Governorates in Egypt. The bacterial symbionts in these strains have been isolated and tested for production parameters using four different media. Experiments showed that both Loria Broth (LB), and Nutrient Broth (NB) gave good results in laboratory production of bacterial symbionts, concerning stability, cell size, and pigment production during culturing. Experiments using different techniques of introducing the bacterial symbionts to larvae of the cotton leafworm showed that the injection technique was the most effective among all the tested techniques. This is followed by oral and forced feeding which seemed to give equal results. On the other hand, toxicity experiments showed that the four bacterial isolates named, G-NRC-A3, SH-NRC-A5, SH-NRC-A6, and N.Sinai-NRC-A8, besides the bacterial symbiont of Steinernema abbasi, Flavimonas oryzihabitans, all give 100% mortalities to Spodoptera littoralis larvae after 24 h post-treatment at the higher dose (5×104 cells/10 μl) but at the lower doses 5×103 and 5×102 cells/10 μl of injection solution, a 100% mortality was reached after 72 h post-treatment.  相似文献

A study was made of the time course and kinetics of [3H]GABA uptake by dispersed cell cultures of postnatal rat cerebellum with and without neuronal cells. The properties of GABA neurons were calculated from the biochemical difference between the two types of cultures. It was found that for any given concentration of [3H]GABA, or any time up to 20 min, GABA neurons in cultures 21 days in vitro had an average velocity of uptake several orders of magnitude greater than that of nonneuronal cells. In addition, the apparent Km values for GABA neurons for high and low affinity uptake were 0.33 × 10−6 M and 41.8 × 10−4 M, respectively. For nonneuronal cells, the apparent Km for high affinity uptake was 0.29 × 10−6 M. The apparent Vmax values for GABA neurons for high and low affinity uptake were 28.7 × 10−6 mol/g DNA/min and 151.5 mmol/g DNA/min, respectively. For nonneuronal cells, the apparent Vmax for high affinity uptake was 0.06 × 10−6 mol/g DNA/min. No low affinity uptake system for nonneuronal cells could be detected after correcting the data for binding and diffusion. By substituting the apparent kinetic constants in the Michaelis-Menten equation, it was determined that for GABA concentrations of 5 × 10−9 M to 1 mM or higher over 99% of the GABA should be accumulated by GABA neurons, given equal access of all cells to the label. In addition, high affinity uptake of [3H]GABA by GABA neurons was completely blocked by treatment with 0.2 mM ouabain, whereas that by nonneuronal cells was only slightly decreased. Most (75–85%) of the [3H]GABA (4.4 × 10−6 M) uptake by both GABA neurons and nonneuronal cells was sodium and temperature dependent.  相似文献

Several new 10-formyl and 10-hydroxymethyl derivatives of 5,8,10-trideazapteroic acid have been synthesized by a novel and convenient enamine alkylation procedure. Two of these compounds (10a and 10b) were shown to be very powerful inhibitors of L. casei (10a, IC50 = 8 × 10−6 M ; 10b, IC50 = 7 × 10−6 M ) and recombinant mouse (10a, IC50 = 3.4 × 10−5 M ; 10b, IC50 = 2.8 × 10−5 M ) glycinamide ribonucleotide formyltransferase (GARFT). These IC50 values are comparable to the classical GARFT inhibitor (6R)-DDATHF (IC50, L. casei 2.3 × 10−6M ; recombinant mouse 2.3 × 10−5 M ) under identical assay conditions. For both compounds, the inhibition of L. casei GARFT increased with time of incubation, but not markedly with the recombinant mouse enzyme. Due to their potential ability to interfere with purine biosynthesis and to penetrate microbial cells the new nonclassical GARFT inhibitors reported here may be useful for the treatment of infections caused by microorganisms that are sensitive and resistant to conventional antimicrobial agents.  相似文献

The metamorphic response of pediveliger larvae of Mytilus galloprovincialis to the neurotransmitter blockers chlorpromazine, amitriptyline, rauwolscine, idazoxan, atenolol and butoxamine, and to tetraethylammonium chloride (TEA) was investigated through a series of bioassays. Chlorpromazine, amitriptyline and idazoxin inhibited larval metamorphosis induced by 10?4 M epinephrine. The concentration that inhibited metamorphosis by 50% (IC50) for chlorpromazine and amitriptyline was 1.6 × 10?6 M and 6.6 × 10?5 M, respectively. Idazoxan was less effective with an IC50 of 4.4 × 1013 M. Moreover, these three inhibitors showed no toxicity at any of the concentrations tested. The larval metamorphic response to K+ was not inhibited by 10?3 M tetraethylammonium chloride after 96 h. Thus, the neurotransmitter blockers chlorpromazine and amitriptyline are inhibitors of larval metamorphosis, and will be useful tools for antifouling studies.  相似文献

