Most asexual species of fungi have either lost sexuality recently, or they experience recombination by cryptic sexual reproduction. Verticillium dahliae is a plant-pathogenic, ascomycete fungus with no known sexual stage, even though related genera have well-described sexual reproduction. V. dahliae reproduces mitotically and its population structure is highly clonal. However, previously described discrepancies in phylogenetic relationships among clonal lineages may be explained more parsimoniously by recombination than mutation; therefore, we looked for evidence of recombination within and between clonal lineages. Genotyping by sequencing was performed on 141 V. dahliae isolates from diverse geographic and host origins, resulting in 26,748 single-nucleotide polymorphisms (SNPs). We found a strongly clonal population structure with the same lineages as described previously by vegetative compatibility groups (VCGs) and molecular markers. We detected 443 recombination events, evenly distributed throughout the genome. Most recombination events detected were between clonal lineages, with relatively few recombinant haplotypes detected within lineages. The only three isolates with mating type MAT1-1 had recombinant SNP haplotypes; all other isolates had mating type MAT1-2. We found homologs of eight meiosis-specific genes in the V. dahliae genome, all with conserved or partially conserved protein domains. The extent of recombination and molecular signs of sex in (mating-type and meiosis-specific genes) suggest that V. dahliae clonal lineages arose by recombination, even though the current population structure is markedly clonal. Moreover, the detection of new lineages may be evidence that sexual reproduction has occurred recently and may potentially occur under some circumstances. We speculate that the current clonal population structure, despite the sexual origin of lineages, has arisen, in part, as a consequence of agriculture and selection for adaptation to agricultural cropping systems.  相似文献

Substrate-borne signals are widely used in Hemiptera and are known to be utilized in mate searching and recognition. Within this Order, the superfamily Psylloidea is a diverse taxon which uses this type of signal modality during mating behavior between the two sexes. This study describes and compares the previously unreported vibrational communication of two closely related species of Macrohomotoma (Homotomidae). Both genders of these two species, Macrohomotoma gladiata Kuwayama 1908 and Macrohomotoma robusta Yang 1984, emit vibrational signals and establish duets during mating. The structure of male calling consists of two chirps while the female response is a single chirp. Males may sometimes follow the female response by emitting a single chirp that sounds similar to the female response with respect to the chirp duration and dominant frequency. This behavior is novel among Psylloidea and its potential function is discussed. Specific comparison of signal characteristics has revealed that the two species of Macrohomotoma are clearly distinguishable from each other which opens the possibility of acoustic signals being used for species delineation.  相似文献

Acacia visco is a native South American tree species that has been extensively used for ornamental purposes and in carpentry, bodywork and parquet due to the hardness and durability of its wood. Little is known about genetic diversity and mating system of A. visco. The main aims of this study were to (1) estimate outcrossing rates in natural Argentinean populations using AFLP markers, (2) test for any difference in mating patterns among a large a patchy and relict population, and (3) compare the mating system of A. visco with other Acacia species. The three primer pairs used in the AFLP analysis revealed a total of 569 variable loci. Most genetic variation was observed among individuals within families (61.2 %). The estimate of multilocus outcrossing rate (t m) was high (≥0.971) in all populations. Average pairwise coancestry between progenies within families for each population ranged from 0.082 to 0.105 or from 0.125 to 0.136, depending on the method θ was estimated. In the three populations studied, the progenies of open pollination were constituted mainly for half-sibs (94.3 %). This work shows a similar mating system in all populations of A. visco in spite of their size differences, hypothesizing that the entire species has a similar mating system of outcrossing preferential. Considering the results obtained here where a high percentage of individuals were half-sibs, sampling large numbers of pods from individual trees for ex situ conservation will result in a genetically diverse sample as a consequence of high outcrossing rates.  相似文献

