Decontamination treatments of burning and biocide application, alone and in combination with tillage, were evaluated for their ability to reduce populations of bacteria applied to the leaves of plants in field plots. In addition, the effects of these control methods on indigenous leaf and soil bacteria and fungi were assessed. Field plots of bush beans (Phaseolus vulgaris), sprayed with the bacterium Pseudomonas syringae, Pseudomonas fluorescens, or Erwinia herbicola, received the following treatments: (i) control, (ii) tillage, (iii) burning, (iv) burning plus tillage (burn-tillage), (v) Kocide (cupric hydroxide), (vi) Kocide plus tillage, (vii) Agri-Strep (streptomycin sulfate), and (viii) Agri-Strep plus tillage. Leaves and soil from the plots were sampled at 1 day before and at 1, 3, 7, 10, 14, 21, and 30 days after application of the decontamination treatments. The burn and burn-tillage treatments produced the most significant reductions in bacterial populations. The Agri-Strep treatment was more effective than the Kocide treatment in eliminating applied bacteria, but neither biocide produced consistent or persistent control. In contrast, the tillage treatment, alone or in combination with the Agri-Strep or Kocide treatments, had a short-term stimulatory effect and increased populations of applied bacteria and also levels of indigenous fungi and bacteria. Agri-Strep and Kocide treatments caused significant reductions in indigenous bacterial populations up to 14 days after application and in indigenous fungal populations on day 7 after application. Our results suggest that conventional plant disease control methods may not provide satisfactory control of genetically engineered microorganisms and indicate a need for further development of effective and selective methods to control release microorganisms at field sites.  相似文献

Density counts were taken for 37 plant species on 12 plots which were assigned to three treatments: spring burn, autumn burn, and control. Post-fire sampling was carried out at intervals of up to 6 years when a second fire treatment was applied, followed by further sampling at 1 year. Most pre-fire species returned by 12 months after fire. Initial recovery was slower on the autumn burn plots and these were surpassed after 12 months by the spring burn plots which remained significantly more diverse until 6 years after fire when the treatments converged. The second fire led to further loss of species, especially on the autumn burn plots. Overall, numbers of individuals were lower in the post-fire community and did not fully recover during the 6 years before the second fire. Of the 37 species followed over the study period, eight had population numbers consistently below pre-fire values, seven showed better recovery on the autumn burn plots, eight showed better recovery on the spring burn plots, and fourteen had population numbers equal to or greater than pre-fire values. Most of the species not fully recovering had relatively poor survival rates. The better recovery rate of species on the autumn burn plots was attributable to better recruitment while the species recovering on the spring burn plots showed better survival and recruitment. The species increasing their numbers after the fire did so through good recruitment or survival, or both. For some species post-fire rainfall or temperature was significantly correlated with greater post-fire recruitment; others were more strongly affected by the treatment itself or more directly by the somewhat different fire intensities in spring and autumn, the differences in soil and litter moisture content, or seasonal variations in soil seed populations.  相似文献

Rangeland degradation is a serious problem in semiarid Africa. Extensive areas of bare, compacted, nutrient‐poor soils limit the productivity and biodiversity of many areas. We conducted a set of restoration experiments in which all eight combinations of soil tilling, fertilization, and seeding with native perennial grasses were carried out in replicated plots. After 6 months, little aboveground biomass was produced in plots without tilling, regardless of seeding or fertilization. Tilling alone tripled plant biomass, mostly of herbaceous forbs and annual grasses. Perennial grasses were essentially limited to plots that were both tilled and seeded. The addition of fertilizer had no significant additional effects. After 7 years, vegetation had declined, but there were still large differences among treatments. After 10 years, one tilled (and seeded) plot had reverted to bare ground, but the other tilled plots still had substantial vegetation. Only one seeded grass (Cenchrus ciliaris) was still a contributor to total cover after 10 years. We suggest that restoration efforts on these soils be directed first to breaking up the surface crust, and second to the addition of desirable seed. A simple ripping trial inspired by this experiment showed considerable promise as a low‐cost restoration technique.  相似文献

