Out of seven fungal species belonging to four genera isolated from pond and wallow soils using feathers of Pavo cristatus as bait, four species viz., Aspergillus flavus, Aspergillus niger, Chrysosporium pseudomerdarium and Trichophyton ajelloi were most frequent. Chrysosporium and Trichophyton spp. were more pathogenic on Culex quinquefasciatus larvae than Aspergillus and Penicillium. The bioefficacy tests conducted as per the protocol of World Health Organization and the LC50 values calculated by the Probit analysis showed that 3rd-instar C. quinquefasciatus were more susceptible to the conidia of above fungi. Highest mortality was observed in the larvae of C. quinquefasciatus when exposed to T. ajelloi. The density of fungal conidia was greatest on the ventral brush, palmate hair and anal region of the mosquito larvae after exposing for 72 hours. The potentiality of these fungi for use in the control of C. quinquefasciatus is discussed which can be exploited as a suitable biocontrol agent in the tropics.  相似文献

Two mirids,Deraeocoris sp. andCampylomma nicolasi Reuter and one lygaeid,Geocoris ochropterus Fieber were found preying onB. tabaci (Gennadius) for the first time in Maharashtra State of India during 1987–88. Their biology and predation capacity onB. tabaci were studied in detail under laboratory conditions. The nymphal stage ofDeraeocoris sp. passed through 6 instars, whereas 5 instars in case of the remaining species.G. ochropterus, Deraeocoris sp. andC. nicolasi consumed on an average 482.5, 275.3 and 128.8 nymphs of 57.3, 25.5 and 20.6 days, respectively. Bioassay procedures for the insect growth regulators diflubenzuron (a chitin synthetase inhibitor) and methoprene (a juvenile hormone mimic) were evaluated on the mosquito Culex quinquefasciatus Say to determine the most sensitive larval stages and the effects of varying exposure periods. Sensitivity to diflubenzuron decreased gradually during instars I and II; instars III and IV were relatively insusceptible. Sensitivity to methoprene increased with larval age. Standard test procedures were devised for diflubenzuron involving continuous exposure from the early third instar and for methoprene as 6 h exposure during instar IV. Selection for ten generations of strains from Dar-es-Salaam which were multiresistant to insecticides increased the LC50 to diflubenzuron by 2.8-fold and increased the LC50 to methoprene by 3.9-fold. When compared to a standard susceptible strain from Colombo the resistance ratios of Dar-es-Salaam strains rose from 2.4 x to 6.6 x for diflubenzuron and from 6.1 x to 21.3 x for methoprene, with no significant effects detected on the pattern of cross-resistance. These results are discussed in relation to standardization of IGR testing and the need for integrated control programmes against Cx quinquefasciatus.
Résistance potentielle de Culex quinquefasciatus Say au méthoprène et au diflubenzuron, régulateurs de croissance des insectes
Résumé La détermination du stade larvaire le plus sensible et les effects de différentes durées d'exposition aux régulateurs de croissance des insectes-diflubenzuron (inhibiteur de la chitine synthétase) et méthoprène (mimétique de l'hormone juvénile)—ont été recherchés sur C. quinquefasciatus. La sensibilité au diflubenzuron diminue graduellement pendant les deux premiers stades, les stades III et IV étant relativement insensibles. La sensibilité au méthoprène augmente avec l'âge des larves. Les essais comportaient une exposition continue au diflubenzuron depuis le début du stade III et une exposition de 6 heures au méthoprène pendant le stade IV. La sélection pendant 10 générations de lignées de Dares-Salam multirésistantes aux insecticides a entraîné une augmentation due 2,8 fois de la DL 50 au diflubenzuron et de 3,9 fois au méthoprène. Comparés aux souches standards sensibles de Colombo, les taux de résistance des lignées de Dar-es-Salam se sont élevés de 2,4 à 6,6 pour le diflubenzuron et de 6,1 à 21,3 pour le méthoprène, sans effet visible significatif sur la résistance croisée. Ces résultats sont discutés en relation avec la standardisation du test IGR et les besoins de programmes de lutte intégrée contre C. quinquefasciatus.

