采用PCR-DGGE指纹、克隆测序和系统发育分析技术较系统地对我国南海贪婪倔海绵(Dysidea avara)和澳大利亚厚皮海绵(Craniella australiensis)共附生的优势细菌进行了研究。研究发现变形菌门(Proteobacteria)细菌是这两种海绵中的主要优势细菌,贪婪倔海绵中的变形菌包含了α、β、γ三种类型,而澳大利亚厚皮海绵中仅有γ一种类型。两种海绵都有拟杆菌(Bacteroidetes),但是具体的种类不同。这些细菌都是第一次在海绵中被发现。澳大利亚厚皮海绵共附生的优势细菌还包括放线菌属(Actinobacterium)及厚壁菌门(Firmicutes)细菌,菌群多样性要比贪婪倔海绵的丰富。两种海绵尽管来自于同一海域但其共附生优势细菌的组成明显不同,这说明海绵共附生微生物具有宿主特异性。  相似文献

黄海中部近岸春夏季鱼卵、仔稚鱼群落结构特征   总被引:5,自引:0,他引:5  
2013年4月下旬至7月上旬,采用浅水I型网垂直拖网的调查方式对黄海中部20m以浅海域(34°30'—35°30'N,119°20'—120°20'E)的鱼卵、仔稚鱼种类组成与数量分布进行了6个航次的综合调查。春夏季共采集鱼卵21种(未定种3种),共计2247粒,隶属于9科15属。仔稚鱼22种(未定种1种),共计286尾,隶属于13科17属。鱼卵优势种为日本鳀Engraulis japonicus、赤鼻棱鳀Thryssa kammalensis、江口小公鱼Stolephorus commersonnii、皮氏叫姑鱼Johnius belengerii和属Callionymus spp.。仔稚鱼优势种为大银鱼Protosalanx chinensis、白姑鱼Argyrosomus argentatus、日本鳀、赤鼻棱鳀和皮氏叫姑鱼。鱼卵丰度和分布与经度呈负相关、与SST呈正相关,但与其他环境因子如纬度、表层盐度(SSS)和水深等无显著相关性。仔稚鱼丰度和分布与上述环境因子均未表现出显著相关性。基于Bray-Curtis指数的聚类分析表明,鱼卵优势种包括集中分布于研究水域34°48'N以南海域(江口小公鱼、皮氏叫姑鱼和青鳞小沙丁Sardinella zunasi)、10m等深线以内(赤鼻棱鳀),以及北部水域(日本鳀和属)3种主要聚集类型;仔稚鱼优势种包括集中分布于34°48'N—35°12'N海域(赤鼻棱鳀和日本鳀)、连云港外海浅水区(白姑鱼、皮氏叫姑鱼和梭鱼Liza haematocheilus,以及南部水域(褐菖鲉Sebastiscus marmoratus和鲬Platycephalus indicus)3种主要聚集类型。研究结果揭示了黄海中部近岸海域鱼卵、仔稚鱼群落的时空分布格局动态,为科学认识环境变动下该海区渔业种群数量动态和资源变化提供了科学依据。  相似文献

【背景】珊瑚适应环境的能力与机体内共附生细菌有关,然而,这些细菌在珊瑚宿主适应环境变化过程中所起的作用尚不清楚。对珊瑚共附生细菌进行纯培养,探究其生物功能和生态作用,是解析珊瑚宿主环境适应机理的重要途径。【目的】研究热耐受性不同的2种造礁珊瑚共附生可培养潜在耐热细菌多样性和功能,为理解珊瑚适应环境的能力提供新的认识。【方法】从涠洲岛选取2种热耐受性差异显著的霜鹿角珊瑚(Acropora pruinosa)和丛生盔形珊瑚(Galaxea fascicularis)为研究对象,采用2216E、海水R2A和海水GYP这3种琼脂培养基,于32℃(珊瑚热耐受阈值)培养条件下对珊瑚共附生潜在耐热细菌进行分离培养,对分离菌株进行16S rRNA基因测序和序列相似性分析。选取代表菌株进行热耐受性检验,并利用平板对峙法进行抗菌活性检测。【结果】2种造礁珊瑚共附生可培养潜在耐热细菌的多样性存在显著差异。从热敏型的霜鹿角珊瑚中获得44株细菌,隶属于4个门22个属,其中弧菌属(Vibrio)、假交替单胞菌属(Pseudoalteromonas)和Tenacibaculum为优势属;从热耐...  相似文献

