The division of labor between the different worker castes of leaf-cutting ants may reflect in their capacity to exchange liquids by trophallaxis. The crop capacity of and trophallactic exchanges between different size classes of worker leaf-cutting ants of the sub-species Acromyrmex subterraneus subterraneus were investigated. Size classes were defined from head capsule widths and crop capacity of each class was determined following ad libitum feeding on dye solution. Experiments were carried out to investigate trophallactic exchanges between donor ants and recipient ants of each class size combination on a one to one basis. An experiment was also performed to investigate dye distribution within mini-colonies following introduction of three classes of donor ants. Worker ants were categorized into four size classes from their head capsule widths (C1 = 0.8-1.0 mm; C2 = 1.2-1.5 mm; C3 = 1.6-2.0 mm; C4 = 2.1-2.4 mm). C1 ants crop capacity was 0.13 μL; C2: 0.21 μL; C3: 0.52 μL; C4: 1.03 μL. Ants of each class previously fed on the dye solution (donors) were placed individually with an unfed ant of each class (recipients) and the presence of dye solution, passed from the donor to the recipient by oral trophallaxis was observed after 1 h. Results showed that all classes of donor ants performed trophallactic exchanges with all recipient classes. However, statistically fewer exchanges were seen for C2 donor ants when placed with C3 recipient ants. Ten donor ants of each of three classes (C2, C3 and C4) were introduced into mini-colonies without queen ants. It was observed that C1 and C2 ants were poor recipients, whilst C3 and C4 received the highest percentages of dye. Within 10 h of introducing the donor ants, 14 to 20% of their nest-mates had received dye solution, with 58 to 77% of dye passed to recipients. These studies show the altruistic nature of “food-laden” leaf-cutters and indicate that ants involved in garden maintenance activity are less likely to receive liquids from foraging workers.  相似文献

Stomodeal trophallaxis plays a major role in ant colony nutrition and communication. While the rate of food distribution at the individual level (worker to worker) is rapid, factors affecting the rate of food distribution at the colony level remain poorly understood. We used the odorous house ant, Tapinoma sessile (Say), as a model species to investigate the factors affecting the rate of spread of liquid carbohydrate food throughout a colony. To track the movement of the food we used protein marking and double antibody sandwich enzyme-linked immunosorbent assay, DAS-ELISA. Increasing colony size while keeping the number of donor workers constant significantly decreased the number of individuals testing positive for the marker. After 8 h of trophallactic interactions with ten donors, 92 ± 5% of recipient workers tested positive in a colony of 125 and 38 ± 5% tested positive in a colony of 1,000. Interestingly, as colony size increased and the percentage of workers testing positive decreased, the proportion of workers actually receiving food increased. Food originating from a single donor fed approximately 12 individuals in colonies comprised of 125 recipients and approximately 38 individuals in colonies comprised of 1,000 recipients. Thus, the per capita consumption of food decreased as colony size increased, most likely because the amount of food reaching the colony was limited. Increasing the number of donors while keeping colony size constant significantly increased the number of recipient ants testing positive for the marker. As the number of donor workers doubled, the percentage of recipients testing positive more than doubled suggesting that the number of individuals receiving food increases with increasing colony size, while the per capita amount of food decreases. When food was available ad libitum and in close proximity to the nest, numerous workers fed directly at the food source. This dramatically increased the rate and the extent of food distribution to both the workers and the queens and colony size had no significant effect on the spread of the marker in the workers or the queens. The rate and the extent of food distribution at the colony level may depend on a number of factors including the number of successful foragers, the size and density of the recipient colony, and the recipient caste.  相似文献

