Pollen data from the atmosphere of Vigo, NW Spain was collected using a Hirst type pollen trap over a seven-year period (1995–2001). A total of 56 different pollen types were identified, among which Urticaceae, Poaceae, Betula and Quercus represent the greatest risk for people suffering from allergic rhinitis (hay fever) or other allergic diseases. Although in the atmosphere of Vigo the presence of allergenic pollen is constant throughout the year, the months of March and April account for 40% of the annual total pollen count. Two main risk periods have been identified for asthma and allergies: (1) March – April, and (2) June – July, the latter is of greater importance due to high concentrations of Poaceae pollen. Correlation analysis with meteorological parameters demonstrates that rainfall, relative humidity, maximum temperature, sun hours and north-easterly winds are the main factors influencing the average daily pollen concentrations in the atmosphere.  相似文献

Background

Evidence is compelling for a positive correlation between climate change, urbanisation and prevalence of allergic sensitisation and diseases. The reason for this association is not clear to date. Some data point to a pro-allergenic effect of anthropogenic factors on susceptible individuals.

Objectives

To evaluate the impact of urbanisation and climate change on pollen allergenicity.

Methods

Catkins were sampled from birch trees from different sites across the greater area of Munich, pollen were isolated and an urbanisation index, NO2 and ozone exposure were determined. To estimate pollen allergenicity, allergen content and pollen-associated lipid mediators were measured in aqueous pollen extracts. Immune stimulatory and modulatory capacity of pollen was assessed by neutrophil migration assays and the potential of pollen to inhibit dendritic cell interleukin-12 response. In vivo allergenicity was assessed by skin prick tests.

Results

The study revealed ozone as a prominent environmental factor influencing the allergenicity of birch pollen. Enhanced allergenicity, as assessed in skin prick tests, was mirrored by enhanced allergen content. Beyond that, ozone induced changes in lipid composition and chemotactic and immune modulatory potential of the pollen. Higher ozone-exposed pollen was characterised by less immune modulatory but higher immune stimulatory potential.

Conclusion

It is likely that future climate change along with increasing urbanisation will lead to rising ozone concentrations in the next decades. Our study indicates that ozone is a crucial factor leading to clinically relevant enhanced allergenicity of birch pollen. Thus, with increasing temperatures and increasing ozone levels, also symptoms of pollen allergic patients may increase further.  相似文献

Cycas circinalis L. or queen sago is a common ornamental gymnosperm in tropics and subtropics. The objectives of the study were (a) to observe the seasonal variation of queen sago pollen in the atmosphere of a rural and an industrial area of West Bengal, India, (b) to visualize its allergenic potential on local population, and (c) to identify and isolate the important IgE-binding protein component present in the pollen extract. A two-year aerobiological survey was performed with Burkard personal volumetric sampler, and Cycas pollen was found to be present in air during April–July. Among 172 respiratory allergic patients of study area, 25.58% showed skin reaction to Cycas pollen extract. The allergenicity of the pollen extract was confirmed by in vivo (skin reaction test) and in vitro (IgE-ELISA and dot blotting) analyses and immunoblotting. Two components of 39.6 and 20.7 kDa were found to be the important IgE-binding proteins in pollen extract. The 20.7 kDa component was purified by two-step gel electrophoresis and it was found to retain its IgE reactivity. This component can be used for further work in diagnostic and therapeutic purpose in susceptible individuals. Pollen material of the two anemophilous plants of known allergic significance Dactylis glomerata (Poaceae) and Betula verrucosa (Betulaceae) was collected at several places (in Sweden and France) and exposed to natural or experimental conditions. This material was analyzed with Electron Probe X Microanalysis (EPMA) (System ORTEC 5000) attached to a Scanning Electron Microscope (Jeol JMS 35), or with EDAX, a 10000 X-Ray Analyser, Link System, connected to a SEM-microscope (Jeol JSM 820).

It is clear that the environment has an effect on such a co-inductor or adjuvant of allergenicity as K. The possible presence of pollutant inorganic (mineral) particles on the pollen grain surface may play a role in the allergenic activity. Pollen, consequently, may be used as sensitive biological indicators of atmospheric pollution.  相似文献

Aerobiological investigations of the last ten years in the former USSR emphasised, preferentially the following problems: content of pollen in the air, pollen transport, and impact of pollen on the health of the human. Pollen grains which cause various allergic diseases have been the main object of extensive aerobiological studies. Ado and his collaborators have made valuable contributions to Aerobiology in the former USSR (Ado 1978, 1980, 1989, 1990; Ado & Gubankova, 1979 a, b). Their investigations confirmed the existence of a correlation between the number of allergic patients and the concentration of pollen grains in the atmosphere, and some peculiarities of the allergic diseases prevalent in towns.

