The dynamics of changes in the content of four groups of phenolic substances was investigated during the growth cycle of the cell suspension culture ofNicotiana tábacum by means of fractionation. The relative contents of free phenolic acids, their esters, phenolic glycosides, and phenole acids non-extractable with methanol changed in dependence on the growth phase of the culture. A sharp increase, especially in the content of ester- and glycoside-bound phenolics and to a lesser extent also of phenolics belonging to the other two groups, occurred at the end of the lag phase. Then, after a temporary decrease at the early linear phase, the level of phenolics in the three fractions representing bound forms considerably increased again at the late linear and early stationary phases. The synthesized phenolic substances were partially released from the cells into the cultivation medium, which contained 15 to 30 % of the total content of the phenolics in the culture at different phases of the growth cycle. Likely causes of these changes are discussed.  相似文献

The time course of growth, biosynthesis and secretion of different phosphohydrolytic activities by Nicotiana tabacum 1507 cell suspension culture were investigated. It was established that the cell culture under study biosynthesised large amounts of phosphohydrolytic activities during the linear phase of growth for a relatively short period (between the 2nd and 5th days of cultivation). The highest enzyme activity was determined with bis-pNPP which is non-specific substrate for phosphodiesterases and for some nucleases (116.103 U/L at pH 5.7 and 51.103 U/L at pH 8.0). The different phosphohydrolytic activities were distingnished using specific substrates at pH 5.7 (20.103 U/L – 5′-phosphodiesterases, 18,8.103 U/L – 3′-phosphodiesterases and 15,5.103 U/L – phosphomonoesterases) and at pH 8.0 (10,2.103 U/L – 5′-phosphodiesterases, 9,5.103 U/L – 3′-phosphodiesterases and 6,4.103 U/L – phosphomonoesterases). Three techniques of the test-tube fertilization by means of an artificial pollen tube culture are described. The essentials for each individual method are:
  1. Transfer of pollen tubes from the sugar solution onto the placentae in the test tube.
  2. Placing the placentae with ovules onto pollen tube culture.
  3. Precultivation of pollen tubes on small cellophane squares placed on the surface of a semi-liquid medium and transfer of cellophane squares along with the pollen culture to the solid medium, over which finally the placentae with ovules are placed.
By means of all the three techniques viable seeds were obtained in vitro. In an artificial medium pollen tubes are able to maintain their fertilizing ability even after the 24 hours cultivation, i.e. at the time after the formation of the gametes.  相似文献

Summary In order to gain a more complete understanding of the organization of the mitotic apparatus (MA) in the generative cells (GCs) of flowering plants, pollen tubes ofNicotiana tabacum were examined using tubulin immunocytochemistry and Hoechst fluorescence. The observations were then compared with previously published information onTradescantia GCs and the MA of somatic cells. At the onset of division, the prominent microtubule (Mt) bundles characteristic of GCs are reorganized into a more random Mt network. At late prophase/prometaphase, kinetochores appear to interact with this network, resulting in the formation of K-fibers that frequently link in tree-like aggregates. The GC MA takes the form of a distinct spindle and often has pointed, focused poles; the metaphase plate is usually oblique. Karyokinesis involves both anaphase A and B; lengthening of interzonal Mts is accompanied by elongation of the spindle. In late anaphase/early telophase, phragmoplast Mts are formed in association with the proximal face of the sperm nuclei. The phragmoplast remains prominent for some time, so that its Mts as well as another population generated from the distal face of the sperm nuclei constitute the initial sperm cytoskeleton. Comparisons indicate that the spindle in tobacco GCs falls on a continuum of organization between that of somatic cells and the MA ofTradescantia GCs.Abbreviations GC generative cell - MA mitotic apparatus - Mt microtubule  相似文献

The interconversion between 5 to 8-membered cycloalkanones and the corresponding cycloalkanols by a cell suspension culture of Nicotiana tabacum was studied. It was found that the interconversion is at equilibrium and the balance in the equilibrium depends on the number of carbon atoms in the carbocyclic ring of the substrates; the equilibrium in the interconversion of the 6-membered compound lies toward the side of the cycloalkanol, while in the cases of the 5, 7, and 8-membered compounds the cycloalkanones are favoured.  相似文献

