Fertilization rate and embryonic mortality were assessed in 636 ewes inseminated in each uterine horn with 50 x 10(6) frozen spermatozoa from four control rams and from four rams submitted to a moderate (1.4-2.2 degrees C), but repeated, intermittent (16 h/day for 21 consecutive days) increase in their subcutaneous scrotal temperature by means of scrotal insulation. Pregnancy was assessed twice in each ewe from concentration of progesterone in blood plasma at 17 days and by ultrasound at 65 days after insemination. No differences were observed in the pregnancy rate at 17 days between ewes inseminated with semen collected from control rams (56.0, 65.2, 66.7 and 60.3%) and from heated rams (60.6, 71.8, 63.6 and 48.2%) before or after 4, 15 and 21 days of heating, respectively. In contrast, the rate of embryonic mortality between 17 and 65 days after insemination was significantly higher at days 4, 15 and 21 in the heated rams (78.7, 78.6 and 93%) than in the control rams (55, 59 and 65.7%). These results indicate that an intermittent slight, but repeated, increase in the subcutaneous scrotal temperature could induce a significant increase in the embryonic mortality rate. As these changes were apparent on day 4 of heating, an effect must have occurred on sperm stored in the epididymis.  相似文献

Epididymal fluid may contain substances which promote development of the fertilizing capacity of testicular spermatozoa under in vitro conditions, provided that the spermatozoa are exposed to such substances for long periods of time. In an attempt to resolve this question, the fertilizing capacity of testicular spermatozoa was assessed before and after storage in cauda epididymal fluid and comparisons made with ejaculated spermatozoa from the same rams. Of the 13 eggs examined from the group of ewes inseminated with ejaculated spermatozoa 61.5% were found to be in the 2-to 8-cell stage. No fertilized eggs were recovered from ewes impregnated with freshly collected testicular spermatozoa. Nor were any cleaved eggs obtained from the group of ewes inseminated with testicular spermatozoa stored in cauda epididymal fluid at 4°C for 7 to 11 days. We suggest there-fore, that in order to develop maximal fertilizing capacity, mammalian spermatozoa must be exposed to specific concentrated testicular and epididymal secretions in a sequential order and within strict time limits.  相似文献

Fertility data from 8 artificial insemination programs, involving more than 5000 ewes and 110 rams in 3 flocks, were analyzed to determine variation due to individual AI program and ram in the reproductive performance of ewes inseminated with frozen-thawed semen by laparoscopy. The semen had been previously frozen by commercial AI centers in either pellets or straws. Both AI program and individual ram affected the proportion of ewes pregnant and the number of fetuses per ewe inseminated, but not the number of fetuses per pregnant ewe. Semen samples from 97 of the rams used were analyzed on a Hamilton Thorn HTM 2000 image analyzer for sperm concentration, percentage of motile and progressively motile spermatozoa, mean progressive velocity, and mean linear index. The correlations between these traits and reproductive performance obtained after insemination were calculated. There was large variation in the quantity and quality of the frozen semen, but only the number of total and motile spermatozoa inseminated per ewe was correlated with fertility (0.25 and 0.26, respectively). Regression analysis showed that none of the traits measured were useful for predicting fertility.  相似文献

Experiments were conducted to determine the influence of sexual stimulation on testicular size and morphology.Scrotal volume decreased by 18–26% in rams mated for 4–6 weeks on two commercial farms, with only part of the decrease being accounted for by the 4–12% decrease in live weight. Similar observations were made over 7–8 weeks on rams exposed to increasing intensity of sexual stimulation by having rams either isolated, non-mated with or without non-oestrous ewes or mated with oestrous ewes. Again live weight change could not account entirely for the decrease in scrotal volume. The decrease in testicular size was confirmed on castration of the rams at the end of the experimental period. In addition to the 15–42% reduction in testicular weight with increased sexual stimulation, there was a reduction in the number of spermatozoa in the testes (21–36%) and an increase in the volumetric proportion of intertubular tissue (16–58%). A decrease in the volumetric proportion of spermatocytes (18–47%) and of round spermatids (13–57%) in the testicular tissue indicated that sexual stimulation was affecting spermatogenesis and decreasing daily production of spermatozoa.A decrease in the number of spermatozoa in the epididymides (24–26%) with increased sexual stimulation reflected an increased outflow of spermatozoa and/or reduced production of spermatozoa.  相似文献

