Changes in the amounts of several components of the photosyntheticelectron-transport system during greening of etiolated barleyleaves were studied on a "per plastid" basis. P700 and QA, whichwere initially absent from etioplasts, appeared 2 h after thestart of illumination in complete complexes of PS I and PS II,respectively. From 6 h, they increased rapidly in amount witha constant stoichiometric ratio of 1:1. Amounts of Cyt f, Cytb6, Cyt b-559 and FeS, initially present in etioplasts at levelsthat were one-third to half of those in mature chloroplasts,also increased rapidly after 6 h of illumination. The molarratio of Cyt f, Cyt b6 and Cyt b-559 was the same in etioplastsand in mature chloroplasts, namely 1:2:2. After 4 h of illumination,levels of FeS increased at nearly the same rate as those ofthe PS I complex. The increase in levels of all components wasmarked after 6 h of illumination, probably due to the energysupplied by developing plastids that had just become photosyntheticallycompetent. PSII, the oxygen-evolving complex of photosynthetic organisms,contains an intriguingly large number of low molecular weightproteins. PsbX, one of these proteins, is ubiquitous in PSIIcomplexes of cyanobacteria and plants. In previous studies,deletion of the PsbX protein in cyanobacteria has not resultedin clear phenotypic changes. Here we report the constructionof an antisense (AS-PsbX) line in Arabidopsis thaliana with<10% of wild-type PsbX levels. AS-PsbX plants are capableof photoautotrophic growth, but biochemical, biophysical andimmunological evidence demonstrates that reduction of PsbX contentsleads to reduced levels of functional assembled PSII core complexes,while the light-harvesting antennae are not affected. In addition,levels of phosphorylation of the core proteins D1, D2 and CP43are severely reduced in the antisense plants relative to theirwild-type counterparts. We conclude that PsbX is important foraccumulation of functional PSII.  相似文献

Photosystem stoichiometry adjustments in Chlamydomonas reinhardtiiwere induced upon a sudden shift in the light quality duringcell growth. Reversible changes in the PSI/PSII ratio were acompensation response to changes in the balance of light absorptionby the two photosystems. Quantitations of PSII, Cyt b6-f complexand PSI revealed a constancy in the cellular content of PSIIand the Cyt b6-f complex, and variable amounts of PSI in C.reinhardtii. These results strengthen the notion that PSI isthe thyla-koid component subject to chromatic regulation andresponsible for the adjustment and optimization of the PSI/PSII ratio in the thylakoid of oxygenic photosynthesis. Additionalresults, obtained upon the use of protein biosynthesis translationinhibitors (chloramphenicol and cyclohex-imide), suggested thata chromatically-induced lowering of the PSI/PSII ratio in C.reinhardtii occurs by suppression of de novo biosynthesis ofPSI components and, therefore, by dilution of the PSI complexin the thylakoid membrane, rather than by active degradationof assembled PSI in chlo-roplasts. In C4 plants, bundle sheath (BS) chloroplasts are arranged inthe centripetal position or in the centrifugal position, althoughmesophyll (M) chloroplasts are evenly distributed along cellmembranes. To examine the molecular mechanism for the intracellulardisposition of these chloroplasts, we observed the distributionof actin filaments in BS and M cells of the C4 plants fingermillet (Eleusine coracana) and maize (Zea mays) using immunofluorescence.Fine actin filaments encircled chloroplasts in both cell types,and an actin network was observed adjacent to plasma membranes.The intracellular disposition of both chloroplasts in fingermillet was disrupted by centrifugal force but recovered within2 h in the dark. Actin filaments remained associated with chloroplastsduring recovery. We also examined the effects of inhibitorson the rearrangement of chloroplasts. Inhibitors of actin polymerization,myosin-based activities and cytosolic protein synthesis blockedmigration of chloroplasts. In contrast, a microtubule-depolymerizingdrug had no effect. These results show that C4 plants possessa mechanism for keeping chloroplasts in the home position whichis dependent on the actomyosin system and cytosolic proteinsynthesis but not tubulin or light.  相似文献

