家蚕雌性附腺及其Ng突变体的蛋白质组差异研究   总被引:4,自引:0,他引:4  
家蚕雌蛾性附腺在化蛾前2到3天开始大量分泌胶状粘性蛋白,其贮存部迅速地膨大,而其Ng突变体的雌蛾性附腺不能正常分泌胶状粘性物质.分别对家蚕(Bombyx mori)的正常及Ng突变体雌蛾性附腺分泌部组织的蛋白质进行提取,并采用双向凝胶电泳和计算机辅助分析方法,对提取的蛋白质混合物进行分离和比较分析,并对主要差异表达的蛋白质用质谱鉴定.实验结果表明,用银染法,平均每张电泳图谱可以分离约700个蛋白质点,其中大部分的蛋白质点分布在pH 4~8范围内,其分子质量主要集中在30~70 ku区域.比较分析发现一些差异表达蛋白,其中No2,3蛋白质点经质谱鉴定为肌动蛋白A3,该蛋白质只在化蛹后期正常雌性附腺组织中特异表达,而Ng突变体中肌动蛋白A3的缺失,暗示了肌动蛋白A3可能与家蚕雌性附腺的胶状粘性物质的胞外分泌有关.  相似文献

分别对家蚕(Bombyx mori.L)正常及Ng突变体雌蛾件附腺分泌部组织的蛋白质进行提取,并采用双向凝胶电泳和计算机辅助分析方法,对提取的蛋白质混合物进行分离和比较分析。用银染的方法,平均每张电泳图谱可以分离约700个蛋白质点,其中大部分的蛋白质点分布在pH4~8范围内,在分子量上主要集中在30~70kD区域:比较分析发现,有4种蛋白只在正常性附腺组织中特异表达,而有2种蛋白只在Ng突变体的组织中特异表达。另外约有29种蛋白在正常性附腺分泌部组织中的表达水平明显高于Ng突变,而约有15种蛋白在Ng突变体的分泌部组织中表达水平较高。这些差异蛋白质可能与Ng突变的形成和导致这种突变体的性附腺不能正常分泌粘性蛋白的性状有关。  相似文献

The mucous glands of Bombyx pupae secrete glue proteins which attach deposited eggs to the mounting sheet. A mutant of a dominant gene, named no glue (Ng) , produces nonadhesive eggs which have a low capacity for glue-protein synthesis. In the present study it was shown that the mucous glands of Ng silkworms showed rapid degradation of mRNA as well as rRNA during development; this may cause the low capacity for glue-protein synthesis in the mutant organ. In contrast, the mucous glands of normal silkworms showed a significant increase in content of RNA's until the maximum rate of glue-protein synthesis was achieved. The degradation of RNA in the Ng mucous gland was inhibited by actinomycin D injected into the body fluid. Thus it is supposed that the Ng gene codes for a presumptive controller RNA, which would be the mediator of RNA instability in the mucous glands of Ng pupae.  相似文献

The honeybee pupae development influences its future adult condition as well as honey and royal jelly productions. However, the molecular mechanism that regulates honeybee pupae head metamorphosis is still poorly understood. To further our understand of the associated molecular mechanism, we investigated the protein change of the honeybee pupae head at 5 time-points using 2-D electrophoresis, mass spectrometry, bioinformatics, quantitative real-time polymerase chain reaction and Western blot analysis. Accordingly, 58 protein spots altered their expression across the 5 time points (13-20 days), of which 36 proteins involved in the head organogenesis were upregulated during early stages (13-17 days). However, 22 proteins involved in regulating the pupae head neuron and gland development were upregulated at later developmental stages (19-20 days). Also, the functional enrichment analysis further suggests that proteins related to carbohydrate metabolism and energy production, development, cytoskeleton and protein folding were highly involved in the generation of organs and development of honeybee pupal head. Furthermore, the constructed protein interaction network predicted 33 proteins acting as key nodes of honeybee pupae head growth of which 9 and 4 proteins were validated at gene and protein levels, respectively. In this study, we uncovered potential protein species involved in the formation of honeybee pupae head development along with their specific temporal requirements. This first proteomic result allows deeper understanding of the proteome profile changes during honeybee pupae head development and provides important potential candidate proteins for future reverse genetic research on honeybee pupae head development to improve the performance of related organs.  相似文献

