Cutaneous receptive fields of dorsal roots T11 through S3 in rats were mapped by recording dorsal root multiple unit activity. The receptive fields, having the shape of transverse bands on the skin, are arranged in order according to their dorsal roots, and there are extensive overlaps between adjacent fields. Routes followed by peripheral sensory innervation of skin were also studied by observing effects of various methods of skin denervation on dorsal root responses to cutaneous stimulation. The results are discussed with respect to the somatosensory control of female rat mating reflexes.  相似文献

It has been thought that spinal dorsal horn neurons receive convergent inputs from not only somatosensory but also visceral pathways. For instance, the referred pain is presumed to be due to the convergence of sensory inputs from cardiac and shoulder receptive fields. However, precise investigation has not been made from dorsal horn neurons yet, because of difficulty in studying the pathways from those regions by means of conventional electrophysiology. The purpose of this study is to clarify the convergent inputs to single dorsal horn neurons from wide receptive fields using an in vivo patch-clamp recording technique from the superficial spinal dorsal horn and an intracellular recording from dorsal root ganglion neurons that keep physiological connections with the peripheral sites. Identified dorsal root ganglion neurons received an input from a quite small area, about 1 x 1 mm in width of the skin. In contrast, substantia gelatinosa neurons in the spinal cord received inputs from an unexpectedly wide area of the skin. Previous extracellular recordings have, however, revealed that substantia gelatinosa neurons have small receptive field. This discrepancy is probably due mainly to an availability of the in vivo patch-clamp method to analyze sub-threshold synaptic responses. In contrast, the extracellular recording technique allows us to analyze predominantly the firing frequency of neurons. Thus, the in vivo patch-clamp recordings from dorsal horn neurons and the intracellular recordings from DRG neurons will be useful for well understanding the sensory processing in the spinal cord.  相似文献

The present study was designed (1) to characterize the subliminal responses of dorsal horn neurons to stimulation of the sural nerve, and (2) to correlate the type of response to this stimulus with the responses to natural mechanical stimulation of the skin. To accomplish this, intracellular and extracellular recordings were carried out in L6 and L7 dorsal horn neurons in the cat. The excitatory responses of each cell to electrical stimulation of the sural nerve and to mechanical stimulation of the skin were noted.

Of 35 dorsal horn cells recorded intracellularly, 11 responded with impulses to sural nerve stimulation, 9 responded with excitatory postsynaptic potentials (EPSPs) but not impulses, and 15 had no excitatory responses to this stimulus. The type of response to sural nerve stimulation was strongly correlated with receptive field modality. Most cells receiving an input from high-threshold cutaneous mechanoreceptors responded with impulses or gave no excitatory response to sural nerve stimulation, whereas most cells that had only low-threshold mechanoreceptor input responded with EPSPs only or gave no response. In cells with only low-threshold (LT) mechanoreceptive input, response to sural nerve stimulation was highly correlated with receptive field locus. Those LT cells with no excitatory responses to sural nerve stimulation had receptive fields confined to the foot and/or toes, whereas those that gave EPSPs had more proximal receptive fields. The possible significance of these data with reference to changes observed after lesions, such as increased response to sural nerve stimulation, increased receptive field size, and somatotopic reorganization, is discussed.  相似文献

The flagellar root system of Entosiphon sulcatum (Dujardin) Stein (Euglenophyceae) is described and compared with kinetoplastid and other euglenoid systems. An asymmetric pattern of three microtubular roots, one between the two flagellar basal bodies and one on either side (here called the intermediate, dorsal, and ventral roots), is consistent within the euglenoid flagellates studied thus far. The dorsal root is associated with the basal body of the anterior flagellum (F1) and lies on the left dorsal side of the basal body complex. Originating between the two flagellar basal bodies, and associated with the basal body of the trailing flagellum (F2), the intermediate root is morphologically distinguished by fibrils interconnecting the individual microtubules to one another and to the over lying reservoir membrane. The intermediate root is often borne on a ridge projecting into the reservoir. The ventral root originates near the F2 basal body and lies on the right ventral side of the cell. Fibrillar connections link the membrane of F2 with the reservoir membrane at the reservoircanal transition level. A large cross-banded fiber joins the two flagellar basal bodies, and a series of smaller striated fibers links the anterior accessory and flagellar basal bodies. Large nonstriated fibers extend from the basal body complex posteriorly into the cytoplasm.  相似文献