Later, aerobiology was elaborated by the investigations of Kobzar (1987), Kobzar & Kharitonova (1990), Kobzar et al. (1990, 1991), etc. The possibility of airborne pollen grains as indicators of environmental contamination and pollution levels was demonstrated. Various data show the periods during which pollen grains are present in atmosphere, their peaks and the levels of concentration which are able to cause allergies in the territory of the former USSR.  相似文献

Pollen proteins that are located in the cytoplasm or on the surface of the exine can function as allergens and evoke immune system responses in sensitive patients, leading to allergic rhinitis and asthma. In this research, the pollen allergenicity and ability to induce IgE response of the pollen of two plant species were studied in rats. Acroptilon repens is an herbaceous, invasive plant with entomophilous pollen, while Juglans regia which is a tree crop produces anemophilous pollen. Immunoblot analysis using sera of sensitised rats revealed IgE reactivity to three protein bands including the 70, 41 and 25.12 kDa bands present in the A. repens pollen extract, while only one single immunogenic band of 11 kDa was detected in J. regia pollen extract. Both pollen extracts increased the eosinophil content and caused some clinical signs of allergy in treated rats. The results showed that both entomophilous and anemophilous pollen can be allergenic.  相似文献

The prevalence of allergic disease has increased world wide during the last decades. Pollen allergy is the most typical form of allergic disease. The increase in its frequency during recent years is the most evident. Environmental factors play an important role in the problem of pollen allergy in large cities. The aim of this research is determination of allergenicity of Canna pollen in polluted and non-polluted conditions, detection of their allergenic proteins and also elucidation of some microscopic effects of air pollutants on pollen structure and proteins. Mature and immature pollen grains of Canna indica were collected from polluted and non-polluted areas. Pollen grains were studied by scanning electron microscopy. Mice were sensitized by injection of pollen extract and an adjuvant for five times. Allergy potency of different pollen extracts were compared by means of skin test, blood eosinophills number and IgE levels in sensitized and treated animals. Pollen proteins were studied by SDS-PGE and allergenic proteins were detected by immunoblotting techniques. Scanning electron microscope study of the pollen grains showed that in polluted areas, air born particles accumulated on the surface of pollen and changed both pollen's shape and pollen's tectum. Also many vesicles were released out of polluted pollen and the pollen material agglomerated on the surface of pollen. SDS-PAGE showed that different proteins exist in mature and immature pollen. In pollen collected from polluted area, some of protein bands between 22 and 45 kDa were disappeared . Also in all polluted pollen grains, protein content of pollen decreased in response to air pollution causing the release of pollen proteins. According to our experiments and regarding induction of allergic symptoms, the polluted pollen is more effective than non-polluted one, and mature pollen has more allergy potency than immature one.  相似文献

Ragweed allergy is one of the primary causes of seasonal allergies in Europe and its prevalence is expected to rise. The leaf beetle Ophraella communa, recently and accidentally established in N-Italy and S-Switzerland, represents a promising approach to control ragweed, but negative side effects should be excluded before its use. Since biotic and abiotic stresses are known to influence the allergenicity of pollen, we set out to assess the effect of sub-lethal defoliation by O. communa on the quantity and quality of ragweed pollen. Seventeen sister pairs (including six clones) of ragweed plants were grown in controlled conditions. One of each pair was exposed to O. communa as soon as the plant started to produce reproductive structures. After 10 weeks of exposure, plant traits were measured as a proxy for pollen quantity. Pollen quality was assessed by measuring its viability and allergenicity. Generally, plants produced very few male flowers and little amount of pollen. Damage by the beetle was severe with most of the leaf tissue removed, but no treatment effect was found on any of the quantitative and qualitative traits assessed. In conclusion, O. communa did not increase the amount or allergenicity of ragweed pollen grains in our experimental conditions.  相似文献