The colleterial glands of insects are accessory reproductive structures which produce secretions that are applied to eggs after fertilization and which serve a number of protective functions. The colleterial glands of lepidoptera are of particular interest in the study of the events of cellular differentiation because they undergo rapid development, generally during the pupal adult transformation, and contain highly specialized cells which produce large amounts of a restricted variety of secretory products. The extreme specialization of these organs facilitates parallel studies of differentiation at the biochemical and morphological level. This communication describes the changes in the ultrastructure of cells which will form the protein-secreting segment of the colleterial gland of the moth Actias luna during the period of transition from the undifferentiated state to the acquisition of secretory ability.
An initial stage of general cellular proliferation by mitosis in the presumptive colleterial tissue mass is followed by differentiation of the cells in the absence of further mitosis. Four distinctive cell types are recognized during the phase of differentiation. These types include a chitogenous cell which forms the chitin lining of the main duct, and three cells which cooperate in the formation of a secretory apparatus. Cell A forms two temporary flagella-like structures which assist in the formation of a ductule, which eventually leads from the secretory cell to the main duct. Near the end of the differentiative phase, Cell A degenerates and is phagocitized by Cell B. Cell B becomes the actual secretory element, and acquires cytoplasmic features such as extensive rough endoplasmic reticulum and Golgi apparatus which are associated with synthesis and secretion of protein. The final element, Cell C, remains associated with the ductule which it helps to construct and which traverses its cytoplasm.  相似文献

Cilia and flagella are rare in nongerminal tissues of anthropods, and are generally thought to be restricted to sperm and sensory cells in insects (2). Whitten (5) has reported the presence of kinetosomes at the base of mitotrichia in the dipteran fly Sarcophaga bullata, but reports no evidence of the organization of fibrous elements characteristic of cilia and or flagella. During an ultrastructural analysis of morphogenesis of the colleterial gland of the silk moth Hyalophora cecropia, we found the first example of paired flagella associated with an insect secretory cell. These structures are also unusual in that they serve a temporary role in morphogenesis and subsequently disappear at the terminal stages of differentiation.  相似文献

Proteomic profiles from the wing discs of silkworms at the larval, pupal, and adult moth stages were determined using shotgun proteomics and MS sequencing. We identified 241, 218, and 223 proteins from the larval, pupal, and adult moth stages, respectively, of which 139 were shared by all three stages. In addition, there were 55, 37, and 43 specific proteins identified at the larval, pupal, and adult moth stages, respectively. More metabolic enzymes were identified among the specific proteins expressed in the wing disc of larvae compared with pupae and moths. The identification of FKBP45 and the chitinase-like protein EN03 as two proteins solely expressed at the larval stage indicate these two proteins may be involved in the immunological functions of larvae. The myosin heavy chain was identified in the pupal wing disc, suggesting its involvement in the formation of wing muscle. Some proteins, such as proteasome alpha 3 subunits and ribosomal proteins, specifically identified from the moth stage may be involved in the degradation of old cuticle proteins and new cuticle protein synthesis. Gene ontology analysis of proteins specific to each of these three stages enabled their association with cellular component, molecular function, and biological process categories. The analysis of similarities and differences in these identified proteins will greatly further our understanding of wing disc development in silkworm and other insects.  相似文献

Oothecins, the structural proteins of the egg case of the cockroach Periplaneta americana, have a high content of a few amino acids. This allowed cDNA cloned in the phage M13 to be directly screened for the presence of sequences derived from glycine-rich oothecins by selective DNA sequence analysis. One cDNA fragment codes for the major oothecin synthesised by the left colleterial gland. Part of its derived amino-acid sequence contains a repeated pentapeptide, Gly.Gly.Leu.Gly.Tyr, which is identical to the tandemly repeated consensus pentapeptide in silkmoth chorion proteins. Oothecin synthesis in the left colleterial gland is dependent on the presence of juvenile hormone. A probe from the cDNA segment coding for the major glycine-rich oothecin was used to measure the induction of the corresponding RNA during maturation of the colleterial gland after adult emergence.  相似文献

