Two Dirofilaria ursi, six Trichodectes pinguis and numerous Haemaphysalis megaspinosa were obtained from an Asiatic black bear (Ursus thibetanus) shot in Kyushu, Japan. The presence of two parasites specific to black bears may indicate the existence of a wild population, although the bears are regarded to be extinct on Kyushu Island.  相似文献

The Asiatic black bear is one of the most endangered mammals in South Korea owing to population declines resulting from human exploitation and habitat fragmentation. To restore the black bear population in South Korea, 27 bear cubs from North Korea and Russian Far East (Primorsky Krai) were imported and released into Jirisan National Park, a reservoir of the largest wild population in South Korea, in 2004. To monitor the success of this reintroduction, the genetic diversity and population structure of the reintroduced black bears were measured using both mitochondrial and nuclear DNA markers. Mitochondrial D-loop region DNA sequences (615 bp) of 43 Japanese black bears from previous study and 14 Southeast Asian black bears in this study were employed to obtain phylogenetic inference of the reintroduced black bears. The mitochondrial phylogeny indicated Asiatic black bear populations from Russian Far East and North Korea form a single evolutionary unit distinct from populations from Japan and Southeast Asia. Mean expected heterozygosity (H(E)) across 16 microsatellite loci was 0.648 for Russian and 0.676 for North Korean populations. There was a moderate but significant level of microsatellite differentiation (F(ST) = 0.063) between black bears from the 2 source areas. In addition, genetic evidences revealed that 2 populations are represented as diverging groups, with lingering genetic admixture among individuals of 2 source populations. Relatedness analysis based on genetic markers indicated several discrepancies with the pedigree records. Implication of the phylogenetic and genetic evidences on long-term management of Asiatic black bears in South Korea is discussed.  相似文献

将采自23 头成年圈养黑熊的精液,分别用3 种稀释液(Ⅰ:Tris - 乳- 果- 卵;Ⅱ:柠- 葡- 蔗- 卵;Ⅲ:Tris - 柠- 果- 葡- 卵)稀释并在4℃ 下保存,通过检测精液在不同稀释液稀释条件下的保存时间,筛选出最适稀释液用于精液的冷冻保存;从精液解冻后精子的活率、活力、畸形率、顶体完整率4 个指标,分别从3种冷冻保护剂(甘油3%、3.5% 、4% )、两种冷冻方法(两步冷冻法和自动冷冻法)两个方面进行了比较试验。结果表明:精子活力在0.3 以上时,稀释液Ⅲ保存时间为175.42 ± 3.04 h,显著高于稀释液Ⅰ和稀释液Ⅱ(P ﹤0.01),稀释液Ⅱ保存时间也明显高于稀释液Ⅰ (P ﹤0.01);含3.5% 甘油浓度的稀释液解冻后精子活率(41.75 ± 3.46% )、活力(32.63 ±5.27% )和顶体完整率(85.62 ± 4.58% )显著高于其他两组(P ﹤0.01),并且精子畸形率(29.32 ± 8.22% )明显低于其他两组(35.95 ± 8.04% ,36.07 ±7.72% )(P ﹤0.01);采用自动冷冻法冷冻保存圈养黑熊精液,解冻后精子活率、活力和顶体完整率分别为41.75 ±3.46% 、32.63 ± 5.27%和85.62 ±4.58% ,都明显高于两步冷冻法(P ﹤0.01);解冻后畸形率为29.32 ± 8.22% ,明显低于两步冷冻法(P ﹤0.01)。  相似文献

This study details the seminal traits in the rare Asiatic black bear, revealing that this species generally produces high quality, concentrated ejaculates containing sperm of high motility and morphological integrity and similar to other members of the Ursine lineage. Semen was collected by electroejaculation and 23 trials were performed on 18 bears. The mean values were obtained for volume (0.61 ml), pH (7.1), sperm concentration (1049 x 10(6) ml(-1)), total sperm counts (502.8 x 10(6)), motility percentage (63.8%), forward progressive status (3.5), abnormal sperm percentage (37.9%) and intact acrosome percentage (76.6%).  相似文献

We describe primers and polymerase chain reaction conditions to amplify 21 tetranucleotide microsatellite DNA loci in black bears (Ursus americanus). We tested primers using individuals from two populations, one each in Georgia and Florida. Among individuals from Georgia (n = 29), primer pairs yielded an average of 2.9 alleles (range, one to four) and an average observed heterozygosity (HO) of 0.50 (range, 0.00 to 0.79). Among individuals from Florida (n = 19), primer pairs yielded an average of 5.7 alleles (range, one to 14) and an HO of 0.55 (range, 0.00 to 1.00). A comparison of previously developed markers with individuals from Georgia suggests that bear populations in Georgia and Florida have reduced allelic diversity relative to other populations.  相似文献

