Myxinol tetra-acetate was examined by both nuclear-magnetic-resonance and mass-spectral techniques. The results are compared with corresponding data for 5α-ranol tetra-acetate and 5α-cyprinol tetra-acetate. The structure deduced for myxinol tetra-acetate, 3β,7α,16α,27-tetrahydroxy-5β-cholestane, is in agreement with that simultaneously arrived at by Haslewood (1966).  相似文献

Two peptides with urotensin II (UII = smooth muscle stimulating) activity have been isolated from urophyses of the sucker, Catostomus commersoni. The amino acid sequences of the two peptides, designated UIIA and UIIB, are as follows: UIIA, H-Gly-Ser-Gly-Ala-Asp-Cys-Phe-Trp-Lys-Tyr-Cys-Val-OH; UIIB, H-Gly-Ser-Asn-Thr-Glu-Cys-Phe-Trp-Lys-Tyr-Cys-Val-OH. These peptides have the part sequence Cys-Phe-Trp-Lys-Tyr-Cys-Val in common with the UII peptide from Gillichthys mirabilis and the part-sequence Phe-Trp-Lys in common with somatostatin.  相似文献

The distribution of urotensin I (UI) and corticotropin-releasing factor (CRF) immunoreactive (IR) structures was studied in the central nervous system (CNS) of the white sucker using the peroxidase-antiperoxidase immunocytochemical procedure. The close sequence homology between both peptides resulted in a high degree of crossreactivity. This was resolved by saturating the antisera solutions with heterologous antigens and specificity tests were done by adding excess of homologous peptides. UI immunoreactivity was seen in all of the identifiable caudal spinal cord neurosecretory cells, in their processes projecting to the urophysis, in thin beaded fibres coursing along the spinal cord, in brain stem, hypothalamus, proximal pars distalis and, especially, in the telencephalon. Some IR-UI specific and IR-CRF specific parvocellular neurons were also identified in the caudo-ventral tuberal region and ventral telencephalon. The IR-CRF was mainly present in parvocellular and magnocellular perikarya of the nucleus preopticus and in the preoptic-neurohypophysial pathway. Dense networks of IR-CRF reacting beaded fibres were also located in the lateral and posterior recessus nuclei. In the pituitary, IR-CRF fibre bundles were seen mainly in the neurointermediate lobe and in the rostral pars distalis. The cells of origin of the extraurophyseal system of IR-UI fibres in the sucker CNS have not been identified. The distribution of CRF immunostaining correlates well with the documented knowledge of CNS structures involved in the control of ACTH secretion in the goldfish. The probability of the occurrence of two UI-CRF related molecules, or of two different forms resulting from a common precursor molecule, forming two separate neuronal systems in the sucker CNS seems likely.  相似文献

cDNA clones encoding two members of the vasotocin hormone precursor gene family have been isolated from the white sucker Catostomus commersoni. The hormone is encoded by at least two distinct genes, both of which are expressed, as indicated by Northern blot analysis. Genomic DNA amplified by the polymerase chain reaction has been used to define exon-intron boundaries. Both vasotocin genes contain introns in positions corresponding to those found in the gene of their mammalian counterpart vasopressin. The predicted vasotocin precursors show a surprising degree of sequence divergence, amounting to 45% at the amino acid level, of which only approximately half can be accounted for by conservative amino acid changes. The precursors include a hormone moiety followed by a putative neurophysin sequence that is longer at the C-terminus by a tract of some 30 amino acids by comparison to their mammalian counterpart. Each of these sequences contains a leucine-rich core segment resembling that found in copeptin, a glycopeptide moiety present in mammalian vasopressin precursors.  相似文献

The distribution of stanniocalcin immunoreactivity was examined in the corpuscles of Stannius of the white sucker (Catostomus commersoni) by using a chum salmon stanniocalcin antiserum, Western blotting, and light and electron microscopy. The white sucker possesses at least two stanniocalcin-immunoreactive corpuscles in the most posterior portions of the kidneys. Immunocytochemistry and ultrastructure revealed two cell-types in the corpuscle parenchyma, only one of which was immunoreactive. The nonimmunoreactive cells contained dense-cored vesicles and long processes that extended between the immunoreactive cells and terminated at perivascular spaces. When corpuscle extracts were subjected to electrophoresis and Western blotting, three nonreduced stanniocalcin-like immunoreactive bands (approximately 56, 61, and 64 kDa) were observed. However, in the presence of a reductant, a diffuse band migrating in the range of 28 to 32 kDa was noted. The results of this study on the white sucker demonstrate the presence of a dimeric stanniocalcin-like molecule and present evidence of a previously uncharacterized cell-type in the corpuscles of Stannius.  相似文献

