Free amino acids are important bioactive ingredients in herbs and herbal products. Coptis herbs contain a variety of free amino acids; however, studies have not yet analyzed the relationship between free amino acids and species of Coptis herbs. In the current study, the contents of 20 free amino acids in Coptis chinensis Franch., Coptis teeta Wall., and Coptis deltoidea C.Y. Cheng et Hsiao were determined using an automatic amino acid analyzer to evaluate the differences between the three species. We found that the major amino acids (Asn, Arg, and γ-aminobutyric acid (GABA)) were significantly more in C. chinensis than in the other two species. In addition, Asp content was significantly more in C. deltoidea. The study concluded that Asn can be used to identify C. deltoidea, C. chinensis, and C. teeta, and Gln and Arg can be used to distinguish C. teeta from C. chinensis and C. deltoidea. These findings suggested that free amino acids as active ingredients can be used to identify Coptis herb species.  相似文献

The non-essential free amino acids of Peneaus japonicus hemolymph (asp, ser, glu, pro, gly, tyr, arg, ala, cys, tau) are more common than the essential ones: 750 pmol μl?1 of hemolymph vs 330 pmol. Under a light-dark (L-D); 12-12 photoperiod, tricircadian variations of males or tetracircadian variations of females are more pronounced for non-essential amino acids then for essential ones. In the first case, free amino acid concentrations of hemolymph can be multiplied by a factor of three, and in the second case by a factor of six; but circadian variations of females are greater than those of males. Differences between the maximum and minimum of essential free amino acid concentrations of male and female hemolymph are more frequent than for non-essential amino acids. A differences of 2 h between the minimum of essential and non-essential amino acid concentrations in males only appeared during the afternoon. The more concentrated free amino acids in P. japnicus hemolymph are glycine, proline, histidine, and alanine; the less concentrated are lysine, cysteine and glutamic acid while others, like leucine, isoleucine, phenylalanine and tyrosine can only be estimated at 10.00 and 24.00 h.  相似文献

Free amino acids, amino sulfonic acids, sugars and sterols have been examined and quantitatively determined in 10 brown seaweeds. Acidic amino acids and their amides are the main components of the amino acid fraction. Cysteinolic acid, taurine and its N-methyl derivatives have been identified in most of the species examined. In all the algae, mannitol is present, sometimes in very large amounts. The sterol fractions of all the species contain fucosterol, cholesterol and 24-methylenecholesterol; minute amounts of 22-dehydrocholesterol and 24-methylcholesta-5, 22-dien-3β-ol have also been frequently detected.  相似文献

2(S),3′(S)-N-(3-Amino-3-carboxypropyl)azetidine-2-carboxylic acid and 2(S),3′(S),3″(S)-N-[N-(3-amino-3-carboxypropyl)-3-amino-3-carboxypropyl]azetidine-2-carboxylic acid have been isolated from seeds of Fagus silvatica L. (beechnuts). The structures have been established by PMR- and 13C-NMR-spectroscopy and by synthesis from l-azetidine-2-carboxylic acid. The second of the new amino acids is identical with nicotianamine. previously isolated from Nicotiana tabacum but assigned a different formula. The ring opening reactions of azetidine-2-carboxylic acid in neutral solution have been studied and the chemical and possibly biochemical precursor role of this amino acid for various amino acids including the two new ones described here, nicotianine [N-(3-amino-3-carboxypropyl)nicotinic acid] and methionine is discussed.  相似文献

The amino acids of four species of Microsporidia parasitizing crustaceans were investigated: three species — Thelohania maenadis, Ormieresia carcini and Ameson pulvis — are parasites of Carcinus mediterraneus; Inodosporus sp. is a parasite of Palaemon serratus. Seventeen protein amino acids were identified of which aspartic acid, glutamic acid and lysine were quantitatively the most important, being quantitatively similar in all four parasites. The relative amounts of the next most abundant amino acids were found to vary and might serve as a systematic criterion at the genus level. For example, phenylalanine is predominant in Ormieresia and leucine in Inodosporus: serine in Thelohania and glycine in Ameson are also predominant, but to a lesser extent. The free amino acids composition shows little qualitative variation among the four genera, but quantitative differences are found in the composition of Microsporidia parasitizing the same host species; this may reflect variations in the amino acid metabolism of the parasite. The urea cycle in Ormeresia is most remarkable in this connection. The free amino acid level in the parasites was generally found to be in inverse proportion to the level in the host; the amino acids which are found to exist at high levels in the parasites correspond to essential amino acids of the Crustacea. Metabolic and adaptative relations are discussed.  相似文献

