A Gram-negative, aerobic, rod-shaped, and red-pigmented bacterial strain, HMC5104T, was isolated from a solar saltern, found in Jeungdo, Republic of Korea (34°59′47″N 126°10′02″E). The major fatty acids were summed feature 4 (comprising iso-C17:1 I and/or anteiso-C17:1 B; 37.2%), iso-C15:0 (20.4%), and iso-C17.0 30H (15.3%). The DNA G+C content was 46.0 mol%. The major isoprenoid quinone was menaquinone-7 (MK-7). A phylogenetic tree based on 16S rRNA gene sequences showed that strain HMC5104T formed a lineage within the genus Pontibacter, and was closely related to Pontibacter korlensis (95.9%), P. roseus (94.9%), and P. actiniarum (94.3%). Similarities to all other Pontibacter species were between 95.9–93.9%. On the basis of the evidence presented in this study, strain HMC5104T represents a novel species of the genus Pontibacter, for which the name Pontibacter salisaro sp. nov. is proposed. A bacterial strain, designated KMM 6244T, was isolated from the sea urchin Strongylocentrotus intermedius and subjected to a polyphasic taxonomic investigation. The bacterium was found to be heterotrophic, aerobic, non-motile and spore-forming. Comparative phylogenetic analysis based on 16S rRNA gene sequencing placed the marine isolate in the genus Bacillus. The nearest neighbor of strain KMM 6244T was Bacillus decolorationis LMG 19507T with a 16S rRNA gene sequence similarity of 98.0%. Sequence similarities with the other recognized Bacillus species were less than 96.0%. The results of the DNA–DNA hybridization experiments revealed a low relatedness (37%) of the novel isolate with the type strain of B. decolorationis LMG 19507T. Strain KMM 6244T grew at 4–45°C and with 0–12% NaCl. It produced catalase and oxidase and hydrolyzed aesculin, casein, gelatin and DNA. The predominant fatty acids were anteiso-C15:0, iso-C15:0, anteiso-C17:0, C15:0, iso-C16:0 and iso-C14:0. The DNA G + C content was 39.4 mol%. A combination of phylogenetic, genotypic and phenotypic data clearly indicated that strain KMM 6244T represents a novel species in the genus Bacillus, for which the name Bacillus berkeleyi sp. nov. is proposed. The taxonomic position of a Gram-positive, endo-spore forming bacterium isolated from a haematite ore sample was analyzed by a polyphasic approach. The strain designated as HIO-4T matched most of the phenotypic and chemical characteristics of the genus Cohnella and represents a novel species. The sequence of the almost complete 16S rRNA (1489 bases) was compared with those of previously studied Cohnella type strains and confirmed that the strain belongs to the genus Cohnella. Strain HIO-4T differs from all other species of Cohnella by at least 3.9% at the 16S rRNA level and the moderately related species are Cohnella phaseoli (96.1%) and Cohnella yongneupensis (96.1%), respectively. Predominant polar lipids are diphosphatidylglycerol (DPG), phosphatidylglycerol (PG), phosphatidylethanolamine (PE); few unknown phospholipids, mannose containing lipid, aminophospholipid and aminophosphoglycolipids. The results of physiological and biochemical tests allowed the genotypic and phenotypic distinctiveness of strain HIO-4T with its phylogenetic relatives and suggest that the strain HIO-4T should be recognized as a novel species, for which the name Cohnella ferri sp. nov. is proposed. A novel actinomycete, designated KLBMP 1221T, was isolated from the surface-sterilized seeds of an oil-seed plant Jatropha curcas L. collected from Sichuan Province, south-west China and was characterized taxonomically by using a polyphasic approach. Phylogenetic analyses based on 16S rRNA gene sequence showed that this strain formed a distinct phyletic line within the radiation of the genus Amycolatopsis. The 16S rRNA gene sequence similarity indicated that strain KLBMP 1221T was most closely