Pollen of some species of the genus Quercus shows rod-shaped substructures in fresh or acetolysed exines, while in other species rod substructure is mostly masked by sporopollenin. Oxidation with potassium permanganate removes exine substance (sporopollenin) from between the rod substructures. We propose that the rods include receptors for sporopollenin. The sporopollenin between rods we refer to as ‘receptor-independent sporopollenin’. Pollen of Typha, when mature, has tectal surfaces with concave tops and sides, whereas during development the tectal surfaces are smoothly rounded. After acetolysis treatment followed by potassium permanganate the tectum surfaces again appear rounded. When these exines are subsequently eroded by a fast atom source, rod-shaped substructures are seen to protrude from the tectum. These structures are equivalent in size and shape to the rods of the exine of Quercus. Sporopollenin that accumulates over and masks rod substrucutre is less resistant to our degradative methods than the sporopollenin in rod structures of exines. We suggest that the exine material we call “receptor-independent sporopollenin” be given a simple positive name, such as masking-sporopollenin or abbreviated to masking-spn.  相似文献

Sporopollenin obtained from wings of Pinus mugo (Turra) pollen was analysed by pyrolysis mass spectrometry. In the spectrum, mass peaks which are characteristic for p-coumaric acid were dominant. p-Coumaric acid was the main degradation compound when the wing material was treated by a gentle method using AII3, and also when the remaining residue of the treated sporopollenin material was saponified. It is therefore assumed that p-coumaric acid is a genuine structural unit in the sporopollenin skeleton. We report the isolation of purified sporopollenin from pollen grains of different species and its complete solubilization. Exine from Pinus pinaster, Betula alba, Ambrosia elatior and Capsicum annuum was extracted by treatment with hydrogen fluoride in pyridine. These exines were purified from their aromatic moieties and from fatty acids linked by ester bonds using acidolysis and saponification treatments. The biopolymer obtained retains almost completely the shape of the original pollen grain. Fourier-transform infrared spectroscopy analysis of the isolated sporopollenin showed the absence of polysaccharide and phenolic material and the presence of carboxylic acid groups joined to unsaturations and ether linkages. Sporopollenin samples were successfully degraded by exhaustive 24-h ozonolysis at room temperature. Gentle ozonolysis (3 h at 0°C) did not completely degrade the biopolymer. The compounds obtained after exhaustive ozonolysis were analysed by gas chromatography-mass spectrometry. Dicarboxylic acids with a low number of carbon atoms were identified as major components of sporopollenin from P. pinaster, A. elatior and C. annuum, representing 28.8%, 63.2% and 88.5%, respectively, of the total compounds obtained. Fatty acids and n-alkanes also were identified in P. pinaster, A. elatior and B. alba sporopollenin. The extremely chemically resistant component of the cell wall of spores, pollens, and some microorganisms, sporopollenin, is generally accepted to be derived from carotenoids or carotenoid esters. However, we report here that 13C NMR analyses of sporopollenin from several sources shows that this widely held view is incorrect, with one possible exception. Sporopollenin is not a unique substance but rather a series of related biopolymers derived from largely saturated precursors such as fatty acids. The biopolymers contain widely varying amounts of oxygen in the form of ether, hydroxyl, carboxylic acid, ester, and ketone groups.  相似文献

Sporopollenin is a natural polymer obtained from Lycopodium clavatum, which is highly stable with constant chemical structure and has high resistant capacity to chemical attack. In this study, immobilization of lipase from Candida rugosa (CRL) on sporopollenin by adsorption method is reported for the first time. Besides this, the enzyme adsorption capacity, activity and thermal stability of immobilized enzyme have also been investigated. It has been observed that under the optimum conditions (Spo-E(0.3)), the specific activity of the immobilized lipase on the sporopollenin by adsorption was 16.3 U/mg protein, which is 0.46 times less than that of the free lipase (35.6 U/mg protein). The pH and temperature of immobilized enzyme were optimized, which were 6.0 and 40 °C respectively. Kinetic parameters Vmax and Km were also determined for the immobilized lipase. It was observed that there is an increase of the Km value (7.54 mM) and a decrease of the Vmax value (145.0 U/mg-protein) comparing with that of the free lipase.  相似文献