A highly active mosquitocidal mutant of Lysinibacillus sphaericus Ahmed 2362, namely, UCR-146, was efficiently produced on cottonseed meal (CSM) medium, using sand as a carrier under solid state fermentation (SSF). The optimum CSM concentration for the highest sporulation and toxin formation was 12%. The maximum toxicity of the tested organism against second instar larvae of Culex pipiens was obtained at 25% moisture content, initial pH 6–7, 1% sodium acetate, 18.9×106 CFU/g inoculum and 6 days incubation period at 30°C. Pilot scale production of UCR-146 under the optimum SSF conditions was assessed in aluminium trays. Spore count, mortality of larvae and LC50 of the final product were 5.5×1010 CFU/g, 72% at 1 part per million (PPM) and 0.54 PPM, respectively. These results were comparable with those obtained from bench-scale production (in flasks). The cost of 1 kg of this bio-larvicide was estimated at US $0.34.  相似文献

The biological activities of two species of bacteria isolated from soil of cotton fields identified as Bacillus subtilis strain NRC313 (BS NRC313) and Bacillus thuringiensis strain NRC335 (BT NRC335) were evaluated against the third larval instar of the cotton leafworm, Spodoptera littoralis (Boisd.). The different entomopathogenic bacteria of BS NRC313and BT NRC335 contained 10 × 108 cell/ml, and caused mortality of 100 and 97.3% for the above mentioned strains, respectively. Concentrations of 2.5 × 108 to 10 × 108 cell/ml of strains BS NRC313 and BT NRC335 were applied to the larvae: LC50 were 3.3 × 108 and 3.9 × 108 cell/ml respectively. The influence of exposure to toxin concentrations manifested in terms of decreasing the adult emergence and prolongation of the generation period. The percentage of larvae that survived and succeeded to pupate increased by decreasing the concentration. The longevity of adult emergence that resulted from larvae treated with Bacillus subtilis were 6.0 ± 0.51 and 9.0 ± 0.63 days at 5 × 108 and 2.5 × 108 cell/ml, respectively compared with 9.8 ± 0.47 in control. The results indicated that Bacillus subtilis was more potent than Bacillus thuringiensis. Field applications of B. thuringiensis, B. subtilis and Reldan achieved 55.6, 67.4 and 89.4% reduction of the cotton leafworm larvae Spodoptera littoralis in clover plants under field conditions.  相似文献

The prognostic signatures play an essential role in the era of personalised therapy for cancer patients including lung adenocarcinoma (LUAD). Long noncoding RNA (LncRNA), a relatively novel class of RNA, has shown to play a crucial role in all the areas of cancer biology. Here, we developed and validated a robust LncRNA-based prognostic signature for LUAD patients using three different cohorts. In the discovery cohort, four LncRNAs were identified with 10% false discovery rate and a hazard ratio of >10 using univariate Cox regression analysis. A risk score, generated from the four LncRNAs’ expression, was found to be a significant predictor of survival in the discovery and validation cohort (p = 9.97 × 10 −8 and 1.41 × 10 −3, respectively). Further optimisation of four LncRNAs signature in the validation cohort, generated a three LncRNAs prognostic score (LPS), which was found to be an independent predictor of survival in both the cohorts ( p = 1.00 × 10 −6 and 7.27 × 10 −4, respectively). The LPS also significantly divided survival in clinically important subsets, including Stage I ( p = 9.00 × 10 −4 and 4.40 × 10 −2, respectively), KRAS wild-type (WT), KRAS mutant ( p = 4.00 × 10 −3 and 4.30 × 10 −2, respectively) and EGFR WT ( p = 2.00 × 10 −4). In multivariate analysis LPS outperformed, eight previous prognosticators. Further, individual members of LPS showed a significant correlation with survival in microarray data sets. Mutation analysis showed that high-LPS patients have a higher mutation rate and inactivation of the TP53 pathway. In summary, we identified and validated a novel LncRNA signature LPS for LUAD.  相似文献

