Nine known or potential components of the aggregation-attachment pheromone of the ticksAmblyomma hebraeum andA. variegatum were tested as attachment stimulants. Unfed adult male and female ticks were confined in linen bags on the ears of rabbits that had been treated with solutions of the test compounds; the numbers attached were recorded after 24 h. In both species, males attached more rapidly and more readily than females. InA. hebraeum, attachment of males was induced by eight compounds (benzaldehyde, benzyl alcohol, 2,6-dichlorophenol, heptadecane, 2-methyl propanoic acid, methyl salicylate, o-nitrophenol and salicylaldehyde) and attachment of females by six compounds (benzaldehyde, benzyl alcohol, 2,6-dichlorophenol, 2-methyl propanoic acid, o-nitrophenol and salicylaldehyde). InA. variegatum, attachment of males was induced by seven compounds (benzaldehyde, benzyl alcohol, 2,6-dichlorophenol, heptadecane, methyl salicylate, o-nitrophenol and salicyladeehyde) and attachment of females by two compounds (methyl salicylate and o-nitrophenol). A mixture of the nine compounds and extracts of prefed males ofA. herbraeum andA. variegatum induced attachment in males of both species and in femaleA. hebraeum; in these tests, attachment of femaleA. variegatum was only induced by extracts of conspecific males.  相似文献

In field assays, laboratory-reared unfed male and female ticks were released 20 cm downwind of CO2/pheromone release sites; the numbers of ticks that aggregated at the release sites were recorded after 30 min. InA. variegatum, aggregation was induced by methyl salicylate,o-nitrophenol, 2,6-dichlorophenol, phenylacetaldehyde and some mixtures containing these compounds; a strong aggregation response was induced by an extract of five prefed malesA. variegatum and a weak response was induced by an extract of 50 prefed males ofA. hebraeum. InA. hebraeum, aggregation was induced by phenylacetaldehyde, mixtures of compounds that included phenylacetaldehyde, extracts of 50 prefed males ofA. hebraeum orA. variegatum and 50 live prefed males ofA. hebraeum. InA. variegatum, aggregation was inhibited if compounds that do not occur naturally in the AAP of the species were included in mixtures. InA. hebraeum, phenylacetaldehyde appeared to act as an arrestant for ticks that had been attracted to release sites by other compounds.  相似文献

In field assays, laboratory-reared unfed male and female ticks were released 4 m downwind of treated filter-paper targets. Carbon dioxide was used to activate the ticks and the numbers attracted to the targets were recorded. Adults ofA. hebraeum were attracted to six compounds (benzaldehyde, 2,6-dichlorophenol, heptadecane, methyl salicylate, nonanoic acid and o-nitrophenol) and adults ofA. variegatum to four compounds (2,6-dichlorophenol, methyl salicylate, nonanoic acid and o-nitrophenol). The most important attractant forA. hebraeum was o-nitrophenol; methyl salicylate and o-nitrophenol were the most important attractants forA. variegatum. Nonanoic acid and o-nitrophenol attracted significantly more female than maleA. hebraeum. Both species were also attracted by a mixture of the nine compounds.  相似文献

The attraction of nymphs of the ticksAmblyomma hebraeum andA. variegatum to CO2 alone and CO2 together with the male-produced aggregation-attachment pheromone (AAP) was investigated. Matching experiments on the attraction of the adults of the two species were run for comparison. Nymphs ofA. hebraeum were strongly attracted to sources of CO2 and CO2/AAP at distances of 10 and 15 m. At distances of 20 and 25 m significantly more nymphs were attracted to CO2/AAP than to CO2 alone. Adults ofA. hebraeum were not attracted to CO2 alone at any distance; these ticks were attracted to CO2 and AAP together for distances of up to 25 m. The patterns of attraction of nymphs and adults ofA. variegatum at 10 m were similar to those recorded inA. hebraeum.  相似文献

Losses in domestic ruminants caused by heartwater (Cowdria ruminantium infection) in Zimbabwe and Mozambique are greater when the vector isAmblyomma hebraeum than when the vector isA. variegatum. It has been suggested that the epidemiology of the disease may be influenced by the rates at which unfed adults of these two tick species attach to uninfested hosts (i.e. in the absence of the male-produced aggregation-attachment pheromone [AAP]). In this study we confined unfed males ofA. hebraeum andA. variegatum on uninfested cattle, sheep and rabbits and recorded their attachment rates. Males of both species attached more rapidly on cattle than on sheep or rabbits. Males ofA. hebraeum attached more rapidly than males ofA. variegatum on all three host species. The differences in the attachment rates between the two species were much greater on sheep and rabbits than on cattle. The findings suggest that in the absence of AAP, pioneer males of both tick species may attach to cattle, and pioneer males ofA. hebraeum may also attach to sheep. The differences in the attachment rates ofA. hebraeum andA. variegatum provide a possible explanation for observed differences in the epidemiology of heartwater associated with these two vector species.  相似文献

