An overview on 5α-reductase inhibitors

Benign prostatic hyperplasia (BPH) is the noncancerous proliferation of the prostate gland associated with benign prostatic obstruction and lower urinary tract symptoms (LUTS) such as frequency, hesitancy, urgency, etc. Its prevalence increases with age affecting around 70% by the age of 70 years. High activity of 5α-reductase enzyme in humans results in excessive dihydrotestosterone levels in peripheral tissues and hence suppression of androgen action by 5α-reductase inhibitors is a logical treatment for BPH as they inhibit the conversion of testosterone to dihydrotestosterone. Finasteride (13) was the first steroidal 5α-reductase inhibitor approved by U.S. Food and Drug Administration (USFDA). In human it decreases the prostatic DHT level by 70-90% and reduces the prostatic size. Dutasteride (27) another related analogue has been approved in 2002. Unlike Finasteride, Dutasteride is a competitive inhibitor of both 5α-reductase type I and type II isozymes, reduced DHT levels >90% following 1 year of oral administration. A number of classes of non-steroidal inhibitors of 5α-reductase have also been synthesized generally by removing one or more rings from the azasteroidal structure or by an early non-steroidal lead (ONO-3805) (261). In this review all categories of inhibitors of 5α-reductase have been covered.     

An α-fucosidase pseudogene on human chromosome 2

Summary In Chinese hamster-human hybrids with overlapping translocations, the major site of hybridization of a cDNA clone for the liver form of human -l-fucosidase was 1p36.31p34, consistent with hybridization to the FUCA1 locus. No hybridization to the FUCA2 locus on chromosome 6 was observed. Hybridization to a genomic sequence on chromosome 2 was, however, detected, thus defining a new FUCA-like locus. The restriction map of the -fucosidase cDNA could be exactly superimposed upon its region of homology within a genomic clone containing this FUCA-like locus, suggesting that it is a processed pseudogene.     

Can rats prey on gull eggs? An experimental approach

Black rat Rattus rattus populations can reach high densities on the Mediterranean islands, as has been the case on the Chafarinas Islands (Western Mediterranean coast) in the last decade. This archipelago holds the second largest breeding population of Audouin's gull Larus audouinii and an important population of yellow-legged gull Larus cachinnans. Circumstantial evidences of rat predation upon Audouin's gull eggs led to conservation concerns about the impact of rat predation on gull productivity. We studied the impact of rats on gull nests experimentally by building artificial nests into the breeding area of yellow-legged gulls. Two kinds of eggs were placed in the artificial nests: manipulated eggs, in which the shell had been previously opened, and intact (unmanipulated) eggs. Rats preyed extensively on opened eggs but only once on intact eggs, apparently due to lack of skills to prey on intact eggs, suggesting that rats do not normally have access to fresh gull eggs. Rats inhabiting breeding areas of gulls can scavenge on broken eggs rolled out of the nest or on eggs with softened shells after nest desertion, but it is unlikely that they prey on natural gull nests guarded by parents. We suggest that introduced black rats on Mediterranean islands can only marginally affect the productivity of gulls owing to egg predation.