Two trisaccharides, two tetrasaccharides, one penta-, one hexa-, two hepta-, one deca- and two undeca-saccharides were isolated from several Japanese black bear milk samples by chloroform/methanol extraction, gel filtration and preparative thin-layer chromatography. Relatively little is known about the nutritional ecology of omnivorous Asiatic black bears (Ursus thibetanus) in Nepal. We characterized the diet of black bears in two seasons (June–July, “summer”; and October–November “autumn”) and two study areas (Dhorpatan Hunting Reserve [DHR]; and Kailash Sacred Landscape [KSL]). We then conducted nutritional analysis of species consumed by black bears in each study area, in combination with nutritional estimates from the literature, to estimate the proportions of macronutrients (i.e., protein [P], lipid [L], and carbohydrate [C]) in the seasonal bear foods and diets, as well as their macronutrient niche breadth. We found that bamboo (Arundinaria spp.) had the highest relative frequency in both study areas and seasons. Ants and termites were found in DHR diets, but not KSL diets. One anthropogenic crop was found in DHR summer diets (Zea mays) and two were found in KSL summer diets (Z. mays; and Kodo millet [Paspalum scrobiculatum]). Other than insects, no animal prey was found in either diet. The proportions of macronutrients in diets (i.e., realized macronutrient niches) were relatively high in carbohydrate for both study areas and seasons: DHRsummer 24.1P:8.7L:67.2C; KSLsummer 16.7P:8.2L:75.1C; DHRautumn 21.1P:10.5L:68.4C; KSHautumn 19.0P:11.0L:70.0C. Macronutrient niche breadth was 3.1 × greater in the DHR than KSL during summer, and 4.0 × greater in the autumn, primarily due to the higher proportion of lipid in ants and termites relative to plant foods. Within‐study area differences in niche breadth were greater during summer than autumn; in the KSH the macronutrient breadth was 1.4 × greater in summer, while in the DHR it was 1.1 × greater in summer. Similarity in dietary macronutrient proportions despite differences in foods consumed and niche breadth are suggestive of foraging to reach a preferred macronutrient balance.  相似文献

Identifying habitat suitability and potential corridors are important tools for biodiversity conservation in the face of climate change. We modeled habitat suitability and simulated possible corridors for movement and gene flow among the Asiatic black bear (Ursus thibetanus) population in the Northern Highlands of Pakistan (NHP). Results indicated that the areas of 13,923 km2 and 21,931 km2 suitable for the Asiatic black bear under current and future scenarios respectively. Under the future scenario, we found an area of 12,657 km2 (57.21%) as increase in suitable habitat (ISHf) and 4649 km2 (33.39%) area as a decrease in current suitable habitat (DSHc). Our model predicted that about >65% (9274 km2) of the current suitable habitat as a climate refugia which is projected from the center to southeast east and northwest of the NHP primarily in the Khyber Pakhtunkhwa (KPK) and Pakistan Administered Kashmir (PAK). The attitudinal range of refugia was projected from 688 m to 4483 m with >56% at the elevations between 2001 m to 3000 m. A very small portion of suitable habitats (current suitable habitat = 2.75%, future suitable habitat = 5.11%) were projected under the protected areas. Maps connecting suitable habitats identified different regions delineated as important for the dispersal of Asiatic black bears, which mainly distributed in the PAK and KPK. Our results help informs conservation strategies and management plans for mitigating the impacts of climate change on Asiatic black bears in the NHP.  相似文献

We developed 29 forensic quality tetranucleotide microsatellite loci for the identification of individual black bear (Ursus americanus). Allele number averaged 5.35 and ranged from 3 to 11. The eight markers selected for forensic use permit the identification and exclusion of individual bear with a probability of identity of approximately 8.1 × 10−8 (approximately one per 12 million).  相似文献

The adult Polar and Asiatic Black Bear have one hemoglobin component each. The complete amino-acid sequences of their alpha- and beta-chains are presented. Their primary structures were determined by sequencing the tryptic and prolyl peptides. The alignment of these peptides was deduced from homology to human hemoglobin chains. The hemoglobin sequences of the two species proved to be identical. The evolutionary aspects of this result are discussed. Sialyl oligosaccharides were separated from two samples of Japanese black bear milk by extraction with chloroform/methanol, gel filtration on Bio Gel P-2, ion exchange chromatography on DEAE-Sephadex A-50 and high-performance liquid chromatography (HPLC) on a TSK gel Amido-80 column. The population genetic status of the Asian black bear Ursus thibetanus in the western part of Japan was examined using microsatellite DNA markers. Although three fragmented populations have been recognized in this region, i.e., the western Chugoku (WC), eastern Chugoku (EC), and northern Kinki (NK) populations, it was recently pointed out that the NK population has also been divided into two subpopulations, the western (WNK) and the eastern (ENK) populations. We examined the genetic status of these four populations based on their heterozygosities and the genetic distances (F ST ) between them. The mean observed heterozygosity, i.e., genetic diversity, of the six microsatellite loci examined, was significantly lower in WC (0.272; n = 52) and WNK (0.311; n = 66) than that in ENK (0.445; n = 67), respectively. Reflecting geographic distances among populations, F ST was higher between WC and WNK (0.244) and between WC and ENK (0.229) than that between WNK and ENK (0.066), respectively. There was also low but significant genetic differentiation between the two nearest populations (WNK and ENK). Although the status of EC was not conclusively determined because of its small sample size (n = 24), its genetic diversity seemed to be at a level similar to that of WC and WNK. These results suggest that WC, which is isolated from the others, has been losing genetic diversity, and that WNK, which had been thought to form a relatively large population together with ENK, might be isolated genetically from other populations. Since the number and range of Asiatic black bear (Ursus thibetanus) are declining due to habitat loss and illegal trade, it is essential to take effective actions to reinforce the conservation of the remaining bear populations. In order to aid such conservation efforts, we developed 12 novel polymorphic microsatellite loci of Asiatic black bear from genomic DNA-enriched libraries in this paper. The number of alleles per locus in 24 individuals ranged from 3 to 10, the average observed heterozygosity per locus from 0.214 to 0.950, and the average expected heterozygosity per locus from 0.243 to 0.891. Eight loci followed Hardy–Weinberg expectations after Bonferroni correction for multiple comparisons. No significant linkage association was found among all these loci. The 12 polymorphic microsatellite loci will be helpful to the conservation of the Asiatic black bear.  相似文献

