It is known that vegetation plays an important role in the retention of heavy metals in salt marshes by taking up and accumulating the metals. In this study, we investigated whether arbuscular mycorrhizal fungi (AMF) increase Cd and Cu uptake and accumulation in the root system of the salt marsh species Aster tripolium L., and whether indigenous AMF isolated from polluted salt marshes have higher capacity to resist and alleviate metal stress in A. tripolium than isolates of the same species originated from non-polluted sites. Plants inoculated with Glomus geosporum, either isolated from a polluted salt marsh site (PL isolate) or from a non-polluted site (NP isolate), and non-mycorrhizal (NM) plants were compared in a pot experiment at four different Cd and Cu concentrations. Cd had no effect in root colonization, whereas high concentrations of Cu decreased colonization level in plants inoculated with the NP isolate. AM colonization did not increase plant dry weight or P concentration but influenced root Cd and Cu concentrations. Inoculation with PL and NP isolates enhanced root Cd and Cu concentrations, especially at highest metal addition levels, as compared to NM plants, without increasing shoot Cd and Cu concentrations. There was no evidence of intraspecific variation in the effects between AMF isolated from polluted and non-polluted sites, since there were no differences between plants inoculated with PL or NP isolate in any of the tested plant variables. The results of this study showed that AMF enhance metal accumulation in the root system of A. tripolium, suggesting a contribution of AMF to the sink of metals within vegetation in the salt marshes.

Growth of mycorrhizal tomato and mineral acquisition under salt stress
High salt levels in soil and water can limit agricultural production and land development in arid and semiarid regions. Arbuscular mycorrhizal fungi (AMF) have been shown to decrease plant yield losses in saline soils. The objective of this study was to examine the growth and mineral acquisition responses of greenhouse-grown tomato to colonization by the AMF Glomus mosseae [(Nicol. And Gerd.) Gerd. and Trappe] under varied levels of salt. NaCl was added to soil in the irrigation water to give an ECe of 1.4 (control), 4.7 (medium) and 7.4 dS m–1 (high salt stress). Plants were grown in a sterilized, low P (silty clay) soil-sand mix. Mycorrhizal colonization was higher in the control than in saline soil conditions. Shoot and root dry matter yields and leaf area were higher in mycorrhizal than in nonmycorrhizal plants. Total accumulation of P, Zn, Cu, and Fe was higher in mycorrhizal than in nonmycorrhizal plants under both control and medium salt stress conditions. Shoot Na concentrations were lower in mycorrhizal than in nonmycorrhizal plants grown under saline soil conditions. The improved growth and nutrient acquisition in tomato demonstrate the potential of AMF colonization for protecting plants against salt stress in arid and semiarid areas.  Halophytes from both coastal and inland Central European salt marshes were examined for colonization by arbuscular mycorrhizal (AM) fungi. Plants from different families were strongly colonized but the degree of colonization varied with the individual plant and apparently during the vegetation period, too. Members of the typical non-mycorrhizal families like Armeria maritima of the Plumbaginaceae and Salicornia europaea of the Chenopodiaceae were found to be colonized, particularly in the drier salt marshes. High numbers of Glomus spores were found in the saline soils, especially those of the inland locations examined. Approximately 80% of these spores were from Glomus geosporum as shown by a typical restriction fragment length polymorphism (RFLP) pattern of the amplified internal transcribed spacer regions. The present study demonstrates that RFLP analysis is useful when screening habitats for the occurrence of mycorrhizal fungi which can be identified only with difficulty by morphological criteria. Responses of Puccinellia distans, a halophytic grass to low (50 mM) and high (200 mM) NaCl salinity, were studied in a sand culture experiment without or with inoculation by arbuscular mycorrhizal fungus (AMF), Claroideoglomus etunicatum isolated from its saline habitat. Plant biomass was not influenced by salinity levels, while a tendency to a higher biomass was observed in AMF plants under both control and saline conditions. Leaf photosynthesis increased by both salinity and AMF inoculation. Despite higher transpiration rate, AMF plants had higher water-use efficiency under sever saline conditions. AMF inoculation decreased proline concentration, but increased significantly leaf osmotic potential. Antioxidative enzymes responded differently to the salt and AMF treatments depending on the salt concentration and plant organ. Nonetheless, salt-induced malondialdehyde accumulation in the leaves diminished by AMF colonization. K and Ca contents were not affected by salt, while fungal colonization increased K in the roots and Ca in both leaves and roots. Our results indicated that enhancement of photosynthesis and ion homeostasis is involved in the tolerance of P. distans to both low and high salinity. AMF inoculation increased plants’ tolerance by augmentation of the above mechanisms accompanied by improvement of water relations and protection against oxidative damage in the leaves.

