Obtaining a reliable estimation of the methane potential of organic waste streams in anaerobic digestion, for which a biochemical methane potential (BMP) test is often used, is of high importance. Standardization of this BMP test is required to ensure inter-laboratory repeatability and accuracy of the BMP results. Therefore, guidelines were set out; yet, these do not provide sufficient information concerning origin of and the microbial community in the test inoculum. Here, the specific contribution of the methanogenic community on the BMP test results was evaluated. The biomethane potential of four different substrates (molasses, bio-refinery waste, liquid manure and high-rate activated sludge) was determined by means of four different inocula from full-scale anaerobic digestion plants. A significant effect of the selected inoculum on the BMP result was observed for two out of four substrates. This inoculum effect could be attributed to the abundance of methanogens and a potential inhibiting effect in the inoculum itself, demonstrating the importance of inoculum selection for BMP testing. We recommend the application of granular sludge as an inoculum, because of its higher methanogenic abundance and activity, and protection from bulk solutions, compared with other inocula.  相似文献

A method is described to estimate the biodegradation potential of soluble, insoluble, and unknown organic chemicals. The method consists of two stages: (i) generation of a microbial inoculum in a bench scale semicontinuous activated sludge system during which microorganisms are acclimated to test material and the removal of dissolved organic carbon is monitored and (ii) biodegradability testing (CO2 evolution) in a defined minimal medium containing the test material as the sole carbon and energy source and a dilute bacterial inoculum obtained from the supernatant of homogenized activated sludge generated in the semicontinuous activated sludge system. Removal and biodegradation are measured using nonspecific methods, at initial concentrations of 5 to 10 mg of dissolved organic carbon per liter. Biodegradability data are accurately described by a nonlinear computer model which allows the rate and extent of biodegradation for different compounds to be compared and statistically examined. The evaluation of data generated in the combined removability-biodegradability system allows the biodegradation potential of a variety of xenobiotic organic chemicals to be estimated.  相似文献

The effect of the inoculum source on the digestion of linear alkylbenzene sulfonates (LAS) under anaerobic conditions has been investigated. The potential for primary and ultimate LAS biodegradation of anaerobic sludge samples obtained from wastewater treatment plants (WWTPs) of different geographical locations was studied applying a batch test system. It was found that only 4–22% of the LAS added to the batch anaerobic digesters was primarily transformed suggesting a poor primary degradation of the LAS molecule in anaerobic discontinuous systems. Regarding ultimate biodegradation, the addition of LAS to the batch anaerobic digesters caused a reduction on the extent of biogas production. Significant differences in the inhibition extent of the biogas production were observed (4–26%) depending on the sludge used as inoculum. Effect of the surfactant on the anaerobic microorganisms was correlated with its concentration in the aqueous phase. Sorption of LAS on anaerobic sludge affects its toxicity by depletion of the available fraction of the surfactant. LAS content on sludge was related to the total amount of calcium and magnesium extractable ions. The presence of divalent cations promote the association of LAS with anaerobic sludge reducing its bioavailability and the extent of its inhibitory effect on the biogas production.  相似文献

