云芝糖肽的液体发酵培养基的研究 总被引:2,自引:0,他引:2
通过液体发酵的摇瓶实验,考察碳源、氮源、碳氮比和乙醇浓度对云芝糖肽的产率影响,确定葡萄糖和玉米淀粉为适宜碳源,蛋白胨和黄豆饼粉为适宜氮源,合适的碳氮比为20∶1,适宜的乙醇体积分数为1.5%。采用以上的培养基进行70 L发酵罐进行扩大实验和10 t工业生产规模试验。发酵周期32 h,最终生物量达到13.0 g/L,云芝糖肽产率平均为2.21 g/L,与摇瓶发酵水平相当。此外工业实验还确定了云芝液体发酵的pH值、残糖浓度以及菌丝体形态等终点指标。 相似文献

利用热水抽提从云芝菌丝体中提取胞内多糖,初步纯化后,用DEAE-Sepharose CL-6B进行分离,从中分离出两个带电荷多糖组分即CVP-I和CVP-Ⅱ,对这两个组分分别用Sepharose CL-6B凝胶柱层析进行纯度鉴定,均出现单一峰,然后用紫外扫描发现这两个组分均出现蛋白多糖的特征吸收,从而可以判断这两个组分是蛋白结合多糖。 相似文献

经试验云芝菌深层培养合适培养基为马铃薯蔗糖综合培养基。其配方:马铃薯20%,蔗糖2%,KH2PO40.1%,(NH4)2SO40.2%,水100ml,pH自然。试验表明,利用云芝菌深层培养发酵滤液可制成营养丰富的保健饮料。经测定,每100ml饮料中云芝糖含量0.82g、粗蛋白含量为0.51g,游离氨基酸为27.06mg饮料卫生指标符合中国食品卫生标准。 相似文献

深层培养云芝菌丝体蛋白多糖的提取及性质 总被引:5,自引:0,他引:5
采用水、3%草酸和0.2mol/L氢氧化钠为溶剂分别提取深层培养的云芝〔Polystictusversicolor(L.)Fr.〕菌丝体蛋白多糖,其得率分别为菌丝体干重的20%、49%和19%。提取的粗多糖经SephadexG100柱层析进一步分离、纯化,纯化的多糖经聚丙烯酰胺凝胶电泳分离后呈现一个斑点,在SephadexG100柱层析中有一个对称峰,紫外及红外光谱分析和蛋白质含量测定表明该多糖为蛋白多糖,其蛋白质和多糖含量在水提多糖中分别为13%和62%、在酸提多糖中分别为5%和91%。在碱提多糖中分别为3%和81%。组成该多糖的主要单糖是葡萄糖和木糖。 相似文献

The evolution of cooperative colony foundation (pleometrosis)in ants has
been attributed to conversion of extra resourcesinto increased competitive
ability. Most cooperative foundingspecies provide these additional resources
from internal fatstores; however, in those species that forage for resources,
theextent and type of individual investment in multiqueen coloniesis not
well understood. We compared singly- and group-foundedlaboratory colonies of
the desert leaf-cutter ant Acromyrmexversicolor to investigate how
cooperation affects colony survival,foraging success, and worker production.
Under laboratory conditions,single foundresses were significantly less likely
to initiatea successful symbiotic fungus garden, which inevitably led to
colonystarvation and death. If gardens were initiated successfully,however,
there was no difference in the growth trajectoriesand foraging patterns
between colony types. Cooperation in thisspecies may more likely be
maintained by survival benefits thanby growth rate differences, which may be
constrained in groupsby individual and colony-level costs. 相似文献

Abstract The optimal fermentation medium and conditions for mycelial growth and water-soluble exo-polysaccharides production by Isaria farinosa B05 were investigated. The medium components and fermentation conditions were optimized according to the one at a time method, while the concentration of medium components was determined by the orthogonal matrix method. The results showed that the optimal fermentation medium was as follows: sucrose 3.5% (w/v), peptone 0.5%, yeast extract 0.2%, K2HPO4 0.1%, and MgSO4 0.05%. The suitable fermentation conditions were as follows: initial pH 7.0, temperature 25°C, medium volume 75 mL/250 mL, inoculum volume 5% (v/v), time 5d. In such optimal nutrition and environmental conditions, the maximal mycelial yield was 2.124 g/100 mL after 4 day's fermentation, while maximal water-soluble exo-polysaccharides production reached 2.144 g/L after 5 day's fermentation. 相似文献

用正交试验法对樟芝菌 (AntrodiacamphorataZang&Su)的发酵培养基进行了研究。选取马铃薯淀粉、葡萄糖、麦芽糖、酵母膏、蛋白胨、麸皮、KH2 PO4 、MgSO4 ·7H2 O等作为试验因素 ,采用L2 7(31. 3)正交表设计五因素、三水平的正交试验 ,对所选各因素的添加量分别作了试验 ,对菌丝干重和发酵液多糖含量结果作了方差分析 ,确定了合适的发酵培养基。试验结果表明 :樟芝菌的深层发酵适宜培养基是 :马铃薯淀粉 1 .0 0 % ,葡萄糖2. 0 0 % ,酵母膏 0 . 10 % ,蛋白胨 0 . 10 % ,麸皮 1 .0 0 % (或 0. 5 0 % ) ,KH2 PO4 0 . 10 % ,MgSO4 ·7H2 O 0 . 0 5 %。 相似文献