Salt tolerance of halophytes corresponds with the habitat requirement of the species. It is an important factor during the germination phase and it can determine successful establishment. This paper presents the effects of alternating temperature–light regimes (4/8°C, 10/20°C, 20/32°C; 12 h dark: 12 h light) and different salinity levels (0, 200, 400, 600 mmol l21 NaCl) on seed germination of five halophytes, Halimione pedunculata, Bupleurum tenuissimum, Aster tripolium, Triglochin maritimum and Armeria maritima. The five species differ with respect to family and life‐form and spatially correspond to a decreasing salt gradient (i.e. distance from salt water, with H. pedunculata being the most tolerant and A. maritima being the least). Armeria maritima, A. tripolium and T. maritimum seeds were additionally subjected to a cold stratification experiment. The results showed that Halimione pedunculata, an annual therophyte of year‐round heavily saline habitats, was dormant under all experimental conditions. Bupleurum tenuissimum, a species typical to sites of varying salinity prone to leaching during spring and autumn rainfall, germinated best under cold and warm temperatures, but only under non‐saline conditions. Aster tripolium and T. maritimum, close neighbours in salt marshes, showed very similar germination behaviour: seeds of both species tolerated high levels of salinity and germinated best in summer temperatures during periods of highest soil salinity, and germination was significantly promoted by cold. Armeria maritima, a species usually found on the marginal fringes of saline habitats, germinated only under low salt levels and maximum germination was under cold (spring) and warm (autumn) temperatures, with no significant effect of cold stratification.

Effects of arbuscular mycorrhizal fungi (AMF) and salt stress on nutrient acquisition and growth of two tomato cultivars exhibiting differences in salt tolerance were investigated. Plants were grown in a sterilized, low-P (silty clay) soil-sand mix. Salt was applied at saturation extract (ECe) values of 1.4 (control), 4.9 (medium) and 7.1 dS m–1 (high salt stress). Mycorrhizal colonization occurred irrespective of salt stress in both cultivars, but AMF colonization was higher under control than under saline soil conditions. The salt-tolerant cultivar Pello showed higher mycorrhizal colonization than the salt-sensitive cultivar Marriha. Shoot dry matter (DM) yield and leaf area were higher in mycorrhizal than nonmycorrhizal plants of both cultivars. Shoot DM and leaf area but not root DM were higher in Pello than Marriha. The enhancement in shoot DM due to AMF inoculation was 22% and 21% under control, 31% and 58% under medium, and 18% and 59% under high salinity for Pello and Marriha, respectively. For both cultivars, the contents of P, K, Zn, Cu, and Fe were higher in mycorrhizal than nonmycorrhizal plants under control and medium saline soil conditions. The enhancement in P, K, Zn, Cu, and Fe acquisition due to AMF inoculation was more pronounced in Marriha than in the Pello cultivar under saline conditions. The results suggest that Marriha benefited more from AMF colonization than Pello under saline soil conditions, despite the fact that Pello roots were highly infected with the AMF. Thus, it appears that Marriha is more dependent on AMF symbiosis than Pello.

