Theory for the evolution of modifiers of the rate of mutation suggests that a lower rate of mutation may evolve after the breakdown of mechanisms that enforce outcrossing. Mutation accumulation (MA) experiments were conducted to compare deleterious mutation parameters in two closely related species of the plant genus Amsinckia, a group that exhibits wide variation in the mating system. One of the two species studied (A. douglasiana) is predominantly outcrossed in natural populations, where as the other species (A. gloriosa) is predominantly self-pollinated. Progeny assays of flower number per plant from generation 1 lines (control) and generation 11 lines (MA treatment) were conducted in both species. Dry weight measurements of progeny from the control and MA treatment in A. douglasiana also were made. Estimation of mutation parameters was conducted using maximum likelihood under the assumption of a gamma distribution of mutational effects. The two species exhibited similar rates and effects of deleterious mutation affecting flower number. Estimates of mutation rate for dry weight in A. douglasiana are close to those for flower number. Overall, the estimates of mutation parameters observed in these species are intermediate within the range reported for fitness components in other eukaryotes. The results are discussed within the context of evolutionary change in deleterious mutation accompanying mating system evolution and with respect to previous estimates of mutation parameters based on assays of inbreeding depression and the assumption of mutation-selection equilibrium.  相似文献

Repeated efforts to estimate the genomic deleterious mutation rate per generation (U) in Drosophila melanogaster have yielded inconsistent estimates ranging from 0.01 to nearly 1. We carried out a mutation-accumulation experiment with a cryopreserved control population in hopes of resolving some of the uncertainties raised by these estimates. Mutation accumulation (MA) was carried out by brother sister mating of 150 sublines derived from two inbred lines. Fitness was measured under conditions chosen to mimic the ancestral laboratory environment of these genotypes. We monitored the insertions of a transposable element, copia, that proved to accumulate at the unusually high rate of 0.24 per genome per generation in one of our MA lines. Mutational variance in fitness increased at a rate consistent with previous studies, yielding a mutational coefficient of variation greater than 3%. The performance of the cryopreserved control relative to the MA lines was inconsistent, so estimates of mutation rate by the Bateman-Mukai method are suspect. Taken at face value, these data suggest a modest decline in fitness of about 0.3% per generation. The element number of copia was a significant predictor of fitness within generations; on average, insertions caused a 0.76% loss in fitness, although the confidence limits on this estimate are wide.  相似文献

Summary Depletion of stratospheric ozone and the resulting increase in ultraviolet-B (UV-B) radiation may negatively impact the productivity of terrestrial ecosystems. This concern has led to a number of studies that report the influence of supplementing UV-B radiation on plant growth and development. However, only two of these field studies have included tree species and both were singleseason experiments. In this study, loblolly pine (Pinus taeda L.) from seven seed sources was grown under natural and supplemental levels of UV-B radiation. Irradiation treatments were continued for three seasons on plants from four of the seven groups and for 1 year only for three groups. The supplemental irradiances simulated those that would be anticipated with stratospheric ozone reductions of 16% and 25% over Beltsville, Md. The effects of UV-B radiation during the 1st year on plant growth varied among the seed sources. The growth of plants from two of the seven seed sources tested showed significant reductions following a single irradiation season and plants from one group tended to be larger under increased UV-B radiation. However, after 3 years of supplemental irradiation, plant biomass was reduced in all four groups by 12–20% at the highest simulated ozone depletion. These results suggest that the effects of UV-B radiation may accumulate in trees and that increased UV-B radiation could significantly reduce the growth of loblolly pine over its lifetime. However, they also point to a need for multiple season research in any analysis of potential consequences of global change on the long-term growth of trees.  相似文献

