胞间连丝为多细胞植物有机体提供了一个直接的细胞间物质运输和信息传递的细胞质通道,把一个个独立的“细胞王国”转变成相互连接的共质体,它是当今细胞生物学中十分活跃的研究领域。日益增多的研究结果揭示,胞间连丝协调基因表达和许多的细胞生理生化过程,对细胞的分裂与分化、形态发生、植物体的生长与发育,以及植物对环境的反应与适应等诸方面都起着十分重要的作用。本文仅就胞间连丝结构的多样性;胞间通道的调节因子;大分子蛋白质和核酸的胞间运输;胞间连丝阻断和共质体分区的形成及其与形态发生、休眠和抗逆性的关系等几个方面的新进展做一个简要的综述,借此例证胞间连丝在植物生命活动中的重要意义。  相似文献

Plasmodesmata (PD) are the communication channels which allow the trafficking of macromolecules between neighboring cells. Such cell-to-cell movement of macromolecules is regulated during plant growth and development; however, little is known about the regulation mechanism of PD size exclusion limit (SEL). Plant viral movement proteins (MPs) enhance the invasion of viruses from cell to cell by increasing the SEL of the PD and are therefore a powerful means for the study of the plasmodesmal regulation mechanisms. In a recent study, we reported that the actin cytoskeleton is involved in the increase of the PD SEL induced by MPs. Microinjection experiments demonstrated that actin depolymerization was required for the Cucumber mosaic virus (CMV) MP-induced increase in the PD SEL. In vitro experiments showed that CMV MP severs actin filaments (F-actin). Furthermore, through the analyses of two CMV MP mutants, we demonstrated that the F-actin severing ability of CMV MP was required to increase the PD SEL. These results are similar to what has been found in Tobacco mosaic virus MP. Thus, our data suggest that actin dynamics may participate in the regulations of the PD SEL.Key words: plasmodesmata, size exclusion limit, movement protein, actin filaments, F-actin severing  相似文献

In plants, cell-to-cell transport of endogenous and viral proteins and ribonucleoprotein complexes (RNPCs) occurs via plasmodesmata. Specificity of this transport pathway appears to involve interaction between such proteins/RNPCs and plasmodesmal chaperones/receptors. Here, KN1 and the cucumber mosaic virus movement protein (CMV-MP) were used, in a modified phage-display screening system, to identify peptides capable of interacting with proteins present in a plasmodesmal-enriched cell wall fraction. Binding/competition assays and microinjection experiments revealed that these phage-displayed peptides and homologous synthetic oligopeptides function as ligand-specific antagonists of macromolecular trafficking through plasmodesmata. A KN1 peptide antagonist had the capacity to interact with a motif involved in the dilation of plasmodesmal microchannels. Although KN1 could still achieve limited movement through plasmodesmata when this SEL motif was blocked, KN1-mediated transport of KN1-sense RNA was fully inhibited. These findings provide direct support for the hypothesis that KN1 requires, minimally, two physically separated signal motifs involved in the dilation of, and protein translocation through, plasmodesmal microchannels, and provide direct proof that plasmodesmal dilation is a prerequisite for the cell-to-cell transport of an RNPC.  相似文献

胞间连丝是细胞间细胞器,是细胞间通讯的直接途径。一般认为,胞间连丝允许通过物质的分子量上限(SEL)是800~1000 Da.近年来研究的许多证据表明,胞间连丝的SEL随组织种类及其生理状况而异。在某些情况下,它可以允许大分子物质通过,如病毒运动蛋白与胞间连丝相互作用,使病毒通过胞间连丝转移。玉米突变体 kn1基因异常表达的KN1可使包括表皮在内的各层组织结瘤,KN1是细胞间移动的信息物,P-蛋白可由伴胞通过胞间连丝转移到筛管。某些组织中胞间连丝很高的SEL和发育过程胞间连丝SEL的变化可能在植物发育调控中有重要作用。本文对大分子通过胞间连丝转移的机理进行了讨论。  相似文献