related to Amycolatopsis eurytherma NT202T (98.9%), Amycolatopsis tucumanensis ABOT (98.8%), Amycolatopsis thermoflava N1165T (98.6%) and Amycolatopsis methanolica IMSNU 20055T (98.5%). Strain KLBMP 1221T had morphological and chemotaxonomic properties that were consistent with its classification in the genus Amycolatopsis. However, DNA–DNA relatedness data and phenotypic differences clearly distinguished the isolate from its closest relatives. Based on the combined genotypic and phenotypic evidence, it is proposed that strain KLBMP 1221T be classified as representative of a novel species for which the name Amycolatopsis endophytica sp. nov. is proposed. A novel bacterium, strain blls-2T was isolated from Pu’er tea. The isolate was Gram-positive, endospore-forming motile rod that grew at 15∼42°C and pH 6.0∼10.2. The DNA G+C content was 48.3 mol%, the predominant isoprenoid quinone was MK-7, and the predominant cellular fatty acid was anteiso-C15:0 (54.2%) followed by C16:0 (15.5%) and iso-C16:0 (8.2%). The polar lipid pattern of blls-2T was characterized by the presence of diphosphatidylglycerol, phosphatidylethanolamine, and phosphatidylglycerol. Phy-logenetic analysis based on 16S rRNA gene sequence showed that the strain was affiliated within the Paenibacillaceae. The strain was most closely related to Paenibacillus granivorans A30T, with a similarity of 97.1%. Based on the phylogenetic and phenotypic characteristics of strain blls-2T, the isolate is thought to represent a novel taxon in the genus Paenibacillus. The name Paenibacillus camelliae sp. nov. is proposed for the fermented tea isolate; the type strain is blls-2T (= KCTC 13220T= CECT 7361T).  相似文献

Five strains (LN12, LN14T, LN15T, LN16 and LN17T) representing three novel methylotrophic yeast species were isolated from the external surface of plant leaves by three-consecutive enrichments. On the basis of morphological, biochemical, physiological and chemotaxonomic characteristics, the sequence analysis of the D1/D2 domain of the large subunit (LSU) rRNA gene and the phylogenetic analysis, the five strains were assigned to be one novel Ogataea species and two novel Candida species. Three strains (LN12, LN14T and LN16) represent a single novel species of the genus Ogataea, for which the name Ogataea phyllophila sp. nov. is proposed. The type strain is LN14T (= BCC 42666T = NBRC 107780T = CBS 12095T). Strain LN15T was assigned to be Candida chumphonensis sp. nov. (type strain LN15T = BCC 42667T = NBRC 107781T = CBS 12096T). An aerobic, non-motile, catalase-positive, Gram-stain positive actinomycete designated YIM 63233T was isolated from the surface-sterilized leaves of Artemisia annua L. and characterized using a polyphasic taxonomic approach. Optimal growth occurred at 20–28°C, pH 6.0–7.0 and in the presence of 0–3% (w/v) NaCl. 16S rRNA gene sequence-based phylogenetic analysis showed that strain YIM 63233T clustered with species of the genus Pseudonocardia, displaying ≥1.2% sequence divergence with recognized species of this genus (from 98.8 to 94.0%). Relatively low levels of DNA–DNA relatedness were found between strain YIM 63233T and Pseudonocardia petroleophila IMSNU 22072T, which supported the classification of strain YIM 63233T within a novel species of the genus Pseudonocardia. The G + C content of genomic DNA was 72.0 mol%. Strain YIM 63233T possessed chemotaxonomic markers that were consistent with classification in the genus Pseudonocardia, i.e. the predominant fatty acids were iso-C16:0 (32.27%), C16:0 10-methyl (8.73%) and C17:1ω8c (8.30%), whilst the predominant menaquinone was MK-8(H4). The diagnostic diamino acid of the cell-wall peptidoglycan was meso-diaminopimelic acid. The major cell wall sugars were glucose, arabinose, galactose, mannose and rhamnose. The results of physiological