本文报道大鼠水盐负荷时,其心房特殊颗粒(ASG)的数目及钙含量发生相关变化。给大鼠禁水5d或饮2%NaCl液4d造成水盐负荷,用电镜形态计量法计数ASG,以反映心房钠尿肽(ANP)的分泌水平,电镜X射线显微分析法测定ASG中钙、硫含量及肌质同终末池(TSR)中钙含量。结果显示,禁水引起ANP分泌减少时,ASG的数密度增加(6.02±2.30增至9.97±3.21个/μm3),同时伴有钙含量增加(64±16增至92±18mmol/kg);饮盐水引起ANP分泌增加时,ASG的数密度减少(6.02±2.30减至2.96±1.62个/μm3),巨伴有钙含量减少(64±16减至38±22mmol/kg),但水盐负荷对TSR中钙浓度无任何影响。据此推论ASG作为细胞内钙库,借某种机制调控其中钙的释放而参与ANP的刺激分泌耦联过程。  相似文献

冷锻炼和ABA诱导水稻幼苗提高抗冷性期间膜保护系统的变化   总被引:16,自引:1,他引:15  
冷锻炼和ABA处理提高了水稻幼苗叶绿体SOD和GR活性及叶片抗氧化剂AsA和GSH的含量,降低了膜电解质泄漏,增强了幼苗的抗冷性.等电聚焦电泳分析表明,冷锻炼和ABA处理苗叶绿体SOD三条同工酶带和GR1、2、3和6同工酶带都有不同程度的增强.低温胁迫后,处理和未处理首的SOD、GR活性和ASA、GSH含量均有所下降.但处理苗的水平仍维持在未处理苗之上.亚胺环已酮可抑制因冷锻炼和ABA诱导增加的SOD和GR活性,并使叶片电解质泄漏增大.本试验结果表明冷锻炼或ABA诱导水稻幼苗抗冷性提高时,对防御活性氧的保护系统有类似的影响。  相似文献

水稻(Oryza sativa L.)和黄瓜(Cucum ism elo L.)幼苗在昼夜温度为15 ℃/10 ℃、白天光照12 h,光强为250 μm ol·m - 2·s- 1的条件下锻炼3 d,明显地提高幼苗叶片中膜保护酶——超氧物歧化酶(SOD)、谷胱甘肽还原酶(GR)活性和内源抗氧化剂——还原型谷胱甘肽(GSH)、抗坏血酸(AsA)的含量。经冷锻炼和未锻炼的幼苗移置4 ℃、光强为250 μm ol·m - 2·s- 1下胁迫处理2 d,未锻炼苗叶片中SOD、GR 活性和GSH、AsA 含量明显下降,而经冷锻炼的苗则相对比较稳定。从脂质过氧化产物——丙二醛(MDA)含量及幼苗的存活率亦看出:冷锻炼苗具有较低的脂质过氧化水平和较高的幼苗存活率。由此认为:冷锻炼能提高水稻和黄瓜幼苗细胞膜的稳定性,从而增强了耐低温光胁迫的能力  相似文献

盐胁迫激活大麦幼苗脯氨酸合成的鸟氨酸途径   总被引:20,自引:2,他引:18  
200mmol/L NaCl处理结合^14C-Glu和^14C-Arg叶面饲喂6d龄大麦(Hordeum vulgareL.)幼苗,结果证明6d龄大麦幼苗体内普遍存在Arg(谷氨酸)→Om(鸟氨酸)→Pro(脯氨酸)途径。直 迫明显激活了Arg→Om→Pro途径,胁迫处理7d,大麦幼苗叶片和根系中该途径对Pro含量上升的贡献是Glu→Pro途径的1.0-1.5倍。而盐的“鉴4”品种Arg→Om→Pro途径对Pro含量上升的贡献是对盐敏感的“科品7号”的1.7-2.0倍,结果表明盐胁迫下Arg→Om→Pro途径的激活对大麦幼苗耐盐性的提高具有重要意义。  相似文献

以杂交稻(汕优63)为试验材料,在木村B营养液中培养至三叶期,用草酸5mmol/L预处理水稻2d,再处以氧化胁迫(用0.1mmol/L浓度的活性氧诱发剂甲基紫精处理)。结果表明:MV诱发的氧化胁迫下,Rubisco及其它可溶性蛋白快速降解。草酸预处理可明显缓解Rubisco及其它可溶性蛋白的降解,降解速率分别降低1/3和1/2左右,植株经草酸处理后其叶片中几种抗氧化酶如AsA-POD、SOD、CA  相似文献