Tuber spp. are filamentous ascomycetes which establish symbiosis with the roots of trees and shrub species. By virtue of this symbiosis they produce hypogeous ascocarps, known as truffles. Filamentous ascomycetes can reproduce by homothallism or heterothallism depending on the structure and organization of their mating type locus. The first mating type locus in a truffle species has been recently characterized in Tuber melanosporum and it has been shown that this fungus, endemic in Europe, is heterothallic. The availability of sequence information for T. melanosporum mating type genes is seminal to cloning their orthologs from other Tuber species and assessing their reproductive mode. Here we report on the organization of the mating type region in T. indicum, the black truffle species present in Asia, which is the closest relative to T. melanosporum and is characterized by an high level of morphological and genetic variability. The present study shows that T. indicum is also heterothallic. Examination of Asiatic black truffles belonging to different genetic classes, sorted according to the sequence polymorphism of the internal transcribed spacer rDNA region, has revealed sequence variations and rearrangements in both coding and non-coding regions of the mating type locus, to suggest the existence of cryptic species within the T. indicum complex. The presence of transposable elements within or linked to the mating type region suggests a role of these elements in generating the genotypic diversity present among T. indicum strains. Overall, comparative analyses of the mating type locus have thus allowed us to tackle taxonomical and phylogenetic issues within black truffles and make inferences about the evolution of T. melanosporum-T. indicum lineage. Our results are not only of fundamental but also of applied relevance as T. indicum produces edible fruit bodies that are imported also into Europe and thus may represent a biological threat for T. melanosporum.  相似文献

Population genetic analyses are especially relevant for species considered threatened or highly endemic and for which other forms of biological information are lacking. Patellapis doleritica is a recently described communally nesting halictid bee of conservation concern because it is rare and endemic to the Succulent Karoo of South Africa. Moreover, its dispersal is considered to be restricted by its specialised nesting requirements and inclement weather conditions during its limited annual flight period, traits which may be common to other bee species of the region. We hypothesised that gene flow in P. doleritica was low, leading to marked genetic differentiation. Using 7 microsatellites, we investigated its mating and population genetic structure in 258 individuals (171 females and 87 males) from 7 populations spanning most of its known range. Deviation from Hardy–Weinberg equilibrium (FIS = + 0.254) suggested P. doleritica to be inbred, as in many other communal nesting bee species. Global FST (0.028) and global G′ST (0.216) revealed modest but significant differentiation between most populations, even across the very limited range of the species (ca. 25 km), with one genetically extreme outlier population. Despite inbreeding, we detected a surprisingly low frequency of diploid males (2 %). Patellapis doleritica nevertheless deserves special conservation attention since it is an endemic species with a low overall abundance and therefore possibly prone to environmental change and local extinction.  相似文献

Two new species of Pristionchus, P. lucani n. sp. and P. bulgaricus n. sp., are described from France and Bulgaria, respectively. Additionally, new morphological and morphometric data are provided for two previously described species from Europe, P. brevicauda (Kotlán, 1928) Paramonov, 1952 and P. clavus (von Linstow, 1901) Sudhaus and Fürst von Lieven, 2003. A phylogeny including these four species was inferred from a dataset including 26 ribosomal protein-coding genes, sequences of which are original for P. bulgaricus n. sp. and P. clavus. Relationships support a radiation of all sequenced European Pristionchus species from a single, gonochoristic common ancestor, and current knowledge of species ranges supports “western” and “eastern” clades. Similar diagnostic morphologies reflect the close relationships among the new and recharacterized species, especially P. bulgaricus n. sp., P. brevicauda, and P. clavus, although mating tests as well as genetic and phylogenetic separation support their identities as unique species. Our results show that Pristionchus species in Europe are more diverse than typological characters suggest, and thus biological and molecular profiling will be essential for future delimitation of Pristionchus species from the region.  相似文献

Panorpa similis Esben-Petersen and Panorpa connexa MacLachlan are the most prominent Mecoptera in the Caucasian region. As our knowledge of mating systems and life histories of the different Panorpa species is unsatisfactory, it is the purpose of this contribution to add some interesting information on the life histories and mating behaviour of these two species. Further, we present results on successful breeding of these species, both in the field and the laboratory. Our data suggest that both species are monovoltine under the natural conditions in the Caucasian region, although our breeding experiments revealed a cryptic photoperiodic responsibility in a certain percentage of individuals of Panorpa connexa, which is completely absent in Panorpa similis. First, results on temporally and spatially differing foraging manners of the two species are presented. Male mating tactics of both species comprise copulations with one salivary mass as nuptial gift or copulations without any nuptial gift, but always without the use of force. This paper also gives information on egg deposition, egg hatching and larval development. Finally, we compare aspects of the mating systems for both species with those that have already been investigated for other Panorpa species.  相似文献

