Muscle, liver, kidney and skin samples taken from 78 harbour porpoises (Phocoena phocoena) were analysed for mercury, cadmium, zinc, copper and selenium. The highest concentrations of mercury were found in the liver (geometric mean 4.17 g/g wet weight), whilst the highest concentrations of cadmium were in the kidney (g.m. 13.2 g/g ww). The levels of cadmium were more than ten times higher than in harbour porpoises from the North Sea and the British NW coast, whilst the mercury levels were about the same. The importance of the cadmium content in the prey is discussed, but this attempt did not revealed the differences. Very high levels of zinc (g.m. 359 g/g ww) and selenium (g.m. 28.6 g/g ww) were found in skin samples, respectively seven and ten times more than in liver. A significant correlation was found between age and the level of mercury and cadmium in all organs. The concentration of mercury and selenium in liver and skin samples and of cadmium and zinc in kidney samples were highly correlated.  相似文献

Muscle, liver, and kidney tissues from 38 polar bears (Ursus maritimus) caught in the Scoresby Sound area, Central East Greenland, were analysed for zinc, cadmium, mercury and selenium. In general, cadmium concentrations were low in muscle, liver and kidney tissue, with geometric means (g.m.) of 0.022 (range: <0.015–0.085), 0.841 (range: 0.092–3.29) and 13.1 (range: 1.04–115) g Cd/g wet weight (ww) respectively. This finding can be explained by low cadmium levels in the blubber of ringed seals. The concentration of mercury in muscle tissue was low (g.m. 0.071; range: 0.039–0.193 g Hg/g ww), whereas concentrations in liver and kidney tissue were relatively high (liver: g.m. 7.87; range: 1.35–24.8 g Hg/g ww, and kidney: g.m. 15.2; range: 1.59–66.6 g Hg/g ww). Mercury and cadmium were positively correlated with age in liver and kidney. Zinc was positively correlated with age in kidney, and selenium was correlated with age in liver. Contrary to other marine mammals, polar bears had higher mercury levels in the kidneys than in the liver. In all three tissues polar bears had significantly lower cadmium levels than ringed seals from the same area. Mercury levels were likewise significantly lower in the muscle tissue of polar bears than in ringed seals, whereas levels in the liver and kidney were significantly higher. The previous geographic trend for cadmium and mercury found in Canadian polar bears could be extended to cover East Greenland as well. Hence cadmium levels were higher in Greenland than in Canada, while the opposite was the case for mercury. Greenland polar bears had higher mercury and cadmium contents in livers and kidneys than polar bears from Svalbard. The mercury levels in muscle and liver tissue from polar bears from East Greenland were twice as high as found in bears from western Alaska, but half the levels found in northern Alaska. Cadmium and zinc were partially correlated in kidney tissue, and this was found for mercury and selenium as well. Cadmium and zinc showed molar ratios close to unity with the highest concentrations occurring in kidney tissue, while the levels of zinc exceeded cadmium in muscle and liver tissue by up to several decades. Concentrations of cadmium, mercury, lead, arsenic, selenium, copper, zinc, manganese and iron in liver, and cadmium in kidneys, were analysed in 95 carcasses of arctic fox (Alopex lagopus) caught in Svalbard during three winter seasons from 1984 through 1986. The hepatic concentration ranges of cadmium, mercury, lead and arsenic were 0.1–2.4, 0.01–2.2, < 0.5–2.9 and 0.01–1.3 g·g–1 WW, respectively. The range of cadmium concentration in the kidneys was from 0.2 to 13 g·g–1 WW. Cadmium and mercury concentrations were higher in adult animals than in juveniles. The average concentrations of cadmium and lead were similar to recently published levels in polar bear from Svalbard, but the mercury concentrations were lower. Significant geographical differences were observed between trapping areas. Foxes caught north of Isfjorden had lower levels of liver iron and higher levels of all other elements analysed than those caught south of Isfjorden. Effects of Pb2+, Ni2+, Hg2+ and Se4+ on cultured human glioma U-343MG cells were investigated considering uptake, toxicity and, in combination with radiation, clonogenic cell survival. The cells were exposed to 0-100 m of the metals for a week before the evaluation. The tests showed a tendency to toxicity with 10 m nickel although not significant (P > 0.05). Selenium, lead and mercury exerted a significant toxicity (P < 0.05) at 2.5 m, 10 m and 1 m, respectively. To challenge the clonogenic cell survival capacity, the cells were irradiated with60Co photons after being exposed to the highest nontoxic concentration of the different metals. The clonogenic cell survival tests, after irradiation, showed no significant change if the cells were exposed to 5 m nickel, 0.5 m selenium or 5 m lead compared with those not exposed. Mercury, 0.1 m, gave a relative reduction in survival compared with only irradiated cells of 58 ± 17%. Thus, only mercury affected the radiation-induced damage and/or repair. When exposed to the highest nontoxic concentrations of the different metals, the cultures did not display a significant uptake ratio (metal concentration ratio of exposed cells to control cells) of nickel (3.1 ± 3.3), only a small uptake ratio of selenium (4.0 ± 0.4), while there was a large uptake ratio of both lead (2.6 ± 1.7) x 102 and mercury (1.5 ± 0.2) x 101. The results indicated that nickel was neither especially toxic nor influenced the clonogenic cell survival after irradiation. Mercury was more toxic and also influenced the radiation sensitivity. Lead was taken up strongly but did not influence the radiation sensitivity. Selenium accumulated but gave no detectable effect on the radiation sensitivity.  相似文献

