Oleic acid was transformed into 10-hydroxy-8E-octadecenoic acid and 7,10-dihydroxy-8E-octadecenoic acid using Pseudomonas sp. 42A2 NCIMB 40045 in different immobilised supports. Celite R633 gave highest conversions in 48 h: the cellular yield (Y p/x) g products g–1 of cellular protein in the immobilised culture was 5 compared with the free-cell culture Y p/x of 3.8. Conversion of oleic acid in the immobilised cell culture was 50% of the carbon source supplied.  相似文献

Controlled acid hydrolysis of poly-(2,8)-linked homopolymers ofN-acetylneuraminic acid (colominic acid) and its homologous poly-N-glycolylneuraminic acid afforded high yields of the corresponding disaccharides useful in block synthesis of disialylated gangliosides. The poly-N-glycolylanalog was derived from de-N-acetylated colominic acid by two different reaction sequences. The first one involved reaction with acetoxyacetyl chloride followed by de-O-acetylation. The second and most interesting one requiredN-acryloylation and reductive ozonolysis.  相似文献

Starting from labdanolic acid, extracted from Cistus ladaniferus plant, the first synthesis of the natural product 3β-hydroxy-labd-8(17)-en-15-oic acid was achieved, via microbial transformation mediated by Penicillium janczewskii followed by dehydration. The structure of a new labdane diterpene, namely 3β-hydroxy-labdanolic acid, was elucidated by X-ray crystallography and spectroscopic data.  相似文献

The cyanobacteriumSpirulina platensis is one of the most promising sources of the polyunsaturated fatty acid -linolenic acid (GLA). The GLA content ofSpirulina can be enhanced by cultivation under light-dark cycles in the laboratory or outdoors. Thus, in strain BP, the GLA content increased from 1.2 to 1.6% when cultivated under light-dark cycles. Moreover, in the derived mutant Z19, the GLA content reached 2.4% when cultivated outdoors. In the aqueous solution of copper(II) ions, bidentate L-α-alaninehydroxamic acid (CH3CH(NH2)-CONHOHHL) binds cupric ion forming of monodimeric and bis(L-α-alaninehydroxamato)copper(II) complexes. These complexes were studied by potentiometric, ESR and spectrophotometric methods.The ESR studies provide important evidence for the formation of different Cu(II) complexes with L-α-alaninehydroxamic acid, depending on pH. The ESR spectra can be used to follow the appearance of the individual complexes, to estimate the coordination sphere around Cu(II) and to observe the equilibria between different complexes.The solution electronic spectra are reported. The experimental curve was resolved into precise- positioned absorption bands by Gaussian analysis for the bis(L-α-alaninehydroxamato)copper(II) species. These data were used in a weak tetragonal ligand field model to calculate ligand field parameters.The distribution and the relevant stability constants of species present in aqueous solutions were obtained by analytical potentiometry.  相似文献

2-(±)-Cyclopentenecarboxylic acid added to the culture medium is incorporated into two new fatty acids by the growing cells of Bacillus subtilis (ATCC 7059). The new fatty acids, amounting to 24% of the total cellular fatty acids, are identified as hydrocarpic [11-(2′-cyclopentenyl)-hendecanoic] and chaulmoogric [13-(2′-cyclopentenyl)-tridecanoic] by gas-liquid chromatography and mass spectrometry. These C16 and C18 fatty acids are optically active, levorotatory, with the specific rotation of ?50.4° as mixture, thus the optical purity of approximately 80%. This indicates that the optical rotation of these bacterial fatty acids are opposite with that of the fatty acids from plant oils.  相似文献

Summary The effect of propionic acid on poly(-hydroxybutyric-co--hydroxyvaleric)acid P(HB-co-HV) copolymer production byAlcaligenes eutrophus ATCC 17699 supplied with fructose and propionic acid under nitrogen limited conditions was studied. The growth ofA. eutrophus was almost completely inhibited when the concentraion, of propionic acid exceeds 1.5 g/L. Specific production rate of HV unit was highest when propionic acid concentration was 0.5 g/L. In batch culture, pH change occurs in proportion to the consumption of propionic acid. Optimal concentration of propionic acid was maintained during the production phase by using a pH-stat feeding method and a total polymer content higher than 70% and the relative HV content upto 50% could be achieved.  相似文献

