Water-soluble salvianolic acid A (Sal A) and salvianolic acid B (Sal B) were successfully isolated and purified from the crude extract of Salvia miltiorrhiza by high-speed counter-current chromatography (HSCCC). The solvent system was n-hexane–ethyl acetate–methanol–water (3:6:6:10, v/v/v/v). 4.27 mg of Sal A and 32.09 mg of Sal B were obtained from 260 mg of the crude sample. The purities of Sal A and Sal B were 96.67% and 97.43%, respectively. Their structures were identified by 1H NMR and 13C NMR. Antioxidant activities of Sal A and Sal B were also evaluated and compared by the methods of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay and 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid (ABTS+) radical cation decolourisation assay. Both Sal A and Sal B showed high radical scavenging activities with their EC50 values being 1.43 ± 0.09 and 1.81 ± 0.01 μg/ml in DPPH radical method. The ABTS results showed that Sal A and Sal B exhibited high total antioxidant activities, their EC50 values were 1.35 ± 0.00 and 1.43 ± 0.01 μg/ml, respectively.  相似文献

The whole seed (W), endosperm (E) and hull (H) of five cultivars of Job’s tears (Coix lachryma-jobi Linn. var. ma-yuen Stapf) including Thai Black Phayao, Thai Black Loei, Laos Black Loei, Laos White Loei and Laos Black Luang Phra Bang were processed before solvent extraction by non-cooking, roasting, boiling and steaming Each part of the Job’s tears was extracted by the cold and hot process by refluxing with methanol and hexane. The total of 330 extracts included 150 methanol extracts and 180 hexane extracts were investigated for anti-proliferative activity on human colon adenocarcinoma cell line (HT-29) by the sulforhodamine B (SRB) assay. The extracts which gave high anti-proliferative activity were tested for apoptotic activity by acridine orange and ethidium bromide double staining and anti-oxidative activities including free radical scavenging and lipid peroxidation inhibition activities. The extract from the hull of Thai Black Loei roasted before extracting by hot methanol (M-HTBL-R2) showed the highest anti-proliferative activity on HT-29 with the IC50 values of 11.61 ± 0.95 μg/ml, while the extract from the non-cooked hull of Thai Black Loei by cold methanol extraction (M-HTBL-N1) gave the highest apoptosis (8.17 ± 1.18%) with no necrosis. In addition, M-HTBL-R2 and M-HTBL-N1 indicated free radical scavenging activity at the SC50 values of 0.48 ± 0.12 and 2.47 ± 1.15 mg/ml, respectively. This study has demonstrated the anti-colorectal cancer potential of the M-HTBL-R2 and M-HTBL-N1 extracts.  相似文献

Boerhaavia diffusa Linn. of family Nyctaginaceae is a known traditional medicinal plant and has been used in the treatment of many free radical mediated diseases. Excessive formation of free radicals, either reactive oxygen species (ROS) or reactive nitrogen species (RNS) is responsible for damaging various biomolecules like DNA, lipids and proteins. The present investigation was initially carried out to explore the possible link between antioxidant, oxidative DNA damage protective and α-amylase inhibitory property of B. diffusa root extract and their bioactive fraction. Our results illustrated an enhanced DPPH radical scavenging activity/antioxidant power of methanol root extract (IC50 < 250 μg/ml) than ethanol (IC50 = 250 μg/ml) and aqueous extract (IC50 > 250 μg/ml). In addition, the methanol root extract also showed better oxidative DNA damage protective activity and α-amylase inhibitory property than ethanol and aqueous root extract. Phytochemical screening of B. diffusa ethanol and methanol root extract showed the presence of saponins, alkaloids, flavonoids, glycosides and terpenoids in large amount. By means of repetitive preparatory TLC and HPLC the potent antioxidant and α-amylase inhibitory fraction was isolated from methanol root extract. Our results illustrated that DPPH radical scavenging activity (IC50 < 250 μg/ml) and oxidative DNA damage protective and α-amylase inhibitory activity of the isolated/purified bioactive compound from methanol extract were significantly closer to that of crude extract, which in turn confirm that antioxidant and antidiabetic property of methanol root extract resides in this fraction and established a significant correlation between antioxidant and inhibitory α-amylase property of this bioactive fraction compound. These profound effects of B. diffusa methanol root extract and its purified fraction against oxidative plasmid DNA damage, antioxidant and α-amylase inhibitory activity may explain its extensive use in daily life and possible health benefits.  相似文献