In social insects, interactions among individuals are important in colony organisation because they can be used in decision making. During trophallaxis in ants, antennal and foreleg contacts between both partners are established. It has been suggested that a modulatory communication channel could be involved in such contacts, but it remains undemonstrated. The aim of this work was to find variables plausible to be encoded in such contacts and quantify the consequent changes in the tactile stimulation the food-donor ant receives. We recorded nectar transference between pairs of workers in experimental arenas once one of them had returned from collecting sucrose solution (15 or 40%w/w), with different situations of colony’s sugar deprivation. The frequency of antennal strokes that the food-donor ant received on her head depended on both the colony’s sugar-deprivation and the concentration, the latter showed differences within 3–5 deprivation days. Antennal and foreleg movements of the food-receiver increase with increasing level of colony’s carbohydrate deprivation, as well as with increasing concentration of the transferred food. Not only does this study reopen an interesting question, but it gives evidence that variables related to the appetitive context are indeed encoded in the tactile stimulation during the trophallaxis as well. Consequently, they have the effective potentiality to play a communicational role in the organization of colony activities. Summary Kin recognition and nepotism between honeybee workers (Apis mellifera L.) was analysed in a trophallactic bio-assay. Donor workers were fed dyed sugar syrup and introduced into a recipient group consisting of 12 to 15 workers of the same colony. After allowing for 1 hour of trophallaxis, the distribution of the dyed food was analysed with spectrophotometry. The subfamily composition in the recipient group was varied such that the donor bees had to discriminate between workers of 2 to 7 different patrilines. Donor bees preferentially fed super sisters if few patrilines were present in the recipient group. However, preferential feeding was not observed if the recipient group consisted of workers of more than three subfamilies. Since the natural degree of polyandry causes intracolonial genetic variance to exceed the genetic variability in the experiments, nepotistic behaviour among workers may not reveal intranidal subfamily recognition in honeybees.  相似文献

Protoplasts from cell suspensions of young-embryo-derived calli, which were nonregenerable for long-term subculture and protoplasts from embryogenic calli with the regeneration capacity of 75% of the same wheat Jinan 177, were mixed as recipient. Protoplasts from embryogenic calli of Avena sativa (with the regeneration capacity of less than 10%) irradiated with UV at an intensity of 300 μW/cm2 for 30 s, 1 min, 2 min, 3 min, 5 min were used as the donor. Protoplasts of the recipient and the donor were fused by PEG method. Many calli and normal green plants were regenerated at high frequency, and were verified as somatic hybrids by chromosome counting, isozyme, 5S rDNA spacer sequence analysis and GISH (genomic in situ hybridization). Fusion combination between protoplasts either from the cell suspensions or from the calli and UV-treated Avena sativa protoplasts could not regenerate green plants.  相似文献

Somatic embryos were induced on roots excised from in vitro plants of Prunus avium× pseudocerasus `Colt'. On medium containing 6-benzylamino purine (BAP, 1.5 μM) and 2,4-dichlorophenoxyacetic acid (2,4-D, 15 μM), a mean of 25 (s.e. ± 2.0) somatic embryos were produced on intact root systems and 15 (s.e. ± 1.7) on roots systems cut into 10 mm pieces. Most somatic embryos were formed directly on intact roots and indirectly (from callus) on sectioned roots. A mean of 2.5 (s.e. ± 0.25) secondary embryos per primary embryo were formed directly on primary embryos after they were transferred to medium containing BAP (1.5 μM), indole-3-butyric acid (10 μM) and 2,4-D (5 μM). After transfer to a medium containing BAP (2 μM) and gibberellic acid (GA3, 3 μM), shoots developed in 75% (s.e. ± 7.3) of the embryos. Somatic embryos were not induced on explants of in vitro roots or shoots of P. avium, and were induced infrequently on zygotic embryos, although a wide range of media were tested. Possible reasons for the contrasting embryogenic ability of `Colt' and P. avium are discussed. This revised version was published online in June 2006 with corrections to the Cover Date.  相似文献

Paired exocrine metapleural glands are present in almost all ants and produce compounds with antibiotic properties towards a variety of pathogenic fungi and bacteria. In Acromyrmex leaf-cutting ants, small workers have relatively large metapleural glands compared to large workers, and thus harbour approximately half the number of gland cells of large workers, despite being only one-fifteenth their body mass. Here we present results showing that when the two worker castes of A. echinatior are treated with spores of the pathogenic fungus Aspergillus nomius in doses that correspond to the difference in metapleural gland cell numbers they do not differ in survival. However, we also show, for the first time, that small workers survive significantly longer than large workers when both are challenged with a dose of spores that corresponds to their difference in body mass. Furthermore, the time until Aspergillus nomius hyphae and spores appear on the cadavers of workers dead from infection, is significantly increased in the small worker caste. In addition to supporting previous findings that the metapleural glands have an important defence function, the results of this study indicate that the relatively large glands in small workers makes this caste particularly well adapted to preventing pathogenic microorganisms from entering the colony. For many plants, the number of pollen genotypes deposited on a flower’s stigma is positively related to fruit maturation and seed number; however, the mechanisms underlying this effect are less understood. Here we examined whether diversity of pollen (1, 3, or 5 donors) affects reproductive success in self-incompatible apple (Malus × domestica). Using paternity analysis, we then assessed the siring rate of individual donors to test whether diversity effects are related to the presence of a superior pollen donor or to donor × donor interactions. Increasing the diversity of compatible pollen enhanced seed number and reduced seed abortion in some but not all recipient genotypes. This effect was associated with an increased number of sires per fruit and non-random siring success among pollen donors. Two donors had consistently high siring rates, regardless of recipient genotype; however, the siring success of pollen donors was not correlated with their siring success in single-donor pollinations. Rather, siring success was affected by the identity of other pollen genotypes present on the stigma. Our results therefore suggest that the effects of pollen diversity are variable but may enhance fecundity by fostering interactions between pollen donors.  相似文献