and biochemical tests and DNA–DNA hybridization allowed the phenotypic and genotypic differentiation of strain YIM 63233T from its closest phylogenetic neighbours. Therefore, the new isolate YIM 63233T represents a novel species of the genus Pseudonocardia, for which the name Pseudonocardia serianimatus sp. nov. is proposed. A strain named A18 was recovered from a compost of button mushrooms. It was characterized using a polyphasic approach. On the basis of 16S rRNA gene sequence comparison, it belonged to the genus Nocardiopsis and was most closely related to the type strains of Nocardiopsis flavescens (sequence similarity 98.0%), Nocardiopsis prasina (97.5%), Nocardiopsis metallicus (97.4%), Nocardiopsis alba (97.3%). The combination of phylogenetic analysis, DNA–DNA hybridization, phenotypic characteristics and chemotaxonomic data supported the proposal that strain A18 represents a new species of the genus Nocardiopsis, for which the name Nocardiopsis yanglingensis sp. nov. was proposed (type strain A18T = KCTC 19723T = CCTCC 209063T).  相似文献

A novel Gram-negative, catalase- and oxidase-positive, strictly aerobic, non spore-forming, rod-shaped bacterium, designated strain JSM 083058T, was isolated from non-saline forest soil in Hunan Province, China. Growth occurred with 0–8% (w/v) NaCl (optimum, 0.5–3%) at pH 6.0–10.0 (optimum, pH 7.0) and at 5–35°C (optimum, 25–30°C). Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain JSM 083058T fell within the cluster comprising species of the genus Sphingomonas, clustering with Sphingomonas aestuarii K4T, with which it shared highest 16S rRNA gene sequence similarity (99.2%). The chemotaxonomic properties of strain JSM 083058T were consistent with those of the genus Sphingomonas. The predominant respiratory quinone was ubiquinone Q-10, and the major cellular fatty acids were summed feature 8 (C18:1ω7c/C18:1ω6c), C16:0, summed feature 3 (C16:1ω7c/C16:1ω6c) and C17:1ω6c. The polar lipids consisted of diphosphatidylglycerol, phosphatidylcholine, phosphatidylethanolamine, phosphatidylglycerol and sphingoglycolipid. The genomic DNA G+C content of strain JSM 083058T was 65.5 mol%. The combination of phylogenetic analysis, DNA–DNA relatedness, phenotypic characteristics and chemotaxonomic data supported the view that strain JSM 083058T represents a novel species of the genus Sphingomonas, for which the name Sphingomonas hunanensis sp. nov. is proposed. A novel actinomycete strain, GW25-5T, was isolated from a soil sample collected from the Fildes Peninsula, King George Island, West Antarctica. The strain was characterized by white to grey aerial mycelia, which were differentiated to straight to flexuous spore chains, with rod-shaped smooth spores. The cell wall of strain GW25-5T contained LL-diaminopimelic acid (A2pm) and traces of meso-A2pm. Whole-cell sugars were galactose and minor amounts of mannose and glucose. The predominant menaquinones were MK-9(H6) (49%), MK-9(H8) (24%) and MK-9(H4) (12%). The phospholipids contained DPG, PE, PI, PIM and PL(s). The major cellular fatty acids were iso-C16:0 and anteiso-C15:0. Genomic DNA G+C content of strain GW25-5T was 70.0 mol%. BLAST result showed that strain GW25-5 has the 16S rRNA gene sequence highest similarity of 97.5% with members of genus Streptomyces and phylogenetic analysis indicated that this strain belongs to the genus Streptomyces. DNA–DNA relatedness values of strain GW25-5T with the closest species of Streptomyces purpureus LMG 19368T and Streptomyces beijiangensis YIM 6T were significantly lower than 70% of the threshold value for the delineation of genomic species. A polyphasic taxonomic investigation based on a judicious combination of genotypic and phenotypic characteristics revealed that the organism represents a novel species of the genus Streptomyces. Thus, we propose strain GW25-5T as the type strain of this novel species, Streptomyces fildesensis (=CGMCC 4.5735T = YIM 93602T = DSM 41987T = NRRL B 24828T).  相似文献

