Lactic acid production was investigated for batch and repeated batch cultures of Enterococcus faecalis RKY1, using wood hydrolyzate and corn steep liquor. When wood hydrolyzate (equivalent to 50 g l−1 glucose) supplemented with 15–60 g l−1 corn steep liquor was used as a raw material for fermentation, up to 48.6 g l−1 of lactic acid was produced with, volumetric productivities ranging between 0.8 and 1.4 g l−1 h−1. When a medium containing wood hydrolyzate and 15 g l−1 corn steep liquor was supplemented with 1.5 g l−1 yeast extract, we observed 1.9-fold and 1.6-fold increases in lactic acid productivity and cell growth, respectively. In this case, the nitrogen source cost for producing 1 kg lactic acid can be reduced to 23% of that for fermentation from wood hydrolyzate using 15 g l−1 yeast extract as a single nitrogen source. In addition, lactic acid productivity could be maximized by conducting a cell-recycle repeated batch culture of E. faecalis RKY1. The maximum productivity for this process was determined to be 4.0 g l−1 h−1.  相似文献

Sweet sorghum juice supplemented with 0.5% ammonium sulphate was used as a substrate for ethanol production by Saccharomyces cerevisiae TISTR 5048. In batch fermentation, kinetic parameters for ethanol production depended on initial cell and sugar concentrations. The optimum initial cell and sugar concentrations in the batch fermentation were 1 × 108 cells ml−1 and 24 °Bx respectively. At these conditions, ethanol concentration produced (P), yield (Y ps) and productivity (Q p ) were 100 g l−1, 0.42 g g−1 and 1.67 g l−1 h−1 respectively. In fed-batch fermentation, the optimum substrate feeding strategy for ethanol production at the initial sugar concentration of 24 °Bx was one-time substrate feeding, where P, Y ps and Q p were 120 g l−1, 0.48 g g−1 and 1.11 g l−1 h−1 respectively. These findings suggest that fed-batch fermentation improves the efficiency of ethanol production in terms of ethanol concentration and product yield.  相似文献

The development of microorganims that efficiently ferment lactose has a high biotechnological interest, particularly for cheese whey bioremediation processes with simultaneous bio-ethanol production. The lactose fermentation performance of a recombinant Saccharomyces cerevisiae flocculent strain was evaluated. The yeast consumed rapidly and completely lactose concentrations up to 150 g l−1 in either well- or micro-aerated batch fermentations. The maximum ethanol titre was 8% (v/v) and the highest ethanol productivity was 1.5–2 g l−1 h−1, in micro-aerated fermentations. The results presented here emphasise that this strain is an interesting alternative for the production of ethanol from lactose-based feedstocks.  相似文献

An innovative consecutive batch fermentation process was developed for very high gravity (VHG) ethanol fermentation with the self-flocculating yeast under high biomass concentration conditions. On the one hand, the high biomass concentration significantly shortened the time required to complete the VHG fermentation and the duration of yeast cells suffering from strong ethanol inhibition, preventing them from losing viability and making them suitable for being repeatedly used in the process. On the other hand, the separation of yeast cells from the fermentation broth by sedimentation instead of centrifugation, making the process economically more competitive. The VHG medium composed of 255 g L−1 glucose and 6.75 g L−1 each of yeast extract and peptone was fed into the fermentation system for nine consecutive batch fermentations, which were completed within 8–14 h with an average ethanol concentration of 15% (v/v) and ethanol yield of 0.464, 90.8% of its theoretical value of 0.511. The average ethanol productivity that was calculated with the inclusion of the downstream time for the yeast flocs to settle from the fermentation broth and the supernatant to be removed from the fermentation system was 8.2 g L−1 h−1, much higher than those previously reported for VHG ethanol fermentation and regular ethanol fermentation with ethanol concentration around 12% (v/v) as well.  相似文献