In order to evaluate the ability of the freshwater bivalve Potomida littoralis for use as a biodetector of aquatic pollution, the short-term effects of some pollutants (metals, antifouling product and effluents) on pumping activity of this mussel were studied qualitatively (pumping behaviour) and quantitatively (filtration rate). The method of the flow sensing at the exhalent siphon showed that the normal activity of P. littoralis was characterized by a continuous pattern of pumping with occasional and brief interruptions. When exposed to toxic stress, the exhalent flow immediately shifted to an intermittent pattern with periods of pumping alternating with periods of pause that could last several minutes. The threshold concentrations of heavy metals/antifouling agent inducing this type of activity within 2 h were: 25 g l–1 for copper, 90 g l–1 for cadmium, 5 g l–1 for mercury, 3000 g l–1 for zinc and 700 g l–1 for the antifouling agent Mexel. Both the effluent samples of the urban wastewater of Marrakech city tested were found to be toxic.The filtration rate of control animals ranged from 0.91 to 1.72 l h–1 g–1 animal, while after 3 h exposure to Cu, Cd, Hg and the effluent E1 and E2, filtration rates were decreased by concentrations above the threshold levels set as toxic for the pumping activity.The sensitivity of P. littoralis to pollution and its usefulness as biodetector for rapid toxicity screening is discussed and a procedure for the detection of pollution in Moroccan freshwater is proposed.  相似文献

The present study evaluates the growth response of two strains of filamentous fungi; a Fusarium sp. and Alternaria tenuis, grown on both solid and liquid Czapek Dox medium amended with different concentrations of CdCl2. Colony extension and the mycelial dry weight of both fungi were significantly inhibited by high concentrations of cadmium. Extended lag phases and low growth rates resulted from cadmium administration. Cadmium drastically affected fungal morphogenesis by the production of stunted sterile thick mycelial filaments of the Fusarium sp. and chains of uncharacterized swellings instead of conidia in A. tenuis. Experiments showed that cadmium accumulation by the Fusarium sp. grown in liquid medium was a concentration dependent, and over the incubation time it displayed a plateau pattern. The cells grown on medium containing 0.25 mmol l–1 CdCl2 accumulated up to 89 ± 12 mol Cd (gm dw)–1 after two days, falling to 29 ± 10 mol Cd (gm dw)–1 after five days. At 0.5 mmol l–1 CdCl2 treatment the maximum cellular cadmium content was 132 ± 14 mol (gm dw)–1, attained after 3 days, and decreased to 98 ± 9 mol (gm dw)–1 at the end of the incubation time. There was a simultaneous marked drop in cadmium content and pH of the growth medium during the first few days. The presence of cadmium markedly altered the cellular essential cations; K+ and Mg2+ being decreased while Na+ increased during the growth period. Such findings resulted a reverse pattern of cellular Na+/K+ ratio for cells grown on cadmium-containing medium in respect to the control treatment. The results are discussed in relation to a further dimension of cadmium effects that might reflect its toxicity, as well as the implication of cadmium extrusion for tolerance during fungal growth.  相似文献