The microbial dynamics expressed in terms of culturable microbial populations i.e. bacteria, fungi, actinomycetes and Azotobacter were measured after 33 years of continuous application of mineral fertilizers and amendments to an acid alfisol. The bacterial, fungal and Azotobacter populations were maximum in plots treated with mineral fertilizers and FYM (100%NPK+FYM) while actinomycetes population was maximum in mineral fertilizes and lime treated plots (100%NPK+Lime). The bacterial population decreased and fungal population increased with increasing levels of NPK i.e. from 50% to 150%NPK. Bacillus species of bacteria and Gliocladium, Aspergillus and Rhizopus species of fungi were the main dominating culturable microorganisms in all the treatments. The FYM and lime amended plots sustained crop productivity and microbial populations at higher levels than rest of the mineral fertilizer treatments. The nitrogenous fertilizers alone had the most deleterious effect on crop productivity and the biological soil environment.  相似文献

In many tropical regions, slash‐and‐burn agriculture is considered as a driver of deforestation; the forest is converted into agricultural land by cutting and burning the trees. However, the fields are abandoned after few years because of yield decrease and weed invasion. Consequently, new surfaces are regularly cleared from the primary forest. We propose a reclamation strategy for abandoned fields allowing and sustaining re‐cultivation. In the dry region of south‐western Madagascar, we tested, according to a split‐plot design, an alternative selective slash‐and‐burn cultivation technique coupled with compost amendment on 30–year‐old abandoned fields. Corn plants (Zea mays L.) were grown on four different types of soil amendments: no amendment (control), compost, ashes (as in traditional slash‐and‐burn cultivation), and compost + ashes additions. Furthermore, two tree cover treatments were applied: 0% tree cover (as in traditional slash‐and‐burn cultivation) and 50% tree cover (selective slash‐and‐burn). Both corn growth and soil fertility parameters were monitored during the growing season 2015 up to final harvest. The amendment compost + ashes strongly increased corn yield, which was multiplied by 4–5 in comparison with ashes or compost alone, reaching 1.5 t/ha compared to 0.25 and 0.35 t/ha for ashes and compost, respectively. On control plots, yield was negligible as expected on these degraded soils. Structural equation modeling evidenced that compost and ashes were complementary fertilizing pathways promoting soil fertility through positive effects on soil moisture, pH, organic matter, and microbial activity. Concerning the tree cover treatment, yield was reduced on shaded plots (50% tree cover) compared to sunny plots (0% tree cover) for all soil amendments, except ashes. To conclude, our results provide empirical evidence on the potential of recultivating tropical degraded soils with compost and ashes. This would help mitigating deforestation of the primary forest by increasing lifespan of agricultural lands.  相似文献

The native Escherichia coli aspartate transcarbamoylase (ATCase, E.C. 2.1.3.2) provides a classic allosteric model for the feedback inhibition of a biosynthetic pathway by its end products. Both E. coli and Erwinia herbicola possess ATCase holoenzymes which are dodecameric (2(c3):3(r2)) with 311 amino acid residues per catalytic monomer and 153 and 154 amino acid residues per regulatory (r) monomer, respectively. While the quaternary structures of the two enzymes are identical, the primary amino acid sequences have diverged by 14 % in the catalytic polypeptide and 20 % in the regulatory polypeptide. The amino acids proposed to be directly involved in the active site and nucleotide binding site are strictly conserved between the two enzymes; nonetheless, the two enzymes differ in their catalytic and regulatory characteristics. The E. coli enzyme has sigmoidal substrate binding with activation by ATP, and inhibition by CTP, while the E. herbicola enzyme has apparent first order kinetics at low substrate concentrations in the absence of allosteric ligands, no ATP activation and only slight CTP inhibition. In an apparently important and highly conserved characteristic, CTP and UTP impose strong synergistic inhibition on both enzymes. The co-operative binding of aspartate in the E. coli enzyme is correlated with a T-to-R conformational transition which appears to be greatly reduced in the E. herbicola enzyme, although the addition of inhibitory heterotropic ligands (CTP or CTP+UTP) re-establishes co-operative saturation kinetics. Hybrid holoenzymes assembled in vivo with catalytic subunits from E. herbicola and regulatory subunits from E. coli mimick the allosteric response of the native E. coli holoenzyme and exhibit ATP activation. The reverse hybrid, regulatory subunits from E. herbicola and catalytic subunits from E. coli, exhibited no response to ATP. The conserved structure and diverged functional characteristics of the E. herbicola enzyme provides an opportunity for a new evaluation of the common paradigm involving allosteric control of ATCase.  相似文献

