Research on the comparative morphology of pollen grains depends crucially on the application of appropriate microscopy techniques. Information on the performance of microscopy techniques can be used to inform that choice. We compared the ability of several microscopy techniques to provide information on the shape and surface texture of three pollen types with differing morphologies. These techniques are: widefield, apotome, confocal and two-photon microscopy (reflected light techniques), and brightfield and differential interference contrast microscopy (DIC) (transmitted light techniques). We also provide a first view of pollen using super-resolution microscopy. The three pollen types used to contrast the performance of each technique are: Croton hirtus (Euphorbiaceae), Mabea occidentalis (Euphorbiaceae) and Agropyron repens (Poaceae). No single microscopy technique provided an adequate picture of both the shape and surface texture of any of the three pollen types investigated here. The wavelength of incident light, photon-collection ability of the optical technique, signal-to-noise ratio, and the thickness and light absorption characteristics of the exine profoundly affect the recovery of morphological information by a given optical microscopy technique. Reflected light techniques, particularly confocal and two-photon microscopy, best capture pollen shape but provide limited information on very fine surface texture. In contrast, transmitted light techniques, particularly differential interference contrast microscopy, can resolve very fine surface texture but provide limited information on shape. Texture comprising sculptural elements that are spaced near the diffraction limit of light (~250 nm; NDL) presents an acute challenge to optical microscopy. Super-resolution structured illumination microscopy provides data on the NDL texture of A. repens that is more comparable to textural data from scanning electron microscopy than any other optical microscopy technique investigated here. Maximizing the recovery of morphological information from pollen grains should lead to more robust classifications, and an increase in the taxonomic precision with which ancient vegetation can be reconstructed.  相似文献

The pollen morphology of twenty‐five Chinese species belonging to two sections of Glochidion and one species from Phyllanthus subgen. Phyllanthodendron was examined using scanning electron microscopy (SEM). The results showed that pollen grains of Glochidion are isopolar, spheroidal to rarely subprolate in shape, (3)4–5(6)‐colporate in aperture pattern, and tectate‐reticulate to tectate‐rugulate in exine ornamentation. Fusion of colpus ends was observed in some pollen grains of G. pseudo‐obscurum and G. wrightii. The pollen grains of Phyllanthus dunnianus are similar to those of Glochidion species in their spheroidal shape and four‐colporate aperture pattern, but are slightly larger in size, and show tectate‐perforate exine ornamentation. In Glochidion, pollen morphology appears to be useful for distinguishing several macromorphologically similar. Additionally, palynological characters provide evidence for the taxonomy of the supergenus Phyllanthus s.l.  相似文献

Scanning and transmission electron microscopy of pollen grains of 29 species, representing the ten extant genera of Cycadales, has provided valuable insight into their relationships. Pollen grains of these taxa are boat-shaped, monosulcate, and bilaterally symmetrical. They range from narrowly to widely elliptical or subcircular when viewed distally, and have an exine surface of psilate, foveolate, or fossulate. Pollen wall ultrastructure of Cycadales is typically tectate with alveolate∗∗∗spongy exine. The nexine is laminated in all genera. Nexine 1 (footlayer) is present in most species as a thin and often discontinuous layer. There is consistent variation in thickness of the sporoderm layers among the genera but relative uniformity within them. Pollen characteristics are well correlated with macro- and micromorphological features, chromosome numbers, geographical distribution, and postulated pollination mode. A close affinity between Encephalartos, Lepidozamia, and Macrozamia is recognized. Pollen characteristics of the genus Bowenia show some similarity with those of the latter group. Except for two species of Macrozamia which are narrowly elliptic, all of the genera have widely elliptic pollen and share a psilate exine surface and the thinnest sexine with nearly identical arrangement of alveoli. Pollen grains of the species in the genus Dioon exhibit a unique morphology but are more similar to Stangeria than they are to those of taxa in Zamiaceae. The circular outline of the grains and the foveolate exine surface are characters shared by these two genera, but several morphological features distinguish Dioon from Stangeria. Ceratozamia and Zamia share a widely elliptic shape, foveolate exine surface and nearly identical sexine, as well as morphological features and chromosome numbers. They differ from Microcycas in sexine thickness, gross morphology and chromosome numbers. The pollen grains of Cycas circinalis and C. revoluta differ in size and structure of the sexine from all other genera and from each other, substantiating their distinct subgeneric delimitations.  相似文献