The factors which may influence temporal and spatial variation in plant arbuscular mycorrhizal (AM) colonization and propagule occurrence were evaluated in a Portuguese salt marsh poor in plant diversity. Two distinct sites were studied: a more-flooded (low marsh) and a less-flooded zone (high marsh). AM root colonization, AM fungal spore number and inoculum potential, soil edaphic parameters and tidal flooding time periods were analysed. Levels of AM colonization were considerable in Aster tripolium and Inula crithmoides but very low in Puccinellia maritima and non-existent in Spartina maritima, Halimione portulacoides, Arthrocnemum fruticosum and Arthrocnemum perenne. Fungal diversity was very low, with Glomus geosporum dominant at both marsh zones. Colonization showed no spatial variation within marsh zones but temporal variation was observed in the high marsh, dependent on plant phenological phases. In the low marsh, no significantly seasonal variation was observed. Apparently, plant phenological events were diluted by stressful conditions (e.g. flooding, salinity). Spore density was significantly different between marsh zones and showed temporal variation in both zones. This study showed that distribution of mycorrhizas in salt marsh is more dependent on host plant species than on environmental stresses.

Maize, genetically modified with the insect toxin genes of Bacillus thuringiensis (Bt), is widely cultivated, yet its impacts on soil organisms are poorly understood. Arbuscular mycorrhizal fungi (AMF) form symbiotic associations with plant roots and may be uniquely sensitive to genetic changes within a plant host. In this field study, the effects of nine different lines of Bt maize and their corresponding non‐Bt parental isolines were evaluated on AMF colonization and community diversity in plant roots. Plants were harvested 60 days after sowing, and data were collected on plant growth and per cent AMF colonization of roots. AMF community composition in roots was assessed using 454 pyrosequencing of the 28S rRNA genes, and spatial variation in mycorrhizal communities within replicated experimental field plots was examined. Growth responses, per cent AMF colonization of roots and AMF community diversity in roots did not differ between Bt and non‐Bt maize, but root and shoot biomass and per cent colonization by arbuscules varied by maize cultivar. Plot identity had the most significant effect on plant growth, AMF colonization and AMF community composition in roots, indicating spatial heterogeneity in the field. Mycorrhizal fungal communities in maize roots were autocorrelated within approximately 1 m, but at greater distances, AMF community composition of roots differed between plants. Our findings indicate that spatial variation and heterogeneity in the field has a greater effect on the structure of AMF communities than host plant cultivar or modification by Bt toxin genes.

The arbuscular mycorrhizal status of fifteen mangroves and one mangrove associate was investigated from 27 sites of three inundation types namely, diurnal, usual springtide and summer springtide. Roots and rhizospheric soil samples were analysed for spore density, frequency of mycorrhizal colonization and some chemical characteristics of soil. Relative abundance, frequency and spore richness of AMF were assessed at each inundation type. All the plant species except Avicennia alba exhibited mycorrhizal colonization. The study demonstrated that mycorrhizal colonization and spore density were more influenced by host plant species than tidal inundation. Forty four AMF species belonging to six genera, namely Acaulospora, Entrophospora, Gigaspora, Glomus, Sclerocystis and Scutellospora, were recorded. Glomus mosseae exhibited highest frequency at all the inundation types; Glomus fistulosum, Sclerocystis coremioides and Glomus mosseae showed highest relative abundance at sites inundated by usual springtides, summer springtides and diurnal tides, respectively. Spore richness of AMF was of the order usual springtide > diurnal > summer springtide inundated sites. The mean spore richness was 3.27. Diurnally inundated sites had the lowest concentrations of salinity, available phosphorus, exchangeable potassium, sodium and magnesium. Statistical analyses indicated that mycorrhizal frequency and AMF spore richness were significantly negatively correlated to soil salinity. Spore richness was also significantly negatively correlated to available phosphorus. The soil parameters of the usual springtide inundated sites appeared to be favourable for the existence of maximum number of AMF. Glomus mosseae was the predominant species in terms of frequency in the soils of the Sundarbans.

