【背景】扎布耶湖位于青藏高原地区,是以水体中富含高浓度CO32−、HCO3和Na+为显著特性的盐碱湖,因其地理位置高寒、高海拔,人迹罕至,涉及该盐湖微生物的研究相对较少。【目的】系统探究湖水中细菌多样性和菌种资源等,发掘可利用的潜在菌种。【方法】采用Illumina测序16S rRNA基因V3−V4区分析扎布耶湖细菌的群落结构组成、物种多样性特征;采用纯培养筛选分离可培养细菌,明确分离菌株分类学地位、生长特性及产酸能力,同时测定分离菌株四氢嘧啶(ectoine)、吲哚乙酸(3-indoleacetic acid, IAA)和胞外聚合物(extracellular polymeric substances, EPS)的积聚量。【结果】Illumina测序明确分类地位的细菌有21门44纲86目583属,其中优势门类群是变形菌门(Proteobacteria, 25.11%−67.60%)、拟杆菌门(Bacteroidetes, 4.84%−35.02%)和厚壁菌门(Firmicutes, 1.24%−11.01%),优势属类群是克雷伯氏菌属(Klebsiella, 0.01%−9.53%)、盐单胞菌属(Halomonas, 0.54%−8.75%)、芽单胞菌属(Gemmatimonas, 2.39%−6.00%)和腈基降解菌属(Nitriliruptor, 1.27%−6.26%)。纯培养法筛选获得嗜盐碱菌38株,其中芽孢杆菌属(Bacillus) 23株(60.53%)和盐单胞菌属(Halomonas) 10株(26.32%),菌株均具有耐盐碱和产酸能力(19.80%−29.68%)。大多数菌株能积聚四氢嘧啶、IAA和EPS,产量范围分别为1.36−175.59、0.27−8.69和0.02−0.22 g/g。【结论】扎布耶湖细菌群落结构与其他地区盐碱湖相似,但存在大量未明确分类学地位的细菌,分离菌株多具有耐盐碱及产酸特性。此外,还发现4株高效生产四氢嘧啶、3株生产吲哚乙酸和3株生产胞外聚合物的潜力菌株,为扎布耶盐湖微生物资源的开发利用奠定了基础。  相似文献

【背景】北桑寄生(Loranthus tanakae)植物中黄酮类化合物成分含量较高,生物活性较好。由于植物资源短缺,导致其生物活性成分的研究受限。【目的】从北桑寄生中分离、筛选和鉴定产黄酮类化合物的内生菌,并评估其体外抗氧化和抑菌活性。【方法】以北桑寄生枝条为研究对象分离内生菌,通过显色反应和薄层色谱法(thin-layer chromatography,TLC)筛选产黄酮类化合物的内生菌,并采用NaNO2-Al (NO3)3比色法测定其总黄酮含量。利用清除自由基和抑菌的体外实验来初步评估产黄酮内生菌的生物活性。【结果】共筛选出4株产黄酮内生菌,其中3株为内生真菌,1株为内生细菌,通过形态学和分子生物学方法将其分别鉴定为Botryosphaeria sp.(ZC020)、Phoma sp.(ZZ105)、Nemania sp.(ZS042)和Pseudomonas sp.(ZC026)。4株菌中ZC020和ZS042的总黄酮含量较高,分别为(44.58±0.72) mg/L和(31.98±0.18) mg/L (P<0.05,n=3),并且ZS042可产生与北桑寄生植物相同类型的黄酮类化合物。ZC026和ZS042表现出优异的抗氧化活性,其1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picrylhydrazyl,DPPH)和2,2''-联氮-二(3-乙基苯并噻唑啉-6-磺酸)二铵盐[2,2''-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt,ABTS]自由基清除率分别为72.85%±0.66%(ZC026)、57.01%±0.89%(ZS042)和85.36%±0.75%(ZC026)、88.17%±0.15%(ZS042)(P<0.05,n=3)。ZZ105对革兰氏阳性菌和革兰氏阴性菌均有抑制作用。【结论】从北桑寄生枝条中分离出4株产黄酮类化合物的内生菌,其中有一株为内生细菌。这项工作为北桑寄生黄酮类化合物的生产提供了新的资源,也为其他珍稀药用植物和材料的开发和保护提供了参考。  相似文献