A small fraction of low potential Cyt b-559, amounting to only13% of total Cyt b-559 in spinach chloroplasts, is analyzedwith the help of a highly selective, computer-controlled spectrophotometer,which simultaneously applies 16 pulse modulated narrow bandmeasuring beams with wavelengths in the cytochrome -band (500–600nm) for recordings of time resolved difference spectra. ThisCyt b-559 fraction remains oxidized upon dark incubation withascorbate and is reduced upon illumination. It can be reducedby cyclic PSI in an antimycin A-sensitive reaction or in thecourse of antimycin A-insensitive linear electron transportvia the Cyt b6/f complex. Reduction by NADPH in the dark requiresferredoxin. Simultaneous recordings of Cyt b-563 and Cyt f revealclose kinetic connection between this Cyt b-559 fraction andthe low potential chain of the Cyt b6/f complex. These resultsconfirm and extend previous observations of Miyake et al. 1995(Plant Cell Physiol. 36: 743) in maize mesophyll thylakoids,which led to the hypothesis that Cyt b-559 (Fd) occupies theposition of the postulated ferredoxin-plastoquinone reductase(FQR) in cyclic electron transport. A desiccation-tolerant cyanobacterium, Nostoc commune, showsunique responses to dehydration. These responses are: (i) lossof PSII activity in parallel with the loss of photosynthesis;(ii) loss of PSI activity; and (iii) dissipation of light energyabsorbed by pigment–protein complexes. In this study,the deactivation of PSII is shown to be important in avoidingphotoinhibition when the Calvin–Benson cycle is repressedby dehydration. Furthermore, our evidence suggests that dissipationof light energy absorbed by PSII blocks photoinhibition understrong light in dehydrated states.  相似文献

Stability of thylakoid components under supra-high irradiancewas studied with the cyanophyte Synechocystis PCC 6714. Theactivity of overall photosynthesis was quickly inactivated (T1/2=20min) under supra-high irradiance (300 W m–2, white light).In parallel with the inactivation of photosynthesis, QA in PSII was also inactivated. Both inactivations were acceleratedby chloramphenicol (CAP) addition. The reactivation of PS IIrequired weak irradiation and was suppressed by CAP. However,PS I measured as P700 was very stable. The level of PS I measuredas P700 was not significantly reduced by the irradiation for12 h even in the presence of CAP while the level of Cyt b559,component of PS II, was decreased markedly. The function ofPS I before and after supra-high irradiation with CAP was examinedby comparing sizes of P700 oxidation induced by a short flash,by a continuous light, and by determination of O2-and ferredoxin-reduction.No difference was observed in PS I actions before and afterthe irradiation treatment. These results indicate that the PSI complex is very tolerant of supra-high irradiation. However,the cells grown under supra-high irradiance contained much fewerPS I and PS II complexes than Cyt b6–f complexes. Theformer levels were reduced to a half to one fourth of thosebefore growth while the level of Cyt b6–f complex wasnot reduced so much. The effect of the Cyt b6-f redox state on the PSI formationwas examined with the cyanophyte Synechocystis PCC 6714 by usinga Q-cycle inhibitor, HQNO (2-n-heptyl-4-hydroxyquinoline N-oxide).HQNO inhibited the rapid reduction of flash-oxidized Cyt f,the reaction correlating with the stimulation of PSI formation,on one hand, and accumulated reduced Cyt b6, on the other, indicatingthat the electron flow in the Q-cycle correlates with regulationof PSI synthesis. HQNO also inhibited the stimulation of PSIformation under PSII light, resulting in a low PSI/PSII ratioeven under PSII light, while the PSI formation under PSI lightwas not suppressed by HQNO. Simultaneous inhibition of Cyt b6oxidation through the Q-cycle and the stimulated PSI formationby HQNO suggests that an HQNO-sensitive Cyt b6 oxidation isinvolved in the mechanism of monitoring the state of electrontransport system for regulation of PSI formation. Light-induced changes in stoichiometry among three thylakoidcomponents, PS I, PS II and Cyt b6-f complexes, were studiedwith the cyanophyte Synechocystis PCC 6714. Special attentionwas paid to two aspects of the stoichiometric change; first,a comparison of the patterns of regulation in response to differencesin light-intensity with those induced by differences in light-quality,and second, the relationship between regulation of the stoichiometryand the steady state of the electron transport