The effects of systematically administered puromycin on the fine structure of the lung were studied. The effects varied depending on the duration of exposure and the time interval between the last injection and sacrifice. After short term exposure most surfactant had separated from the epithelial surface and profound alterations in the tubular myelin structure were seen. After moderate duration of exposure a previously undescribed multilamellar lining layer was observed which was often detached from the alveolar epithelium. Six hours after the last injection the regular tubular myelin pattern reappeared. Puromycin treatment results in inhibition of various proteins synthesized by type II epithelial cells. Inhibition of synthesis of some proteins, most probably that of glycoprotein A, causes a primary effect on the structure of surfactant. The loss of at least some of the cytoskeletal proteins in Type II epithelial cells apparently results in interference with exocytosis of lamellar body contents. 相似文献

This study centres on the effect of puromycin, applied in various concentrations, on the differentiation of choroid plexus cells in tissue cultures. Puromycin added to these cultures in doses of 25, 50 and 100 μg/ml medium merely produced reversible cell changes when present for 2 h. After 3 h, choroid plexus cells react to continuous feeding with puromycin in a dose of 100μg/ml with an almost total loss of the membrane system of the endoplasmic reticulum. At this time many of the free ribosomes have disappeared. The results of this study suggest a continuous renewal of the membrane-structure of the endoplasmic reticulum. 相似文献

The possibility is discussed that puromycin may not inhibit cleavage in marine eggs solely by inhibiting protein synthesis. It is suggested that part of the effect of puromycin is through 6-dimethylaminopurine (DMAP) (the purine component of puromycin) a potent cleavage inhibitor which appears to enhance protein synthesis in sea urchin eggs. 相似文献

Axons of corpora cardiaca in the Nosema infected honeybees contained numerous tightly packed neurosecretion granules. These granules invariably possessed high electron densities. Neurosecretion granules of similar size were also observed in the axons of corpora cardiaca in the healthy honeybees. But these granules were shown to possess various electron densities. Image analysis revealed that some of these granules possessed an electron dense core, and the electron densities decreased gradually from the core, to the fringe of the granule. Some granules appeared to be completely broken down into fine granular substances, while others appeared to be completely broken down into finer particles. It is suggested that in the healthy honeybees, the releasing of neurosecretion from the corpora cardiaca had taken place normally, whereas in the diseased bee, the releasing mechanism had been blocked. The fine-structure of the median neurosecretory cells and corpora cardiaca of the Cecropia silkmoth during the first 7 days after transfer from cold conditions to room temperature was compared to that of similar animals whose development was arrested with aminophylline. The major difference observed was the failure of the intrinsic secretory cells of the corpus cardiacum to degenerate in the arrested animals. This failure to degenerate coincides with the expected period of brain hormone release. After long periods of arrest, the medial neurosecretory cells and their axons became distended with neurosecretory granules. The significance of these observations in the initiation of adult development is discussed. 相似文献

Aminonucleoside (AMS) inhibited the cell cycle of human lung fibroblasts at a point in G1 phase and at another point in G2 phase. Even when this inhibition was fully established, DNA synthesis and mitosis which were in progress proceeded normally. Inhibition of RNA synthesis in the cultures preceded the effects on DNA synthesis and mitosis, but inhibition of protein synthesis could not be detected. These points of potential inhibition do not exist in the cell cycle of HeLa cell, or are not affected by aminonucleoside. Here inhibition of cell proliferation by AMS was less marked, and when inhibition eventually occurred it was not specific for any point of the cell cycle. The rate of entry of the inhibitor was similar in both types of cell. 相似文献

Summary The ultrastructure of the optic and trigeminal nerves of the rat, cryofixed by use of a liquid nitrogenpropane jet, was examined, paying special attention to the myelin sheath and the cytoskeleton of the axoplasm. The cytoskeleton of the axoplasm is formed by a meshwork of neurofilaments and microtubules connected both to each other and also to the cell organelles and axolemma. These cross-linkers are fixed to the longitudinal neurofilaments in a helical arrangement, which could be a morphological substrate for the diverse axonal transport phenomena. The myelin sheath is formed by concentrically apposed membrane pairs, which are not fused together. The corresponding major and intraperiod lines seen using classical electron microscopy are in fact fissures that are obscured by the pattern of the selective deposition of osmium at certain sites and cannot be interpreted as specific structures. The cryofixed myelin membranes have the appearance of predominantly globular subunits arranged in an asymmetrical bilayer. The globular particles are of diverse diameter and occupy varying positions within the membrane. The tight junctions or zonulae occludentes of the myelin are formed by arrays of isolated particles, and consequently the fibril formation seems to be a result of the chemical fixation. 相似文献