Molecular characterization of the insecticide resistance has become a hot research topic ever since the first disease transmitting arthropod (Anopheles gambiae) genome sequence has unveiled in 2002. A recent publication of the Culex quinquefasciatus genome sequence has opened up new opportunities for molecular and comparative genomic analysis of multiple mosquito genomes to characterize the insecticide resistance. Here, we utilized a whole genome sequence of Cx. quinquefasciatus to identify putatively active members of the detoxification supergene families, namely cytochrome P450s (P450s), glutathione-S-transferases (GSTs), and choline/carboxylesterases (CCEs). The Culex genome analysis revealed 166 P450s, 40 GSTs, and 62 CCEs. Further, the comparative genomic analysis shows that these numbers are considerably higher than the other dipteran mosquitoes. These observed speciesspecific expansions of the detoxification super gene family members endorse the popular understanding of the involvement of these gene families in protecting the organism against multitudinous classes of toxic substances during its complex (aquatic and terrestrial) life cycle. Thus, the generated data set may provide an initial point to start with to characterize the insecticide resistance at a molecular level which could then lead the development of an easy to use molecular marker to monitor the incipient insecticide resistance in field environs.  相似文献

Transmission electron microscopy of the invasion of Culex quinquefasciatus by Leptolegnia chapmanii confirmed that it is a primary pathogen and revealed several differences between penetration via the gut and penetration via the integument. The latter often involved aggregations of zoospores, appressoria-like swellings of the invasive hyphae, and lateral growth of hyphae between the epicutical and endocuticle. These features were not detected in the case of gut invasion, but hyphal septa at the point of entry were apparently peculiar to this route. There was no evident tissue specificity, and death presumably resulted from generalized destruction of tissues.  相似文献

Culexpipiens quinquefasciatus (C. quinquefasciatus) is an important vector that can transmit human diseases such as West Nile virus, lymphatic filariasis, Japanese encephalitis and St. Louis encephalitis. However, very limited research concerning the humoral and cellular immune defenses of C. quinquefasciatus has been done. Here we present the research on hemocyte identification and plasma including hemocyte prophenoloxidase from C. quinquefasciatus at all developmental stages in order to obtain a complete picture of C. quinquefasciatus innate immunity. We identified hemocytes into four types: prohemocytes, oenocytoids, plasmatocytes and granulocytes. Prophenoloxidase (PPO) is an essential enzyme to induce melanization after encapsulation. PPO-positive hemocytes and plasma PPO were observed at all developmental stages. As for specific hemocyte types, prophenoloxidase was found in the plasmatocytes at larval stage alone and in the smallest prohemocytes during almost all developmental stages. Moreover, the granulocytes were PPO-positive from blood-fed female mosquitoes and oenocytoids were observed PPO-positive in pupae and in adult females after blood-feeding. As for plasma, there were different patterns of PPO in C. quinquefasciatus at different developmental stages. These results are forming a basis for further studies on the function of C. quinquefasciatus hemocytes and prophenoloxidase as well as their involvement in fighting against mosquito-borne pathogens.  相似文献

叶片生态化学计量是了解生物地球化学循环的窗口,也是认识植物的生长状况及生态适应性的关键,研究人工林叶片生态化学计量特征对进一步优化生态工程建设具有重要意义。为揭示兰州市南北两山主要绿化树种侧柏、圆柏和云杉环境适应性的异同,对其叶片生态化学计量特征进行了对比研究。结果显示:云杉、圆柏叶片C含量(496.02、497.64 g/kg)显著高于侧柏叶片(484.12 g/kg),侧柏叶片N、P含量(17.70、1.66 g/kg)均显著高于圆柏(14.04、1.42 g/kg)和云杉(15.62、1.40 g/kg)。C、N、P的变异系数均为圆柏<侧柏<云杉;云杉、圆柏叶片C∶N (33.32、35.42)、C∶P (389.57、360.52)显著高于侧柏叶片(28.17、306.07),云杉叶片N∶P (11.88)显著高于圆柏叶片(10.20)和侧柏叶片(10.98)。C∶N、C∶P、N∶P的变异系数均为圆柏<侧柏<云杉;侧柏和云杉叶片N含量与P含量正相关、与C∶N、C∶P负相关,而3种乔木叶片P含量与C∶P、N∶P负相关(且C∶P与N∶P正相关)。圆柏叶片C含量与N含量正相关,侧柏叶片C含量与N含量负相关但与叶片C∶N、C∶P正相关,侧柏和云杉叶片C∶N与C∶P正相关。冗余分析表明,SOC∶STN是影响3种乔木叶片化学计量的主导因子,圆柏叶片C、侧柏叶片P与SOC正相关,3种乔木叶片N都与SOC、SOC∶STN和SOC∶STP正相关;云杉叶片N,侧柏、云杉叶片P和STP负相关,而STP与侧柏叶片C∶P、N∶P及云杉叶片C∶N正相关;3种乔木叶片C∶N与SOC∶STN、SOC∶STP负相关。研究结果表明:面对养分贫瘠的土壤环境,3种乔木叶片生态化学计量的适应策略有所不同,其中云杉和圆柏采取防御策略,而侧柏采取竞争倾向策略。虽然云杉是兰州市较适宜的造林树种,但其生长与其他两种乔木一样,都受N元素的限制。  相似文献