Production of lactic acid from glucose by immobilized cells of Lactococcus lactis IO-1 was investigated using cells that had been immobilized by either entrapment in beads of alginate or encapsulation in microcapsules of alginate membrane. The fermentation process was optimized in shake flasks using the Taguchi method and then further assessed in a production bioreactor. The bioreactor consisted of a packed bed of immobilized cells and its operation involved recycling of the broth through the bed. Both batch and continuous modes of operation of the reactor were investigated. Microencapsulation proved to be the better method of immobilization. For microencapsulated cells at immobilized cell concentration of 5.3 g l−1, the optimal production medium had the following initial concentrations of nutrients (g l−1): glucose 45, yeast extract 10, beef extract 10, peptone 7.5 and calcium chloride 10 at an initial pH of 6.85. Under these conditions, at 37 °C, the volumetric productivity of lactic acid in shake flasks was 1.8 g l−1 h−1. Use of a packed bed of encapsulated cells with recycle of the broth through the bed, increased the volumetric productivity to 4.5 g l−1 h−1. The packed bed could be used in repeated batch runs to produce lactic acid.  相似文献

A yeast strain Kluyveromyces sp. IIPE453 (MTCC 5314), isolated from soil samples collected from dumping sites of crushed sugarcane bagasse in Sugar Mill, showed growth and fermentation efficiency at high temperatures ranging from 45°C to 50°C. The yeast strain was able to use a wide range of substrates, such as glucose, xylose, mannose, galactose, arabinose, sucrose, and cellobiose, either for growth or fermentation to ethanol. The strain also showed xylitol production from xylose. In batch fermentation, the strain showed maximum ethanol concentration of 82 ± 0.5 g l−1 (10.4% v/v) on initial glucose concentration of 200 g l−1, and ethanol concentration of 1.75 ± 0.05 g l−1 as well as xylitol concentration of 11.5 ± 0.4 g l−1 on initial xylose concentration of 20 g l−1 at 50°C. The strain was capable of simultaneously using glucose and xylose in a mixture of glucose concentration of 75 g l−1 and xylose concentration of 25 g l−1, achieving maximum ethanol concentration of 38 ± 0.5 g l−1 and xylitol concentration of 14.5 ± 0.2 g l−1 in batch fermentation. High stability of the strain was observed in a continuous fermentation by feeding the mixture of glucose concentration of 75 g l−1 and xylose concentration of 25 g l−1 by recycling the cells, achieving maximum ethanol concentration of 30.8 ± 6.2 g l−1 and xylitol concentration of 7.35 ± 3.3 g l−1 with ethanol productivity of 3.1 ± 0.6 g l−1 h−1 and xylitol productivity of 0.75 ± 0.35 g l−1 h−1, respectively.  相似文献

Previously, a Saccharomyces cerevisiae strain was engineered for xylose assimilation by the constitutive overexpression of the Orpinomyces xylose isomerase, the S. cerevisiae xylulokinase, and the Pichia stipitis SUT1 sugar transporter genes. The recombinant strain exhibited growth on xylose, under aerobic conditions, with a specific growth rate of 0.025 h−1, while ethanol production from xylose was achieved anaerobically. In the present study, the developed recombinant yeast was adapted for enhanced growth on xylose by serial transfer in xylose-containing minimal medium under aerobic conditions. After repeated batch cultivations, a strain was isolated which grew with a specific growth rate of 0.133 h−1. The adapted strain could ferment 20 g l−1 of xylose to ethanol with a yield of 0.37 g g−1 and production rate of 0.026 g l−1 h−1. Raising the fermentation temperature from 30°C to 35°C resulted in a substantial increase in the ethanol yield (0.43 g g−1) and production rate (0.07 g l−1 h−1) as well as a significant reduction in the xylitol yield. By the addition of a sugar complexing agent, such as sodium tetraborate, significant improvement in ethanol production and reduction in xylitol accumulation was achieved. Furthermore, ethanol production from xylose and a mixture of glucose and xylose was also demonstrated in complex medium containing yeast extract, peptone, and borate with a considerably high yield of 0.48 g g−1.  相似文献

