Summary Olfactory receptor cells of the spruce bark beetle,Ips typographus, and its predator, the clerid beetleThanasimus formicarius, were studied using electrophysiological techniques. Recordings were made of nerve impulses from single cells and of the summated receptor potential (electroantennogram).Information from bark beetle pheromones and host volatiles is detected by separate olfactory receptor cells inI. typographus. Those which detected bark beetle pheromones responded to only one key substance. Some receptor cells which responded to spruce bark volatiles were strongly activated by one of the synthetic host compounds tested. However, too few host compounds were tested to reach definite conclusions about the specialization of host odour cells. T. formicarius has evolved olfactory receptor cells for bark beetle pheromones. These have similar specificities (specialist types) to those of the bark beetles. Furthermore, the predator has olfactory receptor cells for many bark beetle pheromones. This indicates thatT. formicarius is able to detect and discriminate between many bark beetle species. No significant differences were found between prey and predator cells which responded to host volatiles.  相似文献

Pithecellobium dulce has been used in traditional medicine to treat various ailments owing to its restorative properties. The biological activities and chemical profiles of the lipophilic fraction of P. dulce bark and leaves were assessed herein. Fatty acid methyl esters (FAME) and unsaponifiable matter (USM) were prepared and analyzed by GC/MS. A total of 40 compounds were identified in the bark saponifiable fraction, whereas 9 compounds were annotated in the leaves. Palmitic acid methyl ester was the major compound identified accounting for 41.48 % of the bark and 19.03 % of the leaves composition. Besides, linolenic acid methyl ester (22.40 %) and linoleic acid (12.69 %) were annotated in the leaves saponifiable fraction. A total of 63 compounds were detected in the bark USM and 4 compounds were identified in the leaves. Phytol represented the major component in the leaves (52.57 %) followed by lupeol (20.68 %) and lupenone (8.60 %). Meanwhile, n‐dodecane dominated in the bark USM accounting for 24.69 % of the total composition. The leaves and bark lipophilic fractions revealed moderate antioxidant and antibacterial activities. Both extracts showed no antifungal activity. No cytotoxicity was observed for both lipophilic fractions. P. dulce offers a good source of antioxidant compounds that can be introduced to food and pharmaceutical industry.  相似文献

The role of olfactory stimuli in host detection and evaluation was studied in two encyrtid Hymenoptera. The first, Epidinocarsis lopezi De Santis, is a monophagous parasitoid of the cassava mealybug Phenacoccus manihoti Matile-Ferrero, itself feeding exclusively on cassava, Manihot esculenta Crantz. The second, Leptomastix dactylopii Howard, is a monophagous parasitoid of the Citrus mealybug, Planococcus citri Risso, but this latter is highly polyphagous. The behaviour of females of both parasitoids (attaction and locomotion) was compared in a tubular olfactometer for the odours of their respective hosts on cassava and poinsettia. Tests were made using: 1) healthy host-plant alone; 2) host-plant infested with unparasitized mealybugs; 3) unparasitized mealybugs only; 4) host-plant infested with parasitized mealybugs and 5) parasitized mealybugs only. Only E. lopezi was attracted by the odour of the host-plant alone, but both species were attracted by the odour of an infested host-plant and that of unparasitized mealybugs. The odour of parasitized mealybugs, alone or on host-plant, induced an undirected activity. The attraction of E. lopezi to the odour of the host-plant alone could be linked to the monophagous diet of its host, whereas the attraction of the two species of parasitoids to the odours of infested host-plants and unparasitized mealybugs could be due to the fact that both parasitoids are specialists. The behavioural response of both species to the odour of parasitized mealybugs revealed a new aspect in host discrimination: the identification of parasitized hosts could be partly mediated through olfactory stimuli, and not only through gustatory stimuli.  相似文献