Synopsis Foraging behaviour of lake charr, Salvelinus namaycush, exposed to 0.5 and 5.0 g l–1 cadmium concentrations for 106–112 d, was examined in a flow-through stream channel using fathead minnows, Pimephales promelas, as prey. Five modes of foraging behaviour were observed and quantified (ending at fixate-orient; ending at approach and/or chase; ending at failed attack; ending at capture and loss; successful consumption). Each mode represented an increasingly progressive end-point from initial sighting of the prey to final consumption. Compared to controls., 0.5 g l–1 Cd-exposed predators captured significantly fewer prey, while 5 g l–1 Cd-exposed predators made fewer attacks. Both Cd-exposed groups consumed fewer prey than controls.  相似文献

The 22 strains of Emmonsia Ciferri & Montemartini 1959, inoculated intranasally to laboratory mice are not equally virulent. One month after the inoculation, 15 of the strains had produced adiaspores 120 to 190 m in diameter in the lung. Another strain produced adiaspores measuring 44 m and 2 others measuring 20 m or 10 m. The remaining 4 strains did not develop in the lung tissue. Four thermophilic strains, which in vitro have adiaspores measuring 8 to 15 m, had adiaspores reaching 120–180 m in vivo. Neither budding nor endosporulation could be observed in any adiaspore.

Résumé Vingt deux souches d'Emmonsia Ciferri et Montemartini, 1959, inoculées par instillations nasales à des souris de laboratoire, ne présentent pas toutes la même virulence. Un mois après l'inoculation, quinze d'entre elles donnent dans le parenchyme pulmonaire des adiaspores de 120–190 m de diamètre, une souche, des adiaspores de 44 m, une autre, des adiaspores de 20 m et une autre, des adiaspores de 10 m. Les quatre dernières ne se développent pas dans les poumons. Quatre souches thermophiles, donnant in vitro de petites adiaspores de 8–15 m, différencient in vivo des adiaspores de 120–180 m. Aucun phénomène de bourgeonnement ou d'endosporulation n'est observé sur aucune adiaspore.

Water, sediment, red swamp crayfish (Procambarus clarkii) and black bass (Micropterus salmoides) from Lake Naivasha were analyzed for selected organochlorine and organophosphorus pesticide residues. The mean p,p'-DDT, o,p'-DDT and p,p'-DDE residue levels recorded in black bass (28.3 (± 30.0), 34.2 (±54.0) and 16.1 (±16.1) g kg–1, respectively) and crayfish (4.6 (±5.1), 3.2 (±2.8), and 1.4 (±1.1) g kg–1, respectively), were higher than previously recorded. This indicated recent usage of technical DDT in the lake's catchment. Levels of p,p'-DDT, higher than those of p,p'-DDE further emphasized this. Mean lindane, dieldrin, -endosulfan and aldrin concentrations in black bass were 100.5, 34.6, 21.6 and 16.7 g kg–1, respectively. The same residues were detected at lower concentrations in crayfish at 2.0, 2.0, 2.0 and 1.9 g kg–1, respectively. The higher fat content (3.7 ± 2.7% SD) in black bass (compared to 0.6 ± 0.3% in crayfish) accounted for the significantly higher residue concentrations in black bass. Organophosphate pesticides were the most commonly used pesticides in the lake's catchment, but none was detected in any of the samples. The results indicate that there is need for further work to identify sources and fate of pesticide contaminants, as well as to improve monitoring of pesticide use throughout the catchment.  相似文献