We examined effects of aboveground herbivory by spider mites (Tetranychus urticae) on colonization and activity of arbuscular mycorrhizal fungi (AMF; Gigaspora margarita) using potted plants (Lotus japonicus). We evaluated changes in arbuscular mycorrhizal (AM) association two ways: (1) conventional trypan blue staining of mycorrhizal hyphae to examine AMF biomass in roots (mycorrhizal colonization) and (2) vital staining for a mycorrhizal enzyme (succinate dehydrogenase, SDH) to examine mycorrhizal activity (SDH activity). Mycorrhizal colonization and SDH activity started to increase 4 days after aboveground herbivory, and returned to the initial levels in the absence of mite herbivory in 7 and 12 days, respectively. These results suggest that the change in AM association in response to mite herbivory is a short-term response.

The mycorrhizal status of epiphytic, rupicolous, and terrestrial bromeliad species from the Brazilian Atlantic Rain Forest has been examined. Roots of 13 species of bromeliads were analyzed for the presence of mycorrhizal structures such as arbuscules, hyphae, and vesicles as well as other fungal structures. Rhizosphere soil was sampled to identify arbuscular mycorrhizal fungal (AMF) species associated only with terrestrial bromeliad species. Most specimens collected were epiphytic bromeliads in the genera Aechmea, Bilbergia, Nidularium, Tillandsia, and Vriesea. Differentiating structures of AMF were found in only three species of bromeliads. The pattern of mycorrhizal colonization was mainly internal, and external mycelium and arbuscules were observed only in the terrestrial Nidularium procerum. Root endophytes with dark brown septate mycelium, thin external hyphae, and Rhizoctonia-like sclerotia were also detected in some root segments. A total of ten spore morphotypes were recovered from the rhizosphere of N. procerum, with Acaulospora mellea, A. foveata, and Glomus sp. being the most common species recovered. Our study demonstrated that most of the epiphytic species are not associated with AMF. We attribute this mainly to the exposed bare root conditions found in epiphytic bromeliads.

Aims: After the determination of the toxic but nonlethal concentration of NaCl for cucumber, we examined the interaction between an ACC (1‐aminocyclopropane‐1‐carboxylate) deaminase producing bacterial strain and an arbuscular mycorrhizal fungus (AMF) and their effects on cucumber growth under salinity. Methods and Results: In the first experiment, cucumber seedlings were exposed to 0·1, 50, 100 or 200 mmol l?1 NaCl, and plant biomass and leaf area were measured. While seeds exposed to 200 mmol l?1 NaCl did not germinate, plant growth and leaf size were reduced by 50 or 100 mmol l?1 salt. The latter salt cancentration caused plant death in 1 month. In the second experiment, seeds were inoculated with the ACC deaminase‐producing strain Pseudomonas putida UW4 (AcdS+), its mutant unable to produce the enzyme (AcdS?), or the AMF Gigaspora rosea BEG9, individually or in combination and exposed to 75 mmol l?1 salt. Plant morphometric and root architectural parameters, mycorrhizal and bacterial colonization and the influence of each micro‐organism on the photosynthetic efficiency were evaluated. The AcdS+ strain or the AMF, inoculated alone, increased plant growth, affected root architecture and improved photosynthetic activity. Mycorrhizal colonization was inhibited by each bacterial strain. Conclusions: Salinity negatively affects cucumber growth and health, but root colonization by ACC deaminase‐producing bacteria or arbuscular mycorrhizal fungi can improve plant tolerance to such stressful condition. Significance and Impact of the Study: Arbuscular mycorrhizal fungus and bacterial ACC deaminase may ameliorate plant growth under stressful conditions. It was previously shown that, under optimal growth conditions, Ps. putida UW4 AcdS+ increases root colonization by Gi. rosea resulting in synergistic effects on cucumber growth. These results suggest that while in optimal conditions ACC deaminase is mainly involved in the bacteria/fungus interactions, while under stressful conditions this enzyme plays a role in plant/bacterium interactions. This finding is relevant from an ecological and an applicative point of view.