The sporopollenin of pollen exines of Ambrosia trifida is soluble in fused potassium hydroxide, in strong oxidizing solutions, and in certain organic bases. It is insoluble in other organic and inorganic acids and bases, in lipid solvents, and in detergents. The outer exine layer of gymnosperm and angiosperm pollen dissolves in 2-aminoethanol. The inner exine layer, as well as the exine of pteridophyte spores, is insoluble. The exine dissolution process in 2-aminoethanol involves swelling and disintegration of exine structures, leaving some residual globules. Sporopollenin shares some solubility properties with lignin and cutin but appears to be chemically distinct from these substances.  相似文献

Sporopollenin can be dissolved in water by oxidative degradation with using potassium permanganate (KMnO4). in mature pollen grain ofMagnolia grandiflora andHibiscus syriacus some organic compounds were extracted from the aqueous phase into hexane, and analyzed by using gas chromatography-mass spectrometry (GC-MS). The analysis shows that the extracted compounds Include some organosilicon compounds. By researching of mass spectra data base, the organosillicon compounds are suggested as 1, 1, 1, 5, 7, 7, 7-heptamethyl-3,3-bis(trimethylsilox))tetrasiloxane and 1, 1, 1, 3, 5, 7, 7, 7-octamethyl-3, 3- bis(trimethylsiloxy)tetrasiloxane. 1,2-benzendicarboxylic acid butyl 2-ethylhexyl ester is also suggested by data base research in the extracted compounds. The present study implies that silicon (Si) is considered to be located in inner part of three dimensional structure of sporopollenin.  相似文献

Sporopollenin is a natural polymer obtained from Lycopodium clavatum, which is highly stable with constant chemical structure and has high resistant capacity to chemical attack. In this study, the Candida rugosa lipase (CRL) was encapsulated within a chemically inert sol–gel support prepared by polycondensation with tetraethoxysilane (TEOS) and octyltriethoxysilane (OTES) in the presence and absence of sporopollenin and activated sporopollenin as additive. The catalytic properties of the immobilized lipases were evaluated into model reactions, i.e. the hydrolysis of p-nitrophenylpalmitate (p-NPP), and the enantioselective hydrolysis of rasemic Naproxen methyl ester that was studied in aqueous buffer solution/isooctane reaction system. The results indicated that the sporopollenin based encapsulated lipase particularly had higher conversion and enantioselectivity compared to the sol–gel free lipase. In this study, excellent enantioselectivity (E > 400) has been noticed for most lipase preparations (E = 166 for the free enzyme) with an ee value ~98% for S-Naproxen. Moreover, (S)-Naproxen was recovered from the reaction mixture with 98% optical purity.  相似文献

This study aims to find likely sources of Ambrosia pollen recorded during 2007 at five pollen-monitoring sites in central Europe: Novi Sad, Ruma, Negotin and Nis (Serbia) and Skopje (Macedonia). Ambrosia plants start flowering early in the morning and so Ambrosia pollen grains recorded during the day are likely to be from a local source. Conversely, Ambrosia pollen grains recorded at night or very early in the morning may have arrived via long-range transport. Ambrosia pollen counts were analysed in an attempt to find possible sources of the pollen and to identify Ambrosia pollen episodes suitable for further investigation using back-trajectory analysis. Diurnal variations and the magnitude of Ambrosia pollen counts during the 2007 Ambrosia pollen season showed that Novi Sad and Ruma (Pannonian Plain) and to a lesser degree Negotin (Balkans) were located near to sources of Ambrosia pollen. Mean bi-hourly Ambrosia pollen concentrations peaked during the middle of the day, and concentrations at these sites were notably higher than at Nis and Skopje. Three episodes were selected for further analysis using back-trajectory analysis. Back-trajectories showed that air masses brought Ambrosia pollen from the north to Nis and, on one occasion, to Skopje (Balkans) during the night and early morning after passing to the east of Novi Sad and Ruma during the previous day. The results of this study identified the southern part of the Pannonian Plain around Novi Sad and Ruma as being a potential source region for Ambrosia pollen recorded at Nis and Skopje in the Balkans.  相似文献