One hundred sixty-four adult male volunteers (29 controls [Group 1] and 135 combi drivers) enrolled in the study. The combi drivers were divided into three groups as nonusers of either Maras powder or cigarette (Group 2), smokers (Group 3), and users of Maras powder (Group 4). Blood lead levels (BLLs) were analyzed by atomic absorption spectrophotometer. BLL was detected as 2.8 ± 2.3 μg/dL in Group 1 (n = 29); however, it was 3.5 ± 1.6 μg/dL in Group 2 (n = 33), 3.8 ± 2.4 μg/dL in Group 3 (n = 62), and 3.9 ± 2.4 μg/dL in Group 4 (n = 40). BLL in Group 1 was found significantly lower than other groups (p < 0.05). The use of cigarette or Maras powder by the drivers did not give rise to a marked difference on the BLLs (p > 0.05). BLL of (combi) drivers was detected to be significantly higher than nondrivers; however, it was still under the hazardous level of 10 μg/dL announced by WHO. Although there are publications reporting that usage of tobacco increases the level of lead in blood, both smoking and use of Maras powder did not affect BLL markedly in our study. Previous evidence indicates that the recognition of the nectar delivered by forager honeybees within the colony may have been a primitive method of communication on food resources. Thus, the association between scent and reward that nectar foragers establish while they collect on a given flower species should be retrieved during trophallaxis, i.e., the transfer of liquid food by mouth, and, accordingly, foraging experience could affect the occurrence of these interactions inside the nest. We used experimental arenas to analyze how crop scents carried by donor bees affect trophallaxis among foragers, i.e., donors and receivers, which differ in their foraging experience. Results showed that whenever the foragers had collected unscented sugar solution from a feeder the presence of scents in the solution carried by donors did not affect the occurrence of trophallaxis nor its dynamics. In contrast, whenever the foragers had previous olfactory information, new scents present in the crop of the donors negatively affected the occurrence, but not the dynamics of trophallaxis. Thus, the association learned at the food source seems to be retrieved during trophallaxis, and it is possible that known scents present in the mouthparts of nest-mates may operate as a triggering stimulus to elicit trophallactic behavior within the hive.  相似文献

The larvae of leaf-cutting ants are maintained within the fungus gardens of their colonies and are fed pieces of fungus by the adult workers. However, little else is known about the nature of the worker-larva interaction in these ecologically important ants. To examine whether workers can gauge the needs of individual larvae, we isolated larvae without adult workers for different lengths of time. We then placed workers with the larvae and recorded the type and frequency of the subsequent behaviours of the workers. Workers scraped the mouthparts of larvae, ingested their faecal fluid, fed them with fungal hyphae, transported them around the fungus garden and, most frequently, licked their bodies. The workers were also observed to ‘plant’ fungal hyphae on the bodies of larvae. Workers interacted more frequently with larvae that had been isolated without workers than with those that had not, but there was no effect of the length of isolation. The results suggest that the interactions are complex, involving a number of behaviours that probably serve different functions, and that workers are to some extent able to assess the individual needs of larvae. The development of stem callus mediated plant regeneration system for Withania somnifera is described. Maximum callus proliferation was obtained on Murashige and Skoog medium supplemented with 2.26 μM 2,4-D. Three-week-old, white, friable callus was used for shoot regeneration. The maximum shoot regeneration (6.2 ± 0.34 shoots/explant) was achieved in four weeks when callus was cultured on MS medium fortified with 4.44 μM BA and 0.57 μM IAA. Regenerated shoots were excised and multiplied (8.4 ± 0.43 shoots/explant) on MS medium supplemented with 4.44 μM of BA. Multiple shoots were divided into single shoots and were rooted (5.1 ± 0.49 rootlets/shoot) on half strength MS medium supplemented with 9.84 μM of IBA. Conventional control of leaf‐cutting ants is mainly carried out by application of solid insecticide baits. However, alternative approaches could increase the efficiency of control methods. Here, we investigated the hypothesis that liquid and semi‐solid baits are more readily ingested by leaf‐cutting ants and are therefore more toxic than solid baits. Furthermore, following increased ingestion rates, ants could perform higher frequencies of trophallactic exchanges with their nest mates, thus increasing dispersal rates within the colony. Each of three diets were offered to Acromyrmex subterraneus subterraneus (Forel) and Atta sexdens rubropilosa Forel (both Hymenoptera: Formicidae) for 1 h under laboratory conditions and subsequently crop volumes were quantified. The highest crop volumes for both species were seen when the diet was offered in liquid form, and the lowest crop volumes were observed after offering solid diets. Survival rates of the ants were monitored following addition of the neonicotinoid insecticide imidacloprid (IMI) to the three diets. The ingestion of liquid and semi‐solid diets + IMI by A. subterraneus resulted in 17 and 6% survival, respectively, whereas these diets caused 100% mortality of A. sexdens. Ingestion of solid diets with IMI resulted in 51% survival of A. subterraneus and 23% survival of Atta workers. Twenty‐three percent of A. subterraneus which had fed on liquid diet carried out trophallactic exchanges, whereas only 10 and 3% of ants carried out trophallaxis when fed on semi‐solid or solid diet, respectively. Lower trophallactic frequencies were observed for A. sexdens, with 13 and 3% of ants that had fed on liquid and semi‐solid diets performing trophallaxis, respectively. The low trophallactic exchange rates following ingestion of solid diets would reduce the dispersal of these compounds throughout the colony. Control strategies using solid baits should be re‐examined in the light of these findings, which indicate the importance of optimizing insecticide ingestion.  相似文献