Compared with steady state, oscillation in continuous very-high-gravity ethanol fermentation with Saccharomyces cerevisiae improved process productivity, which was thus introduced for the fermentation system composed of a tank fermentor followed by four-stage packed tubular bioreactors. When the very-high-gravity medium containing 280 g l−1 glucose was fed at the dilution rate of 0.04 h−1, the average ethanol of 15.8% (v/v) and residual glucose of 1.5 g l−1 were achieved under the oscillatory state, with an average ethanol productivity of 2.14 g h−1 l−1. By contrast, only 14.8% (v/v) ethanol was achieved under the steady state at the same dilution rate, and the residual glucose was as high as 17.1 g l−1, with an ethanol productivity of 2.00 g h−1 l−1, indicating a 7% improvement under the oscillatory state. When the fermentation system was operated under the steady state at the dilution rate of 0.027 h−1 to extend the average fermentation time to 88 h from 59 h, the ethanol concentration increased slightly to 15.4% (v/v) and residual glucose decreased to 7.3 g l−1, correspondingly, but the ethanol productivity was decreased drastically to 1.43 g h−1 l−1, indicating a 48% improvement under the oscillatory state at the dilution rate of 0.04 h−1.  相似文献

The constant-rate fed-batch production of the polygalacturonic acid bioflocculant REA-11 was studied. A controlled sucrose-feeding strategy resulted in a slight improvement in biomass and a 7% reduction in flocculating activity compared with the batch process. When fed with a 3 g l−1 urea solution, the flocculating activity was enhanced to 720 U ml−1 in 36 h. High cell density (2.12 g l−1) and flocculating activity (820 U ml−1) were obtained in a 10-l fermentor by feeding with a sucrose-urea solution, with values of nearly two times and 50% higher than those of the batch process, respectively. Moreover, the residual sucrose declined to 2.4 g l−1, and residual urea decreased to 0.03 g l−1. Even higher flocculating activity of 920 U ml−1 and biomass of 3.26 g l−1 were obtained by feeding with a sucrose-urea solution in a pilot scale fermentation process, indicating the potential industrial utility of this constant-rate feeding strategy in bioflocculant production by Corynebacterium glutamicum.  相似文献

Industrial waste corn cob residue (from xylose manufacturing) without pretreatment was hydrolyzed by cellulase and cellobiase. The cellulosic hydrolysate contained 52.4 g l−1 of glucose and was used as carbon source for lactic acid fermentation by cells of Lactobacillus delbrueckii ZU-S2 immobilized in calcium alginate gel beads. The final concentration of lactic acid and the yield of lactic acid from glucose were 48.7 g l−1 and 95.2%, respectively, which were comparative to the results of pure glucose fermentation. The immobilized cells were quite stable and reusable, and the average yield of lactic acid from glucose in the hydrolysate was 95.0% in 12 repeated batches of fermentation. The suitable dilution rate of continuous fermentation process was 0.13 h−1, and the yield of lactic acid from glucose and the productivity were 92.4% and 5.746 g l−1 h−1, respectively. The production of lactic acid by simultaneous saccharification and fermentation (SSF) process was carried out in a coupling bioreactor, the final concentration of lactic acid was 55.6 g l−1, the conversion efficiency of lactic acid from cellulose was 91.3% and the productivity was 0.927 g l−1 h−1. By using fed-batch technique in the SSF process, the final concentration of lactic acid and the productivity increased to 107.6 g l−1 and 1.345 g l−1 h−1, respectively, while the dosage of cellulase per gram substrate decreased greatly. This research work should advance the bioconversion of renewable cellulosic resources and reduce environmental pollution.  相似文献

Batch cultivation of Ralstonia eutropha NRRL B14690 attained 21 g biomass l−1 and 9.4 g poly(β-hydroxybutyrate) l−1 (0.45 g PHB g−1 dry wt−1) in 60 h. Repeated batch operation (empty-and-fill protocol) to remove 20% (v/v) of the culture broth and to supplement an equal volume of fresh media resulted in 49 g biomass l−1 and 25 g PHB l−1 (0.51 g PHB g−1 dry wt−1) with an overall productivity of 0.42 g PHB l−1 h−1 in 67 h. In the two cycles of repeated batch fermentation there was a 3-fold increase in productivity as compared to batch.  相似文献