Effects of some metals on the growth of cultured human erythroleukemia K-562 cells were investigated when grown in two different types of media based upon RPMI-1640 or Ham's F-10. The study on proliferation, using RPMI-1640 supplemented with sodium selenite, selenomethionine, mercuric chloride, methylmercuric chloride and cadmium nitrate showed no inhibition of growth at concentrations of 2.5, 25, 25, 2.5 and 25 M, while at 75, 250, 50, 5 and 50 M toxicity was apparent. Selenite at 5–50 M and selenomethionine at 50–100 M inhibited the growth. In Ham's F-10 supplemented with the same compounds no inhibition was found at concentrations of 5, 10, 25, 1 and 50 M, while at 50, 100, 50, 5 and 75 M toxic effects were noted. Selenite 10 M and selenomethionine 25-50 M inhibited the proliferation. Measurements of trace element levels in pellets of K-562 cells grown in RPMI-1640 or Ham's F-10 unveiled higher cell contents of cadmium and selenium in cells grown in RPMI-1640, being consistent with higher concentrations of these elements in that medium. Manganese and mercury concentrations were higher in cells grown in Ham's F-10 correlating with a higher medium concentration of these elements. The growth responses and cellular uptake differed between the metals and the selenocompounds and although extrapolating the results to humans is difficult the selenium exposures were in approximately the same order of magnitude as in human exposures. The compounds could be ranked according to decreasing toxicity as: methylmercuric chloride > mercuric chloride, cadmium nitrate, sodium selenite > selenomethionine.  相似文献

Total mercury was measured in different compartments of Lake Xolotlán's (Managua) ecosystemviz., sediments, water, fish and men. Sediments from 18 localities at 5 depths inside the sediment (5, 10, 15, 20 and 25 cm) contained an average concentration of 0.62 g Hg.g–1±0.46 at the surface, with extreme values of 0.16 and 1.8 g.g–1. The highest concentration was observed at 25 cm depth in front of the chlor-alkaly factory (ELPESA). This maximum is associated with the period of highest production of this factory. The highest mercury concentrations in water were also measured close to the discharge of ELPESA,viz. 787 g.Hg–1 in January and 506 g.g–1 in April. The mean mercury concentrations measured in the muscles of the most consumed fish were 0.63 g.g–1±0.22 (extreme values 0.22 and 1.45) inCichlasoma managuense, and 0.07 g.g–1±0.14 (extreme values 0.004 and 0.63) inC. citrinellum. The concentration in the liver was 0.79 g.g–1±1.29 inC. managuense and 0.62 g.g–1±0.44 inC. citrinellum. Human hairs (n=98) of fishermen and their families contained 5.03 g.g–1±6.2 (extreme values 0.02 and 38.22). The mean concentration measured in men was 6.22 g.g–1±6.34 (n=58), and in women 3.39 g.g–1±5.7 (n=40). The average mercury concentration of hairs of workers of ELPESA was 91.24 g.g–1±156.9 (extreme values 0.46 and 724.53; n=32). We conclude that total mercury levels in the various ecosystem compartments are very high and mercury contamination in the lake may be considered as dangerous for human health.  相似文献