To evaluate the species specificity of Cd accumulation and the relationship of Cd with other essential metals and metallothionein (MT), the concentrations of Cd, Zn, Cu, and Fe in the liver and kidney and the MT concentrations in the soluble fractions of the liver and kidney were determined in Cd-uncontaminated nonhuman primates (11 species, 26 individuals) kept in a zoo and two wild-caught Japanese macaques. The compositions of metal-binding proteins in the soluble fractions were also investigated by high-performance liquid chromatography (HPLC). The hepatic Cd concentration was 0.03–14.0 μg/g and the renal Cd concentration was 0.35–99.0 μg/g, both varying greatly and being higher in nonhuman primates, which were more closely related to man. The hepatic Zn concentration was 24.0–176 μg/g and the renal Zn concentration was 13.5–138 μg/g, showing 7- to 10-fold differences, and a correlation (r=0.558, p<0.01) was found between renal Zn and renal Cd concentrations. It was proved that in the liver, MT is more closely correlated with Zn (r=0.795, p<0.001) than with Cd (r=0.492, p<0.01) and that in the kidney MT is correlated with both Cd (r=0.784, p<0.001) and Zn (r=0.742, p<0.001). HPLC analysis of metals bound to MT-like protein in chimpanzees, de Brazza’s monkeys, and Bolivian squirrel monkeys showed that more than 90% of Cd in both the liver and kidney, approx 40% of Zn in liver and 28–69% of Zn in kidney were bound to MT-like protein. Results are presented from experiments on the laser generation of X-ray radiation at the wavelength λ=469 ? (ε=26.4 eV) on the 3p(J=0)−3s(J=1) transition of Ne-like Ar ions. Experiments were carried out on the SIGNAL electrophysical facility with a 3.1-mm-diameter 157-mm-long Al2O3 ceramic capillary filled with argon at a pressure of 0.2–1.0 Torr. The discharge current amplitude was I ∼ 25–40 kA, the current rise rate being dI/dt ∼ 1012 A/s. By a vacuum X-ray diode tuned to detect X-ray photons with energies in the range 10–40 eV, laser pulses with a duration of t 1 ∼ 1 ns and maximum energy of E 1,max ∼ 1 μJ were recorded. The pulses were generated 35 ns after the discharge current was switched on. The line spectra in the wavelength range of 150–500 ? showed the bright λ=469 ? line. Changes in leaf physiology with tree age and size could alter forest growth, water yield, and carbon fluxes. We measured tree water flux (Q) for 14 ponderosa pine trees in two size classes (12 m tall and ∼40 years old, and 36 m tall and ∼ 290 years old) to determine if transpiration (E) and whole-tree conductance (g t) differed between the two sizes of trees. For both size classes, E was approximately equal to Q measured 2 m above the ground: Q was most highly correlated with current, not lagged, water vapor pressure deficit, and night Q was <12% of total daily flux. E for days 165–195 and 240–260 averaged 0.97 mmol m–2 (leaf area, projected) s–1 for the 12-m trees and 0.57 mmol m–2 (leaf area) s–1 for the 36-m trees. When photosynthetically active radiation (I P) exceeded the light saturation for photosynthesis in ponderosa pine (900 μmol m–2 (ground) s–1), differences in E were more pronounced: 2.4 mmol m–2 (leaf area) s–1 for the 12-m trees and 1.2 mmol m–2 s–1 for the 36-m trees, yielding g t of 140 mmol m–2 (leaf area) s–1 for the 12-m trees and 72 mmol m–2 s–1 for the 36-m trees. Extrapolated to forests with leaf area index =1, the 36-m trees would transpire 117 mm between 1 June and 31 August compared to 170 mm for the 12-m trees, a difference of 15% of average annual precipitation. Summary Protoplasts were isolated from leaves ofBetula platyphylla var.japonica using a 0.6M mannitol solution containing 1% Cellulase Onozuka R-10 and 1% Driselase. The cell division and colony formation were largely enhanced using Murashige and Skoog (1962) liquid medium at half strength (1/2 MS), containing 0.6M mannitol, 0.09M sucrose, and factorial combinations of 0.1–30 μM N-(2-chloro-4-pyridyl)-N′-phenylurea (4-pu) and 0.1–10 μM 1-naphthaleneacetic acid (NAA) or 0.1–30 μM 2,4-dichlorophenoxyacetic acid (2,4-D). The optimal protoplast density was 5–7 × 104/ml. Continuous callus proliferation from protoplasts was achieved by transferring colonies to fresh 1/2 MS agar medium containing 1 μM NAA and 1 μM 4-pu with no mannitol. It appeared that supplementation of the medium with phenylurea type cytokinin, 4-pu gave the successful callus proliferation from the protoplasts ofB. platyphylla.  相似文献