The synthesis of novel indolopyrazoline derivatives (P1-P4 and Q1-Q4) has been characterized and evaluated as potential anti-Alzheimer agents through in vitro Acetylcholinesterase (AChE) inhibition and radical scavenging activity (antioxidant) studies. Specifically, Q3 shows AChE inhibition (IC50: 0.68 ± 0.13 μM) with strong DPPH and ABTS radical scavenging activity (IC50: 13.77 ± 0.25 μM and IC50: 12.59 ± 0.21 μM), respectively. While P3 exhibited as the second most potent compound with AChE inhibition (IC50: 0.74 ± 0.09 μM) and with DPPH and ABTS radical scavenging activity (IC50: 13.52 ± 0.62 μM and IC50: 13.13 ± 0.85 μM), respectively. Finally, molecular docking studies provided prospective evidence to identify key interactions between the active inhibitors and the AChE that furthermore led us to the identification of plausible binding mode of novel indolopyrazoline derivatives. Additionally, in-silico ADME prediction using QikProp shows that these derivatives fulfilled all the properties of CNS acting drugs. This study confirms the first time reporting of indolopyrazoline derivatives as potential anti-Alzheimer agents.  相似文献

The objective of this study was to evaluate the antioxidant properties of extracts of the lucanid beetle, Serrognathus platymelus castanicolor Motschulsky, obtained at different growth stages. The antioxidant activities of six different extracts were determined using 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) (ABTS), and singlet oxygen (1O2). The activity level of pupal methanol extracts (PME) was higher in the DPPH and ABTS radical scavenging assays, whereas that of the water extracts was weaker in all assays. The 1O2 quenching ability of the PME was comparable to that of ascorbic acid (effective concentration of 50% 1O2 quenching: EC50 = 0.184 mg/ml-1 and 0.167 mg/ml-1, respectively). The free radical scavenging antioxidant ability of the extracts significantly altered phenolic contents, important factors in the potency of antioxidant capacity. Our results suggest that these extracts may reduce oxidative stress in living organisms and reduce oxidative damage in insects under unfavorable conditions.  相似文献

Two new phenanthrene alkaloids, beilschglabrines A (1) and B (2) were isolated from the stem bark of Beilschmiedia glabra, together with lupeol, taraxerol, and 24-methylenelanosta-7,9-diene-3β-15α-diol. The structures of the isolated compounds were elucidated by extensive spectroscopic data analysis and comparison with respective literature data. The compounds were tested for DPPH radical scavenging, acetylcholinesterase and lipoxygenase inhibitory activities. Compound 1 displayed considerable activity in the acetylcholinesterase (IC50 50.4 μM), the DPPH radical scavenging (IC50 115.9 μM) and the lipoxygenase (IC50 32.8 μM) assays.  相似文献

Liquid-liquid chromatography can lead to the isolation of compounds or enrichment of fractions. To investigate the structural characteristics of antioxidants and UV-B absorbing molecules, a preparative centrifugal partition chromatography (CPC) method was developed to prepare enriched bioactive fractions from the red seaweed Solieria chordalis (Gigartinales, Solieriaceae). After a methanolic extraction of dried Solieria chordalis and liquid-liquid separation using ethyl acetate/water (v/v), the ethyl acetate phase was submitted to CPC using Arizona Y of the quaternary solvent system composed of n-heptane/ethyl acetate/methanol/water (19/1/19/1, v/v) in ascending mode. Among the fifteen collected fractions, three fractions gave up to 23.50% of DPPH radical scavenging activity. The CPC fractionation was monitored by HPLC and, four compounds exhibited UV-B absorbing capacity regarding their absorption at 310 nm.  相似文献