Allergic diseases have been increasing in industrialized countries. The environment is thought to have both direct and indirect modulatory effects on disease pathogenesis, including alterating on the allergenicity of pollens. Certain plant proteins known as pathogenesis-related proteins appear to be up-regulated by certain environmental conditions, including pollutants, and some have emerged as important allergens. Thus, the prospect of environmentally regulated expression of plant-derived allergens becomes yet another potential environmental influence on allergic disease. We have identified a novel pathogenesis-related protein allergen, Jun a 3, from mountain cedar (Juniperus ashei) pollen. The serum IgE from patients with hypersensitivity to either mountain cedar or Japanese cedar were shown to bind to native and recombinant Jun a 3 in Western blot analysis and ELISA. Jun a 3 is homologous to members of the thaumatin-like pathogenesis-related (PR-5) plant protein family. The amounts of Jun a 3 extracted from mountain cedar pollen varied up to 5-fold in lots of pollen collected from the same region in different years and between different regions during the same year. Thus, Jun a 3 may contribute not only to the overall allergenicity of mountain cedar pollen, but variable levels of Jun a 3 may alter the allergenic potency of pollens produced under different environmental conditions.  相似文献

The ambient atmosphere is dominated with pollen and spores, which trigger allergic reactions and diseases and impact negatively on human health. A survey of pollen and fungal spores constituents of the atmosphere of Garki, Abuja (North-Central Nigeria) was carried out for 1 year (June 1, 2011–May 31, 2012). The aim of the study was to determine the prevalence and abundance of pollen and fungal spores in the atmosphere and their relationship with meteorological parameters. Airborne samples were trapped using modified Tauber-like pollen trap, and the recipient solutions were subjected to acetolysis. Results revealed the abundance of fungal spores, pollen, fern spores, algal cysts and diatoms in decreasing order of dominance. The atmosphere was qualitatively and quantitatively dominated by pollen during the period of late rainy/harmattan season than the rainy season. Numerous fungal spores were trapped throughout the sampling periods among which Alternaria spp., Fusarium spp., Cladosporium spp. and Curvularia spp. dominated. These fungi have been implicated in allergic diseases and are dermatophytic, causing diverse skin diseases. Other pathogenic fungi found in the studied aeroflora were Dreschlera spp., Helminthosporium spp., Torula spp., Pithomyces spp., Tetraploa spp., Nigrospora ssp., Spadicoides spp., Puccinia spp. and Erysiphe graminis. Total pollen and fungal spores counts do not show significant correlation with meteorological parameters.  相似文献

The Pinus genus has an elevated pollen production and an anemophilous nature. Although considered to be hypoallergenic, numerous cases of allergies caused by Pinus pollen have been cited and different authors believe that its allergenicity should be studied in more depth. In the city of Vigo several patients have tested positive for Pinus pollen extracts in skin tests, some of them being mono-sensitive to such pollens. In order to ascertain the behaviour of Pinus pollen and its correlation to the main meteorological factors, we carried out an aerobiological study in the city of Vigo from 1995 to 1998 by using a Hirst active-impact volumetric sporetrap, model Lanzoni VPPS 2000, placed on the left bank of the Vigo estuary (42°14’15’’N, 8°43’30’’W). Pinus has high quantitative importance in the airborne pollen spectrum of the city. It is one of the best represented taxa constituting 13%–20% of the total annual pollen levels. The quantity of Pinus pollen present in the atmosphere of the city of Vigo throughout a year is 5751 grains (as the average for the sampled years), with a very long pollination period, from the middle of January until May. The maximum concentration was recorded in 1998 with 1105 grains/m3 on 3 March, a much greater value than those for the previous years. At the end of its pollination period there is usually a final increase in Pinus pollen concentrations coinciding with the pollination of Pinus silvestris, which are more abundant in mountainous areas far from the city. Olive is recognized as a crop with great impact in agricultural, socioeconomic, environmental and public health sectors. The last is becoming more important during recent years as consequence of the increase of the pollen allergy in south Europe prompted by the widespread Olea pollen allergic reactions. The aim of the study was to quantify, for the first time, the variations of the Ole e 1 allergen amount in Olea pollen grains from four cultivars in three regions of Portugal. How weather parameters can affect the allergen production was also assessed. The study was conducted in three olive producer areas of Portugal from 2010 to 2013, Santarém (Central), Elvas (Southeast) and Mirandela (Trás-os-Montes region, Northeast). Mature pollen of four different cultivars (Cobrançosa, Arbequina, Picual and Verdeal) was collected during the olive flowering season. Ole e 1 was quantified using specific 2-site antibody ELISA. Pollen of the olive groves at the boundary Olea bioclimatic distribution in the Mirandela registered the higher allergen content for all varieties in each study year. Arbequina was the variety that showed the lower Ole e 1 allergen concentration, whereas the higher content was registered for Cobrançosa. The main meteorological parameters that influenced the allergen Ole e 1 concentration in the pollen grains were the rainfall and temperatures related variables. The knowledge of the allergenicity in different olive cultivars is an important tool in the selection of the most adequate for planting as ornamental crop and to adjust the pollen extracts used for diagnosis or even immunotherapy.