Purified β-glucosidase fromCellulomonas biazotea had an apparentK m andV for 2-nitrophenyl β-d-glucopyranoside (oNPG) of 0.416 mmol/L and 0.22 U/mg protein, respectively. The activation energy for the hydrolysis of pNPG of β-glucosidase was 65 kJ/mol. The inhibition by Mn2+ vs. oNPG of parental β-glucosidase was of mixed type with apparent inhibition constants of 0.19 and 0.60 μmol/L for the enzyme and enzyme-substrate complex, respectively. Ethanol at lower concentrations activated while at higher concentrations it inhibited the enzyme. The determination of apparent pK a’s at different temperatures and in the presence of 30 % dioxane indicated two carboxyl groups which control theV value. The thermal stability of β-glucosidase decreased in the presence of 10 % ethanol. The half-life of β-glucosidase in 1.75 mol/L urea at 35 °C was 145 min, as determined by 0–9 mol/L transverse urea gradient-PAGE. 为了解白头叶猴(Trachypithecus leucocephalus)的栖息地利用规律及其影响因素,2016年2月至2017年1月,采用瞬时扫描取样法对广西崇左白头叶猴国家级自然保护区一群白头叶猴的栖息地利用进行了研究。结果表明,白头叶猴对山体不同部位的利用存在显著性差异(χ2 =39.467,df=3,P<0.001),其中,对崖壁(56.75±9.55)%的利用比例最大,其次是对山坡(39.42±10.93)%和山顶(2.98±2.54)%的利用,而对山脚(0.84±1.47)%的利用频率最低。白头叶猴对不同微生境类型的利用存在差异(χ2=27.709,df=3,P<0.001),其中对乔木(49.37±12.31)%的利用比例最大,其次是裸岩(24.05±13.61)%,随后依次为藤本(15.48±8.01)%和灌木(10.87±5.45)%。白头叶猴主要在山坡上觅食,利用崖壁移动、休息,进行社会活动;主要利用裸岩进行社会活动,觅食、移动、休息主要发生在乔木上。从整体来看,白头叶猴在雨季对乔木的利用频率显著大于旱季(Z=-2.680,n=12,P=0.007);雨季在山坡觅食频率显著大于旱季(Z=-2.517,n=12,P=0.012),而在崖壁觅食频率刚好相反(Z=-2.842,n=12,P=0.004);白头叶猴雨季在乔木休息的频率显著大于旱季(Z=-2.355,n=12,P=0.019)。白头叶猴对栖息地的利用受到温度的影响。白头叶猴对乔木的总体利用频率随着平均温度的升高而增加(r=0.664,n=12,P=0.018);觅食时,对崖壁、裸岩的利用频率均与平均温度成负相关关系(崖壁:r=-0.685,n=12,P=0.014;裸岩:r=-0.600,n=12,P=0.039);休息时,对乔木的利用频率与平均温度呈正相关关系(r=0.650,n=12,P=0.022)。不同季节,白头叶猴对栖息地的利用方式不同。白头叶猴的栖息地利用模式可能是在觅食利益和捕食风险之间作出的权衡,并受到环境温度的影响。  相似文献