Plasmodesmata and the nuclear pore complex (NPC) mediate the selective trafficking of proteins and protein-nucleic acid complexes. The events underlying the translocation of endogenous and viral proteins through plasmodesmata were investigated to further explore the parallels between these cell-to-cell and intracellular communication systems. Studies performed with crosslinked KNOTTED1 (KN1) revealed that a conformational change is required for the cell-to-cell movement of this protein. Microinjection of gold-conjugated KN1 established that, as with the NPC, a combination of protein unfolding and microchannel dilation appears to be involved in protein translocation. However, during this process the extent of microchannel dilation is much less than observed for the NPC, which may reflect a physical limitation imposed by the cell wall. Co-injection of KN1-gold with unbound KN1 or cucumber mosaic virus movement protein (CMV-MP) established that the KN1-gold probe is highly effective at blocking plasmodesmal transport of KN1 and CMV-MP. This result provided the foundation for competition experiments which demonstrated that KN1 and the viral movement proteins of CMV and tobacco mosaic virus likely utilize a common receptor in the pathway for cell-to-cell transport of proteins. A combination of biochemical fractionation methods, an in vitro binding assay founded on the high affinity between KN1-gold and the putative common plasmodesmal receptor, and microinjection techniques were used to isolate plasmodesmal constituents involved in cell-to-cell transport. A model describing the steps involved in protein transport through plasmodesmata is presented.  相似文献

Summary In the multicellular organisms of higher plants, plasmodesmata provide pathways for intimate symplasmic communication between neighboring cells. The arguments summarized in the present review demonstrate that plasmodesmata are diverse and highly dynamic structures. Differences in the plasmodesmal origin and modifications of the plasmodesmal structure and functioning at the various cell interfaces are the basic means which give rise to a complicated and flexibile symplasmic network. This complex communication system is discussed to serve a significant role in the coordinated development and in the concerted physiological functioning of the cells within the plant tissues, organs, and organisms.  相似文献

Tobacco mosaic virus movement protein (TMV MP) is required to mediate viral spread between plant cells via plasmodesmata. Plasmodesmata are cytoplasmic bridges that connect individual plant cells and ordinarily limit molecular diffusion to small molecules and metabolites with a molecular mass up to 1 kD. Here, we characterize functional properties of Nicotiana clevelandii trichome plasmodesmata and analyze their interaction with TMV MP. Trichomes constitute a linear cellular system and provide a predictable pathway of movement. Their plasmodesmata are functionally distinct from plasmodesmata in other plant cel types; they allow cell-to-cell diffusion of dextrans with a molecular mass up to 7 kD, and TMV MP does not increase this size exclusion limit for dextrans. In contrast, the 30-kD TMV MP itself moves between trichome cells and specifically mediates the translocation of a 90-kD beta-glucuronidase (GUS) reporter protein as a GUS::TMV MP fusion. Neither GUS by itself nor GUS in the presence of TMV MP moves between cells. These data imply that a plasmodesmal transport signal resides within TMV MP and is essential for movement. This signal confers selectivity to the translocated protein and cannot function in trans to support movement of other molecules.  相似文献

Plasmodesmata are intercellular channels that establish a symplastic communication pathway between neighboring cells in plants. Owing to this role, opportunistic microbial pathogens have evolved to exploit plasmodesmata as gateways to spread infection from cell to cell within the plant. However, although these pathogens have acquired the capacity to breach the plasmodesmal trafficking pathway, plants are unlikely to relinquish control over a structure essential for their survival so easily. In this review, we examine evidence that suggests plasmodesmata play an active role in plant immunity against viral, fungal and bacterial pathogens. We discuss how these pathogens differ in their lifestyles and infection modes, and present the defense strategies that plants have adopted to prevent the intercellular spread of an infection.  相似文献

Propagation of viral infection in host plants comprises two distinct and sequential stages: viral transport from the initially infected cell into adjacent neighboring cells, a process termed local or cell-to-cell movement, and a chain of events collectively referred to as systemic movement that consists of entry into the vascular tissue, systemic distribution with the phloem stream, and unloading of the virus into noninfected tissues. To achieve intercellular transport, viruses exploit plasmodesmata, complex cytoplasmic bridges interconnecting plant cells. Viral transport through plasmodesmata is aided by virus-encoded proteins, the movement proteins (MPs), which function by two distinct mechanisms: MPs either bind viral nucleic acids and mediate passage of the resulting movement complexes (M-complexes) between cells, or MPs become a part of pathogenic tubules that penetrate through host cell walls and serve as conduits for transport of viral particles. In the first mechanism, M-complexes pass into neighboring cells without destroying or irreversibly altering plasmodesmata, whereas in the second mechanism plasmodesmata are replaced or significantly modified by the tubules. Here we summarize the current knowledge on both local and systemic movement of viruses that progress from cell to cell as M-complexes in a nondestructive fashion. For local movement, we focus mainly on movement functions of the 30 K superfamily viruses, which encode MPs with structural homology to the 30 kDa MP of Tobacco mosaic virus, one of the most extensively studied plant viruses, whereas systemic movement is primarily described for two well-characterized model systems, Tobacco mosaic virus and Tobacco etch potyvirus. Because local and systemic movement are intimately linked to the molecular infrastructure of the host cell, special emphasis is placed on host factors and cellular structures involved in viral transport.  相似文献