Fifty percent of anAmblyomma variegatum female population were able to find upwind-positioned targets containing the synthetic aggregation-attachment pheromone of this species or the pheromone componento-nitrophenol alone. The ticks were not attracted to the other components of the pheromone: methyl-salicylate and pelargonic acid. Male Amblyomma variegatum ticks feeding on a host release a mixture of o -nitrophenol and methyl salicylate which serves to attract conspecifics. The behavioural responses of A. variegatum on a servosphere to these volatiles presented in an air stream are detailed here. In still air, ticks walked on all eight legs, but with long halts. In contrast, the air stream caused continuous walking and induced a reaching response where the forelegs actively sampled the air. Such reaching increased the angular velocity and reduced walking speed, effects that were amplified in the presence of vapours from o -nitrophenol and methyl salicylate in the air flowing over the ticks. Vapour from a 1:1 mixture of o -nitrophenol and methyl salicylate was attractive over a 104-fold concentration range providing an increase in upwind displacement of 20–40%, significantly higher than the natural ratio where o -nitrophenol vapour predominates. Although the responses to o -nitrophenol vapour were variable when presented alone, this chemical was consistently attractive when delivered with steer hair odour – unattractive on its own. Moreover, the upwind walk to this combination did not cause a change in speed or angular velocity. This supports the hypothesis that the response to the pheromone is enhanced by host odour. It was determined previously that unfed adults ofAmblyomma hebraeum become active in response to carbon dioxide but are not attracted by it. We have now shown in field experiments that males and females which are activated by carbon dioxide are attracted to the aggregation-attachment pheromone produced by males after a period of feeding on the host. Unfed adults were attracted too-nitrophenol, a component of the aggregation-attachment pheromone ofAmblyomma variegatum, pre-fed males ofA. hebraeum, and extracts of such males. Live males were more attractive than the extracts which, in turn, were more attractive thano-nitrophenol. The ticks were attracted to sources ofo-nitrophenol from as far away as 11 m in 90 min. Simple traps, baited with carbon dioxide and aggregation-attachment pheromone (from pre-fed males or extracts of pre-fed males), were used to capture unfed adults that had been released in the field. The response of unfed adults to carbon dioxide and the aggregation-attachment pheromone presents an obvious host-location mechanism and a means for the ticks to discriminate between suitable hosts (those with males already attached) and potentially unsuitable hosts (those with no males attached).  相似文献

Three low molecular weight compounds were found in hexane: diethyl ether extracts of fed males of the African ticks,Amblyomma variegatum (tropical bont tick) andA. hebraeum (bont tick), namely,o-nitrophenol, methyl salicylate and 2,6-dichlorophenol. These same compounds were also found in a rinse of fedA. variegatum males, but were absent or present in only trace amounts in a rinse of fedA. hebraeum males.o-Nitrophenol and methyl salicylate were present in much higher concentrations (i.e., amounts/tick) inA. variegatum than inA. hebraeum. 2,6-Dichlorophenol was also more abundant inA. variegatum than inA. hebraeum, but the differences were not as great as with the former two compounds. Extraction in hexane over a 3-week period revealed four additional compounds, benzaldehyde, benzyl alcohol, benzothiazole and nonanoic acid. The first three compounds were found in males of both species; nonanoic acid was found only inA. hebraeum males. Published reports consistently show strong attraction byo-nitrophenol and methyl salicylate for both sexes of the two bont tick species; 2,6-dichlorophenol and benzaldehyde have been reported to be attractive to both sexes ofA. hebraeum. At the beginning of the 1999 rainy season, three traditional cattle herds were monitored for 48 days while grazing in the bushy savannah of southwestern Burkina Faso. Cattle in each herd were caught on several occasions each day and the attached ticks were counted. This confirmed that Amblyomma variegatum Fabricius (Acari: Ixodidae) adults picked up in the pastures mainly attach to the interdigital areas (87% of the 791 ticks captured), and reach the predilection sites later (chest and udder/inguinal area) when the animals lie down. As many females as males attached to the hosts, but the seasonal distribution was very heterogeneous, with only a few females attaching as long as the humidity rate remained low. It is suggested that this prevents eggs from being laid when conditions are not optimal for their survival and that of the larvae. Ticks attached all day but the number picked up hourly and daily varied greatly according to their density on the pasture. As a general trend, confirmed by another study carried out in 2005, the number of ticks picked up daily increased from less than one tick/animal/day, before the onset of the rainy season, to 6.5 (+/- 1.5) ticks/animal/day on average during the infestation peak, which lasted 6-8 weeks, until early or mid-July. The number then decreased to less than one tick/animal/day from the end of July onwards. The infestation on the predilection sites followed the same trend, with a daily tick burden increase of three to five A. variegatum adults, depending on herd and year, during the infestation peak.  相似文献