To investigate the anti-cancer properties of soil-borne actinobacteria, MJM 8637, the glutathione S-transferase pi (GST-pi) assay, anti-tumor necrosis factor (TNF)-α assay, the level of antioxidant potential by DPPH radical scavenging activity, NO scavenging activity, and ABTS radical scavenging activity in ethyl acetate extract were determined. The 16S rDNA sequencing analysis revealed that Streptomyces sp. strain MJM 8637, which was isolated from Hambak Mountain, Korea, has 99.5% similarity to Streptomyces atratus strain NBRC 3897. The physiological and the morphological characteristics of the strain MJM 8637 were also identified. The ethyl acetate extract of MJM 8637 inhibited TNF-α production approximately 61.8% at concentration 100 μg/ml. The IC50 value of the strain MJM 8637 extract on GST-pi was identified to be 120.2 ± 1.6 μg/ml. In DPPH, NO, and ABTS radical scavenging assays, the IC50 values of the strain MJM 8637 extract were found to be 977.2 μg/ml, 1143.7 μg/ml, and 454.4 μg/ml, respectively. The ethyl acetate extract of the strain MJM 8637 showed 97.2 ± 1.3% of cell viability at 100 μg/ml in RAW 264.7 cell viability assay. The results obtained from this study suggest that the ethyl acetate extract of Streptomyces sp. strain MJM 8637 could be considered as a potential source of drug for the cancers that have multidrug resistance with its GST-pi inhibition and anti-inflammation activities, and low cytotoxicity.  相似文献

Novel hydroxylated benzylideneamino imidazole derivatives were synthesized and their radical scavenging activity was assessed against DPPH and hydroxyl radicals. In the DPPH assay, most of the synthesized compounds showed an IC50 in the range 3.2 μM  IC50  8.4 μM, lower than the reference compound trolox (IC50 = 9.5 μM) or the parent aldehydes (5.4 μM  IC50  11.6 μM). The activity depends mainly on the phenolic subunit (number and position of the hydroxyl groups) and the extent of conjugation with the imidazole ring. In the deoxyribose assay, all the compounds, including parent imidazoles and aldehydes, showed high activity against the hydroxyl radical and the ability to chelate iron ions. At 5 μM concentration, the compounds protected the deoxyribose from degradation by hydroxyl radical between 62% and 38%.  相似文献

Two new diketopiperazines, cyclo(l-Phe-l-NMe-DOPA) (2) and cyclo[l-Phe-l-(NMe-3-(NMe-3-O-α-l-rhamnopyranosyl)-DOPA] (3), along with a known diketopiperazine (1), were isolated from the cultures of Streptomyces sp. SC0581. Their structures were elucidated by extensive spectroscopic analysis, single-crystal X-ray crystallographic analysis, and chemical correlation. Compounds 1  3 exhibited more potent ABTS radical cation scavenging activity (IC50 values: 3.7  14.6 μM) than l-ascorbic acid (IC50: 17.7 μM). In the present work we synthesized a series of hydroxy-3-arylcoumarins (compounds 19), some of them previously described as MAO-B selective inhibitors, with the aim of evaluating their antioxidant properties. Theoretical evaluation of ADME properties of all the derivatives was also carried out. From the ORAC-FL, ESR and CV data it was concluded that these derivatives are very good antioxidants, with a very interesting hydroxyl, DPPH and superoxide radicals scavenging profiles. In particular compound 9 is the most active and effective antioxidant of the series (ORAC-FL = 13.5, capacity of scavenging hydroxyl radicals = 100%, capacity of scavenging DPPH radicals = 65.9% and capacity of scavenging superoxide radicals = 71.5%). Kinetics profile for protection fluorescein probe against peroxyl radicals by addition of antioxidant molecule 9 was also performed. Therefore, it can operate as a potential candidate for preventing or minimizing the free radicals overproduction in oxidative-stress related diseases.  相似文献

Ethyl acetate extraction (EAE) of the steam exploded sugarcane bagasse may be an effective and economic way to extract antioxidants as well as enhance the enzymatic hydrolysis and bioethanol yield from the extracted residue. Therefore, the effects of EAE on steam-exploded sugarcane bagasse (SESB) were studied. Under boiling solvent extraction (BSE), the efficiency of EAE for obtaining phenolics from SESB was approximately 20%. EA extracts obtained under BSE showed an H2O2 scavenging activity (210 μL) of 99%. The IC50 values for 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity and reducing power of BSE40 were 50.89 and 256.38 μg/mL, respectively, while those of vitamin C were 24 and 112 μg/mL, respectively. EAE improved the glucose yield by 30% but had no significant effect on the xylose yield during the enzymatic hydrolysis obtained using Celluclast 1.5L and Novozym 188. EAE also increased the ethanol yield by 8.78% by employing simultaneous saccharification and fermentation. The present study may be of great importance in industrial bioethanol production from steam-exploded biomass environmentally friendly and economically.  相似文献