A potential industrial substrate (liquefied corn starch; LCS) has been employed for successful acetone butanol ethanol (ABE) production. Fermentation of LCS (60 g l−1) in a batch process resulted in the production of 18.4 g l−1 ABE, comparable to glucose: yeast extract based medium (control experiment, 18.6 g l−1 ABE). A batch fermentation of LCS integrated with product recovery resulted in 92% utilization of sugars present in the feed. When ABE was recovered by gas stripping (to relieve inhibition) from the fed-batch reactor fed with saccharified liquefied cornstarch (SLCS), 81.3 g l−1 ABE was produced compared to 18.6 g l−1 (control). In this integrated system, 225.8 g l−1 SLCS sugar (487 % of control) was consumed. In the absence of product removal, it is not possible for C. beijerinckii BA101 to utilize more than 46 g l−1 glucose. A combination of fermentation of this novel substrate (LCS) to butanol together with product recovery by gas stripping may economically benefit this fermentation. An optimized very high gravity (VHG) glucose medium supplemented with low cost nutrient sources was used to evaluate bio-ethanol production by 11 Saccharomyces cerevisiae strains. The industrial strains PE-2 and CA1185 exhibited the best overall fermentation performance, producing an ethanol titre of 19.2% (v/v) corresponding to a batch productivity of 2.5 g l−1 h−1, while the best laboratory strain (CEN.PK 113-7D) produced 17.5% (v/v) ethanol with a productivity of 1.7 g l−1 h−1. The results presented here emphasize the biodiversity found within S. cerevisiae species and that naturally adapted strains, such as PE-2 and CA1185, are likely to play a key role in facilitating the transition from laboratory technological breakthroughs to industrial-scale bio-ethanol fermentations.  相似文献

Optimum growth conditions for the fermentation of non-concentrated whey permeate by Kluyveromyces fragilis NRRL 665 have been defined. Use of 3.75 g yeast extract l?1, a growth temperature of 38°C and a pH of 4.0 allowed a maximum productivity of 5.23 g ethanol l?1 h?1 in continuous culture with a yield 91% of theoretical. Complete batch fermentation of permeate with 100 g lactose l?1 was possible with a maximum specific growth rate of 0.276 h?1 without any change in ethanol yield. Fermentation of concentrated permeate resulted, however, in a general decrease of specific substrate consumption rate, demonstrated by the inability to completely convert an initial 90 or 150 g lactose l?1 in continuous culture, even at dilution rates as low as 0.05 and 0.08 h?1, respectively. The decrease could be related to substrate inhibition, to an increase in osmotic pressure caused by lactose and salts, and to ethanol inhibition of both alcohol and biomass yield. The decrease in specific productivity could be counterbalanced by use of high cell density cultures, obtained by cell recycle of K. fragilis. Fermentation of a non-concentrated permeáte at a dilution rate of 1 h?1 resulted in a productivity of 22 g l?1 h?1 at 22 g ethanol l?1. Cell recycle using flocculating Kluyveromyces lactis NCYC 571 was also tested. With this strain a productivity of 9.3 g l?1 h?1 at 45 g product l?1 was attained at a dilution rate of 0.2 h?1, with an initial lactose concentration of 95 g l?1.  相似文献

The filtration in 1,3-propanediol (1,3-PD) downstream process is influenced by the large amounts of capsular polysaccharides (CPS) produced by Klebsiella pneumoniae CGMCC 1.6366. The morphological and fermentation properties were investigated with the CPS-deficient mutant K. pneumoniae CGMCC 1.6366 CPS. Similar biomass was obtained with CGMCC 1.6366, and the mutant strain in batch cultures indicating the cell growth was slightly inhibited by CPS defection. The viscosity of fermentation broth by mutant strain decreased by 27.45%. The flux with ceramic membrane filter was enhanced from 168.12 to 303.6 l h−1 m−2, exhibiting the great importance for downstream processing of 1,3-PD fermentation. The products spectrum of mutant isolate changed remarkably regarding to the concentration of fermentation products. The synthesis of important 1,3-PD and 2,3-butanediol was enhanced from 9.73 and 4.06 g l−1 to 10.37 and 4.77 g l−1 in batch cultures. The noncapsuled K. pneumoniae provided higher 1,3-PD yield of 0.54 mol mol−1 than that of encapsuled wild parent in batch cultures. The fed-batch fermentation of mutant strain resulted in 1,3-PD concentration, yield, and productivity of 78.13 g l−1, 0.53 mol mol−1, and 1.95 g l−1 h−1, respectively.  相似文献