system. Resultsfor the former indicated that (1) the abundance of PS I on aper cell basis was reduced under white light at the intensityas high as that for light-saturation of photosynthesis, butPS I per cell was increased under low light-intensity, (2) PSII and Cyt b6-f complexes remained fairly constant, and (3)changes in the abundance of PS I depended strictly on proteinsynthesis. The pattern was identical with that of chromaticregulation. For the second problem, the redox steady-statesof Cyt f and P700 under white light of various intensities weredetermined by flash-spectroscopy. Results indicated that (1)Cyt f and P700 in cells grown under low light-intensity [highratio of PS I to PS II (PS I/PS II)] were markedly oxidizedwhen the cells were exposed to high light-intensity, while theyremained in the reduced state under low light-intensity. (2)After a decrease in the abundance of PS I, most of P700 remainedin the reduced state even under high light-intensity, whilethe level of reduced Cyt f remained low. (3) Both Cyt f andP700 in cells of low PS I/PS II were fully reduced under lowlight-intensity, and Cyt f reduction following the flash wasrapid, which indicates that the turnover of PS I limits theoverall rate of electron flow. After an increase in the abundanceof PS I, the electron transport recovered from the biased state.(4) The redox steady-state of the Cyt b6-f complex correlatedwell with the regulation of PS I/PS II while the state of thePQ pool did not. Based on these results, a working model ofthe regulation of assembly of the PS I complex, in which theredox steady-state of the Cyt b6-f complex is closely relatedto the primary signal, is proposed. We analyzed the chlorophyll fluorescence parameters in a 3Dcellular arrangement in vivo by using a modified Nipkow disk-typeconfocal laser scanning microscope (CLSM). We first definedthe 3D values of PSII (photochemical yield of PSII) and NPQ(non-photochemical quenching) in mesophyll, epidermal and guardcell chloroplasts from the leaf surface to several tens of micronsin depth. We also used this CLSM method to analyze the relationshipsbetween actinic light intensity and the chlorophyll fluorescenceparameters for Boston fern and broad bean leaf specimens. Asthe actinic light intensity increased, the mean PSII valuesdecreased and the NPQ values increased in all chloroplasts ofBoston fern and broad bean leaf. These values differed withcell type and species. The Boston fern chloroplasts had lowerPSII values than the broad bean chloroplasts, and vice versafor the NPQ values. The PSII values of Boston fern chloroplastsdecreased in the order mesophyll, epidermal and guard cell chloroplasts.The NPQ values decreased in the order guard cell, mesophylland epidermal chloroplasts, except at 12 µmol m–2s–1 actinic light, when the mesophyll value was slightlylower than that of the epidermis. The trend in the PSII andNPQ values of broad bean mesophyll and guard cell chloroplastswas opposite to that of Boston fern chloroplasts. As 3D CLSMcan provide the PSII and NPQ values of each chloroplast in a3D cellular arrangement, this method has potential for investigatingdifferences in the functions of chloroplasts in vivo.  相似文献

Some plant species show constant rates of respiration and photosynthesismeasured at their respective growth temperatures (temperaturehomeostasis), whereas others do not. However, it is unclearwhat species show such temperature homeostasis and what factorsaffect the temperature homeostasis. To analyze the inherentability of plants to acclimate respiration and photosynthesisto different growth temperatures, we examined 11 herbace-ouscrops with different cold tolerance. Leaf respiration (Rarea)and photosynthetic rate (Parea) under high light at 360 µll–1 CO2 concentrations were measured in plants grown at15 and 30°C. Cold-tolerant species showed a greater extentof temperature homeostasis of both Rarea and Parea than cold-sensitivespecies. The underlying mechanisms which caused differencesin the extent of temperature homeostasis were examined. Theextent of temperature homeostasis of Parea was not determinedby differences in leaf mass and nitrogen content per leaf area,but by differences in photosynthetic nitrogen use efficiency(PNUE). Moreover, differences in PNUE were due to differencesin the maximum catalytic rate of Rubisco, Rubisco contents andamounts of nitrogen invested in Rubisco. These findings indicatedthat the temperature homeostasis of photosynthesis was regulatedby various parameters. On the other hand, the extent of temperaturehomeostasis of Rarea was unrelated to the maximum activity ofthe respiratory enzyme (NAD-malic enzyme). The Rarea/Parea ratiowas maintained irrespective of the growth temperatures in allthe species, suggesting that the extent of temperature homeostasisof Rarea interacted with the photosynthetic rate and/or thehomeostasis of photosynthesis.  相似文献