Tamarins of the genusSaguinus are small-bodied New World monkeys that exhibit clawlike or modified nails. Patterns of positional behavior and habitat utilization are presented for three species,Saguinus fuscicollis, S. geoffroy, andS. mystax. These data were collected on free-ranging tamarin populations in Panama and Peru. Despite considerable differences in body weight, all three species exhibited very similar patterns of positional behavior, with quadrupedal bounding and running accounting for 43 – 52% of travel time. Leaping was the second most common locomotor activity and accounted for 31 – 41% of travel. Although each species leaped principally on small supports in the perimeter of the tree crown, approximately 20% of all leaps inS. fuscicollis involved moderate to large sized vertical trunks located in the undercanopy. Leaping between trunks was rare in the two larger tamarin species. Measurements taken on live wild-trapped adults reveal that compared toSaguinus geoffroyi andS. mystax, S. fuscicollis is characterized by a long legspan and an especially long armspan. It is proposed that inS. fuscicollis, elongated forelimbs play an important role in maneuvering and rotating the body during the in-air phase of trunk-to-trunk leaping, and increase the breaking distance needed to decelerate the body upon impact. Manganese, Mn, is a naturally abundant metal in marine sediments. During hypoxic conditions the metal converts into a bioavailable state and can reach levels that have been shown immunotoxic to the crustacean Nephrops norvegicus. For this species it has previously been shown that exposure to 15 mg L−1 of Mn decreased the number of circulating haemocytes while it for the echinoderm Asterias rubens increased the number of coelomocytes. Here, we compared if five days of exposure to the same concentration of Mn affects the bactericidal capacity of these two species and the mollusc Mytilus edulis when inoculated with the bacterium Vibrio parahaemolyticus. Viable counts of the bacteria were investigated at a time-course post-injection in the blood and the digestive glands of Mn-exposed and un-exposed (controls) animals. Accumulation of Mn was also analyzed in these tissues. When exposed to Mn the haemocyte numbers were significantly reduced in M. edulis and it was shown that the bactericidal capacity was impaired in the mussels as well as in N. norvegicus. This was most obvious in the digestive glands. These two species also showed the highest accumulation of the metal. In A. rubens the bactericidal capacity was not affected and the metal concentration was similar to the exposure concentration. After a recovery period of three days the concentration of Mn was significantly reduced in all three species. However, in M. edulis and N. norvegicus it was still double that of A. rubens which could explain the remaining bactericidal suppression observed in N. norvegicus. This study pointed out that exposure to such Mn-levels that are realistic to find in nature could have effects on the whole organism level, in terms of susceptibility to infections. The effect seemed associated to the accumulated concentration of Mn which differed on species level.  相似文献

Summary Interspecific relationship studies between A, S and D genome diploid species, and between AAGG and AABB allotetraploid species of the genus Triticum were conducted using isoenzymatic characters located in dry mature seeds. Data was analyzed by the factorial analysis of correspondences, and dendograms were obtained by two different genetic distances. The discussion of results was based on the limitations of the study, intraspecific variability differences, isoperoxidase frequency differences and chromosomal location of peroxidases in T. aestivum cv. Chinese Spring. The closest relationship was found between dicoccoides and carthlicum. Relationships found between T. turgidum L. and T. timopheevvi Zhuk., both allotetraploid species and boeoticum; this species and speltoides; tauschi and searsii, and the last two species with bicorne, are discussed at the phylogenetic level.  相似文献