Mowing and burning of emergent vegetation were evaluated as potential management strategies for the control of the malaria vector, Anopheles vestitipennis, in northern Belize, Central America. The primary aim was reduction of tall dense macrophytes (dominated by Typha domingensis) as preferred larval habitat for An. vestitipennis. Nine experimental plots were established in a Typha marsh in Orange Walk District, Belize. Three plots were burned, three were treated by subaquatic mowing, and three were unaltered controls. After treatment, Typha height was most dramatically affected by the mow treatment. Plant heights at 21 and 95 days post-treatment reflected an 89% and 48% decrease, respectively, compared to pretreatment conditions. The Typha height in the burn plots was not as severely affected. Heights at 21 days post-treatment were 39% lower than those of pre-treatment vegetation, with a return to near pre-test heights by 95 days post-treatment. Both treatments resulted in a significant reduction in the number of An. vestitipennis larvae collected as compared to control plots. Conversely, the treatments resulted in increased larval densities of several other vector and pest mosquito species. Larval population densities ofAn. albimanus, Ochlerotatus taeniorhynchus, and Culex coronator were significantly higher in burn plots. In mow plots, there were significant increases in An. albimanus and Oc. taeniorhynchus larval populations. Non-target invertebrate species affected by the treatments were adult Tropisternus collaris, larval Corythrella, and adult Parapleapuella.  相似文献

The effect of three different release rates (1×, 10×, and 20× the recommended rate of 25,000/ha) of Eretmocerus eremicus Rose and Zolnerowich on Bemisia tabaci (Gennadius) populations found in open-field cantaloupe, Cucumis melo L., was evaluated against populations in untreated control plots. Parasitoids were released from a point source in the center of each of nine treatment plots. Whitefly population growth, encompassing all developmental stages, and rates of parasitism were monitored within a 10-m annulus surrounding the center point in all 12 plots over a 52-d period. The rates of B. tabaci population increase during this time were equivalent regardless of the parasitoid release rate. Whitefly densities were not limited in any of our treatment plots when compared to those found in the control plots. Moreover, mean rates of parasitism did not increase with time nor did they differ among the three treatments or control plots (7.9 ± 6.5%). Finally, estimated rates of parasitism were density-dependent responding positively to increasing host numbers. The ineffectiveness of this parasitoid in controlling whitefly populations in the field may be due to its high propensity to disperse at low host densities or to influxes of immigrating whiteflies. Hence, the use of E. eremicus alone is not an efficient means to reduce whitefly populations in melon crops in the southwestern United States.  相似文献

Three crops of cucumber were grown in succession in beds by use of trickle irrigation, plastic film mulch, and soil chemical treatments over a 17-month period, including a fallow winter season. Total yield for the three crops was highest (1208 quintals/ha) in film-mulched plots treated with MBR-CP, and next-highest in film-mulched plots treated with DD-MENCS (1094 quintals/ha); total yield was only 456 quintals/ha in film-mulched control (untreated) plots. Yield in untreated film-mulched plots was 256% of that in untreated unmulched plots (178 quintals/ha). Plant growth and yields were greatest when populations of nematodes and soil-borne fungi were suppressed to very low levels. The residual control by soil treatments lasted longest on Meloidogyne incognita and Fusarium solani.  相似文献