The role of seminal plasma (SP) components on the maintenance of motility, viability and fertilising ability of frozen-thawed spermatozoa is of considerable interest. However, differences observed in constituents of SP among males could explain differences in fertility obtained in vivo. Two experiments were designed to examine the effects of seminal plasma on fertility from cervically inseminated frozen-thawed semen. The objective of Experiment 1 was to investigate if source or type of SP influences pregnancy rate. Seminal plasma was collected from rams previously classified as having either High (HSP; n=3) or Low (LSP; n=3) fertility in vivo. Artificial SP (fructose/sodium solution with 10% BSA; ASP) was made. Frozen semen from the same 6 rams was thawed and inseminated (Control) or resuspended either in HSP, LSP or ASP (20% in semen) prior to insemination of ewes (n=284, over 2 farms). The overall pregnancy rate was 28.1%. Treatments (Control, ASP, HSP and LSP) were not significantly different (P>0.3). There was no difference between HSP and LSP (P>0.5), and no effect of using ASP compared to ram SP (P>0.7), on pregnancy rate. As there was no effect of SP on pregnancy rate a repeat experiment (Experiment 2) was designed to test the effect of washing and selecting motile sperm prior to resuspending in phosphate-buffered saline (PBS) containing SP on pregnancy rate. Frozen-thawed semen from each of 2 rams was centrifuged through a density gradient, pellets were centrifuged through a wash medium and the sperm concentration/ram was counted. Sperm cells were resuspended in: (1) control PBS, (2) PBS containing 30% HSP or (3) PBS containing 30% LSP to give 100 x 10(6) motile sperm in 0.25 mL. Control straws were thawed and inseminated directly. Ewes (n=223 over 2 farms) were inseminated 57 h post-sponge withdrawal and those not returning to oestrus were slaughtered 29-50 days post-insemination for pregnancy determination. In Experiment 2, the pregnancy rate for Control, PBS, HSP and LSP were 15.4%, 2.3%, 0% and 0%, respectively, for Farm 1 (P>0.05) and 17.8%, 11.0%, 3.9% and 12.4%, respectively, for Farm 2. Under the conditions of the current study, addition of SP from different donors of either High or Low fertility status to frozen-thawed ram semen post-thawing did not improve pregnancy rate in ewes. ASP had no effect on pregnancy rate in ewes when added to frozen-thawed semen. Washing and selection of motile sperm prior to resuspension in PBS with or without SP (30%) before insemination had a negative effect on pregnancy rate in cervically inseminated ewes. Hence, the addition of seminal plasma or some of its constituents to semen does not appear to improve pregnancy rate in cervically inseminated ewes.  相似文献

In Exp. 1, 4 groups of 50 recently weaned ewes were exposed to various degrees of contact with rams for 65 days, followed by exposure to novel rams for 4 days. Ovarian activity in the ewes was determined by laparoscopy on Days 29, 65 and 69 of treatment. There were no treatment differences in the percentage of ewes ovulating on Day 4 whereas by Day 29 more ewes in clear fenceline and full ram contact were ovulating compared to controls (P less than 0.05, P less than 0.001). After 65 days ovarian activity was significant only in those ewes in full contact with rams (P less than 0.001). Between 89 and 95% of ewes remaining anovulatory after 65 days ovulated after 4 days of full contact with novel rams. In Exp. 2, 4 groups of about 30 anovulatory ewes were exposed to various degrees of contact with rams for 5 days. Ovarian activity was assessed before and after treatment by laparoscopy. After 5 days, more ewes were ovulating in response to full ram contact than in any other treatment (P less than 0.05) and more ewes in fenceline contact with rams or with rams plus ewes were ovulating than in the isolated control treatment (P less than 0.01). In Exp. 3, 6 groups of about 40 anovulatory ewes were exposed to face masks with and without rams' wool and/or various degrees of contact with rams for 5 days. Two closed lines of sheep selected for high and low prolificacy were established in 1969. Rams within each line were selected on the basis of their dams' lambing records. Foundation ewes were assigned at random to selected rams, and no ewe selection was practiced until replacement yearlings were selected for mating. Selection of the rams used to mate the foundation ewes resulted in more lambs born per 100 ewes exposed to the ram (P<.10) and more lambs born per 100 ewes lambing from the high line rams (P<.05).Seven rams from each line were then used to estimate fertilization rate and embryonic survival rate when mated to an unrelated group of ewes. There was no difference in ovulation rate or fertilization rate of ewes mated to low and high line rams. Percent eggs recovered per CL was 80 and 92 (P<.01) for low and high line rams, respectively. At 35 days 82 and 86% of the ewes were pregnant, and 84 and 93% of the CL were represented by embryos in the pregnant ewes mated to low and high lines, respectively. Estimated embryonic survival rates were 81 and 89% in ewes mated to low and high line rams, respectively.  相似文献

