ELISA – Nachweis von Pflanzenviren mit Hilfe unterschiedlicher Reagenzstreifen

Abstract Detection of plant viruses by ELISA using different reagent strips
A simplified immunoassay for detection of plant viruses under field conditions was developed on the basis of the direct double antibody sandwich ELISA using dry reagent carriers immobilized on PVC-supports which are arranged in a fan-like manner. The fan consists of a first reagent carrier with antivirus IgG covalently bound to cyanuric chloride-activated (CCA) paper while the second and third are filter papers impregnated with alkaline phosphatase labelled antivirus IgG and the fluorogenic subsrate, 4-methylumbelliferylphosphate, respectively. Performing the test, the first carrier is contacted with a liquid sample containing the virus to be identified, the second and third carriers is contacted with a liquid sample containing the virus to be identified the second and third carriers are sequentially put one upon the other and the reactions carried out. The virus is detected by the reacted substrate fluorescing under a UV-light. The applicability of the test is demonstrated with cucumber mosaic virus and potato virus X.

Zusammenfassung Eine Methode zum histochemischen Nachweis der Cholindehydrogenase wird mitgeteilt. In diesem System werden keine zusätzlichen Cofaktoren benötigt. Das Enzym wird durch Hg⁺⁺-Ionen, Stickstofflost und Aureomycin gehemmt. Es ist resistent gegenüber Cyanid, Fluorid und Jodessigsäure. Seinph-Optimum liegt zwischenph 7,3 und 7,8.Im wesentlichen ist das Enzym auf Leber und Niere der untersuchten Nager beschränkt.Mit 3 TextabbildungenDer Deutschen Forschungsgemeinschaft danke ich für die Gewährung eines Stipendiums.     

Nachweis der thyrotropen Zellen der Adenohypophyse mit aromatischen Diaminen

Zusammenfassung Mit einer Perameisensäure-HID (high iron diamine)-Technik ist eine prägnante und elektive Darstellung thyrotroper Zellen der Adenohypophyse möglich. Diese Methode, die auf dem relativ hohen Gehalt an Disulfidgruppen bzw. Cystin des thyrotropen Hormons beruht, ist sehr empfindlich und liefert gut reproduzierbare Ergebnisse.

Summary With a performic acid-HID (high iron diamine) reaction, which is based upon the relative high cystine content of TSH, a selective demonstration of TSH-cells will be obtained. This method is a very sensitive one and gives reproducible results.

Detection of Phytophthora fragariae Hickman in roots of strawberry cultivar ‘Tenira’ by enzyme-linked immunosorbent assay (ELISA) Phytophthora fragariae Hickman is detected by ELISA in roots of strawberry cv. ‘Tenira’. Because of the high sensitivity of ELISA presence on fungal antigen was demonstrated before symptoms are detected in microscopical investigations.     

Genetische Untersuchungen bei Picea abies mit Hilfe der Isoenzym-Identifizierung

Summary Using techniques of starch gel zone electrophoresis, a considerable variation among esterase and leucine aminopeptidase isoenzyme patterns was found in the endosperm of dormant seeds of Norway spruce (Picea abies). Since the so-called primary endosperm of conifers is only a further developmental stage of the haploid female gametophyte, simple Mendelian segregations can be determined in seeds of individual open-pollinated trees. It was therefore possible to identify some esterase and leucine aminopeptidase loci only with regard to phenotypic frequency distributions without difficult crossing procedures.

Zusammenfassung Es wird eine Methode zum cytochemischen Nachweis der B-Sulfatase angegeben. Fixation: Baker-Formol, danach Einlegen der Blöcke in Holts Sucrose-Lösung. Inkubationsmedium: 50 mg 8-Oxychinolinsulfat, 200 mg Kochsalz, 10 ml Michaelis-Puffer pH 5,8–6,0, 0,5 ml hex. Pararosanilin. Das Enzym ist in zahlreichen Organen in den Lysosomen darstellbar. Die Herstellung des Substrates wird beschrieben.

Summary A method is described for the cytochemical demonstration of B-sulphatase. Fixation: Baker-formalin followed by immersing the material into Holt's sucrose solution. Incubation medium: 50 mg 8-Oxichinolinsulphate, 200 mg NaCl, 10 ml Michaelis buffer pH 5.8–6.0, 0.5 ml hex. Pararosanilin. The enzyme is demonstrable in the lysosomes of various organes. The preparation of the substrate is described.

Zusammenfassung Es handelt sich um eine Methode unter Verwendung der Gefriertrocknung am Kryostatschnitt. Vorteilhaft ist die Schnelligkeit des Verfahrens und die Möglichkeit, qualitativ gute, relativ großflächige Präparate zu gewinnen.

Summary A method using freeze-dried cryostat sections is described. The advantages are the rapidity of the procedure and the possibility to obtain relatively large specimens of good quality.

Absolute Viskositätsmessung mit Hilfe der Brown-Schen Molekularbewegung

Zusammenfassung Es wird das Prinzip einer in der Biologie bisher nicht verwendeten Methode zur Messung absoluter Viskositäten von Protoplasma, Zellsaft, beziehungsweise kleiner Mengen beliebiger Körpersäfte ausgeführt. Diese Methode, der die Brownsche Molekularbewegung zugrunde liegt, beruht auf der R. Fürthschen Methode der mittleren doppelseitigen Erstpassagezeiten zur Messung der Brownschen Bewegung. Anschließend werden die erforderlichen Bedingungen zur Auswertung und die notwendigen Einschränkungen kritisch diskutiert.