Appropriate controls in outdoor UV-B supplementation experiments   总被引:7,自引:0,他引:7  
Quercus robur L. saplings were exposed in an outdoor experiment to supplemental levels of UV-8 (280–315 nm) radiation using treatment arrays of cellulose diacetate-filtered fluorescent lamps that also produce UV-A (315–400 nm) radiation. Saplings were also exposed to UV-A radiation alone using control arrays of the same lamps filtered with polyester and to ambient levels of radiation, using arrays of unenergized lamps. The UV-B treatment was modulated to maintain a 30% elevation above the ambient level of UV-B radiation, measured by a broad-band sensor weighted with an erythemal action spectrum. Saplings exposed to UV-B radiation beneath treatment arrays developed thicker leaves than those beneath ambient and control arrays. Despite the fact that supplemental levels of UV-A radiation were only a small percentage of ambient levels, apparent UV-A effects were also recorded. Significant increases in sapling height, lammas shoot length and herbivory by chewing insects were observed under treatment and control arrays, relative to ambient, but there were no differences between the responses of saplings under treatment and control. These data imply that supplemental UV-A radiation or other effects associated with energised lamps can significantly affect plant growth parameters and herbivory in outdoor studies. We conclude that the results from current outdoor UV-B supplementation experiments that lack control exposures using polyester-filtered lamps need to be interpreted with caution and that future supplementation experiments should include appropriate controls.  相似文献

The genetic variability for RAPDs band pattern was studied in a set of 157 mutation accumulation (MA) lines of Drosophila melanogaster. These MA lines were derived from the same isogenic base population and subsequently maintained by full-sib mating during 132 generations. The ancestral pattern of the original isogenic base can be unambiguously established as the consensus pattern of the MA lines and, because these lines are expected to be homozygous, dominance for band pattern is not a concern. Only repeatable changes in band pattern were considered. The number of ancestral bands detected implies that nine-nucleotide targets are enough for repeatable PCR amplification. Compared with the ancestral pattern, one MA line lost one band and two MA lines gained a new one. These results can be accounted for by the insertion of transposable elements occurring at a rate 0.07 < i < 0.21 per whole haploid genome and generation. This range is typical for Drosophila and consistent with the previously observed mobility for the roo family, supporting the generality of previous estimates of spontaneous mutation rates for morphological and fitness traits based on these MA lines. The sequence of one of the new bands suggests that the Idefix family is also active in the lines.  相似文献

We simulated single-generation data for a fitness trait in mutation-accumulation (MA) experiments, and we compared three methods of analysis. Bateman-Mukai (BM) and maximum likelihood (ML) need information on both the MA lines and control lines, while minimum distance (MD) can be applied with or without the control. Both MD and ML assume gamma-distributed mutational effects. ML estimates of the rate of deleterious mutation had larger mean square error (MSE) than MD or BM had due to large outliers. MD estimates obtained by ignoring the mean decline observed from comparison to a control are often better than those obtained using that information. When effects are simulated using the gamma distribution, reducing the precision with which the trait is assayed increases the probability of obtaining no ML or MD estimates but causes no appreciable increase of the MSE. When the residual errors for the means of the simulated lines are sampled from the empirical distribution in a MA experiment, instead of from a normal one, the MSEs of BM, ML, and MD are practically unaffected. When the simulated gamma distribution accounts for a high rate of mild deleterious mutation, BM detects only approximately 30% of the true deleterious mutation rate, while MD or ML detects substantially larger fractions. To test the robustness of the methods, we also added a high rate of common contaminant mutations with constant mild deleterious effect to a low rate of mutations with gamma-distributed deleterious effects and moderate average. In that case, BM detects roughly the same fraction as before, regardless of the precision of the assay, while ML fails to provide estimates. However, MD estimates are obtained by ignoring the control information, detecting approximately 70% of the total mutation rate when the mean of the lines is assayed with good precision, but only 15% for low-precision assays. Contaminant mutations with only tiny deleterious effects could not be detected with acceptable accuracy by any of the above methods.  相似文献