The aluminium (Al) tolerance of 34 temperate legume species (143 genotypes, including 57 from Trifolium repens) was determined in 60 experiments over a 3 year period in a low ionic strength (2.7 × 10-3 M) solution culture. For each genotype, the relationship between solution Al3+ activity (M) and relative yield was determined and the Al3+ activity associated with a 50% reduction in yield (AlRY50) calculated. In addition, plant chemical concentrations were determined in at least one genotype from most species. For white clover, AlRY50 over all genotypes had an approximately normal distribution with mean of 1.31 M for the tops and 1.51 M for the roots, and a standard deviation of about 0.4. This suggested that Al tolerance had a polygenic inheritance. For the other species tested, AlRY50 ranged from 0.15 to 4.53 M in the tops and from 0.21 to 4.89 M in the roots. In the tops and roots, 37% and 26% respectively of the genotypes had an AlRY50 less than 1 M, including all species tested in the genera Melilotus and Medicago. Only 8% or 23% of the genotypes, based on the tops and roots respectively, had an AlRY50 greater than 2, including all genotypes in the species Lotus pedunculatus. Except for Lotus, there were no consistent differences between genera in plant chemical concentrations. In Lotus, concentrations of Ca, Zn, Mn and Cu in the tops and of all elements except B in the roots were lower than that of the other species. The AlRY50 of the species was not related to plant chemical concentrations in the absence of Al. Depending on the plant element, increasing solution Al concentrations had no significant effect on plant chemical concentrations for 56–94% of the species. When a significant effect did occur, increasing Al in solution generally decreased S and K concentrations and increased Mn, Zn, Cu Fe, B and Al concentrations in the tops and roots and decreased Ca concentrations in the tops. Plant P concentrations decreased in the tops but increased in the roots. Increasing Al in solution increase plant Al at the average rate of 44 g g-1 M -1 (range 20–87) in the tops and 333 g M -1 (range 162–616) in the roots.  相似文献

Recently, cadmium has been described to disturb ovarian function in rats. In this paper the direct influence of cadmium on steroid production of ovarian cellsin vitro has been studied. Granulosa and luteal cells were obtained from proestrous and pregnant rats, and incubated with 0, 5, 10, 20 or 40 g ml–1 CdCl2 in the presence or absence of 0.1–1000 ng ml–1 follicle stimulating hormone (FSH) or luteinizing hormone (LH) for 24 or 48 h. Production of progesterone (P) and 17-estradiol (E2) by granulosa and that of P by luteal cells were measured by radioimmunoassay. In FSH-stimulated granulosa cell cultures, 5 and 40 g ml–1 CdCl2 suppressed P accumulation to 65 and 10%, respectively; accumulation of E2 (at 5 g ml–1 CdCl2) decreased to 44%. P production of LH-supported luteal cells dropped to 86 and 66%, respectively, when 5 and 40 g ml–1 CdCl2 was added to the medium. No alteration in basal P accumulation occurred in granulosa and luteal cell cultures following incubations with 20 and 40 g ml–1 CdCl2, whereas basal E2 production of granulosa cells was markedly diminished. It is concluded that CdCl2 suppressing steroid synthesisin vitro exerts a direct influence on granulosa and luteal cell function.  相似文献

The increasing effect of regucalcin, isolated from rat liver cytosol, on neutral proteolytic activity in the hepatic cytosol was characterized. The proteolytic activity was markedly elevated by the addition of regucalcin (0.1–0.5 M) in the absence of Ca2+. This increase was not significantly altered by the presence of diisopropylfluorophsophate (DPF;2.5 mM)—although DFP caused a significant decrease in the proteolytic activity. Regucalcin (0.25 M) additively enhanced the dithiothreitol (DTT; 1.0 mM)—increased proteolytic activity, while the regucalcin or DTT effect was completely abolished by NEM (5 mM), indicating that regucalcin may act on the SH group in proteases. Also, regucalcin (0.25 M) enhanced the effect of Ca2+ (10 M) increasing liver proteolytic activity, suggesting that regucalcin does not influence on the active sites for Ca2+ in proteases. Moreover, the proteolytic activity of regucalcin (0.25 M) was significantly decreased by the presence of calpastatin (24 g/ml), an inhibitor of Ca2+-activated neutral protease (calpain). Now, regucalcin (0.25 M) increased about 7-fold the activity ofm-calpain isolated from rabbit skeletal muscle. These observations demonstrate that regucalcin directly activates cysteinyl-proteases. Regucalcin may have a role as a potent proteolytic activator in the cytoplasm of liver cells.  相似文献