A marine bacterium designated strain IMCC4074T was isolated from surface seawater collected off Incheon Port, the Yellow Sea, and subjected to a polyphasic taxonomy. The strain was Gram-negative, chemoheterotrophic, slightly halophilic, strictly aerobic, and motile rods. Based on 16S rRNA gene sequence comparisons, the strain was most closely related to Marinobacterium litorale KCTC 12756T (93.9%) and shared low 16S rRNA gene sequence similarities with members of the genus Marinobacterium (91.8–93.9%) and the genus Neptunomonas (93.4%) in the order Oceanospirillales. Phylogenetic analyses showed that this marine isolate formed an independent phyletic line within the genus Marinobacterium clade. The DNA G+C composition of the strain was 56.0 mol% and the predominant constituents of the cellular fatty acids were C16:0 (28.0%), C16:1 ω7c and/or iso-C15:0 2-OH (19.3%), C18:1 ω7c (17.8%), and C17:1 cyclo (12.5%), which differentiated the strain from other Marinobacterium species. Based on the taxonomic data collected in this study, only a distant relationship could be found between strain IMCC4074T and other members of the genus Marinobacterium, thus the strain represents a novel species of the genus Marinobacterium, for which the name Marinobacterium marisflavi sp. nov. is proposed. A Gram-stain negative, aerobic, oxidase and catalase positive, non-flagellated, pink coloured bacterium with gliding motility, designated as strain UDD1T was isolated from soil. The bacterium lacked flexirubin-type pigments. Phylogenetic analysis based on its 16S rRNA gene sequence revealed that strain UDD1T formed a lineage within the family Cytophagaceae of the phylum Bacteroidetes, and forms a distinct clade with type strains of the closely related genus Pontibacter with similarities of 91.36–93.62%. Strain UDD1T contained MK-7 as the predominant menaquinone and summed feature 4 (iso-C17:1 I and/or anteiso-C17:1 B) and iso-C15:0 as the major fatty acids. The major polar lipids were phosphatidylethanolamine and an unidentified glycolipid. The DNA G+C content of strain UDD1T was 49 mol%. On the basis of phenotypic, genotypic and phylogenetic analyses, the strain UDD1T represents a novel species of a new genus in the family Cytophagaceae, for which the name Edaphorhabdus rosea gen. nov., sp. nov., is proposed. During the course of our research on new actinobacterial sources, a novel actinomycete strain YIM 63101T was isolated from the surface-sterilized roots of Artemisia annua L. collected from Xishuangbanna, Yunnan province, south-west China and characterized by using a polyphasic approach. The strain formed well-differentiated aerial and substrate mycelia. Phylogenetic analysis based on 16S rRNA gene sequences showed that strain YIM 63101T belongs to the genus Pseudonocardia, with highest similarity to “Pseudonocardia artemisiae YIM 63587T” (99.4%). Sequence similarities between strain YIM 63101T and the other Pseudonocardia species ranged from 97.0 (Pseudonocardia saturnea IMSNU 20052T) to 94.0% (Pseudonocardia compacta IMSNU 20111T). The chemotaxonomic characteristics, such as cell wall diaminopimelic acid, whole-cell sugars, fatty acid components and the major menaquinones suggested that the organism belonged to the genus Pseudonocardia. The G + C content of the genomic DNA was 69.4 mol%. Based on comparative analysis of physiological, biochemical and chemotaxonomic data, including low DNA–DNA hybridization results, it is proposed that strain YIM 63101T represents a novel species of the genus Pseudonocardia, named Pseudonocardia bannaensis sp. nov. A Gram-staining-negative, motile, non-spore-forming and rod-shaped bacterial