Isolated by the cellulolytic enzyme lignin (CEL) process, water-alcohol (1:1, v/v) was introduced as co-solvent in the process of the hydrothermal treatment. The modification parameters such as reaction temperature and time, solid-to-liquid ratio, and catalysts (NaOH and NaOAlO2) have been investigated in terms of the specific lignin properties, such as the phenolic hydroxyl content (OHphen), DPPH free radical scavenging rate, and formaldehyde value. The CELs were also characterized by GPC, FT-IR and 1 H NMR spectroscopy, and Py-GC/MS. The key data are under optimal lignin modification conditions (solid-to-liquid ratio of 1:10 (w/v) and a temperature of 250 °C for 60 min) are: OHphen content: 2.50 mmol/g; half maximal inhibitory concentration (IC50) towards DPPH free radicals: 88.2 mg/L; formaldehyde value: 446.9 g/kg). Both base catalysts decrease the residue rate, but phenol reactivities of the products were also detracted. Py-GC/MS results revealed that modified lignin had a higher phenolic composition than the CEL did, especially the modified lignin without catalyst (ML), which represented 74.51% phenolic content.  相似文献

Edaravone (3-methyl-1-phenyl-2-pyrazoline-5-one) is a neuroprotective drug that has been used for brain ischemia injury treatment. Because its activity is speculated to be due to free radical scavenging activity, we carried out a quantitative determination of edaravone’s free radical scavenging activity against multiple free radical species. Electron spin resonance (ESR) spin trapping-based multiple free-radical scavenging (MULTIS) method was employed, where target free radicals were hydroxyl radical, superoxide anion, alkoxyl radical, alkylperoxyl radical, methyl radical, and singlet oxygen. Edaravone showed relatively high scavenging abilities against hydroxyl radical (scavenging rate constant k = 2.98 × 1011 M−1 s−1), singlet oxygen (k = 2.75 × 107 M−1 s−1), and methyl radical (k = 3.00 × 107 M−1 s−1). Overall, edaravone’s scavenging activity against multiple free radical species is as robust as other known potent antioxidant such as uric acid, glutathione, and trolox. A radar chart illustration of the MULTIS activity relative to uric acid, glutathione, and trolox indicates that edaravone has a high and balanced antioxidant activity with low specificity.  相似文献

Ultrasound complex enzymes (UCE) were used to extract crude polysaccharides (CEP) from Epimedium leaves. The response surface methodology (RSM) based on Box–Behnken Design (BBD) was employed to further optimize the extraction conditions. The optimum conditions were: complex enzyme dose of 50, 250, 200, 100 U/g (papain, pectase, cellulase and α-amylase), extraction temperature 46.8 °C, ultrasound time 42.3 min, pH 4.28, and ultrasound power 311 W. Under these conditions, the experimental yield of CEP was 5.98%, which was well matched with the value predicted by the model. Three major polysaccharide fractions (EP-1, EP-2 and EP-3) were obtained by ultrafiltration, chromatography on DEAE-Sepharose Fast Flow and Sephadex G-100 column. Their chemical–physical characteristics were investigated using chemical methods, gas chromatography (GC) and high-performance size-exclusion chromatography (HPSEC). Biochemical characterization of the polysaccharide fractions was evaluated on the basis of free radical scavenging, ferric-reducing antioxidant power (FRAP), lymphocyte proliferation, macrophage phagocytosis and NK cell activity assay in vitro. The results showed that all fractions contained higher galacturonic acid (GalA) and galactose (Gal), possessed considerable antioxidant activity, and could promote proliferation of splenic lymphocytes, activate macrophages and enhance NK cells’ activity. UCE as a more effective and environment-friendly technique could be employed for extracting active ingredients from plant materials, extensively applied in pharmaceutical and food industries.  相似文献

A mild and efficient method for the conversion of fatty acid methyl esters from lard into ascorbyl esters via lipase-catalyzed transesterification in co-solvent mixture is described. A solvent engineering strategy was firstly applied to improve fatty acid ascorbyl esters production. The co-solvent mixture of 30% t-pentanol:70% isooctane (v/v) was optimal. Response surface methodology (RSM) and central composite design (CCD) were employed to estimate the effects of reaction parameters, such as reaction time (12–36 h), temperature (45–65 °C), enzyme amount (10–20%, w/w, of fat acid methyl esters), and substrate molar ratio of fatty acid methyl esters to ascorbic acid (8:1–12:1) for the synthesis of fatty acid ascorbyl esters in co-solvent mixture. Based on the RSM analysis, the optimal reaction conditions were determined as follows: reaction time 34.32 h, temperature 54.6 °C, enzyme amount 12.5%, substrate molar ratio 10.22:1 and the maximum conversion of fatty acid ascorbyl esters was 69.18%. The method proved to be applicable for the synthesis of ascorbyl esters using Novozym 435 in solvent.  相似文献