Changes in LH, FSH, and testosterone concentrations, testicular firmness and resilience, and scrotal circumference were monitored in 16 Corriedale rams (8 experienced adult and 8 inexperienced young) for 20 days during which they were used to stimulate ewes. The experiment was conducted during November (mid-non-breeding season). Increases (P<0.05) were observed in LH and testosterone concentrations and in testicular firmness and resilience during the first 4 days when rams were in permanent contact with estrual ewes. During the following 13 days, when rams were in contact with non-estrual ewes (i.e. initially estrual ewes were no longer in estrus), LH and testosterone concentrations decreased. When initially anestrous ewes exhibited estrus 17 to 20 days later, concentrations of testosterone increased. Testicular firmness and resilience remained high throughout the period. We conclude that: (1) rams used to stimulate anestrous ewes show an increase in LH and testosterone concentrations beginning at 12 h after joining, and greater concentrations are maintained while estrual ewes and mating are allowed; and (2) estrous and mating activity are probably the most important stimuli for the increase in hormone concentrations.  相似文献

European mouflon sheep are an endangered species of ovidae residing primarily in the mountenous habitat of the islands of Sardinia and Corsica. The purpose of this study was to assess the fertilizing capacity of cryopreserved European mouflon spermatozoa after AI in synchronized mouflon and domestic ewes and after IVF in in vitro matured mouflon and domestic ewe oocytes collected by OPU technique. Domestic ram (Ovis aries) spermatozoa served as control. Semen was collected by artificial vagina from three mouflons and three domestic rams during the breeding season and was cryopreserved. At thawing, no significant differences in sperm viability were found between the wild and the domestic species (53.1 +/- 4.6% versus 56.0 +/- 4.7%) whereas the percentage of acrosome-intact sperm was lower in mouflon (55.5 +/- 4.6%) than in ram semen (62.7 +/- 3.1%; P < 0.05). Lambing rate did not differ between synchronized mouflon and domestic ewes (5/11 versus 8/12) after 150 and 156 days of pregnancy, respectively. After two OPU sessions, 87 and 132 oocytes were collected from three hyperstimulated mouflon and three domestic ewes. Cryopreserved/thawed semen was inseminated with an endoscope into the uterus of corresponding species during the non-breeding season. The oocytes were matured and fertilized in vitro; 61/73 mouflon and 81/101 domestic ewe oocytes were found to be fertilized. From these, we obtained 6/61 and 17/81 blastocysts. After vitrification and thawing, the hatching rate showed no significant difference between mouflon and sheep blastocysts (4/6 versus 14/17). In conclusion, our data showed that cryopreserved mouflon spermatozoa can be successfully used to carry out a genuine and complete program of genetic restoration in small and isolated groups of European mouflons.  相似文献

In a series of experiments spermatozoa were inseminated blindly into the vagina of ewes and then recovered at varying times after insemination. Most of the spermatozoa inseminated were lost by drainage through the vulva. The rate of loss was not affected by the motility of spermatozoa or oestrous state of the ewe. Initially after insemination the loss was not rapid with 82% of the insemination 18% of spermatozoa remained and by 12 h 10% remained. Spermatozoa were removed from the vagina during withdrawal of the penis after intromission and the extent of this loss varied between rams and with the volume of semen already in the vagina. Up to half the inseminate was lost in this way when there was 0.5 ml of semen in the vagina but only 11% was lost when the volume of inseminate was 0.1 ml. The unavoidable loss of spermatozoa may influence the quantity available for fertilizing ova.  相似文献