strain, 20-23RT, was isolated from intestine of bensasi goatfish, Upeneus bensasi, and its taxonomic position was investigated by using a polyphasic study. Phylogenetic analyses based on 16S rRNA gene sequences revealed that strain 20-23RT belonged to the genus Shewanella. Strain 20-23RT exhibited 16S rRNA gene sequence similarity values of 99.5, 99.2, and 97.5% to Shewanella algae ATCC 51192T, Shewanella haliotis DW01T, and Shewanella chilikensis JC5T, respectively. Strain 20-23RT exhibited 93.1–96.0% 16S rRNA gene sequence similarity to the other Shewanella species. It also exhibited 98.3–98.4% gyrB sequence similarity to the type strains of S. algae and S. haliotis. Strain 20-23RT contained simultaneously both menaquinones and ubiquinones; the predominant menaquinone was MK-7 and the predominant ubiquinones were Q-8 and Q-7. The fatty acid profiles of strain 20–23RT, S. algae KCTC 22552T and S. haliotis KCTC 12896T were similar; major components were iso-C15:0, C16:0, C16:1 ω7c and/or iso-C15:0 2-OH and C17:1 ω8c. The DNA G+C content of strain 20-23RT was 53.9 mol%. Differential phenotypic properties and genetic distinctiveness of strain 20–23RT, together with the phylogenetic distinctiveness, revealed that this strain is distinguishable from recognized Shewanella species. On the basis of the data presented, strain 20-23RT represents a novel species of the genus Shewanella, for which the name Shewanella upenei sp. nov. is proposed. An actinomycete strain, designated YIM 60475T, was isolated from the roots of Maytenus austroyunnanensis and was characterized by using a polyphasic approach. The strain was determined to belong to the genus Streptomyces, based on its phenotypic and phylogenetic characteristics. The strain produced spiral spore chains on aerial mycelium. The cell wall contained ll-diaminopimelic acid. Whole-cell hydrolysates contained galactose, glucose, and xylose. The phospholipid was type II. The DNA G+C content of the type strain was 73.3 mol%. DNA–DNA hybridization and comparison of physiological and chemical characteristics suggested that strain YIM 60475T is a new Streptomyces species, for which the name Streptomyces mayteni sp. nov. is proposed. Strain M1-2T was isolated from the black sand from the seashore of Jeju Island, Republic of Korea and was classified using a polyphasic taxonomic approach. Strain M1-2T appeared as Gram-negative, motile rods that could grow in the presence of 1–10% (w/v) NaCl and at temperatures ranging from 4 to 37°C. This isolate has catalase and oxidase activity and hydrolyses aesculin, DNA and l-tyrosine. Based on phylogenetic analysis using 16S rRNA gene sequences, strain M1-2T belongs to the genus Joostella and is clearly distinct from the other described species of this genus, Joostella marina (type strain En5T). The 16S rRNA gene sequence similarity level between M1-2T and J. marina En5T is 97.2%, and the DNA–DNA relatedness value between the two strains is 23.9%. Strain M1-2T contains MK-6 as the major menaquinone and iso-C15:0, summed feature 3 (C16:1 ω7c and/or iso-C15:0 2OH) and iso-C17:0 3OH as major cellular fatty acids. The DNA G + C content is 32.3 mol%. These data suggest that strain M1-2T should be classified as a novel species, for which the name Joostella atrarenae sp. nov. is proposed. The alphaproteobacterial strain JLT2003T was isolated from surface seawater off the coast of Guishan island, Taiwan. The strain was Gram negative, ovoid or coccoid, non-motile and formed pink colonies on marine agar 2216 (MA; DIFCO) medium. The dominant fatty acids were C18:1ω7c, cyclo C19:0ω8c, and C16:0. The polar lipid profile consisted of diphosphatidylglycerol and phosphatidylglycerol. The major respiratory ubiquinone was Q-10. The DNA G+C content was 62.3 mol%. Phylogenetic analysis based on 16S rRNA