Shifts in activity patterns and time budgets in response to environmental variables may yield insights into key factors affecting animal life histories. Activity data on wild Asiatic black bears Ursus thibetanus are limited, and non-existent in tropical portions of their geographic range. During November 1998–November 2000, we monitored the activity (active vs. inactive) of bears within a national park in the tropics of central Taiwan. On the basis of 4630 readings from motion-sensitive transmitters obtained in 24-h monitoring sessions, bears were active 54–57% of the time over the course of the year. However, bears were more active during summer (60±15% sd ) and fall (60±15%) than in spring (47±13%). They were active mainly during daylight hours in spring and summer, and also during fall when acorns, a principal fall food, were scarce. However, extensive nocturnal activity occurred in fall when acorns were abundant. Increased nocturnal activity in fall is consistent with studies of the related American black bear Ursus americanus , and suggests that this behavior is not necessarily driven by preparation for winter denning or because of disturbance by hunters (as posited in previous studies), as bears in our study did not den (except parturient females) and were not hunted or otherwise disturbed by people.  相似文献

We obtained the complete mitochondrial genome of U.thibetanus mupinensis by DNA sequencing based on the PCR fragments of 18 primers we designed. The results indicate that the mtDNA is 16,868 bp in size, encodes 13 protein genes, 22 tRNA genes, and 2 rRNA genes, with an overall H-strand base composition of 31.2% A, 25.4% C, 15.5% G and 27.9% T. The sequence of the control region (CR) located between tRNA-Pro and tRNA-Phe is 1422 bp in size, consists of 8.43% of the whole genome, GC content is 51.9% and has a 6bp tandem repeat and two 10bp tandem repeats identified by using the Tandem Repeats Finder. U. thibetanus mupinensis mitochondrial genome shares high similarity with those of three other Ursidae: U. americanus (91.46%), U. arctos (89.25%) and U. maritimus (87.66%).  相似文献

The Anolis roquet series of Caribbean lizards provides natural replicates with which to examine the role of historical contingency and ecological determinism in shaping evolutionary patterns. Here, we describe 10 polymorphic tetranucleotide microsatellites to facilitate studies on population differentiation and gene flow. All loci successfully amplified in several species from this series. Genotyping 96 individuals from two A. roquet populations demonstrated the markers’ suitability as population genetic markers: genetic diversity was high (9–22 alleles per locus); there were no instances of linkage disequilibrium; and, with one exception, all genotypic frequencies conformed to Hardy–Weinberg equilibrium expectations.  相似文献

研究同域物种的分布格局及重叠状况对物种的区域整合保护管理及区域生物多样性保护具有重要实践价值。本研究基于全国第四次大熊猫调查及长期野外调查数据, 利用MaxEnt模型预测了凉山山系两种同域分布的熊科动物——大熊猫(Ailuropoda melanoleuca)和黑熊(Ursus thibetanus)的适宜生境, 基于适宜生境预测结果, 分析了两个物种的生境需求因子、生境破碎化现状及重叠状况。结果显示: (1)大熊猫和黑熊的适宜生境分布格局相似, 主要分布在凉山山系的山脊地带, 适宜生境面积分别为1,383.84 km2和2,411.49 km2; (2)两个物种的适宜生境都较为破碎, 且存在一些隔离分布区, 相较而言, 黑熊适宜生境的连通性要优于大熊猫; (3)两个物种生态位重叠度较高(D = 0.654, I = 0.901), 适宜生境重叠面积为958.29 km2, 分别占大熊猫和黑熊适宜生境总面积的69.25%和39.74%; (4)两个物种对环境因子的选择和响应表现出了相似性和差异性。相似性在于对两个物种生境分布影响最大的两个因子均为距居民点距离和海拔; 差异性在于对大熊猫生境分布影响次之的因子是植被类型和最冷季均温, 而黑熊的是年最大EVI指数和距道路距离。为了更有效地保护两个物种, 应加强对人类干扰的控制和植被的恢复, 对栖息地实行连通管理, 并建立多物种保护规划。  相似文献