  1. Response properties of neurons in the dorsal granular ridge (DGR) of the little skate, Raja erinacea, were studied in decerebrate, curarized fish. Sensory responses included proprioceptive (426 of 952; 45%) and electroreceptive units (526 of 952; 55%). Electroreceptive units responded to weak electric fields with a higher threshold than lower-order units and had large ipsilateral receptive fields, whose exact boundaries were often unclear but contained smaller, identifiable best areas. Proprioceptive units responded to displacement of the ipsilateral fin and were either position-or movement-sensitive.
  2. Both proprioceptive and electroreceptive units showed a progression of receptive fields from anterior to posterior body in the rostral to caudal direction along the length of DGR. Sensory maps in DGR projected homotopically to the electrosensory somatotopy in the dorsal nucleus. Peak evoked potentials and units responding to local DGR stimulation occurred only in areas of the dorsal nucleus with receptive fields located within the composite receptive field at the DGR stimulation site.
  3. Single shocks to DGR produced a short spike train followed by a prolonged suppression period in the medullary dorsal nucleus. These results have implications for the role of the parallel fiber system in medullary electrosensory processing.

In order to obtain further evidence of putative neurotransmitters in primary sensory neurons and interneurons in the dorsal spinal cord, we have studied the effects of unilateral section of dorsal roots and unilateral occlusion of the dorsal spinal artery on cholinergic enzyme activity and on selected amino acid levels in the spinal cord. One week after sectioning dorsal roots from caudal cervical (C7) to cranial thoracic (T2) levels, the specific activity of choline acetyltransferase (ChAT) was significantly decreased and acetylcholinesterase (AChE) showed a tendency to decrease in the dorsal quadrant on the operated side of the spinal cord. Dorsal root sectioning had little effect on the levels of free glutamic acid or other amino acids in the dorsal spinal cord. These results suggest that primary sensory neurons may include some cholinergic axons, and that levels of putative amino acid transmitters are not regulated by materials supplied by axonal transport from the dorsal root ganglia. By contrast, one week following unilateral occlusion of the dorsal spinal artery, the activities of ChAT and AChE were unchanged in the operated quadrant of the spinal cord, while decreases of Asp, Glu, and GABA, and an increase in Tau were detected. Salinization of crop fields is a pressing matter for sustainable agriculture under desertification and is largely attributed to root absorptive functions of the major crops such as maize. The rates of water and ion absorption of intact root system of maize plants were measured under the salinized condition, and the salt absorptive function of maize roots was analyzed by applying different two kinetic models of root ion absorption (i.e. the concentration dependent model and the transpiration integrated model). The absorption rates for salinization ions (Na+, Cl?, Ca2+ and Mg2+) were found to depend on ion mass flow through roots driven by the transpiration, and therefore the transpiration integrated model represented more accurately rates of root ion absorption. The root absorption of salinization ions was characterized quantitatively by two model parameters of Qmax and KM involved in the transpiration integrated model, which are considered to relate to the potential absorbing power and the ion affinity of transport proteins on root cell membranes, respectively.  相似文献

This study describes the projection of cervical spinal afferent nerve fibers to the medulla in the brush-tailed possum, a marsupial mammal. After single dorsal roots (between C2 and T1) were cut in a series of animals, the Fink-Heimer method was used to demonstrate the projection fields of fibers entering the CNS via specific dorsal roots. In the high cervical spinal cord, afferent fibers from each dorsal root form a discrete layer in the dorsal funiculus. The flattened laminae from upper cervical levels are lateral and those from lower cervical levels are medial within the dorsal columns. All afferent fibers at this level are separated from gray matter by the corticospinal fibers in the dorsal funiculus. All cervical roots project throughout most of the length of the well-developed main cuneate nucleus in a loosely segmentotopic fashion. Fibers from rostral roots enter more lateral parts of the nucleus, and fibers from lower levels pass to more medial areas; but terminal projection fields are typically large and overlap extensively. At more rostral medullary levels, fibers from all cervical dorsal roots also reach the external cuneate nucleus. The spatial arrangement here is more complex and more extensively overlapped than in the cuneate nucleus. Rostral cervical root fibers reach ventral and ventrolateral areas of the external cuneate nucleus and continue to its rostral pole; more caudal root fibers project to more dorsal and medial regions within the nucleus. These results demonstrate that projection patterns of spinal afferents in this marsupial are similar to those seen in the few placental species for which detailed data concerning this system are available.  相似文献