Previous studies showed low selenium (Se) concentrations in Belgian children. Serum α-tocopherol, retinol, total cholesterol, high-density lipoprotein and low-density lipoprotein cholesterol, selenium (Se), and thiobarbituric acid-reactive substances were examined. In order to obtain further information on the Se status in Belgian children, Se, α-tocopherol, retinol, and lipid concentrations were examined and signs of peroxidative lipid damage were evaluated in a subgroup. The study was performed in 524 children (0–14 yr old) during vaccination campaigns. Three age groups were analyzed: 0–1, 1–4, and 4–14 yr. In 87 of them, where sufficient amounts of serum were available, analysis of thiobarbituric acid-reactive substances was done. Infants have high serum α-tocopherol concentrations: (23.2 μmol/L [median and interquartile range: 18.6–30.2]) and low Se concentrations (0.37 mol/L [0.27–0.47]). Se concentrations rise significantly during the first 4 yr (p < 0.0001) (Mann-Whitney U-test, tied p-values): 0.70 μmol/L (0.59–0.82); in the 4–14 yr olds, it was 0.75 μmol/L (0.67–0.86). These values remain low compared to results coming from other parts of the world. α-Tocopherol concentrations decrease significantly after infancy (p < 0.0001). The ratio α-tocopherol/total cholesterol is higher in infants. This is induced by the high vitamin E content of infant formulas. Signs of serum lipid peroxidation could not be detected by analysis of serum malondialdehyde concentrations. High α-tocopherol concentrations, as those observed in infant serum lipids, could be one of the protective mechanisms from the peroxidative lipid damages, sometimes observed in a low-Se status.  相似文献

[目的]为了解决高温的煤化工废水生物脱氮效率不高的技术难题。[方法]本研究从上海某能化集团有限公司的煤化工废水处理系统的活性污泥中筛选得到一株耐热氨氧化细菌A1和一株耐热反硝化细菌D1。[结果]通过形态学观察、生理生化特征及16S rRNA基因序列分析,菌株A1初步鉴定为Aquamicrobium ahrensii,菌株D1初步鉴定为Pseudomonas stutzeri。采用单因子优化实验研究发现,菌株A1和D1的最适生长温度分别高达42℃和40℃。在模拟实际废水处理的初始NH4+-N浓度100 mg/L和42℃的条件下,构建了由菌株A1和D1 (W/W,20%/10%)组成的共培养物,探究该共培养物在不同pH和C/N对短程硝化反硝化脱氮及N2O的释放效应。结果表明,该共培养物在42℃、pH 9.0–10.0和初始C/N为2:1时,处理模拟废水的氮素去除率达>99.0%,最大N2O得率高达51.3%。[结论]本研究的结果可为高温煤化工废水的生物处理提供技术支撑及菌种储备,同时也为高温污水处理过程中N2O的释放规律提供理论参考。  相似文献

【背景】生物防治是“基于自然的解决方案”,有利于生态文明和可持续发展,开展生物防治技术研究的基础是明确菌株的生防作用和抑菌特性。【目的】探究杉木内生菌株T1-3-2的抑菌促生特性,为研制该菌株生防菌剂、防治杉木炭疽病(Cunninghamia lanceolata anthracnose)奠定基础。【方法】通过形态学特征、生理生化特性及16S rRNA基因序列分析,确定菌株T1-3-2的分类地位;通过平板对峙、菌落径向生长抑制率和平板倒扣等方法测定细菌及其挥发性气体和次级代谢产物的抑菌作用;同时,测定其促生作用和室内防效。【结果】菌株T1-3-2与桉生假单胞菌(Pseudomonas eucalypticola)亲缘性较近,属于假单胞菌属。该菌株对分属于6个属的10株靶标菌株具有较强的拮抗作用,尤其对炭疽菌属、拟盘多毛孢属、黑孢霉属和葡萄座腔菌属的6株靶标菌株抑制率高达80%以上。室内盆栽试验显示:菌株T1-3-2用Kings Medium B液体培养基的发酵菌液对杉木炭疽病的防效可达74.20%,同时能有效改善杉木幼苗的生长状况、增加生物量。【结论】菌株T1-3-2隶属于假单胞菌属,对杉木具有良好的抗病促生作用,是一株具有开发潜力的生防菌株。  相似文献