This investigation was undertaken to evaluate practical feasibility of site specific pulmonary delivery of liposomal encapsulated Dapsone (DS) dry powder inhaler for prolonged drug retention in lungs as an effective alternative in prevention of Pneumocystis carinii pneumonia (PCP) associated with immunocompromised patients. DS encapsulated liposomes were prepared by thin film evaporation technique and resultant liposomal dispersion was passed through high pressure homogenizer. DS nano-liposomes (NLs) were separated by ultra centrifugation and characterized. NLs were dispersed in phosphate buffer saline (PBS) pH 7.4 containing different carriers like lactose, sucrose, and hydrolyzed gelatin, and 15% l-leucine as antiadherent. The resultant dispersion was spray dried and spray dried formulation were characterized to ascertain its performance. In vitro pulmonary deposition was assessed using Andersen Cascade Impactor as per USP. NLs were found to have average size of 137 ± 15 nm, 95.17 ± 3.43% drug entrapment, and zeta potential of 0.8314 ± 0.0827 mV. Hydrolyzed gelatin based formulation was found to have low density, good flowability, particle size of 7.9 ± 1.1 μm, maximum fine particle fraction (FPF) of 75.6 ± 1.6%, mean mass aerodynamic diameter (MMAD) 2.2 ± 0.1 μm, and geometric standard deviation (GSD) 2.3 ± 0.1. Developed formulations were found to have in vitro prolonged drug release up to 16 h, and obeys Higuchi's Controlled Release model. The investigation provides a practical approach for direct delivery of DS encapsulated in NLs for site specific controlled and prolonged release behavior at the site of action and hence, may play a promising role in prevention of PCP.  相似文献

A survivorship model was developed for UV-B irradiated Catla catla (17 days) larvae with the help of Kaplan and Meier Product-Limit (PL) method. Larvae were exposed to UV-B radiation (145 μW cm−2) for three different exposure times: 5, 10 and 15 min on every other day. The mean survival time of fish was calculated for each treatment using uncensored and censored survival data during 74 days study period. The mean uncensored and censored survival data for the 5-min exposed fish were 7 and 43, respectively. In 10-min exposure period, the uncensored and censored survival data were 19 and 31, respectively. During maximum exposure of 15 min, the uncensored survival data was 20 and censored data was 30. The mean survival time of fish calculated using PL estimate in 5, 10 and 15-min exposure treatments were 69.61 ± 0.50, 65.25 ± 0.96 and 60.60 ± 1.55 days, respectively. The mean survival time showed a decreasing trend with the increase of exposure period. The survival time was significantly (P < 0.001) higher in 5-min exposure treatment than others. This is clear from the present study that the exposure of UV-B radiation affects the survival rate of surface feeder catla larvae.  相似文献