小菜蛾Plutella xylostella L.是世界性十字花科蔬菜的主要害虫, 已对多种杀虫剂产生抗性, 其中以对拟除虫菊酯类杀虫剂的抗性发展最快。溴氰菊酯是拟除虫菊酯杀虫剂中杀虫毒力最强的品种。我们前期的研究发现, 小菜蛾溴氰菊酯敏感品系(DS)和抗性品系(DR)成虫期的蛋白质双向电泳(2-DE)图谱存在显著差异。本研究通过双向电泳技术从小菜蛾4龄幼虫中分离出89个有明显差异的蛋白点, 从中选出30个进行串联质谱(MALDI-TOF-MS)实验, 并利用蛋白质数据库检索这些在抗性品系中表达而在敏感品系中不表达或者不同品系中差异表达的蛋白质的归属、 性质和功能, 最终成功鉴定出10个蛋白。对其中的3个基因进行了荧光定量PCR验证, 发现这些蛋白质在mRNA水平的表达与在蛋白水平的表达是一致的。这些在溴氰菊酯胁迫下差异表达的蛋白为研究溴氰菊酯的作用靶标和作用机理, 以及筛选与其抗性相关的蛋白质提供了依据。  相似文献

为了探明小峰熊蜂Bombus hypocrita蜂王蛹期发育蛋白质表达调控方面的特点,揭示其发育的分子机理。采用双向电泳法对小峰熊蜂蜂王蛹期发育进行蛋白质组研究,结果在小峰熊蜂蜂王蛹期的白眼期(A期)、褐眼期(B期)和黑眼期(C期)分别检测到81、80和75个蛋白点,特有蛋白质分别为8个、7个和2个,共有蛋白质为61个,A期到B期有4个蛋白质显著上调,5个显著下调,B期到C期有7个蛋白质显著上调,1个显著下调,A期到C期有10个蛋白质显著上调,有4个显著下调。此外,3个蛋白质是在A、B期表达C期关闭,6个蛋白质A、C期表达,B期关闭,5个蛋白质A期关闭,而B、C期表达。初步表明小峰熊蜂蜂王从蛹期发育到成蜂过程中,不仅需要一些保守蛋白质来调控,而且还需要一些特异蛋白质。  相似文献

Whereas in all other vertebrates the Müllerian ducts of genetic males are aborted during development, under the influence of Müllerian-inhibiting substance, in the caecilian amphibians they are retained as a pair of functional glands. It has long been speculated that the Müllerian gland might be the male accessory reproductive gland but there has been no direct evidence to this effect. The present study was undertaken to determine whether the caecilian Müllerian gland secretory proteins would bear antigenic similarity to secretory proteins of the prostate gland and/or the seminal vesicles of a mammal. The secretory proteins of the Müllerian gland of Ichthyophis tricolor were evaluated for cross-reactivity with antisera raised against rat ventral prostate and seminal vesicle secretory proteins, adopting SDS-PAGE, two-dimensional electrophoresis and immunoblot techniques. Indeed there was a cross-reaction of five Müllerian gland secretory protein fractions with prostatic protein antiserum and of three with seminal vesicle protein antiserum. A potential homology exists because in mammals the middle group of the prostate primordia is derived from a diverticulum of the Müllerian duct. Thus this study, by providing evidence for expression of prostatic and seminal vesicle proteins in the Müllerian gland, substantiates the point that in caecilians the Müllerian glands are the male accessory reproductive glands.  相似文献