Questions: What influence does mechanical mastication and other fuel treatments have on: (1) canopy and forest floor response variables that influence understory plant development; (2) initial understory vegetation cover, diversity, and composition; and (3) shrub and non‐native species density in a second‐growth ponderosa pine forest. Location: Challenge Experimental Forest, northern Sierra Nevada, California, USA. Methods: We compared the effects of mastication only, mastication with supplemental treatments (tilling and prescribed fire), hand removal, and a control on initial understory vegetation response using a randomized complete block experimental design. Each block (n=4) contained all five treatments and understory vegetation was surveyed within 0.04‐ha plots for each treatment. Results: While mastication alone and hand removal dramatically reduced the midstory vegetation, these treatments had little effect on understory richness compared with control. Prescribed fire after mastication increased native species richness by 150% (+6.0 species m2) compared with control. However, this also increased non‐native species richness (+0.8 species m2) and shrub seedling density (+24.7 stems m2). Mastication followed by tilling resulted in increased non‐native forb density (+0.7 stems m2). Conclusions: Mechanical mastication and hand removal treatments aided in reducing midstory fuels but did not increase understory plant diversity. The subsequent treatment of prescribed burning not only further reduced fire hazard, but also exposed mineral soil, which likely promoted native plant diversity. Some potential drawbacks to this treatment include an increase of non‐native species and stimulation of shrub seed germination, which could alter ecosystem functions and compromise fire hazard reduction in the long‐term.  相似文献

土壤线虫在农田生态系统中数量丰富且对土壤环境变化敏感, 可用于评估不同田间管理条件下的土壤健康。本文探究了紫色土区长期不同施肥措施及土壤团聚体粒径对线虫群落的分布及其生态功能多样性的影响。试验设置了5个施肥处理: 不施肥(对照, CK)、单施化肥(NPK)、生物炭 + 化肥(BCNPK)、商品猪粪 + 化肥(OMNPK)和秸秆 + 化肥(RSDNPK)。团聚体粒径分为: 原状土(BS)、大团聚体(> 2 mm; LA)和小团聚体(0.25-2 mm; SA)。结果表明, 与对照相比, 施肥可促进线虫数量增长, 其中单施化肥处理下增幅最小(66%); 有机物料与化肥配施对线虫数量的提升更为显著, 秸秆 + 化肥处理下增幅达206%。不同施肥处理间线虫类群相对丰度差异显著, 大小均表现为: 食细菌线虫(BA) > 杂食/捕食线虫(OP) > 植食线虫(PP) > 食真菌线虫(FU)。小团聚体较其他土壤团聚体的杂食/捕食线虫丰度更低, 食细菌线虫丰度较高。秸秆与化肥配施处理下线虫群落结构指数和富集指数显著增加, 且各施肥处理下线虫功能足迹呈现明显差异。有机肥与化肥配施(尤其是秸秆 + 化肥)可提高土壤养分供应且有利于形成稳定健康的土壤生态系统, 助推区域农业的可持续发展。  相似文献

The efficacy and cost of reduced release rates of the parasitoid Eretmocerus eremicus Rose and Zolnerowich (Hymenoptera: Aphelinidae) when combined with application of the insect growth regulator buprofezin were compared to those of a higher parasitoid release rate used alone for whitefly control (Homoptera: Aleyrodidae) on poinsettia (Euphorbia pulcherrima Willd. ex Koltz.). The trial was conducted in seven greenhouses in Methuen, Massachusetts from August through December 1997 and employed commercial poinsettia production practices. Two whiteflies species, Trialeurodes vaporariorum (Westwood) and Bemisia argentifolii Bellows and Perring (= Bemisia tabaci [Gennadius] strain B), were present. Three treatments were examined: (1) E. eremicus used alone at a release rate of three females per plant per week (two greenhouses); (2) E. eremicus at an intermediate release rate of two females per plant per week, combined with mid-season use of buprofezin (two applications, spaced 1 week apart, applied in weeks 9 and 10) (two greenhouses); and (3) E. eremicus at a low release rate of one female per plant per week, combined with mid-season use of buprofezin, applied as in treatment 2 (two greenhouses). In addition, observations were made in one additional greenhouse at the site, in which the grower used pesticides for whitefly control. Prior to the start of the trial, cuttings used for all treatments experienced some pesticide use, first abamectinduring rooting and later buprofezin at potting to reduce whitefly numbers, which were initially very high. At harvest, densities of live whitefly nymphs were not statistically different among the biological control treatments, indicating that a low parasitoid release rate combined with buprofezin was as effective as a higher release rate of the parasitoid used alone. Nymphal densities in separate market samples (based on smaller sample sizes) showed differences among treatments, but all treatments, including the low parasitoid release rate + buprofezin maintained densities of live nymphs + pupae at or below approximately two per leaf, a level commercially acceptable in local markets. Control costs per single-stemmed poinsettia plant were $1.18 for the high parasitoid release treatment, $0.75 for the treatment of weekly releases of two female parasitoids per plant per week + buprofezin, $0.38 for the treatment of releases of one female parasitoid per plant per week + buprofezin, and $0.14 for the chemical control greenhouse.  相似文献