Stoichiometry among 3 thylakoid components, PSI and PSII andCyt b6-f complexes, was determined with the red alga Porphyrayezoensis with special reference to the regulation of PSI/PSIIstoichiometry in response to light regime. The ratio of PSIto PSII abundance was four times greater in thalli grown underorange light which excites mainly phycobilisome, thus PSII,than that under red light which excites preferentially Chl a,thus PSI. Cyt b6-f abundance remained almost constant. The PSIand PSII content was regulated separately under the two growthlight conditions as was also observed with the red alga Porphyridiumcruentum by Cunningham et al. [(1990) Plant Physiol. 93: 888].This differs from the cyanophyte Synechocystis PCC 6714 whereadjustment occurs only in the PSI content [(1987) Plant CellPhysiol. 28: 1547]. However, results on the marine cyanophyteSynechococcus NIBB 1071 indicate that changes in the PSI/PSIIsoichiometry is similar to red algae. In this species, as inthe red algae, more than one PSII is associated with each phycobilisome.The light regime also induced changes in the phycobiliproteincomposition in Porphyra yezoensis. Under PSII light, phycoerythrinincreased, and phycocyanin decreased, while under PSI lightthe response was reversed. The change suggests an occurrenceof complementary chromatic adaptation. This study was done to confirm our previous observation withthe pattern of changes in electron transport composition inducedby an imbalance of the electron transport state. Contents ofphotosystem (PS) I and II complexes and their antennae and Cytb6/f complex were determined for systems of cyanobacterium SynechocystisPCC 6714 of different PS I/PS II ratios. The results indicatedthat (1) the observed changes in the PS I/PS II ratio are not-dueto regulation of the activities of the respective PS's but tochanges in their contents, (2) the molar ratio between PS IIand Cyt b6/f complexes was fairly constant when marked changesoccurred in the PS I content, and (3) the PS II and Cyt b6/fcontents per cell remained fairly constant while the PS I contentchanged markedly. These findings agree with our previous observationwith autotrophic cells of Anacystis nidulans Tx 20 and supportour argument that in cyanobacterial and red algal electron transportsystems, the content of the terminalcomponent(s), such as PSI complex, is regulated in order to maintain a balance betweenthe electron influx by PS II action to the system and the effluxby PS I action from it. Chloroplast development was followed in intact bean leaves illuminatedwith far-red light by extracting chloroplasts at various timesto assay photosynthetic activities. Photochemical activity wasdetected in isolated chloroplasts prior to the times which werepreviously reported for intact leaf discs. Cyclic phosphorylationwas observed in isolated chloroplasts after 8 h of far-red illuminationwhile non-cyclic electron transport and phosphorylation weremeasurable after 12 and 16 h of illumination respectively. TheP/2e ratios were less than 0.5 after 24 h of far-red exposurebut approached a value of 1.0 by 60 h of illumination. Ammoniumchloride (10–3 M) had little effect on electron transportin isolated chloroplasts until after 24 h of far-red illumination.Chlorophyll a accumulated slowly from the onset of far-red illuminationwhile chlorophyll b was not detected until after 48 h of far-redexposure. Leaf fresh weight increased four-fold over the 60h illumination period. Electron microscopy of isolated chloroplasts from far-red-illuminatedleaves indicated the presence of unfused primary thylakoidsby 12 h of exposure and prolamellar bodies throughout the entire60 h illumination period. Grana were not observed in isolatedchloroplasts nor were they induced by a 2 min exposure of thechloroplasts to 172 000 lx of white light. O2 evolution in leaf discs of far-red-illuminated plants wasmeasurable after 16 h of illumination, attained a maximum valueby 36 h of far-red exposure, and then declined. Net CO2 fixationwas observed in leaf discs after 8 h of far-red illuminationand the rates remained constant for an additional 16 h, beforeincreasing at least two-fold.  相似文献