【背景】抗生素污染越来越引起人们的关注。利用微生物处理抗生素污染被认为是一种环境友好型的方法。【目的】筛选林可霉素高效降解菌并研究其降解机制。【方法】经形态学观察、生理生化鉴定和16S rRNA基因测序分析进行鉴定;通过PCR技术和质谱分析技术对该菌抗性基因和降解产物等进行分析。【结果】从林可霉素菌渣堆肥样本中获得一株高效降解林可霉素的假单胞菌(Pseudomonas RST-1),该菌在林可霉素浓度为3.0 g/L的牛肉膏蛋白胨培养基上培养40 h后,林可霉素降解率高达57.3%。该菌含有intI1、sul1、sul2等抗性基因,降解产物为去甲基林可霉素和2-丙基-N-甲基脯氨酸。【结论】菌株RST-1具有高效降解林可霉素的能力,推测可能的降解机制为去甲基化和酰胺键水解作用,该菌株降解特性及降解机制研究为林可霉素降解工程菌及其高效降解菌剂的研制奠定了基础。  相似文献

【背景】昆虫是世界上种类最多、肠道菌群资源最丰富且多样的动物类群之一。昆虫肠道微生物具有产生活性次级代谢产物的能力,是活性天然产物的重要来源。【目的】研究药用昆虫喙尾琵琶甲(Blaps rynchopetera)成虫肠道来源链霉菌(Streptomyces sp.) BPA71的次级代谢产物及其生物活性。【方法】利用正相硅胶柱色谱、葡聚糖凝胶Sephadex LH-20柱色谱等方法分离纯化该菌株的发酵粗提物,采用牛津杯法进行抗菌活性追踪,确定抗菌活性部位,通过ESI-MS、NMR等波谱数据分析对化合物结构进行鉴定,采用微量肉汤稀释法测定最低抑菌浓度(minimal inhibitory concentration, MIC),采用MTS法测定抗肿瘤活性。【结果】从Streptomyces sp. BPA71的固体发酵提取物中共分离得到4个已知化合物,通过对比核磁数据确定为糠酸甲酯(1)、吡咯甲酰胺A (2)、吡咯甲酰胺B (3)和吲哚-3-乙酸甲酯(4)。抗菌活性结果显示化合物2具有广谱抗菌活性。此外,化合物2对宫颈癌细胞HeLa、肺癌细胞A549、肝癌细胞SMMC-7721、乳腺癌细胞MDA-MB-231和结肠癌细胞SW480这5株肿瘤细胞均有明显的抑制活性。【结论】喙尾琵琶甲肠道来源Streptomyces sp. BPA71可产生丰富的生物活性物质,该研究结果为进一步挖掘喙尾琵琶甲肠道链霉菌的活性天然产物奠定了基础,同时丰富了人们对喙尾琵琶甲肠道微生物的认识。  相似文献