The effect of 250 generations of mutation accumulation (MA) on the second chromosome competitive viability of Drosophila melanogaster was analyzed both in homozygous and heterozygous conditions. We used full-sib MA lines, where selection hampers the accumulation of severely deleterious mutations but is ineffective against mildly deleterious ones. A large control population was simultaneously evaluated. Competitive viability scores, unaffected by the expression of mutations in heterozygosis, were obtained relative to a Cy/L(2) genotype. The rate of decline in mean DeltaM approximately 0.1% was small. However, that of increase in variance DeltaV approximately 0.08 x 10(-3) was similar to the values obtained in previous experiments when severely deleterious mutations were excluded. The corresponding estimates of the mutation rate lambda > or = 0.01 and the average effect of mutations E(s) < or = 0.08 are in good agreement with Bateman-Mukai and minimum distance estimates for noncompetitive viability obtained from the same MA lines after 105 generations. Thus, competitive and noncompetitive viability show similar mutational properties. The regression estimate of the degree of dominance for mild-to-moderate deleterious mutations was approximately 0.3, suggesting that the pertinent value for new unselected mutations should be somewhat smaller.  相似文献

During the last few decades many experiments have been performed to evaluate the responses of plants to enhanced solar UV-B radiation (280–320 nm) that may occur because of stratospheric ozone depletion; most of them were performed in controlled environment conditions where plants were exposed to low photosynthetically active radiation (PAR) levels and high UV-B irradiance. Since environmental radiative regimes can play a role in the response of plants to UV-B enhancement, it appears doubtful whether it is valid to extrapolate the results from these experiments to plants grown in natural conditions. The objective of this work was to evaluate the effects on physiology and morphology of a bean (Phaseolus vulgaris L.) cultivar Nano Bobis, exposed to supplemental UV radiation in the open-air. UV-B radiation was supplied by fluorescent lamps to simulate a 20% stratospheric ozone reduction. Three groups of plants were grown: control (no supplemental UV), UV-A treatment (supplementation in the UV-A band) and UV-B treatment (supplemental UV-B and UV-A radiation). Each group was replicated three times. After 33 days of treatment plants grown under UV-B treatment had lower biomass, leaf area and reduced leaf elongation compared to UV-A treatment. No significant differences were detected in photosynthetic parameters, photosynthetic pigments and UV-B absorbing compounds among the three groups of plants. However, plants exposed to UV-A treatment showed a sort of 'stimulation' of their growth when compared to the control. The results of this experiment showed that plants may be sensitive to UV-A radiation, thus it is difficult to evaluate the specific effects of UV-B (280–320 nm) radiation from fluorescent lamps and it is important to choose the appropriate control. Environmental conditions strongly affect plant response to UV radiation so further field studies are necessary to assess the interaction between UV-B exposure and meteorological variability.  相似文献

Intensified ultraviolet-B radiation or UV-B (wavelengths between 280 and 320 nm) can delay flowering and diminish lifetime flower production in a few plants. Here we studied the effects of enhanced UV-B on floral traits crucial to pollination and pollinator reproduction. We observed simultaneous flowering responses of a new crop plant, Limnanthes alba (Limnathaceae), and a wildflower, Phacelia campanularia (Hydrophyllaceae), to five lifetime UV-B dosages ranging between 2.74 and 15.93 kJ·m·d. Floral traits known to link plant pollination with bee host preference, host fidelity and larval development were measured. Intensified UV-B had no overall effect on nectar and pollen production of L. alba and P. campanularia flowers. A quadratic relationship between UV-B and nectar sugar production occurred in P. campanularia and showed that even subambient UV-B dosages can be deleterious for a floral trait. Other floral responses to UV-B were more dramatic and idiosyncratic. As UV-B dosage increased, L. alba plants were less likely to flower, but suffered no delays in flowering or reductions to lifetime flower production for those that did flower. Conversely, an equal proportion of P. campanularia plants flowered under all UV-B treatments, but these same plants experienced delayed onset to bloom and produced fewer flowers at greater UV-B intensities. Therefore, intensified UV-B elicits idiosyncratic responses in flowering phenology and flower production from these two annual plants. Diurnal patterns in nectar and pollen production strongly coincided with fluctuating humidity and only weakly with UV-B dosage. Overall, our results indicated that intensified UV-B can alter some flowering traits that impinge upon plant competition for pollinator services, as well as plant and pollinator reproductive success.  相似文献