The southern cattle tick, Rhipicephalus microplus, is the ixodid species causing the largest economic losses in tropical agrosystems because of its recurrent invasive success, explosive demography on bovine herds, vector competence for diverse pathogens and frequent development of acaricide resistance. Its ecology and the physiological bases of the acaricide resistances it developed, as well as alternative tick control measures, have been intensively studied for decades. By contrast, the tick population genetic structure and its remarkable ability to quickly adapt to new environments have not yet received much attention. We investigated such issues using population genetics analyses in the recently invaded island New Caledonia. In this paper we aim to describe some guidelines for acarologists willing to investigate the processes at play in Acari invasions. Particular emphasis is given to the accuracy of sampling designs and sampling scales for population genetics to be actually informative on the demographical processes of the species (i.e., its mating rules, the determinants of population limits, population sizes, the relationships between genetic exchanges and geographical distances and relevant ecological factors).  相似文献

Obligate endoparasitic oomycetes are known to ubiquitously occur in marine and freshwater diatoms, but their diversity is still largely unexplored. Many of these parasitoids are members of the early-diverging oomycete lineages (Miracula, Diatomophthora), others are within the Leptomitales of the Saprolegniomycetes (Ectrogella, Lagenisma) and some have been described in the Peronosporomycetes (Aphanomycopsis, Lagenidium). Even though some species have been recently described and two new genera were introduced (Miracula and Diatomophthora), the phylogeny and taxonomy of most of these organisms remain unresolved. This is contrasted by the high number of sequences from unclassified species, as recently revealed from environmental sequencing, suggesting the presence of several undiscovered species. In this study, a new species of Miracula is reported from a marine centric diatom (Minidiscus sp.) isolated from Skagaströnd harbor in Northwest Iceland. The morphology and life cycle traits of this novel oomycete parasite are described herein, and its taxonomic placement within the genus Miracula is confirmed by molecular phylogeny. As it cannot be assigned to any previously described species, it is introduced as Miracula islandica in this study. The genus Miracula thus contains three described holocarpic species (M. helgolandica, M. islandica, M. moenusica) to which likely additional species will need to be added in the future, considering the presence of several lineages known only from environmental sequencing that clustered within the Miracula clade.  相似文献

Acacia caven (Mol.) Mol. is native to South America. The species is a leguminous, woody small tree that is considered to have certain potential as a managed silvopastoral crop. Six varieties have been described for the species based on both morphological traits and molecular markers. Little information is available on its mating system. The main objectives of this work were to test the hypothesis that A. caven is an outcrosser and to estimate parameters of its mating system and population structure on the basis of isozyme markers. In the four populations studied, a high homozygote excess was found in the progeny population but not in the mother plant genotypes. The estimate for the multi-locus outcrossing rate (t m) was high (??0.957) in all populations, indicating that Acacia caven is a predominantly outcrosser species. The results of genetic structure analysis within each population indicated that differences in allelic frequencies among families in all of the populations studied are highly significant. The difference in F estimates between progeny and mother plants suggests some selection favouring heterozygotes between the seedling and adult stages. Therefore, a strategy for ex situ conservation might emphasise sampling more populations with a relative large number of trees per site.  相似文献

Almost three-quarters of a century ago, Ernst Mayr (1950) shocked paleoanthropologists into an exceptionalist taxonomic minimalism that lingers today. As a result, a constant stream of new fossil discoveries has been shoehorned wherever possible into existing taxa, almost irrespective of morphology. This compression has been especially true at the level of the genus, where virtually all except very early hominins have over the last couple of decades been classified and named according to the unconscious algorithm, “if it isn’t Australopithecus, it is Homo” (and vice versa). This has led to a vastly overstuffed genus Homo, which has lost any phylogenetic or morphological coherence. Genera are monophyletic clusters of species; and although their boundaries in terms of species content are arbitrary, they do contain phylogenetic structure that ultimately needs to be expressed. Homininae is a large and diverse taxon; and this essay is a plea to paleoanthropologists not only to reflect the phylogenetic structure within it by employing an adequate number of generic and species names, but at the same time to acknowledge the internationally agreed rules (especially of priority) that govern the naming of zoological taxa.  相似文献