This study was aimed at determining the optimal depth and photon irradiance for growth of Sargassum fulvellum. Sampling and measurement of underwater irradiance were carried out at farms cultivating S. fulvellum at Wando, southwestern coast of Korea, from May 2004 to April 2005. Growth of thalli, underwater irradiance and photosynthetic quantum yield were measured over a range of depths for three culture stages. During their nursery cultivation stage (Stage I), length increase was greatest at 1.5 m depth (2.5 ± 0.2 cm), where the average midday irradiance over 28 days was 488 ± 58 μmol photons m−2 s−1. During the pre-main cultivation stage (Stage II), the greatest length increase occurred at 1 m depth (10.9 ± 0.1 cm) with an average irradiance of 845 ± 169 μmol photons m−2 s−1. For the main cultivation stage (Stage III) of the alga, thalli showed maximal length growth in March and early April at depths of 1–2 m and 3 m. These results suggest that growth at each cultivation stage of S. fulvellum could be controlled by depth of cultivation rope. Summary Plants were suecessfully régenerated via somatic embryos from 3-yr-old cell suspension cultures of Medicago truncatula Gaertin. cv. Jemalong line M9-10a. The cultures were originally initiated from callus induced in well-expanded leaflets of 30 d in vitro-grown plants, Suspension cultures were established in stirred-liquid Murashige and Skoog (MS) basal salts and vitamins supplemented with 2.3 μM 2.4-dichlorophenoxyacetic acid (2,4-D) and 2.3 μM kinetin (Kin) and subeultured weekly. Somatic embryogenesis induction step was conducted in liquid MS medium containing 0.45 μM 2,4-D and 0.91 μM zeatin (Zea), during 1,2, and 3wk after subculture. Induced and non-induced cultures were transferred to solid embryo proliferation medium [EPM-MS basal salts and vitamins solidified with 0.2% (w/v) gelrite]. Somatic embryos developed until the late torpedo/dicotyledonary stages. We found that the best condition for the development of somatic embryos was achieved when suspension cultures were not subjected to the induction step. Induction of 1 and 2 wk led to a decrease in the recovery of somatic embryos and the 3-wk treatment resulted in no differentiation of somatic embryos. Plant regeneration was obtained in all conditions (except for 3wk induction) when embryos were transferred to an embryo conversion medium [ECM, similar to EPM but solidified with 0.7% (w/v) agar]. Embryo conversion rates were 54.5±1.6, 52.5±18.5, and 41.6±8.4% for 0, 1, and 2 wk induction treatments, respectively. These plants were successfully transferred to the greenhouse where they matured and produced seeds.  相似文献

Yeasts and filamentous fungi carried by the gynes of leaf-cutting ants   总被引:1,自引:1,他引:0  
Insect-associated microbes exhibit a wide range of interactions with their hosts. One example of such interactions is the insect-driven dispersal of microorganisms, which plays an essential role in the ecology of several microbes. To study dispersal of microorganisms by leaf-cutting ants (Formicidae: Attini), we applied culture-dependent methods to identify the filamentous fungi and yeasts found in two different body parts of leaf-cutting ant gynes: the exoskeleton and the infrabuccal pocket. The gynes use the latter structure to store a pellet of the ants’ symbiotic fungus during nest founding. Many filamentous fungi (n = 142) and yeasts (n = 19) were isolated from the gynes’ exoskeleton. In contrast, only seven filamentous fungi and three yeasts isolates were recovered from the infrabuccal pellets, suggesting an efficient mechanism utilized by the gynes to prevent contamination of the symbiotic fungus inoculum. The genus Cladosporium prevailed (78%) among filamentous fungi whereas Aureobasidium, Candida and Cryptococcus prevailed among yeasts associated with gynes. Interestingly, Escovopsis, a specialized fungal pathogen of the leaf-cutting ant-fungus symbiosis, was not isolated from the body parts or from infrabuccal pellets of any gynes sampled. Our results suggest that gynes of the leaf-cutter ants Atta laevigata and A. capiguara do not vertically transmit any particular species of yeasts or filamentous fungi during the foundation of a new nest. Instead, fungi found in association with gynes have a cosmopolitan distribution, suggesting they are probably acquired from the environment and passively dispersed during nest foundation. The possible role of these fungi for the attine ant–microbial symbiosis is discussed.  相似文献