Abstract 1 When searching for suitable hosts in flight, especially in mixed forests, conifer‐inhabiting bark beetles will encounter not only suitable host trees and their odours, but also unsuitable hosts and nonhost trees. Rejection of these trees could be based on an imbalance of certain host characteristics and/or a negative response to some nonhost stimuli, such as nonhost volatiles (NHV). 2 Recent electrophysiological and behavioural studies clearly indicate that conifer‐inhabiting bark beetles are not only able to recognize, but also to avoid, nonhost habitats or trees by olfactory means. Green leaf volatiles (GLV), especially C6‐alcohols, from the leaves (and partly from bark) of nonhost angiosperm trees, may represent nonhost odour signals at the habitat level. Specific bark volatiles such as trans‐conophthorin, C8‐alcohols, and some aromatic compounds, may indicate nonhosts at the tree species level. Flying bark beetles are also capable of determining whether a possible host is unsuitable by reacting to signals from conspecifics or sympatric heterospecifics that indicate old or colonized host tree individuals. 3 Combined NHV signals in blends showed both redundancy and synergism in their inhibitory effects. The coexistence of redundancy and synergism in negative NHV signals may indicate different functional levels (nonhost habitats, species, and unsuitable hosts) in the host selection process. Combinations of NHV and verbenone significantly reduced the number of mass attacked host trees or logs on several economically important species (e.g. Dendroctonus ponderosae, Ips typographus, and I. sexdentatus). 4 We suggest a semiochemical‐diversity hypothesis, based on the inhibition by NHV of bark beetle host‐location, which might partly explain the lower outbreak rates of forest insects in mixed forests. This ‘semiochemical‐diversity hypothesis’ would provide new support to the general ‘stability‐diversity hypothesis’. 5 Natural selection appears to have caused conifer‐inhabiting bark beetles to evolve several olfactory mechanisms for finding their hosts and avoiding unsuitable hosts and nonhost species. NHV and unsuitable host signals have potential for use in protecting trees from attack. The use of these signals may be facilitated by the fact that their combination has an active inhibition radius of several metres in trap test, and by the observation of area effects for several trees near inhibitor soruces in tree protection experiments. Furthermore, incorporation of negative signals (such as NHV and verbenone) and pheromone‐based mass‐trapping in a ‘push–pull’ fashion may significantly increase the options for control against outbreaks of conifer‐inhabiting bark beetles, especially in high risk areas.  相似文献

Juvenile anemonefishes detect their host sea anemone by olfactory stimuli; in order to investigate whether this behaviour is innate or acquired, the anemonefish species Amphiprion ocellaris was bred in two different ways: 1. With no host sea anemone present at all (–A); and 2. With the specific host sea anemone Heteractis magnifica present in the hatching aquarium, so that these eggs were laid and hatched close to the sea anemone, as in nature (+A). The two different types of juvenile A. ocellaris were presented to the odours of the host sea anemone H. magnifica in two sets of short-term experiments with the host (a) visually hidden in a net cage, and (b) visible but physically separated from the anemonefishes. In both cases, a water flow was established between fishes and host. The +A-fishes found their host by olfactory and not by visual stimuli. In both series, the –A-fishes showed a significantly lower affinity behaviour towards the odour compounds from the host sea anemone than the +A-fishes did. A third type of experiment was a direct confrontation between fishes and host; here, the –A-fishes were indifferent towards the host sea anemone for almost 48 h, while the +A-fishes acclimated to the host sea anemone within the first 5 min of the direct confrontation. The results of this study suggest that Amphiprion ocellaris imprints itself olfactorily to its species-specific host sea anemone Heteractis magnifica, and, furthermore, may be genetically disposed towards olfactory recognition of the host sea anemone.  相似文献

Herbivorous insects use olfactory cues to locate their host plant within a complex olfactory landscape. One such example is the European grapevine moth Lobesia botrana, a key pest of the grape in the Palearctic region, which recently expanded both its geographical and host plant range. Previous studies have showed that a synthetic blend of the three terpenoids, (E)‐β‐caryophyllene, (E)‐β‐farnesene and (E)‐4,8‐dimethyl‐1,3,7‐nonatriene (DMNT), was as attractive for the moth as the complete grape odour profile in laboratory conditions. The same studies also showed that the specific ratio of these compounds in the grape bouquet was crucial because a percentage variation in any of the three volatiles resulted in almost complete inhibition of the blend's attractiveness. Here, we report on the creation of stable grapevine transgenic lines, with modified (E)‐β‐caryophyllene and (E)‐β‐farnesene emission and thus with an altered ratio compared to the original plants. When headspace collections from these plants were tested in wind tunnel behavioural assays, they were less attractive than control extracts. This result was confirmed by testing synthetic blends imitating the ratio found on natural and transformed plants, as well as by testing the plants themselves. With this evidence, we suggest that a strategy based on volatile ratio modification may also interfere with the host‐finding behaviour of L. botrana in the field, creating avenues for new pest control methods.  相似文献

Terrestrial snails Helix aspersa that were exposed to moving air containing a relatively uniform concentration of host-plant odour utilized an anemotactic response and oriented upwind toward the odour source. Also, the snails were able to follow a well-defined trail of host-plant odour via olfactory cues in the absence of directional cues from air movement. In the natural environment, these anemotactic and odour-trail-following mechanisms may operate either independently or in combination to enable location of a host plant.  相似文献