Pheromone detecting sensory neurons in moths are known to be highly sensitive and selective. Female-emitted sexual pheromones are normally mixtures of a few to several components. However, not much is known about how receptor neurons respond to blends of compounds. In the present study we investigated how four physiological types of pheromone component-selective neurons responded to binary mixtures or to the complete blend in the turnip moth Agrotis segetum. We found that responses to mixtures only rarely differed from that to the excitatory component alone. The mixture interactions were exclusively suppressive and occurred only at high concentrations. Therefore we conclude that the, in A. segetum, commonly observed mixture interactions observed in higher brain centra are mainly the result of central nervous processing and that information about the pheromone components reaches the antennal lobes virtually unaltered. In addition, we found a physiological type of receptor neuron, responding selectively to one of the female-emitted pheromone components, that has previously not been observed in the Swedish population.  相似文献

The ticks Amblyomma hebraeum and A. variegatum are the main vectors of heartwater, a disease of ruminants caused by Cowdria ruminantium, in the agricultural areas of Zimbabwe. At present, A. hebraeum is widely distributed in the dry southern lowveld, and occurs in at least seven foci in the higher rainfall highveld. Amblyomma variegatum occurs in the Zambezi valley and surrounding dry lowveld areas in the northwest. The distribution of A. hebraeum has changed considerably over the past 70 years, while that of A. variegatum appears to have remained fairly static. The distribution patterns of both species in Zimbabwe display anomalous features; the ticks occur in areas of lowest predicted climatic suitability for survival and development and in areas where the densities of cattle, the most important domestic host, are lowest. The only factor favouring the survival of the species in the lowveld habitats in which they occur is the presence of alternative wildlife hosts for the adult stage. Their absence from more climatically favourable highveld habitats appears to have been the result of intensive acaricide treatment of cattle over a long period and a historic absence of significant numbers of wildlife hosts. Eradication of A. hebraeum and A. variegatum by intensive acaricide treatment of cattle can be achieved in the absence of significant numbers of alternative hosts, because of the long attachment and feeding periods of the adults of these tick species. However, eradication becomes impossible when alternative hosts for the adult stage are present, because a pheromone emitted by attached males attracts the unfed nymphal and adult stages to infested hosts. The unfed ticks are not attracted to uninfested hosts, such as acaricide-treated cattle.Regular acaricide treatment of cattle is expensive and so, for economic reasons, the Government of Zimbabwe is no longer enforcing a policy of strict tick control. It is likely that reduced tick control will result in the spread of Amblyomma ticks to previously uninfested areas. Added to this, recent introductions of various wildlife species to highveld commercial farming areas have created conditions in which the ticks could become established in higher rainfall areas. Amblyomma hebraeum is more likely to spread than A. variegatum, because its adults parasitize a wider range of wildlife hosts (warthogs, medium to large-sized antelope, giraffe, buffalo and rhinoceros), whereas adults of A. variegatum appear to be largely restricted to one wildlife species (buffalo) in Zimbabwe, the distribution of which is now confined to very limited areas of the country, as part of foot and mouth disease control measures. A model to predict the rate of spread of A. hebraeum through the highveld is described.Possible control options for dealing with the spread of Amblyomma ticks and heartwater to previous unaffected highveld areas, include (1) continuation of intensive acaricide treatment of cattle to prevent the spread, (2) establishment of a buffer zone of intensive tick control around affected areas to contain the spread and (3) allow the spread to occur and control heartwater by means of immunization. An economic analysis to determine the costs and benefits of the control options, which takes into account the development of Amblyomma-specific tick control technologies and improved heartwater vaccines, is recommended.Deceased.  相似文献