We report here the complete cDNA sequence of a nonfibrillar collagen (COLch) isolated from the marine sponge Chondrosia reniformis, Nardo 1847 using a PCR approach. COLch cDNA consists of 2,563 nucleotides and includes a 5′ untranslated region (UTR) of 136 nucleotides, a 3′ UTR of 198 nucleotides, and an open reading frame encoding for a protein of 743 amino acids with an estimated M r of 72.12 kDa. The phylogenetic analysis on the deduced amino acid sequence of C-terminal end shows that the isolated sequence belongs to the short-chain spongin-like collagen subfamily, a nonfibrillar group of invertebrate collagens similar to type IV collagen. In situ hybridization analysis shows higher expression of COLch mRNA in the cortical part than in the inner part of the sponge. Therefore, COLch seems to be involved in the formation of C. reniformis ectosome, where it could play a key role in the attachment to the rocky substrata and in the selective sediment incorporation typical of these organisms. qPCR analysis of COLch mRNA level, performed on C. reniformis tissue culture models (fragmorphs), also demonstrates that this matrix protein is directly involved in sponge healing processes and that soluble silicates positively regulate its expression. These findings confirm the essential role of silicon in the fibrogenesis process also in lower invertebrates, and they should give a tool for a sustainable production of marine collagen in sponge mariculture.  相似文献

A lysimeter study was conducted on Cajeme wheat (Triticum aestivum L.) to investigate the impact of salinity on protein and free amino acid content of the grain. Cross correlations were obtained between 16 different soil-plant-water based parameters and the concentration and total accumulation of amino acids. The results indicated that after 3 years of irrigation, the majority of protein bound and free amino acids increased in concentration in the grain. However, both free tryptophan and free proline revealed decreasing concentrations with increasing salinity. Free tryptophan showed a synergism between total accumulation, yield and concentration. Free proline concentrations decreased in association with increasing protein concentrations. Cross correlations of the 16 soil-plant-water based parameters with free and protein bound amino acids revealed significant correlations for free aspartic acid and glycine with total accumulation but not with concentrations. Only methionine plus cystine was lower than suggested FAO levels for essential amino acids and was lower in the third year than in the first year.  相似文献

Two diastereoisomers of 4-carboxy-4-hydroxy-2-aminoadipic acid have been isolated from leaves and inflorescences of Caylusea abyssinica. Green parts of the plant also contain appreciable amounts of the two diastereoisomers of 4-hydroxy-4-methylglutamic acid, 3-(3-carboxyphenyl)alanine, (3-carboxyphenyl)glycine, 3-(3-carboxy-4-hydroxyphenyl)alanine, (3-carboxy-4-hydroxyphenyl)glycine and in low concentration 2-aminoadipic acid, saccharopine [(2S, 2′S)-N6-(2-glutaryl)lysine] and some γ-glutamyl peptides. The acidic amino acids were separated from other amino acids on an Ecteola ion exchange column with M pyridine as eluant.  相似文献

The marine sponges Dysidea avara and Chondrosia reniformis (globular forms) were cultured in the laboratory on a diet of viable Phaeodactylum tricornutum cells and dissolved nutrients (algae and fish powders). Our growth data were combined with literature data for Pseudosuberites andrewsi (a globular sponge) and for the encrusting sponges Oscarella lobularis, Hemimycale columella, and Crambe crambe. The suitability of three growth models—linear, exponential, and radial accretive—for describing the growth of globular and encrusting sponges was assessed. Radial accretive growth was determined to be the best model to describe growth of both encrusting and globular sponges. Average growth rates of 0.051 ± 0.016 and 0.019 ± 0.003 mm/day (calculated as the increase of the radius of the sponge per day) were obtained experimentally for D. avara and C. reniformis, respectively.  相似文献