Approximately 80 per cent of tyrosine hydroxylase activity in bovine mandibular nerve and rabbit sciatic nerve was soluble, and the rest of the activity was particle-bound. The soluble enzyme in bovine mandibular nerve was isolated by ammonium sulphate fractionation (25–35 per cent saturation). The enzyme had a pH optimum at 5·9 in Tris-acetate buffer, and at 6·5 in Tris-HCl or phosphate buffer. The enzyme required a tetrahydropteridine cofactor. Km values toward various tetrahydropteridines such as l -erythro-tetrahydrobiopterin (a probable natural cofactor), 2-amino-4-hydroxy-6-methyltetrahydropteridine, and 2-amino-4-hydroxy-6,7-dimethyltetrahydropteridine were 2 × 10−5m , 5 × 10−5m and 4 × 10−4m , respectively. The Km value for tyrosine at 1 × 10−3m -2-amino-4-hydroxy-6-methyltetrahydropteridine as a cofactor was 5 × 10−5m . The enzyme activity was markedly stimulated with Fe2+ or catalase, but Fe2+ gave higher activity. The activity was inhibited with α, α′-dipyridyl, l -α-methyl-p-tyrosine, and various catecholamines. Among catecholamines, dopamine was the most potent inhibitor. l -5-Hydroxytryptophan was an inhibitor as potent as dopamine. Neither d -5-hydroxytryptophan nor 5-hydroxytryptamine inhibited the enzyme. The inhibition by l -5-hydroxytryptophan was partially competitive with tetrahydrobiopterin at concentrations higher than 9 × 10−5m , and partially uncompetitive at concentrations lower than 9 × 10−5m . The addition of heparin or lysolecithin did not affect enzyme activity with tetrahydrobiopterin as cofactor.  相似文献

The online photoreaction of the rose bengal photosensitized luminol–copper (II) chemiluminescence (CL) system was used for the determination of β-nicotinamide adenine dinucleotide (NADH) and ethanol (EtOH) in pharmaceutical formulations combined with a flow injection technique. NADH can significantly enhance the CL emission of the reaction. For EtOH, alcohol dehydrogenase in soluble form was utilized in the presence of nicotinamide adenine dinucleotide resulting in NADH production. The limit of detection (3σ blank, 𝑛 = 3) of 4.0 × 10−8 and 2.17 × 10−5 M, and linear range 1.3 × 10−7 to 2.5 × 10−5 M (R2 = 0.9998, n = 6) and 0.11–2.17 × 10−3 M (R2 = 0.9996, n = 6) were obtained for NADH and EtOH respectively. The injection rate was 100 h−1 with a relative standard deviation (n = 3) of 1.5–4.8% in the range studied for both analytes. The procedure was satisfactorily applied to pharmaceutical formulations with recoveries in the range 91.6 ± 3.0% to 110 ± 2.0% for NADH and 88 ± 3.0% to 95.4 ± 4.0% for EtOH. The results obtained were very consistent and did not differ considerably from the reported approaches at a 95% confidence limit. The possible mechanism of the CL reaction is also explained briefly.  相似文献

Miscarriage is one of the main complications occurring in pregnancy. The association between adverse pregnancy outcomes and silent bacterial infections has been poorly investigated. Ureaplasma parvum and urealiticum, Mycoplasma genitalium and hominis and Chlamydia trachomatis DNA sequences have been investigated by polymerase chain reaction (PCR) methods in chorionic villi tissues and peripheral blood mononuclear cells (PBMCs) from females with spontaneous abortion (SA, n = 100) and females who underwent voluntary interruption of pregnancy (VI, n = 100). U. parvum DNA was detected in 14% and 15% of SA and VI, respectively, with a mean of bacterial DNA load of 1.3 × 10−1 copy/cell in SA and 2.8 × 10 −3 copy/cell in VI; U. urealiticum DNA was detected in 3% and 2% of SA and VI specimens, respectively, with a mean DNA load of 3.3 × 10−3 copy/cell in SA and 1.6 × 10−3 copy/cell in VI; M. hominis DNA was detected in 5% of SA specimens with a DNA load of 1.3 × 10−4 copy/cell and in 6% of VI specimens with a DNA load of 1.4 × 10−4 copy/cell; C. trachomatis DNA was detected in 3% of SA specimens with a DNA load of 1.5 × 10−4 copy/cell and in 4% of VI specimens with a mean DNA load of 1.4 × 10−4 copy/cell. In PBMCs from the SA and VI groups, Ureaplasma spp, Mycoplasma spp and C. trachomatis DNAs were detected with a prevalence of 1%–3%. Bacteria were investigated, for the first time, by quantitative real-time PCR (qPCR) in chorionic villi tissues and PBMCs from women affected by SA and VI. These data may help to understand the role and our knowledge of the silent infections in SA.  相似文献