Streptomyces albulus PD-1 can co-produce antimicrobial homo-polymers poly(ε-lysine) (ε-PL) and poly(l-diaminopropionic acid) (PDAP). In this study, a novel feeding strategy of citric acid coupled with glucose-(NH4)2SO4 feeding was employed to S. albulus PD-1. When the pH of the culture broth dropped to 4.0, the feeding solution was added continuously to maintain the concentrations of glucose and citric acid at 10 and 4 g L?1, respectively. As a result, the final concentration of ε-PL increased from 21.7 to 29.7 g L?1 and the final concentration of PDAP decreased from 4.8 to 3.2 g L?1. Assays on intracellular nucleotide levels and key enzyme activities were performed to elucidate the underlying regulation mechanism. The addition of citric acid increased NADH/NAD+ ratio and decreased intracellular ATP level; meanwhile, the activities of pyruvate kinase, citrate synthase and isocitrate dehydrogenase decreased while aspartate aminotransferase activity increased. Therefore, we deduced that citric acid feeding resulted in metabolic flux redistribution at the node of phosphoenolpyruvate; the metabolic pathway from phosphoenolpyruvate directed into tricarboxylic acid cycle was weakened and thus PDAP production was inhibited. On the other hand, the metabolic pathway from phosphoenolpyruvate directed into oxaloacetate and l-aspartate was enhanced, thereby improving ε-PL production. This fermentation strategy may be potentially useful in ε-PL production because it can effectively inhibit the formation of by-products, such as PDAP.  相似文献

(p-Chlorophenoxy)isobutyric acid (PCIB) inhibited indole-3-acetic acid (IAA)-induced ethylene production in etiolated mung bean hypocotyl sections. The endogenous level of 1-aminocyclopropane-1-carboxylic acid (ACC) was not significantly affected by PCIB, indicating that PCIB exerted its effect primarily by inhibiting the activity of the ethylene-forming enzyme (EFE). This conclusion was supported by the observations that PCIB inhibited the conversion of exogenously applied ACC to ethylene. The inhibitory effect of PCIB was already evident with 0.05 mM PCIB, and it increased with time after application of the inhibitor. PCIB also significantly inhibited ethylene production in apple fruit tissues, but it only slightly reduced the level of endogenous ACC. Similar to mung bean, EFE activity in apple tissue was significantly inhibited by PCIB. The possibility that PCIB also inhibits auxin-induced ACC synthase activity is discussed.  相似文献

 Eight strains of the genus Aureobasidium obtained from culture collections were tested for their capability to produce poly(β-L-malic acid) (PMA). Four of the tested strains showed positive results. The most productive strain, A. pullulans CBS 591.75, was used to study the production of PMA in stirred-tank reactors. It was found that PMA was mainly produced in the late exponential phase, and the production related positively to glucose consumption. At the beginning of the fermentation the pH increased from 4.0 to about 7.0; subsequently the pH decreased and remained stable at around 3.0–3.5 for several days. Temperatures higher than 25°C were detrimental to PMA production and cell growth. PMA production and cell growth at 20°C and 25°C exhibited no significant differences. PMA production and cell growth were studied under pH-controlled fermentation (at pH 2.0, 4.0, 5.5). The highest PMA production occurred at pH 4.0. PMA production was reduced at pH 2.0 although quite reasonable cell growth occurred at this pH value. Under optimized conditions 9.8 g PMA/l was produced during 9 days of fermentation in the stirred-tank reactors with an overall yield of 0.11 g PMA/g glucose. A procedure for the isolation of PMA and its separation from the other components of the fermentation broth was developed. The isolated PMA was characterized by 1H and 13C-NMR spectroscopy as well as by infrared absorption spectroscopy. Gel-permeation chromatography revealed a relative molecular mass of approximately 3000–5000 by comparison with polyethylene glycol standards. The assembly of lipoprotein(a) (Lp(a)) is a two-step process which involves the interaction of kringle-4 (K-IV) domains in apolipoprotein(a) (apo(a)) with Lys groups in apoB-100. Lys analogues such as tranexamic acid (TXA) or δ-aminovaleric acid (δ-AVA) proved to prevent the Lp(a) assembly in vitro. In order to study the in vivo effect of Lys analogues, transgenic apo(a) or Lp(a) mice were treated with TXA or δ-AVA and plasma levels of free and low density lipoprotein bound apo(a) were measured. In parallel experiments, McA-RH 7777 cells, stably transfected with apo(a), were also treated with these substances and apo(a) secretion was followed. Treatment of transgenic mice with Lys analogues caused a doubling of plasma Lp(a) levels, while the ratio of free:apoB-100 bound apo(a) remained unchanged. In transgenic apo(a) mice a 1.5-fold increase in plasma apo(a) levels was noticed. TXA significantly increased Lp(a) half-life from 6 h to 8 h. Incubation of McA-RH 7777 cells with Lys analogues resulted in an up to 1.4-fold increase in apo(a) in the medium. The amount of intracellular low molecular weight apo(a) precursor remained unchanged. We hypothesize that Lys analogues increase plasma Lp(a) levels by increasing the dissociation of cell bound apo(a) in combination with reducing Lp(a) catabolism.  相似文献