Summary Using the label-fracture technique, an ultrastructural comparison was made of the number and distribution of wheat germ agglutinin (WGA)-binding sites between human normal and sickle red blood cells. The WGA was adsorbed to colloidal gold, and quantitative analysis of the electron micrographs revealed that more binding sites were present on the sickle erythrocytes than on the normal erythrocytes. Moreover, the sites were more clustered on the sickle red cells than on the normal red cells. Use of another lectin, Bandieraea simplicifolia-II, revealed that it did not bind to normal or sickle red cells. Because of the affinity of the WGA for sialic acid residues, it is probable that the WGA is binding to a transmembrane sialoglycoprotein, glycophorin A. The conformation and/or distribution of the glycophorin A molecules may be altered by the sickle hemoglobin that binds to the red cell membrane. Hence, as detected by WGA, new surface receptors, which could play a role in the adhesion of sickle cells to endothelium may be exposed.  相似文献

Concentrations of manganese were determined in the liver, kidney, muscle and bone of white suckers (Catostomus commersoni) from five acid (pH < 5.8), and two circumneutral lakes in south-central Ontario. Manganese tissue concentrations were greater in fish captured from the most acidified lakes with the greatest concentrations of dissolved manganese. These fish had increased concentrations of manganese in the liver, as indicated by a comparison of liver:kidney manganese concentration ratios among the seven fish populations. Tissue concentrations of manganese from all populations either were negatively correlated (P < 0.05) or remained constant with fish size indicating homeostatic regulation of this metal. Manganese concentrations of the benthic fauna were positively correlated to sediment concentrations (R=0.30). Lake sediment manganese concentrations were significantly correlated to maximum lake depth (R=0.80, P < 0.03), with the concentrations in the top 0–1 cm dependent on the redox conditions in the seven lakes. Based on the seven lakes studied, manganese concentrations in the benthic-feeding white sucker correlated better with dissolved manganese, than with either the concentrations in food or surficial sediments.  相似文献

Synopsis Epizootics of skin neoplasms in teleost species have been documented within the Great Lakes region over the last decade. The white sucker,Catostomus commersoni, has been proposed as a sentinel species to monitor environmental health in these systems. The prevalence of skin neoplasia is elevated in white suckers and other fish taken from chemically polluted sites or from lake regions adjacent to heavy industry. Lip papillomas regress and proliferate spontaneously in captive wild suckers. It is important to investigate the relevance of these observations to papilloma etiology to determine whether the prevalence of this disease is a suitable biomarker for environmental health. White suckers were captured in the spring of 1992 and 1993 during annual spawning runs (mid to late April) at the Ganaraska River, which discharges into Lake Ontario at Port Hope, Ontario. Under crowded laboratory conditions, there was either proliferation of existing papillomas or development of new papillomas. However, in uncrowded conditions, existing papillomas either regressed completely or there was no development of new papillomas. Protein Kinase C (PKC), a proposed marker enzyme for hyperplasia and neoplasia, was used to determine if regressing and proliferating papillomas could be differentiated on the basis of biochemical activity. PKC activity was lower in proliferating papillomas, but not significantly different from papillomas sampled initially from Ganaraska River suckers. Regressing papillomatous tissue displayed a significantly higher level of enzyme activity than either proliferating or unchanged papillomas, but the PKC activity of regressing papillomas was not significantly different from that of normal lip epidermis.  相似文献

The alpha-subunit of a Na+/K+ ATPase has been cloned by analysing a lambda gt11 library constructed from polyA+ RNA from the hypothalamic region of the teleost fish Catostomus commersoni (white sucker). The cDNA clone consists of 3853 bp and predicts a protein of 1027 amino-acid residues. Alignment of the sucker sequence with protein sequences previously published for alpha-subunits from various species reveals a high degree of homology throughout the entire sequence containing five potential sites for N-glycosylation, a phosphorylation site and a site for binding fluorescein 5'-isothiocyanate (FITC). A hydropathy profile predicts a secondary structure of the Na+/K+ ATPase alpha-subunit with at least eight membrane-spanning domains. Northern and southern blot analyses suggest the existence of two distinct Na+/K+ ATPase alpha-subunit genes in the sucker genome.  相似文献