The proteins from the posterior silk gland of silkworm hybrids and their parents reared under high temperatures were studied by using comparative proteomic and phosphoproteomic analysis. A total of 82.07, 6.17 and 11.76 % protein spots showed additivity, overdominance and underdominance patterns, respectively. Fifteen differentially expressed protein spots were identified by peptide mass fingerprinting. Among these, four spots, including sHSPs and prohibitin protein that were directly relevant to heat response, were identified. Eleven protein spots were found to play an important role in silk synthesis, and nine protein spots expressed phosphorylation states. According to Gene ontology and KEGG pathway analysis, these nine spots played an important role in stress-induced signal transduction. Expression of most silk synthesis-related proteins was reduced, whereas stress-responsive proteins increased with heat exposure time in three breeds. Furthermore, most proteins showed under- or overdominance in the hybrids compared to the parents. The results suggested that high temperature could alter the expression of proteins related to silk synthesis and heat response in silkworm. Moreover, differentially expressed proteins occurring in the hybrid and its parents may be the main explanation of the observed heterosis.  相似文献

The process of mucoprotein (glue) accumulation is described. This substance is secreted by salivary glands of the III instar larvae of Drosophila melanogaster. Granules of the secretory substance appear in the glandular cells 90 hrs after egg laying. Its secretion takes place within 120 hrs. The template RNA of glue proteins appears to be synthesized within the range of 72-85 hrs since actinomycin treatment of larvae at this time suppresses the formation of secretory substance. The portion of secretory proteins amounts to 23-32% of the total gland protein. In a mutant, 13tl, deficient by the absence of pupation, no PAS-positive, staining of glands or secretory granules identified under the phase-contrast microscope was found. A possible participation of fat body in the formation of mucoprotein secretory substance is discussed.  相似文献

目的:研究樟子松突变丛生枝的发生机理。方法:对其正常枝和突变丛生枝进行蛋白质双向电泳,通过ImageMaster2-DE计算机分析系统对结果进行定性、定量分析。结果:共有59个蛋白点在表达的质和量上有变化,其中35个蛋白点在突变丛生枝2-DE图谱中表达量均高于正常枝的蛋白点,20个蛋白点在突变丛生枝2-DE图谱中表达量均低于正常枝的蛋白点,4个蛋白点在突变丛生枝图谱上发生缺失。结论:差异表达的蛋白点可能与突变丛生枝的发生有关。  相似文献

We have previously identified three lesion-mimic mutants, cell death and resistance (cdr), in rice. These mutants induce a series of defense responses, including expression of defense-related genes and high accumulation of phytoalexins, indicating that the cdr mutants are useful materials to study programmed cell death and defense signaling in rice. Here, we carried out a proteome analysis of the cdr2 mutant. Total proteins prepared from the wild type and the cdr2 mutant at three different stages of lesion formation were compared using two-dimensional electrophoresis. We found a total of 37 proteins that were differentially expressed between cdr2 and wild type. Among them, 28 spots were up-regulated and nine were down-regulated in the cdr2 mutant. All the protein spots were identified by mass spectrometric analysis. These differentially regulated proteins included defense-related proteins. In addition, 27 proteins were classified as metabolic enzymes, suggesting that the programmed cell death that occurs in the cdr2 mutant is associated with active metabolic changes. Our study shows that proteome analysis is a useful approach to study programmed cell death and defense signaling in plants.  相似文献