Chicken liver fatty acid synthetase is rapidly inhibited by 5,5′-dithiobis(2-nitrobenzoi acid). The inhibition results from the reaction of 5,5′-dithiobis(2-nitrobenzoic acid) with the cysteine-SH residue of the β-ketoacyl synthetase site. The adjacent pantetheine-SH of the other subunit displaces 2-nitro-5-thiobenzoic acid from the mixed disulfide resulting in the formation of a disulfide bond between the two residues and thereby cross-linking the two subunits. Scatchard analysis of the 5,5′-dithiobis(2-nitrobenzoic acid) inhibition indicated that there are two β-ketoacyl synthetase sites in the homodimer. The mixed disulfide formed between the pantetheine-SH and the cysteine-SH was reduced by 2-mercaptoethanol resulting in restoration of enzyme activity.  相似文献

Summary A new strategy for stereoconservative and stereoselective syntheses of several types of amino acids starting from-functional carboxylic acids employing hexafluoroacetone as protecting and activating reagent is described. Outstanding features of this new method are the mild reaction conditions and the high yields for introduction and cleavage of the protective group allowing sensitive functional groups in the side chain to survive. The effect of various diets containing linoleic and/or -linolenic acids was studied on n-6 fatty acid composition of the rotifer Brachionus plicatilis. The rotifer's abilities for transformations of n-6 fatty acids were evaluated. Diets containing only linolenic acid as n-6 fatty acid induced low levels of other n-6 fatty acids in rotifers while a diet containing also -linolenic acid led to substantial amounts of di homo -linolenic acid in the rotifers through elongation. Desaturation of -linoleic acid to gamma linolenic appears to be the limiting factor of n-6 highly unsaturated fatty acid biosynthesis by the rotifer. Two sets of experiments were compared using different techniques and different sources of -linolenic acid: Spirulina in inert food or borage oil in emulsion with baker's yeast. Rotifers fed with inert diet with Spirulina contained arachidonic acid while those fed with borage oil had very low arachidonic content. High level of n-3 fatty acids incorporated into the diets seemed to exert inhibitory effects on n-6 transformation rate.  相似文献

Summary Myrmicacin (-hydroxydecanoic acid) applied externally inhibited the cytoplasmic streaming ofChara cells in acidic (pH 4.5) but not in neutral (pH 7.5) solution. Application by intracellular perfusion at neutral pH also did not inhibit the streaming. Other carboxylic acids could inhibit the streaming when given as an acidic external solution. Inhibition of the cytoplasmic streaming was concluded to occur due to acidification of the intracellular pH upon application of an acidic external medium of these carboxylic acids. We constructed a metabolically engineered glutamate-independent Bacillus amyloliquefaciens strain with considerable γ-PGA production. It was carried out by double-deletion of the cwlO gene and epsA-O cluster, as well as insertion of the vgb gene in the bacteria chromosome. The final generated strain NK-PV elicited the highest production of γ-PGA (5.12 g l−1), which was 63.2% higher than that of the wild-type NK-1 strain (3.14 g l−1). The γ-PGA purity also improved in the NK-PV strain of 80.4% compared with 76.8% for the control. Experiments on bacterial biofilm formation experiment showed that NK-1 and NK-c (ΔcwlO) strains can form biofilm; the epsA-O deletion NK-7 and NK-PV strains could only form an incomplete biofilm.  相似文献