Herbivorous insects use highly specific volatiles or blends of volatiles characteristic to particular plant species to locate their host plants. Thus, data on olfactory preferences can be valuable in developing integrated pest management tools that deal with manipulation of pest insect behaviour. We examined host plant odour preferences of the tomato leafminer, Liriomyza bryoniae (Kaltenbach) (Diptera: Agromyzidae), which is an economically important agricultural pest widespread throughout Europe. The odour preferences of leafminers were tested in dependence of feeding experiences. We ranked host plant odours by their appeal to L. bryoniae based on two‐choice tests using a Y‐tube olfactometer with five host plants: tomato, Solanum lycopersicum Mill.; bittersweet, Solanum dulcamara L.; downy ground‐cherry, Physalis pubescens L. (all Solanaceae); white goosefoot, Chenopodium album L. (Chenopodiaceae); and dead nettle, Lamium album L. (Lamiaceae). The results imply that ranking of host plant odours by their attractiveness to L. bryoniae is complicated due to the influence of larval and adult feeding experiences. Without any feeding experience as an adult, L. bryoniae males showed a preference for the airflow with host plant odour vs. pure air, whereas females did not display a preference. Further tests revealed that adult feeding experience can alter the odour choice of L. bryoniae females. After feeding experience, females showed a preference for host plant odour vs. pure air. Feeding experience in the larval stage influenced the choice by adults of both sexes: for males as well as females reared on bittersweet the odour of that plant was the most attractive. Thus, host feeding experience both in larval and/or adult stage of polyphagous tomato leafminer L. bryoniae influences host plant odour preference by adults.  相似文献

Olfactory receptors pertaining to G protein-coupled receptor (GPCR) are integral membrane proteins composed of seven transmembrane spanning domains. It has been reported that these receptor proteins are difficult to overexpress, solubilize, and purify because of their complicated structures and strong hydrophobicity. In this study, full-length human olfactory receptor (hOR) 2AG1 was overexpressed in E. coli as a fusion protein with a glutathione S-transferase (GST) tag mainly as an inclusion body without any mutations or deletions in the gene. This protein was difficult to solubilize with detergents and chaotropic agents, and only N-lauroyl sarcosine was found to be suitable for solubilizing it. In contrast, Trition X-100 was found to solubilize most of the impurity proteins from the insoluble fraction in E. coli. Based on this observation, we applied a simple and efficient column-free method using these two detergents for the purification of the olfactory receptor protein. In this method, the insoluble fraction of the cell lysate was first treated with Triton X-100 to remove impurity proteins. The remaining insoluble fraction was then further treated with N-lauroyl sarcosine to solubilize the olfactory receptor protein. Milligram quantity of the human olfactory receptor was produced. This is the first report to produce full-length of the olfactory receptor from E. coli.  相似文献

Action potentials from individual olfactory cells and receptor potentials were recorded from antennae of female cabbage white butterflies, Pieris brassicae, using a new ‘surface-contact’ recording technique. Stimuli used were plant volatiles and conspecific egg odours. Most cells were activated by some stimuli and inhibited by others. Hence, the ‘odour spectra’ of most cells included activating as well as inhibiting substances. In addition, the ‘response profile’ of a given stimulus contained both positively and negatively responding cells.Cluster analysis of the odour spectra revealed the existence of moderately separated clusters of olfactory receptors. No groups of cells specifically tuned to odours of eggs or cabbage leaves were found. Analysis of the response profiles did not show a clear clustering of stimuli. The results suggested that the membranes of different receptor cells contain several types of acceptor sites in various proportions.No correlation between EAG and receptor potential amplitude or spike frequency was found. It was concluded that the EAG may be useful in comparing the relative receptor sensitivities only when chemically related compounds are involved.  相似文献

Bovine odour excites olfactory receptor(s) in a wall-pore olfactory sensillum on the anterior pit of Haller's organ in Amblyomma variegatum. Gas chromatography-coupled electrophysiology recordings from this sensillum reveal the presence of 4 active compounds in bovine odour. The two strongest stimulants were identified as 2-nitrophenol and 4-methyl-2-nitrophenol by gas chromatography-coupled mass spectrometry, and by matching electrophysiological activity of synthetic analogues. Synthetic analogues of known vertebrate-associated volatiles also stimulate other olfactory receptors in sensilla on the surface of tarsus I: a lactone receptor responding to -valerolactone and 6-caprolactone; different fatty acid receptor types responding best to either pentanoic acid, 2-methylpropanoic acid or to butanoic acid; three receptors responding to NH3; and one receptor responding to 3-pentanone. Gas chromatography-coupled mass spectrometry analysis of vertebrate volatiles revealed presence of a number of these olfactory stimulants in concentrates of rabbit and steer odour, i.e. 2-methylpropanoic acid, butanoic acid, 3-methylbutanoic acid, pentanoic acid, and -valerolactone.Abbreviations GC-EL gas chromatography-coupled electrophysiological recording - GC-MS gas chromatography-coupled mass spectrometry  相似文献