作为蚁播植物种子的重要传播者,蚂蚁不但取食种子上附着的油质体,也喜食其它富含蛋白质、脂类、糖和维生素等的食物,因此环境中其它可利用食物的存在可能会影响蚂蚁对种子的搬运进而影响种子散布,但目前对于这种影响是如何发生的仍不清楚。在野外研究了蚂蚁对小花宽瓣黄堇(Corydalis giraldii Fedde)种子、肉、蜂蜜、苹果、馒头等食物的趋性和偏好程度,以及添加食物后蚂蚁对种子的拜访频率和搬运效率,以揭示其它可利用食物如何影响蚂蚁觅食和取食偏好,进而影响小花宽瓣黄堇种子散布。结果显示,在所诱捕的8种蚂蚁中,玉米毛蚁(Lasius alienus(Foerster))和丝光蚁(Formica fusca Linnaeus)是小花宽瓣黄堇种子的主要搬运者,不同食物诱捕的玉米毛蚁数量无显著性差异(P0.05),但蜂蜜和苹果诱捕的丝光蚁数量均显著大于种子(P0.05)。玉米毛蚁和丝光蚁均为杂食性,在觅食中分别行使群体募集和简单协作性募集。在仅有种子的对照处理中,玉米毛蚁和丝光蚁对种子的拜访频率分别为(38.73±4.57)头和(30.8±2.87)头(40min,n=15),两种蚂蚁对种子的拜访频率差异不显著(P0.05);玉米毛蚁和丝光蚁搬运种子的效率分别为(33.87±4.22)粒和(16.27±3.35)粒(40min,n=15),玉米毛蚁的搬运效率显著高于丝光蚁(P0.05)。与对照相比,添加馒头、苹果和蜂蜜后丝光蚁对种子的拜访频率显著降低(P0.05),分别为(15.6±3.61)头、(9.07±1.4)头和(7.67±1.58)头(40min,n=15);添加苹果和蜂蜜后丝光蚁对种子的搬运效率显著降低(P0.05),分别为(3.47±1.17)粒和(2.87±0.9)粒(40min,n=15);添加不同食物后玉米毛蚁对种子的拜访频率和搬运效率均无显著变化(P0.05)。研究结果表明行使群体募集的玉米毛蚁比行使简单协作募集的丝光蚁有更高的种子搬运效率,添加食物后影响丝光蚁对种子的拜访频率和搬运效率,这说明其它可利用食物对小花宽瓣黄堇种子散布的影响与搬运蚂蚁的种类及其觅食的募集方式有关。研究结果可为进一步研究蚂蚁与植物(种子)间的互利共生关系及其影响因素提供资料。  相似文献

Summary An efficient protocol for in vitro propagation of the valuable medicinal plant, Wasabia japonica (Miq.) Matsumura is described through shoot tip proliferation and direct regeneration. Multiple shoots were induced from shoort tips cultured on Murashige and Skoog (MS) semi-solid medium containing various concentrations (0.5–50 μM) of N6-benzyladenine (BA), thidiazuron, kinetin, and zeatin. A comparison was made on shoot multiplication between semi-solid and liquid culture media. Well-developed shoots were obtained using full-strength MS semi-solid medium containing 5.0 μM BA. However, the greatest shoot proliferation was achieved on either full- or half-strength MS liquid media supplemented with 5.0 μM BA for 4 wk (15.3±0.9 and 15.0±0.7 shoots per explant, respectively), and on half-strength MS liquid medium for 6 wk (25.8±1.3 shoots per explant) in culture. In contrast, the maximum number of shoots per explant on full-strength MS semi-solid medium was achieved with either 5.0 μM BA (10.4±0.6 shoots per explant) or 10.0 μM kinetin (10.9±0.8 shoots per explant). Fresh weight of explants and length of shoots derived from full-strength MS liquid medium (1055±77 mg and 34.2±1.0 mm, respectively) were significantly higher than those derived from full-strength MS semisolid medium (437.6±17.3 mg and 15.4±0.7 mm, respectively). Quarter-strength MS liquid medium had no significant difference in shoot proliferation when compared to quarter-strength MS semi-solid medium. Elongated shoots were separated and rooted on half-strength MS semi-solid media fortified with 1-naphthaleneacetic acid (NAA), indole-3-butyric acid (IBA), or indole-3-acetic acid (IAA) ranging from 0.1 to 10.0 μM. Root formation was greatest with IBA when compared with IAA and NAA. One hundred percent of shoots were rooted on half-strength MS medium with 5.0 μM IBA, while vigorous roots were obtained with 10.0 μM IBA. Micropropagated plantlets were successfully established in soil with 95% survival rate after heardening.  相似文献