Amino acid compositions of the eggs of five lepidopteran hosts for Trichogramma minutum were compared with each other and with a non-host species, Rhodnius prolixus, in which T. minutum oviposits but does not develop. Host eggs are quite homogeneous, particularly when compared according to groupings of potentially interconvertible amino acids. Combined mole percent values for glycine, serine and alanine were higher in hosts (27.5–29.2 mole%) than in R. prolixus eggs (21.5 mole%), in bovine serum albumin (14.9%), which has been used as a protein source in artificial diets for T. minutum, or in many of the mixtures used in published diets for this species. Since these three amino acids make up 26.3 mole% of the adult amino acid content of T. minutum, their deficiency in diets could require metabolic compensation detrimental to development.Adult T. minutum arising from eggs of Manduca sexta, Choristoneura fumiferana, and Sitotroga cerealella are similar in amino acid composition to each other and, in general, to their hosts. Variability appears greater in hosts than in adult wasp composition, suggesting some interconversion of host amino acids to accommodate inflexible nutritional requirements of T. minutum.In the three host species tested, free amino acids constituted 15.8–19.3% by weight of the amino acid in egg contents. In M. sexta eggs, glycine, serine and alanine together make up 28.4% by weight of the total free amino acid, a much higher proportion than in many published diets. The four free amino acids (isoleucine, leucine, phenylalanine and histidine) reported to be oviposition stimulants in experiments on encapsulated diets are present in sufficient concentrations to induce oviposition in the host species tested and in R. prolixus. S. cerealella egg contents having approximately 1.8 g amino acid, yield one or rarely two adult T. minutum (1g amino acid/insect). In contrast, M. sexta eggs with 94 g amino acid each yield an average of 10–12 adults (8.2g amino acid/insect). This suggests that small hosts are allocated few eggs which can only develop into small adults because of nutrient supply (parasitoid size in metabolically restricted), whereas much larger hosts are allocated proportionately fewer eggs than the former resulting in larger, and presumably more viable and fecund, adults (parasitoid size is established behaviourally).  相似文献

Transplantation was employed to determine how marine sponges grow in different in situ conditions of light and current. The growth of Verongia aerophoba (Schmidt), which contained symbiotic cyanobacteria. was enhanced in light, particularly when sediment was excluded by a clear shield. V. cavernicola Vacelet and Chondrosia reniformis Nardo. which did not contain cyanobacteria, grew preferentially in the shade and were inhibited by light. The growth of Petrosia ficiformis (Poirct) and Chondrilla nucula Schmidt, which may or may not contain cyanobacteria. did not appear to be favoured by either light or shade. The growth of sponges in lower current speeds was considerably reduced ; this reduction was. however, partially reversed in those sponges with cyanobacteria when grown in the light. Presumably symbiotic cyanobacteria are beneficial to sponges growing in the light because they transfer photosynthetically fixed nutrient and shield the underlying tissue. Significant morphological differences were observed in sponges grown under different environmental conditions.  相似文献

The amino acid composition of the acid soluble fraction of Euglena gracilis was determined from cells grown in 4 different culture media. Glutamic acid is the major free amino acid. Hydrolysis of this fraction increases the amount of free amino groups, the major amino acids found are then glutamic acid, aspartic acid, glycine and arginine. The pattern of amino acid distribution is similar in all 4 culture media. L-arginyl-L-glutamine was isolated and identified in extracts from all 4 culture conditions. It was shown to be a metabolic intermediate by radioactivity chase experiments.  相似文献