A cultivation-based approach was employed to compare the culturable actinobacterial diversity associated with five marine sponge species (Craniella australiensis, Halichondria rugosa, Reniochalina sp., Sponge sp., and Stelletta tenuis). The phylogenetic affiliation of the actinobacterial isolates was assessed by 16S rDNA-RFLP analysis. A total of 181 actinobacterial strains were isolated using five different culture media (denoted as M1–M5). The type of medium exhibited significant effects on the number of actinobacteria recovered, with the highest number of isolates on M3 (63 isolates) and the lowest on M1 (12 isolates). The genera isolated were also different, with the recovery of three genera on M2 and M3, and only a single genus on M1. The number of actinobacteria isolated from the five sponge species was significantly different, with a count of 83, 36, 30, 17, and 15 isolates from S. tenuis, H. rugosa, Sponge sp., Reniochalina sp., and C. australiensis, respectively. M3 was the best isolation medium for recovery of actinobacteria from S. tenuis, H. rugosa, and Sponge sp., while no specific medium preference was observed for the recovery of actinobacteria from Reniochalina sp., and C. australiensis. The RFLP fingerprinting of 16S rDNA genes digested with HhaI revealed six different patterns, in which 16 representative 16S rDNAs were fully sequenced. Phylogenetic analysis indicated that 12 strains belong to the group Streptomyces, three strains belong to Pseudonocardia, and one strain belongs to Nocardia. Two strains C14 (from C. australiensis) and N13 (from Sponge sp.) have only 96.26% and 96.27% similarity to earlier published sequences, and are therefore potential candidates for new species. The highest diversity of three actinobacteria genera was obtained from Sponge sp., though the number of isolates was low. Two genera of actinobacteria, Streptomyces, and Pseudonocardia, were isolated from both S. tenuis and C. australiensis. Only the genus of Streptomyces was isolated from H. rugosa and Reniochalina sp. Sponge species have been demonstrated here to vary as sources of culturable actinobacterial diversity, and the methods for sampling such diversity presented may be useful for improved sampling of such diversity.  相似文献

A comparison of branchial enzyme profiles indicates that the gills of Periophthalmodon schlosseri would have a greater capacity for energy metabolism through glycolysis than those of Boleophthalmus boddaerti. Indeed, after exposure to hypoxia, or anoxia, there were significant increases in the lactate content in the gills of P. schlosseri. In addition, exposure to hypoxia or anoxia significantly lowered the glycogen level in the gills of this mudskipper. It can be deduced from these results that the glycolytic flux was increased to compensate for the decrease in ATP production through anaerobic glycolysis. Different from P. schlosseri, although there was an increase in lactate production in the gills of B. boddaerti exposed to hypoxia, there was no significant change in the branchial glycogen content, indicating that a reversed Pasteur effect might have occurred under such conditions. In contrast, anoxia induced an accumulation of lactate and a decrease in glycogen in the gills of B. boddaerti. Although lactate production in the gills of these mudskippers during hypoxia was inhibited by iodoacetate, the decreases in branchial glycogen contents could not account for the amounts of lactate formed. The branchial fructose-2,6-bisphosphate contents of these mudskippers exposed to hypoxia or anoxia decreased significantly, leaving phosphofructokinase and glycolytic rate responsive to cellular energy requirements under such conditions. Summary Turfgrass, like other major crop species, is recalcitrant to manipulation in vitro. To perform efficient genetic transformation of turfgrass, it is necessary to optimize tissue culture conditions. In most reports, plant tissue culture techniques have been applied to propagate a single cultivar or several cultivars in one species of turfgrass. In this experiment, four turfgrass genera were used, namely common bermudagrass, Cynodon dactylon [L.] Pers. (California origin); red fescue, Festuca rubra L. var. rubra ‘Shadow’; perennial ryegrass, Lolium perenne L. ‘Barbal’; and Kentucky bluegrass, Poa pratensis L. ‘Merion.’ Mature seeds were surface-sterilized and cultured on basal Murashige and Skoog (MS) media supplemented with 30–250 μM 2,4-dichlorophenoxyacetic acid (2,4-D) for callus induction. Regeneration media consisted of MS supplemented with 5–10 μM 6-benzyladenine (BA). Among the genera, Poa had the higest callus induction percentage (CIP) regardless of 2,4-D concentration, followed by Cynodon, Lolium, and Festuca, respectively. Cynodon and Lolium had the highest callus regeneration percentage (CRP) and overall regeneration rate (ORR). Festuca had a poor CIP, CRP, and ORR compared to other studied genera. Cynodon produced the highest shoot number per explant. Based on the results of the present study, MS medium supplemented with 60 μM 2,4-D (for callus induction) and 7.5 μM BA (for regeneration) can be used in multi-generic transformation studies with the genera used.  相似文献