The pollen grains of Ambrosia spp. are considered to be important aeroallergens in parts of southern and central Europe. Back-trajectories have been analysed with the aim of finding the likely sources of Ambrosia pollen grains that arrived at Poznań (Poland). Temporal variations in Ambrosia pollen at Poznań from 1995–2005 were examined in order to identify Ambrosia pollen episodes suitable for further investigation using back-trajectory analysis. The trajectories were calculated using the transport model within the Lagrangian air pollution model, ACDEP (Atmospheric Chemistry and Deposition). Analysis identified two separate populations in Ambrosia pollen episodes, those that peaked in the early morning between 4 a.m. and 8 a.m., and those that peaked in the afternoon between 2 p.m. and 6 p.m.. Six Ambrosia pollen episodes between 2001 and 2005 were examined using back-trajectory analysis. The results showed that Ambrosia pollen episodes that peaked in the early morning usually arrived at Poznań from a southerly direction after passing over southern Poland, the Czech Republic, Slovakia and Hungary, whereas air masses that brought Ambrosia pollen to Poznań during the afternoon arrived from a more easterly direction and predominantly stayed within the borders of Poland. Back-trajectory analysis has shown that there is a possibility that long-range transport brings Ambrosia pollen to Poznań from southern Poland, the Czech Republic, Slovakia and Hungary. There is also a likelihood that Ambrosia is present in Poland, as shown by the arrival of pollen during the afternoon that originated primarily from within the country.  相似文献

The basis for the resistance of certain algae to microbial decomposition in natural waters was investigated using Pediastrum duplex, Staurastrum sp., and Fischerella muscicola as test organisms. Enzyme preparations previously found to convert susceptible algae into spheroplasts had no such effect on the resistant species, although glucose and galacturonic acid were released from P. duplex walls. Little protein or lipid but considerable carbohydrate was found in the walls of the refractory organisms, but resistance was not correlated with the presence of a unique sugar monomer. A substance present in Staurastrum sp. walls was characterized as lignin or lignin-like on the basis of its extraction characteristics, infrared spectrum, pyrolysis pattern, and content of an aromatic building block. Sporopollenin was found in P. duplex, and cellulose in Staurastrum sp. Cell walls of the algae were fractionated, and the fractions least susceptible to microbial degradation were the sporopollenin of P. duplex, the polyaromatic component of Staurastrum sp., and two F. muscicola fractions containing several sugar monomers. The sporopollenin content of P. duplex, the content of lignin or a related constituent of Staurastrum sp., and the resistance of the algae to microbial attack increased with age. It is suggested that resistance results from the presence of sporopollenin in P. duplex, a lignin-like material in Staurastrum sp., and possibly heteropolysaccharides in F. muscicola.  相似文献

Question: We present a study of positive and negative interactions between the invasive grass Schismus barbatus (Poaceae) and Ambrosia dumosa (Asteraceae). Ambrosia facilitates seedling establishment, and such facilitation may accelerate invasion of exotic species, which, in turn, may reduce establishment of native plants. Location: Joshua Tree National Park, California, USA. Methods: During 2003‐2004, we used field surveys to characterize the natural spatial distribution of Schismus in relation to native shrubs, and experimentally manipulated seed rain of Ambrosia and Schismus at three distances from adult Ambrosia canopies. We measured percentage germination and individual performance of both species. Field data were complemented by a greenhouse experiment that measured competition between Ambrosia seedlings and Schismus planted at three densities and five relative abundances under controlled conditions. Results: Field surveys showed that the density of Schismus is independent of Ambrosia shrubs, but growth is enhanced near shrub canopies. In our field experiment Schismus is facilitated by adult Ambrosia. Under controlled conditions, Schismus does not respond to the density of Ambrosia seedlings, but changes in density of Schismus decreased performance of Ambrosia seedlings. Conclusion: Schismus invasion may be detrimental to native perennial plant populations. Although a reduction of seedling establishment is not usually expected to slow population growth of long‐lived perennials, recent unprecedented adult mortality in this community, and the well‐documented facilitative role of Ambrosia, suggest that Schismus invasion may be of high ecological significance.  相似文献