We conducted three experiments to examine the influence of ultraviolet-B radiation (UV-B; 280–320 nm) exposure on reproduction in Brassica rapa (Brassicaceae). Plants were grown in a greenhouse under three biologically effective UV-B levels that simulated either an ambient stratospheric ozone level (control), 16% (“low enhanced”), or 32% (“high enhanced”) ozone depletion levels at Morgantown, WV, USA in mid-March. In the first experiment, we examined whether UV-B level during plant growth influenced in vivo pollen production and viability, and flower production. Pollen production and viability per flower were reduced by ≈50% under both enhanced UV-B levels relative to ambient controls. While plants under high-enhanced UV-B produced over 40% more flowers than plants under the two lower UV-B treatments, whole-plant production of viable pollen was reduced under high-enhanced UV-B to 17% of that of ambient controls. Whole-plant production of viable pollen was reduced under low-enhanced UV-B to 34% of ambient controls. In the second experiment, we collected pollen from plants under the three UV-B levels and examined whether source-plant UV-B exposure influenced in vitro pollen germination and viability. Pollen from plants under both enhanced-UV-B treatments had initially lower germination and viability than pollen from the ambient level. After in vitro exposure to the high-enhanced UV-B levels for 6 h, viability of the pollen from plants grown under ambient UV-B was reduced from 65 to 18%. In contrast, viability of the pollen from plants grown under both enhanced UV-B treatments was reduced to a much lesser extent: only from ≈43 to 22%. Thus, ambient source-plant pollen was more sensitive to enhanced UV-B exposure. In the third experiment, we used pollen collected from source plants under the three UV-B levels to fertilize plants growing under ambient-UV-B levels, and assessed subsequent seed production and germination. Seed abortion rates were higher in plants pollinated with pollen from the enhanced UV-B treatments, than from ambient UV-B. Despite this, seed yield (number and mass) per plant was similar, regardless of the UV-B exposure of their pollen source. Our findings demonstrate that enhanced UV-B levels associated with springtime ozone depletion events have the capacity to substantially reduce viable pollen production, and could ultimately reduce reproductive success of B. rapa.  相似文献

Aims Information about how species respond to extreme environments, such as high UV-B radiation, is very useful in estimating natural ecosystem structure and functions in alpine areas. Our aim is to examine the effect of enhanced UV-B radiation on the fitness of an alpine meadow annual species on Qinghai-Tibet Plateau.Methods Plants of Cerastium glomeratum Thuill. were exposed to ambient (control) or ambient plus supplemental UV-B radiation (enhanced), simulating a 9% ozone depletion over Gannan, China (102°53′E, 34°55′N, 2900 m in altitude), up to leaf senescence and fruit maturation. Plant height, flower phenology, biomass allocation and reproductive parameters of the species were measured.Important findings Plant height in C. glomeratum was reduced by enhanced UV-B radiation at early growth stages and compensated with ongoing development. Fruit biomass, aboveground biomass, total biomass and reproductive effort (fruit dry mass/aboveground biomass) were not affected by enhanced UV-B radiation, but a significant increase in root/shoot ratio was found. Enhanced UV-B radiation delayed onset of flowering by 1 day and shortened duration of flowering by 5 days in C. glomeratum. But because of the long period of flowering time (83–88 days), this did not make any significant effect on flower number, seed number, pollination success (number of seeds per fruit) or reproductive success (fruit to flower ratio) in C. glomeratum. Enhanced UV-B radiation had no effect on seed germination and seed mass either. And the high production and low germination rate of the seed might be the strategy of C. glomeratum to survive the extreme environments on alpine meadow. All these results showed that C. glomeratum was tolerant to enhanced UV-B radiation.  相似文献

Soybean, Glycine max (L.) Merr. cv Essex, plants were grown in the field in a 2 × 2 factorial design, under ambient and supplemental levels of ultraviolet-B (UV-B) radiation (supplemental daily dose of 5.1 effective kilojoules per square meter) and were either well-watered or subjected to drought. Soil water potentials were reduced to −2.0 megapascals by the exclusion of natural precipitation in the drought plots and were maintained at approximately −0.5 megapascal by supplemental irrigation in well-watered plots. Plant growth and gas exchange characteristics were affected under both drought and supplemental UV-B radiation. Whole-leaf gas exchange analysis indicated that stomatal limitations on photosynthesis were only significantly affected by the combination of UV-B radiation and drought but substrate (ribulose bisphosphate) regeneration limitations were observed under either stress. The combined effect of both drought and UV-B radiation on photosynthetic gas exchange was a reduction in apparent quantum efficiency and the rapid appearance of biochemical limitations to photosynthesis concomitant with reduced diffusional limitations. However, the combination of stresses did not result in additive effects on total plant growth or seed yield compared to reductions under either stress independently.  相似文献