This study introduces a novel chemiluminescence (CL) approach utilizing FeS2 nanosheets (NSs) catalyzed luminol–O2 CL reaction for the measurement of three pharmaceuticals, namely venlafaxine hydrochloride (VFX), imipramine hydrochloride (IPM), and cefazolin sodium (CEF). The CL method involved the phenomenon of quenching induced by the pharmaceuticals in the CL reaction. To achieve the most quenching efficacy of the pharmaceuticals in the CL reaction, the concentrations of reactants comprising luminol, NaOH, and FeS2 NSs were optimized accordingly. The calibration curves demonstrated exceptional linearity within the concentration range spanning from 4.00 × 10−7 to 1.00 × 10−3 mol L−1, 1.00 × 10−7 to 1.00 × 10−4 mol L−1, and 4.00 × 10−6 to 2.00 × 10−4 mol L−1 with detection limits (3σ) of 3.54 × 10−7, 1.08 × 10−8, and 2.63 × 10−6 mol L−1 for VFX, IPM, and CEF, respectively. This study synthesized FeS2 NSs using a facile hydrothermal approach, and then the synthesized FeS2 NSs were subjected to a comprehensive characterization using a range of spectroscopic methods. The proposed CL method was effective in measuring the aforementioned pharmaceuticals in pharmaceutical formulations as well as different water samples. The mechanism of the CL system has been elucidated.  相似文献

Counting growth-layer groups (GLGs) in teeth is one of the most precise and widely accepted methods for aging marine mammals. Male narwhals have a large erupted tusk that can be used for aging, but this tusk is often difficult or expensive to obtain from hunters and most females do not display the tusk; thus, alternative methods for narwhal aging are needed. In this study, we aged narwhals by counting annual GLGs in embedded tusks and by measuring the change in the ratio of D- and L-enantiomers of aspartic acid in the eye lens nucleus that occurs as the animal ages (the aspartic acid racemization [AAR] technique). Absolute age estimates were estimated for seven tusks aged ≤15 yr. Estimated age was a significant predictor of aspartic acid D/L ratios with a racemization rate (Kasp) of 9.72 × 10−4/year ± 2.28 × 10−4 and a (D/L)0 of 3.46 × 10−2 ± 1.78 × 10−3 (r2 = 0.74). Results from our study, which included more younger GLG-aged animals than previously evaluated, confirms AAR can be used to generate age estimates for narwhals.  相似文献

A flow injection (FI) methodology using the acidic potassium permanganate (KMnO4)–rhodamine-B (Rh-B) reaction with chemiluminescence (CL) detection was established to determine acetochlor and cartap-HCl pesticides in freshwater samples. Experimental parameters were optimized, and Chelex-100 cationic exchanger mini column and solid-phase extraction (SPE) were used as phase separation techniques. Linear calibration curves were observed for the standard solutions of acetochlor and cartap-HCl over the ranges 0.005–2.0 mg L−1 [y = 1155.8x + 57.551, R2 = 0.9999 (n = 8)] and 0.005–1.0 mg L−1 [y = 979.76x + 14.491, R2 = 0.9998 (n = 8)] with LODs and LOQs of 7.5 × 10−4 and 8.0 × 10−4 mg L−1 (3σ blank) and 2.5 × 10−3 and 2.7 × 10−3 mg L−1 (10σ blank), respectively, with an injection throughput of 140 h−1. These methods were used to estimate acetochlor and cartap-HCl with or without the SPE procedure, respectively, in spiked freshwater samples. Results obtained were not significantly different at a 95% confidence level to those of other reported methods. Recoveries for acetochlor and cartap-HCl were obtained over the ranges 93–112% (RSD = 1.9–3.6%) and 98–109% (RSD = 1.7–3.8%), respectively. 1 Granulovirus PoGV is a strong candidate to substitute for chemical insecticides in integrated pest management (IPM) of the potato tuber moth Phthorimaea operculella Zeller (Lepidoptera: Gelechiidae). Generally, susceptibility to baculoviruses decreases with increasing larval age. For effective field applications, knowledge of the age‐related change in susceptibility is crucial. 2 The relative susceptibility of various instars of P. operculella larvae to PoGV was studied by leaf‐disc bioassays in the laboratory at 26 °C incubation temperature. The LC50 increased from 4 × 106 granules/mL for neonate larvae to 1.2 × 107, 1.1 × 108, 4.4 × 108, and 4.2 × 109 granules/mL, signifying resistance ratios of 3, 29, 110, and 1051, for 5, 6, 7, and 9‐day‐old larvae, respectively. 3 The relationship between log‐LC50 values and log‐larval weight was significantly linear. A logistic function described significantly the relation between larval weights and physiological age measured in degree‐days (DD > 13.4 °C). Both functions allowed prediction of the activity ratio of PoGV for different larval ages by using temperature summation to express physiological age. 4 PoGV was found to be highly active against P. operculella larvae up to a physiological age of approximately 50 DD (>13.4 °C) (i.e. the first third of the total larval development time). Thereafter, the virus rapidly lost its activity against older larvae. Prospects for applying this knowledge in the field are discussed.  相似文献