Summary Protoplasts ofNicotiana tabacum var. Xanthi were incubated with liposomes containing the plasmid plGVneo23 encoding kanamycin resistance. Transformed protoplasts and calli and plants derived from transformed protoplasts were treated with the demethylating agent 5-azacytidine. Three lines of evidence indicate that 5-azacytidine can increase NPT II activity in transformed cell lines and plants: a) Addition of azacytidine to the protoplast medium increased the proportion of kanamycin-resistant transformants recovered. b) NPT II activity could not be detected in approximately 50% of calli derived from transformed protoplasts although such calli grew slowly on medium containing kanamycin. Treatment of NPT-negative calli with 5-azacytidine restored detectable gene activity and increased the growth rate of the callus in the presence of kanamycin. c) Shoot tips regenerated from transformed calli were either NPT-positive or NPT-negative. When shoots were NPT-negative, treatment with 5-azacytidine restored detectable gene activity and improved growth in the presence of kanamycin.  相似文献

Chlorophyll containing callus cells of Marchantia polymorpha are able to grow under dim illumination in the presence of an organic carbon source and retain the ability to produce polyunsaturated fatty acids (PUFA), including C(20) fatty acids. Highest PUFA production was achieved using 2,4-dichlorophenoxyacetic acid as growth regulator. Inoculum size, illumination intensity, organic carbon source, and ferrous ion are the major factors affecting PUFA productivity. Maximum PUFA productivity is attained under low light intensity, with a photon flux density ca. 20 micromol m(-2) s(-1). Optimal inoculum size and glucose concentration for PUFA production are 8-12% and 20-30 g l(-1), respectively. Ferrous ion can promote PUFA productivity by increasing the intracellular lipid content. Highest productivities for PUFA, arachidonic acid (ARA), and eicosapentaenoic acid (EPA) were 35.0+/-2.1, 6.7+/-0.4 and 6.6+/-0.4 mg l(-1) day(-1), respectively. PUFA production in the M. polymorpha culture is shown to be strongly growth-associated. Environmental stress (osmotic pressure) is ineffective in promoting PUFA productivity. Chitosan, an elicitor, also has no effect on intracellular PUFA content in cultured M. polymorpha cells.  相似文献

The oxidation of gallic acid by an enzyme preparation isolated from the culture medium ofNicotiana tabacum cell suspension could be followed by means of fast scan differential pulse polarography (DPP). Summary The effect of daylenght and temperature for the donor plants (Nicotiana tabacum var. Badischer Burley) on the formation of pollen competent for embryogenesis (P-pollen) by the three possible routes (during normal flower developmentin situ (pollen dimorphism), during cold-treatment of excised flower buds, in cultured anthers) was studied. In all three routes, P-pollen frequency (premitotic pollen, before 1. sporophytic division, PPF) was affected in essentially the same way. At 24 °C and long days, PPF was low and short days had only a slightly increasing effect. At 18 °C and long days, PPF was higher and short days further increased it. Correlated with PPF under the different growth regimes was the percentage of units with more than one vegetative-type nucleus (normal embryos + abortive embryos + multinucleate pollen) in 3 weeks old anther cultures. Under greenhouse conditions, PPF was generally higher than at 24° in growth rooms and showed a maximum in the winter months. Plant age did not affect PPF. These results give further evidence that pollen embryogenesis is predetermined before excision and culture of the pollen or anthers.  相似文献