Proteins synthesized by soma located in L4 dorsal root ganglia and supplied to the axonal branches extending centrally in the dorsal root and peripherally towards the sciatic nerve were analyzed for radioactivity following injections of [3H] leucine into the L4 dorsal root ganglia. All proteins located in the dorsal root and sciatic nerve were analyzed by SDS acrylamide gel electrophoresis at various times post injection. The differences in radioactivity between the dorsal root and sciatic nerve proteins were mainly quantitative and not qualitative, with many proteins of various molecular weight ranges being transported into both segments. Generally, it appears that in both axonal branches the high molecular weight proteins are transported at the highest rate, medium weights slower and low molecular weight proteins slowest. More proteins of high and low molecular weights are transported into the dorsal root whereas more of those of medium molecular weight are transported towards the sciatic nerve.  相似文献

The intensity of axon transport (AT) of3H-hydrocortisone via fibers of the ventral (anterograde AT) and dorsal (retrograde AT) roots was shown to depend on the concentration of labelled hormone injected into the gray matter of theL 5-L6 spinal segments. The concentration dependence of anterograde flow (axons of motoneurons) was manifested both in the velocity and amount of transported hormone, while only velocity was modified in retrograde AT (in central processes of dorsal root ganglion neurons). In old rats, a concentration-dependent increase in the amount of labelled hormone was observed only within rather narrow range of activity of injected3H-hydrocortisone (from 0.036 to 0.108 MBq/μl). An increase in the activity of labelled hormone to 0.14–0.18 MBq/μl suppressed its uptake into the ventral root fibers. At the same time, modifications of the AT velocity showed direct dependence on the concentration within the entire studied range of3H-hydrocortisone activity. similar modification of the velocity were observed in the dorsal roots of old rats. It is supposed that an increase in the intracellular concentration of a glucocorticoid activates the mechanisms of hormone removal from the motoneurons, and these mechanisms become sharply weaked in old age.  相似文献

Summary Morphological changes in the motor and sensory neurons in the lumbar spinal cord and the dorsal root ganglia were investigated at different survival times following the injection of the B subunit of cholera toxin (CTB) into the medial gastrocnemius muscle. Unconjugated CTB, visualized immunohistochemically, was found to be retrogradely transported through ventral and dorsal roots to motor neurons in the anterior horn, each lamina in the posterior horn, and ganglion cells in the dorsal root ganglia at L3–L6. The largest numbers of labeled motor neurons and ganglion cells were observed 72 h after the injection of CTB. Thereafter, labeled ganglion cells were significantly decreased in number, whereas the amount of labeled motor neurons showed a slight reduction. Motor neurons had extensive dendritic trees filled with CTB, reaching lamina VII and even the pia mater of the lateral funiculus. Labeling was also seen in the posterior horn, but the central and medial parts of laminae II and III had the most extensively labeled varicose fibers, the origin of which was the dorsal root ganglion cells. The results indicate that CTB is taken up by nerve terminals and can serve as a sensitive retrogradely transported marker for identifying neurons that innervate a specific muscle.  相似文献

The changes in cell-wall peroxidase (POD) activity and H2O2 level in roots of NaCl-stressed rice seedlings and their correlation with root growth were investigated. Increasing concentrations of NaCl from 50 to 150 mM progressively reduced root growth and increased ionically bound cell-wall POD activity. NaCl had no effect on covalently bound cell-wall POD activities. The reduction of root growth by NaCl is closely correlated with the increase in H2O2 level. Exogenous H2O2 was found to inhibit root growth of rice seedlings. Since ammonium and proline accumulation are associated with root growth inhibition caused by NaCl, we determined the effects of NH4Cl or proline on root growth, cell-wall POD activity and H2O2level in roots. External application of NH4Cl or proline markedly inhibited root growth, increased cell-wall POD activity and increased H2O2 level in roots of rice seedlings in the absence of NaCl. An increase in cell-wall POD activity and H2O2 level preceded inhibition of root growth caused by NaCl, NH4Cl or proline. NaCl or proline treatment also increased NADH-POD and diamine oxidase (DAO) activities in roots of rice seedlings, suggesting that NADH-POD and DAO contribute to the H2O2 generation in the cell wall of NaCl- or proline-treated roots. NH4Cl treatment increased NADH-POD activity but had no effect on DAO activity, suggesting that NADH-POD but not DAO is responsible for H2O2 generation in cell wall of NH4Cl-treated roots.  相似文献