Using physiological assays coupled with ultrathin tissue sections, we investigated the impacts of exogenous selenium (Se) on the growth, antioxidant enzymes, osmotic regulation and ultrastructural modifications of leaf mesophyll and root tip cells of 100 mM NaCl-stressed sorrel (Rumex patientia × R. tianshanicus) seedlings. At low concentrations (1–5 M), Se tended to stimulate the growth, the activities of superoxide dismutase and peroxidase enzymes, as well as the accumulation of water-soluble sugar in leaves of sorrel seedlings. At higher concentrations (10–30 M), Se exerted diminished beneficial effects on growth and enzyme activities. CAT activity did not change with Se addition (1–30 M). Electrolyte leakage of leaf cells declined, and K+ and Na+ ions increased in leaves with Se treatment, notably at 5 M of Se. TEM observations revealed that treatment with 5 M of Se positively promoted the integrity of membrane systems and cellular organelles, such as chloroplasts and mitochondria in leaf mesophyll and root tip cells. These results strongly suggest that an appropriate concentration of exogenous Se functions positively to promote the antioxidative and osmoregulatory capacity, and enhance the salt-resistance in sorrel seedlings.  相似文献

Toxicity of 14 selected acaricides was tested for a laboratory strain of femaleD. gallinae. The most toxic were carbaryl (LC50=5.0 g/m2), deltamethrin (LC50=7.8 g/m2), bendiocarb (LC50=11.1g/m2), and permethrin (LC50=12.6 g/m2). High level resistance to DDT inD. gallinae was recorded, with factors of resistance at LC50/ LC90 reaching 57/73.  相似文献

Ninety-eight samples of feeds associated with 44 cases of equine leukoencephalomalacia (ELEM) and 83 samples of feed associated with 42 cases of a porcine pulmonary edema syndrome (PPE) were analyzed for fumonisin B1 (FB1). For comparison purposes, 51 feed samples not associated with PPE or ELEM were also analyzed. Feed associated with ELEM contained FB1 ranging from less than 1 g/g to 126 g/g with 75% of the cases having at least 1 sample above 10 g/g. Feeds associated with PPE ranged from less than 1 g/g to 330 g/g with 71% of the cases having at least 1 sample greater than 10 g/g. Quantitation was by high performance liquid chromatography (HPLC)/fluorescence using the fluorescamine derivative with confirmation by thin layer chromatography (TLC) and/or gas chromatography/mass spectroscopy (GC/MS).  相似文献

The effect of lead on the filtration rate of the zebra musselDreissena polymorpha was investigated, together with the accumulation of Pb in the soft tissues of the mussels. The NOEC-filtration was 116 g.l–1 (0,56 mol.l–1) and the EC50-filtration was 370 g.l–1 (1.79 mol.l–1). The NOEC-accumulation was the concentration found in the control water (1.4g.l–1). These experiments show that the EC50-filtration for Pb is similar to that for Cd, higher than that for Cu and lower than that for Zn. The water quality criteria for lead allow 25 g Pb.l–1 in surface water. This will not cause short-term effects. Long-term effects may, however, occur, since an accumulation of Pb as low as 16 g.l–1 was recorded in this study.  相似文献

Pot culture experiments containing sand and soil, at two levels of mercury (2 and 5 g mL-1) added through irrigation with increasing supplementation of selenium (selenite and selenate) led to a decrease in the uptake of mercury by tomato (Lycopersicum esculentum) plant. Both the forms of selenium (selenite and selenate) were found to be equally effective in reducing the mercury accumulation by plants. The observed reduction pattern of mercury accumulation in plant tissues has been discussed on the basis of the formation of insoluble HgSe complex in soil-root environment.  相似文献

Summary The inactivation of repressor was followed by the specific DNA binding assay during the course of lysogenic induction provoked by incubation at 42°C of an E. coli tif-1 lysogenic strain. The presence of up to 400 g/ml chloramphenicol during the inducing treatment did not impair the loss of repressor binding activity, whilst concentrations of 200 g/ml neomycin and 100 g/ml rifampicin effectively inhibited the inactivation of repressor.Residual protein synthesis in the presence of chloramphenicol, neomycin and rifampicin was 5%, 5% and 27% respectively of that observed in the drug-free control. This residual synthesis did not appear to involve amplification of the X-protein. These results suggest that tif-mediated inactivation of the repressor requires the activation of some specific gene(s), the translation of which appears to be resistant to chloramphenicol.  相似文献