Cell-to-cell communication is essential for plant development and adaptation to environmental changes. As a strategy for efficient intercellular communication, plants have evolved a plant-specific symplasmic network connected via plasmodesmata that allows a locally restricted information exchange from cell to cell. A rapid information transfer over long distances is enabled via the phloem transport tubes that pervade the complete plant and thus connect even the most distant organs. While communication by small molecules like metabolites and phytohormones is comparably well studied, the intercellular trafficking of proteins and RNAs has only recently emerged as a novel mechanism of cell-to-cell and long-distance signalling in plants. In particular the non-cell-autonomous and systemic transport pathway for specific RNAs seems to play a key role in the co-ordination of important physiological processes, including virus defence, gene silencing, regulation of development, and nutrient allocation. This review is a summary of the current knowledge on RNAs contained in the phloem long-distance transport system, their transport mechanism, and their potential functions.  相似文献

Cell-to-cell communication is a pivotal process in the determination of cell fate during development and physiological adaptation in response to environmental stimuli. The intercellular trafficking of proteins and RNAs has emerged as a novel mechanism of cell-to-cell signaling in plants. As a strategy for efficient intercellular communication, plants have evolved plant-specific symplasmic communication networks via plasmodesmata (PD) and the phloem. PD are symplasmic channels connecting the cytoplasm of neighboring cells and are responsible for the local exchange of metabolites and signaling molecules. The phloem is the sieve-tube system that allows rapid, long-distance translocation of molecules. Together, PD and phloem conduits have been shown to allow the transport of proteins and RNAs in non-selective or/and selective modes. This review describes the current understanding of macromolecule trafficking through PD and the phloem.  相似文献

The complexity associated with post-translational processing, in terms of protein sorting and delivery is now well understood. Although such studies have been focused almost exclusively on the fate of proteins within the cell in which they are synthesized, recent studies indicate that it is time to broaden this focus to incorporate the concept of intercellular targeting of proteins. Direct evidence is now available that viral and endogenous proteins can be synthesized in a particular cell and subsequently transported into neighboring (or more distant) cells. Plasmodesmata, plasma membrane-lined cytoplasmic pores, are thought to establish the intercellular pathway responsible for this cell-to-cell trafficking of macromolecules (proteins and nucleic acids). These recent findings establish a new paradigm for understanding the manner in which higher plants exert control over developmental processes. We discuss the concept that programming of plant development involves supracellular control achieved by plasmodesmal trafficking of informational molecules, herein defined as supracellular control proteins (SCPs). This novel concept may explain why, in plants, cell fate is determined by position rather than cell lineage. Finally, the circulation of long-distance SCPs, within the phloem, may provide the mechansm by which the plant signals to the shoot apical meristem that it is time to switch to the reproductive phase of its development.  相似文献

Summary Using Lucifer Yellow we have demonstrated that the phloem-loading pathway from the mesophyll to the bundle sheath—vascular parenchyma interface inZea mays source leaves follows a symplasmic route in small and intermediate vascular bundles in control as well as in the green sections of mutant sucrose-export-defective (SXD-1) plants. In the anthocyanin-rich mutant leaf sections, Lucifer Yellow transport was prohibited along the same path, at the bundle sheath—vascular parenchyma interface in particular. Plasmodesmata at the latter interface in SXD-1 anthocyanin-rich leaf sections appear to be structurally altered through callose deposition at the plasmodesmal orifices. We suggest that a transport bottleneck at the bundle sheath—vascular parenchyma interface is thus orchestrated and regulated through callose formation, preventing symplasmic transport across this important loading interface.  相似文献