The general morphology, surface sculpturing, and exine ultrastructure have been studied in dispersed monosulcate pollen from the Early Cretaceous of Transbaikalia, Russia. The pollen grains dominate the palynological assemblage extracted from coal deposits of the Khilok Formation in the Buryat Republic, which also contain ginkgoalean leaves of Baierella averianovii as the only constituent of the assemblage of plant megafossils. The relationship between the pollen grains and ginkgoalean leaves from this autochthonous burial is hypothesised on the basis of taphonomical analysis and palaeobiogeographical data. It is shown that the ectexine of the pollen grains includes a thick solid tectum, a thin granular infratectum and a thin foot layer; the endexine is fine-grained, slightly more electron-dense than the ectexine, and is preserved only in places. The distal aperture is formed by a thinning of the exine. No analogous ultrastructure has been described so far in fossil pollen grains of this morphotype studied ultrastructurally from in situ material. For comparison, we also studied the exine ultrastructure of pollen grains Ginkgo biloba. The fossil pollen is not identical to pollen of extant G. biloba, but shows several significant similarities in the exine ultrastructure, which does not contradict the presumable ginkgoalean affinity of the fossil pollen.  相似文献

A morphological study of dispersed Circumpolles pollen grains from the Upper Triassic of the Southern Alps has been initiated with the genus Duplicisporites. Individual pollen grains were studied by means of scanning electron microscopy (SEM) and transmission electron microscopy (TEM). Seen with SEM the pollen surface is finely verrucate with low verrucae of different sizes. A sub‐equatorial continuous rimula is clearly visible. The proximal trilete scar is small and indistinct. TEM images reveal a bi‐layered exine. The ectexine is formed by numerous small, closely packed, granulae subdivided by irregularly‐spaced cavities. In the region of the subequatorial canal, the ectexine becomes thinner, about 1/3 of the usual thickness. At places, the ectexine is slightly separated from the underlying endexine. The endexine is prominent and significantly darker than the ectexine. It is homogeneous and of constant thickness. On the basis of its older age, with respect to Classopollis, the present ultrastructural dataset provides information on the possible origin of cheirolepidiaceous‐type morphology.  相似文献

The pollen morphology and tapetal membranes of 17 species of Trillium from North America have been examined by scanning and transmission electron microscopy. The pollen grains of the genus are consistently spherical and inaperturate. The exine is unstratified and ultra-thin. Five types of exine structure are recognized, namely, echinate-, corrugate-, granulate-, verrucate- and spinulate-type, based on the variations in the components of the exine. The granulate-type is the most common and is divided into six subtypes. The features of tapetal membranes also vary in relation with the variation of the exine structures. This study demonstrates the potential of palynological evidence in assessment of taxonomic relationships within the genus.  相似文献

The pollen morphology of 11 genera and 11 species of the Hydrocharitaceae and one species of the Najadaceae was studied using scanning and transmission electron microscopies, and the exine structures and sculptures are discussed in relation to pollination mechanisms and the molecular phylogeny. The pollen grains of the Hydrocharitaceae are spherical, inaperturate, and form monads or tetrads, while those of the Najadaceae are elliptical, inaperturate, and form monads. The entomophilous genera Egeria, Blyxa, Ottelia, Stratiotes, and Hydrocharis share pollen grains that have projections like spines or bacula. The anemophilous genus Limnobium has reticulate pollen grains. The hypohydrophilous genera Thalassia and Najas are characterized by pollen grains with reduced exine structures. The pollen-epihydrophilous genera Elodea and Hydrilla have tightly arranged small spinous pollen grains, and the male flower-epihydrophilous genera Enhalus and Vallisneria have reduced reticulate or gemmate exines. Character state reconstruction of the exine structures and sculptures using a molecular phylogenetic tree suggests that variation in the exine is generally correlated with the pollination mechanism; the selective pressures acting on the pollination mechanisms have reduced the exine structure in hypohydrophilous plants and resulted in various exine sculptures that are adapted to the different pollination mechanisms in entomophilous, anemophilous, and pollen-epihydrophilous plants.  相似文献

Background and Aims

Hydration, rupture and exine opening due to the sudden and large expansion of intine are typical of taxoid-type pollen grains. A hemispheric outgrowth external to the exine was observed on Cupressus and Juniperus pollen grains before the intine swelling and exine release. However, the actual existence of this permanent or temporary structure and its precise role in pollen hydration is still being debated. The aim of this paper is to collect information on the actual presence of this peculiar outgrowth on the surface of the Cupressus pollen grain, its structure, composition and function.

Methods

Pollen grains of several Cupressus species were observed using various techniques and methodologies, under light and fluorescence microscopy, phase-contrast microscopy, confocal microscopy, scanning electron microscopy, and an environmental scanning electron microscope. Observations were also performed on other species with taxoid-type pollen grains.