The aerobiological study of the Ambrosia pollen in Sofia, Bulgaria began in 1991. The quantitative dynamics of Ambrosia pollen grains during the last seven years was determined by Chebishev's method of parabolic interpolation with orthogonal polynoms. Thesensitization to ragweed pollen was investigated in random groups of patients with clinical manifestation of hay fever.This investigation proves the previouslyobserved tendency of an increase in thespreading of Ambrosia pollen in Bulgaria.  相似文献

The pollen antigens of various Ambrosia and related species were studied to learn whether substances closely related to antigen E (the major allergen of Ambrosia artemisiifolia) were present. After conventional immunoelectrophoresis, pollen extracts from six Ambrosia species each produced at least one pronounced precipitin line with antiserum for purified antigen E. Electrophoretic mobility was the same for several species (A. artemisiifolia, A. bidentata, A. psilostachya, and A. trifida) but was relatively lower for A. acanthicarpa and A. ambrosioides. Precipitin rings were also produced when pollen extracts of the various Ambrosia species were subjected to radial immunodiffusion in agarose which contained antiserum for purified antigen E. There was great variation among the Ambrosia species with respect to precipitin ring diameters. The variation may be due to differences among species in content of the antigen E-like substances or to altered interaction with the immobilized antibody. Crossed (2-dimensional) immunoelectrophoresis was shown to be useful for characterizing Ambrosia pollen antigens. Pollen extracts from A. artemisiifolia produced eight pronounced precipitin bands and at least eight faint, relatively fast-moving bands after crossed immunoelectrophoresis with antiserum against a whole pollen extract from the same species. Sporopollenin from the pollen of Typha angustifolia L. was exposed to a series of 36 subsequent acidic methanolysis procedures. The remaining decomposition products were investigated using several spectroscopic methods including Fourier transform infrared spectroscopy (FT-IR), solid state 13C nuclear magnetic resonance spectroscopy (13C-CPMAS-NMR) and X-ray photoelectron spectrometry (XPS). Substantial weight losses of the sporopollenin material occur after each acidic methanolysis step, while FT-IR and 13C-CPMAS-NMR spectra display no noticeable differences after 12, 24 and 36 steps. These findings are interpreted as a hint that the sporopollenin polymer has a uniform composition, i.e. relatively small monomer moieties of similar primary structure are present. Moreover, the weight losses account for the presence of substantial amounts of ether linkages in the sporopollenin polymer.  相似文献

The relationship ofAmbrosia (ragweed) andFranseria has long been debated. Their treatment as separate genera has been repeatedly challenged. In this study, chloroplast DNA restriction site variation was examined for species from bothAmbrosia andFranseria as well as taxa from the closely related genusHymenoclea. The chloroplast genomes of members of these three genera were examined using 21 restriction endonucleases and the restriction mutations were used to construct phylogenetic trees. Wagner and Dollo parsimony as well as weighted parsimony were employed to compare the different phylogenies. The results support a close relationship betweenAmbrosia andFranseria, but indicate that the two groups are well separated. Compared toFranseria, Ambrosia is a much more strongly supported group, and the results indicate thatHymenoclea is closer toFranseria than toAmbrosia. The cpDNA phylogeny was used as a framework to examine evolutionary trends in morphology and secondary chemistry.  相似文献