Sprouting of basal buds in rose plants with a completely removed canopy was restrained by inhibitors in a water soluble fraction of an ethanolic extract from the bark tissue of the plant stump (the crown) extracted before the top removal. The inhibitory effect of this fraction disappeared if the bark was extracted one week after the removal of the plant canopy. A similar inhibition was obtained with extracts from stem segments. The organic fraction did not show any inhibitory activity. Both fractions, aqueous and organic, were inhitory in the lettuce hypocotyl elongation bioassay. High levels of ABA in the cis-configuration were found in extracts before the top removal. The amount of cis-ABA decreased in extracts made one week after top removal and a different peak of activity, probably corresponding to trans-ABA or phaseic acid was shown in the GLC-system used.A high content of cytokinins has been found in the bark tissue of the crown. There were qualitative differences in cytokinin activity between the tissue from plants after the removal of the canopy and from plants with main branches left intact. A balance between cytokinins and inhibitory complex was proposed to be involved in the regulation of basal buds outgrowth and plant regeneration.  相似文献

Insect olfactory receptor (Or) genes are large, rapidly evolving gene families of considerable interest for evolutionary studies. They determine the responses of sensory neurons which mediate critical behaviours and ecological adaptations. We investigated the evolution across the genus Drosophila of a subfamily of Or genes largely responsible for the perception of ecologically relevant aliphatic esters; products of yeast fermentation and fruits. Odour responses were recorded from eight classes of olfactory receptor neurons known to express this Or subfamily in D. melanogaster and from homologous sensilla in seven other species. Despite the fact that these species have diverged over an estimated 40 million years, we find that odour specificity is largely maintained in seven of the eight species. In contrast, we observe extensive changes in most neurons of the outgroup species D. virilis, and in two neurons across the entire genus. Some neurons show small shifts in specificity, whilst some dramatic changes correlate with gene duplication or loss. An olfactory receptor neuron response similarity tree did not match an Or sequence similarity tree, but by aligning Or proteins of likely functional equivalence we identify residues that may be relevant for odour specificity. This will inform future structure–function studies of Drosophila Ors.  相似文献

Studies of Varroa destructor orientation to honey bees were undertaken to isolate discrete chemical compounds that elicit host-finding activity. Petri dish bioassays were used to study cues that evoked invasion behaviour into simulated brood cells and a Y-tube olfactometer was used to evaluate varroa orientation to olfactory volatiles. In Petri dish bioassays, mites were highly attracted to live L5 worker larvae and to live and freshly freeze-killed nurse bees. Olfactometer bioassays indicated olfactory orientation to the same type of hosts, however mites were not attracted to the odour produced by live pollen foragers. The odour of forager hexane extracts also interfered with the ability of mites to localize and infest a restrained nurse bee host. Varroa mites oriented to the odour produced by newly emerged bees (<16 h old) when choosing against a clean airstream, however in choices between the odours of newly emerged workers and nurses, mites readily oriented to nurses when newly emerged workers were <3 h old. The odour produced by newly emerged workers 18–20 h of age was equally as attractive to mites as that of nurse bees, suggesting a changing profile of volatiles is produced as newly emerged workers age. Through fractionation and isolation of active components of nurse bee-derived solvent washes, two honey bee Nasonov pheromone components, geraniol and nerolic acid, were shown to confuse mite orientation. We suggest that V. destructor may detect relative concentrations of these compounds in order to discriminate between adult bee hosts, and preferentially parasitize nurse bees over older workers in honey bee colonies. The volatile profile of newly emerged worker bees also may serve as an initial stimulus for mites to disperse before being guided by allomonal cues produced by older workers to locate nurses. Fatty acid esters, previously identified as putative kairomones for varroa, proved to be inactive in both types of bioassays.  相似文献