Key Results

A temporary structure located just above the pore was observed on Cupressus pollen grains, as well as on other taxoid-type pollens. It is hemispheric, layered, and consists of polysaccharides and proteins. The latter are confined to its inner part. Its presence seems to regulate the entrance of water into the grains at the beginning of pollen hydration.

Conclusions

The presence of a temporary structure over the pore of taxoid-type pollen grains was confirmed and its structure was resolved using several stains and observation techniques. This structure plays a role in the first phases of pollen hydration.  相似文献

Pollen grains of 30, mainly annual, species from 134 populations of the genus Veronica (Plantaginaceae, formerly Scrophulariaceae) from the Mediterranean Region, have been studied with light and scanning electron microscopy. Three pollen types are defined based on pollen size and exine surface sculpture. In some cases within the study group pollen size can be useful in species determination. Hypothetical relationships of pollen grain size and aperture number with ploidy level, style length and corolla diameter are discussed. The ornamentation of the exine in Veronica, although generally a conservative character within the genus, gives some additional support to the most recent infrageneric classification of Veronica.  相似文献

The results of a study on pollen morphology of the species of the genus Ptilostemon growing in Italy: Ptilostemon niveus, P. greuteri, P. gnaphaloides, P. casabonae, P. strictus, P. stellatus by light and scanning electron microscopy were presented and discussed including in relation to their taxonomic position. The exine shows two different ornamentation patterns: echinae and scabrae. This last ornamentation pattern together with other features was previously described in the genus Ptilostemon. Of 6 species analysed, P. stellatus is the only one that shows a scabrate ornamentation of exine. Two pollen types were recognized through the exine ornamentation, length of ectocolpus and polar outline: P. stellatus type and P. niveus type. A multivariate analysis (principal component analysis and linear discriminant analysis) was carried out with the aim of examining potential morphological characters which could be used to identify taxa. The data suggest that several characteristics can be used to delimit the species in particular the exine ornamentation. The results of our studies support the actually subgeneric classification, and several features of the pollen grains analysed seem to have a palynoecological role and an important taxonomic significance. A dichotomous key based on palynological data is also given.  相似文献

The pollen morphology of two species of the Neotropical genus Ceratostema (Ericaceae) was examined by light, scanning and transmission electron microscopy. The Ceratostema species examined have 3-colporate pollen grains united in permanent tetrahedral tetrads that show a common condition encountered in the Ericaceae. But the septal exine was absent between two neighboring grains in each pollen tetrad of Ceratostema. The pollen tetrads without septa are the first report for the Ericaceae as well as other angiosperm families.  相似文献

The developmental stages of the pollen wall and tapetum, together with exine morphology were studied in a number of Crocus species, by light and scanning electron microscopy. Gametogenesis was characterized by: 1) development of a thick intine, 2) single mitosis, and 3) terminal amylolysis. The tapetum was of the secretory type. In C. cartwrightianus cv. albus, abnormal sporogenesis and gametogenesis produced vacuolate pollen grains with a reduced-or no intine layer, and rich with starch granules; the tapetum was either of the parietal-or amoeboid type. The exine was echinate and the pollen grains had different types of aperture: furrows, colpi or pores. The ornamentation varied from microreticulate to irregularly perforate. The exine framework was overlaid by a pellicle resistant to chloroform-carbon disulphide, on which a layer of pollenkitt was deposited. The results are discussed from both cytological and evolutionary viewpoints.  相似文献

Angiosperm male reproductive organs (anthers and pollen grains) have complex and interesting morphological features, but mechanisms that underlie their patterning are poorly understood. Here we report the isolation and characterization of a male sterile mutant of No Pollen 1 (NP1) in rice (Oryza sativa). The np1‐4 mutant exhibited smaller anthers with a smooth cuticle surface, abnormal Ubisch bodies, and aborted pollen grains covered with irregular exine. Wild‐type exine has two continuous layers; but np1‐4 exine showed a discontinuous structure with large granules of varying size. Chemical analysis revealed reduction in most of the cutin monomers in np1‐4 anthers, and less cuticular wax. Map‐based cloning suggested that NP1 encodes a putative glucose‐methanol‐choline oxidoreductase; and expression analyses found NP1 preferentially expressed in the tapetal layer from stage 8 to stage 10 of anther development. Additionally, the expression of several genes involved in biosynthesis and in the transport of lipid monomers of sporopollenin and cutin was decreased in np1‐4 mutant anthers. Taken together, these observations suggest that NP1 is required for anther cuticle formation, and for patterning of Ubisch bodies and the exine. We propose that products of NP1 are likely important metabolites in the development of Ubisch bodies and pollen exine, necessary for polymerization, assembly, or both.  相似文献