Blue light effects on the acclimation of energy partitioningcharacteristics in PSII and CO2 assimilation capacity in spinachto high growth irradiance were investigated. Plants were grownhydroponically in different light treatments that were a combinationof two light qualities and two irradiances, i.e. white lightand blue-deficient light at photosynthetic photon flux densities(PPFDs) of 100 and 500 µmol m–2 s–1. The CO2assimilation rate, the quantum efficiency of PSII (PSII) andthermal dissipation activity / in young, fully expanded leaves were measured under 1,600 µmol m–2 s–1white light. The CO2 assimilation rate and PSII were higher,while / was lower in plants grown under high irradiancethan in plants grown under low irradiance. These responses wereobserved irrespective of the presence or absence of blue lightduring growth. The extent of the increase in the CO2 assimilationrate and PSII and the decrease in / by high growth irradiance was smaller under blue light-deficient conditions. These resultsindicate that blue light helps to boost the acclimation responsesof energy partitioning in PSII and CO2 assimilation to highirradiance. Similarly, leaf N, Cyt f and Chl contents per unitleaf area increased by high growth irradiance, and the extentof the increment in leaf N, Cyt f and Chl was smaller underblue light-deficient conditions. Regression analysis showedthat the differences in energy partitioning in PSII and CO2assimilation between plants grown under high white light andhigh blue-deficient light were closely related to the differencein leaf N.  相似文献

Cytochrome composition of the cyanobacterial photosyntheticsystem was studied with Anacystis nidulans (Tx 20) in relationto the chromatic regulation of photosystem composition. Comparisonof cytochrome compositions in cells with a high PS I/II ratio(3.0, grown under weak orange light) and with a low ratio (1.6,grown under weak red light) indicated that cytochrome compositionwas also changed in the chromatic regulation of photosystemcomposition. Two types of cytochrome change were observed: 1)contents of cytochromes C553 and c548 were changed in parallelwith the changes in PS I content, and 2) cytochrome b553 andcytochrome b6-f complex were held at a constant molar ratioto PS II. The molar ratio, PS II : cytochrome b559 : cytochromeb6-f complex : cytochrome c553 : PS I : cytochrome C548, inthe red-grown cells was 1 : 2.5 : 1.3 : 0.17 : 1.6 : 0.67, andthe ratio in the orange-grown cells, 1:2.4:0.9:0.32:3.0:1.2.In both types of cells, almost all cytochrome f in the cytochromeb6-f complex was rapidly oxidized after multiple flash activation,indicating that all cytochrome b6-f complexes in cells of bothtypes are functionally connected to PS I, even when the molarratio to PS I is largely changed. Arabidopsis halleri is a Cd hyperaccumulator; however, the mechanismsinvolved in the root to shoot translocation of Cd are not wellunderstood. In this study, we characterized Cd transfer fromthe root medium to xylem in this species. Arabidopsis halleriaccumulated 1,500 mg kg–1 Cd in the shoot without growthinhibition. A time-course experiment showed that the releaseof Cd into the xylem was very rapid; by 2 h exposure to Cd,Cd concentration in the xylem sap was 5-fold higher than thatin the external solution. The concentration of Cd in the xylemsap increased linearly with increasing Cd concentration in theexternal solution. Cd transfer to the xylem was completely inhibitedby the metabolic inhibitor carbonyl cyanide 3-chlorophenylhydrazone(CCCP). Cd concentration in the xylem sap was decreased by increasingthe concentration of external Zn, but enhanced by Fe deficiencytreatment. Analysis with 113Cd-nuclear magnetic resonance (NMR)showed that the chemical shift of 113Cd in the xylem sap wasthe same as that of Cd(NO3)2. Metal speciation with Geochem-PCalso showed that Cd occurred mainly in the free ionic form inthe xylem sap. These results suggest that Cd transfer from theroot medium to the xylem in A. halleri is an energy-dependentprocess that is partly shared with Zn and/or Fe transport. Furthermore,Cd is translocated from roots to shoots in inorganic forms.  相似文献