Very few studies have evaluated the effects of UV-B radiation on trees. especially deciduous species. In this study we evaluate the effects of supplemental UV-B radiation on the growth and photosynthetic capacity of sweetgum (Liquidambar styraciflua L.). Sweetgum seedlings were grown for 2 years in the field under either ambient or supplemental UV-B radiation. Artificial UV-B radiation was supplied by fluorescent lamps at a maximum daily supplementation of either 3.1 or 5.0 kJ of biologically effective UV-B radiation. Over the 2-year period, supplemental UV-B radiation had little effect on total plant biomass or photosynthetic capacity. However, subtle changes in leaf physiology, carbon allocation, and growth were observed. Supplemental UV-B radiation reduced photosynthetic capacity only during the first year, while leaf area and biomass were reduced in the second year. Alterations in carbon allocation included an increase in branch number and root to shoot ratio. While these data do not indicate that the productivity of sweetgum would likely be compromised by an increase in solar UV-B radiation, they do suggest that the UV-B portion of the solar spectrum contributes to the regulation of sweetgum growth and development. Therefore the possibility of significant consequences to sweetgum due to possible increases in UV-B radiation cannot be ruled out.  相似文献

Little is known about the role of transposable element (TE) insertion in the production of mutations with mild effects on fitness, the class of mutations thought to be central to the evolution of many basic features of natural populations. We propagated mutation-accumulation (MA) lines of two RNAi-deficient strains of Caenorhabditis elegans that exhibit germline transposition. We show here that the impact of TE activity was to raise the level of mildly deleterious mutation by 2- to 8.5-fold, as estimated from fecundity, longevity, and body length measurements, compared to that observed in a parallel MA experiment with a control strain characterized by a lack of germline transposition. Despite this increase, the rate of mildly deleterious mutation was between one and two orders of magnitude lower than the rate of TE accumulation, which was approximately two new insertions per genome per generation. This study suggests that high rates of TE activity do not necessarily translate into high rates of detectable nonlethal mutation.  相似文献

Spring wheat (Triticum aestivum) was grown in the field under ambient and supplemental levels of ultraviolet-B (UV-B, 280–315 nm) radiation to determine the potential for alteration in plant nutrients, decomposition, leaf quality and dry matter yield. Supplemental UV-B radiation simulating a 12, 20 and 25% stratospheric ozone depletion significantly decreased dry matter yield, but had no significant impact on harvest index. UV-B radiation resulted in an increase of the concentrations of N and K in all plant parts; changes of the concentrations of P, Mg, Fe and Zn varied in a tissue-dependent manner, as the decrease of P in leaves and stems, and its increase in spikes and grains. The mass of N, P, K, Mg, Fe and Zn in various plant parts and whole plant was generally decreased except leaf N mass was increased by enhanced UV-B radiation. Enhanced UV-B radiation decreased the concentrations of soluble carbohydrates in leaves and increased that of holocellulose and soluble proteins. After 60 and 100 days of decomposition of leaves and stems in the field, enhanced UV-B radiation stimulated the loss of organic C. As a consequence, the nutrient content of soils might be less diminished under enhanced UV-B radiation.  相似文献