Pollen morphology was examined in 40 samples from 19 species in the genus Erythronium (Liliaceae) using scanning (SEM) and transmission electron microscopy (TEM). Pollen grains are monosulcate monads, except for a plant of E. oregonum which has dyad grains. North American and European species are characterized by reticulate exine sculpture composed of various degrees of wavy muri and variably-sized lumina. The most distinctive feature of exine sculpture in the genus is exhibited by the Asian species E. japonicum, which has a unique reticulate pattern composed of striate muri and a distinctive exine structure without columellae. The distinctness of E. japonicum pollen suggests that it has specialized in isolation from species in North America and Europe.  相似文献

Pollen morphology of Gyrostemonaceae, Bataceae, andKoeberlinia, which have been affiliated with glucosinolate-producing taxa, was examined by field emission scanning and transmission electron microscopy. Pollen grains of Gyrostemonaceae are tricolpate with scabrate-spinulate surface and have a thick, unstratified exine, while those of Bataceae are tricolporoidate with granular surface and have a thin exine with a single, outermost granular layer. Gyrostemonaceae and Bataceae, which had often been considered sister taxa based on palynological similarity and now are considered more distantly related, have a similar spongy ektexine, but differences between them are evident.Koeberlinia, which is recently considered a sister group to Bataceae+Salvadoraceae (with no spongy ektexine), has tricolporoidate pollen composed of a plesiomorphic, stratified exine with columellae. The totality of evidence indicates that, contrary to earlier observations, pollen of Gyrostemonaceae and Bataceae does not closely resemble each other, and that the spongy ektexine, which looks to be similar in TEM sections, is a homoplasy that evolved independently in the two families. Dedicated to the late Prof. Emer. Kankichi Sohma (August 28, 1926–June 26, 1995), who supervised us for our M. Sc. and D. Sc. programs at Tohoku University, Sendai. The pollen of six taxa of the genus Rubus endemic to Poland (R. capitulatus, R. chaerophylloides, R. ostroviensis, R. posnaniensis, R. seebergensis and R. spribillei)was studied using light and scanning electron microscopy. The study objective was to extend the knowledge of the species pollen morphology and to investigate whether pollen morphology may help to taxonomically distinguish a very rare bramble species. Study material was derived from six natural localities where the species occur in the Wielkopolska region (Poland). Ten quantitative pollen grain features were analysed as well as the following qualitative ones: outline, shape, type of bridge and exine sculpture. Only mature, correctly formed pollen grains (30 per specimen) were measured and 180 pollen grains were examined in total. It was found that R. capitulatus and R. seebergensis had the highest mean values of P, E and Le features. In addition, pollen grains of R. capitulatus were most flattened (mean P/E ratio equals 1.14), while those of R. seebergensis were most elongated (mean P/E ratio equals 1.27). R. spribillei was the only species with striae of similar width as muri, whereas striae in the remaining species were wider. R. chaerophylloides and R. posnaniensis were characterised by specific, similar exine sculpture. Nevertheless, it should be stated that differences between pollen features of the species studied are so small and the variability of these features are so large that it is difficult to clearly identify the species studied. Therefore, pollen grain morphology can serve only as an auxiliary feature for the determination of these species.  相似文献

利用扫描电镜对8种石竹属(Dianthus)植物的花粉形态进行了观察。结果表明:本属植物花粉为单粒花粉,圆球形,大小为30.5~44.0 μm,具多个萌发孔,孔数为12~16个,分散,孔圆形,孔径4.1~7.0 μm,具孔膜,孔膜上着生有若干棵小刺,花粉外壁具刺状和穴状纹饰,表面有或无穿孔。研究表明:花粉大小、表面纹饰、特别是萌发孔的形态特征存在较明显差异。可为石竹属植物的分类及野生资源利用等相关研究提供了可靠的孢粉学参考。  相似文献

The pollen of nine endemic taxa of the Gonospermum Less., Lugoa DC. and Tanacetum L. complex and of T. parthenium L. (introduced species from the Canary Islands) was studied using light and scanning electron microscopy with the aim of extending the knowledge of their morphology and to investigate whether pollen morphology may help clarifying the controversial taxonomy of these taxa. Twenty six populations in the Canary Islands (Spain) were studied and the following parameters were measured: polar axis (P), equatorial diameter (E), polar exine thickness (Ex1), length (L) and thickness (A) of the spines in the meridional optical section, exine thickness at the centre of the mesocolpium in the interapertural area (Ex2) and distance between spines (De) in the equatorial optical section. The studies show that the pollen of the three genera is very similar regarding shape, apertures, and ornamentation, but may be distinguished by size and exine thickness.  相似文献