Several screening methods at the so-called ready biodegradability level are suitable to test poorly soluble substances. Typical for these tests is that mineralization is evaluated from monitoring oxygen uptake or carbon dioxide production. Unfortunately, they suffer from a rather low precision in the calculated percentage of mineralization caused by subtracting a too high inoculum control measurement from the response in the test system. Criteria for blank oxygen consumption, due to the metabolic activity of the inoculum, are proposed from which maximum amounts of activated sludge or secondary effluent per litre test medium can be derived to be used as an appropriate inoculum. Both for current and future standardized tests the precision of the method can be kept within acceptable margins. Inoculum material was sampled from 40 communal biological waste water treatment plants. Microbial aggregates of an aerobic granular sludge can be used for the treatment of industrial or municipal wastewater, but their formation from a microbial activated sludge requires several weeks. Therefore, the aim of this research was the selection of microbial cultures to shorten the granule-forming period from several weeks to a few days. An enrichment culture with the ability to accelerate granulation was obtained by repeating the selection and batch cultivation of fast-settling microbial aggregates isolated from the aerobic granular sludge. Bacterial cultures of Klebsiella pneumoniae strain B and Pseudomonas veronii strain F, with self-aggregation indexes of 65 and 51%, respectively, and a coaggregation index of 58%, were isolated from the enrichment culture. A mixture of these strains with the activated sludge was used as an inoculum in an experimental sequencing batch reactor to start up an aerobic granulation process. Aerobic granules with a mean diameter of 446±76 μm were formed in an experiment after 8 days of cultivation, but microbial granules were absent in controls. Nylon-6, a man-made polymer that finds its application in the manufacture of car tires, ropes, fabrics, automobile parts etc., is manufactured with ε-caprolactam. Waste water generated during production of nylon-6 contains the unreacted monomer. Owing to the polluting and toxic nature of ε-caprolactam, its removal from waste streams is necessary. Pseudomonas aeruginosa MCM B-407 was isolated from activated sludge used to treat waste from a factory producing nylon-6. This organism was able to remove ε-caprolactam with simultaneous reduction in chemical oxygen demand (COD). The degradation of ε-caprolactam in waste water was found to be optimal over a wide range of pH from 5.0 to 9.0, temperature of 30°C, and under shake or aerated conditions, with an inoculum density of 105 cells/ml and with an incubation period of 24 – 48 h. Thus, Pseudomonas aeruginosa MCM B-407 isolated from the activated sludge exposed to ε-caprolactam may play an important role in the bioremediation of ε-caprolactam from the waste waters of industries manufacturing nylon-6. The effectiveness of bioaugmentation in the improvement of the start-up of a biofilm airlift reactor to perform partial nitrification was investigated. Two identical biofilm airlift reactors were inoculated. The non-bioaugmented reactor (NB-reactor) was inoculated with conventional activated sludge, whereas the bioaugmented reactor (B-reactor) was seeded with the same conventional activated sludge but bioaugmented with nitrifying activated sludge from a pilot plant performing full nitritation under stable conditions (100% oxidation of influent ammonium to nitrite). The fraction of specialized nitrifying activated sludge in the inoculum of the B-reactor was only 6% (measured as dry matter). To simplify comparison of the results, operational parameters were equivalent for both reactors. Partial nitrification was achieved significantly faster in the B-reactor, showing a very stable operation. The results obtained by fluorescence in situ hybridization assays showed that the specialized nitrifying biomass added to the B-reactor remained in the biofilm throughout the start-up period.  相似文献

 The present work investigates 1-anthraquinone sulphonate (1-AS) biodegradation under (i) aerobic conditions using domestic activated sludge as inoculum, (ii) anaerobic conditions using sludge from an anaerobic domestic wastewater treatment digestor in a sulphate-containing or methanogenic environment, (iii) a combination of anaerobic followed by aerobic conditions. The process was evaluated in terms of primary degradation, i.e. 1-AS elimination and ultimate degradation, as total dissolved organic carbon removal. It was shown that aerobic conditions lead to the complete primary and ultimate degradation, of 1-AS. By contrast, neither under sulphato-reductive nor methanogenic conditions does anaerobic digestion lead to the significant degradation of 1-AS. The use of anaerobic treatment followed by aerobic treatment did not improve degradation. Indeed aerobic post-treatment resulted in the re-appearance of pollutant in the medium even though this had been partly degraded under anaerobic conditions. The use of activated sludge (AS) to assess the environmental impact of chemicals and wastewaters suffers from several drawbacks related to the heterogeneity, absence of standardization and health risk associated with such a mixed sewage population. To search for reliable testing inoculum alternatives, the potential of a standardized soil inoculum and a pure culture (Pseudomonas sp.), as reference material in a 5-day BOD test, was evaluated using the dilution and manometric methods and testing the standard glucose-glutamic acid solution. The results obtained for the BOD5 of the standard solution fall in the range referred for the AS in the standards of the cited methods, demonstrating the potential of these inocula as an alternative to AS in the BOD5 test. Moreover, testing on real wastewater samples showed highly significant correlations (P < 0.001) between the BOD5 values obtained with these two inocula and with the standard inoculum. Analysis of metabolic patterns also pointed to a similar catabolic profile between soil inoculum and AS and to the potential of a mixed Pseudomonas-soil inoculum as an AS surrogate culture.  相似文献