Although the prevalence of asthma and allergic rhinitis has been increasing in tropical regions, little is known about the allergenicity of pollens from tropical plant families or the importance of ongoing environmental changes. We investigated associations between daily average pollen counts of several tropical plant families and sales of medications for the treatment of allergic rhinitis in Darwin, Australia—a tropical setting in which grass abundance has increased due to increased fire frequencies and the introduction of African pasture grasses. Daily pollen counts with detailed identification of plant species were undertaken in conjunction with a weekly survey of flowering plant species from April 2004 to November 2005. Five pharmacies provided daily sales data of selected medications commonly used to treat allergic rhinitis. We used generalized linear modeling to examine outcomes. All analyses accounted for the potential confounding effects of time trends, holidays, respiratory viral illnesses, meteorological conditions, and air pollution. The peak total pollen count was 94 grains/m3. Despite the low levels of Poaceae (grass) pollen (maximum daily count, 24 grains/m3), there was a clear association with daily sales of anti-allergic medications greatest at a lag of 1 day. Sales increased by 5% with an interquartile range rise (3 grain/m3) in Poaceae pollen (5.07%, 95%CI 1.04%, 9.25%). No associations were observed with pollen from other plant families. Although further testing is required, we suggest that an overlooked aspect of the “grass-fire cycle” that is degrading many tropical landscapes, could be an increase in the prevalence of allergic rhinitis.  相似文献

Ragweed pollen is the main cause of allergenic diseases in Northern America, and the weed has become a spreading neophyte in Europe. Climate change and air pollution are speculated to affect the allergenic potential of pollen. The objective of this study was to investigate the effects of NO2, a major air pollutant, under controlled conditions, on the allergenicity of ragweed pollen. Ragweed was exposed to different levels of NO2 throughout the entire growing season, and its pollen further analysed. Spectroscopic analysis showed increased outer cell wall polymers and decreased amounts of pectin. Proteome studies using two‐dimensional difference gel electrophoresis and liquid chromatography–tandem mass spectrometry indicated increased amounts of several Amb a 1 isoforms and of another allergen with great homology to enolase Hev b 9 from rubber tree. Analysis of protein S‐nitrosylation identified nitrosylated proteins in pollen from both conditions, including Amb a 1 isoforms. However, elevated NO2 significantly enhanced the overall nitrosylation. Finally, we demonstrated increased overall pollen allergenicity by immunoblotting using ragweed antisera, showing a significantly higher allergenicity for Amb a 1. The data highlight a direct influence of elevated NO2 on the increased allergenicity of ragweed pollen and a direct correlation with an increased risk for human health.  相似文献

The aim of this study was to construct a picture of the influence of meteorological conditions on the start and duration of the airborne Betulaceae pollen season and the pollen concentrations in the atmosphere of Zagreb, Croatia. The study during three seasons (2002–2004) used a 7‐day Hirst‐type volumetric pollen and spore trap. Total annual airborne pollen of Alnus, Corylus and Betula greatly varied from year to year. The differences in the dates of onset of airborne pollen presence of Alnus, Corylus and Betula noted in Zagreb in 2002–2004 were controlled by weather conditions, particularly temperature and precipitation. In all years studied, airborne pollen peaks were recorded on days with temperature above 0°C and without or minimal precipitation. The mean number of days with airborne pollen concentrations exceeding levels which provoke symptoms of an allergic reaction was 15, 16 and 29 days for alder, hazel and birch, respectively. The results of the present study may provide useful data for allergologists to reach accurate diagnoses, and timely information on concentrations of airborne pollen types and concentrations for individuals with pollen hypersensitivity.  相似文献