Abstract. Mainland salt marshes in Schleswig-Holstein (northern Germany) have been grazed intensively by sheep for several decades. In 1988 experimental sites were established in the lower and middle salt marsh of Sönke-Nissen-Koog and subjected to different grazing intensities. From 1989 to 1993 the impact of sheep grazing on the composition and structure of the salt marsh vegetation was studied through the yearly analysis of permanent plots, vegetation mapping and measurements of the vegetation height. The intensively grazed site (10 sheep/ha) is covered by a short monotonous Puccinellia maritima sward with Salicornia europaea and Suaeda maritima. Halimione portulacoides and Aster tripolium, especially flowering plants, are rare. On the sites with 1.5 and 3 sheep/ha Puccinellia maritima remained dominant. The population density of Salicornia europaea decreases after reduction of the grazing intensity, whereas Suaeda maritima finds optimal growing conditions. Stands of Halimione portulacoides and flowering Aster tripolium plants are rare near the sea dike but their cover and size increases further away from the dike. In the plot with 1.5 sheep/ha the height of the vegetation increases along the gradient from the dike towards the tidal flats, due to local differences in actual grazing intensity. When grazing is stopped, Puccinellia maritima is successively replaced by Festuca rubra, Halimione portulacoides and Aster tripolium. High variability of vegetation height indicates structural diversity. Patches of higher and lower vegetation correspond with the distribution pattern of different plant species. In terms of nature conservation cessation of grazing is recommended.

Here, we examined the colonization by fungal root symbionts in the cultivated Andean grain Chenopodium quinoa and in 12 species that dominate plant communities in the Bolivian Altiplano above 3,700 m elevation and explore for the possible relationships between fungal colonization and fine root proportion. The 12 most abundant species in the study area were consistently colonized by AMF and DSE. In contrast, the annual Andean grain C. quinoa showed negligible or absence of mycorrhizal fungi colonizing roots. On the other hand, C. quinoa, Junelia seriphioides and Chersodoma jodopappa were infected to a varying degree by the root pathogen Olpidium sp. We observed no relationship between AMF and DSE colonization and proportion of fine roots in the root system, but instead, the ratio between DSE and AMF colonization (ratio DSE/AMF) negatively related with proportion of fine roots. Our findings support the hypothesis regarding the importance of DSE at high altitudes and suggest a functional relationship between the rate of DSE/AMF and proportion of fine roots. The colonization by the root pathogen Olpidium sp. in C. quinoa deserves further study since this Andean grain is increasingly important for the local economy in these marginal areas.

Two wheat (Triticum aestivum L.) cultivars, Sids 1 and Giza 168, were grown under non-saline or saline conditions (4.7 and 9.4 dS m?1) with and without arbuscular mycorrhizal fungi (AMF) inoculation. Salt stress considerably decreased root colonization, plant productivity and N, P, K+, Fe, Zn and Cu concentrations, while it increased Na+ level, particularly in Giza 168. Mycorrhizal colonization significantly enhanced plant productivity and N, P, K+, Fe, Zn and Cu acquisition, while it diminished Na+ uptake, especially in Sids 1. Salinity increased putrescine level in Giza 168, however, values of spermidine and spermine increased in Sids 1 and decreased in Giza 168. Mycorrhization changed the polyamine balance under saline conditions, an increase in putrescine level associated with low contents of spermidine and spermine in Giza 168 was observed, while Sids 1 showed a decrease in putrescine and high increase in spermidine and spermine. Moreover, mycorrhizal inoculation significantly reduced the activities of diamine oxidase and polyamine oxidase in salt-stressed wheat plants. Modulation of nutrient acquisition and polyamine pool can be one of the mechanisms used by AMF to improve wheat adaptation to saline soils. This is the first report dealing with mycorrhization effect on diamine oxidase and polyamine oxidase activities under salt stress.