【目的】研究青海干旱地区蚕豆根瘤菌的遗传多样性,获得与蚕豆品种共生匹配且具有耐旱性的根瘤菌株,促进蚕豆耐旱根瘤菌在青海干旱地区生产中的应用。【方法】以分离自青海干旱地区一株菌株QHCD22为材料,利用细菌形态学、生理生化指标鉴定、Biolog细菌鉴定系统、16S rRNA基因序列分析、全基因组分析等进行菌种鉴定和系统发育分析,进一步通过PEG6000模拟干旱胁迫、盆栽回接干旱胁迫处理及旱作田间接种验证试验对该菌株的耐旱性进行综合评价。【结果】QHCD22菌株属快生型根瘤菌属(Rhizobium),Rhizobium indicum种。随着PEG6000模拟干旱胁迫程度的加剧,在−0.6 mPa这一更低渗透势时菌株存活数量增高,浊度由61.48%上升到69.42%,表现出较强的耐旱性。盆栽试验表明,接种根瘤菌处理(NA)的株高、植株鲜干比、根瘤数、根瘤鲜重、叶绿素含量(SPAD)、叶片相对含水量(RWC)、脯氨酸含量(PRO)、超氧化物歧化酶活性(SOD)、根系活力(TCC)均高于不接种根瘤菌处理(NN),并且在正常供水条件下,NA处理的各指标也均高于NN处理。旱作田间验证试验表明接种该菌株显著提高固氮酶活性,青海13号蚕豆根瘤固氮酶活性由不接种的42.07 C2H4 nmol/(g·h)显著增加到221.78 C2H4 nmol/(g·h),青蚕14号蚕豆由40.60 C2H4 nmol/(g·h)显著增加到109.78 C2H4 nmol/(g·h),马牙蚕豆由33.41 C2H4 nmol/(g·h)显著增加到643.15 C2H4 nmol/(g·h)。接种根瘤菌对于增加产量具有促进作用,其中青蚕14号的增产效果显著,增产幅度达32.3%。【结论】QHCD22菌株可能为快生型根瘤菌属的一个种Rhizobium indicum,具有一定的耐旱性,研究表明接种根瘤菌可以提高蚕豆的耐旱性,尤其对干旱敏感型蚕豆品种增产效果显著,具有潜在的应用前景。  相似文献

[背景] 1,3-二甲基-2-咪唑烷酮(1,3-Dimethyl-2-Imidazolidinone,DMI)作为一种强极性非质子溶剂,在生产和应用过程的环境中有稳定残留问题,存在安全隐患。[目的] 分离筛选具有降解DMI能力的微生物菌株,为清除环境中残留的DMI提供优良的微生物菌种资源。[方法] 从DMI生产区域土壤采集样品分离DMI抗性微生物,采用形态学及分子生物学鉴定确定其分类地位,并对DMI降解能力进行测定。[结果] 分离到最高能够耐受5%(体积分数) DMI的微生物菌株,形态学及分子生物学鉴定初步表明获得的菌株DT-1和DT-2为贝莱斯芽孢杆菌(Bacillus velezensis);全细胞及细胞提取液均具有降解DMI的能力;其中菌株DT-1及其细胞提取液对1%(体积分数) DMI的降解率分别达到48%和68%。[结论] 从DMI生产区域土壤中分离到具有DMI降解能力的芽孢杆菌,不但可为DMI污染的微生物治理提供优良微生物资源,而且扩展了人们对芽孢杆菌生物学功能的认识。  相似文献

【目的】探究化能自养硫氧化细菌Halothiobacillus sp. LS2介导的以乙炔为电子受体的厌氧硫氧化反应。【方法】稀释涂布法测定细胞生长情况,离子色谱仪测试硫氧化动力学中SO_4~(2–)和S_2O_3~(2–)以及基于相对荧光定量法的基因表达分析。【结果】尽管菌株LS2在以氧气为电子受体时的最大反应速率V_(max)更高,但在厌氧条件下且以乙炔为电子受体时,菌株LS2的生长量是氧气为电子受体时的2倍,且硫氧化酶基因soxB的表达量显著高于氧气作为电子受体时。【结论】菌株LS2不仅可以以乙炔为电子受体完成厌氧硫氧化反应,且这一代谢过程的产能效率较有氧硫氧化过程更高。本研究首次发现了微生物介导的以乙炔为电子受体的厌氧硫氧化反应,对丰富硫的生物地球化学循环理论有积极意义。  相似文献