Being major ornamental street trees, species of Platanus are widely planted in the Shanghai urban area. A great deal of allergenic Platanus pollen is released from the trees and suspended in the atmosphere during its flowering season, ultimately causing allergic respiratory diseases. Few papers have focused on the distribution of this type of pollen and its expression of allergenic proteins. In order to investigate any differences in protein expression in Platanus pollen following exposure to gaseous and particulate pollutants, a special apparatus was designed. Exposure condition (such as temperature, humidity, and exposure time) of Platanus pollen and gaseous pollutants can be simulated using of this apparatus. Fresh Platanus orientalis pollen, pollutant gases (NO2, SO2, NH3), and typical urban ambient particles (vehicle exhaust particles, VEPs) were mixed in this device to examine possible changes that might occur in ambient airborne urban pollen following exposure to such pollutants. Our results showed that the fresh P. orientalis pollen became swollen, and new kinds of particles could be found on the surface of the pollen grains after exposure to the pollutants. The results of SDS-PAGE showed that five protein bands with molecular weights of 17–19, 34, 61, 82, and 144 kDa, respectively, were detected and gray scale of these brands increased after the pollen exposure to gaseous pollutants. The two-dimensional gel electrophoresis analysis demonstrated that a Platanus pollen allergenic protein (Pla a1, with a molecular weight of 18 kDa) increased in abundance following exposure to pollutant gases and VEPs, implying that air pollutants may exacerbate the allergenicity of pollen.  相似文献

This study reports the hourly distribution of the allergenic airborne pollen types more abundant in the atmosphere of Porto (Portugal) during the studied period. This knowledge will allow an adequacy daily routine for allergic patients during the hours of higher airborne concentrations. The airborne pollen concentration was continuously performed from January 2003 to December 2007 in the city of Porto using a Hirst-type volumetric sampler. Urticaceae, Cupressaceae, Acer spp., and Plantago spp. airborne pollen presented higher concentrations in the morning, while Alnus spp. and Betula spp. pollen were mainly present during the afternoon. Olea europaea and Platanus spp. pollen were regularly distributed along the day, while Poaceae and Pinus spp. pollen presented two diurnal maxima.  相似文献

A continuous aeropalynologic survey of the atmosphere of La Plata was carried out between July 1998 and June 2001 in order to study flowering development from winter to summer using a Lanzoni volumetric spore trap. The total pollen spectrum was represented by 79 pollen types. Between 10 and 12 pollen types showed a relative concentration of more than 1% of the annual total. Airborne pollen was mainly represented by Platanus, Fraxinus, Cupressaceae, Poaceae, Urticaceae, Cyperaceae, Myrtaceae, Celtis, Casuarina and Morus during the 3-year period. Acer and Ambrosia pollen types were only dominant in the first 2 years. Maximum absolute concentrations were recorded in the the July 1998–June 1999 period, and the minimum concentrations were recorded in the July 2000–June 2001 period. The contribution of the arboreal pollen grains was higher than 68% relative to the annual total for each year. Two periods of maximum pollen emissions were found for each year: pollen from aboreal taxa predominated from July to October, and pollen from herbaceous taxa predominated from November to March. There was very little pollen in the atmosphere between April and June. The maximum arboreal and herbaceous pollen emissions were recorded during hours of daylight: at 10:00 and 14:00 hours.  相似文献

Grasses (Poaceae) are very common plants, which are widespread in all environments and urban areas. Despite their economical importance, they can represent a problem to humans due to their abundant production of allergenic pollen. Detailed information about the pollen season for these species is needed in order to plan adequate therapies and to warn allergic people about the risks they take in certain areas at certain moments. Moreover, precise identification of the causative species and their allergens is necessary when the patient is treated with allergen‐specific immunotherapy. The intrafamily morphological similarity of grass pollen grains makes it impossible to distinguish which particular species is present in the atmosphere at a given moment. This study aimed at developing new biomolecular tools to analyze aerobiological samples and identifying major allergenic Poaceae taxa at subfamily or species level, exploiting fast real‐time PCR. Protocols were tested for DNA extraction from pollen sampled with volumetric and gravimetric methods. A fragment of the matK plastidial gene was amplified and sequenced in Poaceae species known to have high allergological impact. Species‐ and subfamily‐specific primer–probe systems were designed and tested in fast real‐time PCRs to evaluate the presence of these taxa in aerobiological pollen samples. Species‐specific systems were obtained for four of five studied species. A primer–probe set was also proposed for the detection of Pooideae (a grass subfamily that includes also major cereal grains) in aerobiological samples, as this subfamily includes species carrying both grass allergens from groups 1 and 5. These, among the 11 groups in which grass pollen allergens are classified, are considered responsible for the most frequent and severe symptoms.  相似文献