The methane emitted from rice fields originates to a large part (up to 60%) from plant photosynthesis and is formed on the rice roots by methanogenic archaea. To investigate to which extent root colonization controls methane (CH4) emission, we pulse‐labeled rice microcosms with 13CO2 to determine the rates of 13CH4 emission exclusively derived from photosynthates. We also measured emission of total CH4 (12+13CH4), which was largely produced in the soil. The total abundances of archaea and methanogens on the roots and in the soil were analysed by quantitative polymerase chain reaction of the archaeal 16S rRNA gene and the mcrA gene coding for a subunit of the methyl coenzyme M reductase respectively. The composition of archaeal and methanogenic communities was determined with terminal restriction fragment length polymorphism (T‐RFLP). During the vegetative growth stages, emission rates of 13CH4 linearly increased with the abundance of methanogenic archaea on the roots and then decreased during the last plant growth stage. Rates of 13CH4 emission and the abundance of methanogenic archaea were lower when the rice was grown in quartz‐vermiculite with only 10% rice soil. Rates of total CH4 emission were not systematically related to the abundance of methanogenic archaea in soil plus roots. The composition of the archaeal communities was similar under all conditions; however, the analysis of mcrA genes indicated that the methanogens differed between the soil and root. Our results support the hypothesis that rates of photosynthesis‐driven CH4 emission are limited by the abundance of methanogens on the roots.  相似文献

This paper aims to investigate the effect of the root architecture on the shear strength increment provided by plant roots in the soil. In situ shear tests were conducted for this purpose. Five plant species – Hibiscus tiliaceus L. (Linden hibiscus), Mallotus japonicus (Thunb.) Muell.-Arg. (Japanese Mallotus), Sapium sebiferum (L.) Roxb. (Chinese tallow tree), Casuarina equisetifolia L. (ironwood), and Leucaena leucocephala (Lam.) (white popinac) – were used in this study. Tensile tests on roots of various diameters and surveys on the root system structure were carried out for each of the plant species tested. The shear strength increments (ΔS) provided by the roots of Linden hibiscus, Japanese Mallotus, Chinese tallow tree, ironwood, and white popinac were 0.34tR, 0.462tR, 0.688tR, 0.3tR, and 0.87tR, respectively, when tR was estimated on the basis of the tensile root strength crossing through the shear plane. The shear strength increment provided by plant roots with conspicuous oblique and vertical roots was greater than that of root structures in which lateral roots were dominant. In comparison with other types of root architecture, the R-type root architecture was found to be the most effective root system against shear failure in the soil. Its shear strength increment was slightly greater than that with the V-type root architecture, followed by the VH-type root architecture. The shear strength increment provided by plants with the H-type root architecture was less effective than that contributed by plant species with other types of root architecture.  相似文献

Evoked electrical discharges in the spinal cord roots and dorsal surface ipsilateral to the previously severed sciatic nerve (as well as on the contralateral side) were investigated in rats one, three, seven, and 14 days after tractotomy. Monosynaptic reflex discharges in the ventral roots were found to return to 20–40% of the level of this parameter as measured on the contralateral side within seven and 14 days after tractotomy. Mean amplitude of antidromic dorsal root discharges, afferent peak, and the N1 component of potential(s) at the dorsal surface ipsilateral to the severed nerve barely altered, remaining significantly lower than on the contralateral side. Although monomeric Al species are often toxic in acidic soils, the effects of the aluminate ion (Al(OH) 4 ) on roots grown in alkaline media are still unclear. Dilute, alkaline (pH 9.5) nutrient solutions were used to investigate the effects of Al(OH) 4 on root growth of mungbean (Vigna radiata L.). Root growth was reduced by 13% after 3 d growth in solutions with an Al(OH) 4 activity of 16 μM and no detectable polycationic Al (Al13). This decrease in root growth was associated with the formation of lesions on the root tips (due to the rupturing of the epidermal and outer cortical cells) and a slight limitation to root hair growth (particularly on the lateral roots). When roots displaying these symptoms were transferred to fresh Al(OH) 4 solutions for a further 12 h, no root tip lesions were observed and root hair growth on the lateral roots improved. The symptoms were similar to those induced by Al13 at concentrations as low as 0.50 μM Al which are below the detection limit of the ferron method. Thus, Al(OH) 4 is considered to be non-toxic, with the observed reduction in root growth in solutions containing Al(OH) 4 due to the gradual formation of toxic Al13 in the bulk nutrient solution resulting from the acidification of the alkaline nutrient solution by the plant roots.  相似文献