The specialist parasitoidMacrocentrus grandii Goidanich (Hymenoptera: Braconidae) appears to parasitize its polyphagous host, European corn borer (Ostrinia nubilalis (Hübner)) (Lepidoptera: Pyralidae), in only certain habitats. To determine whether it differed in its olfactory response to host-habitat odours, volatiles from four plants were isolated using Tenax. Wind tunnel bioassays of the extracts revealed that, besides corn which was tested in an earlier study, olfactory stimuli for attraction ofM. grandii females were present in potato and snap bean but not in pepper or soybean. To further characterize the response to pepper and soybean, these extracts were bioassayed in combination with an attractive extract. The results indicated that pepper volatiles evoked a neutral response inM. grandii while response to soybean volatiles appeared to be neutral or slightly negative. The innate response to soybean volatiles was altered to one of attraction after oviposition experience on soybean. Seven days after oviposition, experienced females continued to respond positively to soybean volatiles. Components of soybean volatiles responsible for the change in flight behaviour resulting from oviposition experience were eluted by nonpolar and slightly polar solvents. These results support the idea that plant odour may be a factor determining the range of plants on whichM. grandii parasitizes its host. The study indicates the occurrence of associative learning of plant-related volatiles during oviposition inM. grandii, and suggests the involvement of diverse plant compounds in the learning process.  相似文献

The recent shift of Rhagoletis pomonella from its native host hawthorn to introduced, domestic apple has been implicated as an example of sympatric speciation. Recent studies suggest that host volatile preference might play a fundamental role in host shifts and subsequent speciation in this group. Single sensillum electrophysiology was used to test a proposed hypothesis that differences in R. pomonella olfactory preference are due to changes in the number or odor specificity of olfactory receptor neurons. Individuals were analyzed from apple, hawthorn, and flowering dogwood-origin populations, as well as from the blueberry maggot, Rhagoletis mendax Curran (an outgroup). Eleven compounds were selected as biologically relevant stimuli from previous electroantennographic/behavioral studies of the three R. pomonella populations to host fruit volatiles. Cluster analysis of 99 neuron responses showed that cells from all tested populations could be grouped into the same five classes, ranging from those responding to one or two volatiles to those responding to several host volatiles. Topographical mapping also indicated that antennal neuron locations did not differ by class or fly taxa. Our results do not support the hypothesis that differences in host preference among Rhagoletis populations are a result of alterations in the number or class of receptor neurons responding to host volatiles.  相似文献

Extracellular recordings were performed from olfactory bulbneurons in the frog. The odour stimuli were the same as thosepreviously used for studying the receptor cells in the sameanimal species and were delivered at similar concentrations(Revial et al., 1982). The general properties of the neuronresponses are presented and discussed with reference to homologousproperties of olfactory receptor cells. The response rates elicitedby different stimuli from the bulbar neurons were found to behighly correlated with those elicited from receptor cells. Theindividual cell selectivity was better in the bulb than in theolfactory epithelium. The olfactory bulb neurons seemed to improvethe discrimination between stimuli (enantiomers) poorly distinguishedby the receptor cells. Reducing odor concentration caused therate of suppressive response to decrease faster than that ofexcitatory ones, suggesting that the manifestations of inhibitoryprocesses in some neurons requires a high level of excitationin others.  相似文献

The response of antennal olfactory receptor neurons (ORNs) of Monochamus galloprovincialis to several odourants was tested using single sensillum electrophysiology. Behaviourally active pheromone, and kairomone (host and sympatric bark beetle pheromone) odours were tested alongside smoke compounds released by burnt wood that are potentially attractive to the insect. The antennae bore several types of sensilla. Two plate areas in the proximal and distal ends of each antennal segment were covered with basiconic sensilla that responded to the odour stimuli. Sensilla basiconica contained one or two cells of different spike amplitude. The 32 male and 38 female ORNs tested responded with excitations or inhibitions to the different plant odours. In general the response of male and female receptors was very similar so they were pooled to perform a cluster analysis on ORN responses. Six ORNs were clearly specialised for pheromone reception. Responses to kairomone and smoke odours were less specific than those of pheromone, but a group of 9 cells was clearly excited by smoke compounds (mainly eugenol and 4-methyl 2-methoxyphenol), a group of 8 cells was very responsive to α-pinene, β-pinene and cis-verbenol, and a group of 14 cells responded to a wider range of compounds. The rest of the cells (47%) were either non-responsive or slightly inhibited by smoke compounds. Dose–response curves were obtained for several compounds. Different compounds induced significantly different latencies and these appeared to be unrelated to their boiling point.  相似文献

Several fractions of ether extracts of beet molasses separated by chemical and chromatographic methods were found to be toxic for Aspergillus niger. Addition of these fractions to growth medium caused sinking of the mycelium in the culture broth and irregular growing of the test strains. Fermentation tests carried out on molasses media in the presence of the toxic fractions showed a different susceptibility depending on the used strain. From one fraction n-alkanes with 16 to 36 carbon atoms were isolated by thin-layer chromatography and identified using IR and GC methods. These compounds originated in molasses probably from chemical additives used in the sugar manufacture plant.  相似文献