The natural host of Ophraella notulata is Iva frutescens (Asteraceae); its close relative feeds on a related plant, Ambrosia artemisiifolia. We reared beetles on both plants, obtained progeny from the four possible crosses (two sexes X two parental hosts), and reared the progeny on both plant species. Survival to the imaginal stage of progeny reared on Iva varied with both maternal and paternal host. Hatchling feeding response to both plants showed a maternal host X paternal host interaction. Consumption of Ambrosia by adult beetles was, counter to expectation, higher for progeny of Iva-reared males than Ambrosia-reared males. Oviposition response, although based on too few data to be definitive, was peculiar: parental host did not affect oviposition on Ambrosia; on Iva daughters of Iva-reared males laid significantly more eggs than did daughters of Ambrosia-reared males, but only if they had been reared on Iva; those reared on Ambrosia displayed the reverse pattern. We discuss the possibility that nongenetic paternal transmission of host plant effects may explain these results, but offer a somewhat uncomfortable hypothesis of selection as a preferable explanation. An important outcome of the experiment is that it provided no evidence of maternal effects of host plant on offspring feeding or oviposition.  相似文献

Summary In order to identify factors necessary for the establishment of the reticulate pollen wall pattern, we have characterized a T-DNA tagged mutant ofArabidopsis thaliana that is defective in pattern formation. This study reports the results of an ultrastructural comparison of pollen wall formation in the mutant to wall development in wild-type plants. Pollen wall development in the mutant parallels that of wild-type until the early tetrad stage. At this point in wild-type plants, the microspore plasma membrane assumes a regular pattern of ridges and valleys. Initial sporopollenin deposition occurs on the ridges marking the beginning of probacula formation. In contrast, the plasma membrane in the mutant appears irregular with flattened protuberances and rare invaginations. As a result, the wild-type regular pattern of ridges and valleys is not formed. Sporopollenin is randomly deposited on the plasma membrane and aggregates on the locule wall; it is not anchored to the membrane. Our finding that the mutation blocks the normal invagination of the plasma membrane and disrupts the proper deposition of sporopollenin during wall formation suggests that the mutation could be in a gene responsible for pattern formation. These results also provide direct evidence that the plasma membrane plays a critical role in the establishment of the pollen wall pattern.  相似文献

Understanding the causes of interannual variability in atmospheric pollen concentration is an important but elusive goal for public health and environmental change. We analyzed long-term daily records of pollen counts from urban Kansas City, Missouri, USA collected from 1997 to 2009 for three pollen groups: Ambrosia, Poaceae, and a third group which is mostly composed of arboreal pollen types. The annual pollen index varied from 8,368 to 80,822 over the thirteen-year period. Although Ambrosia pollen is often thought to be associated with droughts and disturbance, years with high Ambrosia pollen were associated with high summer precipitation to the south of Kansas City. Years with high Poaceae pollen were associated with high spring precipitation to the south of the city. In support of the southern influence to Kansas City pollen, Ambrosia and Poaceae pollen mostly arrived on southern winds. In contrast to the other two pollen groups, the arboreal pollen was most associated with growing season precipitation to the east of Kansas City, although it was still highest on days with southern winds. Based on the correlations with climate, the severity of an upcoming allergy season may be predicted with early-season precipitation data, while short-term severity can be forecast from local weather patterns.  相似文献

Summary Extensive tracer experiments were carried out on Tulipa with the aim of determining the structure and biosynthesis of sporopollenin. The radiolabeled precursors were applied using an improved technique previously selected. The sporopollenin fraction was purified using either a gentle method — hydrolyzing enzymes (pronase, amylase, amyloglucosidase, cellulase, pectinase and lipase) and alkaline hydrolysis (method A) — or by a conventional aggressive procedure, where the material was enriched by alkaline hydrolysis and treated several days with 80% phosphoric acid (method B). The 14C-labeled precursors applied were mevalonate, glucose, acetate, malonic acid, phenylalanine, tyrosine, p-coumaric acid. Regardless of the method of enrichment, a higher level of incorporation into the sporopollenin fraction was always seen with [U-14C]-phenylalanine. The level of radioactivity found in sporopollenin labeled by phenylalanine or malonate was sufficiently high for the labeled polymer to be degraded and the products released analyzed for the first time. In the case of phenylalanine-labeled sporopollenin, the main degradation component, p-hydroxybenzoic acid, was also the most heavily labeled substance. This result was not dependent on the procedure used for sporopollenin enrichment. These findings are interpreted as meaning that phenylpropane metabolism via phenylalanine-ammonia lyase is involved in sporopollenin biosynthesis.  相似文献