Factors governing the rate and direction of prairie dog (Cynomys spp.) colony expansion remain poorly understood. However, increased knowledge and ability to control these factors may lead to more effective reintroductions of prairie dogs and restoration of grassland habitats. We present density and directional analyses of the establishment of new burrows on three reintroduced colonies of Black‐tailed prairie dog (Cynomys ludovicianus) in southern New Mexico; the study colonies had been subjected to mow and burn treatments in the second year of the study. Our hypotheses were that prairie dogs will preferentially dig new burrows in the treatment plots versus control plots and that the colonies will expand in the direction of the treatment plots. The results support these hypotheses; analysis of burrow counts by site and treatment shows that prairie dogs preferentially colonized both mow and burn treatments compared to untreated areas at the periphery of the colonies. Directional analysis showed a significant posttreatment orientation of new burrows toward the treatment plots for all colonies. Our results show that the direction of expansion of prairie dog colonies can be manipulated. Effective control of the expansion of prairie dog colonies may lead to more successful reintroductions.  相似文献

The effect of different dosages of thymol alone, iprodione alone and combinations of thymol and iprodione on white rot disease of garlic and its impact on soil microbial community structure were investigated under greenhouse conditions. Thymol alone or in combination with the fungicide iprodione did not appear to reduce either white rot incidence or soil sclerotia density as compared to an infected control. However, iprodione alone or in combination with thymol reduced soil fungal biomass. In addition, iprodione alone decreased soil microbial activity as estimated by fluorescein diacetate (FDA). Soil bacterial community structure as estimated by phospholipid fatty acid (PLFA) profiles was also was affected by both thymol and iprodione applications. The correlation biplot of the individual PLFAs and biocide treatment indicated that the treatments with thymol alone increased cyclopropyl fatty acid (cy17:0 and cy19:0), while the treatments with iprodione alone increased some saturated and branched fatty acids (principally i16:0, a15:0 and 18:0). In addition, taking into account PLFA biomarkers, thymol applications reduced Gram-negative bacteria in soil. To our knowledge, this research is the first report about the effect of a monoterpene (thymol) on soil microflora.  相似文献

Understanding the effects of disturbances such as fire on plant demography helps elucidate the mechanisms that cause changes in community composition. I studied the effects of spring and fall fires on Silene spaldingii, an endangered perennial herb of grasslands in northwest Montana. Individual S. spaldingii plants were mapped, and size and flowering were recorded for 1 yr prior and 5 yr subsequent to the burn treatments. Enhanced seedling recruitment (70–410%) and a 22% increase in population size were the principal effects of fire on S. spaldingii, and fall burn plots had lower recruitment than spring burn plots. These effects were apparent for 2–3 yr following the treatments. Fire had no detectable effect on the survival of adults or recruits of S. spaldingii. Silene spaldingii exhibits prolonged dormancy in which plants do not produce aboveground vegetation for one to several consecutive years. Results suggest that fire has a positive effect on the population dynamics of S. spaldingii by removing litter and creating safe sites for recruitment. Prescribed fire should be an important tool for managing populations of this rare plant.  相似文献