Two soybean [ Glycine max (L.) Merr.] cultivars, Essex and Williams, were grown in the field for 6 consecutive seasons under ambient and supplemental levels of ultravio-Set-B radiation to determine the potential for alterations in yield or seed quality with a reduction in the stratospheric ozone column. The supplemental UV-B fluences simulated a 16 or 25% ozone depletion. The data presented here represent the first field experiment conducted over multiple seasons which assesses the effects of increased UV-B radiation on seed yield. Overall, the cultivar Essex was found to be sensitive to UV-B radiation (yield reductions of 20%) while the cultivar Williams was tolerant. However, the effectiveness of UV-B radiation in altering yield was strongly influenced by the seasonal microclimate, and the 2 cultivars responded differently to these changing factors. Yield was reduced most in Essex during seasons in which water availability was high and was reduced in Williams only when water was severely limiting. The results of these experiments demonstrate the necessity for multiple-year experiments and the need to increase our understanding of the interaction between UV-B radiation and other environmental stresses in order to assess the potential consequences of stratospheric ozone depletion.  相似文献

The combined effects of additional UV-B radiation and artificial wounding on leaf phenolics were studied in a short term field experiment with the drought semi-deciduous Mediterranean shrub Phlomis fruticosa L. The seedlings were grown under ambient or ambient plus supplemental UV-B radiation (biologically equivalent to a 15% ozone depletion over Patras, 38.3° N, 29.1° E) for 7 months before wounding. Unexpectedly, supplemental UV-B radiation decreased leaf phenolics. Subsequently, wounding was effected by removing leaf discs from some of the plants, while the rest remained intact and served as controls. Wounding significantly increased phenolics of the wounded leaves and the increase was more pronounced under supplemental UV-B radiation. In addition, wounding had a significant positive effect on the phenolics of the opposite, intact leaf, but only under additional UV-B radiation. We conclude that UV-B radiation, wounding and their combination may affect the chemical defensive potential of Phlomis fruticosa. In addition, increased levels of phenolics after herbivore attack under field conditions may afford extra protection against enhanced UV-B radiation levels.  相似文献

Seedlings of the Mediterranean shrub Cistus creticus L. were grown in the field under ambient or ambient plus supplemental UV-B radiation (simulating a 15% ozone depletion over Patras, 38.3°W, 29.1°E) for 20 months. During this period, measurements of photosynthetic capacity, photochemical efficiency of PS II, chlorophylls and carotenoids were performed once per season. Supplemental UV-B radiation had no significant effect on these parameters nor on the total, above ground biomass accumulation, plant height and leaf specific mass measured at plant harvest. It was observed, however, that UV-B supplementation increased the number of seeds per fruit as well as mean individual seed mass. As a result, seed number and total seed mass per plant were considerably increased. Germination rates of produced seeds were not affected. We may conclude that C. creticus is a UV-B resistant plant whose competitive ability may be improved by enhanced UV-B radiation through an increase in its reproductive effort and a higher contribution to the seed bank.  相似文献

The fitness effects of spontaneous mutations in Caenorhabditis elegans   总被引:1,自引:0,他引:1  
Abstract. Spontaneous mutation to mildly deleterious alleles has emerged as a potentially unifying component of a variety of observations in evolutionary genetics and molecular evolution. However, the biological significance of hypotheses based on mildly deleterious mutation depends critically on the rate at which new mutations arise and on their average effects. A long-term mutation-accumulation experiment with replicate lines of the nematode Caenorhabditis elegans maintained by single-progeny descent indicates that recurrent spontaneous mutation causes approximately 0.1% decline in fitness per generation, which is about an order of magnitude less than that suggested by previous studies with Drosophila . Two rather different approaches, Bateman-Mukai and maximum likelihood, suggest that this observation, along with the observed rate of increase in the variance of fitness among lines, is consistent with a genomic deleterious mutation rate for fitness of approximately 0.03 per generation and with an average homozygous effect of approximately 12%. The distribution of mutational effects for fitness appears to have a relatively low coefficient of variation, being no more extreme than expected for a negative exponential, and for one composite fitness measure (total progeny production) approaches constancy of effects. These results are derived from assays in a benign environment. At stressful temperatures, estimates of the genomic deleterious mutation rate (for genes expressed at such temperatures) is sixfold lower, whereas those for the average homozygous effect is approximately eightfold higher. Our results are reasonably compatible with existing estimates for flies, when one considers the differences between these species in the number of germ-line cell divisions per generation and the magnitude of transposable element activity.  相似文献