ABSTRACT. Field tests showed ( S )-(-)- cis -verbenol and ( R )-(+)- trans -verbenol in combination with a second pheromonal component, 2-methyl-3-buten-2-ol, more attractive than the combination with their optical antipodes. Inhibition of response to the attractant component, ( S )-(-)- cis -verbenol, by high concentrations of its optical antipode did not occur. No significant differences were noted for response to the attractant, ( S )-(-)- cis -verbenol and 2-methyl-3-buten-2-ol, with the addition of either ipsdienol enantiomer or a racemic mixture of ipsdienol enantiomers. Electroantennogram (EAG) studies correlated well with the behavioural studies. EAGs recorded from male and female beetles revealed both sexes to have a lower threshold for the pheromone, ( S )-(-)- cis -verbenol, than its host terpene precursor, (-)- alpha -pinene. EAGs showed a greater number of acceptors for (-)- alpha-pinene in males than in females. EAGs at acceptor saturation to the enantiomers of alpha -pinene and the verbenol isomers showed males more responsive to (-)- alpha -pinene, (±)- cis -verbenol, and ( R )-(+)- trans -verbenol. Significantly greater EAGs were elicited in females than in males to (-)- alpha -pinene, and (±)- and ( S )-(-)- cis -verbenol. No significant differences in EAGs of females to the enantiomers of trans -verbenol were noted. EAGs showed similar thresholds in males and females to the pheromone component, 2-methyl-3-buten-2-ol; however, female response at threshold was significantly greater than male response. The results are discussed with regard to olfactory acceptor evolution.  相似文献

Nonlinear interactions in the human visual system were studied using visual evoked potentials (VEPs). In one experiment (superimposed condition), all segments of a dartboard pattern were contrast reversed in time by a sum of two sinusoidal signals. In a second experiment (lateral condition), segments in some regions of the dartboard pattern were contrast reversed by a single sinusoid of one frequency, while segments in other (contiguous) regions of the pattern were contrast reversed by a single sinusoid of another frequency. An identical set of ten frequency pairs was used in each experiment. The frequency pairs were chosen such that the difference between frequencies in each pair was 2 Hz. Amplitudes and phases of the sum and difference frequency components of the VEP (intermodulation terms) were retrieved by Fourier analysis and served as measures of nonlinear interactions. The use of input pairs with a fixed separation in frequency enabled the estimation of the temporal characteristics of the visual pathways prior to a second linear stage. We investigated the effect of physiological state of adult ticks (Amblyomma hebraeum Koch; Acari: Ixodidae) on their responses to a CO2-enriched atmosphere. Ticks were exposed to CO2 or air in an experimental chamber; we measured the effects on questing, movement and latency (the time after exposure to CO2 or air when ticks began questing or moving). This study confirmed the hypothesis that responsiveness to CO2 is enhanced during host-seeking periods of the life cycle and reduced at other times. Thus, whereas newly moulted ticks quested infrequently when exposed to CO2, questing increased over the 6 week period following moulting. Likewise, in 6 week old ticks, movement in CO2 was significantly greater than in air. When a partially fed female below approximately ten times its unfed weight is removed from a host, it reattaches if subsequently presented to another host. Such females quested vigorously in CO2 but not in air. When a partially fed female above approximately ten times its unfed weight is removed from a host, it usually does not subsequently reattach. Such females quested infrequently, there being no significant difference between air and CO2. Males removed from a host at any stage of feeding reattach readily when given the opportunity. Combining a new in vitro bioassay with the analytical method capillary gas chromatography-mass spectrometry, the aggregation-attachment pheromone produced by fed males of the tropical bont tick Amblyomma variegatum was shown to consist of o-nitrophenol, methyl salicylate and pelargonic acid in the approximate amounts of 2/1/8 × 10?6 g/tick. A synthetic pheromone blend composed of those three volatile compounds evoked an aggregation response in unfed males and females in a bioassay comparable to the response to a natural pheromone source. Of the individual components. only o-nitrophenol induced a significant, although not complete aggregation response. Methyl salicylate and pelargonic acid contribute to complete pheromone activity, but induce no aggregation response at all, when offered separately.  相似文献

Recently, chemical analysis of solvent rinses of the external surfaces of pheromone glands from female Manduca sexta revealed a blend of 12 aldehydes, including the previously identified sex pheromone component, (E,Z)-10,12-hexadecadienal (bombykal). Previous electrophysiological studies showed that olfactory (deutocerebral) interneurons in the antennal lobes of males exhibited a wide range of responsiveness to pheromonal stimulation of the ipsilateral antenna. These experiments were performed with crude extracts of pheromone glands as well as two synthetic compounds: the major pheromone component, bombykal, and (E,Z)-11,13-pentadecadienal, a mimic of a second component of the female's pheromone blend. Using intracellular methods, we have now reexamined similar olfactory interneurons, using each of the 12 chemically identified components as well as synthetic blends of various combinations of them. Eight of the 12 components isolated from female glands elicited some form of response in olfactory interneurons in males. In accordance with biochemical and behavioral data, the most potent are bombykal and two trienals, (E,E,E)- and (E,E,Z)-10,12,14-hexadecatrienal. We also conclude that the C15 dienal is selective for one of the trienal receptors on the antenna, but is much less potent than the natural trienal stimulant.  相似文献