Alternative Norwegian sheep breeding schemes were evaluated by stochastic simulation of a breeding population with about 120 000 ewes, considering the gain for an aggregate genotype including nine traits and also the rate of inbreeding. The schemes were: a scheme where both young unproven rams (test rams) and proven rams (elite rams) are used in artificial insemination (AI scheme), a scheme with test rams in natural mating in ram circles and elite rams (from one and a half years of age) in AI across all flocks in the country (NMAI2 scheme), a scheme where, in addition to testing rams, the youngest elite rams (one and a half years of age) are also used in natural mating in ram circles, while older elite rams are used in AI (NMAI1 scheme), and a scheme, acting as a control, where both test and elite rams are used in natural mating (NM scheme). Within the NMAI- and AI-schemes, experimentation was performed for percent ewes inseminated to elite rams v. test rams (EM%), numbers of ewes inseminated per elite ram (EAIn), and numbers of ewes mated per test ram by natural service (TNMn) or by AI (TAIn), respectively. With a restriction on the rate of inbreeding (⩽0.8% per generation), the AI scheme gave similar gain to the NMAI2 scheme (and about 40% more than did the NM scheme). Less gain was generated by the NMAI1 scheme, but it was still considerably more than for the NM scheme (about 25%). In the AI scheme, relatively few ewes (200/300) should be inseminated to each test/elite ram, and a low EM% should be chosen (10%). In the NMAI schemes, TNMn should be relatively high (40 to 50), combined with average and somewhat larger than average EAIn (NMAI2: 700 ewes, NMAI1: 900 ewes), and EM% medium (30%).  相似文献

In three experiments, the onset of oestrus, time of ovulation and lambing after intrauterine insemination with frozen-thawed semen were examined following synchronisation of oestrus using intravaginal progestagen-impregnated sponges (inserted for 12 days) and an injection of PMSG at sponge removal.

The number (and percentage) of ewes detected in oestrus 12, 24, 36, 48, 60 and 72 h after sponge removal was 1 (0.3), 2 (0.6), 17 (5.2), 120 (36.7), 65 (20.0) and 10 (3.1) respectively. One hundred and twelve ewes (34.3%) remained unmarked. Egg fertilisation rates were not different between ewes irrespective of time of onset of oestrus or whether or not ewes were marked.

The median time of ovulation with respect to sponge removal (with 95% fiducial limits) for ewes joined with vasectomised rams (10:1) at spronge removal (teased ewes) was 55.8 h (54.61–57.09) and for unteased ewes 59.7 h (58.27–61.12).

In the third experiment, a total of 394 ewes were inseminated by laparoscopy with frozen-thawed semen. The percentage of ewes lambing and lambs born per ewe inseminated, and number of lambs born per ewe lambing for inseminations 48, 60, 72 and 78 h after sponge removal were 45.9, 57.7 and 1.25; 55.1, 72.0 and 1.31; 57.4, 80.9 and 1.41; and 39.3, 60.7 and 1.54, and for 59 control ewes receiving fresh semen by cervical insemination 47.5, 69.5 and 1.46 respectively. The lambing data after insemination with frozen semen was not different to that of the controls. The percentage of ewes lambing and lambs born per ewe inseminated increased with time of insemination at 48, 60 and 72 h (linear, P < 0.01) but was lower for inseminations at 78 h after sponge removal. Number of lambs born per ewe lambing increased with time of insemination after sponge removal (linear, P < 0.05).  相似文献