Drosophila hydei mating system characteristics were studied and compared to what has been observed for two other Drosophila species. Hydei females will copulate when they are 3 days old, while males do not exhibit courtship behaviour until they are 9 days old. Unlike any other Drosophila, D. hydei females will re-mate as often as four times in one morning. However, re-mating in the same morning does not increase the number of progeny a female produces. Male D. hydei appear to deal with the continual receptivity of females and the pressures of sperm competition by passing less material to any given female but maintaining a constant level of fertility across numerous successive copulations. D. hydei is a cosmopolitan species which utilizes a wide variety of resources. As in D. melanogaster, another cosmopolitan species which is not closely related to it, male success appears to be dependent upon genetic quality only. This pattern differs from that observed in its relative D. mojavensis, a cactiphilic species endomic to the Sonoran desert, in which males contribute material benefits to females in their ejaculate.  相似文献

Paecilomyces variotii is a common cosmopolitan species that is able to spoil various food- and feedstuffs and is frequently encountered in heat-treated products. However, isolates from heat-treated products rarely form ascospores. In this study we examined by using molecular techniques and mating tests whether this species can undergo a sexual cycle and form ascospores. The population structure of this species was examined by analyzing the nuclear ribosomal internal transcribed spacer 1 (ITS1) and ITS2 and the 5.8S rRNA gene, as well as partial β-tubulin, actin, and calmodulin gene sequences. Phylogenetic analyses revealed that P. variotii is a highly variable species. Partition homogeneity tests revealed that P. variotii has a recombining population structure. In addition to sequence analyses, mating experiments indicated that P. variotii is able to form ascomata and ascospores in culture in a heterothallic manner. The distribution of MAT1-1 and MAT1-2 genes showed a 1:1 ratio in the progeny of the mating experiments. From the sequence analyses and mating data we conclude that P. variotii is the anamorph of Talaromyces spectabilis and that it has a biallelic heterothallic mating system. Since Paecilomyces sensu stricto anamorphs group within Byssochlamys, a new combination Byssochlamys spectabilis is proposed.  相似文献

Cryptococcus neoformans is a human fungal pathogen that exists as three distinct varieties or sibling species: the predominantly opportunistic pathogens C. neoformans var. neoformans (serotype D) and C. neoformans var. grubii (serotype A) and the primary pathogen C. neoformans var. gattii (serotypes B and C). While serotypes A and D are cosmopolitan, serotypes B and C are typically restricted to tropical regions. However, serotype B isolates of C. neoformans var. gattii have recently caused an outbreak on Vancouver Island in Canada, highlighting the threat of this fungus and its capacity to infect immunocompetent individuals. Here we report a large-scale analysis of the mating abilities of serotype B and C isolates from diverse sources and identify unusual strains that mate robustly and are suitable for further genetic analysis. Unlike most isolates, which are of both the a and α mating types but are predominantly sterile, the majority of the Vancouver outbreak strains are exclusively of the α mating type and the majority are fertile. In an effort to enhance mating of these isolates, we identified and disrupted the CRG1 gene encoding the GTPase-activating protein involved in attenuating pheromone response. crg1 mutations dramatically increased mating efficiency and enabled mating with otherwise sterile isolates. Our studies provide a genetic and molecular foundation for further studies of this primary pathogen and reveal that the Vancouver Island outbreak may be attributable to a recent recombination event.  相似文献

To assess the potential for mating in several Fusarium species with no known sexual stage, we developed degenerate and semidegenerate oligonucleotide primers to identify conserved mating type (MAT) sequences in these fungi. The putative α and high-mobility-group (HMG) box sequences from Fusarium avenaceum, F. culmorum, F. poae, and F. semitectum were compared to similar sequences that were described previously for other members of the genus. The DNA sequences of the regions flanking the amplified MAT regions were obtained by inverse PCR. These data were used to develop diagnostic primers suitable for the clear amplification of conserved mating type sequences from any member of the genus Fusarium. By using these diagnostic primers, we identified mating types of 122 strains belonging to 22 species of Fusarium. The α box and the HMG box from the mating type genes are transcribed in F. avenaceum, F. culmorum, F. poae, and F. semitectum. The novelty of the PCR-based mating type identification system that we developed is that this method can be used on a wide range of Fusarium species, which have proven or expected teleomorphs in different ascomycetous genera, including Calonectria, Gibberella, and Nectria.  相似文献