Testosterone induced papillomata in 85% (11/13) of initially non-papillomatous white suckers Catostomus commersoni and increased papillomata growth in 100% (16/16) of initially papillomatous suckers. 17β-oestradiol induced papillomata in 83% (10/12) of initially nonpapillomatous suckers and increased papillomata growth in 100% (16/16) of initially papillomatous suckers. Less than 29% (4/14) of white suckers injected with tamoxifen developed papillomata, while complete papillomata regression was observed in 71% (10/14) of initially papillomatous suckers. In control groups 59% (27/46) of suckers either retained or developed papillomata and 27% (6/24) of suckers exhibited tumour regression. Protein kinase C (PKC) activity was significantly depressed and ornithine decarboxylase (ODC) activity was significantly elevated in steroid-treated papillomata v. normal lip epidermis. ODC activity was significantly depressed in tamoxifen-injected, regressing papillomata. There were no significant differences in plasma oestrogen and testosterone levels between papillomatous and nonpapillomatous female fish from a site contaminated with persistent organic chemicals and an uncontaminated reference site. Similarly, no significant differences in testosterone and 11-ketotestosterone levels were observed between papillomatous and non-papillomatous male fish.  相似文献

Summary Scotopic visual pigments measured in the creek chub and the white sucker are porphyropsins with mean max values located at 538.3 and 536.5 nm, respectively. There is a shift of the max towards shorter wavelengths during the winter in both of these species coinciding with similar changes in the quality of downwelling light. max is significantly correlated to the P50 and spectrum width of the downwelling light and dissolved oxygen. An analysis of variance shows that there are significant differences between the max values of: fish in the two lakes, fish at different times, the two species at different times and fish in different lakes at different seasons. The offset visual pigments of both species appear to be well adapted to their photic environment in terms of the contrast hypothesis. The proton nuclear-magnetic-resonance spectra of the cyano-met complexes of the cooperative dimeric and tetrameric hemoglobins from the mollusk Scapharca inaequivalvis have been investigated and compared to those of other structurally characterized oxygen binding hemoproteins. For these proteins, cooperativity is displayed even in the homodimer and preliminary X-ray structural data reveal an unusual back-to-front assembly with intersubunit contacts involving the EF helices [Royer, W. E., Love, W. E. + Fenderson, F. F. (1985) Nature (Lond.) 316, 277-280]. The pattern of hyperfine shifts is very similar for the dimer and tetramer chains, but distinctly different from those of previously characterized low-spin, ferric heme proteins. Individual heme resonances are identified by reconstituting the protein with specifically deuterated hemes. While the axial interactions involving the proximal and distal histidines are very similar to that in myoglobins and other hemoglobins, both the heme contact shift pattern and the amino acid dipolar shift pattern reflect a significantly reduced asymmetry. The decreased spread of the non-cordinated amino acid signals is interpreted in terms of a rotation of the magnetic axes relative to those in myoglobin or other hemoglobins, rather than a change in the magnetic anisotropy. The decreased spread of the heme methyl contact shifts supports this conclusion and is consistent with an orientation of the proximal histidine with the imidazole ring rotated by about 30-40 degrees relative to that in other structurally characterized proteins. Although resonances associated with a complex pattern of alternate heme orientations can be detected immediately after reconstitution of the protein, the isolated protein was found to exhibit insignificant equilibrium heme rotational disorder.  相似文献