通过对铜陵地区不同河段的上覆水和沉积物的理化性质进行分析,结合聚合酶链式反应-变性梯度凝胶电泳(PCR-DGGE)的方法,研究了铜陵不同河段中附生细菌的多样性及其与环境因子的关系。结果表明,8个河段中上覆水和沉积物的营养水平及重金属含量存在较大差异,不同河段附生细菌的多样性指数也存在差异。数据分析表明,铜陵河流中黄浒河附生细菌的多样性最高,顺安河下游和钟仓河附生细菌的多样性最低。这种差异主要与河段间上覆水和沉积物不同的营养水平、重金属含量水平等因素有关。附生细菌的香农-威纳多样性与上覆水中的氨态氮(NH3-N)、总氮(TN)、总磷(TP)、可溶性正磷酸盐(PO3-4-P)和高锰酸盐指数(COD Mn)呈显著或极显著负相关,与沉积物中镉(Cd)、铜(Cu)和锌(Zn)的含量具有显著或极显著正相关性。  相似文献

为了解调水调沙(Water-Sediment Regulation Scheme, WSRS)对黄河口海域鱼卵、仔稚鱼的群落结构和时空分布格局的影响,分别于2017年(非调水调沙年)和2018年(调水调沙年)的6—7月在黄河口海域利用水平拖网开展了6个航次的鱼卵、仔稚鱼调查。运用聚类分析和冗余分析(RDA)等方法对该海域非调水调沙年和调水调沙年的鱼卵、仔稚鱼丰度和群落结构的空间分布及环境因子状况等进行了综合的分析比较,结果表明:6个航次调查共采获鱼卵6212粒,仔稚鱼431尾,隶属于8目12科14属,其中,河口定居性鱼类短吻红舌鳎(Cynoglossus joyeri)为控制黄河口及邻近海域夏季鱼类浮游生物群落演替格局的主要种类;鱼卵受环流和调水调沙的共同影响,其空间分布在各调查阶段有所不同,仔稚鱼则趋向于在河口近岸海域分布;河口东北部及东南部靠近莱州湾区域是黄河口及邻近海域鱼类浮游生物群落的主要聚集区;调水调沙年鱼卵、仔稚鱼的采获总量以及底层鱼类鱼卵的采获数量均明显高于非调水调沙年,调水调沙带来的丰富径流和泥沙有利于黄河口海域渔业资源的补充;冗余分析结果表明,海洋鱼类的群落结构与环境...  相似文献

莱州湾春季鱼卵仔稚鱼群落结构和物种多样性   总被引:3,自引:0,他引:3  
于2007—2010年春季(5月)利用大型浮游生物网对莱州湾海域位置相同的12个站位进行4个航次拖网调查,研究分析了该海域鱼卵仔稚鱼群落组成、优势种、空间分布及其多样性特征。结果表明:4个航次共获得鱼卵、仔稚鱼26种,隶属于6目16科24属,1种衔科鱼卵鉴定到科,另有1种鱼卵未能鉴定种类。鱼卵、仔稚鱼分布不均匀,莱州湾西部还湾底海域分布较多。斑鰶(Konosiruspunctatus)为该海域鱼卵的第一优势种;鳀(Engraulisjaponicus)在2007年和2008年作为仔稚鱼的第一优势种出现,而2009年和2010年仔稚鱼的第一优势种则是虾虎鱼。调查期间该海域鱼卵、仔稚鱼的均匀度指数和生物多样性指数变化趋势一致,均表现为从2007年开始随年代递减的趋势;种类数和丰富度指数则从2008年开始呈现随年代递减的趋势。2007—2010年4a鱼卵仔稚鱼聚类结果显示,2007年和2008年鱼卵仔稚鱼群落结构相似,与2009年和2010年均有差异。与1982年山东省近岸调查结果相比,莱州湾海域鱼卵仔稚鱼群落结构发生明显变化,斑鰶取代鳀成为第一优势种,小黄鱼等经济鱼类数量逐渐降低。  相似文献

采用超声波粉碎法、涡旋振荡法、超声波清洗法、研磨匀浆法等4种不同的方法处理龙须菜,分离其体表的附生细菌,并对分离细菌的数量、种类、形态结构、细胞壁特性等进行了观察和分析。通过对不同方法和相间方法的不同处理所得结果比较显示,超声波清洗法和研磨匀浆法对分离细菌的数量和种类效果都较差;超声波粉碎法和涡旋振荡法效果较好,尤以超声波粉碎法的30W30s处理效果最好,该方法分离到本项目4个方法13个处理获得的16个菌株中的12个菌株,龙须菜的细菌数1.75×106cells/g。  相似文献