Ewes of the Préalpes-du-Sud breed (n=112) were mated with fertile rams and were used to investigate the effect of the presence of vasectomized rams during pregnancy on reproductive outcomes. Ewes in the control group (n=56) were isolated from rams during the whole period of pregnancy, whereas those in the experimental group (n=56) were kept with vasectomized rams from day 10 post-mating until lambing. At day 10 post-mating, a series of blood samples was collected every 15 min for 8 h from five control ewes and five experimental ewes to determine their patterns of LH secretion. The introduction of the ram was associated with a rapid increase of pulsatile LH release. The lag between the introduction of the ram and the onset of the first episodic LH release was less than 15 min. The mean(±sem) number of LH pulses/4 h after the introduction of the ram (2.8±0.4) was significantly higher (P<0.01) than that observed/4 h before the introduction of the ram (1.4±0.2). Although more ewes were pregnant in the control group (87.5%) than in the ram-exposed group (82.1%), the difference was not significant. The presence of rams did not affect gestation length (145.8 days), overall lamb mortality (3.5%) or birth weights of single (3.96 kg), twin (3.24 kg) or triplet (2.59 kg) lambs. The proportion of ewes with multiple births in the control group (69.4%) was significantly greater (P<0.05) than that in the ram-exposed ewes (47.8%). The ewes in the control group had significantly more (P<0.01) twin lambs born alive (72.3%) than the ewes in the ram-exposed group (50.0%). In conclusion, the presence of vasectomized rams during early pregnancy affected the incidence of multiple births but did not affect pregnancy rate or gestation length. The altered fertility of ewes exposed to vasectomized rams may reflect changes in embryonic loss during early pregnancy.  相似文献

Two experiments were performed to determine the endocrine and ovarian changes in medroxyprogesterone acetate (MAP)-primed ewes after ram introduction. Experiment 1 was performed during the mid-breeding season with 71 ewes primed with an intravaginal MAP sponge for 12 days. While the control (C) ewes (n = 35) were in permanent contact with rams, the ram effect (RE) ewes (n = 36) were isolated for 34 days prior to contact with rams. At sponge withdrawal, all ewes were joined with eight sexually experienced marking Corriedale rams and estrus was recorded over the next 4 days. The ovaries were observed by laparoscopy 4–6 days after estrus. Four weeks later, pregnancy was determined by transrectal ultrasonography. In eight ewes from each group, ovaries were ultrasonographically scanned; FSH, LH, and estradiol-17β were measured every 12 hours until ovulation or 96 hours after estrus. The response to the rams was not affected by the fact that ewes had been kept or not in close contact with males before teasing. No differences were found in FSH, LH, estradiol-17β concentrations, growth of the ovulatory follicle, onset of estrus, ovulation rate, or pregnancy rate. Experiment 2 was performed with 14 ewes during the nonbreeding season. Ewes were isolated from rams for 1 month, and received a 6-day MAP priming. Ovaries were ultrasonographically scanned every 12 hours, and FSH, LH, estradiol-17β, and progesterone were measured. Ewes that ovulated and came into estrus had higher FSH and estradiol-17β levels before introduction of the rams than did ewes that had a silent ovulation. The endocrine pattern of the induced follicular phase of ewes that came into estrus was more similar to a normal follicular phase, than in ewes that had a silent ovulation. The follicle that finally ovulated tended to emerge earlier and in a more synchronized fashion in those ewes that did come into estrus. All ewes that ovulated had an LH surge and reached higher maximum FSH levels than ewes that did not ovulate, none of which had an LH surge. We conclude that (a) the effect of ram introduction in cyclic ewes treated with MAP may vary depending on the time of the breeding season at which teasing is performed; (b) patterns of FSH, and estradiol-17β concentrations, as indicators of activity of the reproductive axis, may be used to classify depth of anestrus; and (c) the endocrine pattern of the induced follicular phase, which is related to the depth of anestrus, may be reflected in the behavioral responses to MAP priming and the ram effect.  相似文献