The results of rheological measurements on 10 different plant cell suspension cultures are presented. Nicotiana tabacum (tobacco) suspension cultures grown in serial batch subculture display high viscosity and power law rheology. This "undesirable" rheology is shown to be a result of elongated cell morphology. The rheology of Papaver somniferum (poppy) cell suspensions is quite different; poppy suspensions behave as Newtonian fluids and have relatively low viscosity (less than 15 cP) at fresh cell densities up to 250 g/L. This flow behavior can be attributed to a lack of elongation in batch-grown poppy cells. A simple correlation for the viscosity as a function of cell density is developed for poppy suspensions up to 300 g fresh weight (FW)/L. It is shown that tobacco cells do not elongate when grown in semicontinuous culture (daily media replacement). These semicontinuously cultured cells have rheological behavior that is indistinguishable from that of poppy, further confirming the dependence of rheology on plant cell morphology. The rheology of a wide variety of other plant suspensions at 200 g FW/L is presented. Most cell suspensions, including soybean, cotton, bindweed, and potato, display low viscosities similar to poppy suspensions. Only carrot and atriplex exhibit slight pseudoplastic behavior which corresponded to a slight degree of cellular elongation for these cultures. This demonstrates that complex rheology associated with elongated cell morphology is much less common than low-viscosity Newtonian behavior. High viscosity in plant cell culture is therefore not an intrinsic characteristic of plant cells but, instead, is a result of the ability to grow cultures to extremely high cell densities due to low biological oxygen demand. The inorganic phosphate of the liquid nutrient medium was completely taken up by freshly inoculated cells of Nicotiana tabacum L. within the first 2 d of culture. Thus intracellular ortho-phosphate concentrations of approx. 0.06 M were accumulated, which upon growth of the cultures were diluted by cell division and subsequent cell growth. Cells from different stages of the growth cycle containing progressively decreasing levels of phosphate were transferred to a phosphate-free medium which normally stimulates the formation of cinnamoyl putrescines. The effect of amino acids on nitrate transport was studied in Zea mays cell suspension cultures and in Zea mays excised roots. The inclusion of aspartic acid, arginine, glutamine and glycine (15mM total amino acids) in a complete cell-culture media containing 1.0 mM NO3 - strongly inhibited nitrate uptake and the induction of accelerated uptake rates. The nitrate uptake rate increased sharply once solution amino acid levels fell below detection limits. Glutamine alone inhibited induction in the cell suspension culture. Maize seedlings germinated and grown for 7 days in a 15 mM mixture of amino acids also had lower nitrate uptake rates than seedlings grown in 0.5 mM Ca(NO3)2 or 1 mM CaCl2. As amino acids are the end product of nitrate assimilation, the results suggest an end-product feed-back mechanism for the regulation of nitrate uptake.  相似文献

The influence of 3,4-dihydroxybenzoic acid (protocatechuic acid), a naturally occurring benzoic acid derivative, on tobacco (Nicotiana tabacum L.) cell and tissue cultures was examined. The response to 0.1, 10 and 1000 µM 3,4-dihydroxybenzoic acid was tested with regards to cell proliferation in leaf explants, callus growth and shoot formation. Effects on shoot and root growth in micropropagated plants were also analysed. The highest concentration of 3,4-dihydroxybenzoic acid strongly inhibited the proliferation of leaf tissues, callus growth, shoot regeneration and root growth in micropropagated plants. On the contrary, the lowest concentration (0.1 µM) showed auxin-like activity by stimulating cell dedifferentiation, callus induction and rooting of leaf tissues. The presence of auxins and cytokinins in the media contrasted 3,4-dihydroxybenzoic acid inhibition of callus growth at all tested concentrations.  相似文献

Summary Burley tobacco varieties (Nicotiana tabacum L.) which differed in degrees of susceptibility to calcium deficiency were analyzed for calcium and oxalic acid content. Calcium accumulated by the total plant top was not related to susceptibility to calcium deficiency. Upper leaves and buds of plants of susceptible varieties contained less calcium per unit weight than did upper leaves and buds of non-susceptible varieties.The upper half of stalks of susceptible varieties contained more oxalic acid per unit weight than did stalks of non-susceptible varieties. The calcium content minus the oxalic acid content in buds and young leaves of the plants of the susceptible varieties was considerably less than that of buds and young leaves of the plants of the non-susceptible varieties. High levels of oxalic acid in the upper stalks and young leaves of varieties susceptible to calcium deficiency apparently induce calcium deficiency by interfering with the translocation and utilization of absorbed calcium.Contribution of the Department of Agronomy, Kentucky Agr.