A method for quantifying bacterial populations introduced into an activated-sludge microbial community is described. The method involves extraction of DNA from activated sludge, appropriate dilution of the extracted DNA with DNA extracted from nonintroduced activated sludge, PCR amplification of a gyrB gene fragment from the introduced strain with a set of strain-specific primers, and quantification of the electrophoresed PCR product by densitometry. The adequacy of the method was examined by analyzing the population dynamics of two phenol-degrading bacteria, Pseudomonas putida BH and Comamonas sp. strain E6, that had been introduced into phenol-digesting activated sludge. The density of each of the two populations determined by the PCR method immediately after the introduction was consistent with the density estimated from a plate count of the inoculum. This quantitative PCR method revealed different population dynamics for the two strains in the activated sludge under different phenol-loading conditions. The behavior of both of these strains in the activated sludge reflected the growth kinetics of the strains determined in laboratory axenic cultures.  相似文献

The primary and ultimate biodegradability of phthalic acid, monobutyl phthalate, and five structurally diverse phthalic acid ester plasticizers in river water and activated sludge samples were determined via ultraviolet spectrophotometry, gas chromatography, and CO2 evolution. The compounds studied underwent rapid primary biodegradation in both unacclimated river water and acclimated activated sludge. When activated sludge acclimated to phthalic acid esters was used as the inoculum for the CO2 evolution procedure, greater than 85% of the total theoretical CO2 was evolved. These studies demonstrate that the phthalic acid ester plasticizers and intermediate degradation products readily undergo ultimate degradation in different mixed microbial systems at concentrations ranging from 1 to 83 mg/liter.  相似文献

Release of nitrogen and phosphorus into effluent of activated sludge process was reported in the recent researches of sludge reduction induced by oligochaetes. In this study, batch test and radioisotope 32P tracer test were therefore carried out to further investigate nutrients release and phosphorus distribution among supernatant, sludge and worm during predation of oligochaetes on sludge. Results showed that more nutrients release into supernatant occurred in the tests of worms with sterilized sludge than that of worms with activated sludge, and release of nitrogen and phosphorus was few in the tests of worms with activated sludge. Statistical analysis showed that no factor was significantly correlated with phosphorus concentration in the supernatant, but time and worm were only two factors significantly affecting total nitrogen (TN) concentration in the supernatant in the test of worms with activated sludge. Through test of radioisotope 32P tracer, 32P mainly distributed in activated sludge and supernatant, and more release of 32P into the supernatant occurred in the test of sludge with worms. After 24 h, 32P concentration of supernatant in the test of sludge with worms was 9% higher than that in the test of sludge without worms, and 32P concentration of worm increased by 2.7%. Additionally, the release rate of phosphorus into supernatant caused by worm's predation on activated sludge was 0.1211 mgTP/gWorm (wet weight) h.  相似文献