Evolutionary dynamics at the population level play a central role in creating the diversity of life on our planet. In this study, we sought to understand the origins of such population-level variation in mating systems and defensive acylsugar chemistry in Solanum habrochaites—a wild tomato species found in diverse Andean habitats in Ecuador and Peru. Using Restriction-site-Associated-DNA-Sequencing (RAD-seq) of 50 S. habrochaites accessions, we identified eight population clusters generated via isolation and hybridization dynamics of 4–6 ancestral populations. Detailed characterization of mating systems of these clusters revealed emergence of multiple self-compatible (SC) groups from progenitor self-incompatible populations in the northern part of the species range. Emergence of these SC groups was also associated with fixation of deleterious alleles inactivating acylsugar acetylation. The Amotape-Huancabamba Zone—a geographical landmark in the Andes with high endemism and isolated microhabitats—was identified as a major driver of differentiation in the northern species range, whereas large geographical distances contributed to population structure and evolution of a novel SC group in the central and southern parts of the range, where the species was also inferred to have originated. Findings presented here highlight the role of the diverse ecogeography of Peru and Ecuador in generating population differentiation, and enhance our understanding of the microevolutionary processes that create biological diversity.  相似文献

Mating systems have broad impacts on how sexual selection and mate choice operate within a species, but studies of mating behavior in the laboratory may not reflect how these processes occur in the wild. Here, we examined the mating behavior of the neotropical butterfly Heliconius erato in the field by releasing larvae and virgin females and observing how they mated. H. erato is considered a pupal‐mating species (i.e., males mate with females as they emerge from the pupal case). However, we observed only two teneral mating events, and experimentally released virgins were almost all mated upon recapture. Our study confirms the presence of some pupal‐mating behavior in H. erato, but suggests that adult mating is likely the prevalent mating strategy in this species. These findings have important implications for the role of color pattern and female mate choice in the generation of reproductive isolation in this diverse genus.  相似文献

Three new species of Nothacrobeles are described from localities in the Mojave Desert, southern California. Nothacrobeles triniglarus n. sp. is characterized by the presence of a long post-vulval sac and three tubular adoral projections. Both N. spatulatus n. sp. and N. nanocorpus n. sp. are smaller than any other known species within the genus. Nothacrobeles spatulatus n. sp. has labial probolae that are short and spatulate without a basal ridge, whereas those of N. nanocorpus n. sp. are flattened and plate-like. Furthermore, N. nanocorpus n. sp. is unique by its extremely short esophageal corpus (less than 25 µm long in adult females) and the small size of its guard processes. An emended diagnosis of the genus is given to accommodate distinctive characteristics of these new species. A table comparing the 11 valid species of Nothacrobeles is presented.  相似文献

Aster tianmenshanensis G. J. Zhang & T. G. Gao, a new species of Asteraceae from southern China is described and illustrated based on evidence from morphology, micromorphology and molecular phylogeny. The new species is superficially similar to Aster salwinensis Onno in having rosettes of spatulate leaves and a solitary, terminal capitulum, but it differs by its glabrous leaf margins, unequal disc floret lobes and 1-seriate pappus. The molecular phylogenetic analysis, based on nuclear sequences ITS, ETS and chloroplast sequence trnL-F, showed that the new species was nested within the genus Aster and formed a well supported clade with Aster verticillatus (Reinw.) Brouillet et al. The new species differs from the latter in having unbranched stems, much larger capitula, unequal disc floret lobes, beakless achenes and persistent pappus. In particular, A. tianmenshanensis has very short stigmatic lines, only ca. 0.18 mm long and less than 1/3 of the length of sterile style tip appendages, remarkably different from its congeners. This type of stigmatic line, as far as we know, has not been found in any other species of Aster. The very short stigmatic lines plus the unequal disc floret lobes imply that the new species may have a very specialized pollination system, which may be a consequence of habitat specialization. The new species grows only on the limestone cliffs of Mt. Tianmen, Hunan Province, at the elevation of 1400 m. It could only be accessed when a plank walkway was built across the cliffs for tourists. As it is known only from an area estimated at less than 10 km2 and a walkway passes through this location, its habitat could be easily disturbed. This species should best be treated as Critically Endangered based on the International Union for Conservation of Nature Red List Categories and Criteria B2a.  相似文献