【目的】NEDD8是一种重要的蛋白质翻译后修饰蛋白,对底物蛋白的功能具有重要的调节作用。本研究旨在探索家蚕Bombyx mori中NEDD8的功能。【方法】利用RT-PCR技术,从家蚕Bm N细胞中克隆了家蚕NEDD8完整的开放阅读框。通过实时荧光定量PCR(qRT-PCR)技术检测家蚕NEDD8在不同发育阶段、5龄第3天幼虫不同组织中以及Bm NPV感染Bm N细胞后的相对表达量。通过构建GFP融合表达的重组Bm NPV(B.mori nucleopolyherovirus)感染家蚕Bm N细胞,在共聚焦显微镜下观察NEDD8在细胞中分布情况,用GFP抗体进行Western blot验证。【结果】克隆获得了NEDD8基因。序列分析表明,家蚕NEDD8高度保守,与家蚕泛素蛋白氨基酸序列一致性最高。qRT-PCR分析结果表明,NEDD8在家蚕的不同组织中均有表达,其中头部中表达量最高,其次是丝腺中,而在精巢和卵巢中表达量最低;在家蚕5龄第3天幼虫始到化蛹后第3天NEDD8的表达量开始逐渐增加,化蛾后降至低水平;在家蚕杆状病毒感染Bm N细胞的早期和极晚期NEDD8的表达量都有明显增加。GFP-NEDD8融合表达定位显示NEDD8在Bm N细胞内普遍存在,分布于整个细胞中,并且在感染48 h后存在细胞质内的聚集现象。【结论】NEDD8编码序列在物种间高度保守;NEDD8在家蚕幼虫头部中表达量最高,在化蛹阶段表达量逐渐增加;NEDD8在Bm N细胞内普遍存在并且可能与参与Bm NPV复制。本研究所得结果为进一步研究NEDD8在家蚕中的生物学功能及修饰底物蛋白的作用机制奠定了基础。  相似文献

Gamma irradiation doses applied to inhibit potato sprouting were tested against potato tuber moth larval and pupal stages. Young larvae and pupae were more susceptible to irradiation injuries than older ones. When larvae of different ages were exposed to doses > or = 100 Gy, only 13-35% pupated, but no adult emergence was obtained. The exposure of 1-1.5- or 3-3.5-d-old pupae to 150 Gy induced high level of sterility and remarkable reduction in female mating ability and fecundity, whereas the reduction was less noticeable for 5-5.5-d-old pupae. Postharvest gamma irradiation of potatoes could be considered as an efficient control approach against potato tuber moth larval and pupal infestations.  相似文献

In order to explore the molecular mechanism underlying self-incompatibility (SI) in the apricot (Prunus armeniaca L.) at the proteome level, we examined the style proteomes at different stages of flower development: small bud, big bud, 24h after self-pollination and 24h after cross-pollination with cultivar Badanshui in the SI apricot cultivar Xinshiji and the self-compatible (SC) apricot cultivar Katy by 2D fluorescence difference gel electrophoresis (2D-DIGE) and mass spectrometry (MS). About 1500 style protein spots were detected; 66 were expressed differently in the four stages in Xinshiji. About 1600 style protein spots were detected; 143 were expressed differently in the four stages of flower development in Katy. In Xinshiji, one protein was expressed specifically, four proteins showed up-regulated expression and twenty-nine proteins showed down-regulated expression in the cross-pollinated style compared to the self-pollinated style. Thirteen proteins were identified unambiguously. In Katy, three proteins were expressed specifically, five proteins showed up-regulated expression and thirteen proteins showed down-regulated expression in the cross-pollinated style compared to self-pollinated style. Seven proteins were identified unambiguously. The different reactions of the style at the proteomic level were triggered in Xinshiji and Katy by self pollen and non-self pollen.  相似文献

In the present study, the expression of secretory components and vesicular transport proteins in the canine lacrimal gland was examined and morphometric analysis was performed. The secretory epithelium consists of two types of secretory cells with different morphological features. The secretory cells constituting acinar units (type A cells) exhibited higher levels of glycoconjugates, including β-GlcNAc, than the other cell type constituting tubular units (type T cells). Immunoblot analysis revealed that antimicrobial proteins, such as lysozyme, lactoferrin and lactoperoxidase, Rab proteins (Rab3d, Rab27a and Rab27b) and soluble N-ethylmaleimide-sensitive fusion protein attachment protein receptor (SNARE) proteins (VAMP2, VAMP4, VAMP8, syntaxin-1, syntaxin-4 and syntaxin-6), were expressed at various levels. We immunohistochemically demonstrated that the expression patterns of lysozyme, lactoferrin, Rab27a, Rab27b, VAMP4, VAMP8 and syntaxin-6 differed depending on the secretory cell type. Additionally, in type T cells, VAMP4 was confined to a subpopulation of secretory granules, while VAMP8 was detected in almost all of them. The present study displayed the morphological and histochemical characteristics of the secretory epithelium in the canine lacrimal gland.