本文发展了一种从百日咳杆菌Ⅰ相菌株中纯化69KDa外膜蛋白的简易方法,将细菌体经加热浸提、乙醇沉淀蛋白、DEAE-Sephadex A50柱层析精制而成。用SDS-PAGE、免疫印迹、光密度仪扫描分析,证明纯化制剂为均一的、特异性的69KDa外膜蛋白,其收率为54.2%,纯度达99.2%,每微克69KDa蛋白制剂中的内毒素含量低于0.85EU;PT残留量小于0.105ng。抗69KDa蛋白抗血清能  相似文献

NaCl胁迫对螺旋藻生长及抗氧化酶活性的影响   总被引:21,自引:0,他引:21  
在01%~5.0%NaCl浓度范围的培养基中培养极大螺旋藻(Spirulinamaxima),发现NaCl浓度高于2.0%时螺旋藻生长受到明显抑制。培养7天后测定超氧化物歧化酶(SOD)、抗坏血酸过氧化物酶(ASAPOD)、过氧化氢酶(CAT)活性和丙二醛(MDA)含量。结果表明:在盐胁迫下,SOD酶活性升高;抗坏血酸过氧化物酶和过氧化氢酶活性在低盐胁迫下活性升高,高盐胁迫下抗坏血酸过氧化物酶活性迅速降低,过氧化物酶则完全失活;MDA含量先随盐胁迫程度增加而降低,后随盐胁迫的进一步增强恢复至对照水平。  相似文献

冷锻炼处理提高了水稻(Oryza sativa L.)幼苗叶片中抗氧化剂(还原型谷胱甘肽,GSH;抗坏血酸,AsA)含量和膜保护酶(超氧化物歧化酶,SOD)的活性,同时也提高了可溶性蛋白质中热稳定蛋白的含量。CaCl2 浸种处理对上述冷锻炼的作用有加强的效果,且明显地提高了过氧化氢酶(CAT)和过氧化物酶(POD)的活性。有无CaCl2 处理的冷锻炼处理均减轻冷胁迫引起的GSH 及AsA 含量、SOD 活性及热稳定蛋白质含量的下降程度,有利于幼苗在恢复过程中GSH、AsA、CAT、SOD、POD及热稳定蛋白质水平迅速回升。结合CaCl2 处理的冷锻炼苗在冷胁迫恢复生长时增长迅速,且苗健壮浓绿,说明CaCl2浸种对冷锻炼处理提高水稻幼苗的抗冷力有明显的促进作用,这与CaCl2 浸种结合冷锻炼能更有效的提高细胞膜保护能力有关  相似文献

Bean ( Phaseolus vulgaris L. ) seedlings were grown in a greenhouse condition under UV-B (280 ~ 320 nm) radiation and an UV-B-free control condition. At early stage of bean growth, UV-B radiation(UV- BBE: 16 kJ·m-2·d-1 ) increased the synthetic rate of protein in bean leaves. UV-B radiation decreased the synthetic rate of protein and the content of soluble protein, increased the activities of caseolytic enzymes and the content of total amino acids in bean leaves at later growth stage. The results of SDS-PAGE indicated that UV-B radiation increased the contents of polypeptide in a range of 99 kD, 88 kD, 76 kD, 42 kD, 35 kD and 33 kD at the early stage, and increased that of 10 ~ 14 kD, 29 kD, 33 kD and 35 kD at later growth stage. It is suggested that the increase of these polypeptides is an adaptation response of plants to the enhanced UV-B radiation.  相似文献

SDS-polyacrylamide gel electrophoresis of seed protein extracts showed synthesis of major legumin and vicilin polypeptides from 30 OAF onwards though very faint polypeptide bands could be seen at early stage of 10 OAF. Globulin synthesis enhanced and dominated in the second half of seed development. In germinating seeds, legumin polypeptides (mol wt 75–66, 45–41 kD) and vlcllin polypeptides (mol wt 54 kD) were degraded Increasingly from 2nd to 6th day of germination. Responses of soluble amino acids and organic acids to either ambient (36 Pa) or elevated (100 Pa) CO2 treatments were determined using barley primary leaves (Hordeum vulgare L. cv. Brant). Total soluble amino acids were increased 33% by CO2 enrichment 9 days after sowing (DAS), but a decrease relative to the ambient CO2 treatment was observed with increasing leaf age. Marked declines of glutamine and asparagine were observed under CO2 enrichment, both diurnally and with advancing leaf age. Consequently, total soluble amino acids were 59% lower in the elevated compared to the ambient CO2 treatment 17 DAS. It was likely that chlorosis in response to CO2 enrichment negatively impacted soluble amino acid levels in older barley primary leaves. In contrast to the ambient CO2 treatment, glutamine and most other soluble amino acids decreased as much as 60% during the latter half of a 12 h photoperiod in primary leaves of 13-day-old seedlings grown under enhanced CO2. Malate was decreased about 9 percent by CO2 enrichment and citrate and succinate were increased by similar amounts when measured 9 and 13 DAS. Malate accumulation was also decreased about 20% by CO2 enrichment on a diurnal basis. The onset of CO2-dependent leaf yellowing had much less of an effect on organic acids than on soluble amino acids. This above results emphasized the sensitivity of N metabolism to CO2 enrichment in barley. Increased levels of citrate and succinate in response to CO2 enrichment suggested that the tricarboxylic acid cycle was upregulated in barley by CO2 enrichment. In summary, organic and amino acid levels in barley primary leaves were dynamic and were altered by age, diurnally and in response to CO2 enrichment.  相似文献