We purified a novel antioxidant peptide from Ruditapes philippinarum (R. philippinarum) and investigated its free radical scavenging activities. To prepare the peptide, eight proteases were tested for enzymatic hydrolysis. α-chymotrypsin hydrolysate, which showed clearly superior hydroxyl radical scavenging activity (p < 0.05), were further purified using a flow filtration system and consecutive chromatographic methods. Finally, a novel antioxidant peptide was obtained, and the sequence was identified as Ser-Val-Glu-Ile-Gln-Ala-Leu-Cys-Asp-Met. The peptide from R. philippinarum effectively scavenged hydroxyl, DPPH, alkyl and superoxide radicals, with observed IC50 values of 0.042, 0.091, 0.107 and 0.372 mg/ml, respectively. This is the first report of an antioxidant peptide derived from the hydrolysates of R. philippinarum which, further, possesses competitive free radical quenching potential.  相似文献

The present study was carried out to determine the free radical scavenging potential of culture filtrate of Streptomyces sp. AM-S1. Antioxidant activity of culture filtrate, lyophilized culture filtrate and ethyl acetate extract of Streptomyces sp. AM-S1 was determined by various in vitro assays such as ferric reducing power assay, phosphomolybdenum reduction, DPPH and ABTS radical scavenging activities. The results revealed that the culture filtrate of Streptomyces sp. AM-S1 effectively scavenged DPPH (IC50 90.2 μl/ml) and ABTS (IC50 13.2 μl/ml) radicals in a concentration dependent manner. In all the assays, ethyl acetate extract registered higher antioxidant activity when compared with the lyophilized culture filtrate (LCF). In addition, ethyl acetate extract (1123.4 μmole Fe(II)/mg extract) exhibited higher ferric reducing activity than the standard BHA (814.4 μmole Fe(II)/mg extract). Further works are needed on the isolation and identification of antioxidant molecules from the ethyl acetate extract of Streptomyces sp. AM-S1 culture filtrate.  相似文献

Three new phenolic glycosides (1–3) together with nine known ones were isolated from the roots of Tecoma mollis using DPPH radical scavenging bioassay-guided chromatographic separation. The structures of the new compounds were established using extensive spectroscopic data and HR-MS. The antioxidant, COX-2 inhibition, and cytotoxic activities were evaluated for the isolated compounds. Compound 4 displayed the strongest radical scavenging activity relative to ascorbic acid with IC50 8.7 μM. Compounds 5, 6, and 10 showed promising COX-2 inhibitory action, IC50 values of 11.3 μM, 9.4 μM, and 13.4 μM, respectively. All compounds exhibited weak cytotoxic activity against Hela and A549 cancer cell lines.  相似文献

Nine rotenoids were isolated from the hexane and dichloromethane extracts of Derris trifoliata stems and were tested for nitric oxide (NO) inhibitory activity using RAW264.7 cells. The result indicated that 12a-hydroxyrotenone (7) possessed very potent NO inhibitory activity with an IC50 value of 0.002 μM, followed by 1 (deguelin, IC50=0.008 μM), 9 (12a-hydroxyelliptone, IC50=0.010 μM) and 2 (α-toxicarol, IC50=0.013 μM), respectively. In addition, the DPPH scavenging activity of rotenoids was also investigated. It was found that 6a,12a-dehydrodeguelin (5) possessed the highest activity against DPPH with an IC50 value of 7.4 μM, followed by deguelin (1, IC50=27.4 μM). All compounds did not show any cytotoxicity at their IC50 values for NO inhibitory activity.Structure–activity relationships (SARs) of these rotenoids against NO release are as follows: (1) hydroxylation at C12a dramatically increased activity, (2) prenylation at furan ring increased activity markedly and (3) hydrogenation of a double bond at C6a–C12a conferred higher activity. For the DPPH radical scavenging effect, it was found that (1) introduction of a double bond at C6a–C12a increased activity and (2) hydroxylation of C11 at the D-ring decreased activity. As regards active compounds of Derris trifoliata stems, the isolated compounds are responsible for the NO inhibitory effect, especially 7, 1, 9 and 2, whereas 5 and 1 are those for the DPPH scavenging activity.  相似文献