The initial stage of information processing by the visual system reduces the information contained in the continuous image on the retina into a discrete set of responses which are carried from the lateral geniculate nucleus (LGN) to the visual cortex.-1. The optimal sampling of the light intensity distribution in the visual environment is achieved only if each channel in the visual pathways carries undistorted information corresponding to an image element. The visual system approaches as closely as possible the scheme of optimal spatial sampling, retaining the full information on the low spatial frequency content of the object light intensity. The ideal receptive field of a sustained LGN cell is then of the form J 1 (Kr)/Kr.-2. The experimentally determined receptive fields of sustained LGN cells (and to some extent retinal ganglion cells as well) in cat closely resemble the functional form J 1 (Kr)/Kr. The centre-surround organization of the receptive fields is therefore understood as a scheme which leads to a maximal information flow through the visual pathways.-3. The optimal sampling scheme cannot be realized by the retina alone, because of restrictions on the size of neural networks. It is therefore constructed in two stages, ending at the LGN level. A recombination of ganglion cell signals into optimal receptive fields is a major role of the LGN.  相似文献

The contrasting hydraulic properties of wheat (Triticum aestivum), narrow-leafed lupin (Lupinus angustifolius), and yellow lupin (Lupinus luteus) roots were identified by integrating measurements of water flow across different structural levels of organization with anatomy and modeling. Anatomy played a major role in root hydraulics, influencing axial conductance (Lax) and the distribution of water uptake along the root, with a more localized role for aquaporins (AQPs). Lupin roots had greater Lax than wheat roots, due to greater xylem development. Lax and root hydraulic conductance (Lr) were related to each other, such that both variables increased with distance from the root tip in lupin roots. Lax and Lr were constant with distance from the tip in wheat roots. Despite these contrasting behaviors, the hydraulic conductivity of root cells (Lpc) was similar for all species and increased from the root surface toward the endodermis. Lpc was largely controlled by AQPs, as demonstrated by dramatic reductions in Lpc by the AQP blocker mercury. Modeling the root as a series of concentric, cylindrical membranes, and the inhibition of AQP activity at the root level, indicated that water flow in lupin roots occurred primarily through the apoplast, without crossing membranes and without the involvement of AQPs. In contrast, water flow across wheat roots crossed mercury-sensitive AQPs in the endodermis, which significantly influenced Lr. This study demonstrates the importance of examining root morphology and anatomy in assessing the role of AQPs in root hydraulics.  相似文献

The dragonfly, (Aeshna, Anax) which feeds on small flying insects, requires a visual system capable of signaling the movements of airborne prey. A group of 8 descending feature detectors in the dragonfly are tuned exclusively to moving contrasting objects. These target-selective descending neurons project from the brain to the thoracic ganglia. Their activity drives steering movement of the wings.In this study, we recorded target-selective descending neuron activity intracellularly.To define their receptive fields, we recorded responses to the movement of black square targets projected onto a screen in front of the animal. Each neuron was identified by dye injection.Target-selective descending neurons exhibit several receptive field properties. Our results show that they are strongly directionally selective. Two TSDNs, exclusively tuned to small targets, have receptive fields restricted to visual midline. Others, which are not selective for target size, have asymmetric receptive fields centered laterally.We suggest that the behavioral function of these specialized feature detectors is to steer the dragonfly during prey-tracking so as to fix the position of the prey image on the retina.