萌发绿豆子叶自然衰老过程中可溶性蛋白质含量一直下降;从衰老开始到衰老前期,总游离氨基酸含量明显上升;但游离氨基酸各组分在子叶衰老期间的变化趋势并不相同。~3H-亮氨酸掺入蛋白质试验和多聚核糖体的相对量及其与总核糖体的比值(P/T)测定都证明在子叶衰老前期有蛋白质的新合成。子叶衰老期间。氨肽酶活性明显降低;而以酪蛋白为底物的蛋白水解酶活性却急剧上升,承担着催化蛋白质降解的主要功能。  相似文献

对盆栽9叶龄的3个烟草近等基因系进行5℃低温处理。测定处理2d、4d和6d以及处理6d后恢复生长2d和4d烟草叶片的生理生化指标,观察处理2d、4d和6d后恢复生长7d烟草的生长情况。结果表明随着低温处理时间的延长,细胞质膜透性、丙二醛含量、可溶性蛋白质和糖含量均升高;SOD活性则开始下降随后升高,但始终低于对照;低温造成株高、叶片数和生物量显著下降。  相似文献

对欧洲榛子(Corylus avellanaL.)在沙藏、萌发、幼苗生长阶段的种仁养分和保护酶活性动态变化特点进行了研究,结果表明:种子萌发后107 d营养耗尽;沙藏过程中种仁的干重、粗脂肪含量略下降,萌发后下降迅速且与时间呈线性负相关;可溶性糖在沙藏至萌发后12 d内下降而后上升,萌发24 d后上升迅速,72 d后进入高峰期;淀粉含量在沙藏过程中下降,萌发后上升并在萌发48 d取得11.12%的峰值后下降;游离氨基酸总量在沙藏期间下降,萌发前12 d开始升高,在萌发36~48 d达155.31 mg/100 g的高峰后降低;可溶性蛋白50.87 mg/g的峰值出现在萌发前12 d;SOD在萌发前12 d至萌发后12 d、CAT由萌发起24 d内、POD在萌发36 d后分别出现活性高峰,对种仁营养按测定时间进行的聚类分析结果与依据幼苗干重划分的幼苗生长阶段相吻合。  相似文献

Abstract— Changes in morphology and in transformations of [U-14C]glucose and [1-14C]acetate into amino acids of the brain cortex were followed on the Sth, 10th and 21st days after production of mechanical lesions and compared with control tissue. In the experimental tissue, proliferation of astroglia and reduction of the number of neurons had taken place. On the 10th day, accumulation of mitochondria and of some gliofilaments in the cytoplasm of astroglia was observed. On the 21st day, the gliofilaments occupied a substantial portion of the astroglial cytoplasm and the mitochondria were reduced in number and compressed to the cell membrane. Incorporation of 14C from acetate into amino acids was substantially increased on the 10th day (up to 240% with respect to controls) and normalized again on the 21st day. Incorporation of [14C]glucose into amino acids decreased somewhat during the experimental period. It has been proposed that the proliferation of astrocytes and their ultrastructural changes may account for the increased transformation of [14C]acetate into amino acids, in particular into glutamine which is formed from the small glutamate pool.  相似文献

Changes in the levels of protein and free amino acids in theseeds and placentae of Nicotiana tabacum were studied duringseed development. Seed maturation was completed 24 days afteranthesis. During maturation, protein rapidly accumulated inthe seeds between the 6th and 18th day, along with an appreciablecompositional change in the protein amino acids as the proportionsof glutamic acid and arginine increased. The amount of freeamino acids in the seeds gradually decreased throughout maturation.The major free amino acid on the 6th day after anthesis wasglutamine, which then drastically decreased between the 6thand 12th day with increases of glutamic acid, proline, arginineand alanine. The latter amino acids decreased thereafter untilthe 24th day. On the other hand, the amount and composition of the proteinsin the placentae did not change significantly throughout seedmaturation. In the early stage of development, the major freeamino acids in the placentae were glutamine, asparagine andglutamic acid, while in the later stage asparagine was mostabundant.