BIOLOG GN plates are increasingly used to characterize microbial communities by determining the ability of the communities to oxidize various carbon sources. Studies were done to determine whether the BIOLOG GN plate assay accurately reflects the catabolic potential of the inoculum used. To gain insight into which populations of microbial communities contribute to the BIOLOG patterns, denaturing gradient gel electrophoresis and temperature gradient gel electrophoresis (TGGE) were used to assess the diversity of ribotypes in the inocula and individual wells of BIOLOG plates following incubation. These studies were done with microbial communities from the rhizosphere of potatoes and an activated sludge reactor fed with glucose and peptone. TGGE analyses of BIOLOG wells inoculated with cell suspensions from the potato rhizosphere revealed that, compared with the inoculum, there was a decrease in the number of 16S rRNA gene fragments obtained from various wells, as well as a concomitant loss of populations that had been numerically dominant in the inoculum. The dominant fragments in TGGE gels could be assigned to the gamma subclass of the class Proteobacteria, suggesting that fast-growing bacteria adapted to high substrate concentrations were numerically dominant in the wells and may have been primarily responsible for the patterns of substrate use that were observed. Similarly, the community structure changed in wells inoculated with cells from activated sludge; one or more populations were enriched, but all dominant populations of the inoculum could be detected in at least one well. This study showed that carbon source utilization profiles obtained with BIOLOG GN plates do not necessarily reflect the functional potential of the numerically dominant members of the microbial community used as the inoculum.  相似文献

Salmonella destruction efficiencies of 99% were obtained after 10 h aeration at 15°C in a laboratory model of the activated sludge process. This study demonstrated that, in a batch process, the removal of salmonellas occurred in three phases. (i) By 4 h, 90% of the original inoculum had disappeared from the activated sludge, probably due mainly to predation by ciliated protozoa. The remaining 10% was distributed between the liquid phase (approximately 90%) and the sludge floc (approximately 10%). (ii) During the next 2 h this situation was inverted so that, by 6 h, more than 80% of the remaining salmonellas were then adsorbed to floc, leaving less than 20% in liquid suspension. (iii) From 6 h onwards there was a much slower decline of the remaining salmonellas attached to floc. The addition of dioctyl sodium sulphosuccinate after 4 h, inactivated the ciliated protozoa populations and completely eliminated the continued reduction of salmonellas from activated sludge observed previously.  相似文献

The effect of cationic polymer additives on biomass granulation and COD removal efficiency had been examined in lab-scale upflow anaerobic sludge blanket (UASB) reactors, treating low strength synthetic wastewater (COD 300-630 mg/l). Under identical conditions, two reactors were operated with and without polymer additives in inoculum under four different organic loading rates (OLRs). The optimum polymer dose was adopted based upon the results of jar test and settling test carried out with inoculum seed sludge. With the use of thick inoculum, SS greater than 110 g/l and VSS/SS ratio less than 0.3, granulation was observed in UASB reactor treating synthetic wastewater as well as actual sewage, when OLR was greater than 1.0 kg COD/m(3) d. Polymer additive with such thick inoculum was observed to deteriorate percentage granules and COD removal efficiency compared to inoculum without polymer additives. At OLR less than 1.0 kg COD/m(3) d, proper granulation could not be achieved in both the reactors inoculated with and without polymer additive. Also, under this low loading, drastic reduction in COD removal efficiency was observed with polymer additives in inoculum. Hence, it is rational to conclude that biomass granulation for treatment of low strength biodegradable wastewater depends on the applied loading rate and selection of thick inoculum sludge.  相似文献

Aerobic biodegradation of tripropylene glycol (PG3) was investigated under the conditions of the OECD screening test 301E and the Continuous Flow Activated Sludge Simulation test (CFAS). A modified two-chamber facility with a denitrification stage was used for the CFAS test. Primary PG3 biodegradation was measured by the HPLC with fluorimetric detection and analyte derivatisation. Metabolites were identified by LC-MS with electrospray ionisation and GC-MS with electron impact ionisation, as well as semiquantitatively determined by the LC-MS technique. PG3 was found to be inherently biodegradable and it exhibits a strong poisonous effect on activated sludge after exceeding the threshold concentration (10 mg l−1). Metabolite accumulation onto the activated sludge is probably responsible for this poisonous effect. Probable biotransformation products of tripropylene glycol under the aerobic conditions include metabolites with a single terminal aldehyde or a ketone group and metabolites with two terminal aldehyde or ketone groups. Their concentration rises at the end of the OECD screening test.  相似文献