The synthesis of phytohemagglutinin (PHA), the major seed lectin ofPhaseolus vulgaris, was investigated inXenopus oocytes injected with RNA isolated from developing bean cotyledons. As is the case for normal PHA, oocyte-synthesized PHA polypeptides were found to contain two asparagine-linked oligosaccharide chains, one of which was of the high-mannose type and the other one of the Golgi-modified type, being largely resistant to endo--N-acetylglucosaminidase H digestion and containing fucose. The modified oligosaccharide chain of oocyte-synthesized PHA appeared to be much larger and more heterogeneous with respect to the modified chain normally present on PHA. When the oocytes were injected with purified mRNA for PHA, isolated by hybrid-selection using a PHA complementary-DNA clone, the results were the same as those obtained by injecting total cotyledonary RNA. On the whole, these results indicate that plant glycoproteins are directed to the Golgi complex even when synthesized in an animal cell, and that correct sorting of the oligosaccharide chains to be processed is independent of the cell-type in which protein synthesis occurs. Summary The central and visceral nervous systems of the cockroach Periplaneta americana were studied by means of the peroxidase-antiperoxidase immunocytochemical method, with the use of antibody to bovine pancreatic polypeptide (PP). PP-like immunoreactive neuron somata are most numerous in the brain; at least 6 pairs of cell groups occur in clearly defined regions. Three pairs of cells each are also present in the suboesophageal ganglion and the thoracic ganglia, one pair of a single cell each in the first abdominal and the frontal ganglia, and 4 to 6 pairs of single cells in the terminal ganglion. No reactive cells were found in the retrocerebral complex and the second to the fifth abdominal ganglia. The axons containing PP-like immunoreactivity issue many branches that are distributed in the entire brain-retrocerebral complex, ventral cord, and visceral nervous system. PP-like immunoreactive material produced in the brain seems to be transported by three routes: protocerebrum to corpora cardiaca (-allata) through the nervi corporis cardiaci, tritocerebrum to visceral nervous system through frontal commissures, and to ventral cord through circumoesophageal connectives.A possible homology between the mammalian brain-GEP (gastro-enteropancreatic) system and the brain-midgut system of this insect is discussed.  相似文献

Lake Huetzalin (Xochimilco) is a shallow waterbody in Mexico City with a dominance of the littoral cladocerans Simocephalus vetulus, Alona rectangula, and Pleuroxus aduncus. Here, we present data on the life table and population dynamics of these three cladoceran species on two types of diets (alga Scenedesmus acutus and detritus). We also analyzed the effect of competition on population growth using Scenedesmus, which proved to be the better diet than detritus. We hypothesize that feces of the larger cladoceran species (S. vetulus) could be a food source for the smaller taxa (A. rectangula and P. aduncus). Experiments were conducted in 100 ml beakers with 50 ml of medium in each at a temperature of 23 ± 1°C. We studied the demography of three test species using two food types with four replicates for each treatment in the life table experiments. Based on the data thus obtained we further studied the competition between the cladocerans on a diet of Scenedesmus. The maximal growth rates obtained for P. aduncus, S. vetulus, and A. rectangula fed Scenedesmus acutus were: 0.1, 0.3, and 0.5 d−1, respectively. Among the test species, S. vetulus had the highest survivorship of 77 days when cultured on detritus and P. aduncus the lowest of 20 days on Scenedesmus. The gross reproductive rate (offspring female−1 lifespan−1) ranged from 6 neonates in Pleuroxus to 160 in Simocephalus per female. From the competition experiments conducted using alga as the diet, we observed that A. rectangula and, especially P. aduncus, had higher growth rates in the presence of S. vetulus than in the controls. This facilitation could be one of the reasons for higher species diversity in the littoral of water bodies.  相似文献

为了揭示鄱阳湖湿地活体植物和自然枯落物在水淹条件下分解过程的差异,以鄱阳湖湿地优势植被物种灰化薹草(Carex cinerascens)和虉草(Phalaris arundinacea)为研究对象,采用分解袋法开展室内模拟实验。植物样品设置新鲜和风干两种处理,分别模拟活体植物和自然枯落物在水下的分解过程。研究结果表明:(1)在180 d的淹水实验过程中,两种处理下的灰化薹草和虉草的分解过程都具有阶段性,前期分解速率较快,后期较慢。(2)两种植物枯落物的分解速率与植物C/N比有显著相关性,而活体植物的分解速率与植物C/N比相关性不强。(3)活体植物和自然枯落物的水下分解速率与物种有关,虉草活体比虉草枯落物分解速率快,而薹草活体比薹草枯落物分解速率慢。(4)模拟实验结束时活体植物仍有34%—43%未分解,推测鄱阳湖丰水期退水后大量洲滩植被消失的原因并非是植被在水淹下完全分解,而是一部分植被被水生动物啃噬或被水流冲走。  相似文献