This study compared the affect of short-term and continuous exposure to rams during the transition between anoestrus and the breeding season on the distribution of mating and subsequent lambing. Further, within ewes continuously exposed to rams we investigated the effect of replacing these rams every 17 days with 'novel' rams. During August (late anoestrus, Northern Hemisphere), multiparous, North of England mule ewes were allocated to one of four groups: SVR ewes were exposed to vasectomised rams for 24h on Day 0 (short term; n=109), RVR ewes were exposed to vasectomised rams for 24h on Days 0, 17 and 34 (short term; n=113); PVR ewes were exposed to vasectomised rams on Day 0 and remained with the same rams for the duration of the pre-mating period (continuous; n=104); NVR ewes were continuously exposed to vasectomised rams from Day 0 with the rams replaced with 'novel' rams every 17 days (continuous; n=113). Blood samples were collected from a subset of ewes (n=22 per group) to monitor progesterone. On Day 50, harnessed, entire rams were introduced for mating and raddle marks recorded daily for the first 17 days. The median date of mating occurred 1 day earlier in NVR ewes than PVR ewes (P<0.05). A synchrony score calculated from the blood sampled ewes showed that the distribution of mating was more synchronised in PVR and NVR ewes than SVR and RVR ewes (P<0.001). PVR and NVR ewes had an earlier onset of cyclic activity than RVR ewes (P<0.01). However, only NVR ewes differed from SVR ewes in this variable (P<0.05). Within ewes lambing to first service, the median date of lambing of PVR, NVR and SVR ewes occurred at least 2 days earlier than RVR ewes (at least P<0.05). Further, PVR and NVR ewes had a more compact distribution of lambing than SVR and RVR ewes (P<0.05) and lambing was more compact in NVR ewes than PVR ewes (P<0.05). In conclusion, ewes in continuous contact with rams prior to mating had a more synchronised distribution of mating and lambing than ewes given only short-term exposure to rams. This distribution of mating in continuous ram exposed ewes can be further enhanced by periodic exposure to novel rams.  相似文献

A total of 415 fat tailed ewes were randomly assigned to two groups to assess the effect of duration of melengestrol acetate (MGA) (9 versus 12d) administration on reproductive parameters associated with laparoscopic artificial insemination. At the end of MGA treatment, ewes in each group were subdivided and inseminated with one of two different insemination doses (10×10(7) or 20×10(7) sperm per 0.5 ml insemination dose) of fresh diluted semen. Inseminations were carried out 11-18 h after first detected estrus. Ewes were screened for their return to oestrus from 10 to 21 days post AI and inseminated at their returned oestrus. Pregnancy diagnosis was done from approximately 55 days after insemination in both synchronized and return estrus. For short (9-day) and long (12-day) term MGA treated groups, estrus rates were 62% versus 89% (P<0.0001), respectively. Ewes (n=115) that returned to estrus were inseminated (7-11h after estrus detection) with fresh diluted semen at different doses (20×10(7) or 40×10(7) or 60×10(7) sperm per 0.5 ml insemination dose). Pregnancy rates were 41% and 44% for short term and long term MGA treated ewes, respectively. Pregnancy rate of ewes which returned to oestrus was 53.4%. There was a significant (P<0.05) increase in pregnancy rates (38-52% for 11-16 h; 63% for 17-18 h) when insemination was held at 17-18 h after first detected estrus following MGA treatments. Pregnancy rates were found to be similar in ewes inseminated with 10×10(7) (36%) or 20×10(7) (47%) motile spermatozoa at first AI, and 20×10(7) (44%) or 40×10(7) (59%) or 60×10(7)(48%) at second AI. It was concluded that short term MGA treated ewes were recorded with lower estrus rates but was similar to pregnancy rates with long term MGA treatment. Acceptable pregnancy rates were achieved in MGA induced estrus when insemination is conducted at 17-18 h after estrus onset and with 20×10(7) sperm per insemination dose.  相似文献

Ewes that were untreated, fed lupins or fed lupins and immunized against androstenedione were artificially inseminated. The percentage of ewes pregnant at 36-45 days after insemination (fertility) was 8% higher in ewes that had more than one ovulation than in those that had only one ovulation. Maximum fertility was achieved with 50 x 10(6) spermatozoa and this did not vary with the number of ovulations that ewes had. Among the pregnant, twin-ovulating ewes, embryo survival increased as the number of spermatozoa inseminated increased from 25 x 10(6) to 400 x 10(6). Immunization of ewes against androstenedione increased ovulation rate but reduced fertility, and reduced embryo survival among twin-ovulation ewes.  相似文献