利用稀释平板法从中华真地鳖体内分离得到52株共附生放线菌,活性筛选表明菌株BD2C2具有较好的抗菌活性,其发酵液对金黄色葡萄球菌的抑菌圈直径超过23mm,对枯草芽胞杆菌的抑菌圈直径约17.5mm,对大肠杆菌也有一定的抑制作用。进一步研究表明其活性物质主要集中在中等极性部位。在供试浓度为每张滤纸片30μg时,菌株BD2C2发酵液的乙酸乙酯提取物对金黄色葡萄球菌、枯草芽胞杆菌和大肠杆菌的抑菌圈直径分别约27.5、22.4和10.7mm,其抑制活性与阳性对照硫酸庆大霉素相当。利用形态学观察和分子生物学方法初步鉴定该菌为Streptomyces sp.,为一个潜在新种。作为微生物源杀菌剂,DB2C2具有一定开发潜力,值得进一步研究。  相似文献

Over 20 marine fish species have been studied forfarming or stock enhancement in Australia. However,commercial production has been dominated by the cageculture of Salmo salar in Tasmania, Thunnusmaccoyiiin South Australia, and to a lesser extent Latescalcarifer in Queensland. A major impediment to thecommercial production of new species has been thelarge-scale production of juvenile fish. Thedevelopment of marine fish larval rearing technologyin Australia has had four main influences over thelast decade: culture system technology from France,live food culture and nutritional enhancement fromBelgium, artificial diets from Japan and extensivepond culture from the USA. Microalgae and live foodculture is based on traditional aquaculture speciesand methods. Recent Australian research has focusedon induced spawning, the role of stress in inhibitingovulation, factors influencing initial swim bladderinflation in larvae, larval nutrition, extensiveculture and diagnosis of disease. Over the next 5years, Australian aquaculturists should be able toproduce industrial quantities of a range of nativemarine fish, either in intensive fish hatcheries, orin combination with extensive pond culture.  相似文献

共生珊瑚的异养营养指有虫黄藻共生的珊瑚在同化作用过程中,除通过虫黄藻光合作用获取营养之外,还可以从外界环境中直接摄取现成的有机物,经消化吸收后转变为自身的组成物质或储存为能量.国内外异地养殖或繁育虫黄藻共生珊瑚的研究多集中在光照、水流、水质等条件对珊瑚生长的影响,对共生珊瑚异养营养需求与供应方面的关注较少.本文从共生珊瑚异养营养来源、影响共生珊瑚异养营养供应的因素以及研究手段等方面,对国内外研究进展进行了综述,探讨异养营养供应对共生珊瑚的意义.总的来说,目前共生珊瑚异养营养的研究尚处于起步阶段,不论是研究方法或者珊瑚选择摄食的内在机制,都需要深入探究.  相似文献

Intact antigen uptake in intestinal epithelial cells of marine fish larvae   总被引:1,自引:0,他引:1  
Endocytosis of bacteria by epithelial cells in the hindgut of 4- to 6-day-old cod ( Gadus morhua L. ) larvae and of 10- to 12-day-old herring ( Clupea harengus L. ) larvae was demonstrated by fluorescence microscopy and transmission electron microscopy. Fixed sheep erythrocytes and bacteria-sized latex particles were not ingested by these cells. By immunohistochemical methods intact bacterial antigens were observed in columnar epithelial cells in the foregut of 4-day-old yolk-sac larvae of cod. In these cells a preferential uptake of Vibrio fischeri and Vibrio salmonicida cells as compared to Flavobacterium sp. was found. The possible role of these findings in the establishment of an indigenous microflora, defence against pathogens or nutrient absorption in fish larvae is discussed.  相似文献

Aims: To evaluate the importance of the bacterial composition on the resilience of the organic matter assimilation in the sea. Methods and Results: Chemostats were inoculated with coastal and offshore bacterial communities. Bacterial density and protein synthesis increased before stabilizing, and this response to confinement was more marked in the offshore chemostats. Before the toluene perturbation the community structure in the coastal chemostats remained complex whereas the offshore chemostats became dominated by Alteromonas sp. After the perturbation, bacterial protein synthesis was inhibited before peaking briefly at a level fivefold to that observed before the perturbation and then stabilizing at a level comparable to that before the perturbation. Alteromonas dominated both the coastal and the offshore communities immediately after the perturbation and the coastal communities did not recover their initial complexity. Conclusions: Cell lysis induced by the toluene perturbation favoured the growth of Alteromonas which could initiate growth rapidly in response to the nutrient pulse. Despite their different community structure in situ, the resilience of protein synthesis of coastal and offshore bacterial communities was dependent on Alteromonas, which dominated in the chemostats. Significance and Impact of the Study: Here we show that although Alteromonas sp. dominated in artificial offshore and coastal communities in chemostats, their response time to the shock was different. This suggests that future perturbation studies on resilience in the marine environment should take account of ecosystem history.  相似文献