The sterol composition of four sponges was determined by a combination of gas chromatography and mass spectrometry. Cliona viridis and Chondrosia reniformis contained mainly C27-C29Δ5 mono- and di-unsaturated sterols. Halichondria bowerbanki and Hymeniacidon sanguinea contained stanols and Δ5-sterols. In hydrolysates of the eastern subterranean termite, Reticulitermes flavipes, the most abundant protein amino acids (μmoles) were glycine, alanine, and glutamic acid; the least abundant were methionine and histidine. Sawdust from both sound and Lenzites trabea-decayed sapwood blocks of sugar maple, loblolly pine, and slash pine was force-fed to termites. A diet of decayed rather than sound wood had little effect on protein amino acid composition of the termites; glycine content varied the most. In contrast, diet affected the free amino acid composition. Except for glutamic acid, the major protein amino acids of the termites were not the predominant free amino acids. Tyrosine and histidine were relatively more abundant as free than as protein amino acids. Greatest differences in protein amino acid compositions of sound and decayed wood were in contents of glycine, leucine, lysine, and arginine.  相似文献

Developing eggs of vendace (Coregonus albula L.) and whitefish (C. lavaretus L.) were experimentally delayed in hatching by incubation at low water temperature (1–2°). Some eggs were taken during this period to a water temperature which was gradually raised up to 8° to provoke mass hatching of embryos. The pattern of free amino acids was followed in eggs incubated at both temperatures. During a 56 days period, the content of several essential amino acids significantly decreased in eggs of both species. For instance, the lysine content dropped from 703 to 270 mg/g dry matter and the arginine content from 257 to 13.3 mg/100 g dry matter in whitefish eggs. A similar pattern of decreasing level of free amino acids in embryonated ova up to hatching was characteristic for essential amino acids and serine. Methionine was exceptional; its level remained approximately the same. On the other hand, non-essential amino acids showed a significant increase in concentration during the experimental period. For instance, the glycine level increased 4.9 and 2.1 times in whitefish and vendace eggs, respectively. Transfer of eggs to 8° accelerated the decrease of nearly all free amino acids before hatching. The consequence of such amino acid metabolism for newly hatched larvae is discussed.  相似文献

Copper(II) complexes of N2-octyl-(S)-phenylalaninamide (Noc-Phe-NH2), N2-dodecyl-(S)-phenylalaninamide (Ndo-Phe-NH2), and N2-octyl-(S)-norleucinamide (Noc-NLeu-NH2), dynamically adsorbed on a reversed-phase C18 column, were able to perform the direct enantiomeric separation of unmodified amino acids, amino acid amides and esters, hydroxy acids, and dipeptides by elution with aqueous or mixed aqueous-organic solutions containing copper(II) sulphate or acetate. The role played by several parameters in the separation procedure was examined with the copper(II) complex of Noc-Phe-NH2 [concentration of the copper(II) ion in the eluent, pH and eluent polarity, amount of adsorbed selector]. The separation was shown to occur entirely on the stationary phase. The mechanism of chiral discrimination is discussed in terms of the chromatographic parameters and of the structure of the copper(II) complexes in solution and in the solid state. The chiral stationary phase maintained its separation ability for about 3 months. However, the column could be easily restored by recovering the selector with methanol and repeating the loading procedure. © 1996 Wiley-Liss, Inc.  相似文献

In all the species examined, aspartic acid, glutamic acid, alanine, glycine and serine dominate. The two diastereoisomers, (2S,4R)-4-hydroxyglutamic acid and (2S,4S)-4-hydroxyglutamic acid, occur in characteristic concentration ratios in Phlox species. The second of these compounds is the principal free amino acid in the green parts of the plants. The presence of (2S,4R)-4-hydroxyglutamic acid in plants is reported for the first tiine. No other 4-substituted acidic amino acids were detected in the Phlox species analysed, although special attention was paid to the possible presence of 4-hydroxy-4-methylglutamic acids which have previously been reported in plants. It was found, however, that both diastereoisomers of (2S)-4-hydroxy-4-methylglutamic acid co-exist in Ledenbergia roseoaenea and also in Pandanus veitchii. Although the presence of 3,4-dihydroxyglutamic acids in green parts of Lepidium sativum and Rheum rhaponticum has been previously reported, we were not able to detect or isolate any of the possible diastereoisomers from the green parts or seeds of these plants. We did isolate glutathione which was found to have some properties in common with those reported for the dihydroxy compounds.  相似文献