gene sequences showed that the strain was most closely related to Pontibaca methylaminivorans GRP21T with 94.8% similarity. The isolate was distinguishable from members of the family Rhodobacteraceae based on phenotypic and biochemical characteristics. On the basis of the taxonomic data presented, strain JLT2003T is considered to represent a novel species of a new genus, for which the name Oceaniovalibus guishaninsula gen. nov., sp. nov. is proposed. A short coccoid-rod-shaped, nonmotile actinobacteria strain MSL-11T was isolated from soil in Bigeum Island, Korea. A polyphasic study was undertaken to establish the taxonomic position of this strain. Phylogenetic analysis based on the 16S rRNA gene sequence revealed that strain MSL-11T forms an evolutionary lineage within the radiation of the genus Nocardioides. The cell wall peptidoglycan of strain MSL-11T contained ll-diaminopimelic acid, indicating wall chemotype I. The predominant menaquinone was MK-8(H4). Strain MSL-11T had a cellular fatty acid profile containing straight-chain, branched, unsaturated, and 10-methyl fatty acids, with iso-C16:0 as a major fatty acid component detected. The DNA G + C content of the strain was 71.8 mol%. Comparative 16S rRNA gene sequencing revealed that the strains constituted a distinct subclade within the genus Nocardioides, displaying a 16S rRNA gene sequence similarity of about 95.68% with Nocardioides jensenii DSM 20641T. On the basis of both phenotypic and phylogenetic evidence, the strain is separated from previously described Nocardioides species and should be assigned to represent a novel species of the genus Nocardioides, for which the name Nocardioides dilutes sp. nov. is proposed. Four gram-negative, aerobic, motile, non-spore, forming rods with a wide pH and temperature range for growth (pH 7.0–11.0, optimum pH 8.0; 20–45°C, optimum 28°C) strains were isolated from root nodules of Sphaerophysa salsula and characterized by means of a polyphasic approach. Phylogenetic analysis based on 16S rRNA gene sequences revealed that the four strains formed a new lineage related to the genus Rhizobium and the sequence similarities between the isolate and the most related type strain Rhizobium giardinii was 96.5%. These strains also formed a distinctive group from the reference strains for defined Rhizobium species based on housekeeping gene sequences (atpD and recA), BOX-PCR fingerprinting, phenotypic features and symbiotic properties. The representative strain CCNWGS0238T has DNA-DNA relatedness of less than 33.4% with the most closely related species R. giardinii. It is therefore proposed as a new species, Rhizobium sphaerophysae sp. nov., with isolate CCNWGS0238T (=ACCC17498T = HAMBI3074T) as the type strain.  相似文献

A novel marine bacterium, designated strain CNURIC014T was isolated from coastal seawater of Jeju Island in Korea. Strain CNURIC014T formed yellow colonies on marine agar 2216 and the cells were Gram-negative, non-motile, strictly aerobic, rod-shaped. The temperature, pH and NaCl ranges for growth were 15–37°C, pH 6.0–9.0 and 1.0–7.0% NaCl. Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain CNURIC014T was most closely related to Gaetbulibacter marinus and Gaetbulibacter saemankumensis, with a sequence similarity of 95.1% and 94.6%, respectively. The DNA G+C content of the strain was 33.1 mol% and the major respiratory quinone was menaquinone-6. The major cellular fatty acids were iso-C15:1 (22.8%), iso-C15:0 (18.8%), summed feature 3 (iso-C15:0 2-OH/C16:1 ω7c, 12.9%) and iso-C17:0 3-OH (11.5%). On the basis of phenotypic, phylogenetic, and genotypic data, strain CNURIC014T represents a novel species within the genus Geatbulibacter, for which the name Gaetbulibacter jejuensis sp. nov. is proposed. The type strain is CNURIC014T(=KCTC 22615T =JCM 15976T).  相似文献