长江口水域夏季鱼卵和仔稚鱼年间变化   总被引:3,自引:0,他引:3  
基于2005年、2008年、2009年和2011年8月(夏季)在长江口水域(30°30'—31°45'N,121°15'—123°10'E)4个航次的浮游生物拖网资料,分析了长江口水域鱼卵和仔稚鱼的种类组成、数量分布特征及其年间变化。结果表明:4个航次采集的鱼卵和仔稚鱼鉴定到种的种类有17种,隶属于8目13科,以鲈形目种类最多,11种,其次是鲱形目,5种,其他各目种类均小于5种;种类数存在明显年间差异,2005年种类数最多(鱼卵3种,仔稚鱼8种),其次是2009年和2011年,2008年种类数最少(鱼卵1种,仔稚鱼5种)。优势种年间更替明显,长蛇鲻(Saurida elongata)、虾虎鱼(Gobiidae spp.)和中华小公鱼(Stolephorus chinensis)在2005年是优势种,2008年优势种为鳀鱼(Engraulis japonicus),2009年优势种为鳀鱼、寡鳞飘鱼(Pseudolaubuca engraulis)、虾虎鱼等,2011年虾虎鱼和小公鱼(包括小公鱼属未定种Stolephorus spp.和中华小公鱼Stolephorus chinensis)成为优势种。2005年鱼卵和仔稚鱼数量分布的密集区在嵊泗列岛附近水域,2008年鱼卵和仔稚鱼出现较少,未出现明显的数量密集区;2009年鱼卵数量较少,仔稚鱼数量较多,密集区主要分布在在长江口以外123°E附近水域;2011年鱼卵主要分布在在长江北支口门外附近水域,仔稚鱼在调查区内分布相对均匀。  相似文献

Freeze dehydration provides instant immobilisation and fixation of living materials to capture particular events and avoids the need for dehydration through an ethanol series for SEM viewing. Examples of the technique are given from freshwater epiphytic and epizoic communities. Seasonal successional trajectories of transplanted and experimental diatom communities were studied during the summer of 1973. Manipulation of these diatom assemblages was accomplished through the development of an in situ experimental apparatus capable of incubating these organisms without doing violence to the community itself.In one set of experiments the effects of water quality were tested on the development of inocula from grossly similar but fairly distant locations. In another set the native inoculum was incubated in the continual presence of Fe, Cu, Cr, Pb, Zn and crude oil. Seasonal succession took place in all of the control and experimental communities. During the course of succession the trajectories of assemblages under some conditions converged while others diverged. Since the random fluctuations in the environment were constant for all assemblages, the abundances of individual species were due to niche packing and interspecific restraints on the biotic potential of selected diatom species as well as the changes in water quality. The most diverse community was an experimentally mixed one. The high diversity of this community was probably due to niche packing a phenomenon which restricted the realized niches of many species and hindered their blooming. Paradoxically twice as many species (40%) had their greatest niche breadth in the mixed community as did species in the transplanted (20%) or native controls (17%). Enrichment of the incubation medium with heavy metals and oil altered the community structure and diversity of most experimental vessels. Growth of some species was depressed while that of others was enhanced. The trajectories of the communities enriched with oil and Pb were quite similar throughout the summer. The Cu and Cr assemblages initially followed the trajectory of oil and Pb but later diverged. Greatest species diversity was consistantly found in the community incubated in the presence of Cu. The Fe enriched community generally diverged from all the others. Recurrent group analysis identified distinctive groups of species for each of the experimental assemblages as well as controls.The response of these diatom communities incubated under natural conditions were characteristic of resilient communities in which populations with different structures were quickly established.Supported by ERDA Contract E (11-1) 3254, Reference Number (COO) 3254-28. 依据1999年6~8月在东海(26°30’~32°30’N、121°00’~128°00’E)海域的鱼卵仔鱼资源调查资料,对东海鱼卵仔鱼种类组成结构的特征进行了分析,并就其与水温、盐度的关系进行了探讨。结果表明:鱼卵仔鱼种类组成丰富,整个调查海域,种类数平面分布呈南部高,北部低的分布特征。仔鱼的种类结构相对鱼卵要稳定,多样性指数较高,其种类组成复杂,种间分布不如鱼卵均匀,优势种明显,优势度较大。温度和盐度是影响鱼卵仔鱼种类分布的重要环境因子。多样性指数与种类数分布的空间差异,与台湾暖流、黑潮水以及江浙沿岸流的消长有关。  相似文献