The controlled disposal of tannery sludge in agricultural soils is a viable alternative for recycling such waste; however, the impact of this practice on the arbuscular mycorrhizal fungi (AMF) communities is not well understood. We studied the effects of low-chromium tannery sludge amendment in soils on AMF spore density, species richness and diversity, and root colonization levels. Sludge was applied at four doses to an agricultural field in Rolandia, Paraná state, Brazil. The sludge was left undisturbed on the soil surface and then the area was harrowed and planted with corn. The soil was sampled at four intervals and corn roots once within a year (2007/2008). AMF spore density was low (1 to 49 spores per 50 cm3 of soil) and decreased as doses of tannery sludge increased. AMF root colonization was high (64%) and unaffected by tannery sludge. Eighteen AMF species belonging to six genera (Acaulospora, Glomus, Gigaspora, Scutellospora, Paraglomus, and Ambispora) were recorded. At the sludge doses of 9.0 and 22.6 Mg ha−1, we observed a decrease in AMF species richness and diversity, and changes in their relative frequencies. Hierarchical grouping analysis showed that adding tannery waste to the soil altered AMF spore community in relation to the control, modifying the mycorrhizal status of soil and selectively favoring the sporulation of certain species.

The effect of sulphide on the growth of several species of salt-marsh plants was investigated. Relative growth rates were significantly reduced in two upper-marsh species, Festuca rubra and Atriplex patula, and in the lower-marsh species Puccinellia maritima. However the growth of Salicornia europaea, a species frequently associated with sulphide-containing sediments, was unaffected. In a separate experiment the wide ranging halophyte Aster tripolium, also appeared to be tolerant of sulphide at a concentration frequently encountered in salt marshes. Sulphide pretreatment inhibited the activity of two metallo-enzymes, polyphenol oxidase and external phosphatase, in plants from the upper marsh, but had no effect on enzymes from P. maritima or S. europaea. The rate of respiration by root tissue was significantly reduced in all of the species investigated but whereas the uptake of 86rubidium was markedly inhibited in the other three species, uptake by S. europaea showed a significant stimulation. Similarly, whereas sulphide-grown plants of F. rubra, A. patula and P. maritima had a considerably reduced tissue iron content, the total iron concentration in S. europaea tissues was comparable to that of the controls. When the sulphide-tolerant species A. tripolium was grown in sulphide-containing media there was no significant effect on the tissue concentration of any of the elements investigated. These results are discussed in relation to possible mechanisms of sulphide toxicity and resistance.

This study was conducted to examine the role of arbuscular mycorrhiza fungi (AMF) in alleviating the adverse effects of drought stress on damask rose (Rosa damascena Mill.) plants. Four levels of drought stress (100, 75, 50, and 25% FC) were examined on mycorrhizal and non-mycorrhizal plants in pots filled with sterilized soil. Our results showed that increasing drought stress level decreased all growth parameters, nutrient contents, gas exchange parameters, and water relations indicators. Under different levels of drought stress, mycorrhizal colonization significantly increased all studied parameters. Pn, gs, and E of the mycorrhizal plants was higher than those of non-mycorrhizal plants under different levels of drought stress. The increase in those rates was proportional the level of the mycorrhizal colonization in the roots of these plants. Majority of growth, nutrition, water status and photosynthetic parameters had a great dependency on the mycorrhizal colonization under all levels of drought stress. The results obtained in this study provide a clear evidence that AMF colonization can enhance growth, flower quality and adaptation of rose plants under different drought stress levels, particularly at high level of drought stress via improving their water relations and photosynthetic status. It could be concluded that colonization with AMF could help plants to tolerate the harmful effects caused by drought stress in arid and semi-arid regions.