Multiple-species natural enemy approach for the biological control of the alfalfa snout beetle, Otiorhynchus ligustici (L.) (Coleoptera: Curculionidae), was compared with using single-species of natural enemies in the alfalfa ecosystem by using entomopathogenic nematodes with different dispersal and foraging behaviors. Steinernema carpocapsae NY001 (ambush nematode), Heterorhabditis bacteriophora Oswego (cruiser nematode), and Steinernema feltiae Valko (intermediate nematode) were applied in single-species, two-species combinations, and one three-species combination treatments at 2.5 x 10(9) infective juveniles per hectare. All nematode species persisted for a full year (357 d). S. carpocapsae NY001 protected the plants from root-feeding damage better than H. bacteriophora Oswego but allowed for higher larval survival than all other nematode treatments. S. feltiae Valko protected the plants better than H. bacteriophora Oswego and controlled alfalfa snout beetle larvae better than S. carpocapsae NY001. H. bacteriophora Oswego allowed for similar root damage compared with control plots but reduced larval populations better than S. carpocapsae NY001. The combination of S. carpocapsae NY001 and H. bacteriophora Oswego provided significantly better protection for the plants than the control (unlike H. bacteriophora Oswego alone) and reduced host larva survival more than S. carpocapsae NY001 alone. The combination S. feltiae Valko and H. bacteriophora Oswego could not be statistically separated from the performance of S. feltiae Valko applied alone.  相似文献

The study aimed to evaluate the effect of cow urine and combination of antioxidants against lindane induced oxidative stress in Swiss mice. Male healthy mice, 8–10 weeks old, weighing 30 ± 5 g were randomly selected and divided into eight groups, namely, control (C); lindane (L); antioxidant (A), antioxidant+lindane (A+L), cow urine (U), cow urine+lindane (U+L), cow urine+antioxidants (U+A) and cow urine+antioxidants+lindane (U+A+L). Group C animals were administered only the vehicle (olive oil); doses selected for other treatments were: lindane: 40 mg/kg b.w.; antioxidants: 125 mg/kg b.w. (vitamin C: 50 mg/kg b.w., vitamin E: 50 mg/kg b.w., α-lipoic acid: 25 mg/kg b.w.) and cow urine: 0.25 ml/kg b.w. In group A+L and U+L antioxidants and cow urine were administered 1 h prior to lindane administration and in group U+A and U+A+L cow urine was administered 10 min before antioxidants. All treatments were administered orally continuously for 60 days. Lindane treated group showed increased lipid peroxidation, whereas glutathione, glutathione peroxidase, superoxide dismutase, catalase, protein and endogenous levels of vitamin C and E were significantly decreased compared to control. Administration of cow urine and antioxidants alleviated the levels of these biochemical parameters.  相似文献

The effects of six burning treatments combined with two insecticide treatments of alfalfa (Medicago sativa L.) stubble on alfalfa plant bug, Adelphocoris lineolatus (Goeze) and Lygus spp. populations were evaluated over an 8-yr period (1982–1989). The burn treatments were: burning every spring and autumn, burning in alternate springs, burning at 50–100 mm and 150–200 mm of spring growth, and an unburned control. Alfalfa plant bug, which overwinters in the egg stage in alfalfa stems, were reduced by the spring burns. First generation populations of Lygus spp., were enhanced by the burn treatments before spring growth appeared, but were reduced when alfalfa was burned over 50 mm of spring growth. Counts of the minute pirate bug, Onus tristicolor White, were enhanced in 1983 by all burn treatments, and in 1985 by some of the burn treatments. Prescribed fire has become a common tool of natural area managers for removal of non‐indigenous invasive species and maintenance of barrens plant communities. Certain non‐native species, such as tall fescue (Festuca arundinacea), tolerate fire and may require additional removal treatments. We studied changes in soil N and C dynamics after prescribed fire and herbicide application in remnant barrens in west central Kentucky. The effects of a single spring burn post‐emergence herbicide, combined fire and herbicide treatments, and an unburned no‐herbicide control were compared on five replicate blocks. In fire‐plus‐herbicide plots, fescue averaged 8% at the end of the growing season compared with 46% fescue cover in control plots. The extent of bare soil increased from near 0 in control to 11% in burned plots and 25% in fire‐plus‐herbicide plots. Over the course of the growing season, fire had little effect on soil N pools or processes. Fire caused a decline in soil CO2 flux in parallel to decreased soil moisture. When applied alone, herbicide increased plant‐available soil N slightly but had no effect on soil respiration, moisture, or temperature. Fire‐plus‐herbicide significantly increased plant‐available soil N and net N transformation rates; soil respiration declined by 33%. Removal of non‐native plants modified the chemical, physical, and biological soil conditions that control availability of plant nutrients and influence plant species performance and community composition.  相似文献