The stereochemistry of the hydroxyl group at C-24 in 5 beta-ranol (27-nor-5 beta-cholestane-3 alpha,7 alpha,12 alpha,24,26-pentol) a principal bile alcohol of the bullfrog which is structurally related to the major human urinary bile alcohol, 27-nor-5 beta-cholestane-3 alpha,7 alpha,12 alpha,24,25-pentol, is described. Two isomers (IIIa and IIIb) at C-24 of 27-nor-5 beta-cholest-25-ene-3 alpha,7 alpha,12 alpha, 24-tetrol were synthesized from cholic acid (I) by the conversion to 3 alpha, 7 alpha, 12 alpha-triacetoxy-5 beta-cholan-24-al (II) followed by a Grignard reaction with vinylmagnesium bromide. The absolute configurations at C-24 of the unsaturated tetrols (IIIa and IIIb) were elucidated as S and R, respectively, by means of the difference of the reactivity to Sharpless oxidation, a stereoselective epoxidation. Catalytic hydrogenation of each delta 25-tetrol (IIIa or IIIb) gave (24R)- or (24S)-27-nor-5 beta-cholestane-3 alpha,7 alpha,12 alpha, 24-tetrol (IVa or IVb). The configurations at C-24 of two isomeric 3 alpha,7 alpha,12 alpha,24-tetrahydroxy-27-nor-5 beta-cholestan-26-oic acids (Va and Vb) were determined as S and R, respectively, by means of their conversion into the saturated tetrols (IVa and IVb) of known absolute configurations by a Kolbe electrolytic coupling with acetic acid. The lithium aluminum hydride reduction product of the 24R-acid (Vb) was identical with the naturally occurring 5 beta-ranol, hence 5 beta-ranol has the 24R configuration.  相似文献

Synopsis White suckers,Catastomus commersoni, use olfactory cues to return to the same spawning stream year after year. If we assume that they follow a model similar to the well-known salmon model, olfactory imprinting must occur very early in their development. We describe the time of migration from the nursery stream in relation to the development of the white sucker olfactory system to determine if the requisite anatomical structures are present which would permit imprinting. At hatching the olfactory placode is present and beginning to differentiate, the lumen of the olfactory capsule is starting to form, and the olfactory tract projects into the telencephalon. Larvae migrate approximately 2 weeks later or at a size of 14 mm TL. At this time olfactory cilia are present, the olfactory tract is robust and the telencephalon is beginning to differentiate. Therefore, it appears that the fundamental neural structures necessary for imprinting are present. A comparison with salmon, however, clearly demonstrates that the white sucker olfactory apparatus is not as well developed as that of salmon at time of migration. This raises the question of the ability of white suckers to imprint in the same manner as salmon and whether the salmonid model is applicable to white suckers. Alternative imprinting hypotheses are discussed.  相似文献

The levels of DNA adducts in the hepatic tissue of the white sucker fish speciesCatostomus commersoni were determined by32P-postlabelling. The fish were caught at four sites: two sites near the city of Windsor (Québec, Canada) on the St. François River, a downstream tributary of the St. Lawrence River, and two sites in the St. Lawrence River itself, near the city of Montréal (Québec, Canada). The latter sites are known to be contaminated by many pollutants including polycyclic aromatic hydrocarbons. Total adduct levels in all fish ranged from 25.1–178.0 adducts per 109 nucleotides. White sucker from the selected sites of the St. Lawrence River had a significantly higher mean level of DNA adducts than those of the St. François River (129.4 vs 56.8, respectively). These results suggest that the effluents of many heavy industries (e.g. from a Soderberg aluminium plant) flowing in the St. Lawrence River are more likely to produce genotoxic damage to fish than those released in one of its tributary, and mainly associated to the activities of a small town and a nearby pulp and paper mill.  相似文献

Boron-neutron capture therapy (BNCT) and magnetic resonance imaging (MRI) are quite attractive techniques for treatment and diagnosis of cancer, respectively. In order to develop practical tools for BNCT and MRI, novel compounds containing both the trifluoromethyl group and 10B atom in a single molecule were designed. In the present study, p-boronophenylalanine and p-boronophenylalaninol with the trifluoromethyl group were synthesized, and 19F NMR measurements of these compounds were carried out.  相似文献

Light and electron microscopic studies of the morphological features of immature and mature rodlet cells in Catostomus commersoni are presented emphasizing the cells' association with epithelial tissues. The peripheral fibrillar layer is lacking from the apex and from the base of the cell. A cytoplasmic extension from the base may be a feeding mechanism whereby the rodlet cell obtains nutrient at the expense of adjacent cells leaving intercellular spaces often containing myelin figures. RNAase digestion studies demonstrate the presence of RNA in the electron dense rodlet core.
The structure and histochemistry of the rodlets which do not appear to disintegrate upon expulsion from the cell are compared to the cytoplasmic inclusions of both normal fish cells and protozoan parasites. The possible association of the rodlet cell with various pathological conditions is briefly reviewed and the authors conclude that it is premature to disregard the possibility that this cell could be a parasite or infective agent.  相似文献