Instead of the conventional carbon sources used for propionic acid biosynthesis, the utilization of glycerol is considered here, since the metabolic pathway involved in the conversion of glycerol to propionic acid is redox-neutral and energetic. Three strains, Propionibacterium acidipropionici, Propionibacterium acnes and Clostridium propionicum were tested for their ability to convert glycerol to propionic acid during batch fermentation with initially 20 g/l glycerol. P. acidipropionici showed higher efficiency in terms of fermentation time and conversion yield than did the other strains. The fermentation profile of this bacterium consisted in propionic acid as the major product (0.844 mol/mol), and in minimal by-products: succinic (0.055 mol/mol), acetic (0.023 mol/mol) and formic (0.020 mol/mol) acids and n-propanol (0.036 mol/mol). The overall propionic acid productivity was 0.18 g l−1h−1. A comparative study with glucose and lactic acid as carbon sources showed both less diversity in end-product composition and a 17% and 13% lower propionic acid conversion yield respectively than with glycerol. Increasing the initial glycerol concentration resulted in an enhanced productivity up to 0.36 g l−1h−1 and in a maximal propionic acid concentration of 42 g/l, while a slight decrease of the conversion yield was noticed. Such a propionic acid production rate was similar or higher than the values obtained with lactic acid (0.35 g l−1h−1) or glucose (0.28 g l−1h−1). These results demonstrated that glycerol is a carbon source of interest for propionic acid production. Mucor indicus can be used to produce ethanol from a variety of sugars, including pentose’s. An extract of it, produced by autolysis, could replace yeast extract in culture medium with improved production of ethanol. At 10 g l−1, the extract gave a higher ethanol yield (0.47 g g−1) and productivity (0.71 g l−1 h−1) compared to medium containing yeast extract (yield 0.45 g g−1; productivity 0.67 g l−1 h−1).  相似文献

The effect of glucose concentration on erythritol production by Torula sp. was investigated. The maximum volumetric productivity of erythritol was obtained at an initial glucose concentration of 300 g l−1 in batch culture. The volumetric productivity was maximal at a controlled glucose concentration of 225 g l−1, reducing the lag time of the erythritol production. A fed-batch culture was established with an initial glucose concentration of 300 g l−1 and with a controlled glucose concentration of 225 g l−1 in medium containing phytic acid as a phosphate source. In this fed-batch culture, a final erythritol production of 192 g l−1 was obtained from 400 g l−1 glucose in 88 h. This corresponded to a volumetric productivity of 2.26 g l−1 h−1 and a 48% yield. The potential use of spin filter device to retainPropionibacterium acidipropionici in the bioreactor under continuous mode of fermentation and improve propionic acid productivity, was examined. The yield of propionic acid based on lactose concentration was 51% in batch and 54% in continuous (dilution rate=0.05 h−1) operation. The yield in continuous fermentation with cell retention using spin filter of 10 micron size (dilution rate=0.05 h−1) was even higher at 70% (w/w). The volumetric productivity under batch and continuous mode of operation were 0.312 g L−1 h−1 and 0.718 g L−1 h−1 respectively. Continuous fermentation with cell retention demonstrated even higher volumetric productivities at 0.98 g L−1 h−1 with out clogging problems It could be used for utilization of cheese whey to produce propionic acid at higher yield and productivities.  相似文献

A two-stage fed-batch process was designed to enhance erythritol productivity by the mutant strain of Candida magnoliae. The first stage (or growth stage) was performed in the fed-batch mode where the growth medium was fed when the pH of the culture broth dropped below 4.5. The second stage (or production stage) was started with addition of glucose powder into the culture broth when the cell mass reached about 75 g dry cell weight l−1. When the initial glucose concentration was adjusted to 400 g l−1 in the production stage, 2.8 g l−1 h−1 of overall erythritol productivity and 41% of erythritol conversion yield were achieved, which represented a fivefold increase in erythritol productivity compared with the simple batch fermentation process. A high glucose concentration in the production phase resulted in formation of organic acids including citrate and butyrate.
