The tracks of normal organisms of Oxytricha bifaria and of stage IA, IB, II, III, IV and V doublets were studied to test the hypothesis that the doublet might function as a dispersal form. Stage IA, the only stage to swim, swims straight with only rare interruptions; its rate of mobility (Rmo = 443 micro/s) is roughly twice that of singlets (Rmo = 218 micro/s). Stage IA doublets swim in three-dimensional movement which enables them to be carried away by water currents. The other stages seem to represent passage back towards the normal singlet form. The ethological evidence reported here together with other results already published supports the working hypothesis that the doublet of O. bifaria is a dispersal form suggests that the doublet might well represent a special fourth differentiation state of this species in addition to pairs, giants, and cysts. 相似文献

The relationship between femoral neck superior and inferior cortical thickness in primates is related to locomotor behavior. This relationship has been employed to infer bipedalism in fossil hominins, although bipeds share the same pattern of generalized quadrupeds, where the superior cortex is thinner than the inferior one. In contrast, knuckle‐walkers and specialized suspensory taxa display a more homogeneous distribution of cortical bone. These different patterns, probably related to the range of movement at the hip joint and concomitant differences in the load stresses at the femoral neck, are very promising for making locomotor inferences in extinct primates. To evaluate the utility of this feature in the fossil record, we relied on computed tomography applied to the femur of the Late Miocene hominoid Hispanopithecus laietanus as a test‐case study. Both an orthograde body plan and orang‐like suspensory adaptations had been previously documented for this taxon on different anatomical grounds, leading to the hypothesis that this fossil ape should display a modern ape‐like distribution of femoral neck cortical thickness. This is confirmed by the results of this study, leading to the conclusion that Hispanopithecus represents the oldest evidence of a homogeneous cortical bone distribution in the hominoid fossil record. Our results therefore strengthen the utility of femoral neck cortical thickness for making paleobiological inferences on the locomotor repertoire of fossil primates. This feature would be particularly useful for assessing the degree of orthograde arboreal locomotor behaviors vs. terrestrial bipedalism in putative early hominins. Aim Rainbow trout (Oncorhynchus mykiss, Walbaum 1792) is an exotic salmonid invading eastern Canada. First introduced for recreational fishing in Ontario, Quebec and the Maritime provinces, the species is now spreading in salmon rivers located in Eastern Quebec, where its stocking is strictly forbidden. Newly established populations have been found along the St Lawrence Estuary. To effectively mitigate the potential threat the invasion poses to native salmonids, we aimed to document the invasion’s origin and progress in the St Lawrence River and estuary. We first determined genetic origins among several potential wild and cultured source populations, found at the upstream and downstream extremities of the St Lawrence system. Thereafter, we studied the range expansion, predicting that the invasion process conforms to a one‐dimensional stepping‐stone dispersion model. Location Recently invaded salmon rivers that flow into the Estuary and Gulf of St Lawrence in Quebec, and watercourses supporting naturalized populations (Lake Ontario, Lake Memphremagog and Prince‐Edward‐Island rivers). Methods Rainbow trout from 10 potential source populations (wild and domestic strains) and 243 specimens captured in salmon rivers were genotyped at 10 microsatellite loci. Genetic origins of specimens and relationship between colonies were assessed using assignment analyses based on individuals and clusters. Results Invasion of rainbow trout in Eastern Quebec is directed downstream, driven by migrants from upstream naturalized populations, found in the Ganaraska River (Lake Ontario), and, to a lesser extent, in Lake Memphremagog. Populations from the Maritime provinces and domestic strains do not contribute to the colonisation process. A recently established population in Charlevoix (Eastern Quebec) supplies other downstream colonies. Main conclusions Rainbow trout is spreading from Lake Ontario downstream to Eastern Quebec using the St Lawrence River system as an invasion corridor. Range expansion in a downstream direction is driven by a more complex stepping‐stone dispersion model than predicted. Management options to protect native salmonids include reducing the effective size of the Charlevoix population, impeding reproduction in recently colonized rivers, halting the upstream migration of anadromous spawners, and curtailing stocking events inside the stocking area. 相似文献

Seaweeds have physiological mechanisms to acquire, utilize, and store various forms of nitrogen in environments where nitrogen levels vary tremendously in space and time. Knowledge of the nitrogen relationships of seaweeds is required for the development of successful seaweed mariculture. For example, it would seem at first that continuous nitrogen enrichment would be desirable in such systems because maximal seaweed yields are possible only when growth is not nitrogen-limited. Yet such fertilization is wasteful and can result in yield reductions due to the enhancement of epiphyte growth. Because most seaweeds can rapidly taken up high concentrations of nitrogen, far in excess of what is required for current growth demands, enrichments are needed only when internal nitrogen concentrations fall to near the critical level (i.e., the minimal tissue concentration of nitrogen required for maximal growth). With the aid of measurements of the fluorescence yield, the efficiency of the various deexcitation mechanisms of an exciton in the light-harvesting system has been determined. For this purpose, the fluorescence of dark-adapted as well as of 3-(3,4-dichlorophenyl)-1,1-dimethylurea (DCMU)-treated and preilluminated leaves of Zea mays L. was excited by single ultrashort laser pulses of different energies. The experimental results have served for the fitting of solutions of rate equations, which describe the deexcitation by linear relaxation processes like fluorescence and radiationless transitions, by annihiation of excitons, and by traps both in the ground state and in an excited state. We have obtained the following results: a ratio of antenna chlorophyll molecules to Photosystem II traps of 600:1, an annihilation constant γ = 2·10?8 cm3·s?1, a mean trapping time of , a trapping probability for traps in the ground state of 2·10?8 cm3·s?1, and 6·10?9 cm3·s?1 for traps in an excited state. 相似文献

CD24 is a cell surface, heavily glycosylated glycosylphosphatidylinositol-anchored mucin-like protein that is overexpressed in various human malignancies. To accurately analyze CD24 function and dissect its biological role in a defined genetic background, it is critical to tightly regulate its expression and be able to turn it on/off in a restricted environment and at a specific time. The tetracycline-induced expression system is most promising as it exhibits such regulation, lack of pleiotropic effects, and high and rapid induction levels. To evaluate the oncogenic and immunotherapeutic potential of CD24 by applying the Tet-On system, the human CD24 gene was cloned downstream to two tetracycline operator sequences, resulting in pCDNA4/TO-CD24, which was then transfected into tetracycline (Tet) repressor-expressing cells (293T-REx), allowing tight on/off regulation, thereby resulting in a very low background or leaky CD24 expression. Selected clones were chosen for further studies and characterized in vitro and in vivo, and several treatment modalities were examined. In addition, the role of CD24 in promoting cell proliferation and tumor growth was studied. The tetracycline-dependent system was successfully implemented. Tetracycline treatment induced CD24 expression in a dose- and time-dependent fashion, which was abrogated following treatment with anti-CD24 monoclonal antibodies (mAbs). CD24-induced expression led to an increased proliferation rate that was inhibited by mAb treatment. In vivo, significantly larger tumors were developed in tetracycline-fed mice. The CD24 Tet-On system is a good model to unravel the role and underlying CD24 pathogenesis in vivo. This valuable tool allows the successful study of novel treatment options, whose effectiveness depends on the CD24 expression level. This set of experiments supports CD24 oncogenic properties. 相似文献