Bioaugmentation of activated sludge systems with specialised bacterial strains could be a powerful tool to improve several aspects in wastewater treatment processes, such as improved flocculation and degradation of recalcitrant compounds. This review focuses on the addition of strains to activated sludge to enhance the biodegradation of recalcitrant compounds, either through the activity of the inoculated strain or after transfer of degradative plasmids to activated sludge bacteria. Different factors that improve the aggregation of the sludge flocs and their influence on biodegradation are described. This review further deals with the role of bacterial plasmids in natural genetic exchange between inoculated and indigenous sludge bacteria, and in the construction of new genetically modified organisms. The few successful cases of bioaugmentation described in this review, together with future research, must lead to a better understanding of sludge bioaugmentation. Summary In this study the gas chromatographic method of Poly-β-hydroxy butyric acid determination of Braunegg et al. (1978), was optimised for activated sludge samples. The poly-β-hydroxy butyric acid was extracted and quantified gravimetrically to confirm the accuracy of the gas chromatographic method. The authenticity of the extracted material was confirmed by several methods. It was also confirmed that the mixed liquor of an activated sludge process did not interfere with the esterification. The sample size required was 25 ml of mix liquor, or 50 mg of freeze-dried sludge.  相似文献

The process of nitrification–denitrification via nitrite for nitrogen removal under real-time control mode was tested in two laboratory-scale sequencing batch reactors (SBRs) with flocculent activated sludge (R1) and aerobic granular sludge (R2) to compare operational performance and real-time control strategies. The results showed that the average ammonia nitrogen, total inorganic nitrogen (TIN), and chemical oxygen demand (COD) removal during aeration phase were 97.6%, 57.0%, and 90.1% in R2 compared with 98.6%, 48.7%, and 88.1% in R1. The TIN removed in both SBRs was partially due to the presence of simultaneous nitrification–denitrification via nitrite, especially in R2. The specific nitrification and denitrification rates in R2 were 0.0416 mgNH4+–N/gSS-min and 0.1889 mgNOX–N/gSS-min, which were 1.48 times and 1.35 times that of R1. The higher rates for COD removal, nitrification, and denitrification were achieved in R2 than R1 with similar influent quality. Dissolved oxygen (DO), pH, and oxidization reduction potential, corresponding to nutrient variations, were used as diagnostic parameters to control the organic carbon degradation and nitrification–denitrification via nitrite processes in both SBRs. The online control strategy of granular SBR was similar to that of the SBR with flocculent activated sludge. However, a unique U-type pattern on the DO curve in granular SBR was different from SBR with flocculent activated sludge in aerobic phase.  相似文献

The use of a granular inoculum prevented acidification during the anaerobic batch biodegradation of a kitchen waste for waste/inoculum ratios in the range of 0.5–2.3 g VS/g VS, when the alkalinity/COD ratio was 37 mg NaHCO3/g COD. In similar experiments but using a suspended sludge with a significantly lower activity, the methane production rates and the biodegradability were significantly lower and the pH decreased below 5.5 at the waste/inoculum ratio of 2.3 g VS/g VS. When the added alkalinity was decreased to 2 mg NaHCO3/g COD, the ratio waste/inoculum was clearly more important than the inoculum activity, since, irrespective of the sludge used, acidification occurred at waste/inoculum ratios higher than 0.5 g VS/g VS. The advantage of using granular sludge was further investigated in order to define reasonable condition of waste/inoculum ratio and added alkalinity that could be applied in practice. For a waste/inoculum ratio of 1.35, there were no significant differences between the results obtained for the biodegradability and maximum methane production rate (MMPR), when the alkalinity decreased from 44 to 22 mg NaHCO3/g COD.  相似文献