[背景] 酪氨酸酶是黑色素合成过程中的关键酶,也是引起人体色素障碍性疾病和产生果蔬酶促褐变的主要原因。目前,酪氨酸酶抑制剂的开发已引起广泛关注,但一些酪氨酸酶抑制剂如熊果苷、曲酸等均存在一定的安全隐患。微生物资源丰富且具有许多优点,从微生物中寻找特异性强、高效的酪氨酸酶抑制剂已成为该领域研究的热点。[目的] 通过测定分离自新疆乌鲁木齐达坂城盐湖的盐水球菌Salinicoccus ventosaetal B2-3-5和B6-1-4代谢物提取物对酪氨酸酶活性的影响,比较2株菌发酵过程中代谢物的差异,了解所筛选菌株B2-3-5抑制酪氨酸酶活性的机制。[方法] 以曲酸为阳性对照分别测定B2-3-5和B6-1-4这2个菌株发酵产生的代谢物提取物对蘑菇酪氨酸酶的抑制活性;应用LC-MS代谢组学方法检测2株菌在相同条件下产生的所有代谢物质;采用单变量、多元变量、正交偏最小二乘判别分析(Orthogonal Partial Least Squares-Discrimination Analysis,OPLS-DA)法识别差异代谢物;利用层次聚类分析(Hierarchial Cluster Analysis,HCA)法对识别的差异物进行聚类分析;通过Kyoto Encyclopedia of Genes and Genomes (KEGG)代谢通路对比法分析这些差异代谢物主要参与的代谢途径。[结果] 菌株B2-3-5代谢物提取物对蘑菇酪氨酸酶二酚酶活性的抑制率为67%,其IC50为0.277 mg/mL,同属菌株B6-1-4代谢物提取物则对酪氨酸酶无抑制活性。采用代谢组学的检测方法从2株菌的代谢物中筛选出63个差异代谢物,其中氨基酸类化合物、维生素类化合物和羧酸类化合物的种类及相对含量均是B2-3-5菌株明显高于B6-1-4菌株。通过代谢途径分析发现这些差异代谢物主要参与15个代谢通路,其中维生素B6生物合成通路的影响较为显著。[结论] 推测B2-3-5菌株可能是通过增加一些氨基酸类、维生素类及羧酸类等小分子化合物的含量来抑制酪氨酸酶活性。维生素B6代谢途径的上调也表明菌体细胞可通过产生维生素B6与酪氨酸酶中的必需氨基作用或清除酶催化循环过程中产生的活性氧自由基(reactive oxygen species,ROS)来抑制酪氨酸酶活性。  相似文献

【背景】类诺卡氏菌(Nocardioides sp.) InS609-2是一株分离自南极洲罗斯海特拉诺瓦湾的恩克斯堡岛土壤中潜在的极地放线菌新种。尚无研究报道Nocardioides sp.InS609-2的全基因组序列,缺少对其功能基因、代谢产物合成途径及比较基因组学等的研究。【目的】解析Nocardioides sp.InS609-2的基因组序列信息,以深入挖掘次级代谢产物基因资源。【方法】利用Illumina HiSeq高通量测序平台对菌株InS609-2进行全基因组完成图测序,使用相关软件对测序数据进行基因组组装、基因预测和功能注释、预测次级代谢产物合成基因簇和共线性分析等。【结果】基因组最后得到的总长度为4 524 052 bp,G+C含量为69.42%,预测到4 656个基因、56个tRNA和6个rRNA。根据Nocardioides sp.InS609-2的全基因组测序结果,分别有3 761、3 052、1 767、4 134和2 725个基因在COG、GO、KEGG、NR和Swiss-Prot数据库中提取到注释信息。同时,还预测得到19个次级代谢产物合成基因簇。基因组测序数据提交至NCBI获得GenBank登录号为CP060034。Nocardioides sp.InS609-2与N. dokdonensis CP015079、N. yefusunii CP034929、N. euryhalodurans CP038267、N. seonyuensis CP038436、N. daphniae CP038462和N. okcheonensis CP087710这6株基因组同源性比较高的类诺卡氏菌进行共线性分析和蛋白聚类分类分析,得到的结果是7个基因组间既有保守性又各自有独特性。七个基因组共有44个蛋白聚类簇。最后进行16S rRNA基因系统发育树、泛基因组、core基因组和基因组进化树分析,进一步挖掘了Nocardioides sp.InS609-2的基因组信息。【结论】从基因组层面上预测了Nocardioides sp.InS609-2的次级代谢产物的生物合成基因簇,为InS609-2的后续相关研究提供了参考信息,具有重要意义。  相似文献