Frequency, density and branching of plasmodesmata were counted in successive tangential and transverse walls in the cambial zone of tomato stems in order to examine development of the plasmodesmal network in a chronological order. Coincident with progress of cell development, plasmodesmal connectivity increased, both at the xylem- and phloem-side. In transverse walls, the number of secondary plasmodesmata enhanced considerably. The same held for tangential walls, with a superimposed plasmodesmal doubling during the first phase of phloem development. This plasmodesmal doubling was interpreted to result from the deposition of wall material between branched plasmodesmal strands. Structural plasmodesmal development was correlated with production of hydroxyl radicals which control local cell wall alterations. Successive phases of plasmodesmal deployment and modification were distinguished which may coincide with differential functional capacities as documented by intracellular injection of fluorochromes. Diffusion-driven symplasmic transport appeared to be transiently interrupted during cell maturation.  相似文献

Summary Plasmodesmata mediate intercellular transport of proteins, nucleic acids, and small molecules in plants. We show that transiently produced green-fluorescent protein (GFP) trafficked intercellularly in the epidermis of sink leaves, but not of source leaves, in tobacco and cucumber. In contrast, the protein did not traffic in either sink or source leaves of tomato. On the other hand, the protein spread extensively from cell to cell in the epidermis of all leaves and stems ofArabidopsis thaliana as well as in young hypocotyls and cotyledons of tomato and cucumber. GFP could traffic from epidermis to ground tissues in hypocotyls but not in cotyledons of cucumber. GFP fused to a number of mutant forms of the cucumber mosaic virus 3a movement protein (CMV 3a MP) failed to traffic from cell to cell, suggesting that GFP does not have a specific motif for plasmodesmal trafficking. Our data, together with previous findings, indicate that plasmodesmata can mediate both specific and nonspecific intercellular trafficking of proteins. Furthermore, our data suggest that nonspecific protein trafficking is controlled by species-, development-, organ-, and tissue-specific factors. Since GFP can readily traffic from cell to cell, it raises the questions of how metabolites are compartmentalized intercellularly in a plant and of whether some endogenous plant proteins traffic nonspecifically from cell to cell to perform physiological functions yet to be elucidated.Abbreviations CMV cucumber mosaic virus - GFP green-fluorescent protein - MP movement protein - SEL size exclusion limit  相似文献

胞间连丝作为一种细胞质结构将相邻的细胞连系起来而形成植物的共质体。胞间连丝通过调控许多离子和分子的共质体运输而广泛地参与植物的生命活动。胞间连丝的主要构成部分是细胞质膜、连丝小管、以及位于二之间的环层细胞质。这三都很容易在电子显微镜下观察到。细胞骨架的成分(肌动蛋白和肌球蛋白)起到稳定胞间连丝的作用。同时,钙结合蛋白可能具有调节间连丝功能的作用。在胞间连丝里,环层细胞质为大多数溶质提供共质体运输的通道,而有些 共质体运输则可能是通过连丝小管的内腔、连丝小管的壳层、甚或是细胞质膜来实现的。共质体可以细分为数个区块,它们各自允许不同大小的分子(从低于1000到高于10000道尔顿)通过。从发生上看,胞间连丝可以是初生的,也可以是次生的。前是伴随着新细胞壁的形成则产生的,而后则是在已有的细胞壁上产生的。胞间连丝的动态性质还表现在它们的频率是处于变化之中,这是由于组织或植物整体的发育和生理状态决定的。虽然共质体运输的基本形式是扩散,但胞间连丝对于某些离子和分子却是选择性的。在病毒感染细胞时,病毒的移动蛋白作用于胞间连丝的受体蛋白,结果,胞间连丝被显地扩张(其机理尚不清楚)。于是,病毒的移动蛋白连同与之结合在一起的病毒基因组进入毗邻的健康细胞。一些植物源性的蛋白质也能够通过胞间连丝来运输;推测其方式类似于病毒的移动蛋白。有些植物蛋白质本身就是信号分子,它们调节分化和其他活动。与此相反,还有一些植物蛋白质的共质体运输并不是通过特异的方式来实现的。  相似文献

Plant virus cell-to-cell movement and subsequent systemic transport are governed by a series of mechanisms involving various virus and plant factors. Specialized virus encoded movement proteins (MPs) control the cell-to-cell transport of viral nucleoprotein complexes through plasmodesmata. MPs of different viruses have diverse properties and each interacts with specific host factors that also have a range of functions. Most viruses are then transported via the phloem as either nucleoprotein complexes or virions, with contributions from host and virus proteins. Some virus proteins contribute to the establishment and maintenance of systemic infection by inhibiting RNA silencing-mediated degradation of viral RNA. In spite of all the different movement strategies and the viral and host components, there are possible functional commonalities in virus-host interactions that govern viral spread through plants.  相似文献