An experiment was undertaken to assess the fertilizing capacity of sex-sorted, frozen-thawed ram spermatozoa, artificially inseminated into superovulated ewes, and the quality and survivability of the resultant pre-sexed embryos. Synchronized (intravaginal progestagen pessary and GnRH) donors were superovulated using PMSG and repeat ovarian stimulation with FSH before insemination. Ewes (n=67) were inseminated with either 30x10(6) or 15x10(6) motile non-sorted (control) or 15x10(6) motile sex-sorted (sorted) frozen-thawed spermatozoa (control: C30 or C15; sorted: S15, respectively) and the resultant embryos transferred immediately into synchronized recipients (n=160). The percentage of transferable embryos, pregnancy rate and embryo survival were similar (P>0.05) across all treatments. Oocyte cleavage rate was higher for ewes inseminated with S15 (172/230; 74.8%; P<0.05) than for C15 (97/151; 64.2%) or C30 (89/141; 63.1%) spermatozoa. Of the lambs resulting from embryos produced with sex-sorted spermatozoa, 86/93 (92.5%) were born of the predicted sex. This study demonstrated for the first time that pre-sexed offspring derived from superovulated sheep can be produced following transfer of embryos. Furthermore, sex-sorting by flow cytometry did not compromise the in vivo fertilizing capacity of ram spermatozoa in superovulated sheep, nor did it affect the quality or survivability of the resultant embryos.  相似文献

The effects of active immunization against oestradiol-17 beta on the ovarian response to pregnant mare serum gonadotrophin (PMSG) was investigated in Merino ewes. Immunized (79) and control (41) ewes were synchronized with intravaginal sponges, given either 750 or 1500 i.u. PMSG and then mated to rams or inseminated laparoscopically with fresh diluted semen. All control ewes mated naturally exhibited oestrus and 40 out of 41 control ewes ovulated. The ovulation rate was higher in the controls receiving 1500 i.u. PMSG than in those ewes which received 750 i.u. PMSG (10.2 v. 3.3). Immunization against oestradiol-17 beta resulted in antibody titres varying from 100 to more than 100 000 in plasma taken 1-4 days after mating. The ovarian response increased significantly in the lowest titre group (100-1000) in conjunction with stimulation with 1500 i.u. PMSG. In these ewes the ovulation rate increased over controls (16.7 v. 10.2) as did the total ovarian response, which includes follicles greater than 10 mm diameter (22.3 v. 11.1). The total ovarian response was also increased in those ewes given 750 i.u. PMSG which had titres in the 1000-10 000 and 10 000-100 000 range, but this was not accompanied by significant increases in the ovulation rate. In general, the higher titre levels (greater than 1000) were correlated with decreases in the proportion of ewes showing oestrus and ovulating and in the embryo recovery rate. The 1500 i.u. PMSG treatment group with the highest titres (greater than 10 000) also showed a significant drop in the ovulation rate as compared to the 1500 i.u. PMSG controls.  相似文献

The experiment measured lamb responses to supplementation of the pregnant ewe diet with vitamin E above requirement. Crossbred ewes were mated with either Suffolk or Texel rams. Twin-bearing ewes were randomly allocated (approximately 21 months of age at allocation) to one of four treatment groups (20 ewes per group, 10 mated with Suffolk and 10 with Texel rams). Treatments imposed were 50, 100, 150 or 250 IU supplementary vitamin E per ewe per day to give a four treatment by two sire-type factorial experimental design. Ewes were fed concentrates to meet energy requirements for stage of pregnancy and hay ad libitum. Diets were introduced approximately 6 weeks before lambing. Blood samples were obtained prior to introduction of diets, 17 days after introduction of diets and within 24 h of lambing from a subset of eight ewes per treatment (32 total). Colostrum samples were obtained from 10 ewes per treatment, 12 h after birth of the first lamb. All births were observed and a lamb vigour score was assigned to each lamb 5 min after birth. At 1 and 12 h after birth, rectal temperature, and at 12 h after birth, sex, crown-rump length and BW of each lamb were recorded. Mean ewe plasma α-tocopherol concentration prior to introduction of the diets was 1.5 μg/ml (s.e.m. 0.09) and did not differ between groups. There were positive linear (P < 0.001) effects of dietary vitamin E on plasma (17 days after introduction of diets) and colostrum (12 h after birth) α-tocopherol concentrations. Lamb vigour scores were superior (P < 0.001) for lambs sired by Texel rather than Suffolk rams but there were no differences as a result of vitamin E supplementation. Lamb mortality was low and unrelated to either sire or supplementary vitamin E. Lamb birth and weaning weights were also unaffected by vitamin E supplementation. Supplementing the ewe with vitamin E therefore had no effect on any lamb measurements.  相似文献
